Plant breeding aspects of ryegrasses (Lolium sp.) infected with endophytic fungi

Some aspects of the presence of systemic endophytic fungi in agriculturally important New Zealand grasses were studied in relation to plant breeding. Seedling resistance to adult Argentine stem weevil feeding in perennial ryegrass, Italian ryegrass and tall fescue was found to be related to the presence of their respective Acremonium endophytes in the seed rather than to plant genetic resistance. In addition a study of perennial ryegrass revealed that this resistance was independent of endophyte viability. The seedling resistance conferred by the endophyte of Italian ryegrass was found to be beneficial for field establishment. This endophyte differs from that in perennial ryegrass and tall fescue in that it does not confer resistance to Argentine stem weevil on mature plants, but only on seedlings. The extent of plant genetic seedling tolerance to adult Argentine stem weevil feeding was limited to broad inter-specific differences, with tall fescue more tolerant than perennial ryegrass and both of these more tolerant than Italian ryegrass. This ranking corresponds with previous observations on feeding preference on mature plants. A study of factors affecting the concentration of endophyte mycelia in infected seed of perennial ryegrass revealed that plant genetic factors had little effect. The major factors studied were: 1) the endophyte concentration in the maternal parent plant directly influenced the endophyte concentration in the seed. 2) nitrogen fertilizer applications to a seed crop reduced the concentration of mycelia in the seed, with earlier applications having a greater effect. 3) application of the fungicide propiconazole (Tilt) to a seed crop reduced the endophyte concentration in the seed. 4) the endophyte concentration in the seed was found to directly influence the endophyte concentration in seedlings, six month old plants and that of seed harvested from a first year seed crop. As there have been no previous reports of tetraploid perennial ryegrass cultivars with endophyte an experiment was conducted to determine if these could be developed by the standard procedure of colchicine treatment. The results revealed that endophyte was retained following colchicine treatment.

De-icing salt and the roadside environment : Air-borne exposure, damage to Norway spruce and system monitoring

After decades of investigation, we still have to deal withthe problem of environmental effects of the use of de-icingsalt on roads. Lacking useful indicators of the system, westill do not know the environmental utility of ameliorativemeasures taken. The thesis aims at i) describing the system of de-icing practices and theirenvironmental effects with special reference to the exposure ofthe roadside environment to air-borne salt and damage to Norwayspruce seedlings and ii) at proposing indicators for afollow-up system. By collecting bulk deposition and relatingthe deposition pattern to factors concerning wind androad-maintenance characteristics, the influence of thesefactors on air-borne exposure is investigated. By exposingNorway spruce (Picea abies(L.) Karst.) seedlings to roadsideconditions the influence ofsalt exposure on the degree ofdamage is investigated. Based on a comparison of severalsystems of monitoring, improvements are suggested by proposingnew indicators for salt use and its environmental effects. A large part of the salt that is applied on the road surfacewill be transported by air and deposited on the ground in theroadside. While the vast majority of the salt will be depositedwithin some tens of metres of the road, some may still be windtransported several hundred metres away. The wind directionplays an important role for the deposition already at adistance of some ten metres from the road. Wind speedinfluences the distance to which the salt is transported.Chloride concentration in unwashed needles collected after thesalting season is positively related to the bulk deposition ofCl during the exposure. The degree of damage can be describedby a sigmoid curve of response to the Cl concentration inneedles. Plant community dynamics governed by red harvester ant (Pogonomyrmex barbatus) activities and their role as drought refugia in a semi-arid savanna

This study examined modifications made by Pogonomyrmex barbatus, by their processes of granivory and nest construction, to forb and grass dynamics under large-scale disturbances of fire, recent drought and long-term, large-mammalian herbivory using comparative studies, field experimental manipulations, and a simulation model on the Edwards Plateau, Texas. Ant nests are refugia for grass survival during extreme droughts as demonstrated during the drought of 1998 to 2002. Significantly greater cover of grasses and lower abundance and cover of forbs was found beside nests compared with surrounding habitat throughout the drought and recovery. Grasses near nests may be the seed source for surrounding habitats during recovery. Seeds were differentially collected among most forbs and grasses despite seed abundance. Harvest was significantly reduced in the fall relative to spring. During preference experiments, harvest differences were found between grazing treatments for two of four species, but only during the spring. High lipid content seeds were unpreferred in fall compared to high protein and carbohydrate content seeds. Granivory influences on seedling establishment were studied by comparing seedling recruitment among sown and naturally occurring seeds excluded and open to foragers. Exclosures were placed in three nest densities and two burn treatments. Seeds in exclosures produced significantly more seedlings than open arenas only during the first year of drought recovery. Densities of grasses and annual forbs were higher in open arenas the second year due to indirect effects of granivory. By reducing seeds ants release seedlings from competition. Sown seedling abundance was unaffected by colony density and fire. Colony density and distribution were influenced by topography, soil types, soil depth and woody cover, but not by historical grazing treatments. Cleared vegetation on nest disks impacted less than 1% of total surface area and losses were compensated by greater basal cover of grasses next to disks compared to surrounding habitats. Foraging areas influenced 17.3-73.6% of surface area and could diminish seed populations for potential seedlings. Model results agree with experimental observations that communities may be modified by P. barbatus presence due to differential responses of grass species to interaction between nests or granivory and rainfall amounts.

