O objetivo desse trabalho foi o de avaliar a atividade antimicrobiana utilizando-se óleos essenciais de Cymbopogon citratus (D.C.) Stapf, Curcuma zedoaria Roscoe e Tagetes minuta L. sobre cepas de Staphylococcus spp., Streptococcus mutans e Candida spp. em crescimento planctônico e em biofilme. Para estudo dos microrganismos em crescimento planctônico, foram determinadas a Concentração Inibitória Mínima e a Concentração Fungicida Mínima de nove cepas clínicas e uma cepa padrão para cada espécie: C. albicans, C. tropicalis, C. glabrata, S. aureus, S. epidermidis e S. mutans. Para avaliação dos efeitos dos óleos essenciais em biofilme, foram utilizadas cepas padrão de Candida albicans (ATCC 18804), Staphylococcus aureus (ATCC 6538) e Streptococcus mutans (ATCC 35688) isolados e em associações em corpos-de-prova durante cinco dias. A seguir, os corpos-de-prova em resina acrílica foram lavados e colocados em contato com óleos essenciais durante 5 min. O número de unidades formadoras de colônias obtidas em cada biofilme (UFC/mL) foram submetidos à análise estatística descritiva e teste t-Student utilizando-se o software Minitab considerando-se o nível de significância p ≤ 0,05. Também foi realizada microscopia eletrônica de varredura (MEV) nos corpos-de-prova em resina acrílica com biofilme com tratamento e controle. Foram observadas reduções significativas do número de unidades formadoras de colônias (UFC/mL) tanto no biofilme como em associação. As maiores reduções ocorreram no tratamento do óleo essencial de C. citratus, seguidas pelo óleo de T. minuta e C. zedoaria. Os óleos essenciais de Cymbopogon citratus, Tagetes minuta e Curcuma zedoaria apresentaram... / In the search for preventive and curative means for periodontal disease and tooth decay, herbal-purpose fungicide, antibacterial and anti-inflammatory have been investigated. The aim of this study was to evaluate the antimicrobial activity using the essential oils of Cymbopogon citratus (D.C.) Stapf, Curcuma zedoaria Roscoe and Tagetes minuta L. on strains of Staphylococcus spp., Streptococcus mutans and Candida spp. in planktonic and biofilm growth. For the study of microorganisms in planktonic growth, were determined Minimum Inhibitory Concentration and Minimum Microbicidal Concentration of nine clinical strains and one ATCC for each species: C. albicans, C. tropicalis, C. glabrata, S. aureus, S. epidermidis and S. mutans. To assess the effects of essential oils in biofilm, was used strains of Candida albicans (ATCC 18804), Staphylococcus aureus (ATCC 6538) and Streptococcus mutans (ATCC 35688) alone and in associations in acrylic resin discs immersed in sterile brain heart infusion broth (BHI) containing 5% sucrose, inoculated with microbial suspension and incubated for 5 days. On the fifth day, the discs were washed in sterile saline solution in order to remove loosely bound material. Next, the acrylic resin discs were washed and placed in contact with essential oils for 5 min. The number of CFU obtained in each biofilm (CFU/mL) were subjected to descriptive statistical analysis using Minitab software considering with significance level p ≤ 0.05. It was also performed scanning electron microscopy (SEM) acrylic resin disc with biofilm with treatment and control. It was identified significant reductions in the number of colony forming units (CFU/mL) in both planktonic and biofilm associated. The largest reductions occurred in the treatment of essential oil of C. citratus, followed by oil of T. minuta and C. zedoaria. We conclude that the essential oils of Cymbopogon citratus... A cárie dentária é uma doença bucal de alta prevalência e impactante em países em desenvolvimento. É causada pela presença de biofilme dentário rico em bactérias acidogênicas e acidúricas, como Streptococcus mutans e Lactobacillus casei. Neste