THE EVOLUTION OF FITNESS AFTER PROLONGED SPERM STORAGE

A series of recent studies using Drosophila melanogaster suggest that while males may benefit from having access to many partners, female fitness is reduced by extended cohabitation and sexual interaction with males. Yet, even if repeated sexual interactions are harmful to females, limited male exposure will ultimately be detrimental due to sperm-depletion and infertility. Females are therefore expected to balance mating opportunities and sperm storage capacity to maximize lifetime reproductive success. I introduced extended mating deprivation as a selective pressure to experimentally evolve lines of D. melanogaster for characters related to mating and postcopulatory sexual selection. Evolution of the mate-deprived lines over several dozen generations demonstrated that restricted sexual access was indeed a potent selective pressure. I consistently found that when males were removed for an extended time period, female fitness declined substantially, suggesting that mate-deprivation over nine days was harmful. Under these conditions, selected-line males responded to mate-reduced conditions and demonstrated a 13% increase in reproductive success compared to controls. Experimental females had a 15% increase in fertility compared to controls. I investigated a series of developmental characteristics that may have been altered by the selection regime, and while there was some evidence of evolved change, these results were not consistent. Although the data at hand do not substantiate a detailed characterization, both sexes in the experimental populations demonstrated increased fitness after extended mate-deprivation, thus evolutionary change appears to have occurred via selection on one or both relevant male ejaculate characteristics: sperm number and survival, and factors affecting female late-life fertility. / Thesis (Master, Biology) -- Queen's University, 2008-04-28 23:05:42.835

Recentes estudos do gênero Micrurus, o qual é popularmente conhecido como o das “cobras-corais verdadeiras”, dividem este táxon em dois grupos morfologicamente distintos, o grupo das cobras corais com anéis negros em mônades (BRM) e o grupo das cobras corais com anéis negros em tríades (BRT) e, com base na morfologia macroscópica, supõe-se que esses grupos possuam estratégias reprodutivas diferenciadas. Assim, este estudo analisa e descreve a biologia reprodutiva de machos e fêmeas da espécie de serpente de tríades Micrurus lemniscatus de algumas regiões do Brasil. Micrurus lemniscatus é um complexo, com evidências polifiléticas, sendo composto por quatro subespécies: Micrurus l. carvalhoi, Micrurus l. helleri, Micrurus l. diutius e M. l. lemniscatus que habitam Mata atlântica, Cerrado e Floresta Amazônica brasileiros. Para isto foram utilizados animais preservados em coleções herpetológicas, dos quais obteveram-se dados biométricos e merísticos. Além disso, estes espécimes foram dissecados e suas gônadas medidas e coletadas para análises morfológicas e histológicas. Outras características, relacionadas à biologia reprodutiva, como presença de dimorfismo sexual, morfologia macro e microscópica, diferenciação das gônadas durante as estações e estocagem de espermatozoides também foram estudadas. Reproduction and immunity in ant queens: Reproduction et immunité chez les reines de fourmis

Ants and other social Hymenoptera (social bees and wasps) have a remarkable mating strategy. Social Hymenoptera live in societies where reproduction is monopolized by a fertile caste consisting of males and queens. On the other hand, the logistical tasks of the colony are carried out by a sterile female caste known as workers. Reproductive individuals mate during a single bout early in their life and will never engage in additional reproductive events later on. Males die soon after mating while queens store millions of sperm cells in a specialized organ, the spermatheca. Queens will use this sperm stock to fertilize eggs during the rest of their life that can last up to several decades. With a record of 28.5 years in the black garden ant, ant queens have the longest lifespan recorded to date among the social Hymenoptera. In my thesis, I addressed three aspects of ant queen reproduction. First, I tested the effect of mating on the expression of several genes involved namely in fecundity, longevity and immunity. I found that mating induces an up-regulation of the yolk precursor vitellogenin and of the antimicrobial peptide defensin. Second, I measured the intensity of different immune responses in male and queen genital organs in order to determine which