Modulatory effect of lipid compositions on phospholipase A2 activityChiou, Yi-ling 17 July 2012 (has links)
The goal of the present study is to elucidate the modulatory effect of lipid compositions on phospholipase A2 (PLA2) activity. Sphingomyelin (SM) incorporation inhibited catalytic activity and membrane-damaging activity of native and mutated PLA2 toward egg yolk phosphatidylcholine (EYPC) vesicles. The inhibitory effects were through the reduction of membrane fluidity and modulation of the mode of membrane binding of PLA2 at water/lipid interface. The modulated effect of SM depended on inherent structural elements of PLA2. Moreover, cholesterol (Chol) incorporation into EYPC/egg yolk sphingomyelin (EYSM) vesicles relieved the inhibitory effect of sphingomyelin on PLA2 activity via lipid domain formation by SM and Chol. The effects on the interactive mode of PLA2 with phospholipids induced by the physical state changes of membrane bilayers abolished the inhibition of SM on catalytic activity and membrane-damaging activity of PLA2. Additionally, quercetin incorporation increased PLA2 activity and membrane-damaging activity toward EYPC/SM vesicles via its raft-making effect. Quercetin incorporation reduced PLA2 activity and membrane-damaging activity toward EYPC/SM/Chol vesicles via its raft-breaking effect. Membrane-inserted quercetin affected on membrane structure and membrane-bound mode of PLA2 to modulate PLA2 interfacial activity and membrane-damaging activity. Finally, studies on the effects of phosphatidylserine (PS) content on the sensitivity of lipid vesicles mimicking inner and outer plasma membrane toward PLA2 activity revealed that the membrane-binding mode adopted by PLA2 depended on the lipid composition. The effects of PS content on the extent of lipid domain formation and the conformation of PLA2 adopted at water-lipid interface modulate PLA2 catalytic activity. Collectively, these results indicate that lipid composition modulates PLA2 activity via its effects on membrane structure and membrane-bound mode of PLA2

In this study, the settlement behavior of the piled raft foundations resting on overconsolidated clays under uniform loading, is investigated for different pile configurations and load levels. A total of 100 plane &ndash / strain and three &ndash / dimensional finite element analyses are carried out and the results of these analyses are compared both with each other and with the results presented by Reul & / Randolph (2004). The material parameters used in the analysis are selected mainly referring to the previous studies cited above on the same subject and slight modifications are made for convenience in the analysis. The analysis method and the applied pile configurations and load levels are directly taken from the reference study, excluding the soil model employed. A drained Mohr &ndash / Coulomb failure criteria is employed in the analysis of this study in modeling the soil instead of an elastoplastic model which was used in the analysis of the reference study. The results are evaluated for the average and differential settlements of the foundations and it is seen that / although the average and differential settlements calculated in this study are not always very close to the values calculated in the reference study, the calculated settlement reduction factors due to piles (especially for the average settlements) compared well with the findings of the reference study for all pile configurations and load levels considered. Based on this, a new approach is suggested to estimate the average settlements of the piled raft foundations. Moreover, correction factors are recommended in order to estimate the average settlements of the piled rafts by directly using the programs employed throughout the thesis.

Living Radical Polymerization Of Hydroxyethyl Methacrylate And Its Block Copolymerization With Poly(dimethyl Siloxane) MacroazoinitiatorVargun, Elif 01 June 2009 (has links) (PDF)
Hydrophilic poly(2-hydroxyethyl methacrylate), PHEMA, and hydrophobic poly(dimethyl siloxane), PDMS, segments containing copolymers have been widely used as a biomaterial. These amphiphilic copolymers also used as an emulsifying agent in polymer solutions and compatibilizer in polymer blends. In this case, solution polymerizations of HEMA by radiation, ATRP and RAFT methods were studied. The thermal degradation mechanism of PHEMA, which was prepared in aqueous solution by gamma radiation technique, was studied in detail. The DSC, TGA and Mass Spectroscopy analyses revealed that the degradation is linkage and depolymerization with a combination of monomer fragmentation. The ATRP of HEMA was performed with ethyl-2-bromoisobutyrate (EBriB) initiator and CuCl/bipyridine catalyst in MEK/1-propanol solvent mixture. Cu(II) complexes and PHEMA obtained via ATRP were characterized by UV-vis, FTIR and 1H-NMR analysis. The RAFT polymerization of HEMA with different [RAFT]/[AIBN] ratios were also investigated in three solvents (methyl ethylketone, ethyl acetate and toluene). The controlled polymerization of HEMA with the ratio of [RAFT]/ [AIBN]=18 at 80 oC in MEK and ethyl acetate, shows the first-order kinetic up to the nearly 40 % conversion Macroazoinitiator PDMS-MAI was synthesized from bifunctional PDMS and then copolymerized with MMA, EMA, HEMA and TMS-HEMA monomers Different characterization methods such as FTIR, 1H-NMR, solid state NMR, GPC, XPS, SEM, DSC, etc. have been used for the characterization of block copolymers. P(DMS-b-TMSHEMA) was converted to the P(DMS-b-HEMA) block copolymer by deprotection of TMS groups. The phase separated morphology was observed for the P(DMS-b-HEMA) copolymer, which was different from P(DMS-b-MMA) and P(DMS-b-EMA) copolymers.