sentido, a fitoterapia tem sido aplicada na odontologia devido ao seu conhecido efeito antimicrobiano, tendo potencial para prevenir doenças como a cárie dentária. Portanto, o presente estudo tem como objetivo testar o potencial antimicrobiano de extratos bruto e etanólico das folhas de Myracrodruon urundeuva (M. urundeuva.) e Qualea grandiflora (Q. grandiflora) sobre S.mutans e L. casei. Para tal, determinaram-se a Concentração Inibitória Mínima (CIM) e a Concentração Bactericida Mínima (CBM). A CIM foi definida como a menor concentração do agente antimicrobiano capaz de inibir 100% o crescimento microbiano (absorbância) em relação aos controles negativos. Para a CBM, alíquotas foram removidas dos poços que não apresentaram nenhuma absorbância (viabilidade, concentrações CIM) e semeadas em placas de ágar BHI, incubadas por 24 h a 37°C em estufa de CO2 5%. A CBM foi determinada considerando a menor concentração dos extratos capaz de impedir o crescimento bacteriano visível. Cepas de S. mutans (ATCC 21175) e L. casei (ATTC 334) foram ativadas em BHI e caldo Rogosa, respectivamente. A CIM foi determinada pela técnica de diluição em microplacas de 96 poços (100 l de extrato + 80 l BHI/Rogosa + 20 l da bactéria diluída em BHI/Rogosa equivalente a 5x105 UFC/mL), as quais foram incubadas por 24 h (S. mutans) e 48 h (L. casei) a 37°C em estufa de CO2 5%. Os extratos de M. urundeuva e Q. grandiflora inicialmente foram diluídos em BHI/Rogosa variando as concentrações entre 2 mg/ml a 0,00012207 mg/ml e os mesmos extratos diluídos em álcool foram avaliados nas concentrações entre 20 mg/ml a 0,00244 mg/ml para S. mutans e L. casei. Não foi possível determinar a CIM e a CBM para os extratos diluídos no BHI/Rogosa. Foram utilizados como controle positivo a clorexidina e como controles negativos BHI/Rogosa com e sem álcool a 5%. As CIMs (CBMs) da M. urundeuva e Q. grandiflora, diluídas em álcool, e clorexidina contra S. mutans foram 2,5 mg/ml (2,5 mg/ml), 5,0 mg/ml (--) e 0,00468 mg/ml (0,00937 mg/ml), respectivamente. Em relação ao L. casei, as CIMs (CBMs) da M. urundeuva e Q. grandiflora, diluídas em álcool, e da clorexidina foram 0,156 mg/ml (0,312 mg/ml), 0,156 0,625 mg/ml (0,312 0,625mg/ml) e 0,00468 mg/ml (0,3 mg/ml), respectivamente. Como conclusão, nosso estudo mostrou que L. casei (ATTC 334) é mais susceptível aos extratos que S. mutans (ATCC 21175) e o extrato M. urundeuva apresenta melhor efeito antimicrobiano que a Q. grandiflora em S. mutans (ATCC 21175), porém os dois extratos apresentam efeito similar sobre L. casei (ATTC 334) e ambos foram inferiores à CHX. / Dental caries is an oral disease of high prevalence and impact in developing countries. It is caused by the presence of a dental biofilm rich in acidogenic and aciduric bacteria, such as Streptococcus mutans and Lactobacillus casei. Accordingly, phytotherapy has been applied in dentistry due to its known antimicrobial effect, having potential to prevent diseases such as dental caries. Therefore, the present study aims to test the antimicrobial potential of crude and ethanolic extracts of Myracrodruon urundeuva (M. urundeuva) and Qualea grandiflora (Q. grandiflora) leaves on S. mutans and L. casei. For this, the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) were determined. MIC was defined as the lowest concentration of the antimicrobial agent capable of inhibiting 100% the microbial growth in comparison to the negative controls. For MBC, aliquots were removed from the wells that did not show any absorbance (viability, concentrations than MIC) and seeded on BHI agar plates, incubated for 24 h at 37°C and 5% CO2. The MBC was determined considering the lowest concentration of extracts capable of preventing visible bacterial growth. Strains of S. mutans (ATCC 21175) and L. casei (ATTC 334) were activated