Ce travail avait pour objectif, de caractériser le poids des infections au cours de la grossesse et d’étudier la construction de l’immunité anti-PfEMP1 chez le jeune enfant. La première partie, aborde la conséquence des infections et l’efficacité des traitements chez la mère. Cette étude réalisée sur la cohorte de femmes d’une étude prospective au Bénin, a démontré l’impact des infections à bas bruit sur le taux d’hémoglobine maternel et le faible poids de l’enfant. Le TPI-SP, a montré les limites quant à sa capacité à débarrasser les femmes enceintes infectées au moment du traitement de leur parasite. Nos résultats soutiennent davantage la nécessité de trouver des moyens alternatifs de prévention qui offrent une meilleure couverture de la grossesse. La deuxième partie aborde la construction de l’immunité anti-PfEMP1 dans la première année de vie chez l’enfant. Un résultat majeur de cette étude est la démonstration que l’acquisition des anticorps contre les PfEMP1 associés aux complications du paludisme est dépendante des infections patentes de l’enfant. La troisième partie aborde les phénotypes des parasites responsables de diverses formes cliniques du paludisme chez l’enfant Africain âgé de 0 à 5 ans. Cette étude a permis de mettre en évidence un marqueur de mauvais pronostic du paludisme cérébral. Sur un deuxième volet, nous avions montré que les isolats adhérant faiblement à ICAM-1, transcrivent fortement les gènes codant pour les PfEMP1 contenant des motifs DC8. Ces résultats soulèvent la question du rôle de EPCR dans la physiopathologie du neuropaludisme. Le travail développé dans cette thèse a permis de décrire pour la première fois la construction de l’immunité anti-PfEMP1 dans la première année de vie, de mettre à jour les connaissances sur la physiopathologie du paludisme cérébral chez le jeune enfant et de dégager des pistes à explorer prioritairement dans la perspective du développement d'un vaccin contre les formes graves du paludisme. / This work aimed to characterize the burden of P. falciparum infections during pregnancy and to study the construction of the anti-PfEMP1 immunity in the early life. The first part discusses the consequence of infection and the effectiveness of treatments in the mother. This study on the cohort of women from a prospective study in Benin, demonstrated the impact of infections with low parasitemia on the maternal hemoglobin and low weight of the child. IPT-SP, has shown the limits of its ability to rid infected pregnant women in the processing of their parasite. Our results further support the need to find alternative means of prevention that provide better coverage of pregnancy. The second part treated the construction of the anti-PfEMP1 immunity in the first year of life in children. A major finding of this study is the demonstration that the acquisition of antibodies against the PfEMP1 associated with complications of malaria depends on patentes infections in children. The third part studies parasites phenotypes responsible of various clinical forms of malaria in African children aged 0-5 years. This study allowed finding a marker of poor prognosis of cerebral malaria. On a second component, we showed that isolates bind to ICAM-1, highly transcribe the genes encoding PfEMP1 containing DC8. These results raise the question of the role of EPCR in the pathophysiology of cerebral malaria. The work developed in this thesis has allowed describing for the first time the construction of the anti-PfEMP1 immunity in the first year of life, to update knowledge on the pathophysiology of cerebral malaria in young children and identify Options to be primarily from the perspective of developing a vaccine against severe forms of malaria.

