Effect of Ion Channels on Intracellular Localization of REV-ERBα in Glioma-Initiating Cells

The number of children and young adolescents diagnosed with cancer is increasing, leading to a need for new therapeutic strategies with diminished neurodegenerative side- effects. This report presents preliminary observations on glioma-initiating cells (GICs) in the way to develop a strategy that induces cell-cycle arrest or quiescence in neural stem cells (NSCs). To test how changes in membrane potential due to pharmacological treatments have effects on localization and levels of REV-ERBα protein, proneural (PN) and mesenchymal (MES) cells were treated with varying concentrations of REV-ERBα agonist SR9009 drug and T-type calcium channel blocker mibefradil. Treatments showed that both drugs do not relocalize REV-ERBα to the nucleus. However, SR9009 decreases the levels of REV-ERBα protein, whereas mibefradil does not have a similar effect.  Our preliminary data on mouse NSCs showed they engage with REV-ERBα protein while going into contact inhibition. Therefore, we investigated whether high confluency put PN and MES GICs into quiescence and the role of the main molecular clock protein REV-ERBα in this process. Cells were grown up to certain confluency, and following qPCR gene expression analysis revealed PN cells go into contact inhibition whereas MES cells continue proliferating even after they are grown to confluency. Moreover, REV-ERBα protein does not have any role in both outcomes.

The study focuses on the establishment of the Central Presbyterian Church Garden of Olives, where through the literature and documentary focuses in Presbyterianism and its operations in the city of São Paulo. Protestantism is established in Brazil so incipient in the late nineteenth century through the early missionaries who came here with the purpose of establishing its various ramifications generated by the reform movement of the sixteenth century. A set of economic, social and judicial greatly facilitated the Protestant Gospel message found good acceptance among Brazilians, mainly arising between the liberal elite of the republican regime. At the turn of the twentieth century the City of St. Paul plays a predominant role in driving the development guidelines and policies of the country and Protestantism with its progressive message rises this locomotive and enjoy a positive opportunities that are available and establish strong communities in the capital and surroundings. But Presbyterianism shares the same divisive symptom of his peers retired and soon experienced its first internal divisions. In 1903 occurs the first major division, resulting in two denominations: the Presbyterian Church of Brazil and the Independent Presbyterian Church. A few years later, this second term simultaneous experiences two divisions: the Conservative Presbyterian Church and the Christian Church of St. Paul (1942). The Presbyterian Church Garden of Olives is also the result of a process dividing the ecclesiastical history of the United Presbyterian Church of Sao Paulo. In this research takes place there is an effort to explain the reasons and motivations that contributed most to the formation of a Presbyterian church in the city center and its efforts to establish itself. We highlight the charismatic person of Rev. Joseph Borges dos Santos Jr. who initiated, conducted and concluded this process separatist. / A presente dissertação analisa a implantação do protestantismo na capital paulista, através de um de seus ramos o presbiterianismo. O estudo tem como foco central o estabelecimento da Igreja Presbiteriana Jardim das Oliveiras, onde através da pesquisa bibliográfica e documental, enfoca-se o presbiterianismo e sua atuação na cidade de São Paulo. O protestantismo estabelece-se no Brasil de forma ainda incipiente no final do século dezenove através dos primeiros missionários que aqui chegaram com o propósito de estabelecerem suas diversas ramificações geradas pelo movimento reformador do século dezesseis. Um conjunto de fatores econômicos, sociais e judiciais facilitou muito para que a mensagem evangélica protestante encontrasse boa aceitação entre os brasileiros, principalmente entre a elite liberal advinda do regime republicano. Na virada para o século vinte a Cidade de São Paulo assume um papel predominante na condução do desenvolvimento e diretrizes políticas do país e o protestantismo com sua mensagem progressista sobe nesta locomotiva e aproveita de forma positiva as oportunidades que lhe são disponibilizadas e estabelece comunidades fortes na capital e aos arredores. Mas o presbiterianismo compartilha do mesmo sintoma divisionista de seus pares reformados e logo experimenta suas primeiras cisões internas. Em 1903 ocorre a primeira grande divisão, dando origem a duas denominações: a Igreja Presbiteriana do Brasil e a Igreja Presbiteriana Independente. Poucos anos depois, esta segunda denominação experimenta duas divisões simultâneas: a Igreja Presbiteriana Conservadora e a Igreja Cristã de São Paulo (1942). A Igreja Presbiteriana Jardim das Oliveiras também é resultado de um processo divisório na história eclesiástica da Igreja Presbiteriana Unida de São Paulo. No transcorre desta pesquisa há um esforço para expor as razões e motivações que contribuíram para a formação de mais uma igreja presbiteriana no centro da cidade e seus esforços para se estabelecer. Destaca-se a pessoa carismática do Rev. José Borges dos Santos Jr. que iniciou, conduziu e concluiu este processo separatista.