in BHI and Rogosa broth. MIC was determined by the dilution technique in 96-wells microplates (100 l of extract + 80 l BHI/ Rogosa + 20 l of bacterium diluted in BHI/Rogosa equivalent to 5x105 CFU/ml), which were incubated for 24 h (S. mutans) and 48 h (L. casei) at 37°C and 5% CO2. The extracts were firstly diluted in BHI/Rogosa varying the concentrations between 2 mg/mL and 0.00012207 mg/ml; the same extracts were also diluted in alcohol at concentrations ranging from 20 mg/ml to 0.00244 mg/ml and both tested against S. mutans and L. casei. It was not possible to determine the MIC and MBC for the extracts diluted in BHI/Rogosa. Chlorhexidine was used as positive control, while BHI/Rogosa with or without 5% alcohol were used as negative controls. The MICs (MBCs) of M. urundeuva and Q. grandiflora diluted in alcohol, and clorexidine against S. mutans were 2.5 mg/ml (2.5 mg/ml), 5.0 mg/ml (--) and 0.00468 mg/ml (0.00937 mg/ml), respectively. In respect to L. casei, the MICs (MBCs) of M. urundeuva and Q. grandiflora, diluted in alcohol, and chlorhexidine were 0.156 mg/ml (0.312 mg/ml), 0.156 0.625 mg/ml (0.312 0.625 mg/ml) and 0.00468 mg/ml (0.3 mg/ml), respectively. In conclusion, our study showed that L. casei (ATTC 334) is more susceptible than S. mutans (ATCC 21175) to the extracts and the extract of M. urundeuva has a better antimicrobial effect than Q. grandiflora against S. mutans (ATCC 21175), but both extracts have similar effect on L. casei (ATTC 334) and they were inferior to CHX.

Also had as objective to determine the sensitivity of C. neoformans variety neoformans and variety gattii, isolated of the líquor of patients with infectious meningite in the State of Sergipe, through the microdilution method (NCCLS) and to determine the sexual type - mating type of isolated physicians of Cryptococcus neoformans through the Reaction in Chain of DNA Polimerase (PCR) in Sergipe. Samples of the LCR had been used that had given entered in the IPH/LACEN/SE in the period of 2001 the 2004. Direct microscopy through the coloration with Nanquim Ink was carried through. An aliquot one of the líquor (0,1mL) was transferred to pipes contends Agar Sabouraud with clorafenicol and incubado in ambient temperature 25ºC and 37ºC. Tests for presumptive, fenotípica identification had been carried through in half Níger and of the varieties in half CGB. In the analyzed period, only one sample presented fenotípicas characteristics in half Níger differentiated of the others. Of the total of 15 samples analyzed in half CGB, 02 samples (13,33%) if had presented positive, or either, had been identified as variety gattii, being one of them isolated one of soronegativo patient for HIV (patient not aidético). The others 13 samples (86,67%) had presented negative CGB, or either, they had been identified as variety neoformans. The majority of the isolated samples of Cryptococcus neoformans of the líquor of patients with infectious meningite is of the variety neoformans in Sergipe. For the accomplishment of the sensitivity test variety had been used two samples of C. neoformans var. gattii and 11 of C. neoformans listed variety neoformans above. For control of the tests standard ATCC 32608 was used. After that suspension of fúngicas cells. The following antifungal had been used: Anphotericin B, Fluconazol, Cetoconazol, Miconazol, Itraconazol, 5- Fluorocitosina and Terbinafrina. The fungal suspension was adjusted in a final concentration of 0,5x10³ for 2,5x10³ UFC in the half RPMI. The experiment was carried through in microtitulação plates. After the inoculation, the microdilution plates had been incubadas 37ºC and deal in 24, 48 and 72 hours. The end point of the CIM was determined in the readings of 48 and 72 hours. The isolated physicians of C. neoformans had presented high values of CIM for