Tracking Tobacco Mosaic Virus Infection from Infected Seeds to Seedlings Confirms Seed Transmission in Tobacco (Nicotiana tabacum L.)Hoak, Jessica 10 July 2019 (has links)
The Tobacco mosaic virus (TMV) is a positive sense single stranded RNA virus and is found across the world. TMV can impact the overall yield and quality of the crop resulting in an economic loss. Plants that are infected with TMV show a variety of symptoms such as mosaic pattern, mottling, necrotic lesions and stunted growth. Historically, TMV has caused controversy on whether this economically significant virus is seedborne or seed transmitted. The objective of this study is to track TMV infection from infected seeds to seedlings to determine the percentage of seed transmission. This experiment used three pods from three different TMV infected cultivar K 326 flue-cured tobacco plants. Seeds from each pod were germinated in a growth chamber for approximately ten days. Samples were separated into seed coat, root and leaves after germination. Total RNA was extracted from each part and synthesized into cDNA for analysis. A quantitative real-time PCR (RT-qPCR) assay was used to determine TMV concentration of each sample. Endpoint RT-PCR was used to determine a conservative threshold value from the RT-qPCR results. These results demonstrated that TMV influenced percent germination with a range from 94% to 50%. Seed coats had a significantly higher virus titer concentration (P < 0.05) when compared to the roots and leaves. Statistical analysis revealed highly significant (P < 0.0001) differences among pods for virus titer and there is a highly significant plant by pod interaction (P < 0.0001). Endpoint RT-PCR confirmed TMV infection in leaves, roots and seed coats. Percent infection in leaves ranged from 2% to 24% and percent infection for roots ranged from 8% to 40%. Results demonstrate that TMV is seed transmitted in flue-cured tobacco. / Master of Science / The Tobacco mosaic virus (TMV) is an RNA virus that occurs globally in areas where tobacco is grown. TMV is a tobamovirus and infects over 350 plant species. TMV can reduce the yield and quality of the crop which will result in an economic loss for the grower. Plants that are infected with TMV show a variety of symptoms such as mosaic pattern, necrotic lesions, and stunted growth, and there are no effective ways to eradicate the virus. There has been controversy on whether to categorize TMV as a seedborne virus or a seed-transmitted virus because the location of the virus within a seed is unknown. This study examined seeds from three pods grown on three different TMV-infected flue-cured tobacco plants of cultivar K 326 to track TMV infection from infected seeds to seedlings. Seeds from each pod were germinated in a growth chamber for ten days and samples were separated into leaves, root and seed coat. Each sample had total RNA extracted and synthesized into cDNA for analysis. A quantitative real-time PCR (RT-qPCR) assay was used to determine TMV concentration of each sample since this technology can detect small amounts of virus. Em cana-de-açúcar, a colonização do caule por fungos oportunistas, como Fusarium verticillioides e Colletotrichum falcatum, está diretamente ligada ao ataque da lagarta Diatraea saccharalis (Lepidoptera: Crambidae). Duas proteínas, SUGARWIN1 e SUGARWIN2 são produzidas em cana-de-açúcar, em resposta ao dano mecânico e ao ataque de D. saccharalis, porém estas proteínas não afetam o inseto, e sim ocasionam alterações fisiológicas e morfológicas em F. verticillioides e C. falcatum, ocasionando a morte destes fungos por apoptose. Dietas artificiais suplementadas com estes fungos oportunistas ocasionaram o ganho de peso da D. saccharalis. Esses dados indicam uma interação mais íntima entre o inseto e estes patógenos de cana, sendo que, neste estudo procuramos identificar relações simbióticas entre os indivíduos, analisando se a forma de transmissão desses fungos é mediado pela D. saccharalis. Os resultados mostraram a presença do F. verticillioides em todas as fases de desenvolvimento da D. saccharalis após contato com o fungo, ou seja, depois de se alimentarem em dieta suplementada por F. verticillioides no 4º instar, permaneceram infectadas pelo fungo ao longo de toda a fase pupal e adulta, em ambos os sexos. Além disso, o F. verticillioides foi transmitido para os descendentes de D. saccharalis, sendo que o fungo foi detectado nos ovos, ou seja, um caso original de transmissão vertical. Por meio de microscopia, também foi possível verificar a alta intensidade de F. verticillioides no interior do intestino de lagartas. Estes dados inferem em uma relação simbiótica entre F. verticillioides e D. saccharalis, onde o simbionte é transferido verticalmente para as gerações