Peripheral and central pathways linking metabolic status and reproduction in male sheep

[Truncated abstract] Reproductive activity is affected by external factors such as photoperiod, social cues, stress and nutrition, all of which can alter the pulsatile activity of the GnRH neurons, which is the major neuroendocrine system used by the brain to control gonadal function. In the male Merino sheep, nutrition is one of the most powerful factors that affect pulsatile LH secretion, used commonly to bioassay GnRH neuronal activity. More accurately, the reproductive system responds to “metabolic status”, rather than “nutrition”, and the three factors that contribute to metabolic status are food intake, the amount of body reserves and the rate of energy expenditure ... In this thesis, I tested the general hypothesis that the metabolic hormones and hypothalamic neuropeptides that are known to control food intake also mediate the effect of metabolic status on the activity of the GnRH neurons ... In conclusion, the results from my experiments provide some insight into the mechanisms by which metabolic status affects reproductive activity in male sheep. Plasma insulin, which changes with alterations in metabolic status, appears to play a critical role in the regulation of GnRH neuronal activity. The level of leptin seems to have a permissive role only in lean animals. Orexins acting via OX2 receptors could be involved in the activation of reproductive function following an acute increase in nutrition. However, the neuropeptidergic systems can not be ruled out because they might be involved in very early steps of responses to nutrition.

Objetivou-se com este trabalho avaliar o consumo, parâmetros ruminais e produção de calor pela ingestão de alimento em carneiros Santa Inês confinados, recebendo tratamentos sem e com inclusão de 2 e 4% de farinha de algas marinhas Schizochytrium sp. em substituição ao grão de milho moído, assim como a digestibilidade e produção de gases in vitro das dietas. Foram utilizados 6 carneiros castrados com 55,6 ± 5,2 kg de peso corporal, providos de cânula ruminal e alojados em baias individuais. O delineamento experimental foi o quadrado latino duplo 3 x 3, tendo as dietas relação volumoso:concentrado 40:60, em que os tratamentos foram: controle D0, sem inclusão de farinha de algas marinhas; D2 e D4 contendo 2 e 4% de farinha de algas marinhas na matéria seca respectivamente, em substituição ao grão de milho moído. Cada período experimental teve duração de 21 dias com 14 dias de adaptação aos tratamentos e 7 dias de coleta. Os dados foram analisados por intermédio do Software R (versão 3.2.2), sendo as comparações das médias feitas pelo teste de Tukey a 5% de significância. This trial aimed to evaluate intake, ruminal parameters and heat production by feed intake in confined Santa Inês rams, receiving treatments without and with inclusion of 2 and 4% of marine algae Schizochytrium sp. meal, replacing ground corn grain, as well as in vitro digestibility and gas production. Six castrated rams with 55.6 ± 5.2 kg of body weight, provided with ruminal cannula, were confined in individual pens. The experimental design was double Latin square 3x3, where diets had a roughage:concentrate ratio of 40:60, in which the treatments were: control D0, without marine algae meal inclusion; D2 and D4 containing 2 and 4% marine algae meal in dry matter respectively, replacing ground corn grain. Each experimental period had duration of 21 days, with 14 days of adaptation to treatments and 7 days of collection. The data were analyzed using R Software version 3.2.2 and comparison between treatments means made with Tukey test at 5% of significance. O critério de exclusão utilizado foi à identificação de pacientes com dados insuficientes relativos ao internamento, que inviabilizassem a verificação das reações. A análise da evitabilidade das RAMs foi realizada segundo os critérios de Schumock e Thornton. RESULTADOS: Foram incluídos no estudo 110 pacientes, representando 7,75% dos casos existentes no banco, com 130 medicamentos implicados e 135 reações adversas, apresentando uma média de 1,22 reações/paciente. O sexo feminino representou 60,9% dos casos de admissão hospitalar por RAM. A mediana de idade dos pacientes foi de 48 anos. As principais classes terapêuticas envolvidas foram: tuberculostáticos, imunossupressores, antirretrovirais, antibióticos, antipsicóticos, anticonvulsivantes e antiinflamatórios não-esteroidais. 32,8% das RAMs foram classificadas como preveníveis. CONCLUSÕES: As características da população admitida por RAM corroboram com os dados descritos na literatura e foram influenciadas pelo perfil de atendimento da Instituição. A avaliação da evitabilidade das RAMs foi uma estratégia que favoreceu a identificação dos casos e é útil para o delineamento de ações futuras para prevenção de danos aos pacientes, objeto principal da farmacovigilância. METHODS: It was a retrospective longitudinal study on patients admitted for ADRs was performed from December 2000 to December 2016, aiming at characterizing the individuals, medications, adverse effects, outcomes and preventability. The study included all patients admitted for ADRs involving notification of the Institution’s Pharmacovigilance (PV) Center. The exclusion criterion was the patients with insufficient hospitalization data, which would hinder and/or preclude the assessment of the reactions. The predictability of ADRs was based on the criteria of Schumock and Thornton. RESULTS: The study included 110 patients, representing 7.75% of the existing cases in the database, with 130 implicated drugs and 135 adverse reactions, for an average of 1.22 reactions/patient. Women represented 60.9% of hospital admission cases with an ADR. The mean patient age was 48 years old. The main drug classes involved were tuberculostatics, immunosuppressants, antiretrovirals, antibiotics, antipsychotics, anticonvulsants, and nonsteroidal anti-inflammatories. 32.8% of adverse reactions were classified as preventable. Simulation-Based Study to Quantify Data-Communication Benefits in Congested Airport Terminal Area