the antifungal agents tested, what he mainly symbolizes one high tax of resistance Anfotericina B and to the Fluconazol, given that he can be correlated with the clinical data. For analysis of mating type sexual of C. neoformans from the líquor of patients with infectious meningite in Sergipe during the period of 2001 and 2004 had been used 13 samples. The related laboratoriais procedures to molecular biology (extration of DNA and accomplishment of the PCR) had followed as Chatuverdi reference et al. (2000). All the samples of C.neoformans tested, as much that pertaining to the variety neoformans as gattii are matting type, considered of bigger virulence. / Cryptococcus neoformans é um fungo oportunista que desencadeia meningite tanto em pacientes imunocompetentes como em pacientes hígidos Este trabalho teve como objetivo estudar as variedades (neoformans e gattii) de Cryptococcus neoformans isolados no Estado de Sergipe a partir do LCR de pacientes com meningite infecciosa. Também teve como objetivo determinar a sensibilidade de C. neoformans variedade neoformans e variedade gattii, isolados do líquor de pacientes com meningite infecciosa no Estado de Sergipe, através do método de microdiluição (NCCLS) e determinar o mating type de isolados clínicos de Cryptococcus neoformans através da Reação em Cadeia da DNA Polimerase em Sergipe. Foram utilizadas amostras do LCR que deram entrada no IPH/LACEN/SE no período de 2001 a 2004. Foi realizada microscopia direta através da coloração com Tinta Nanquim. Uma alíquota do líquor foi transferida para tubos contendo Agar Sabouraud com clorafenicol e incubado em temperatura ambiente 25ºC e a 37ºC.Foram realizadas provas para identificação presuntiva, fenotípica em meio Níger e das variedades em meio CGB. No período analisado, apenas uma amostra apresentou características fenotípicas em meio Níger diferenciada das outras. Do total de 15 amostras analisadas em meio CGB, 02 amostras (13,33%) se apresentaram positiva, ou seja, foram identificadas como variedade gattii, sendo uma delas isolada de paciente soronegativo para HIV (paciente não aidético). As outras 13 amostras (86,67%) apresentaram CGB negativo, ou seja, foram identificadas como variedade neoformans. A maioria das amostras de Cryptococcus neoformans isolados do líquor de pacientes com meningite infecciosa são da variedade neoformans em Sergipe. Para a realização do teste de sensibilidade foram utilizadas duas amostras de C. neoformans variedade gattii e 11 de C. neoformans variedade neoformans listadas acima. Para controle dos testes foi utilizada a cepa padrão ATCC 32608. Em seguida foi realizada a preparação do inóculo. Foram utilizados os seguintes antifúngicos: Anfotericina B, Fluconazol, Cetoconazol, Miconazol, Itraconazol, 5- Fluorocitosina e Terbinafrina. A suspensão fúngica foi ajustada numa concentração final de 0,5x10³ para 2,5x10³ UFC no meio RPMI. O experimento foi realizado em placas de microtitulação. Após a inoculação, as placas de microdiluição foram incubadas a 37ºC e lida em 24, 48 e 72 horas. O ponto final da CIM foi determinado nas leituras de 48 e 72 horas. Os isolados clínicos de C. neoformans apresentaram valores altos de CIM para os agentes antifúngicos testados, o que simboliza uma alta taxa de resistência principalmente a Anfotericina B e ao Fluconazol, dado que pode ser correlacionado com os dados clínicos. Para análise do mating type sexual foram utilizadas 13 amostras de C. neoformans a partir do líquor de pacientes com meningite infecciosa em Sergipe durante o período de 2001 e 2004. Os procedimentos laboratoriais relacionados à biologia molecular seguiram como referência Chatuverdi et al. (2000). Todas as amostras de C. neoformans tanto aquelas pertencentes à variedade neoformans como gattii são matting type a, considerado de maior virulência.

