Étude génétique et fonctionnelle des Interferon-producing Killer Dendritic Cells

L’idée qu’une cellule puisse effectuer la cytolyse de cellules transformées, comme une cellule Natural Killer (NK), tout en ayant la capacité de présenter des antigènes, comme une cellule dendritique (DC), peut sembler fantaisiste. Cependant, de telles cellules furent bel et bien identifiées chez la souris en 2006. Ces cellules, nommées Interferon-producing Killer Dendritic Cells (IKDC), furent l’objet d’une caractérisation extensive qui révéla leur énorme potentiel immunologique. La combinaison de fonctions associées à des cellules NK et à des DC a doté les IKDC d’un pouvoir antitumoral remarquable. D’ailleurs, il a été démontré que les IKDC sont plus efficaces que les cellules NK pour limiter la croissance tumorale. Ainsi, suite à leur découverte, les IKDC ont suscité beaucoup d’intérêt. Cependant, une controverse émergea sur la nature des IKDC. Plusieurs groupes indépendants tentèrent de reproduire les expériences attestant les fonctions de DC des IKDC, sans y parvenir. De plus, des études additionnelles révélèrent que les IKDC possèdent des similitudes très importantes avec les cellules NK. Ces observations ont mené la communauté scientifique à suggérer que les IKDC sont des cellules NK en état d’activation (aNK). Malgré cette controverse, les caractéristiques antitumorales des IKDC sont si uniques et considérables qu’il est primordial de poursuivre l’étude de ces cellules. Pour y arriver, il est essentiel de déterminer la nature des IKDC et de mettre fin à ce débat. Par la suite, il sera important d’identifier des façons de cibler spécifiquement les IKDC pour permettre leur usage dans le cadre de thérapies antitumorales. Ainsi, l’objectif de cette thèse est de définir l’identité des IKDC, puis de déterminer les facteurs génétiques responsables de la régulation de ces cellules. Nous avons démontré que les IKDC ne sont pas des cellules aNK, contrairement à ce qui avait été suggéré. Nous avons constaté que les IKDC prolifèrent activement et possèdent un phénotype unique, des caractéristiques associées à des cellules NK très immatures. Afin de déterminer si les IKDC peuvent acquérir un phénotype mature, nous avons effectué des expériences de transfert adoptif. Suite à leur injection in vivo, les IKDC acquièrent un phénotype de cellules matures, mais étonnamment, elles se différencient aussi en cellules NK. Ainsi, nous avons révélé que les IKDC sont un intermédiaire dans la différenciation des cellules NK. En parallèle, nous avons démontré que la proportion d’IKDC varie grandement entre des souris de fond génétique différent, indiquant que des facteurs génétiques sont impliqués dans la régulation de ces cellules. Nous avons alors effectué une analyse génétique qui a révélé que les IKDC sont régulées par des facteurs génétiques compris dans une région distale du chromosome 7. Les résultats présentés dans cette thèse constituent une avancée importante pour la recherche sur les IKDC. Ils ont permis de définir la nature des IKDC et d’identifier un intervalle génétique impliqué dans la régulation de ces cellules. Ces découvertes sont des connaissances précieuses pour l’identification des IKDC chez l’Homme et la création de nouvelles thérapies dans la lutte contre le cancer. / The idea that a cell could kill transformed cells, like a Natural Killer (NK) cell, all the while exhibiting also the capacity to present antigens to T cells, like a Dendritic Cell (DC), may seem farfetched. However, in mice, a cell presenting these specific properties was identified in 2006. These cells were named Interferon-producing Killer Dendritic Cells (IKDC) and extensive studies revealed that they were endowed with an important immunological potential. Indeed, the fact that IKDCs exhibit properties of both DC and NK cells conferred them with an exceptional anti-tumor potential. Notably, on a per cell basis, the in vivo anti-tumor activity of IKDCs is more efficient than NK cells. Therefore, following their identification, IKDCs showed great therapeutic promise. However, a debate on the cell lineage origin of IKDCs emerged. Several independent groups could not replicate the finding that IKDCs showed functional antigen-presentation properties similar to DCs. Also, additional studies revealed