Fenotypning av trombocytantigen HPA1a med flödescytometri: screening för att finna blodgivare som saknar HPA1a

HPA1a är ett antigen på trombocytytan som kan orsaka alloimmunisering, såsom neonatal alloimmun trombocytopeni och post-transfusion purpura, vilket kan ge svåra blödningssymptom. I fall med antikroppar mot HPA1a måste kompatibla trombocyter finnas tillgängliga. Syftet med studien var att etablera en flödescytometrisk screeningmetod för fenotypning av HPA1a antigen på trombocyter samt att finna HPA1a negativa blodgivare. Före flödescytometrisk analys poolades två till fem blodprover samman till ett prov och vid förekomst av HPA1a negativa trombocyter i poolen analyserades proverna individuellt. Fluorokrommärkta anti-humana antikroppar mot CD42a och CD61 användes för att särskilja HPA1a negativa trombocyter från HPA1a positiva. Totalt fenotypades 177 blodprover varav sju (4%) typades som HPA1a negativa. Av de sju fynden genotypades fyra vid ett externt laboratorium vilket bekräftade att de var HPA1a negativa. Flödescytometrisk screening av HPA1a är snabb, pålitlig och lämplig för storskalig screening. För att fastställa prevalensen av HPA1a negativa individer behöver mer omfattande studier utföras där en större population ingår. Att ha flera HPA1a negativa blodgivare registrerade ger möjligheten att hjälpa patienter i andra regioner. / HPA1a is an antigen on the platelet surface that can cause alloimmunization, such as neonatal alloimmune thrombocytopenia and post transfusion purpura, which can cause severe bleeding symptoms. In case of antibodies against HPA1a, compatible platelets must be available. The purpose of the study was to establish a flow cytometric screening method for phenotyping HPA1a antigen on platelets and to find HPA1a negative donors. Before the flow cytometric analysis, two to five blood samples were pooled into one sample and in the presence of HPA1a negative platelets in the pool, the samples were analyzed individually. Fluorochrome –labeled anti-human antibodies to CD42a and CD61 were used to distinguish HPA1a negative platelets from HPA1a positive. A total of 177 blood samples were phenotyped, of which 7 (4%) were HPA1a negative. Of the seven findings, four samples were genotyped at an external laboratory confirming that they were HPA1a negative. Flow cytometric screening of HPA1a is fast, reliable and suitable for large scale screening. In order to determine the prevalence of HPA1a negative individuals, more extensive studies need to be performed involving a larger population. By having many registered HPA1a negative donors, it can provide opportunities to help patients in other regions.

To complement the research, I intend to present, based on a section of the same magazine, composed of articles written, who the characters are presented and what is, in fact, the relevance of these within the scope of science. / O objetivo deste estudo é analisar como é feita a divulgação da ciência nas capas da revista Galileu. Através de uma apresentação de como se configura a prática jornalística e de arcabouços teóricos filiados à Análise do Discurso de linha francesa, pretendo discutir como se estabelece a relação imagética e textual na capa da publicação de modo a apresentar qual é o efeito pretendido junto ao leitor: apresentar a produção científica ou vender o número. Para complementar a pesquisa, pretendo apresentar, com base em uma seção da mesma revista, composta por artigos assinados, quem são os personagens apresentados e qual é, de fato, a pertinência dos mesmos dentro do âmbito da ciência.