O gênero Neoscytalidium está crescentemente sendo associado às infecções superficiais e profundas causadas em pacientes imunocomprometidos e imunocompetentes. O fungo filamentoso Neoscytalidium dimidiatum é um microrganismo saprofítico e fitopatogênico, encontrado no solo e vegetação de regiões de clima tropical e subtropical. N. dimidiatum var. hyalinum e N. dimidiatum estão envolvidos em infecções superficiais de pele e unha sendo que a espécie produtora de melanina, N. dimidiatum, está associado preferencialmente a infecções profundas sugerindo que a espécie variante não produtora de melanina, N. dimidiatum var. hyalinum, pode ser menos virulenta que a pigmentada. Pouco é descrito na literatura sobre as variedades em questão tanto em relação à sensibilidade a drogas antifúngicas e ao tratamento fotodinâmico antimicrobiano (TFDA), bem como a características fisiológicas referentes à tolerância a variações de temperatura e pH. Assim, o presente trabalho teve como objetivo o estudo de isolados clínicos de N. dimidiatum e N. dimidiatum var. hyalinum obtidos de infecções de pele e unha. Neste estudo foram realizadas a identificação molecular, a filogenia multilocus, a verificação in vitro do desenvolvimento em diferentes temperaturas e pHs e a caracterização in vitro do perfil de susceptibilidade a antifúngicos e TFDA com fotossensibilizadores (FSs) fenotiazínicos, bem como os efeitos do TFDA nas biomoléculas dos artroconídios das variedades estudadas. A filogenia multilocus foi realizada pelo sequenciamento de sete diferentes loci onde foi evidenciado polimorfismo na região Internal Transcribed Spacer (ITS) 1 e nos genes ?-Tubulina (TUB), Fator de Alongamento de cadeia 1? (EF1) e Histona H3 (HH3). O agrupamento dos isolados clínicos deste estudo em genótipos distintos permitiu a separação em 6 tipos de sequência (TSs), sendo que o TS5 foi composto apenas pela espécie variante N. dimidiatum var. hyalinum. A análise de desenvolvimento em diferentes temperaturas e pHs demonstrou um tamanho de colônia reduzido para todos os isolados de N. dimidiatum var. hyalinum. Anfotericina B, voriconazol e terbinafina foram os antifúngicos mais eficientes para ambas variedades. Os valores das concentrações inibitórias mínimas (CIMs) encontrados para os derivados azólicos foram baixos para todos os isolados clínicos de N. dimidiatum var. hyalinum. Os isolados clínicos de N. dimidiatum mostraram ser menos sensível ao TFDA com os FSs azul de metileno (MB), novo azul de metileno N (NMBN), azul de toluidina O (TBO) e novo derivado sintético (S137) quando comparado à variedade hialina. NMBN e S137 mostraram maior eficiência para inativação de Neoscytalidium spp. Adicionalmente, no TFDA todos os FSs apresentaram efeito de permeabilidade de membrana plasmática, embora somente NMBN e S137 apresentaram a produção de Malondialdeido (MDA), isto é, causaram a peroxidação lipídica nos artroconídios de N. dimidiatum e da variedade hialina / Neoscytalidium sp. is increasingly being associated with superficial and deep infections in immunocompromised and immunocompetent patients. The filamentous fungus Neoscytalidium dimidiatum is a saprophytic and plant pathogenic microorganism that is found in soil and vegetation of tropical and subtropical regions. N. dimidiatum var. hyalinum and N. dimidiatum are involved in superficial skin and nail infections. The melanin producer N. dimidiatum is preferably associated with deep infections suggesting that the clinical isolate without melanin, N. dimidiatum var. hyalinum, may be less virulent than the pigmented variety. Little is described in the literature regarding the varieties N. dimidiatum and N. dimidiatum var. hyalinum about the sensitivity to antifungal drugs and photodynamic antimicrobial chemotherapy (PACT), and the physiological characteristics related to the growth at different temperature and pH. Therefore, the present study aimed to investigate clinical isolates of N. dimidiatum and N. dimidiatum var. hyalinum obtained from skin and nail infections. Here in this study were performed the molecular identification, multilocus phylogeny, the in vitro characterization of development at different temperatures and pHs, and the characterization of in vitro susceptibility to antifungal agents and PACT with phenotiazinium photosensitizers (PSs), as well the PACT effects on the biomolecules of the Neoscytalidium spp. arthroconidia. The multilocus phylogeny was performed by sequencing seven different loci where polymorphism were identified in the Internal Transcribed Spacer (ITS) 1 region of rDNA and in the genes ?