subsequentes. As respostas obtidas com o fungo C. falcatum diferiram daquelas obtidas com F. verticillioides, uma vez que não se detectou a presença do fungo a partir da fase pupal. Neste caso, a relação de simbiose entre o fungo e o inseto pode resultar em uma transmissão horizontal. Com este estudo foi possível identificar diferentes formas de transmissão por D. saccharalis para dois fungos envolvidos em podridão de colmo em cana-de-açúcar. Estes dados mudam a forma como é vista a transmissão de F. verticillioides por D. saccharalis em cana-de-açúcar, podendo influenciar a forma de manejo da podridão de Fusarium e da broca nos canaviais. / In sugarcane, stem colonization by opportunistic fungi, such as Fusarium verticillioides and Colletotrichum falcatum, is directly linked to the attack of Diatraea saccharalis (Lepidoptera: Crambidae) caterpillar. Two proteins, SUGARWIN1 and SUGARWIN2 are produced in sugarcane, in response to mechanical damage and attack of D. saccharalis, however these proteins do not affect the insect, but cause physiological and morphological changes in F. verticillioides and C. falcatum, causing the death of these fungi by apoptosis. Artificial diets supplemented with these opportunistic fungi caused the weight gain of D. saccharalis. These data indicate a more intimate interaction between the insect and the sugarcane pathogens. In this study, we sought to identify symbiotic relationship among individuals, analyzing whether the transmission of these fungi is mediated by D. saccharalis. The results showed the presence of F. verticillioides in all stages of D. saccharalis development after contact with the fungus, in the 4th instar. The caterpillars remained infect by the fungus throughout the pupal and adult phase, in both sexes. In addition, F. verticillioides was transmitted to D. saccharalis offspring, being detected in eggs, an original case of vertical transmission. Through the microscopy results, it was also possible to verify the high intensity of F. verticillioides inside the intestines of caterpillar. These data infer in a symbiotic relationship between F. verticillioides and D. saccharalis, where the symbiont is transferred vertically to the offspring. The responses obtained with C. falcatum differed from those obtained with F. verticillioides, since the presence of the fungus was not detected from the pupal phase. In this case, the symbiont relationship between fungus and insect can result in a horizontal transmission. With this study was possible to identify different forms of fungi transmission by D. saccharalis. These data change the way the transmission of F. verticillioides by D. saccharalis in sugarcane is viewed, and may influence the management of Fusarium rot and sugarcane borer attack in sugarcane.

Previous issue date: 2017-03-15 / Introduction: Chikungunya fever (FCHIK) is a disease of abrupt onset, transmitted by
arthropod mosquitoes intermediate hosts of the Chikungunya virus (CHIKV). The illness has
a significant impact on the quality of life of the affected person. Since a disease causes intense
and prolonged symptoms of polyarthralgia and myalgia, it requires health care, during a
recovery, more than other arboviruses. The objective of this study was to study clinicians and
clinicians suggestive of FCHIK, residing in the States of Amapá and Goiás, aiming to
correlate the results of laboratory tests with the presented symptomatology. Materials and
methods: The study was carried out at the Center for Immunological Studies and Research of
the Pontifical Catholic University of Goiás, Goiânia, and in Emergency Care Units in the
cities of Macapá, Oiapoque and Santana-AP. The study population consisted of 80 individuals
with suspected FCHIK and for investigators of inflammatory markers, the control group
consisted of 20 blood samples from healthy donors from Goiana Central de Serologia e
Imunohematologia. Viral RNA extraction was performed, followed by RNA detection by
Real-Time Polymerase Chain Reaction. In addition to ELISA for detection of IgM and IgG
against Chikungunya virus. Participants symptoms were correlated with serology and Creactive
protein (CRP), which was evaluated in healthy subjects and in people with FCHIK.
Results: No data presented for detection of viral RNA by RT-qPCR for CHIKV, but three
samples were positive in this technique for zika virus and one for dengue subtype 1 (DENV1).
In an enzyme-linked immunosorbent assay, 26 samples were positive for IgG and 3 for IgM.
Regarding the stage of the disease, 10 were in the acute phase, 04 in the subacute phase and
12 in the chronic phase. Correlated the results of the serology with a symptomatology it was
observed that the acute phase, all have fever, 90% headache, 70% arthralgia and 60% edema.