Development of Short Term Storage Techniques for Grafted Vegetable Seedlings

Vegetable grafting confers soil-borne disease resistance through the use of selected rootstocks. Additional costs associated with grafted vegetable seedling propagation limit grower access to this technology. The use of low temperature storage in grafted seedling production reduces labor costs and allows propagators to meet the seasonal and narrow-window demand of growers. For the first part of the experiment 22 genotypes of Solanaceae or Cucurbitaceae seedlings were evaluated in low temperature storage conditions (5 or 12°C). Seedling performance was better in 12°C storage than in 5°C storage. For the second part of the experiment watermelon, the most low-temperature sensitive species found in our first study, was grafted onto two different commercial rootstocks or on to other watermelon seedlings and stored at 12°C for two and four weeks. Both 'Strong Tosa' and 'Emphasis' rootstocks conferred chilling tolerance during storage to watermelon scions, allowing storage of grafted plants for two weeks.

Lietuviškų šilauogių veislių tyrimas / Research of Lithuanian Cultivars of Blueberry

Blueberry though one of the most valuable berry plants is limited in its occurrence in Lithuania. One of the reason – is lack of local cultivars. Over 1993 – 2004 the largest blueberry germ plasm was established in Kaunas Botanical Garden at VDU. This collection has been used for breeding of new cultivars. Seedlings ‘No.11, ’‘No.16’ and ‘No.17’ were investigated in Kaunas Botanical Garden over 2002 – 2004. The investigation aimed at estimating of productivity, character of fruiting and biochemical composition of berries. The obtained result were compare with those of halth-highbush ‘Northcountry’. Fruiting of 4 – 6 year old bushes was estimated, mean earliness of fruiting, berry size, volume and average yield for 3 seedling numbers were determined. Seedlings numbers ‘No.11, ��‘No.16’ and ‘No.17’might be classified as early cultivars. The main port of the blueberry yield was obtained in the first picking, due to this mechanised picking might be applied. The plants produce tasty, good marketable appearance and biochemical composition. The best marketable appearance typical of ‘No. 17’, the biggest berries – ‘No. 11����. The most productive were ‘No. 16’ and ‘No. 17’, their mean productivity is two times higher than that of ‘Northcountry’.

Ectomycorrhizal Fungi Facilitate Competitive Interactions Between Tree Taxa: Host Preference, Seedling Recruitment, and Forest Succession

<p>The mycorrhizal mutualism is one of the earliest and most influential of all terrestrial symbioses. As the primary method used by most plants to acquire nutrients from the soil, mycorrhizal fungi help to shape the structure and composition of many ecosystems. Ectomycorrhizal (EM) fungi play an especially significant role because most EM fungi prefer a limited number of host taxa, and EM plant species likewise associate with only a fraction of the available EM fungi. This host preference issue, combined with the high diversity of EM fungi in forest ecosystems, complicates interspecies competition both among fungi and among plants, because these plant and fungal communities interact.</p><p>Despite recent attempts at documenting mycorrhizal fungi in the context of ecological succession, many questions remain about the underlying causal relationships among EM fungi, soil conditions, and plant community assembly. The succession of mycorrhizal fungi often mirrors the succession of plants, and ectomycorrhizal (EM) community composition may affect the outcome of competition among trees during succession. In a pine-oak seral system, we tested the ability of <italic>Pinus taeda</italic> and <italic>Quercus alba</italic> seedlings to associate with EM fungi when planted under both conspecific and heterospecific adults. We found that EM communities under pine and oak canopy were distinct regardless of seedling identity, indicating that the fungal associations of adult trees determine which EM species are available in the soil. In addition, pine seedlings planted under oak canopy showed decreased mycorrhization and growth compared to those planted under pine canopy, while oak seedlings showed no negative effects of heterospecific planting. This impaired ability of pine seedlings to associate with the EM community established under oaks may deter pine recruitment and facilitate the late-seral replacement of pines with oaks.</p><p>While EM fungal communities correlate with the dominant species of host tree, soil properties do as well, making it difficult to establish causality among these three variables. Soil was collected from oak- and pine-dominated stands and dried to kill off mature mycelium, leaving only the spore bank as a source of inoculum for pine and oak seedlings. EM root tips were collected for molecular identification of fungal species based on ITS barcoding, and soil samples from field and laboratory conditions were analyzed for fungal diversity using 454 sequencing. We found a reduced influence of canopy type and a more pronounced influence of seedling identity when compared to the EM communities on seedlings planted in the field, suggesting that adult trees do alter the availability of fungi by directly promoting the growth of their preferred EM associates.</p><p>The availability of EM fungi can also affect seedlings at the interface between EM- an AM-dominated forest. We tested the hypothesis that seedlings of <italic>Dicymbe corymbosa</italic> which recruit outside of monodominant stands have limited access to EM symbionts compared with those which recruit inside <italic>D. corymbosa</italic> stands. EM root tips and rhizosphere soil were collected from seedlings along two transects inside monodominant stands and three transects in the transition zone into mixed forest dominated by AM associates. Seedlings inside monodominant stands yielded both a greater quantity of mycorrhized root tips and a higher diversity of EM species than transition zone seedlings. Of the fungal families commonly found on adult roots, the Boletaceae were notably underrepresented on all seedlings. In the transition zones, high-throughput sequencing of soil also detected a decrease in EM diversity with distance from the parent tree.</p><p>Seedlings of <italic>D. corymbosa</italic> may benefit from recruiting within monodominant stands by tapping into common mycorrhizal networks (CMNs) to acquire low-cost nitrogen and, potentially, photosynthates produced by conspecific adults. Leaves of stand adults, stand seedlings, and mixed-forest seedlings were collected for stable isotope analysis to track the transfer of nitrogen and carbon through CMNs. The &delta;<super>13</super>C and &delta;<super>15</super>N results contradicted each other, suggesting that more complicated interactions may be playing out among adults, seedlings, and fungi.</p> / Dissertation

