Dissection du programme développemental du noyau paraventriculaire de l'hypothalamus.

Une cascade de facteurs de transcription composée de SIM1, ARNT2, OTP, BRN2 et SIM2 est requise pour la différenciation des cinq types cellulaires qui peuplent le noyau paraventriculaire (PVN) de l’hypothalamus, un régulateur critique de plusieurs processus physiologiques essentiels à la survie. De plus, l’haploinsuffisance de Sim1 est aussi une cause d’hyperphagie isolée chez la souris et chez l’homme. Nous désirons disséquer le programme développemental du PVN, via une approche intégrative, afin d’identifier de nouveaux gènes qui ont le potentiel de réguler l’homéostasie chez l’individu adulte. Premièrement, nous avons utilisé une approche incluant l’analyse du transcriptome du PVN à différents stades du développement de la souris pour identifier de tels gènes. Nous avons comparé les transcriptomes de l’hypothalamus antérieur chez des embryons de souris Sim1+/+ et Sim1-/- à E12.5 issus de la même portée. De cette manière, nous avons identifié 56 gènes agissant en aval de Sim1 dont 5 facteurs de transcription - Irx3, Sax1, Rxrg, Ror et Neurod6. Nous avons également proposé un modèle de développement à deux couches de l’hypothalamus antérieur. Selon ce modèle, les gènes qui occupent un domaine médial dans la zone du manteau caractérisent des cellules qui peupleront le PVN alors que les gènes qui ont une expression latérale identifient des cellules qui donneront plus tard naissance aux structures ventrolatérales de l’hypothalamus. Nous avons aussi démontré que Sim1 est impliqué à la fois dans la différenciation, la migration et la prolifération des neurones qui peuplent le PVN tout comme Otp. Nous avons également isolé par microdissection au laser le PVN et l’hypothalamus médiobasal chez des souris de type sauvage à E14.5 pour en comparer les transcriptomes. Ceci nous a permis d’identifier 34 facteurs de transcription spécifiques au PVN et 76 facteurs spécifiques à l’hypothalamus médiobasal. Ces gènes représentent des régulateurs potentiels du développement hypothalamique. Deuxièmement, nous avons identifié 3 blocs de séquences au sein de la région 5’ d’Otp qui sont conservés chez l’homme, la souris et le poisson. Nous avons construit un transgène qui est composé d’un fragment de 7 kb contenant ces blocs de séquences et d’un gène rapporteur. L’analyse de 4 lignées de souris a montré que ce transgène est uniquement exprimé dans le PVN en développement. Nous avons généré un deuxième transgène dans lequel le fragment de 7 kb est inséré en amont de l’ADNc de Brn2 ou Sim1 et de Gfp. Nous avons obtenu quatre lignées de souris dans lesquels le profil d’expression de Brn2 et de Gfp reproduit celui d’Otp. Nous étudierons le développement du PVN et la prise alimentaire chez ces souris. En parallèle, nous croisons ces lignées avec les souris déficientes en Sim1 pour déterminer si l’expression de Brn2 permet le développement des cellules du PVN en absence de Sim1. En résumé, nous avons généré le premier transgène qui est exprimé spécifiquement dans le PVN. Ce transgène constitue un outil critique pour la dissection du programme développemental de l’hypothalamus. Troisièmement, nous avons caractérisé le développement de l’hypothalamus antérieur chez l’embryon de poulet qui représente un modèle intéressant pour réaliser des études de perte et de gain de fonction au cours du développement de cette structure. Il faut souligner que le modèle de développement à deux couches de l’hypothalamus antérieur semble être conservé chez l’embryon de poulet où il est aussi possible de classer les gènes selon leur profil d’expression médio-latéral et le devenir des régions qu’ils définissent. Finalement, nous croyons que cette approche intégrative nous permettra d’identifier et de caractériser des régulateurs du développement du PVN qui pourront potentiellement être associés à des pathologies chez l’adulte telles que l’obésité ou l’hypertension. / A cascade of transcription factors composed of SIM1, ARNT2, OTP, BRN2 and SIM2 is required for the differentiation of the five major cell types populating the paraventricular nucleus (PVN) of the hypothalamus, a critical integrator of several homeostatic processes that are required for the survival of vertebrates. Haploinsufficency of Sim1 also causes isolated hyperphagia in mice and humans. The goal of our study is to dissect the developmental program of the PVN using an integrative approach in order to identify new