-tubulin (TUB), elongation factor 1? (EF1) and Histone H3 (HH3). The grouping of the clinical isolates from this study in different genotypes allowed the clustering in 6 sequence types (ST), in which the ST5 was composed exclusively by all N dimidiatum var. hyalinum isolates from this study. The analysis of development at different temperatures and pHs showed a reduced colony size for all N. dimidiatum var. hyalinum isolates. Amphotericin B, voriconazole and terbinafine were the most effective antifungal for both varieties. The minimal inhibitory concentrations (MICs) values found for the azoles derivatives were low for all N. dimidiatum var. hyalinum isolates. N. dimidiatum clinical isolates have shown to be less sensitive to PACT with the PS methylene blue (MB), new methylene blue (NMBN), toluidine blue O (TBO) and new synthetic derivative (S137) when compared to hyaline variety. NMBN and S137 have shown more effectiveness for the inactivation of Neoscytalidium spp. Additionally, all PS in PACT have caused plasma membrane permeability, although only NMBN and S137 showed the production of malondialdehyde (MDA), i.e., caused lipid peroxidation in both N. dimidiatum and hyaline variety

Avec le développement rapide des modèles mathématiques et des outils de simulation, le besoin des processus de quantification des incertitudes a été bien augmenté. L'incertitude paramétrique et la groupe des nombreux décisions sont aujourd’hui les deux barrières principales dans la résolution des grandes problèmes systématiques.Capable de proportionner l'incertitude de la sortie sur celle des entrées, l’Analyse de Sensibilité Globale (GSA) est une solution fiable pour la quantification de l’incertitude. Parmi plusieurs algorithmes de GSA, Fourier Amplitude Sensitivity Analysis (FAST) est l’un des choix les plus populaires des chercheurs. Basé sur ANOVA-HDMR (ANalysis Of VAriance - High Dimensional Model Representation), il est solide en mathématique est efficace en calcul.Malheureusement, la décomposition unique d’ANOVA-HDMR se dépend sur l’indépendance des entrées. À cause de cela, il y a pas mal de cas industriels qui ne peut pas se traiter par FAST, particulièrement pour ceux qui donnent uniquement les échantillons mais sans lois de distribution. Sous cette demande, deux méthode extensifs de FAST avec design de corrélation sont proposées et étudiées dans la recherche. Parmi les deux méthodes, FAST-c s’est basé sur les distributions et FAST-orig s’est basé sur les échantillons.Comme applications et validations, multiples problèmes vibroacoustiques se sont traités dans la recherche. Les matériaux acoustiques avec soustructures, sont des candidats parfaits pour tester FAST-c et FAST-orig. Deux application sont présentées dans la première partie de la thèse, après l’état de l’arts. Les modèles choisis sont matérial poroélastique et structures composite sandwich, dont les propriétés mécaniques sont tous fortement influencées par les paramètres géométriques microscopique ou mesoscopique. D’avoir la méthode de FAST originale comparée avec les deux nouvelles, on trouve bien plus d’information sur la performance vibroacoustique de ces matériaux.Déjà répondu à la demande de GSA sur les modèles avecs les variables dépendantes, la deuxième partie de la thèse contient plus de recherches reliées avec FAST. D’abord FAST est pris en comparaison avec Random Forest, une algorithme bien connu de data-mining. Leurs erreurs potentiels et la possibilité de fonctioner ensemble sont discutés. Et dans les chapitres suivies, plus d’application de FAST sont présentées. Les méthodes sont appliquées sous plusieurs différente conditions. Une modèle de structure périodique qui contient des corrélation parmi les unités nous a en plus forcé à développer une nouvelle FAST-pe méthode. Dans ces applications, les designs des processus préliminaires et les stratégies d’échantillonages sont des essences à présenter. / With rapid development of mathematical models and simulation tools, the need of uncertainty quantification process has grown higher than ever before. Parametric uncertainties and overall decision stacks are nowadays the two main barriers in solving large scale systematic problem.Global Sensitivity Analysis (GSA) is one reliable solution for uncertainty quantification which is capable to assess the uncertainty of model output on its inputs’. Among several GSA algorithms, Fourier Amplitude Sensitivity Test (FAST) is one of the most popular choices of researchers. Based on ANOVA-HDMR (ANalysis Of VAriance - High Dimensional Model Representation), it is both mathematically solid and computationally efficient.One unfortunate fact is that the uniqueness of ANOVA-HDMR relies on the independency of input variables. It makes FAST unable to treat many industrial cases especially for those with only datasets but not distribution functions to be found. To answer the needs, two extended