Sensitivity control of optical fiber biosensors utilizing turnaround point long period gratings with self-assembled polymer coatingsGifford, Erika Lea 25 July 2008 (has links)
Biosensors have a multitude of important applications in basic research, environmental monitoring, biodefense, and medicine. This research aims to show that Ionic Self-Assembled Multilayers (ISAMs) adsorbed on Long Period Gratings (LPGs) can serve as a highly sensitive, robust, inexpensive optical-based biosensor platform. The ISAM technique is a layer-by-layer deposition method that builds nanometer-thick films based on the principle of Coulomb attraction between oppositely charged polyelectrolyte solutions while LPGs cause strong attenuation bands that enable an optical fiber to be extremely sensitive to changes in the surrounding environment. LPGs have been shown to be highly sensitive to the adsorption of nanoscale self-assembled films on the optical fiber cladding surface. In this work, we utilize Turnaround Point (TAP) LPGs, which possess even greater sensitivity than standard LPGs. This thesis focuses on evaluation of approaches to increasing the sensitivity of the sensor platfom, implementation of a biosensor for detection of several biomolecules, and preliminary evaluation of the potential for pH sensing.
For a thin-film coated TAP LPG, we have demonstrated that shifts in the transmitted light intensity at the resonant wavelength of the LPG can result from the variation in film thickness and/or refractive index. We have observed decreases in intensity as large a 7 dB for one bilayer of ISAM film (~1 nm), which corresponds to an 80% decrease in the transmitted light intensity at the resonant wavelength. We have also shown that the sensitivity of the TAP LPG sensor can be increased by implementing nm-thick ISAM films that have a refractive index greater than silica. Furthermore, it is shown that incorporation of silica nanoparticles into the ISAM films significantly increases sensitivity through increased surface area and thickness.
The biotin-streptavidin system was used as a model for implementaion and optimization of the ISAM-coated TAP LPG biosensor platform. Through evaluation of various biotin derivatives to maximize the amount functionalized onto the ISAM film, optimization of the ISAM film properties, and use of LPGs designed for higher sensitivity, the minimum detectable concentration of streptavidin was decreased from 0.0125 mg/ml to 12.0 ng/ml. The biosensor platform was then tested on prostate specific antigen (PSA), which is used as a clinical marker for early diagnosis of potential prostate cancer. Using a direct crosslinking approach of the monoclonal antibody to PSA into the ISAM film, a sensitivity level of 11.64 ng/ml PSA was obtained through combined optimization of the ISAM film and antibody surface coverage. Finally, the potential of ISAM TAP LPGs as pH sensors was examined based on the pH dependent swelling of ISAM films. / Ph. D.

Biosensors have widespread applications in many areas. Currently the Surface Plasmon Resonance (SPR) biosensor is one of the most prevalent types of biosensor. However, it has several disadvantages such as being delicate, expensive, and non-portable. Ionic Self-Assembled Multilayers (ISAMs) adsorbed on Long Period Fiber Gratings (LPGs) provides an attractive platform for building optical sensors, which could potentially overcome the disadvantages of SPR biosensors. The ISAM technique is a type of layer-by-layer deposition technique for building nanoscale thin films. An LPG is a type of fiber device that is sensitive to physical property changes of the ambient environment. LPGs have been extensively investigated for use as optical sensors. We have carried out a study on combining these two techniques to build efficient biosensors.
In this thesis, we demonstrate ultra-sensitive LPGs whose attenuation can be changed by 25 dB (~99.7%) over a 48-nm spectral band, with ambient-index changes of only 2.7E-4. The device schematic allows arbitrarily high index sensitivities to be achieved, which makes it an attractive platform for realizing sensors and modulators that respond to small index changes. For a thin-film coated LPG, we find theoretically that the resonant wavelength shift of the LPG can result from either the variation of the thickness of the film and/or the variation of its refractive index. Furthermore, results illustrate that the sensitivity of the sensor could be enhanced using a nm-thick thin-film (e.g. ISAM films) whose refractive index is greater than silica. Experimentally, we demonstrate the fabrication of nm-thick ISAM films deposited on LPGs, which induces dramatic shifts in the resonant wavelength. The refractive index and the thickness of the ISAM film was precisely controlled by altering the relative fraction of the anionic and cationic materials combined with layer-by-layer deposition. Finally, we demonstrate that ISAM-coated LPGs can function effectively as biosensors by using the biotin-streptavidin system. These demonstrations confirm that the ISAM-LPG scheme provides a thermally-stable, reusable, and robust platform for building efficient optical sensors. / Ph. D.