genes that could potentially be implicated in the regulation of homeostasis in adults. First, we used a comparative approach to analyse the PVN transcriptome at different developmental stages in mice embryos in order to identity new genes implicated in PVN development. We compared gene expression in the anterior hypothalamus of E12.5 Sim1-/- and Sim1+/+ littermate embryos using a microarray approach. We identified 56 genes acting downstream of Sim1 including 5 transcription factors - Irx3, Sax1, Rxrg, Ror and Neurod6. We proposed a model for the development of the anterior hypothalamus. In this model, the genes expressed in the medial domain of the mantle layer characterise cells that will form the PVN and genes expressed in the lateral domain identify cells that will give rise to ventrolateral areas of the hypothalamus. We also showed that Sim1, like Otp, is implicated in the differentiation, migration and proliferation of the neurons populating the PVN. Furthermore, we have isolated by laser captured microdissection the PVN and ARC nucleus in wild type mice at E14.5 and compared their transcriptomes. This technique allowed us to identity 34 transcription factors specific to the PVN and 76 factors specific to the ARC. These genes represent potential regulators of hypothalamic development. Second, we identified 3 blocks of sequence in the 5’ region of Otp that are conserved between human, mouse and fish. We constructed a transgene which included a 7 Kb fragment encompassing these sequences followed by a reporter gene. The analysis of 4 mice strains showed that this transgene is specifically expressed in the prospective PVN. We have generated a second transgene in which the 7 Kb fragment is located upstream of the cDNA encoding Brn2 or Sim1 and Gfp. We obtained 4 mice strains in which the Brn2 and Gfp expression pattern is similar to the Otp expression pattern. These mice will be used to study PVN development and food intake. Also, to determine if Brn2 expression only – without Sim1 gene expression - allows the development of PVN cells, we are presently crossing these mice with Sim1 deficient mice. In conclusion, we have generated the first transgene that is specifically expressed in the PVN. This transgene constitutes a critical tool for dissecting the developmental program of the hypothalamus. Third, we have characterised the development of the anterior hypothalamus of chick embryos which represent an interesting model for loss and gain of function experiments during the development of this brain region. Interestingly, our proposed model for the development of the anterior hypothalamus seems to be conserved in chick embryos. As a matter of fact, it is possible to classify genes according to their medio-lateral expression patterns and the outcome of the regions that they are defining. Finally, we believe that this integrative approach will allow us to identify and characterize factors implicated in PVN development. From a clinical point of view, these factors could potentially be associated with pathologies such as obesity or arterial hypertension.

Régulation de l’identité des membres postérieurs par le facteur de transcription à boîte T Tbx4

Bien que partageant une homologie structurelle évidente, les membres antérieurs (MA) sont toujours différents des membres postérieurs (MP). Ceci suggère l’existence d’un programme générique de formation d’un membre, un bauplan, qui doit être modulé de façon spécifique pour engendrer cette différence antéro-postérieure de l’identité. Nous avons donc voulu identifier les mécanismes déployés durant l’évolution pour permettre la mise en place de l’identité des membres. Le laboratoire avait précédemment caractérisé, chez les souris où le gène Pitx1 est inactivé, une transformation partielle des MP en MA couplée à une perte de croissance. Nous avons donc cherché à comprendre les mécanismes en aval de Pitx1 dans la détermination de l’identité postérieure. Notre démarche nous a permis d’identifier les gènes affectés par la perte de Pitx1 dans les MP, où nous avons confirmé une dérégulation de l’expression de Tbx4. Tbx4 et Tbx5 sont des candidats évidents pour déterminer l’identité, leur expression étant restreinte aux MP et MA, respectivement, mais leur implication dans ce processus était sujette à controverse. Nous avons donc évalué l’apport de Tbx4 en aval de Pitx1 dans les processus d’identité en restaurant son expression dans les MP des souris Pitx1-/-. Ce faisant, nous avons