The scope of this study was to evaluate the impact of the air traffic controller-to-pilot communication standard known as CPDLC or Data-Communication on the future air traffic operations. The impact was evaluated from the double viewpoint of airport delays and air traffic controllers' workload. RAMS simulation software is used to perform all the runs and from its output data the values of terminal area delays and controllers workload are obtained. The New York Metroplex terminal area was used as a case study. Because of its complexity, where three major airports (i.e. JFK, Newark, and La Guardia) interact and constraint each other, this area was particularly interesting to be studied and the data analyzed gave a valuable insight on the possible future impact of Data-Communication in congested terminal areas. The results of the study, based on some previous man-in-the-loop simulations performed by the FAA in the nineties, showed that significant potential benefits could be obtained with the complete implementation of such technologies in the workload experienced by air traffic controllers. Moreover some small but not negligible benefits were obtained in the total delays accrued by each airport studied. On the other hand, the simulations of the future demand predicted by the FAA demonstrated that without a significant increment in capacity or limitation on the traffic growth intolerable delays would be recorded across the NAS in the future. For the complexity of the simulation model calibration and for the very time-consuming run time not all the scenarios described in the methodology were tested, demonstrating the weakness of RAMS as a ground simulation model. / Master of Science

Effects of breed and ram exposure on Spring estrous behavior and Summer fertility in domestic ewes

The present studies were conducted to check the effects of acute ram introduction into a flock of anestrus ewes in Virginia. Ewes were bled via jugular venipuncture twice weekly and serum samples were radioimmunoassayed for progesterone (P4) content as an indicator of estrous activity. All rams were fitted with crayon equipped marking harnesses for use as an indicator of mating behavior in ewes. The first study tested the effects of introduction of vasectomized rams into a flock: of' 50 Dorset (D) and 50 Hampshire (H) purebred ewes in either May or June. More D ewes ovulated (96% vs 72% for H ewes) and mated (80% vs 20% for H ewes) in May. Of ewes which mated in May 65% D but no H ewes continued to cycle in June after removal of rams. Of ewes exposed to rams in June no difference among breeds was observed in percentage of ewes ovulating but more D ewes (72%) mated than H ewes (44%). Twenty-four percent of D but no H ewes cycled continuously throughout the 68 d or the study. Lambing date significantly affected mating behavior in H but not D ewes. The second study tested the effects of ram breed on incidence of mating and subsequent lambing in Rambouillet x Q Suffolk ewes In June and July of 1984 and 1985. Ewes were Q pre-exposed to either confined Suffolk (S) or Dorset (D) Q yearling rams or no (N) ram for 2 wk prior to breeding by Q either S or D rams. Lambing date was significantly affected by breeding treatment in 1984 and by pre-exposure treatment in 1985. Analys av underhållsprocessen för truckdäck i Aitikgruvan : Nulägesbeskrivning och RAMS-analys