FAST methods with correlation design are proposed and further studied in this research. Among them FAST-c is distribution-based and FAST-orig is data-based.As a frame of validation and application, a number of vibroacoustic problems are dealt with in this research. Vibroacoustic materials with substructures, are perfect test candidates for FAST-c and FAST-orig. Two application cases are presented in the first part of this thesis, following the literature review. The models chosen here are poroelastic material and sandwich composite structures, both having their mechanical properties hugely influenced by their microscopic and mesoscopic geometric parameters. Getting the original FAST method compared to the two with correlation design, many different features on materials’ vibroacoustic performance are latter discovered.Having got an answer for GSA on models with dependent variables, the second part of this thesis contains more extended researches related to FAST. It is taken into comparison with Random Forest, a well-known data-mining algorithm. The potential error of both algorithms are analyzed and the possibility of joint application is discussed. In the following chapters, more applications of FAST-series methods are reported. They are applied under various conditions where another improved version named FAST-pe is developed to treat a model of periodic structures with correlation among each units. Upon these FAST application cases, the design of preliminary process and the sampling strategies is the core part to be introduced.

Cette thèse aide à approfondir les connaissances sur les différentes sources d’incertitude dans l’évaluation économique du risque inondation. Elle explore plusieurs disciplines afin d’analyser l’impact des stratégies utilisées pour modéliser l’aléa inondation et la vulnérabilité d’un territoire, sur l’évaluation des dommages potentiels. On a mesuré la variabilité des estimations en fonction des bases de données, modèles, méthodes et échelles considérés pour : analyser la probabilité des inondations (hydrologie) ; modéliser et cartographier l’aléa inondation (hydraulique) ; caractériser la vulnérabilité des enjeux et leur susceptibilité à subir des dommages (génie civil, géographie et économie de l’environnement). Les analyses et méthodes développées devront appuyer la prise en compte d’incertitudes, la détermination de priorités et l’optimisation de la distribution des ressources entre les différents modules de l’évaluation. Afin d’explorer un degré de complexité supplémentaire d’analyse de risque, nous avons développé une méthode d’analyse de la vulnérabilité systémique des réseaux d’infrastructure en lien avec leur résilience. / This thesis aims at exploring different sources of uncertainty related to the economic analysis of the flood risk. It embraces several fields of knowledge in order to determine how the selection of strategies used to model flood hazard and assess the vulnerability of a territory may affect damage potential estimations. We measured the variability of damage estimations as a function of the datasets, methods, models and scales considered to: analyse the probability of floods (hydrology); model and map flood hazard (hydraulics); assess the vulnerability and susceptibility of properties to floods (civil engineering, geography and environmental economics). The methods and analyses developed here should bring support for practitioners in the investigation of uncertainties, determination of evaluation priorities and optimisation of the distribution of resources between the different modules of the evaluation process. In order to explore a second level of complexity of flood risk evaluations, we developed a method for analysing the systemic vulnerability of infrastructure networks, in relation with their resilience.

This thesis deals with a load testing of web applications. The text is introduced with a definition of the load testing, reasons for its execution and a description of its types. Recommended procedures are provided for preparation, execution and evaluation of the testing. Several commercial and open-source load testing tools are described. Cloud testing -- the contemporary phenomenon -- is also included. The entire process of load testing depicted in this text is demonstrated on a selected application. A following chapter briefly deals with a profiling of web applications. It, again, covers its definition and its purpose, accompanied with a practical example. The aim of this thesis is to introduce the reader to the world of load testing, which is primarily accomplished by describing an example, which provides an in-depth explanation for every step necessary in order to perform the web application testing successfully.