pu montrer que Tbx4 est capable de pallier la perte de Pitx1 dans le MP, en rétablissant à la fois les caractères d’identité postérieure et la croissance. En parallèle, nous avons montré que Tbx5 était capable de rétablir la croissance mais non l’identité des MP Pitx1-/-, démontrant ainsi de façon définitive une propriété propre à Tbx4 dans la détermination de l’identité des membres postérieure. La caractérisation de l’activité transcriptionnelle de Tbx4 et Tbx5 nous a permis de mettre en évidence un domaine activateur conservé mais aussi un domaine spécifique à Tbx4, répresseur de la transcription. Par ailleurs, une mutation faux-sens de TBX4 dans les patients atteints du syndrome coxo-podo-patellaire, TBX4Q531R, inactive le domaine répresseur, empêchant la compensation de l’identité mais non de la croissance des MP dépourvus de Pitx1, démontrant l’importance de cette fonction dans l’identité postérieure. La caractérisation de l’activité répressive de Tbx4, qui se manifeste seulement dans les membres postérieurs démontre l’importance de cette fonction dans l’identité postérieure. Nous avons aussi été en mesure d’identifier un corépresseur qui est suffisant pour supporter cette activité de Tbx4. Enfin, nous avons pu aussi démontrer l’activité transcriptionnelle d’un représentant du gène ancestral, présent chez Amphioxus, qui se comporte strictement comme un activateur et semble dépourvu du domaine répresseur. En somme, nous avons précisé le rôle de Tbx4 et Tbx5, ainsi que leur mécanisme, dans la détermination de l’identité des membres. Globalement, nos travaux permettent d’élaborer une théorie où une divergence d’activité transcriptionnelle de Tbx4 et Tbx5 est responsable de l’identité des membres et même entrevoir que cette divergence d’activité soit à la base de son apparition durant l’évolution. / Forelimbs and hindlimbs are a classical example of serial homology, suggesting they share an evolutionary common generic program that directs their formation. Identity is presumably derived from specific modulations of that program in different limb type. Transcription factors are prime candidates to link these structural differences to specific modulations and three factors with limb-specific expression have been identified. Pitx1 and Tbx4 expression is restricted to the hindlimbs while Tbx5 is restricted to the forelimbs and they have all been ascribed functions in both growth and identity from knockout and overexpression studies. Recent studies have produce evidence that Tbx4 and Tbx5 are interchangeable, sharing identical properties to support growth but not identity of the limbs, the latter being a direct consequence of Pitx1 expression. Indeed, Pitx1 deficient mice have been previously described as undergoing a hindlimb-to-forelimb transformation in addition of growth defects. To better assess the shared and specific properties of Tbx4 and Tbx5, we assessed their capacities to rescue identity and growth defects by expression studies in Pitx1-/- hindlimbs. Specifically, previous studies had shown that Pitx1 deficiency causes the loss of hindlimb features, the transformation of hindlimb features toward forelimb-like morphology, the gain of a forelimb feature and the asymmetric loss of growth at the level of the femur. Targeted expression in the limbs of both Tbx4 and Tbx5 rescued the growth defects similarly. Interestingly, only Tbx4 was able to restore identity features affected in absence of Pitx1. To further assess these shared and specific properties, we conducted transcriptional assays that revealed the presence of a shared and conserved transactivating domain in the C-terminal moiety of these proteins. Moreover, we could identify a repressor domain specific to Tbx4. Human small patella syndrome maps to TBX4 and a coding mutation, TBX4Q531R, that specifically inactivates the repressive properties of Tbx4 prevents it from rescuing identity to the Pitx1-/- hindlimbs but not from rescuing growth. We also conducted a yeast two-hybrid assay that allowed the identification of putative co-factors of Tbx4, of which one seems to act as a co-repressor. Together, our results support the presence of a Tbx4/Tbx5 conserved activating domain required for limb outgrowth that is an integral part of the limb bauplan. Importantly, we identified a molecular basis for the determination of limb identity through the Tbx4-specific repressor domain and reveal a novel path through which limb identity may have emerged during evolution.

