Centrum odpočinku v Lavičkách / Center of Rest in Lavičky

The topic of this master´s thesis is a new building design of recreation centre in Lavičky and processing of project documentation for this building realization. This thesis is furthermore completed with two specializations. It deals specifically with a specialisation in the field of technical facilities of buildings – heating and air conditioning. The recreation centre is a complex of two units that will provide restaurant and accommodation services with leisure activities facilities. In one of the units there is designed an accommodation facility with smaller wellness area, in the other unit there is designed a restaurant. The estate where the units are located occurs in the north part of the village Lavičky. This locality is built up by family houses and apartment blocks. The architectural solution of the designed units follows on from the concept of simple country object where the object has the shape of rectangle ground plan and is roofed by typical saddle roof. The building of the restaurant and also the guest house is in the shape of rectangle and is roofed by double sheathed saddle roof with metal folded roofing in dark grey colour. As well it utilises modern elements of architecture as big window openings or dormers. The filling of the openings will be accomplished by wooden windows in natural shade. The façade of the units is designed in that way to match both unis together. It is solved by white façade paint in combination with cement splinter panels imitating wood or cement splinter panels in grey colour and is completed by outdoor wooden pergola or wooden storm porch. The wall base part is formed by mosaic façade in grey colour. The layout solution of the units follows on from operation requirements of the particular services and they are separated in space for customers and for operation space. The building is designed as rectangle and will serve as support area for guests´accommodation. The building has saddle roof. The main material of vertical and horizontal structures is cast-in-place concrete using formwok VELOX. The second building is horse stables. The building has two floors, partial basement and partial exploited attic for storing fodder. The L-shaped building will serve as stable for eleven horses, storage of fodder and garage for agricultural equipment. The building has saddle roof with truss girders. The main material of vertical and horizontal structures is cast-in-place concrete using formwok VELOX. These buildings are designed on slightly sloping terrain from east to west.

The diploma thesis is focused on a new building guest house with a restaurant, located in the town of Valašské Klobouky. The land has a sloping character. The structure of the guest house consists of one basement and two above-ground floors. The capacity of the restaurant with a terrace is for 84 people, accommodation for 22 guests in 9 rooms and wellness for 15 visitors. The object is a rectangular shape of a rectangle measuring 25.4x14.25 m. The external and internal vertical structures are made of sand-lime blocks of the KM BETA SENDWIX system, horizontal structures are made of reinforced concrete ceiling. The guest house is insulated with the ETICS contact isulation system. The diploma thesis has the form of design documentation for the building process.

L’aromaticité et l’antiaromaticité demeurent un sujet d’actualité au niveau fondamental puisque ces concepts permettent d’expliquer la réactivité de diverses classes de molécules. Certains composés polycycliques ayant une conjugaison π étendue, comme par exemple les tétraoxa[8]circulènes, ont des cycles aromatiques et antiaromatiques qui affectent leurs propriétés physicochimiques. Les oxacirculènes sont formés d’un anneau de quatre furanes et de quatre benzènes positionnés en alternance et créant un cœur formellement antiaromatique de cyclooctatétraène (COT). Malgré cela, ces composés démontrent une grande stabilité et peuvent servir en tant que composantes de dispositifs optoélectroniques comme des diodes électroluminescentes. Les propriétés optoélectroniques de ces composés et leur organisation moléculaire à l’état solide sont à l’origine d’un comportement unique. Le présent mémoire décrit la synthèse de tétraoxa[8]circulènes par une réaction très spéciale de cyclocondensation de 1,4-quinones catalysée par des acides, permettant d’accéder à des grandes molécules conjuguées en une étape. Les composés synthétisés ont une géométrie qui empêche leur empilement efficace et crée des vides dans la structure cristalline pouvant servir à l’inclusion de molécules invitées. En particulier, l’inclusion de C60 effectuée par cocristallisation cause l’uniformisation des longueurs des liens du cœur COT des oxacirculènes, suggérant des interactions de transfert de charge. Enfin, les propriétés optiques étudiées par spectroscopie UV-vis et par fluorescence indiquent que les oxacirculènes sont sensibles à leur environnement moléculaire et pourraient donc être utilisés comme des senseurs chimiques. / Aromaticity and antiaromaticity remain important topics of chemical research because these concepts explain the reactivity of various classes of molecules. For instance, certain polycyclic compounds with π-extended conjugation, such as tetraoxa[8]circulenes, contain both aromatic and antiaromatic rings, which affect their physicochemical properties. Oxacirculenes consist of a circular arrangement of four furan and four benzene rings positioned in an alternating fashion, thus creating a formally antiaromatic cyclooctatetraene (COT) core. Despite this, oxacirculenes exhibit great stability. They can serve as components in optoelectronic devices such as electroluminescent diodes, and their molecular organisation in the solid state can be controlled in ways that influence their properties. The present memoir describes the synthesis of tetraoxa[8]circulenes by a very special acid-catalysed cyclocondensation reaction starting from suitable 1,4-quinones, thereby allowing the preparation of large conjugated molecules in a single step. The compounds prepared have an awkward molecular structure that inhibits efficient packing and creates voids in the crystal structure, allowing the inclusion of guests. In particular, the inclusion of C60, achieved by cocrystallization, appears to create charge-transfer interactions as indicated by uniformization of bond lengths in the COT core. Optical properties studied by UV-vis spectroscopy and fluorescence show that oxacirculenes are sensitive to their local chemical environment, suggesting that they can be used as chemical sensors for the rapid detection of guests.