comparing participants with negative serology, n = 54, the most prevalent symptoms were
rash, headache, fever, and arthralgia. The CRP levels in individuals infected with more than
four symptoms were higher when compared with healthy individuals. Conclusion: The study
focused on people with a clinical picture characteristic of FCHIK. The most common
symptom in the three phases presented for arthralgia, followed by edema and myalgia, a fever
was frequent only in the acute phase. All participants were negative in the evaluation of viral
RNA by RT-qPCR for CHIKV, for the virus has a short duration in the body, and this
methodology is limited to the time of symptom onset and sample collection, DENV and
ZIKV. IG G. Those with negative serology for CHIKV, despite taking into account the joints,
symptoms common to other arboviruses. CRP levels have been shown to be high relative to
healthy subjects. / Introdução: A Febre Chikungunya (FCHIK) é uma doença de início abrupto, transmitida por
mosquitos artrópodes hospedeiros intermediários do vírus Chikungunya (CHIKV). A
enfermidade representa um significativo impacto na qualidade de vida da pessoa afetada. Uma
vez que a doença causa sintomas intensos e prolongados de poliartralgia e mialgia,
requerendo atenção de saúde, durante a recuperação, mais do que outras arboviroses.
Objetivou-se estudar aspectos clínicos e sorológicos de indivíduos apresentando quadro
clínico sugestivo de FCHIK, residentes nos Estados de Amapá e Goiás, visando correlacionar
os resultados de testes laboratoriais com a sintomatologia apresentada. Materiais e métodos:
O estudo foi realizado no Núcleo de Estudos e Pesquisa Imunológica da Pontifícia
Universidade Católica de Goiás, em Goiânia, e em Unidades de Pronto Atendimento de Saúde
das cidades de Macapá, Oiapoque e Santana-AP. A população de estudo foi constituída de 80
indivíduos com suspeita de FCHIK e para comparar os marcadores inflamatórios, o grupo
controle foi constituído de 20 amostras de sangue de doadores saudáveis da Central Goiana de
Sorologia e Imunohematologia. Foi realizada a extração do RNA viral, seguido de detecção
do RNA por meio de Reação em Cadeia de Polimerase em Tempo Real. Além de ELISA para
detecção de IgM e IgG específicos para o CHIKV. Os sintomas dos participantes foram
correlacionados com o resultado da sorologia e da proteína C reativa (PCR), que foi avaliada
em indivíduos saudáveis e em pessoas com FCHIK. Resultados: Nenhuma amostra
apresentou limiar de detecção do RNA viral por RT-qPCR para CHIKV, porém três amostras
foram positivas nessa técnica para vírus zika (ZIKV) e uma para dengue subtipo 1 (DENV1).
Em ensaio imunoenzimático, 26 amostras foram positivas para IgG e 3 dessas para IgM. Em
relação ao estágio da doença, 10 encontravam-se em fase aguda, 04 em fase subaguda e 12 em
fase crônica. Correlacionados os resultados da sorologia com a sintomatologia observou-se
que os de fase aguda, todos tiveram febre, 90% cefaleia, 70% artralgia e 60% edema.
Enquanto que, os de fase subaguda tiveram: artralgia e cefaleia (100%), mialgia e edema
(75%). Os de fase crônica tiveram edema (100%), artralgia (92%) e mialgia (75%). Quando
comparados os participantes com sorologia negativa, n=54, os sintomas mais apresentados
foram exantema, cefaleia, febre e artralgia. Os níveis de PCR nos indivíduos infectados e que
apresentavam mais de quatro sintomas foram maiores quando comparados com indivíduos
saudáveis. Conclusão: O estudo focou em pessoas com quadro clínico característico para
FCHIK. O sintoma mais comum nas três fases apresentadas foi a artralgia, seguido de edema
e mialgia, a febre foi frequente somente na fase aguda. Todos os participantes foram
negativos na avaliação do RNA viral por RT-qPCR para CHIKV, pois o vírus tem uma curta
duração no organismo, e esta metodologia é limitada ao tempo de início dos sintomas e coleta
de amostra, ainda assim foi encontrado RNA viral do DENV e ZIKV. Alguns participantes
foram positivos para sorologia IgG. Aqueles com sorologia negativa para CHIKV, apesar de
terem dor nas articulações, tinham sintomas comuns a outras arboviroses. Os níveis de PCR
demonstraram-se elevados em relação aos indivíduos saudáveis.