that IKDCs are very similar to NK cells. These and other observations led the scientific community to believe that IKDCs were activated NK cells. Despite this controversy, IKDCs clearly exhibit a unique and outstanding anti-tumor potential, highlighting the relevance to further explore these cells. We must first close the debate regarding the lineage origin of IKDCs. We subsequently need to identify a means to specifically target IKDCs to facilitate their use in novel anti-tumor therapies. Thus, the objective of my thesis is first, to define the identity of IKDCs and second, to determine the genetic factors implicated in the regulation of these cells. For the first objective, we demonstrated that IKDCs do not represent activated NK cells, as previously suggested. We show that IKDCs are highly proliferative and exhibit a unique phenotype associated with very immature NK cells. In an attempt to verify if IKDCs could acquire a mature phenotype, we conducted an adoptive transfer experiment. We found that, after adoptive transfer, IKDCs adopt a mature phenotype, but also surprisingly differentiate into NK cells. These findings indicate that IKDCs represent an intermediate in NK-cell differentiation. For the second objective, we demonstrated that the IKDC proportion was highly variable between strains of different background origins, indicating that these cells are regulated by genetic factors. A genetic study revealed that genetic factors in distal arm of chromosome 7 associate with the proportion of IKDCs. The results presented in this thesis represent an important breakthrough for the research on IKDCs. They allowed to define the cell lineage origin of IKDCs and to identify a genetic region involved in the regulation of this cell type. These discoveries are valuable knowledge for the identification of human IKDCs and the development of novel anti-tumor therapies.

Syfte: De globala växthusutsläppen är en stor fråga både för landet och världen. Därför måste man hitta alternativ samt lösningar till att producera förnyelsebar energi. En vision för förnyelsebar energi är att producera fastighetsnära förnybar energi. Målet med studien var att undersöka förnyelsebar energi för egenproducering i nybyggnationer med hänsyn till certifieringen för Miljöbyggnad för att bidra till en mer positiv utveckling i miljöarbetet. Metod: Studien är en kvalitativ inriktad forskning. För att lyckas besvara frågeställningar samt uppnå studiens mål samlades data genom semistrukturerade intervjuer och dokumentanalys. Urvalet av respondenter baserades på yrkesroll på det valda företaget. De dokument som analyserat var en checklista och en intern handbok som finns i det samarbetande företaget samt dokument från Sweden Green Building Council som behandlar Miljöbyggnad. Resultat: Arbetet kring förnyelsebar energi är något som prioriteras trots den höga engångskostnaden, eftersom det finns en långsiktig lönsamhet. Kommunikationen är därför viktig under projekteringen men att det finns bristande faktorer i efterarbetet. Nödvändigtvis behöver man inte installera elproduktionsanläggningar i Miljöbyggnad då det endast är en del av kravet på Guld-nivå. Ur ett hållbarhetsperspektiv bör man ändå installera det vid nyproduktion och komplettera det med mätinstrument till anläggningarna för att följa upp resultatet. Konsekvenser: Den ekonomiska aspekten har en stor roll i projekteringen då det kan hindra hållbarhetslösningar. Därför är ekonomiska stöd inom energifrågor ett bra incitament för företag. Miljöbyggnad är sedan ett bra stöd i projektering för nyproduktioner eftersom en certifieringsprocess inte alltid är nödvändig. För att utveckla strukturarbetet bör kunskapsöverföring prioriteras mer. Begränsningar: Studien begränsas till Miljöbyggnad där enbart energiindikatorer har studerats. Fallstudien är begränsad till flerbostadshus för att minska studiens omfattning. De resultat och rekommendationer som presenteras är generellt giltiga för de företag som har liknande arbetssätt och rutiner. / Purpose: Global greenhouse emissions are a major issue for both the country and the world. Therefore, one has to find alternatives and solutions to produce renewable energy. A vision for renewable energy is to produce property-related renewable energy. The aim of the study was to investigate renewable