Realizaram-se dois experimentos: I) Aspectos relacionados à maturação in vitro e análise ultra-estrutural de complexos cumulus-oophorus (COC) de felinos domésticos e II) Produção in vitro e análise ultra-estrutural de embriões de gato doméstico. O objetivo do experimento I foi avaliar as taxas de recuperação e maturação in vitro de COC e a descrição de suas características ultra-estruturais. O experimento II visou à avaliação das taxas de clivagem, de formação de mórulas, de produção de blastocistos; à avaliação da qualidade dos embriões felinos em fase inicial de desenvolvimento e à descrição das características ultra-estruturais dos embriões obtidos. No experimento I, para verificação das taxas de recuperação oocitária e de maturação in vitro foram avaliados 473 oócitos grau I no período I e 482 oócitos grau I no período II, provenientes de 20 e de 10 pares de ovário, respectivamente. O número médio de oócitos obtidos por gata foi de 23,65 e 48,2 nos períodos I e II, respectivamente. Esses COC foram maturados in vitro e classificados quanto à configuração nuclear em vesícula germinativa (VG), quebra de vesícula germinativa (QVG), metáfase I (MI), metáfase II (MII) e degenerados/não identificados (D/NI). This study was divided in two experiments: I) In vitro oocyte maturation and ultrastructural analysis of cumulus-oocyte complexes (COC) of domestic cats and II) In vitro embryo production and ultrastructural analysis of embryo of domestic cats. The aim of the experiment I was to analyze the COC recovery rate and in vitro nuclear maturation (IVM) rate and the description of its ultrastructural characteristics. The experiment II aimed to evaluate cleavage and morulae rates and blastocyst formation; to evaluate the quality of domestic cat embryos and to describe ultrastructural characteristics of these embryos. In experiment I, to evaluate COC recovery rate and in vitro maturation rate; 473 Grade I COC recovered from 20 ovarian pairs and 482 Grade I COC recovered from 10 ovarian pairs were evaluated during season I and season II, respectively. These COC were in vitro matured and classified according to nuclear configuration as germinal vesicle (GV), germinal vesicle breakdown (GVBD), metaphase I (MI), metaphase II (MII) and degenerated/non-identified (D/NI). Desta forma, o presente estudo teve por objetivo pesquisar em uma área endêmica para leishmaniose visceral canina, a possibilidade de infecção em gatos, por meio de exame parasitológico direto e da pesquisa de anticorpos anti-Leishmania chagasi pelas técnicas de ensaio imunoenzimático indireto (ELISA) e reação de imunofluorescência indireta (RIFI). Para tanto, foram colhidas amostras de soro de 200 gatos, encaminhados ao Centro de Controle de Zoonoses do município de Araçatuba - São Paulo - Brasil, bem como realizadas punções biópsias aspirativas de linfonodo, medula óssea, baço e fígado, utilizados para a confecção de preparados citológicos para a pesquisa direta de formas amastigotas de Leishmania sp. A prevalência da doença nessa população de gatos foi de 6,5%. Dos 200 animais avaliados, oito (4,0%) apresentaram resultado parasitológico positivo, seis (3,0%) apresentaram títulos sorológicos acima do ponto de corte (0,332) pela técnica de ELlSA e um (0,5%) evidenciou título superior ao ponto de corte (1:40) pela RIFI, totalizando 13 gatos considerados positivos. / In spite of some reports of the occurrence of feline visceral leishmaniasis, the literature is scarce about its research on populations of cats in endemic areas for the disease. The present work aimed to study, in an endemic area for canine visceral leishmaniasis, the infection possibility in cats using the direct parasitological test, enzyme-linked immunosorbent assay (ELISA) and indirect immunofluorescent antibody test (IFAT). For this purpose, a total of 200 cats directed to the Zoonosis Control Center of the municipal district of Araçatuba - São Paulo - Brazil were employed. Lymph node, bone marrow, spleen and liver aspiration biopsies were carried out and observed under optical microscope to search for amastigote forms of the parasite and serum samples were submitted to serological methods in order to detect anti-Leishmania chagasi circulating antibodies. The prevalence of the disease in this population of cats was 6.5%. Amastigote forms of the parasite were observed in eight (4.0%) cats; by ELISA method, six (3.0%) cats presented titer above the specie's cut off point (0,332) and one cat (0,5%) showed a titer above 1:40, a positive serological reaction, by IFAT, totalizing 13 positive cats.