The role of seed dispersal, seed predation and drought in the restoration of Ngel Nyaki Forest, Nigeria.

Abstract The restoration of degraded landscapes has become one of our most valuable tools for conservation, however there are many factors which can restrict natural regeneration and impede active restoration attempts. The purpose of this study was to investigate three key processes which commonly limit the establishment of forest tree species into abandoned pasture in tropical forests: i) dispersal limitation, ii) seed predation, and iii) competition from the grass sward. Seed dispersal I identified 59 species of birds that were using the grassland habitat. Through 216 hours of focal tree observations I established that isolated trees in the grassland that had larger canopies, and those that were providing a food source (i.e. flowers or fruit) had significantly higher bird visitation rates and average stay lengths. I found evidence of the “perch effect” as patches of remnant trees encouraged more birds into areas of grassland, and the density of seedlings under tree canopies was significantly positively correlated with bird visitations. 95% of the seedlings found beneath tree canopies in grassland were of a different species to that of the tree canopy above them, demonstrating the dispersal of seeds from elsewhere into these microhabitats. 98% of these seedlings are grassland or forest edge species showing forest core species are still dispersal or microsite limited despite the effect of these trees. Seed predation Removal rates of seeds from experimentally laid out seed piles varied among seed species, the habitat the pile was in, and the predator guild able to access the piles. Preliminary results indicate that these trends are driven by the ecology of the seed predator. Removal of seeds by vertebrates was highest in the core forest, while ant predation was constant across all habitats. Vertebrates removed the larger seeds (Entandrophragma angolense and Sterculia tragacantha) while ants preferred the smaller Celtis gomphophylla and Croton macrostachyus. Overall predation rates in grassland were lower than those in the forest, and the presence of remnant trees did not influence predation rates, a positive sign for regeneration and the survival of seeds dispersed into these areas. Competition from the grass sward While the grass sward provides shade for seedlings of forest tree species it is also a harsh environment for them, as the grass competes with seedlings for water. Removing the grass and covering planted seedlings with artificial shading structures significantly increased both the survival and growth of these seedlings. Recommendations From this study I was able to make recommendations for a low input restoration program at Ngel Nyaki. Planting seedlings in small „islands‟ takes advantage of the natural increase in dispersal of seeds under isolated trees, while low seed predation rates increase the chance of survival of these seeds to germination. Planting these seedlings under shade will lead to increases both their growth rates and their survival. Once the secondary forest develops, under-planting seedlings of core forest trees will introduce them to the system, as the natural establishment of these seeds appears to be limited in the current environment. This study has also served to remind us how little we know about this particular forest-grassland system, and has led to the development of ideas for further investigations into several more aspects of regeneration.

The development of established shrub seedlings in persistent historical reindeer milking grounds

This study focuses on how established shrub seedlings (Salix myrsinifolia x phycilifolia and Vaccinium vitis-idaea) develop in historical milking grounds. Historical milking grounds are cultural remains from the intensive reindeer herding era when the Sami migrated with closely controlled herds. Although the places were never fenced, the high concentration of reindeer close to the tenting grounds created patches of grass and forb dominated vegetation in areas outherwise dominated by deciduous or ericoid shrubs. Despite about 100 years of abandonment the shrubs have not come back and the milking grounds are still clearly visible in the landscape. One theory why the former milking grounds are so stable is that shrubs cannot establish from seedlings due to unfavorable abiotic conditions, or due to competitive dominance of already established forbs and grasses. I tested this hypothesis by planting shrub seedlings in the milking grounds and in reference areas, with and without neighboring vegetation and investigated seedling survival and growth. The results show that shrub seedlings are able to both survive and develop in milking grounds suggesting that the seedling state is not the limiting factor in shrub encroachment in the milking grounds.