immune pathways are activated to protect sperm. Antimicrobial peptide genes are expressed in the genital tract of both sexes and the queen spermatheca is capable of strongly inhibiting bacterial growth. The immune melanization response is, however, overall inactive in the organs tested probably because its unspecific mode of action and cytotoxic by-products are likely to damage sperm cells. Immunity thus seems to be closely regulated in organs that are in contact with sperm. Third, I determined if activation of the queen immune system had an impact on the survival of sperm stored in the spermatheca. There is no detectable effect in young newly mated queens whereas, in one year old queens, immune activation induces a significant reduction in sperm viability. Life stage thus seems to influence queen ability to preserve sperm viability in the event of an immune challenge. In addition, one year old queens have higher sperm viability than newly mated queens suggesting queens are able to displace dead sperm cells from their spermatheca. Finally, I relied on the well-established sequence of behaviors inherent to the early life of ant queens to try to uncover the largely unknown roles of inotocin, the insect ortholog of the vertebrate hormones oxytocin and vasopressin, in regulating insect behavior. I measured gene expression of the inotocin receptor and found that it is highly expressed during social and reproductive behaviors, which is consistent with previous results in vertebrates. Inotocin might thus also be involved in modulating these behaviors in insects, but further studies are needed to be able to fully understand this complex signaling system. Overall, I show that reproduction and immunity are closely linked in ant queens and that the latter provide promising models for investigating the roles of hormones in insects. / Doctorat en Sciences / info:eu-repo/semantics/nonPublished

Nas últimas décadas tem se dado maior atenção à reprodução de animais silvestres, visto que para se conhecer e preservar uma espécie se faz necessário o conhecimento da história natural aliado aos processos da fisiologia e morfologia reprodutivas. Ciclos reprodutivos podem ser sazonais ou assazonais e apresentar variações em ambos os padrões. Por sua natureza e diversidade de fatores associados (e.g., atividade, dieta, temperatura), as estratégias reprodutivas vêm sendo investigadas sob uma abordagem evolutiva. Estudos do comportamento reprodutivo e dimorfismo sexual também têm sido destacados nesse âmbito. No entanto, poucos trabalhos são realizados para se conhecer as estratégias reprodutivas em serpentes neotropicais. Na maioria das vezes tais trabalhos abordam apenas alguns aspectos do ciclo reprodutivo de serpentes, como época de nascimentos, cópula e gestação, podendo ainda ser descritos de forma divergente pelo fato de muitos destes estudos serem norteados por dados de serpentes em cativeiro. Estudos microscópicos e macroscópicos, delineados com animais recém-chegados da natureza e preservados em coleções científicas, pode fornecer uma descrição e uma análise mais efetiva do ciclo reprodutivo permitindo a integração destes fatores. Assim, este trabalho investigou diversos aspectos da biologia reprodutiva de fêmeas e machos de populações de Bothrops alternatus do Brasil. Além disso, foram analisadas e descritas as estratégias reprodutivas empregadas por ambos os sexos durante o ciclo reprodutivo anual. Para isso, os espécimes foram dissecados ventralmente retirando-se fragmentos das gônadas e vias genitais de fêmeas e machos, em diferentes épocas do ano, a fim de se identificar as principais fases do ciclo vitelogênico, espermatogênico e do segmento sexual renal. Igualmente, para determinar se há estocagem de esperma nas fêmeas e nos machos, será investigada o útero aglandular e região do infundíbulo, vias genitais e ductos deferentes respectivamente. Tais dados permitirão compor um padrão reprodutivo, para essa espécie, levando-se em conta processos morfofisiológicos em ambos os sexos. Também foi realizada uma comparação dos ciclos reprodutivos entre populações de Bothrops alternatus e análises de possíveis influências ambientais na duração dos eventos reprodutivos / Over the last decades a greater attention has been given to the reproduction of wild animals, as the knowledge of natural history combined with the processes of reproductive physiology and morphology is essential for the preservation of a species. Reproductive cycles may be seasonal or not, and both patterns may have variations. Due to its nature and to the diversity of the associated factors (e.g., activity, diet, temperature), reproductive strategies have been investigated in an evolutionary approach. Studies of reproductive behavior and sexual dimorphism have also been highlighted in this context. However, few studies have been conducted to determine reproductive strategies in neotropical snakes. Most studies address only some aspects of the reproductive cycle of snakes, such as time of birth, copulation and pregnancy, and may present divergent descriptions as many of them are based on data from snakes in captivity. Microscopic and macroscopic studies carried out with animals newcomers from nature and preserved in scientific collections can provide a more effective analysis of the reproductive cycle allowing the integration of these factors. In this context, this project aims to investigate various aspects of the reproductive biology of female and male populations of Bothrops alternatus from Brazil. The reproductive strategies employed by both sexes during the annual reproductive cycle was analyzed and described. The specimens was dissected ventrally and fragments of gonads and genital tracts of females and males was removed at different times of the year in order to identify the main phases of the vitellogenic cycle, spermatogenesis and sexual segment of the kidney. Also, to determine whether sperm storage is done by females and males, we will investigate the aglandular uterus, the posterior infundibulum, genital tracts and vas deferens respectively. Such data was compose a reproductive pattern for the species that will take into account the morphophysiologic processes of both sexes. It was also performed a comparison of reproductive cycles between populations of Bothrops alternates and analysis of possible environmental influences in the duration of reproductive events

Padrões de estocagem de esperma e variações cíclicas ovidutais em serpentes Xenodontinae / Sperm storage patterns and oviductal cyclical variation in Xenodontinae snakes

Os padrões de estocagem de esperma e as variações morfológicas cíclicas do trato reprodutivo feminino constituem táticas reprodutivas muito pouco estudadas em serpentes neotropicais, se comparadas às serpentes de regiões temperadas. Assim, o presente estudo teve por objetivo caracterizar e compreender as estratégias reprodutivas empregadas por fêmeas de quatro espécies de serpentes brasileiras da subfamília Xenodontinae. Amostras de gônadas e vias genitais foram coletadas em diferentes estações do ano e relacionadas aos estágios reprodutivos da fêmea. Os resultados indicaram que as serpentes estudadas possuem dois locais de estocagem de esperma distribuídos no infundíbulo posterior e na junção útero-vaginal. Liophis miliaris, Oxyrhopus guibei e Philodryas patagoniensis apresentaram estocagem por meio de glândulas túbulo-alveolares ramificadas e glândulas tubulares ciliadas no infundíbulo posterior. Das quatro espécies estudadas, apenas Tomodon dorsatus e P. patagoniensis apresentaram grupos de espermatozóides armazenados nos sulcos da junção útero-vaginal. No entanto, unicamente P. patagoniensis parece ter a capacidade de produzir várias ninhadas com esperma estocado proveniente dessa região. Em ambos os locais de estocagem as células secretoras aumentaram a produção de carboidratos neutros na presença de esperma, o que sugere um processo de nutrição. Em fêmeas vitelogênicas o aumento da atividade secretora do epitélio ovidutal com produção de vacúolos eletrodensos parece estar sobre controle hormonal. Durante o ciclo reprodutivo as glândulas uterinas de espécies ovíparas e vivíparas também mostraram um aumento da secreção em fêmeas vitelogênicas. No entanto, as glândulas uterinas das espécies ovíparas apresentaram uma maior atividade do retículo endoplasmático rugoso, o que indicou produção de proteínas para formação da membrana da casca. O ciclo reprodutivo e a época de acasalamento variaram entre as populações das diferentes espécies. Em L. miliaris, O. guibei e P. patagoniensis essas variações estiveram ligadas a mudanças na temperatura e pluviosidade, já T. dorsatus não apresentou variações no ciclo reprodutivo, provavelmente influenciada pela inércia filogenética. / Sperm storage patterns and cyclic morphological variations in the female reproductive tract are poorly studied reproductive tactics in Neotropical snakes if a comparison to snakes from temperate regions is established. Therefore, the aim of this study is to characterize and understand the female reproductive strategies of four species of Xenodontinae snakes from Brazil. Samples of the gonads and genital ducts were collected during the different seasons of the year