energy for self-production in new construction, taking into account the certification for Miljöbyggnad to contribute to a more positive development in environmental work. Method: The study is a qualitatively oriented research. To succeed in answering questions and attaining the objective of the study, data were gathered through semi-structured interviews and document analysis. The selection of respondents was based on the professional role of the chosen company. The documents that were analyzed were a checklist and an internal manual available in the cooperating company as well as documents from the Sweden Green Building Council that deals with Miljöbyggnad. Findings: The work on renewable energy is something that is prioritized despite the high one-time cost, as there is long-term profitability. Communication is therefore important during the design process, but there is a lack of factors in the finishing work. You do not necessarily need to install electricity production facilities in Miljöbyggnad as it is only part of the requirement for the Gold level. From a sustainability perspective, one should nevertheless install it in new production and supplement it with measuring instruments for the plants in order to follow up the result. Implications: The economic aspect has a major role in the design as it can prevent sustainability solutions. Therefore, financial support in energy issues is a good incentive for companies. Miljöbyggnad is then a good support in project planning for new production since a certification process is not always necessary. To develop the employee work structure, knowledge transfer should be given more priority. Limitations: The study is limited to Miljöbyggnad where only energy indicators have been studied. The case study is limited to apartment buildings to reduce the scope of the study. The results and recommendations presented are generally valid for those companies that have similar working methods and routines.

Bioatividade e efeito residual de nanoformulações de nim sobre Spodoptera frugiperda (J.E. Smith) / Bioactivity and residual effect of neem nanoformulations to Spodoptera frugiperda (J.E. Smith)

Na tentativa de reduzir a biodegradação dos compostos ativos do nim e melhorar seu efeito residual, extratos de nim foram nanoformulados com diferentes tipos de nanoparticulas e de polímeros. Estas nanoformulações foram testadas sobre Spodoptera frugiperda (J. E. Smith) e sua eficiência comparada a um produto comercial. Os polímeros poli--caprolactona (PCL), poli-- hidroxibutirato (PHB) e polimetilmetacrilato(PMMA), foram utilizados nas quantidades 0,25, 0,50 e 0,75g, e em dois tipos de nanopartículas (cápsulas e esferas). Foram utilizados Tween®80 e polivinil álcool (PVA) como agentes tensoativos. As nanoformulações foram produzidas em suspensão aquosa e em pó, sendo diluídas para aplicação. Essas nanoformulações foram aplicadas sobre dieta artificial oferecida a lagartas de segundo instar de S. frugiperda, e em plantas de milho das quais as folhas foram cortadas e fornecidas a lagartas de primeiro instar. A eficiência de diferentes nanoformulações foi testada considerando a mortalidade e o peso 10 dias após o tratamento. Quando aplicado em dieta artificial foi observada redução no peso larval. Entretanto, quando aplicadas em folhas de milho, quatro nanoformulações causaram mortalidade entre 40 a 46%. Essas quatro nanoformulações foram avaliadas quanto à preferência alimentar e efeito residual. Foram conduzidos três bioensaios, nos quais as folhas de milho tratadas com as quatro nanoformulações foram oferecidas às lagartas aos 1, 3 e 7 dias após a pulverização (DAP), considerando-se mortalidade e peso das lagartas após 10 dias. Simultaneamente, foram realizados testes de preferência alimentar com dupla escolha (tratamento x controle) com discos foliares de milho aos 1, 3 e 7 DAP. A preferência alimentar foi determinada por meio de um índice de preferência, calculando-se a área foliar consumida pela lagarta após 24h. O efeito residual das nanoformulações foi menor que do produto comercial avaliado. Apenas uma nanoformulaçao e Organic Neem® apresentaram fagodeterrência a S. frugiperda, mas somente em 1 DAP. Embora as nanoformulações tenham causado mortalidade e redução de peso larval, sua eficiência foi inferior a do