The efficacy of Cat hair 9cH and Histaminum 9cH in treating the symptoms of cat allergy

M.Tech. / Cat allergy is the allergic reaction of a person’s body when exposed to allergens borne in the cat’s dander (shed skin cells), saliva or urine. The major cat allergen, called Fel d 1, is found in cat saliva, dander from sebaceous glands in the skin, fur and anal sebaceous glands. Symptoms of cat allergies may include sneezing, itchy and watery eyes, coughing and wheezing and in rare circumstances anaphylaxis occurs. The aim of this study was to determine the efficacy of the homoeopathic complex Cat hair 9cH and Histaminum 9cH in treating the symptoms of cat allergy using the skin prick test and a symptom score card which rates the severity, frequency and duration of symptoms experienced. This was a four-week double blind, placebo-controlled study in which thirty participants, male and female between the ages of 18 and 45 were randomly divided into two groups. Advertisements was placed at the University of Johannesburg Doornfontein campus and at the Weleda pharmacy (Fourways). Potential participants that meet the inclusion criteria were requested to sign the Participant Information and Consent Form. This study had to form part of a group study related to cat allergy analysis. Participants were requested to report to Ampath laboratories to undergo a skin prick test to establish the presence of a cat allergy. Those participants that tested positive for cat allergies were to undergo an initial consultation involving a case history and a focused physical examination. Participants were given a score card each week to evaluate their severity, frequency and duration of their symptoms to be completed at the end of each week. Participants were given a 25ml bottle of either the homoeopathic complex of Cat hair 9cH and Histaminum 9cH or the placebo in tablet form, and each participant was instructed to take two tablets twice daily throughout the four week study. At the second consultation the physical exam was repeated and a second bottle of medication were given. At the end of week four, participants had to report to Ampath laboratories for the final skin prick test. This was to be followed by the third and final consultation, where the physical examination was done and all score cards collected. All data will be analysed using the Shapiro Wilk test, Mann-Whitney test, Friedman test and Wilcoxon test. With the aid of results from each participant the researcher arrived at the conclusion that the homoeopathic complex Histaminum 9cH and Cat Hair 9cH was effective on alleviating the symptoms of cat allergy.

The efficacy of Cat saliva 9cH and Histaminum 9cH in the desensitization of cat allergy

(Homoeopathy) / Cat allergy is an Immunoglobulin E (IgE) response by the body in hypersensitive individuals, triggered by exposure to cat allergen, which can be found in cat saliva and cat dander. Cat allergy produces a number of symptoms which negatively influence daily living and may be exacerbated in hypersensitive individuals to halt daily functioning. Conventional treatment for cat allergy comprises of antihistamines, anti-inflammatories, leukotrine blockers and decongestants, which have many adverse effects, are not always effective and may lead to drug tolerance. To date, research has been conducted by Naidoo et al., (2011) on the efficacy of the homoeopathic complex of Cat saliva 9cH and Histaminum 9cH in treating the symptoms of cat allergy, which yielded positive and statistically significant results according to the skin prick test. The desensitising effect of this complex on blood IgE levels has not yet been studied. The aim of this study was to determine the efficacy of Cat saliva 9cH and Histaminum 9cH in the desensitization of cat allergy, by use of cat specific RAST tests and a symptom score card. The complex of Cat saliva 9cH and Histaminum 9cH was prepared according to homoeopathic principles and prescribed according to isopathic principles. Thirty male and female participants between the ages of 18 and 45, who had a positive RAST test result for cat allergy, were selected for this four week, double-blind, placebo-controlled study. The RAST tests were performed at the beginning of the study (week 1) and at the end of the study (week 4). The participants were randomly divided into treatment and control groups. The treatment group received the homoeopathic complex of Cat saliva 9cH and Histaminum 9cH and the control group received the placebo. Participants were asked to take two tablets in the morning and two tablets at night sublingually for the duration of the study period. Each participant received a symptom score card which was to be completed at the end of each week. The symptom score card rated the severity, frequency and duration of their symptoms. The results were statistically analysed using, the Friedman-K Related Samples Test, the Wilcoxon Signed Ranks Test and the Mann-Whitney U Test. The results showed that the homoeopathic complex of Cat saliva 9cH and Histaminum 9cH did not improve the IgE levels or IgE grading in the treatment group. When looking at the symptoms, the treatment group had a statistically significant improvement in the severity, duration and frequency of the symptom “red itchy eyes”, as well as improved duration of the symptoms “runny itchy stuffy nose” and “sneezing”; while the control group was found to have a statistically significant improvement in the frequency of the symptom “red itchy eyes”, “scratchy throat”, as well as the duration of “red itchy eyes”. In conclusion it can be said that the homoeopathic complex of Cat saliva 9cH and Histaminum 9cH did not produce desensitization to cat allergen, thus leading to the rejection of the hypothesis and the acceptance of the null hypothesis.