Introgression of genes from rape to wild turnip

Introgression of genes from crops into ruderal populations is a multi-step process requiring sympatry, synchronous flowering, chromosomal compatibility, successful pollination and development of the zygote, germination, establishment and reproduction of hybrid progeny. The goal of this thesis was to generate data on as many steps in this process as possible and integrate them into a predictive statistical model to estimate the likelihood of successful introgression under a range of scenarios. Rape (Brassica napus) and wild turnip (B. rapa var. oleifera) were used as a model system. A homozygous dominant mutation in the rape genome conferring herbicide resistance provided a convenient marker for the study of introgression. Potential differences between wild turnip populations from a wide range of geographic locations in New Zealand were examined. Hand pollination established the genetic compatibility of rape and wild turnip and a high potential for gene introgression from rape to wild turnip. Interspecific hybrids were easily generated using wild turnip as the maternal plant, with some minor differences between wild turnip populations. The frequency of successful hybridisation between the two species was higher on the lower raceme. However, the upper raceme produced more dormant interspecific hybrid seed. Field trials, designed to imitate rare rape crop escapes into the ruderal environment, examined the ability of rare rape plants to pollinate wild turnip plants over four summers. At a ratio of 1 rape plant for every 400 wild turnip plants, the incidence of interspecific hybridisation was consistently low (<0.1 to 2.1 % of total seed on wild turnip plants). There was a significant year effect with the first season producing significantly more seed and a greater frequency of interspecific hybrid progeny than the other years. The frequency of interspecific hybrid progeny increases when the ratio of rape: wild turnip plant numbers increases. The relative importance of anemophily and entomophily in the production of interspecific hybrids was examined. Wild turnip plants produced twice as many seeds with bee pollination relative to wind pollination. However, the frequency of interspecific hybrids under wind pollination was nearly twice that for bee pollination. Light reflectance patterns under UV light revealed a marked difference between wild turnip and rape flowers compared to near identical appearance under visible light. The data indicates that bees are able to distinguish between rape and wild turnip flowers and exhibit floral constancy when foraging among populations with these two species. Hybrid survival in the seed bank, germination and seedling establishment in the field are important components of fitness. Seed banks established in the soil after the field trials described above germinated in subsequent spring seasons. The predominantly brassica weed populations were screened for herbicide resistance and the numbers of interspecific hybrids germinating compared to the original frequency in the field trial results. Frequency of interspecific hybrids was reduced in the populations compared to the original seed deposit. Seed with a known frequency of interspecific hybrid seed was sown in a separate trial, and the frequency of interspecific hybrids compared at 0, 4, 6, and 8 weeks after sowing. Poor germination resulted limited competition between seedlings, however the frequency of interspecific hybrids declined over time indicating low plant fitness. There were no significant population effects on any parameters tested. Interspecific hybrids grown in a glasshouse were backcrossed to the parental species and selfed within the plant and within populations. Pollen from the interspecific hybrids was found to have markedly reduced fertility. Interspecific hybrid plants had low female fertility, with the majority (88%) of the pollinated flowers aborting the siliques. Of the remaining siliques, most (98%) had only one to three seeds per silique. Inheritance of the herbicide resistance gene was regular in backcrosses but highly skewed following self pollination with an excess of herbicide-sensitive progeny. Production of a stochastic predictive model integrated the information acquired over the practical work phase of this thesis and utilised the capabilities of @risk, a new application of a risk analysis tool. The three outputs examined were the number of flowering plants resulting from backcrosses to rape and wild turnip and self pollination of the interspecific hybrid progeny. Five scenarios were modelled and all demonstrated the high likelihood of introgression failure in this system. In all scenarios, >75% of simulations resulted in no interspecific hybrid progeny surviving to flowering in the third generation. In all scenarios, and for all three outputs, the seed set on the interspecific hybrids of the second generation was the major factor that limited the number interspecific hybrid progeny surviving to flowering in the third generation.

Neurological disorders affect more than 14% of the population worldwide and together with traumatic brain and spinal cord injuries represent major health, public and economic burden of the society. Incidence of inherited and idiopathic neurodegenerative disorders and acute CNS injuries is growing globally while neuroscience society is being challenged by numerous unanswered questions. Therefore, research of the CNS disorders is essential. Since animal models of the CNS diseases and injuries represent the key step in the conversion of the basic research to the clinics, we focused our work on generation of new animal models and on their use in pre-clinical research. We generated and characterized transgenic minipig model of Huntington's disease (HD) which represents the only successful establishment of a transgenic model of HD in minipig which should be valuable for testing of long term safety of HD therapeutics. Next, we crossed the well characterized R6/2 mouse HD model with the gad mouse model which lacks the expression of UCHL1 which led to results that support the theory of "protective" role of mutant huntingtin aggregates and suggest that UCHL1 function(s) may be affected in HD disturbing certain branches of Ubiquitin Proteasome System.
