Regeneration of Scots pine (Pinus sylvestris L.) under droughtMacAllister, Sarah Louise January 2016 (has links)
Drought-induced tree mortality is a phenomenon affecting many forest ecosystems and is predicted to increase under ongoing climate change. Forest stability partly depends on regeneration: the process of renewing mature forest with subsequent generations. As seedlings are more susceptible to drought effects than mature trees, mortality of the seedling bank can represent a major bottleneck controlling forest structure and species composition. Scots Pine (Pinus sylvestris L.) is the most widely distributed of the Pinus species, covering a broad latitudinal gradient of ecological conditions. The thesis aims to deepen understanding of drought-induced mortality, while analysing intra-specific variation in the phenotypic and metabolic profile of Pinus sylvestris seedlings subjected to drought stress. I also consider the relevance of the results to the broader conceptual framework of drought-induced mortality. The experiments utilise seeds from different populations of origin (provenances) across the north-south axis of the European range of Pinus sylvestris, in order to determine the extent of regeneration capacity in this species under drought. Seeds were collected from different populations (provenances) that, along with other climatic and edaphic differences, span a gradient of water availability: from wet (Scotland) to intermediate (Austria, Poland) to dry (Spain). In Chapter 2, the effects of osmotic stress on the initial seedling establishment stage were studied by comparing phenotypic responses across provenances. Seedling germination, early growth, osmotic stress tolerance and survival were investigated using a polyethylene glycol irrigation treatment as a proxy for rapid and severe drought. Treatment, provenance and interaction effects were found for rate of germination, final proportion of seeds germinated, seedling size, and superoxide dismutase activity (an antioxidant enzyme). Root investment was affected by both provenance and time to germination. Although there was no significant effect of provenance on survival, a trend towards increased probability of survival under osmotic stress was indicated for the southernmost (driest) as compared with the northernmost (wettest) provenance. Chapter 3 investigates the responses of older seedlings (at 10 months) to a drying down of soil moisture for 40 days. Morphological and physiological data were collected to assess intra-specific and intra-population variation in the seedling stress response under drought. A metabolomics analysis using Ultra performance Liquid chromatography followed by mass spectrometry (UPLC/MS) was carried out to investigate whether metabolic markers could be identified that are suggestive of heightened oxidative stress and whether populations in different climatic and edaphic environments show variation in metabolic activity under drought. Preliminary results suggest large intra-population variability yet clear differentiation in metabolic responses to drought over the time course of the experiment. Univariate and multivariate analyses indicated that among the most significant increases in response to drought were those involved in osmoprotective and antioxidant capabilities, including the free amino acid proline and a quercetin derivative (a flavonoid). Interestingly, provenances, either under experimental drought or not, did not show significantly different metabolite profiles, even though provenance and its interaction with drought treatment did significantly affect seedling biomass and photochemical efficiency. In Chapter 4 the effects of provenance, maternal parentage and seed weight on germination rate, final germination percentage, as well as seedling drought responses in biomass allocation and the expression of selected antioxidant genes were analysed. Seed weights were measured individually and seed weight was found to have a strong positive effect on: germination rate, seedling dry weights, and number of needles. Expression of two antioxidant enzymes increased under drought. Seed weight was strongly determined by provenance and maternal parentage as well as their interaction. However, root to shoot biomass allocation depended on provenance and maternal effects that were not mediated by seed weight effects. Principal component analysis indicated that the Spanish provenances could be characterised by a higher root to shoot ratio and stem weight. Specific leaf area was also found to be lowest for the Spanish provenances.