produto comercial. / Neem extracts were nanoformulated with several types of nanoparticles and polymers in a tentative to reduce the biodegradation of neem active compounds and improve their residual effect. These nanoformulations were tested against Spodoptera frugiperda and their efficacy compared to a commercial product. The polymers poly(-caprolactone) (PCL), poly(- hidroxibutirate) (PHB) and poly(methyl methacrylate) (PMMA) were used in the quantities 0.25, 0.50 and 0.75g and two sorts of nanoparticles (capsules and spheres). Tween®80 and poly(vinyl alcohol) (PVA) were utilized as surfactants. The nanoformulations were produced as aqueous suspension and powder and further diluted before application. These nanoformulations were sprayed on artificial diet offered to second instars of S. frugiperda, and on corn plants which had their leaves offered to first instars. The efficacy of the different formulations was tested by assessing larval mortality and weight 10 days after treatment. When applied to artificial diets, reduced larval weight was observed, but four of them caused 40 to 46% mortality when applied on corn plants. The four most effective nanoformulations were further tested to evaluate their effects on S. frugiperda larval feeding preference and their residual effect. Three bioassays were set up using corn leaves treated with these, wich were offered to larvae at days 1, 3 and 7 after spraying (DAS), having the larval mortality and weight assessed 10 days after larval exposure. Simultaneously, choice tests for larval feeding preference (treatment x control) were carried out using corn leaf discs 1, 3 and 7 DAS. Feeding preference was evaluated by using a preference index calculated after the assessment of the leaf area fed by the larva 24h after exposure. The residual effect of the neem nanoformulations was lower than the commercial product evaluated. Only one nanoformulation and Organic Neem® were feeding deterrents to S. frugiperda, but only in the first day after the treatment. Although the nanoformulations caused larval mortality and weight reduction, their efficacy was not any better than that of the commercial product tested.

Identificação e quantificação do gene pirrolnitrina (prnD) em Terra Preta Antropogênica da Amazônia por PCR em tempo real / Identification and quantification of pyrrolnitrin gene (prnD) in Anthropogenic Dark Earth by Real-time PCR

A Terra Preta Antropogênica (TPA) é considerada um dos solos mais férteis do mundo e recebe essa denominação por ser originada da ação antrópica, provavelmente de populações pré-colombianas que viveram nestes sítios arqueológicos. O crescente aumento por agricultura sustentável torna a utilização de bactérias produtoras de antibióticos uma alternativa de controle para doenças de plantas. Pirrolnitrina (PRN) é um antibiótico que tem ampla atividade antimicrobiana produzida por várias estirpes de Burkholderia e Pseudomonas que foram isoladas de diferentes solos. Entretanto, não se tem conhecimento de isolados bacterianos de TPA que produzem PRN, assim com, sobre a ecologia e freqüência do gene para este antibiótico. A PRN é codificada por um operon composto por 4 genes sendo o gene prnD responsável por catalisar a oxidação que forma a pirrolnitrina. Neste trabalho, foi estudado o gene prnD através de métodos dependente e independente de cultivo. Foram utilizados isolados bacterianos para detectar o gene prnD através de PCR convencional e a identificação feita pelo seqüenciamento. As bactérias com amplificação positiva tiveram suas sequências do gene analisadas no programa MOTHUR o qual reuniu as em 10 grupos. Um representante de cada grupo foi empregado no teste de antagonismo contra o fitopatógeno Fusarium oxysporum. Amostras de solo de TPA e seus solos adjacentes foram coletados de dois sítios: Caldeirão Capoeira (floresta secundária por mais de 20 anos) e Cultivado (cultivado com mandioca por pelo menos 30 anos). Os DNAs totais das amostras de solo extraído foram usados como molde nas reações de PCR quantitativo em tempo real para verificar a abundância do gene prnD nas amostras de solo. Em amostras de solo também foi quantificado o gene 16S rRNA. No estudo dependente de cultivo, do total de 219 isolados (175 Burkholderia e 44 Pseudomonas), 60 isolados do gênero Burkholderia e 