It is estimated that the elderly population will triple in the coming decades, from 20 million in 2010 to 65 million in 2050. Many actors are responsible for implementing the National Health Policy for the Elderly, among them are the local health managers and articulators health of the elderly. The aim of this study is to analyze the social representations of the National Health Policy for the Elderly in a regional health department, the light field of science, technology and society. This is a qualitative, exploratory and descriptive. The methodological approach was to document analysis and field exploration, through semi-structured interviews. The results point to incipient front practices to a well-structured policy, the social representations of groups influence their deployment. Low public participation, especially the elderly and the distancing of the research to the practice of these municipalities are short of expectations of policy. The field of science, technology and society contributes to reflections on the appropriation of science and technology, it was observed that in his views prevail the harsh health technologies. The intention of this thesis was to analyze the policy and practices in the current scenario, with no claim assessment, was intended to bring reflections on this policy by the look of social representations and studies of science, technology and society We can conclude that the regions implement the National Policy differently, with demands and perceptions appropriate to their reality, even if they receive national and international guidelines. / O envelhecimento e a consequente mudança demográfica brasileira nos apontam para uma profunda transformação socioeconômica do país. A estimativa é que a população idosa triplique nas próximas décadas, passando de 20 milhões em 2010 para 65 milhões em 2050. Muitos atores são responsáveis pela implementação da Política Nacional de Saúde da Pessoa Idosa, entre eles estão os gestores municipais de saúde e os articuladores de saúde do idoso. O objetivo deste estudo é analisar as representações sociais da Política Nacional da Saúde da Pessoa Idosa em um Departamento Regional de Saúde, à luz do campo da Ciência, Tecnologia e Sociedade. Trata-se de uma pesquisa qualitativa, de natureza exploratória e descritiva. O percurso metodológico foi o de análise documental e exploração de campo, através de entrevistas semiestruturadas. Os resultados apontam para práticas incipientes frente a uma política bem estruturada, as representações sociais dos grupos influenciam na sua implantação. A baixa participação pública, em especial do idoso e o distanciamento da pesquisa com a prática destes municípios estão aquém das expectativas da política. O campo da ciência, tecnologia e sociedade contribui com reflexões sobre a apropriação da ciência e tecnologia, observou-se que em suas concepções prevalecem às tecnologias de saúde duras. A intencionalidade desta tese foi trazer reflexões sobre esta política pelo olhar das representações sociais e estudos da ciência, tecnologia e sociedade. Pode-se concluir que as regiões implementam de forma diferente a Política Nacional, com demandas e percepções adequadas a sua realidade, mesmo que estes recebam diretrizes nacionais e internacionais.

A Mapping of Historical Discourses in STEM Advocacy Literature

abstract: Efforts to privilege STEM (Science, Technology, Engineering, and Mathematics) disciplines, initiatives, and industries in American discourse are arguably the foremost expressions of scientific authority in contemporary educational policy. Citing a diverse body of STEM literature, I discuss the histories and rationales that sustain the promotion of STEM. In doing so, I appropriate two concepts -Michel Foucault's Regime of Truth and Hayden White's Emplotment- for the purpose of analyzing the complex interests embodied by STEM discourse. I argue that the Sputnik Narrative is the prevailing story in STEM advocacy discourse. I claim that STEM advocates typically emplot this history as a Romance. Furthermore, I classify two major bases of appeal (rationales) that appear within this literature to justify STEM projects and proposals, "competition" and "equity." Throughout my writing, I cite discursive strategies for challenging and reimagining STEM history. Differences between objective and subjective measurements ofphysical activity with regards to gender and age in obese children

Background: Overweight and obesity is increasing worldwide and more and more peopleare dying due to their overweight rather than of starvation. Obesity in young children isbecoming more common and the physical activity is decreasing with age. To measurechildren’s daily physical activity sveral methods can be used, such as Actical activity monitorand activity diary.Purpose: The aim of this study was to investigate if there is a difference between the twomethods mentioned above, current physical activity, age and gender in obese children and toinvestigate how many of these children that reached the recommendations of daily activity.Materials and Methods: All included were subjected to the two activity methods,collection of data from anthropometric measurements and body composition measurements.In total 122 children between the ages 10 to 17 years, 38 girls and 67 boys where included inthis study.Results: None of the children reached the recommended daily activity. Correlations betweenobjective and subjective methods showed a stronger association with the activity monitor thanwith the activity diary. The study showed that girls had a higher fat mass (FM) than boys,while boys had higher fat free mass (FFM).Conclusion: This study showed that physical activity decline with age and boys had higherdaily physical activity. Boys had more tendencies to improve their fat free mass. Girls hadhigher fat mass and lower fat free mass. Which method that gives the most adequate resultsrequires a larger population and more studies of the topic.

Evaluation of Correlation between Platelet Function in Platelet apheresis Donors and Function in Thrombapharesis Concentrates Measured with Impedance Aggregometry (Multiplate® Analyzer)