Boliden Mineral AB är ett metallföretag som producerar metaller för det moderna samhället. Företaget har två dagbrottsgruvor belägna i Aitik strax utanför Gällivare i norra Sverige, varav en är landets största dagbrott. Stora gruvtruckar av två olika modeller transporterar malm från gruvan till anrikningsverket där koppar och andra metaller utvinns. Truckarna utgör en viktig del av logistiken som influerar den totala produktiviteten i verksamheten. Skadade truckdäck har visat sig vara en av de största orsakerna till tillgänglighetsförluster för truckarna. Studiens syfte var att skapa en nulägesbeskrivning gällande underhållskedjan av skadade truckdäck i företaget samt ge förslag på förändringar av däckunderhållet för att förbättra tillgängligheten på truckarna genom att minska nedtiden för däckskador. Intervjuer med inblandade personalgrupper i underhållskedjan samt analyser av underhållsdata ligger till grund för resultatet i studien. En RAMS-analys utfördes för att finna felorsaker i underhållskedjan som bidrar till lägre driftsäkerhet av gruvtruckarna med avseende på däckskador. RAMS-analysen påvisade att de flesta flaskhalsarna i underhållskedjan finns inom området underhållssäkerhet, vilket innebär att underhållsorganisationen reagerar för långsamt för att underhållet ska kunna bedrivas optimalt. Däckunderhållet tar generellt för lång tid i förhållande till de ideala feltider som vore möjliga att uppnå för båda truckmodellerna. Tillräcklig utbildning för att bedömma skadade truckdäck saknas stora delar av dygnet, resursbrist kan förekomma i truckverkstaden samt otydliga direktiv råder mellan arbetsgrupper hur kommunikationen skall förmedlas vilket förlänger nedtiderna. I syfte att optimera underhållssäkerheten bör tydliga rutiner mellan arbetsgrupperna skapas, vidare bör utbildningen utökas till berörda arbetsgrupper vilket innebär att bedömning av däckskador kan ske oavsett tid på dygnet. Arbetsfördelningen för de inblandade arbetsgrupperna bör omfördelas för att skapa en mer effektiv underhållskedja. Utifrån funktionssäkerheten visade studien att den nyare truckmodellen har större problem med däckskador än den äldre truckmodellen. Av de truckdäck där drifttiden analyserats visade sig ett av däckmärkena ha för korta drifttider jämfört med Bolidens internt uppsatta gränsvärden. Underhållsmässigheten visade att den nyare truckmodellen är svårare att underhålla eftersom komponenterna är större samt att utrustning saknas för att utföra fler än ett däckbyte samtidigt. I övrigt har Boliden två olika avtal gällande däck och studien presenterar förändringar i dessa som möjliggör att företaget ska kunna höja tillgängligheten. För att kunna göra bättre kartläggningar och analyser om underhållskedjan utifrån underhållsdata kring däckskador på truckarna bör företaget skapa bättre rutiner för inrapportering och dokumentation. / Boliden Mineral AB is a mining company producing metals for the modern society. The company have two open-pit mines in Aitik, situated in Gällivare in the northern part of Sweden. One of these open-pit mines is the largest in the country. Two models of large mining trucks transports ore from the mine to the concentrator where copper and other metals are recovered. The trucks play a key role in the logistics of ore, therefore their availability affect the total production capabilities of the mine. Damaged truck tires is one of the main reasons of downtime of the trucks. The aim of this study was to present a status report of the maintenance chain regarding damaged truck tires in the company and give suggestions about changes in tire maintenance to obtain better availability for the trucks regarding damaged truck tires. The study is based on interviews with workers from different working places who are involved in the maintenance chain as well as studies of maintenance data regarding truck tires. A RAMS (Reliability, Availability, Maintainability and Supportability) analysis was performed in order to discover causes of breakdown and bottlenecks in supportability, maintainability and reliability in the maintenance chain, all of which affect the availability of the plant. Findings in the RAMS analysis demonstrated that most bottlenecks were found in the supportability of the maintenance organization underlining unsatisfactory reaction times during tire breakdowns. The study further exposed the fact that downtime on behalf of damaged truck tires is too long in comparison to the ideal times achievable for the organization. Main causes to this problem is the fact that educated personnel with sufficient knowledge to inspect tire damage is missing most of the day, occasional resource shortages in the truck workshop as well as uncertainty in the communication between different working places which increases the downtime. To optimize the supportability of the maintenance chain the company have to introduce clear routines between the different departments and make sure that necessary education is provided to all sectors so that inspections are possible at any time. The study shows that the reliability for the newer truckmodel have greater problems with damaged truck tires compared to the older model. The lifespan for one of the analyzed truck tires in the study was shorter than the limit put up by the company. The analysis regarding the maintainability displayed the fact that maintenance is more difficult to perform because of larger components. The workshop have merely one device to perform multiple tire changes at the same time which also increases the downtime. This study underlines the fact that changes in the two existing agreements for truck tires may increase availability of the trucks for the company.