Abstract: After the standards implanting for "Produção de muda Certificada de Citros no Estado de São Paulo", in the year of 1998, there was a big change in the production system of citric seedling, passing from the production system outdoors to a protected atmosphere, or the production of the seedling in greenhouses, and this directly affected the traditional producers, which until then were to produce outdoors. Those changes were necessary due to the appearance of diseases and nuisance that were putting in risk the Brazilian citric production, considered today the biggest of the world. The disease "Clorose Variegada dos Citros" (CVC), is the main disease that unchained such changes. Transmitted by a kind of insect vector, it demanded that the seedling production was made in a greenhouse atmosphere to avoid contamination, which before this, on outdoors used to proliferate a lot. As most part of the seedling production was made by producers with limited technical knowledge, the new system demanded lots of changes in the production system, adoption of new techniques of production, new handlings and high investments to build greenhouses. Even having rules to be followed, each producer adapted in its own way to the new production system, not committing to the quality. This essay analyzed influences of those changes on the producer, through questionnaires and the answers were Tabbed and compared to the Seedling Production Directions. It was concluded in this work that the main objectives of directions were attained with a higher control on the seedling diseases and knowledge on the origin of the genetic material. The initial concern about the producers difficulty in adapting to the new directions, did not come true, once there was a high comphance to the changes by the producers. / Mestre

Frutos, sementes e plântulas de três espécies de Mimosa Linnaeus(Fabaceae: Mimosoideae) : aspectos morfoanatômicos e considerações ecológico-filogenéticas /Iwazaki, Maísa de Carvalho. January 2008 (has links)
The genus exhibits varied morphological and anatomical characteristics, habits, and fruit types. Mimosa daleoides Benth., M. dolens Vell. var. anisitsii (Lindm.) Barneby, and M. orthacantha Benth. are shrubs that commonly occur in remnant areas of cerrado vegetation in São Paulo State and are often considered invasive plants. The present work describes the morphology, anatomy, and ontogeny of the fruits and seeds of this species, examines the morphology of the seedlings, the anatomy and venation of the cotyledons and eophylls, and compares the three species and information concerning them in the published literature. The plant material was processed according to traditional techniques in light microscopy, including embedding in methacrylate, sectioning in a rotary microtome, and staining with toluidine blue. Additionally, semi-permanent slides were prepared for maceration analyses, clearing leaves, and free-hand sections. The ovary of the three species is short-stipitate, with 3-5 anatropous ovules, tending towards a campylotropous only in M. daleoides. Emergences form in the pericarp during fruit development; the mesocarp is parenchymal, with an outer and inner hypoderm in M. dolens var. anisitsii but only an inner hypoderm with phenolic cells in M. daleoides. An adaxial meristem develops, from which the outer and inner endocarp are derived; the outer is lignified at maturation, and an inner is parenchymatous. Mimosa orthacantha demonstrates fibers and sclerids oriented in opposite directions in the outer endocarp, a characteristic that differentiates this species from the others. Anatomical analyses indicate that the fruits of M. daleoides and M. orthacantha are craspedia and are fixed to the inflorescence axis of the raceme ...(Complete abstract click electronic access below) / Mestre

They present a distinct spatial distribution along the topographic gradient of the region, characterized by lowlands, slopes and plateaus. Seedlings of this species are found in all habitats. However, adults of O. bataua only occur in lowland habitats and, by instane, O. bacaba and O. minor are found only in the slopes and plateaus. The aim of this study was to describe the spatial distribution of individuals' ontogenetic stages of three species of the genus Oenocarpus and to understand how these individuals are affected by environmental factors that typify the habitats of lowland, slope and plateau in the Central Amazon region. A total of 72 plots of 50x5 m were set up at the three habitats of three continuous forest reserves of the Biological Dynamics of Forest Fragments Project (BDFFP), located about 80 km from Manaus, AM. The three habitats can be distinguished by some soil characteristics, especially texture and water content. The canopy cover of the trhee habitats were similar. Five ontogenetic stages were determined for the three species: seedling, infant, juvenile, immature adult and reproductive adult. The patterns of spatial distribution for the three species was not correlated with any of the soil variables, except for juveniles of O. bataua, which showed a positive correlated with water content. This study allows us to assert that differences in the physical environment of Central Amazonia may be important on a local scale for the distribution of palms trees. Although only some ontogenetic stages of three species have responded to the soil characteristics, spatial segregation amongst habitats was evident and the relationship between each species and the environment varies accordantly to the ontogenetic development, as a niche segregation form on a local scale, visible in adults of this co-generic species / Mestrado / Ecologia / Mestre em Ecologia