Transfusion of platelet concentrates (PC) can be necessary for patients to maintain coagulability. It is vital that the platelets maintain viability and function during processing and storage to obtain enhanced coagulability in the transfused patient. Today, no test is used to verify platelet function in either donors or in PC’s. Observing swirling effect is the only test applied to control platelets before transfusion but the method is based on platelet morphology and does not directly evaluate platelet function.Impedance aggregometry (IA) (Multiplate analyzer, Roche Diagnostic) is a promising method for measuring platelet function, measuring changes in impedance over time when platelets adhere to electrodes. IA has been well evaluated for the purpose of analyzing whole blood but analyzing PC’s is a relatively new application of the method.Samples from platelet donors and PC’s were analyzed with IA to evaluate correlation in function between donors and PC’s, in the hope of being able to predict function in PC. Different platelet concentrations were also analyzed to evaluate the impact of varying concentration on impedance. Adenosine diphosphate (ADP), collagen and thrombin receptor activating peptide 6 (TRAP-6) were used to induce aggregation. Platelet function was measured in PC’s on day 1 and 4 after donation.A significant correlation was observed between platelet function in donors and in PCs on day 1, measured with ADP. An important finding was also that platelet concentration does affect impedance, in collagen-induced aggregation more than ADP-induced. It is therefore possible that a correlation would also have been found between donors and PC’s analyzed with collagen if the platelet concentration would have been standardized.

Detection of Thymidine Kinase 1 Activity in Whole Blood Using an Oligonucleotide System

In today’s medical science studies, many tumor markers are being used to monitor cancer cell proliferation, but the number of assays for analysis of these markers are few. The aim of this study was to find an easier and more time-efficient way to measure the activity of a specific tumor marker called tymidine kinase 1 (TK1). This tumor marker is an important enzyme involved in cell proliferation and is a key enzyme in the salvage pathway. TK1 activity is related to the occurrence of hematological malignancies and cell activity and therefore have been used as a marker when monitoring this group of patients in treatment. Measurement of the enzyme activity in this study was performed by using an oligonucleotide assay. Detection of the enzyme activity in whole blood and in plasma has not previously been shown. The TK1 activity measured in whole blood and plasma correlated with TK1 activity measured in serum (R2=0,8651 and R2 =0,9845, respectively). It was found that it is possible to determine the TK1 activity in whole blood but only if the activity was measured on the same day as the blood samples were taken. The results shows that the activity measurement of TK1 in plasma and whole blood can be used as a marker to verify patients' therapy in cancer care. This study is only the beginning and further investigations should be made in the future to determine if the method that is subject to this study has the requested effects.

The influence of a blood donors sitting position during time of waiting on the change of haemoglobin concentration during blood donation.

The routines for blood testing were changed during 2010 at the blood bank in UAS. At first, the blood test was taken before the donation and now it is taken after donation. Along with this, the blood bank increased the lowest level for allowance of blood donation with 10 g/L both for men and women. The level is now on 125 g/L and 135 g/L for women respectively men. After the increase, it was noticed that the amount of blood donors were deferred due to low Hb levels in creased. A study made in year 2013, investigated how much the Hb-levels actually was changed during a blood donation. It showed that it was lowered in means by 6 g/L and not 10 g/L as previously thought.The aim of this study was to see if the sitting position of the blood donor during waiting time and the supine position during the time of blood donation may had any effect on the difference of the Hb-level during the blood donation.Data from the 120 blood donors in the earlier study was collected. Hb values, before and after blood donation, were taken from the earlier study and registered times were taken from the database Prosang. The waiting time, time of blood donation and the difference of Hb-levels were calculated and correlated with Spearmanns correlation coefficient.The results did not show any correlation between the times and the difference in Hb-levels. One of the reasons may be that the blood donor physiology differ and thus the change in Hb-level can vary.