Africans, Cherokees, and the ABCFM Missionaries in the Nineteenth Century: An Unusual Story of Redemption

My dissertation, “Africans, Cherokees, and the ABCFM Missionaries in the Nineteenth Century: An Unusual Story of Redemption,” assesses the experience of American missionaries in the Cherokee nation and in Western Africa during the nineteenth century. The American Board of Commissioners for Foreign Missions (ABCFM), founded in 1810, was the first successful foreign missionary society in the U.S., and its campaign among the Cherokees served as springboard for its activities in “Western Africa”—Liberia, Ivory Coast, Gabon, and South Africa. Although the Cherokees and the West Africans were two different peoples, the ABCFM used the same method to Christianize them: the Lancasterian method with which the missionaries planned to “civilize” the Cherokees and West Africans before Christianizing them. Scholars such as William McLoughlin and Theda Purdue studied the missionary perspective and the Cherokee perspective as separate entities and convincingly maintained that the Cherokees embraced the ABCFM’s civilization and Christianization program partly to relieve the pressures on their lands and partly to adapt to the cultural pressures of their times. However, as my dissertation argues, the conversion story of the Cherokees takes a different turn if told simultaneously from the missionary and the Cherokee perspectives. Regarding the West African experience, authors such as Lamin Sanneh and Richard Gray have recently exposed the missionary and African sides of the stories with new questions that had been waiting to be asked for a long time. My dissertation, taking a unique comparative perspective, reveals first that West Africans did not face the same pressures as those faced by the Cherokees, yet, they still embraced the ABCFM’s civilization and Christianization program, though with a lesser sense of urgency and with more assertiveness than did the Cherokees despite the white missionaries’ racism. More importantly, by way of a method I call parallel agency, my dissertation offers a revisionist interpretation of the history of missions, which has traditionally emphasized the power of the white missionaries by calling into question the very assumption that the white missionaries had significantly more power than did their Cherokee and African converts.

Rôle du récepteur nucléaire Rev-erba dans les mécanismes d'anticipation des repas et le métabolisme / Role of the nuclear receptor Rev-erb alpha in circadian food anticipation and metabolism