Two different concepts and design procedures namely settlement reducing piles and piled raft foundations have been studied independently in this thesis.
A laboratory study is conducted on model rafts with differing number of model settlement reducing piles. Pile length, pile diameter, type of soil and size of raft are kept constant and settlements are measured under sustained loading. Remolded
kaolin is consolidated under controlled stresses before tests are performed in model boxes. The tests are conducted under two sustained loadings of 75 kPa and 40 kPa. 0(raft), 16 and 49 number of piles are used. During the tests, all of the skin friction is mobilized. Several tests are conducted for each combination to see the variability. It is concluded that increasing the pile number beyond an optimum value is inefficient as far as the amount of settlement is considered. Also an analytical procedure has been followed to calculate settlements with increasing number of piles. In the second part of this thesis, finite element analyse have been performed on a piled raft foundation model, using Plaxis 3D Foundation Engineering software. This analyse are supported with analytical methods. The piled raft model is loaded with
450 kPa raft pressure. The studies are conducted in two sets in which different pile lengths are used / 25 m and 30 m respectively. The numbers of piles are increased from 63 to 143. All other parameters are kept constant. The results showed that again an optimum number of piles will be sufficient to reduce the settlement to the acceptable level. The analytical methods indicate a similar behavior. The comparison and results are presented in the study.

The oyster culture site is located at the nearshore of Tainan city where it not only has an abundance of phytoplankton for feeding oysters but also has suitable water temperature for growing oysters. Each year the harvest of oyster has brought a good fortune to local fishermen and their household, therefore raising oyster is a very important industry for the Tainan city. Up to the present, the land-based fishery has many difficult problems such as the increasing cost of feed, over pumping of groundwater and lack of efficient treatments for fish diseases. All these difficult situations have given the offshore oyster culture a great opportunity to develop since no feed cost is required and if clean offshore sites are selected for growing oysters. This paper focuses on the problems met in the growing process including oyster raft engineering, biotechnology, marine environmental monitoring and the hygiene problems etc. Finally, several suggestions of oyster value improvement have been provided for the fishermen.

Une investigation des propriétés d'émission de fluorescence à l'état condensé a été effectuée sur des dérivés encombrés du Boron DIPYrromethene (BODIPY). L'objectif est de déterminer la nature de l'encombrement stérique afin d'optimiser l'efficacité d'émission en minimisant les interactions intermoléculaires. L'étude en films de PMMA du 3,5-di-adamantane, 8-mésityle BODIPY a mis en évidence la formation prépondérante d'agrégats H et celle de dimères J, dont le rendement quantique de fluorescence n'est diminué que d'un facteur 5 par rapport à la molécule non agrégée. Une deuxième étude spectroscopique en films évaporés a également été effectuée sur une série de quatre dérivés substitués sur les positions 3 et 5 par des groupements [2.2]paracyclophane et sur les positions 2 et 6 par les groupements respectifs : propyle, éthyle, méthyle et H (BODIPY H-, Alkyl-PcP). L'absence de substitution sur les positions 2 et 6 favorise la formation des agrégats H tandis que la présence d'une chaîne alkyle conduit à la formation prépondérante d'agrégats J. La mesure des rendements quantiques relatifs ainsi que l'analyse des images de fluorescence résolues en temps démontrent une augmentation de l'efficacité d'émission et de la durée de vie de fluorescence de la phase solide lorsque la chaîne alkyle s'allonge. Les composés BODIPY Et-PcP et Pr-PcP présentent tous deux une émission de fluorescence supérieure à un dérivé non encombré. Enfin, une étude préliminaire des phases solides de dérivés BODIPY substitués par des groupements insaturés planaires (benzothiophène, p-bromo-diphényle) a été menée afin d'étudier l'influence de la géométrie des substituants. La synthèse de l'ensemble des résultats permet de conclure que les substituants insaturés planaires ne conduisent pas systématiquement à une perte du rendement quantique de fluorescence à l'état solide, en comparaison aux substituants saturés et insaturés sphériques. Deux méthodes d'élaboration de particules ont été explorées : la première consiste à greffer des dérivés d'aza-BODIPY sur des billes de polystyrène afin d'obtenir des particules sondes de pH - la deuxième approche repose sur la polymérisation d'un dérivé BODIPY selon la méthode RAFT suivie de la formulation du polymère amphiphile obtenu en micelles de taille nanométrique. Ces particules offrent la possibilité d'une post-fonctionnalisation permettant d'élaborer des capteurs.