and a correlation with female reproductive stages was established. Results show that these snakes have two sperm storage sites in the posterior infundibulum and uterovaginal junction. Liophis miliaris, Oxyrhopus guibei and Philodryas patagoniensis store sperm by means of branched tubuloalveolar glands and tubular ciliated glands in the posterior infundibulum. Only Tomodon dorsatus and P. patagoniensis showed sperm stored in the furrows of the uterovaginal junction. Anyway, only P. patagoniensis seems to be capable of producing multiple clutches using sperm stored in this portion. Secretory cells increased the production of neutral carbohydrates in the presence of sperm in both sperm storage sites, which suggests the existence of a nutrition process. In vitellogenic females, the increase of the secretory activity in the oviductal epithelium with production of eletrodense vacuoles seems to be under hormonal control. During the reproductive cycle, uterine glands of oviparous and viviparous species also showed an increase in secretion in vitellogenic females. However, uterine glands of oviparous species showed more intense activity of the rough endoplasmic reticulum, which indicates the production of proteins for the formation of the egg shell. Reproductive cycles and timing of mating varied between different populations. In L. miliaris, O. guibei and P. patagoniensis, these variations were linked to the changes in temperature and rainfall patterns, but T. dorsatus did not show any variation in the reproductive cycles between populations, probably influenced by phylogenetic inertia.

Estudos microscópicos e macroscópicos, delineados com animais recém-chegados da natureza e preservados em coleções científicas, pode fornecer uma descrição e uma análise mais efetiva do ciclo reprodutivo permitindo a integração destes fatores. Assim, este trabalho investigou diversos aspectos da biologia reprodutiva de fêmeas e machos de populações de Bothrops alternatus do Brasil. Além disso, foram analisadas e descritas as estratégias reprodutivas empregadas por ambos os sexos durante o ciclo reprodutivo anual. Para isso, os espécimes foram dissecados ventralmente retirando-se fragmentos das gônadas e vias genitais de fêmeas e machos, em diferentes épocas do ano, a fim de se identificar as principais fases do ciclo vitelogênico, espermatogênico e do segmento sexual renal. Igualmente, para determinar se há estocagem de esperma nas fêmeas e nos machos, será investigada o útero aglandular e região do infundíbulo, vias genitais e ductos deferentes respectivamente. Tais dados permitirão compor um padrão reprodutivo, para essa espécie, levando-se em conta processos morfofisiológicos em ambos os sexos. Também foi realizada uma comparação dos ciclos reprodutivos entre populações de Bothrops alternatus e análises de possíveis influências ambientais na duração dos eventos reprodutivos / Over the last decades a greater attention has been given to the reproduction of wild animals, as the knowledge of natural history combined with the processes of reproductive physiology and morphology is essential for the preservation of a species. Reproductive cycles may be seasonal or not, and both patterns may have variations. Due to its nature and to the diversity of the associated factors (e.g., activity, diet, temperature), reproductive strategies have been investigated in an evolutionary approach. Studies of reproductive behavior and sexual dimorphism have also been highlighted in this context. However, few studies have been conducted to determine reproductive strategies in neotropical snakes. Most studies address only some aspects of the reproductive cycle of snakes, such as time of birth, copulation and pregnancy, and may present divergent descriptions as many of them are based on data from snakes in captivity. Microscopic and macroscopic studies carried out with animals newcomers from nature and preserved in scientific collections can provide a more effective analysis of the reproductive cycle allowing the integration of these factors. In this context, this project aims to investigate various aspects of the reproductive biology of female and male populations of Bothrops alternatus from Brazil. The reproductive strategies employed by both sexes during the annual reproductive cycle was analyzed and described. The specimens was dissected ventrally and fragments of gonads and genital tracts of females and males was removed at different times of the year in order to identify the main phases of the vitellogenic cycle, spermatogenesis and sexual segment of the kidney. Also, to determine whether sperm storage is done by females and males, we will investigate the aglandular uterus, the posterior infundibulum, genital tracts and vas deferens respectively. Such data was compose a reproductive pattern for the species that will take into account the morphophysiologic processes of both sexes. It was also performed a comparison of reproductive cycles between populations of Bothrops alternates and analysis of possible environmental influences in the duration of reproductive events