3 de Pseudomonas exibiram amplificação positiva. A análise filogenética do gene prnD mostrou que a maioria das seqüências obtidas deste estudo não agruparam com as seqüências do banco de dados do GenBank indicando que há diversidade do gene prnD nos isolados de solos amazônicos e que podem ser distintos dos descritos anteriormente. O teste de antagonismo demonstrou que isolados com potencial genético para produção de pirrolnitrina também são bioativos contra Fusarium oxysporum, apresentando forte atividade antimicrobiana. No estudo independente de cultivo, no sítio Caldeirão Cultivado, solo de TPA apresentou maior número de cópias do gene prnD e do gene 16S rRNA em relação ao solo ADJ. No sítio Caldeirão Capoeira, o solo adjacente apresentou maior quantidade do gene prnD (1,74x105 cópias/g solo) que o solo de TPA (2,48x104 cópias/ g de solo), no entanto, na TPA as bactérias totais foram mais abundante. Estes resultados evidenciam que a metodologia de PCR quantitativo em tempo real desenvolvida foi altamente sensível e específica permitindo a detecção de diferenças sensíveis e significativas entre os solos na quantificação do gene prnD. A abundância do gene prnD correlacionou significativamente com parâmetros químicos do solo tais como pH, fósforo, cálcio, magnésio e micronutrientes / Anthropogenic Dark Earth (ADE) is considered one of the world\'s most fertile soils and receives this name because it originated from human action, probably by pre-Columbian populations who lived in these archaeological sites. Because of the common trend in agriculture towards sustainability antibiotic-producing bacteria is an alternative for biocontrol to plant diseases. Pyrrolnitrin (PRN) is a broad-spectrum antibiotic produced by various strains of Burkholderia and Pseudomonas that have been isolated from different soils. However, little is known about PRN-producing bacteria screened from ADE, even as the ecology and frequency of pyrrolnitrin gene. PNR is encoded by an operon comprised by four genes and prnD gene is responsible for catalyze the oxidation to form pyrrolnitrin. In this work, we studied the gene prnD through culture dependent and independent methods. Conventional PCR were established to detect prnD gene in bacterial isolates screened in ADE and their adjacent soils (ADJ) and identification were done by sequencing. Based on the results generated by MOTHUR prnD sequences from isolates were grouped into 10 groups. A representative of each group was used in the test of antagonism against the plant pathogen Fusarium oxysporum. Soil samples of ADE and its adjacent soils were collected from two sites: Caldeirão Capoeira (secondary forest for over 20 years) and Caldeirão Cultivado (cultivated with cassava for at least 30 years). The total DNA extracted from soil samples was used as template in quantitative PCR reactions to determine the abundance of prnD gene. In soil samples was also quantified the 16S rRNA. In the study culture-dependent, in a total of 219 isolates (175 Burkholderia and 44 Pseudomonas), 60 isolates of Burkholderia and 3 Pseudomonas exhibited positive amplification for prnD gene. Phylogenetic analysis of prnD gene showed that most of the sequences obtained in this study grouped distinctly from sequences of the GenBank database. It indicates that there is diversity in prnD gene of isolates from amazonian soils and they may differ from those previous described. The antagonism test showed that isolates with genetic potential for production of pyrrolnitrin are also bioactive against Fusarium oxysporum, exhibiting strong antimicrobial activity. In culture-independent study, the site Caldeirão Cultivado, ADE soil showed higher copies number of prnD gene and 16S rRNA gene than in ADJ soil. At the site Caldeirão Capoeira, surrounding soil had a higher amount of prnD gene (1.74 x105 copies / g soil) than ADE soil (2.48 x104 copies / g soil), however, in ADE total bacteria was more abundant. These results show that the real-time PCR assay developed in this study was highly sensitive and specific enabling the detection of sensitive and significant differences between soils in the quantification of prnD gene. As Enfermidades Transmitidas por Alimentos representam um crescente e relevante problema de saúde pública. Além do prejuízo social, a contaminação