Development of coffee seedlings produced in plastic tubes, under termo-reflective and black screens.Vinicius Maia Costa 12 April 2004 (has links)
This experiment evaluated, in greenhouse conditions, the development of coffee seedlings produced in plastic tubes, under the effect of screens used for shading. The treatments were black screen 50%, known commercially as "sombrite" or as "black screen", considered standard for the development of these seedlings, and thermo-reflective screens with four different percentages of attenuation of the solar radiation, being meshes 40%, 50%, 60%, and 70%. The adopted experimental delineation was randomized blocks with 5 treatments and 4 replicates, making up 20 parcels. The parcels were represented by 20 benches; each one with superior closing system with one type of screen, and also side closing system, attenuating the side solar radiation. Each parcel was composed by 175 seedlings, from which 161 were used as a border, and 14 used for the evaluations. The irrigation, daily carried out, was implemented with the use of a semi-automated motorized bar that dislocated longitudinally along the greenhouse in both directions. The desired water amount was set in accordance to the bar displacement speed that was established by means of a frequency inverter, responsible for this control. To monitor the microclimate, one psicrometer was installed inside each parcel and, from time to time, global solar radiation, photosynthetically active radiation, and net radiation (SR) measures were taken, establishing, this way, relations of global transmissivity (TRG) and photosynthetically active transmissivity (TRFA). The physiological variables used to evaluate the development of the seedlings were leaf area, seedlings height, and stalk diameter. Analyses on the dry matter of aerial part and root system and total dry matter were carried out at the end of the experiment. The evaluation of the psicrometers data showed that the temperature of air gently varied in the parcels, as well as the air relative humidity. Relating to TRG and to TRFA, it was verified that screens with less attenuation of the solar radiation presented higher transmissivity in general, as expected. For the variable net radiation, the presence of environment interference made most data impracticable, verifying a partial trend of higher SR in the screens 40% and 50%. The evaluation of the physiological variables showed that the leaf area did not present significant difference among the treatments. Yet, it was verified that, for seedlings height and stalk diameter, the seedlings produced under thermo-reflective screens 40% and 50% were superior to those produced under black screen 50% and thermo-reflective screen 60% and 70%. It was also verified that, for the evaluations of dry matter, the seedlings produced under thermo-reflective screens 40% and 50%, and black screen 50% comparatively accumulated more dry matter than those produced under thermo-reflective screens 60% and 70%. Thus, under the conditions where the experiment was carried out, it can be concluded that the thermo-reflective screens 40% and 50% and black screen 50% presented better results than the thermo-reflective screens 60% and 70%.

Predation on Early Recruitment in Mediterranean Forests after Prescribed FiresSagra, Javier, Moya, Daniel, Plaza-Álvarez, Pedro, Lucas-Borja, Manuel, Alfaro-Sánchez, Raquel, De Las Heras, Jorge, Ferrandis, Pablo 08 July 2017 (has links)
Wildfires play a significant role in many different elements of Mediterranean forest ecosystems. In recent years, prescribed fires have started being used more often as a fuel reduction tool, and also as silvicultural treatment to help the regeneration and health improvement of stands. Apart from the fact that fire may alter microsite conditions, very little is known about the impact of prescribed burning on natural regeneration or plant species renewal in Mediterranean pine forests. Likewise, knowledge about the influence of seedling predators on post-fire regeneration is still scarce. In this study, we aimed to compare the effects of seedling predation on recruitment in earlier stages after prescribed burnings in three pine stands in Central Spain: a pure stand of Pinus nigra; a mixed stand of Pinus halepensis and Pinus pinaster and a mixed stand P. nigra with P. pinaster. In situ we superficially sowed seeds from two different species. In the sowing experiment, we tested two different seed provenances (drier and more humid spanish regions) for each species. In all, 60 plots (30 burned, 30 unburned) per site, with 10 seeding units per plot and more than 20,000 seeds, were used in the whole study. Seedling predation was evaluated by replicating the seeding units inside and outside a wire cage as protection for rodents and birds. Our results showed that prescribed fires alter initial seedling predation intensity: predation was significantly higher in the seedlings grown in the plots affected by prescribed fire. The individuals sown before the fire passed showed slightly more predation than those sown after fire passage. Provenances did not appear as an important predation drive. Understanding the role of the predation associated with these treatments can help improve Mediterranean pine forest management.