Eine der drei Säulen der Krebsbehandlung ist die Strahlentherapie. Einer der neuesten Ansätze hierbei ist die Bestrahlung mit Ionen, zurzeit insbesondere Protonen und Kohlenstoffionen. Diese Hochpräzisionstherapie erfordert ein hohes Maß an Kontrolle, da die applizierte Dosisverteilung sehr empfindlich von Dichteveränderungen im durchstrahlten Gewebe abhängt. Das bisher einzige klinisch eingesetzte Verfahren zur in vivo Überwachung der Dosisapplikation bei Ionenbestrahlungen ist die Positronen-Emissions-Tomographie (PET). Sie ermöglicht eine Verifikation der Teilchenreichweite sowie der Lage des Bestrahlungsfeldes.
Die mit der PET-Methode gemessene Aktivitätsverteilung lässt sich jedoch nicht direkt mit der geplanten Dosisverteilung vergleichen. Daher ist eine Vorherberechnung der erwarteten Aktivitätsverteilung auf der Grundlage des Bestrahlungsplanes notwendig, welche dann mit der Messung verglichen wird und eine qualitative Beurteilung der Bestrahlung ermöglicht. Die Vorherberechnung der erwarteten Aktivitätsverteilung erfordert bislang die Kenntnis einer Vielzahl von Wirkungsquerschnitten. Nur für wenige dieser Wirkungsquerschnitte liegen jedoch Messdaten im benötigten Energiebereich und mit ausreichender Genauigkeit vor. Daher verwenden viele Monte-Carlo-Simulationen intrinsische Kernmodelle oder semi-empirische Modellierungen, die häufig eine unzureichende Genauigkeit aufweisen.
In Fachkreisen ist bisher noch nicht geklärt, welches die optimale Ionensorte für die Tumortherapie ist. Insbesondere Lithiumionen weisen aufgrund ihrer physikalischen und radiobiologischen Eigenschaften ein großes Potenzial auf. Auch für Bestrahlungen mit diesen Ionen ist ein PET-Monitoring der Therapie erstrebenswert. In der vorliegenden Arbeit wird zunächst die Anwendbarkeit der Reichweite-Verifikation mittels PET bei Bestrahlung mit Lithiumionen gezeigt. Des Weiteren wird ein Konzept zur Modellierung der Positronenemitter-Verteilung ohne Kenntnis der Wirkungsquerschnitte entwickelt. Diese Vorhersage beruht auf in Referenzmaterialien (Wasser, Graphit und Polyethylen) gemessenen tiefenabhängigen Positronenemitter-Yields, mit welchen durch geeignete Linearkombination die Verteilung der Positronenemitter in beliebigen Materialien bekannter Stöchiometrie vorausberechnet werden kann. Here, the application of ion beams, in particular protons and carbon ions, is of growing importance. This high precision therapy requires a consequent monitoring of the dose delivery since the induced dose deposition is very sensitive to density changes in the irradiated tissue. Up to now, positron emission tomography (PET) is the only in vivo method in clinical use for monitoring the dose deposition in ion beam therapy. It allows for the verification of the particle range as well as the position of the irradiation field.
The distribution of activity measured by means of PET cannot be compared directly to the planned dose distribution. Thus, a calculation of the expected activity distribution is required which then can be compared to the measurement. Simulation of the expected activity distribution requires the exact knowledge of various cross sections. Only a few of them have been measured in the required energy range so far. Therefore, in Monte Carlo simulations often intrinsic nuclear models or semi-empirical parametrization are used which often exhibit insufficient accuray.
Among experts the question on the optimum ion species for tumour therapy is still open. Especially lithium ions exhibit a great potential due to their favourable physical and radiobiological properties. Also for these ions a PET monitoring is highly desirable. The presented work shows the feasibility of range verification by means of PET for lithium irradiation. Furthermore, a concept for modeling positron emitter distributions without the knowledge of cross sections is developed. This prediction is based on depth-dependent positron emitter yields measured in reference materials (water, graphite and polyethylene). With these data the positron emitter distribution in any material of known stoichiometry can be calculated by means of an appropriate linear combination. The feasibility of the yield concept is shown for lithium and carbon irradiation, respectively, of homogeneous polymethyl methacrylate (PMMA) as well as various inhomogeneous targets.

本篇論文主要論點有三,分述如下:
(一)在相似的統合治理結構中,制度內的差異造成了統合治理模式運作成效的不同,也導致相異的經濟發展、勞動市場與社會福利表現。特別是國家治理能力與社會治理能力上的差異。就業改革成功的重要因素是國家在改革過程中發揮高度的國家治理能力,促使社會伙伴締結以改革為導向的社會契約,並且化解分配聯盟抵制的難題,進行制度與政策的改革。
(二)高度統合治理模式促成了高度經濟成長、失業率的降低與總體就業的增加。相對的,當統合治理模式退化時,就會導致經濟成長降低與遲緩,失業率升高與總體就業增長不足的情形。因此,統合治理模式與經濟表現、勞動市場之間存在著高度的相關性。但統合治理模式與社會安全支出之間的關係取決於政黨政治的中介影響。政黨擴張社會安全支出以爭取選民的支持,特別是爭取勞工階級的支持。惟在失業率高漲與福利國家危機之際,政黨都會採取社會安全支出的合理化的政策。
(三)統合治理制度調適的可能,在於啟動良性循環來促進與維持高度統合模式。經濟成長與低失業率都得力於高度的統合治理模式。高度國家治理能力,協助勞資社會伙伴重整利益匯集的代表性組織,並授與社會行政管理權力與參與經濟社會政策制定的門徑,達成了高度的統合結構的制度整合。同時高度的社會共識源自於國家與社會伙伴之間的社會伙伴精神與主導經濟發展與社會分配的意識型態確立,也相當程度地反映在當時和諧的勞資關係上,糾合了統合治理所需的社會性支持。高度制度性治理與社會性支持的統合治理模式促成了那一時期內經濟快速成長與低失業率。

The desintegration of the USSR, the resulting release of censorship, and the related publication of earlier prohibited texts, as well as the new charakter of the book market entirely tranformed the shape of the literary field of the nineties. This thesis concerns itself with a specific segment of the professional literary production of this period. With its topic, this work aspires to contribute to the serious research of the little explored postcommunist period in the Russian literature. In order to assess and interpret the evolution of opininions in this period of time, the chosen method is the one of commented overview of literary criticism and of literary-theoretical texts published mainly between 1986 and 2000 in Russian publications (mostly in so-called "thick magazines"). The thesis focuses on studies interested in the work of the middle and younger generation of the Russian prose writers, and more precisely on studies concerning the structural aspects of the literary situation in the nineties. On the background of the specific cultural and social situation, the method of the ample commentary is an appropriate way regarding how to interpret the literary theorists' basis of thoughts and opinions, and at the same time how to affect these experts' relationship to the new tendencies in the...