On the ground floor is located a restaurant with facilities, also rooms for sport and relaxation. On the second and the third floor is the centre’s accommodation with a total capacity of 38 persons. In the basement are mainly storerooms and utility rooms. The building is made of ceramic bricks. The ceiling construction is monolithic and the building has a flat roof. The building is constructed on footing foundations and strip foundations.

The guest house has got fourteen residential units, one of which is wheelchair accessible. Recreational guesthouse is not designed in a passive standard. However, it is built with respect to the appropriate orientation to the cardinal, taking into account the orientation of the main entrance to the south. Recreational guest house, is designed to match the local architecture.

Designed building is placed at flat terrain at plot of number 2016 in municipality Hradec nad Moravicí, cadastral area of Hradec nad Moravicí [646814]. Guest house with restaurant has approximately shape of two rectangles, that together looks like capital letter t. Building has overall dimensions in length 36 m and in width 22,54 m. Detached building is building without cellar, when the part of restaurant is single-storeyed with mono-pitched roof and there is placed the restaurant, kitchen, facilities guests, staff facilities and technical facilities. The part of quest house with mono-pitched roof is two-storeyed and there is placed quests rooms and facilities. Biocatalytic Production, Preparation and Characterization of Large-ring Cyclodextrins

Cyclodextrins (CD) are cyclic oligosaccharides composed of six to more than sixty glucose units. Large-ring cyclodextrins (LR-CD) are novel CD comprised of more than eight glucose units with cavity structures and sizes different from that of commercially available CD<sub>6</sub> – CD<sub>8</sub>. LR-CD may offer unique molecular recognition properties and can be produced biocatalytically from starch using cyclodextrin glucanotransferase (CGTase, E.C. 2.4.1.19) in a short reaction time. LR-CD were isolated from glucose, CD<sub>6</sub> – CD<sub>8</sub> and other compounds by complexation of CD<sub>6</sub> – CD<sub>8</sub> as well as precipitation techniques. The yield of LR-CD (degree of polymerization from 9 to 21) was optimized using central composite design. Addition of polar organic solvents to the synthesis resulted in higher yields of LR-CD. LR-CD composed of 9 to 21 glucose units were successfully separated using reversed-phase of ODS-AQ chromatography and normal-phase of polyamine II chromatography. Maintaining optimized reaction conditions aided in a high yield of CD<sub>9</sub>; it could be separated with reasonable yield using a single step of polyamine II chromatography. A co-grinding method helped to obtain higher solubilization levels of glibenclamide, vitamin A acetate and vitamin D<sub>3</sub> in CD<sub>13</sub>, CD<sub>10</sub> and CD<sub>11</sub>, respectively when compared to other CD. Vitamin K<sub>1</sub> was solubilized in distilled water with CD<sub>6</sub> – CD<sub>13</sub> using a co-precipitation method. When compared with other CD, CD<sub>9</sub> was seen to be the best solubilizer. The analysis of complexes using ESI MS showed spironolactone and glibenclamide complexed with CD<sub>9</sub> and CD<sub>13</sub>, respectively.