Seedling quality, plant growth and fruit yield and quality of tomato (Solanum lycopersicum L.) in response to Trichoderma harzianum and arbuscular mycorrhizal fungiNzanza, Bombiti 04 September 2012 (has links)
Existing evidence suggested that nursery inoculation with Trichoderma harzianum and arbuscular mycorrhizal fungi (AMF) could reduce deleterious effects of biotic and abiotic stresses and improve seedling quality, fruit yield and quality of tomato (Solanum lycopersicum L.). However, studies of their combined inoculation on seedling growth, fruit yield and quality of tomato plants are not well-documented. Experiments were carried out to investigate the combined effect of T. harzianum and AMF on tomato crop performance under various conditions. When combined with a T. harzianum and AMF mixture, seaweed extract from Ecklonia maxiama inhibited AMF root colonisation of tomato seedlings. Treating seedlings with a mixture of T. harzianum and AMF reduced the incidence of Verticillium wilt in tomato grown in a nethouse at early season, with negligible effect on fruit yield. Further investigations were initiated to find out whether T. harzianum and AMF were efficient when applied as a mixture or alone, at different inoculation times. Co-inoculation with T. harzianum and AMF (Glomus mosseae) improved seedling growth and development, except when both fungi were simultaneously applied two weeks after sowing. When the seedlings were allowed to grow up until full harvest in a greenhouse, both fungal inoculants increased total yield and marketable yield, but these increases were not significant. Furthermore, inoculation with AMF increased the percentage of extra-large fruit. Field experiments conducted under commercial tomato production confirmed greenhouse studies. Inoculation of tomato with T. harzianum and AMF, either alone or in combination increased early fruit yield (four first harvesting weeks). Throughout the studies, percentage AMF root colonisation in seedlings and plants remained low, despite nursery inoculation. Field experiments investigated the effects of AMF-inoculated transplants combined with biochar-amended soils on AMF root colonisation and their resultant effects on overall crop performance and microbial community structure. Biochar had no effect on AMF root colonisation, and also when combined with AMF, it had no influence on tomato productivity. Interestingly, biochar altered the fungal community while AMF might have influenced the bacterial community such as plant-growth promoting rhizobacteria, which are associated with improved plant growth, nutrient uptake and disease control in the rhizosphere. These benefits could contribute to improved yield and fruit quality. In conclusion, although the results were variable, there was a clear indication that T. harzianum and AMF can play an important role in tomato production. / Thesis (PhD)--University of Pretoria, 2012. / Plant Production and Soil Science / unrestricted

Identification et caractérisation d'une lipase exprimée pendant l'hydrolyse des réserves chez Arabidopsis thaliana / Identification and characterization of a lipase expressed during Arabidopsis thaliana reserves hydrolysisZallot, Rémi 10 November 2011 (has links)
In germinating seedlings of Arabidopsis thaliana, fat storage breakdown is initiated by lipases. A protein capable to bind to a lipase inhibitor was identified from an extract of rape seedlings and its amino acid sequence found to resemble that of known lipases. Transient expression of the Arabidopsis orthologous gene led to a 100-fold increase in lipase activity in Nicotiana bethamiana leaves. Taken together, these data strongly suggest that this protein is indeed a lipase. In vivo localization studies using a GFP fusion protein in Nicotiana benthamiana as a transcient expression host showed a peroxisomal localization. In Arabidopsis, the gene coding for this lipase was found to be mainly expressed in seedlings during fat storage breakdown. Most lipase activity was abolished in germinating seedlings of an Arabidopsis mutant for this gene. These data suggest that this lipase is likely involved in the breakdown of fat storage in germinating seedlings of Arabidopsis. However, oil mobilization was not affected in Arabidopsis mutant plants. This might suggest that the effect of the mutation could be compensated for by other lipases.
