The diploma thesis covers partial area of investment evaluation. It's main objective is to test the possible application of the real options theory on the Reverse Termination Fee valuation. This thesis considers the Reverse Termination Fee as a real option to abandon project and evaluates it using a binomial pricing model based on the volatility of the stock market. The result of this valuation is a price map. The map itself is analyzed together with the market analysis of announced trades using this fee as a deal protection. The conclusion of this thesis is formed based on observed trends and correlation between partial outputs of the binomial pricing model and market analysis.

Creation, termination and suspension of function of a member of a limited company body Abstract This thesis deals with the issues of creation, termination and suspension of function of a member of a limited company body and focuses on a relatively narrow aspect of the said topic related to the institute of codetermination in relation to the creation, termination and suspension of function of a member of a supervisory board of a joint stock company. Currently effective legislation contains significant gaps and it causes certain interpretation ambiguities and prevents the proper use of the institute of codetermination in practice. The present work is mainly aimed at identification of problem points and analysis of the most important controversial issues of the current legislation in the field of codetermination and aspires to their comprehensive solution. This diploma thesis deals withthe studyof electrochemical behaviorand detection of two structurally different neurotransmitters, dopamine (DA) and serotonin (5-HT), in solutions commonly used for neuron cultivation, namely Neurobasal (NB), NB with phenol red and in phosphate buffer (PB) of a pH close to the physiological value. An electrode based on boron-doped diamondwas used for the study, examiningtwo types of surfaces obtained after oxidation (O-BDD) and mechanical polishing (p-BDD). The results were obtained by two voltametric techniques,namely cyclic and differential pulse voltammetry.The studyrevealedthat DA oxidationis a quasi-reversible process,whereas 5-HT oxidizes irreversibly on O-BDD and p-BDD. Nevertheless, for both neurotransmitters their anodic oxidationonboth BDD surfaces is controlledby diffusion. Due to the passivation of the electrode surface by the oxidation products, anodic reactivation(Eact = +2400 mV, t = 30 s) was first testedto regenerate the O-BDD surface during DA and 5-HT measurements. There was no continuous decrease in DA peak currents on O-BDD and the measuredsignals were characterizedby high repeatabilityin all studiedmedia (sr (Ip) 1.1% in PB of pH 7.0, 1.7% in NB of pH 7.34, 0.9% in NB with phenol red of pH 7.48). Role of general regulatory factors in the control of gene expression and transcription fidelity / Rôle des facteurs de transcription dans le contrôle de l'expression des gènes et de la fidélité de la transcription

Ces dernières décennies ont été marquées par la découverte de la transcription dite « cachée » ou « pervasive ». Il a été en effet montré que la majeure partie du génome des eucaryotes est transcrite, donnant naissance à la formation de nombreux ARNs non-codants. La délimitation des unités de transcription apparait essentielle dans le contrôle de l’expression des gènes mais également dans le maintien de l’intégrité des processus associés à l’ADN en limitant notamment l’apparition de conflits avec la transcription. Dans ce contexte, l’initiation et la terminaison de la transcription représentent des étapes clés dans le partitionnement du génome et le métabolisme des ARNs. Nous avons montré que certains facteurs de transcription, appelés GRFs (General Regulatory Factors) chez la levure S. cerevisiae, jouent un rôle important dans le contrôle de la transcription pervasive à la fois au niveau de l’initiation mais également de la terminaison de la transcription et sont également requis pour assurer la fidélité de la transcription des gènes codant les ARN messagers. Nous avons prouvé que les GRFs liés au niveau des régions promotrices sont capables d’induire la terminaison de la transcription en bloquant physiquement la progression d’ARN polymérases issues de la translecture des terminateurs situés en amont. D’après nos études, cette voie de terminaison appelée « roadblock » est très répandue à l’échelle du génome et joue un rôle important dans la protection des promoteurs contre l’interférence transcriptionnelle. Nous avons également découvert que les GRFs limitent la transcription pervasive en obstruant les sites d’initiations ectopiques situés à proximité de leur site de fixation sur l’ADN. Ces facteurs sont aussi impliqués dans le contrôle de l’expression des gènes codants en favorisant l’utilisation de sites d’initiations les plus appropriés, c’est-à-dire, permettant la synthèse d’ARNs ayant un fort potentiel codant. Le rôle des GRFs dans le contrôle de l’initiation apparait intimement lié à leur capacité à correctement positionner les nucléosomes au niveau des promoteurs en collaboration avec les facteurs de remodelage de la chromatine. / The last decades have been marked by the discovery of pervasive transcription. Indeed, many studies have shown that transcription by RNA polymerase II is not restricted to annotated regions but is widespread in eukaryotic genomes, leading to the production of a plethora of non-coding RNAs. Precise delimitation of transcriptional units appears to be essential to ensure robust fidelity of gene expression and to maintain the integrity of DNA-associated events by preventing the occurrence of conflicts with transcription. In this respect, accurate transcription initiation and termination represent crucial mechanisms to partition the genome and define the correct processing of RNA molecules. Here, we show that yeast general regulatory factors (GRFs), a class of highly expressed transcription regulators, control pervasive transcription at the level of initiation and termination and are also involved in the fidelity of initiation of mRNA-coding genes. We demonstrate that GRFs bound at promoter regions can elicit transcription termination by physically impeding the progression of polymerases mainly deriving from readthrough transcription at upstream canonical termination sites. We provide evidence that this termination pathway named roadblock is widespread throughout the yeast genome and protects promoter regions from transcriptional interference. Furthermore, we establish that the presence of general regulatory factors also limits pervasive transcription at the level of initiation, notably by occluding spurious transcription start sites present in the vicinity of their binding sites. We also unveil the importance of these factors in promoting correct transcription start site selection at mRNA-coding genes thus favouring the synthesis of transcripts with an appropriate coding potential. Finally, we determine that the role of GRFs in controlling proper initiation is intimately linked to their ability to correctly position nucleosomes in promoters, a role that occurs independently from but in cooperation with chromatin remodelers.