Use of Poultry Collagen Coating and Antioxidants as Flavor Protection for Cat Foods Made with Rendered Poultry Fat

Poultry skins and rendered poultry fat are by-products produced in excess at rendering plants. The use of low value by-products such as poultry collagen, from poultry skins, and fat to improve flavor and quality in dry pet food could be economically attractive. This study examined a poultry collagen coating as a protective barrier against oxidation in dry cat food made with rendered poultry fat. Collagen was extracted from chicken skins, dissolved in an acidic solution, applied to dry cat food and dried to form a surface film. Six treatments were examined: kibble, kibble with fat, kibble with collagen, kibble with fat and collagen, kibble with fat, BHA/BHT and collagen and kibble with fat, tocopherol and collagen. There were two storage conditions: ‘jungle condition’ (42°C and 83% relative humidity) and ‘ambient condition’ (21°C and 51% relative humidity). In ‘jungle conditions’, thiobarbituric acid reactive substances (TBARS) was measured over an eight-day period at day 0, 2, 4, 6, and 8. In ‘ambient conditions’, TBARS was measured over a thirty-day period at day 0, 7, 14, 21, and 30. Water activity and moisture contents were measured. There were significantly higher TBARS (P<0.05) for the control kibble at both storage conditions. There was significantly higher fat percentage (P<0.05) in all treatments with the additional fat coatings. Fatty acid compositions showed slight changes during storage. There were some changes in the aroma profile of the kibble with fat treatment having musty, moldy and plastic aromas at both storage conditions. The volatile aromas might be an indication of oxidation in the poultry fat. / Master of Science

Intelligent Goal-Oriented Feedback for Java Programming Assignments

Within computer science education, goal-oriented feedback motivates beginners to be engaged in learning programming. As the number of students increases, it is challenging for teaching assistants to cater to all the doubts of students and provide goals. This problem is addressed by intelligent visual feedback which guides beginners formulate effective goals to resolve all the errors they would incur while solving a programming assignment. Most current automated feedback mechanisms provide feedback without categorization, prioritization, or goal formulation in mind. Students may overlook important issues, and high priority issues might be hidden among other issues. Also, beginners are not well equipped in formulating goals to resolve the issues provided in the feedback. In this research, we address the problem of providing an effective, intelligent goal-oriented feedback to student's code to resolve all the issues in their code while ensuring that the code is well tested. The goal-oriented feedback would eventually implicitly navigate the students to write a logically correct solution. The code feedback is summarized into four categories in the descending order of priority: Coding, Student's Testing, Behavior, and Style. Each category is further classified into subcategories, and a simple visual summary of the student's code is also provided. Each of the above-mentioned categories has detailed feedback on each error in that category to provide a better understanding of the errors. We also offer enhanced error messages and diagnosis of errors to make the feedback very useful. This intelligent feedback has been integrated into Web-CAT, an open-source automated grading tool developed at Virginia Tech that is widely used by many universities. A user survey was collected after the students have utilized this feedback for a couple of programming assignments and we obtained promising results to claim that our intelligent feedback is effective. / Master of Science / Within computer science education, goal-oriented feedback motivates beginners to be engaged in learning programming.