La première partie de mon travail de thèse a été de définir le rôle joué par le récepteur nucléaire Rev-erb alpha dans les mécanismes de synchronisation par la nourriture d’une horloge circadienne putative, non encore localisée, appelée « horloge alimentaire ». La seconde partie de mon travail a consisté à étudier la participation de Rev-erb alpha dans les régulations des métabolismes glucidique et lipidique. L’ensemble de nos données indique que le répresseur transcriptionnel Rev-erb alpha joue un rôle charnière dans les fonctions circadiennes ainsi que dans le métabolisme. En effet, d’un point de vue circadien, l’absence de Rev-erb alpha altère la synchronisation à l’heure des repas – démontré par une réduction des sorties comportementales et physiologiques de l’horloge alimentaire, ainsi que par l’absence d’ajustement du rythme de la protéine d’horloge PER2 dans l’oscillateur cérébelleux. Sur le plan métabolique, la délétion de ce gène modifie notamment le métabolisme des lipides – démontré par une accumulation excessive de tissu adipeux, une utilisation préférentielle des acides gras, ainsi qu’une perte de contrôle de l’expression de la Lipoprotéine lipase. / The work performed during this PhD thesis aimed at investigating the role of the transcriptional silencer Rev-erbα in both the circadian clockwork of the food-entrainable oscillator and metabolic regulations. Firstly, by evaluating food-anticipatory components in animals fed once a day at the same time, we showed that mice lacking Rev-erbα display a reduction in locomotor activity prior to food access compared to littermate controls. Accordingly, the rises in body temperature and corticosterone that anticipate mealtime are also diminished. Interestingly, daily p-ERK expression in hypothalamic regions and daily PER2 expression in the cerebellum of Rev-erbα KO mice are not phase-adjusted to feeding time. These results indicate that Rev-erbα participates in the integration of feeding signals and in food-seeking behaviors. Secondly, by investigating energy balance in fasted, normal chow or high-fat fed animals, we revealed that Rev-erbα KO mice exhibit greater reliance on lipid fuels as energy substrates, contributing to a mild hyperglycemic state. We also found that Lipoprotein lipase (Lpl) expression, is strongly up-regulated in peripheral tissues of Rev-erbα KO mice, predisposing mice to obesity. In this regard, we uncovered a new molecular pathway that ties clock-driven Lpl expression to energy homeostasis. These findings highlight the significance of daily Rev-erbα oscillations to prevent the appearance of the metabolic syndrome.In conclusion, we provide evidence that REV-ERBα may be a part of the food-entrainable oscillator clockwork that triggers food-anticipatory components, and represents a pivotal player to link the core clock machinery to metabolic pathways.

Definition Extraction From Swedish Technical Documentation : Bridging the gap between industry and academy approaches

Terminology is concerned with the creation and maintenance of concept systems, terms and definitions. Automatic term and definition extraction is used to simplify this otherwise manual and sometimes tedious process. This thesis presents an integrated approach of pattern matching and machine learning, utilising feature vectors in which each feature is a Boolean function of a regular expression. The integrated approach is compared with the two more classic approaches, showing a significant increase in recall while maintaining a comparable precision score. Less promising is the negative correlation between the performance of the integrated approach and training size. Further research is suggested.

Subject of the thesis is to create extended model for the evaluation of information security controls. Evaluation of security controls is one from many processes of risk management which is part of information security management system ISMS. Thesis contains the outline of issue of information security and introduce various publications of information security management. Two of these publications were chosen and are used in this thesis. It is ČSN ISO/IEC 27001:2014 and NIST 800_53. These two standards are used for creation of introduced model. Model itself is introduced in second part of the thesis. Model is connecting security controls from these two standards. If organization implements security controls from NIST 800_53, meet requirements defined in ČSN ISO/IEC 27001:2014; Apendix A. This model is also customized for evaluation of security controls and giving feedback to evaluator about state of implementation of security controls. This evaluation process is setup as evaluation of NIST 800_53 security controls and after that these data are recalculated into percentage value of implementation of security controls from Apendix A. Results of this process are most valuable for risk management, for planning an implementation of security controls and for improvement of already implemented.

On the determination and evolution of fabric in representative elementary volumes for a sand specimen in triaxial compression

The soil response in triaxial compression tests, that are commonly treated as element tests, is known to be inhomogeneous. Several studies have revealed the localisation of deformation throughout the whole specimen by digital image correlation techniques on X-ray tomographies. The fabric of a soil specimen has so far only been studied on complete specimens as a bulk measurement or in chosen subsets. In this contribution, we present a study on the spatial and temporal distribution of the fabric throughout one Hostun sand sample in triaxial compression. Therefore, we calibrated the minimum representative element size first for three chosen fabric variables considering three different criteria. By distributing the elements in a regular grid over the specimen, we are able to clearly identify the onset of the localisation in terms of void ratio, coordination number and contact fabric anisotropy. Spatially and temporally the contact fabric variables precede the void ratio changes as they are much more sensitive to small changes.