Suppression traductionnelle des codons stop chez les mammifères / Translational suppression of stop codons in mammals

Entre 10% et 30% des maladies humaines sont liées à l'apparition d'une mutation non-sens (PTC). La synthèse protéique est alors arrêté prématurément. Cet arrêt peut être inhibé par des molécules inductrices de translecture qui permettent l’incorporation d’un ARNt suppresseur naturel au niveau du PTC (translecture). Le ribosome peut alors franchir le PTC et restaurer l’expression de la protéine.Au cours de ma thèse, je me suis intéressé à la suppression des codons stop en caractérisant de nouvelles molécules inductrices de translecture et en analysant les mécanismes de la fidélité de la traduction.J’ai tout d’abord mis au point un système de criblage innovant avec lequel j’ai testé plus de 17 000 molécules et identifié la molécule TLN468. J’ai pu mettre en évidence que cette molécule est capable d’induire la réexpression d’une protéine p53 active.J'ai aussi caractérisé de nouveaux composés dérivés d’aminoglycosides. J’ai pu montré que le NB124 est capable d’induire l’apoptose de cellules tumorales via la réexpression de la protéine p53 tout ayant une toxicité bien plus faible que la gentamicine.En parallèle, j’ai développé une approche en molécule unique permettant d’étudier les erreurs programmées du ribosome (recodage). J’ai ainsi pu analyser la cinétique d’élongation des ribosomes eucaryotes et montré que l’initiation de la traduction sur un site d’entrée interne (IRES) ralentit le ribosome lors des premiers cycles d’élongation. / Nonsense mutations, also known as premature termination codons (PTCs) are responsible for 10% to 30% of all human genetic diseases. Nonsense translation suppression can be induced by readthrough inducers. The presence of such PTC leads to premature translation termination. These stop therapeutic strategies have emerged which attempt to use molecules that facilitate tRNA incorporation at the PTC (readthrough). The, translation continue in the same reading frame until the next stop codon. I first developed an innovative screening system I used to test more than 17,000 molecules and have identified one hit, TLN468 molecule. I have shown that this molecule is able to induce re-expression of an active p53 protein.I also characterized new compounds derived from aminoglycosides. I have shown that the NB124 induces apoptosis of tumor cells by re-expressing p53 protein while having a much lower toxicity than gentamicin.I developed a single molecule approach for studying the ribosome programmed errors (recoding). I was able to analyze the kinetics of elongation eukaryotic ribosomes and showed that the initiation of translation at an internal entry site (IRES) slows the ribosome during the first elongation cycle.

This thesis is devoted to the study of electrochemical oxidation of meta-substituted phenolic substances on a boron-doped diamond electrode. In the first phase, it was studied the effect of pH on their electrochemical oxidation by the methods of classical "direct current" voltammetry (DCV) and differential pulse voltammetry (DPV). The study was performed on three types of boron doped diamond (BDD) surfaces: polished, anodic and cathodically activated. In the second phase, attention was paid to the Hammett correlation to study the effect of the substituent on the potential of electrochemical oxidation of selected phenolic compounds. Among the substituents studied were (-H, -CH3, -OCH3, -(CH2)2COOH, -COOH a -NO2). In general, the oxidation potentials were lower in the basic medium than in the acidic medium on all studied surfaces. An exception was the studied phenolic acids, which could not be detected in an alkaline medium on the O-terminated BDD, probably due to electrostatic repulsion between the partially negative electrode surface and the acid dianion. The highest correlation coefficient for the dependence composed of all studied phenols was obtained for the polished and H-terminated surface at pH 11, ie the dissociated form of phenolic substances.