Using a more reliable sampling system Aitik can have a greater control over the enrichment process and thus have a major prerequisite for increasing the profitability of the company. An increased production, according to the new production target, are putting an increased pressure on the analysis precision and process control to maintain the high quality of the copper concentrate. By reconstruction of the waste weir for separation of Mp and Mp Rep + S Rep is a further need to invest in a more reliable operation and a more well-devised maintenance of the sampling system. By taking the entire life-cycle thinking in procurement Aitik can save large resources and also increase the overall reliability of the equipment in long term. The project aims to increase the reliability of the analyzes in the concentration plant and the goal of the study is to provide a guide for how Aitik, in different ways, can obtain more representative samples and higher availability for sampling. With the help from experienced experts in the areas of operation, maintenance and analysis in Aitik, Boliden and Garpenberg improvement suggestions have been collected, and together with perspectives from a literature study, several potential solutions has been found for the sampling system. Redesign will make sampling more functional and easier to work with and an optimal sampling will require resources to optimize other parts of the system: from the test socket until the sampling equipment. To maintain the system functions for the entire sampling system preventive maintenance activities, primarily in the form of cleaning and replacement of wearing parts, is required. And by identifying the item number and 5S routines to the application Quicktag better conditions for their staff regard the maintenance of the equipment can be created. Additional responsibilities of the company to enable longer system life is to develop a detailed specification for the supplier containing drawings, exploded diagrams, maintenance manuals and maintenance instructions, warranty support and inventory management regarding the spare parts warranty, standard parts and recommendations for inventory planning. In addition, the company should look into the possibility of coordinating parts management within the koncern and enhance sharing information and experiences within the koncern and for staff in Aitik to jointly optimize both operations at large and to improve sampling systems in all concentration plants. To apply NBW (New Boliden Way) for work on the sampling equipment would also be a step towards Boliden's goal for NBW implementation of activities for continuous improvement of sustainable development, improved quality and greater competitiveness in the market. / Med hjälp av ett mer tillförlitligt provtagningssystem kan Aitik få en större kontroll över anrikningsprocessen och därmed ha en större förutsättning för att öka lönsamheten i företaget. En ökad produktion, enligt det nya produktionsmålet, sätter ökad press på analysprecision och processtyrning för att bibehålla hög kvalitet för kopparkoncentratet. I och med planen av ombyggnation av avfallsdammen för separation av Mp och Mp Rep + S finns ytterligare en anledning att satsa på en säkrare drift och ett mer välplanerat underhåll för provtagningssystemet. Genom att företaget tar hänsyn till hela livscykelperspektivet vid upphandling kan Aitik spara stora resurser och dessutom långsiktigt öka den totala driftsäkerheten för utrustningen. Syftet med projektet är att öka tillförlitligheten hos analyserna i anrikningsverket och målet med studien är att ge en vägledning för Aitik hur de, på olika sätt, kan få mer representativa prover och högre tillgänglighet för provtagningsutrustningen. Med hjälp från erfarna experter inom områdena drift, underhåll och analysering från Aitik, Boliden och Garpenberg har flertalet förbättringsförslag inhämtats och i samverkan med kunskap ur en litteraturstudie har flertalet potentiella lösningar hittats för provtagningssystemet. Omkonstruktion kommer att göra provtagningsutrustningen mer funktionell och lättare att arbeta med och för en optimal provtagning kommer det krävas resurser för optimering av övriga delar av systemet: från provuttag fram till provtagningsutrustningen. För att upprätthålla systemfunktionerna för hela provtagningssystemet krävs förebyggande underhållsåtgärder framför allt i form av rengöring och byte av förslitningsdelar, och genom att identifiera objektnummer och 5S-rutiner till applikationen Quicktag kan företaget skapa bättre förutsättningar för sin personal för underhållet av utrustningen. Ytterligare ansvar från företagets sida för att möjliggöra en längre systemlivslängd är att utforma en detaljerad kravspecifikation åt leverantören innehållande ritningar, sprängskisser, underhållsmanualer och underhållsrekommendationer, supportgaranti samt lagerhantering avseende reservdelsgarantier, standardiserade delar och rekommendationer för lagerplanering. Dessutom bör företaget se över möjligheten för att samordna reservdelshanteringen inom koncernen samt öka informations- och erfarenhetsutbytet inom både koncernen och inom Aitikpersonalen för att tillsammans optimera både verksamheterna i stort samt för att förbättra provtagningssystemen i alla anrikningsverk. Att kunna tillämpa NBW (New Boliden Way) för arbetet med provtagningsutrustningen skulle dessutom vara ett steg mot Bolidens mål för NBW-implementering i verksamheten för ständiga förbättringar avseende hållbar utveckling, ökad kvalitet och större konkurrenskraft på marknaden.