Essa pesquisa estuda o desenvolvimento de novas tecnologias de participação sociocultural da população atendida em Terapia Ocupacional. O foco da pesquisa esteve no processo de acompanhar os sujeitos e grupos vinculados ao Programa Permanente Composições Artísticas e Terapia Ocupacional (PACTO), projeto didático-assistencial do Laboratório de Estudos e Pesquisa Arte e Corpo em Terapia Ocupacional ligado ao Departamento de Fonoaudiologia, Fisioterapia e Terapia Ocupacional da Faculdade de Medicina da Universidade de São Paulo. Num território híbrido de encontro entre a Terapia Ocupacional e os campos da Cultura, da Arte e da Cidadania, as questões de pesquisa foram, principalmente: como auxiliar e agenciar o trânsito e a circulação das pessoas e de suas produções realizadas em terapia ocupacional, com os espaços de produção artística e com os espaços de cultura, no contexto sociocultural da cidade de São Paulo na atualidade? Que tecnologias socioculturais favorecem essa participação? Amparados pela perspectiva do pensamento complexo e pelo viés cultural, buscou-se trabalhar num eixo transversal de análise, que parte das experiências artísticoculturais contemporâneas, vinculadas às produções éticas, estéticas e políticas na organização da vida. A pesquisaintervenção resultou na configuração de um estudo de caso, com uso dos procedimentos metodológicos da pesquisa-ação, e coleta de dados no período entre 2007 e 2008. Constatouse, a partir da análise qualitativa dos dados, a relevância de temas, tratados nas seguintes categorias: o tempo e a imaterialidade do trabalho; as relações de afeto, amizade e vínculo que estruturam redes de trocas e de criação; a diferenciação entre a produção de sujeitos e valores e a geração de renda; além da constatação da importância do terapeuta ocupacional como articuladores das redes sociais, para a efetivação do trabalho de criação e circulação das produções artísticas dos sujeitos e grupo estudados, operando na sustentação da artesania dos encontros. Como desdobramentos da pesquisa realizada, vislumbrou-se o surgimento de novas territorialidades no universo cultural da cidade, com ações voltadas para a invenção de mundos, onde as relações se pautam na cooperação, na convivência, no exercício da diferença e nos afetos. Instaurou-se a abertura de fendas no espaço-tempo contemporâneo, engendrando novas configurações da vida coletiva, que sustentem a existência de forma criativa e pulsante, e que possam ser multiplicadas. / This research studies the development of new technologies of socio-cultural participation of the population assisted in Occupational Therapy. The focus of the research was on the process of following up the subjects and groups bound to the Permanent Artistic Compositions and Occupational Therapy Program (PACTO), a didactic-assistance project of the Art ad Body Laboratory of Studies and Research in Occupational Therapy linked to the Department of Speech Therapy, Physiotherapy and Occupational Therapy of the Medical School of the University of São Paulo. Within a hybrid meeting territory between Occupational Therapy and the fields of Culture, Art and Citizenship, the matters researched were mainly: how to help and enable the transit and circulation of people and their productions in occupational therapy in the spaces of artistic production and with the cultural spaces within the current socio-cultural context of the city of São Paulo? Which socio-cultural technologies favor such participation? Supported by the perspective of the complex thinking and under a cultural approach, we tried to work in a cross-sectional axel of analysis which starts in the contemporary artistic-cultural experiences linked to ethical, aesthetical and political production in the organization of life. The research-intervention resulted in the configuration of a case study by using research-action methodological procedures and data collection from 2007 to 2008. We verified, based on the qualitative analysis of the data, the relevance of the themes dealt with under the following categories: the time and immateriality of the work; affective relations, friendship and the bond structuring exchange and creation networks; the difference between the production of subjects and values and income generation; in addition to verifying the importance of occupational therapists as articulators of social networks to enable creative work and circulation of artistic productions of the subjects and group studied, operating to support citizenship in meetings. As the research unfolded, we could see that new territorialities in the cultural universe of the city came up with actions oriented towards inventing worlds where relations were based on cooperation, living together, exercising differences and affections. Cracks in the contemporary space-time were opened engendering a new configuration of collective life which sustains the existence in a creative and pulsing way and which can be multiplied.

Optimization of PCR protocols used for genotyping transgenic mice & Evaluation of a method for co-detecting mRNA and protein / Optimering av PCR-protokoll som används för genotypning av transgena möss och utvärdering av en metod för att detektera mRNA och protein

The aim of the current study was divided into two separate goals, (i) optimization of a number of PCR-based protocols employed for genotyping transgenic mouse lines and (ii) evaluating a protocol for co-detection of mRNA and its correlated protein in the mouse midbrain. The optimization was performed on PCR protocols for genotyping the following transgenic mouse lines; Dat-Cre, Vglut2-Lox, Vglut2-Cre and Vmat2-Lox. Also, two different polymerases were evaluated parallel to each other – KAPA and Maxima Hot Start. One of the main findings from the PCR optimizations were that for the Vglut2-Lox protocol. By decreasing the annealing temp and increasing the MgCl2 the bands appeared brighter.  For the second part of the project, in-situ hybridization (ISH) was used to detect the mRNA expression with a `non-radioactive in situ hybridization´ protocol, using digoxigenin or fluorescein labelled riboprobes (mRNA probes). To detect the correlated protein a basic immunohistochemistry (IHC) protocol with the use of primary and secondary antibodies was implemented. The combined protocol was tested with Nd6 and Grp markers. Before testing to combined the protocols the ISH protocol was performed alone with riboprobes for Girk2, Lpl and Fst. The combined protocol detected mRNA and protein for both the control marker Th and the Nd6 marker. In conclusions, the optimized PCR protocols were optimal when used with the Maxima Hot Start polymerase and the new combined ISH and IHC protocol worked for markers Th and Nd6.