Expressão diferencial de genes induzidos por antracnose em feijoeiro em resposta à indução da resistência por silício / Differential expression of genes activated by anthracnose in response to silicon induced resistanceBeraldo, Ana Luiza Ahern 08 August 2012 (has links)
O feijão é importante fonte carboidratos, vitaminas, minerais e fibras. No Brasil, a produtividade desta leguminosa é baixa e um dos fatores é a ocorrência de doenças como a antracnose causada pelo Colletothrichum lindemuthianum, que gera perdas de até 100% da produção. Plantas possuem diversos mecanismos de defesa contra patógenos e relatos apontam que o silício é capaz não só de promover mudanças morfológicas nas folhas, mas também de ativar os genes de resistência. O presente trabalho foi dividido em três estudos que tinham como objetivo: (1) entender a resposta de três cultivares de feijoeiro ao silício disponível na solução nutritiva; (2) identificar a contribuição do Si na expressão de genes relacionados à infecção pelo fungo através da construção de duas bibliotecas subtrativas por supressão (SSH), visando selecionar genes diferencialmente representados durante a infecção da planta com a raça 65 de C. lindemuthianum (a) e durante a infeção da planta na presença de uma maior dose silicato de potássio (75 ppm) no substrato (b); (3) identificar a resposta de dez transcritos selecionados no Estudo 2 para tentar entender a resposta dos mesmos em diferentes períodos (0; 6; 42; 72 h) após a inoculação, com ou sem suplemento de Si. Como resultados, foi observado que para as três cultivares avaliadas o Si começa a ser absorvido 14 dias após o transplante. Também foi identificado por de microscopia de varredura (MEV) que não há diferença significativa entre o número de tricomas e cada cultivar, mas que para o número de estômatos a cultivar IAC-Harmonia destacou-se das demais. Além disso, quando as três cultivares foram suplementadas com Si, houve a formação de uma cera epicuticular descrita como mecanismo de defesa da planta contra fungos; e que através de EDX (Energy-dispersive X-ray spectroscopy) foi possível constatar que plantas tratadas com Si apresentam maior teor deste elemento nas folhas. Através de inoculações com a raça 65 do patógeno verificou-se o efeito do mineral na redução da severidade da doença nas cultivares IAC-Harmonia e Pérola. No segundo estudo, duas bibliotecas de hibridização subtrativa por supressão (SSH), foram construídas visando selecionar os genes diferencialmente expressos entre plantas inoculadas e não-inoculadas (A) e entre plantas inoculadas e tratadas ou não com 75 ppm de Si (B). Foram geradas 991 sequências únicas, anotadas através do GeneOntolgy. Quinze genes de cada biblioteca foram selecionados para os experimentos de validação por RT-qPCR. Para a Biblioteca A, 11/15 genes foram positivamente regulados, e em B, 14/15. No terceiro estudo ficou evidenciado que a inoculação com o patógeno alterou positivamente a expressão de sete genes, enquanto que o tratamento com 75 ppm de Si alterou a expressão de oito genes, em pelo menos um dos tempos avaliados / Beans are an important source of carbohydrates, vitamins, minerals and fibers. In Brazil, this legume still has low productivity and one of the factors involved is the occurrence of diseases such as anthracnose, caused by the fungus Colletothrichum lindemuthianum, which causes losses in production of up to 100%. Plants present several defense mechanisms against pathogens and the reports indicate that silicon does not only promote morphological changes in leaves, but also activates resistance genes. This work was divided into three studies aiming: (1) to understand the response of three bean cultivars to a silicon source in a nutrient solution, (2) to identify the contribution of Si in the expression of genes related to the infection by the fungus by constructing two subtractive suppression libraries (SSH), to select genes differentially represented during infection of the plant with race 65 of C. lindemuthianum (a) and during infection of the plant in the presence of higher dose of potassium silicate (75 ppm) in the substrate (b), (3) to identify the response of ten selected transcripts in Study 2 in various periods (0, 6, 42, 72 h) after inoculation, with or without supplemental Si. As a result, it was observed that for all three cultivars Si begins to be absorbed 14 days after transplantation. Was also identified by microscopy (SEM) that there is no significant difference between