Teknikämnet har, sedan det fick en egen kursplan, åsidosatts i skolorna. Deltagande observationer och intervjuer med lärare verksamma i varierade åldersgrupper, visade samma resultat gällande tankar kring teknikundervisningen som de uppfattningar som litteraturstudien visat. Det framgår att många lärare inte uppfattar sin ämneskompetens som tillräcklig och att de inte vet hur de ska förhålla sig till ämnet i undervisningen. Resultaten av dessa studier ligger till grund för ett läromedel, bestående av en elevbok och en lärarhandledning, som utgår både från Lpfö 98 och centrala innehållet för år 1-3 och 4-6 ur kursplanen i teknik i Lgr 11. / Technology has been neglected at schools, since the subject got it’s own syllabus. After making observations of teaching in various age groups, and also conducted interviews with teachers, their opinions and thoughts about technology were in accordance with the findings from the literature study. It seems that many teachers don’t have full confidence in their own knowledge of the subject, and they were also unsure on how to approach the subject in their teaching. The results of the studies inspired the creation of a teaching-aid which consists of two books, one is a teaching-guide and the other is for the pupils. It is based on Lpfö 98 (revised 2010), as well as the core content in year 1-3 and 4-6 from the technology syllabus in Lgr 11.

Les orosomucoïdes, membres de la superfamille des lipocalines, sont parmi les protéinesplasmatiques les plus abondantes. Ce sont des protéines de la phase aiguë de l’inflammationsecrétées par les hépatocytes en réponse à un stress (inflammation, cancer, cirrhose…), etutilisées couramment en clinique comme marqueur d’un état pathologique. Elles vont lorsd’un état inflammatoire inhiber la prolifération des neutrophiles et des lymphocytes. J’ai pumontrer que le récepteur nucléaire FXR, impliqué dans de nombreuses voies métaboliques etplus récemment comme ayant un rôle anti-inflammatoire dans le foie, régule l’expression desorosomucoïdes exclusivement au niveau hépatique chez la souris. Cela suggère que larégulation de certains processus inflammatoire par FXR pourrait être expliqué par larégulation transcriptionelle des orosomucoïdes.Le second sujet met en évidence une relation entre le récepteur nucléaire Rev-Erba et la OGlcNAcylationau niveau hépatique. La O-GlcNAcylation est une modification posttraductionnelledes protéines qui permet l’ajout d’un groupement O-GlcNAc. De nombreusesprotéines impliquées dans de multiples processus biologiques comme la régulation du cyclecellulaire, du rythme circadien, ou encore dans de nombreuses voies métaboliques, vont êtreO-GlcNAcylées. La dérégulation de la O-GlcNAcylation est également impliquée dans denombreuses pathologies telles que le diabète, le cancer ou encore la maladie d’Alzheimer. Lerythme circadien est un important régulateur du métabolisme. De plus en plus d’indicesindiquent que la O-GlcNAcylation est un intermédiaire essentiel dans la régulationcircadienne du métabolisme. Des modulations circadiennes de la O-GlcNAcylation deBMAL1, CLOCK et PER ont été mises en évidence. Ce lien entre la O-GlcNAcylation et lerythme circadien nous a poussé à essayer d’identifier le rôle potentiel de Rev-Erba, récepteurnucléaire impliqué dans le contrôle de la ryhtmicité circadienne, dans la régulation de la OGlcNAcylation.Nous avons pu montrer par ces travaux que Rev-Erba est un régulateurimportant de la O-GlcNAcylation. La déficience en Rev-Erba induisant une baisse importantede la O-GlcNAcylation. Rev-Erba ne régule pas le niveau de transcription des enzymesimpliquées dans la O-GlcNAcylation mais est capable d’intéragir avec l’OGT (enzymeresponsable de la O-GlcNAcylation) et d’inhiber sa dégradation par le protéasome. Par cemécanisme de stabilisation de l’enzyme OGT, Rev-Erba se révèle être un régulateurimportant de la O-GlcNAcylation. / Orosomucoïdes, members of the superfamily of lipocalines, are among proteins plasmatiques the most plentiful.