Bothrops atrox é uma serpente de grande importância ecológica devido a sua ampla distribuição geográfica no vasto e diversificado habitat amazônico. Informações sobre a biologia reprodutiva de B. atrox são escassas e pontuais, sendo a maioria dos dados provenientes de serpentes mantidas em cativeiro. Dessa forma, este trabalho teve como objetivo descrever o ciclo reprodutivo de B. atrox, bem como as estratégias reprodutivas relacionadas a este ciclo tais como: maturidade e dimorfismo sexual, fecundidade, estocagem de espermatozoides e hipertrofia do segmento sexual renal nos machos. Além disso, o trabalho visou ainda relacionar tais estratégias com as condições ambientais (temperatura e precipitação). Para tanto, foram examinados 325 espécimes de B. atrox, sendo machos e fêmeas (maduros e imaturos), provenientes da Amazônia brasileira, preservados em nove coleções herpetológicas. Resultados mostraram haver diferenças relacionadas ao dimorfismo sexual: filhotes machos exibiram a coloração da ponta da cauda amarela, enquanto fêmeas apresentaram a ponta da cauda com coloração escura. Foi também observado que filhotes machos possuíram pigmentação escura na região gular e fêmeas apresentaram coloração clara. As fêmeas adultas foram significativamente maiores que os machos e atingiram a maturidade sexual com maiores tamanhos corpóreos. A vitelogênese foi sazonal (janeiro a agosto), entretanto, não houve sincronia entre a ocorrência de fêmeas prenhes e o período de nascimento dos filhotes. Assim, a extensão observada no ciclo reprodutivo das fêmeas foi possível devido à estocagem de espermatozoides, já que a cópula é sazonal. O ciclo reprodutivo dos machos foi descontínuo, sazonal semi-sincrônico. A produção de espermatozoides ocorreu ao longo do ano, no entanto, a espermiogênese foi observada principalmente no início da estação chuvosa (IEC) e a regressão testicular no início da estação seca (IES). Estocagem de espermatozoides nos ductos deferentes foi observada durante todos os meses do ano e o segmento sexual renal apresentou hipertrofia no IEC e final da estação chuvosa (FEC), sincronizado, portanto com a espermiogênese. Dessa forma, as condições ambientais (temperatura e pluviosidade) foram fundamentais na determinação do tipo de ciclo reprodutivo em B. atrox. Thus, this paper aimed to describe the reproductive cycle of B. atrox, as well as reproductive strategies related to this cycle as: maturity and sexual dimorphism, fecundity, sperm storage, and the hypertrophy of sexual segment of the kidney in males. Moreover, this work also aimed to relate such strategies with environmental conditions (temperature and precipitation). For that, it were examined 325 specimens of B. atrox, being males and females (mature and immature) from the Brazilian Amazon, preserved in nine herpetological collections. Results showed some differences related to sexual dimorphism: neonates males had the coloration of the tail tip yellow, whereas females showed the tail tip with dark coloration. It was also observed that neonate males had dark pigmentation in the throat region, whereas neonate females showed lighter coloration. Adult females were significantly larger than males, and attained sexual maturity at larger body sizes than males. Vitellogenesis was seasonal (January to August), however, there was no synchrony between the occurrence of pregnant females and birth periods. Thus, the extent observed in the female reproductive cycle was possible due to the sperm storage, since mating is seasonal. Male reproductive cycle was discontinuous, seasonal semi-synchronous. Sperm production occurred throughout the year, however, spermiogenesis was observed mainly in the beginning of the rainy season (BRS) and testicular regression in the beginning of the dry season (BRS). Sperm storage in the ductus deferens was observed during all months of the year and the sexual segment of the kidney presented hypertrophy in BRS and in the end of the rainy season (ERS), thus synchronized with spermiogenesis. Therefore, environmental conditions (temperature and rainfall) were essential in determining the type of reproductive cycle of B. atrox.