the number of trichomes among cultivars, but that the number of stomata for the IAC-Harmonia stood out from the rest. Moreover, when the three cultivars were supplemented with Si, thus forming an epicuticular wax described as a defense mechanism against plant fungi, and that by EDX (Energy-dispersive X-ray spectroscopy) it was found that plants treated with Si have higher content of this element in leaves. Through inoculations with race 65 of the pathogen it was verified the effect of the mineral in reducing disease severity in IAC-Pérola and IAC - Harmonia. In the second study, two libraries from suppression subtractive hybridization (SSH) were constructed in order to select the differentially expressed genes between inoculated and non-inoculated (A) and between plants inoculated and treated or not with 75 ppm of Si (B). In total, 991 unique sequences were generated, those recorded by GeneOntolgy. Fifteen genes from each library were selected for the validation experiments by RT-qPCR. For library A, 11/15 genes were positively regulated, and in B, 14/15. In the third study it is showed that inoculation with the pathogen positively altered expression of seven genes, whereas treatment with 75 ppm of Si changed the expression of eight genes, in at least one of the times analyzed

Le bassin d’Arcachon est une lagune semi-fermée qui concentre de forts enjeux économiques grâce à la pratique de l’ostréiculture. Cette activité est affectée depuis une trentaine d’années par des phénomènes de mortalités estivales, et plus récemment par des surmortalités du naissain. Ces travaux se sont intéressés au rôle des polluants majeurs du bassin d’Arcachon dans ce contexte de crise en étudiant leurs effets sur la biologie de Crassostrea gigas. Une approche in situ a d’abord été adoptée afin d’identifier les contaminants les plus présents dans le milieu naturel. Des opérations de transplantation d’huîtres et des prélèvements de sédiments ont permis de quantifier divers contaminants et d’associer leur présence à des réponses biologiques. Les polluants ainsi identifiés ont ensuite été employés en conditions contrôlées au laboratoire. Trois expérimentations ont été réalisées : la première concerne l’étude des voies de contamination par le tributylétain ; la seconde concerne les effets des pesticides et du cuivre ; la troisième concerne l’effet des HAP sur des huîtres diploïdes et triploïdes. Nos résultats indiquent que les organismes transplantés au coeur de la lagune sont plus exposés aux polluants, en lien avec les caractéristiques hydrodynamiques du système. La plupart des paramètres biologiques étudiés sur le terrain ont par ailleurs montré une saisonnalité liée aux processus de gamétogenèse. Au laboratoire, la plupart des contaminants testés ont induit une réponse adaptative chez les huîtres exposées. Notre étude souligne l’importance de coupler approche de terrain et approche expérimentale pour comprendre le fonctionnement des écosystèmes côtiers. / The Arcachon Bay is a semi-enclosed lagoon and represents the core of strong economic stakes through the practice of oyster-farming. This activity has been affected for around thirty years by summer mortality events, and more recently by abnormally high death rates of juveniles. This work focused on the role of the Arcachon Bay’s main contaminants in this crisis, by studying their effects on the cupped oyster’s biology. Firstly, an in situ approach was adopted in order to identify the major pollutants of the bay: caged oysters were transplanted and sediments were sampled. The presence of contaminants in the samples was associated to biological responses. Secondly, the contaminants identified in situ were used in controlled conditions at the laboratory. Three experimentations were performed; the first dealt with the contamination pathways of tributyltin; the second focused on the biological effects of pesticides and copper; the third concerned the effects of PAH towards diploid and triploid oysters. Our results indicate that the inner stations present higher accumulation of metals and PAH, in accordance with the hydrodynamic features of the bay. The bioindicators used in situ exhibited seasonal trends related to the oysters’ gametogenesis. In the laboratory, most of the contaminants used at environmental levels induced an adaptive response of the exposed oysters. Our study highlights the importance of coupling in situ and laboratory approaches in order to understand the functioning of coastal ecosystems.