de alimentos com microrganismos patogênicos gera um enorme prejuízo econômico. Técnicas de Análise de Risco permitem mensurar de forma mais adequada o impacto dos microrganismos contaminantes de alimentos na saúde da população. Uma Análise de Riscos, associada a uma combinação patógeno-alimento, envolve três passos: avaliação do risco, gestão do risco e comunicação do risco. Uma das etapas da avaliação do risco é a avaliação da exposição, baseada em dados sobre freqüência e nível de contaminação dos alimentos pelo patógeno avaliado no alimento em questão, o nível atingido pelo patógeno no momento do consumo e os padrões de consumo. Os produtos cárneos são os principais alimentos responsáveis pela veiculação de patógenos ao homem e os microrganismos de maior relevância nestes produtos são Listeria monocytogenes, Salmonella spp., Campylobacter spp. e Escherichia coli produtora de toxina de Shiga. O objetivo do presente estudo foi levantar informações qualitativas e quantitativas desses quatro patógenos em produtos cárneos (salsicha bovina, lingüiça suína, carne bovina moída e coxa de frango) comercializados no município de São Paulo, de forma a contribuir com dados para futuras avaliações de risco em relação a estes microrganismos nestes produtos. Das 552 amostras de produtos cárneos analisadas, L. monocytogenes foi o patógeno isolado com maior freqüência, sendo detectado em 48,7% das amostras, seguido por Campylobacter spp. em 6,0% e Salmonella spp. em 5,8%. E. coli produtora de toxina de Shiga não foi detectada em nenhuma das amostras estudadas. Listeria monocytogenes foi detectada em todos os tipos de produtos cárneos estudados, com freqüências mais elevadas nas amostras de carne bovina moída (59,4%), seguido de coxa de frango (58,0%), lingüiça suína (39,8%) e salsicha bovina (37,7%). Na maioria das amostras (94,4%), as contagens de L. monocytogenes foram inferiores a 102 UFC/g. As cepas de L. monocytogenes apresentaram ampla distribuição, sendo detectados os quatro grupos de sorotipos: 28,7% pertenceram ao Grupo 1 (sorotipos 1/2a e 3a), 21,0% ao Grupo 2 (sorotipos 1/2c e 3c), 17,0% ao Grupo 3 (sorotipos 1/2b, 3b e 7) e 13,8% ao Grupo 4 (sorotipos 4b, 4d e 4e). Salmonella spp. foi detectada em 32 amostras, sendo 20 (14,5%) de lingüiça e 12 (10,6%) de coxa de frango. As contagens foram baixas, variando de 3,0 a 9,3x10 NMP/g e os sorovares mais freqüentemente isolados foram S. Typhimurium (28,1%), S. Enteritidis (12,5%), S. Derby (12,5%) e S. I 4,[5],12:i:- (12,5%). Campylobacter spp. foi detectado em 33 amostras (6,0%), sendo 27 de coxa de frango (19,6%) e seis amostras de carne moída (4,3%). A presença de L. monocytogenes, Salmonella spp. e Campylobacter spp. nos produtos cárneos analisados representa um risco à saúde da população. O consumo destes produtos quando submetidos à cocção inadequada e/ou a contaminação cruzada com outros alimentos pode levar a ocorrência de Enfermidades Transmitidas por Alimentos. / Foodborne Diseases represent an increasingly important public health problem. Besides the social losses, contamination of food with pathogenic microorganisms generates an enormous economic damage. A more accurate measurement of the impact of microorganisms in food health can be achieved using Risk Analysis techniques. A risk analysis is composed by three elements: risk assessment, risk management and risk communication. One of the four steps of a risk assessment is the exposure assessment, based on data on frequency and level of contamination of a food by the pathogen under evaluation, levels of the pathogen in the food at the time of consumption and consumption patterns. Meat products are the main vehicles of pathogens to humans, where Listeria monocytogenes, Salmonella spp., Campylobacter spp. and Shiga toxin-producing Escherichia coli are the most relevant pathogens. The aim of this study was to obtain qualitative and quantitative information on these four pathogens in four types of meat products (beef sausage, pork sausage, ground beef and chicken leg) marketed in the city of Sao Paulo in order to contribute with data for future risk assessments for these microorganisms in these products. L. monocytogenes is the most frequent pathogen in the 552 samples of meat products analyzed, being detected in 48.7% of the samples, followed by