Liquid chromatography is an important separation technique for a vast number of analytes. This thesis mainly focuses on the development of stationary phases for liquid chromatography based on zwitterionic sulfobetaine polymers. In the thesis, various ways to prepare zwitterionic polymers in an aqueous environment using reversible addition fragmentation chain transfer (RAFT) polymerization are described. Both telomers, i.e. short soluble polymer chains containing a functional terminal group, as well as graft polymers on various supports have been synthesized. The RAFT polymerization technique provides an increased degree of control of the final polymers, which may aid in the preparation of more specifically tailored separation materials. Sulfobetaine polymers carry both a positive and a negative charge within a single entity, which results in interesting solution properties as well as highly biocompatible features. These unique features make them especially suited for separation of highly polar and/or charged compounds. An example of the successful separation of short peptides using a stationary phase synthesized with the RAFT technique is given. The unusual properties of sulfobetaine-type polymers are believed to be associated with the structure of water close to the polymer. A study of water structure in some silica based stationary phase grafted with zwitterionic sulfobetaine polymers was conducted. The impact of water structure on retention characteristics was investigated.

Poly (co iso-butyl vinyl ether-alt-maleic anhydride), an alternating copolymer, was synthesised. For this class of copolymers the formation of an electron-donor complex is invoked to explain their microstructure in which the two comonomers strictly alternate. Due to its polarity, this copolymer constitutes a potential additive for imparting hydrophilic properties to a hydrophobic matrix. In order to obtain narrow molecular weight polymers and study the relation between the molecular weight of this additive and its ability to migrate to the host polymer surface, chain transfer agents were introduced in the system and also the Reversible Addition-Fragmentation chain Transfer (RAFT) process was employed. Free radical polymerisation was first carried out to allow for a comparison with the RAFT process and kinetics of copolymerisation was studied by NIR-FTIR and 1H NMR spectroscopy in order to analyse the rate of reaction of each comonomer. Dibenzyl trithiobenzoate, 3-benzyl sulfanyl thiocarbonyl sulfanyl-propionic acid and dibenzyl trithiobenzoate were used as RAFT agents. Results demonstrate that only benzyl dithiobenzoate is able to control the molecular weight of this copolymer and decrease its polydispersity index; possible reasons laying behind this result are discussed. It was also found that, in particular in the presence of benzyl dithiobenzoate, poly(iso-butyl vinyl ether) forms. This is an unusual phenomenon considering that the free radical polymerisation affords alternating copolymers and that iso-butyl vinyl ether is a monomer that polymerises through the cationic process. Experiments were carried out in various solvents in an attempt to counteract this side reaction, but no appreciable correlation between the properties of the solvents and the formation of homopolymer were found. Various hypothesis are considered, however it is likely that, in the conditions adopted, the presence of the RAFT agents alters the equilibrium constant of complex formation favouring the synthesis of the homopolymer. In addition to this side???reaction also inhibition of the copolymerisation reaction was at times encountered and an investigation into this phenomenon was also conducted.

The aim of my thesis was to evaluate two methods of monitoring of the American mink and polecat, monitoring with live traps and by floating rafts. While the first method is commonly used in our country, where the second method is the pilot project here, but it has been used in the UK successfully. Observation was from August to November in 2008 and from April to November in 2009. Observation took place in the area near PP Černíč near Dačice, where were chosen 4 different habitats. All detected tracks of Mustelids found on rafts were from the American mink. It corresponds with captures, where were captured also only minks and no polecat. Occupation of rafts was higher in 2008 than in 2009 because of the eradication of mink in November 2008. In both years the highest occupancy rate in rafts was in September and the lowest in October. There were differences between habitats and number of captured minks and tracks. In compare between rafts and traps, rafts were more successful method.