Campylobacter spp. 6.0% and Salmonella spp. 5.8%. Shiga toxin-producing E. coli was not detected in any sample. L. monocytogenes was detected in all types of meat products, with highest frequency in ground beef (59.4%), followed by chicken leg (58.0%), pork sausage (39.8%) and beef sausage (37.7%). In most samples (94.4%), the counts of L. monocytogenes were below 102 CFU/g. L. monocytogenes strains were widely distributed in the four groups of serotypes: 28.7% belonged to Group 1 (serotypes 1/2a and 3a), 21% to Group 2 (serotypes 1/2c and 3c), 17% to Group 3 (serotypes 1/2b, 3b and 7) and 13.8% to Group 4 (serotypes 4b, 4d and 4e). Salmonella spp. was detected in 32 samples, being 20 (14.5%) of pork sausage and 12 (10.6%) of chicken leg. The counts were low, ranging from 3.0 to 9.3 x 10 MPN/g and the most frequent serovars were S. Typhimurium (28.1%), S. Enteritidis (12.5%), S. Derby (12.5%) and S. I 4, [5], 12: i: - (12.5%). Campylobacter spp. was detected in 33 samples (6.0%), being 27 of chicken leg (19.6%) and six samples of ground beef (4.3%). The presence of L. monocytogenes, Salmonella spp. and Campylobacter spp. in the tested meat products represent a risk to health. The consumption of inadequately cooked products and/or subjected to cross-contamination with other foods may lead to occurrence of foodborne diseases.

Esta pesquisa aborda as políticas territoriais implementadas no Distrito Federal com foco na área de expansão urbana de Águas Claras, levando em consideração os diversos fatores que contribuíram e contribuem para a produção deste espaço como tal, sua dinâmica e suas transformações. A produção do espaço no Distrito Federal intermediada pelo Estado proporciona áreas de expansão urbana, que são definidas em diferentes períodos, direcionando o mercado imobiliário para uma área pré-estabelecida, até que esta área fique saturada, deslocando esse mercado para outra área determinada pelo Estado, dando continuidade nesse processo, proporcionando sempre novas áreas para a reprodução do capital. Águas Claras surge como exemplo dessas áreas de expansão urbana no DF, parte integrante deste processo, que são estabelecidas a partir de uma demanda de ordem política e econômica. É importante salientar a propriedade da terra, visto que, as terras onde se encontra Águas Claras anteriormente pertenciam ao Governo do Distrito Federal e foram vendidas aos promotores imobiliários com o discurso de construir um local para habitação da classe média entre duas áreas já urbanizadas. Com isso, ao longo desta pesquisa, percebe-se que Águas Claras se materializa como sendo mais um espaço de realização de reprodução do capital no DF, porém com características peculiares que tornam o enfoque desta pesquisa interessante e diferente das outras análises feitas no Distrito Federal. / This research boards the territorial politics implemented in the Distrito Federal with focus in the urbane expansion of Águas Claras, taking into account several factors that contributed and still contribute to the production of this space such as its dynamic and transformations.The production of the space in the Distrito Federal is intermediated by the State. This intermediation provides areas of urbane expansion, which are defined in different periods, taking the building market for an established area, until this area is saturated, moving this market for another area determined by the State, giving continuity in this process, always providing new areas for the reproduction of the capital. Águas Claras is an example of an area in urbane expansion in the DF. Águas Claras is integrant part of this process, which is established from a demand of political and economical order. It is important point out that the property of the lands where Águas Claras is situeated previously were belonging to the Government of the Distrito Federal and they were sold to the property promoters with the speech of building a place for dwelling of the middle class between two already built-up areas. With that, along this research, it is realized which Águas Claras it materializes like being one more space of realization of reproduction of the capital in the DF, however with special characteristics that make the approach of this research interesting and different from other analyses done in the Federal area.
