Optimization of Two-photon Excited Fluorescence Enhancement between Tunable and Broadband Femtosecond Laser Pulse Excitations

This project explores optimization of two-photon excited fluorescence (TPEF) enhancement between tunable narrowband and un-tuned broadband femtosecond (fs) laser pulse excitations for two-photon microscopy (TPM). The research is conducted preliminarily in time domain and comprehensively in frequency domain to understand the physics behind TPEF enhancement by un-tuned sub-10 fs nearly transform-limited pulse (TLP) versus tunable 140 fs pulse. The preliminary study on inverse proportionality of TPEF yield to fs-pulse duration delimits a general lower-bound to narrowband fs-pulse regime (pulse duration > 40 fs) with assumption of dye-molecule frequency invariant response. Deviations from this inverse proportionality in broadband fs-pulse regime (pulse duration < 40 fs) highlights dye-molecule frequency variant response, necessity of group delay dispersion (GDD) compensation, and broadband TLP for TPEF enhancement. The follow-up comparative study is made on un-tuned sub-10 fs TLP versus tunable 140 fs pulse excitations using three dye-phantoms (Indo-1, FITC, and TRITC) representative of fluorescent probes with similar TPEF characteristics. The integrated experimental system, with custom-designed GDD compensation, dispersion-less laser-beam expanding and focusing, and compound-lens for efficient fluorescence collection with good spectral resolution, ensures accurate TPEF measurements. Differentiated TPEF enhancements of Indo-1 (1.6), FITC (6.7), and TRITC (5.2) proportionally agree with calculated ones due to the overlap of fs-pulse second harmonic (SH) power spectrum with dye-molecule two-photon excitation (TPE) spectrum. Physically speaking, with broadband sub-10 fs TLP readily involved in both degenerate (v1 = v2) and non-degenerate (v1 ≠ v2) two-photon absorption (TPA), this un-tuned ultrashort fs-pulse excitation simultaneously allows for more accessibility to TPA-associated final states and diversely promotes population of thus excited dye-molecules with the three dye-phantoms. Under environmental influences (mutual quenching through one-photon absorption(s) and solvent effect), multicolor TPEF enhancement observed from a mixture of the three dyes shows promise of sub-10 fs TLP as simultaneous excitation for multiple-dye labeled samples in contrast to compromised excitation with narrowband fs-pulse tuning. Both single- and multicolor TPEF enhancements clarify tradeoff between tunability of narrowband fs-pulse and un-tuned broadband fs-pulse excitations, being instructive to further considerations on optimization of TPEF enhancement by strategic utilization of broadband fs-pulse for better performance of TPM.

New generation three-phase rectifier

This thesis describes the development of a new generation of three-phase rectifier, used to power telecommunications equipment. The traditional topology for such power converters is a single-phase two-stage design, with a boost converter providing power factor correction at the input to the first stage and an isolated dc-dc converter making up the second stage. A two-stage design results in the output power being processed twice and this cascade effect results in an overall reduction in efficiency. A rectifier solution is sought that meets with all the requirements of the telecommunications industry, while not displaying the inherent weaknesses associated with a boost-derived topology, and which can be realised in a single-stage design. A number of common three-phase topologies exist that could be realised as telecommunication power supplies, however, they do not completely satisfy all the industry requirements. A new three-phase rectifier, which is a single-stage buck-derived topology, is proposed. As a consequence of incorporating a buck-derived topology, the three-phase rectifier does not exhibit any issues resulting from startup inrush currents, or high currents due to an output short circuit condition, as would result in a boost-derived topology. The new proposed rectifier is modular in nature, which has the added benefit of redundancy. As a result of the new three-phase rectifier having a single-stage topology, it is expected that the overall efficiency would able to reach close to 95%. This is due to the traditional two-stage designs having efficiencies around the 90% mark, and therefore by removing a stage, out of the power conversion process the overall losses would also be halved, resulting in the 5% gain in efficiency. The rectifier system requires only one controller as a result of being a single-stage design, thus also reducing the overall system cost. Simulations show that if this topology is combined with a three-phase phase-locked loop controller it can meet the industry compliance standards. The thesis follows the development of the three-phase power converter from the simulation stage to the realisation of the control hardware and stability modelling. It also provides a detailed report of an investigation into the power converter system’s performance. The thesis concludes with discussions concerning the viability of the new topology as a commercial product and indicates areas of possible future research and development.

Asynchronous bi-directional relay-assisted communication networks

We consider an asynchronous bi-directional relay network, consisting of two singleantenna transceivers and multiple single-antenna relays, where the transceiver-relay paths are subject to different relaying and/or propagation delays. Such a network can be viewed as a multipath channel which can cause inter-symbol-interference (ISI) in the signals received by the two transceivers. Hence, we model such a communication scheme as a frequency selective multipath channel which produces ISI at the two transceivers, when the data rates are relatively high. We study both multi- and single-carrier communication schemes in such networks. In a multi-carrier communication scheme, to tackle ISI, the transceivers employ an orthogonal frequency division multiplexing (OFDM) scheme to diagonalize the end-to-end channel. The relays use simple amplify-and-forward relaying, thereby materializing a distributed beamformer. For such a scheme, we propose two different algorithms, based on the max-min fair design approach, to calculate the subcarrier power loading at the transceivers as well as the relay beamforming weights. In a single-carrier communication, assuming a block transmission/reception scheme, block channel equalization is used at the both transceivers to combat the inter-blockinterference (IBI). Assuming a limited total transmit power budget, we minimize the total mean squared error (MSE) of the estimated received signals at the both transceivers by optimally obtaining the transceivers??? powers and the relay beamforming weight vector as well as the block channel equalizers at the two transceivers.

Aboriginal Two-Spirit and LGBTQ mobility: meanings of home, community and belonging in a secondary analysis of qualitative interviews

This thesis reports on a secondary analysis of individual and focus group interviews from the Aboriginal Two-Spirit and LGBTQ Migration, Mobility and Health research project (Ristock, Zoccole, and Passante, 2010; Ristock, Zoccole, & Potskin, 2011). This was a community-based qualitative research project following Indigenous and feminist methods, involving two community Advisory Committees, and adopting research principles of Ownership Control Access and Possession (OCAP) (First Nations Centre, 2007). This analysis reviews data from 50 participants in Winnipeg and Vancouver and answers: How do Aboriginal Two-Spirit and LGBTQ people describe home, community and belonging in the context of migration, multiple identities, and in a positive framework focusing on wellbeing, strengths and resilience? Findings demonstrate how participants experience marginalization in both Aboriginal and gay communities. Their words illustrate factors such as safety required to facilitate positive identities, community building, belonging, and sense of home. For participants in this study home is a place where they can bring multiple identities, a geographical place, a physical or metaphorical space (with desired tone, feeling), and a quality of relationships. Community is about places, relationships, participation, and shared interests. Belonging is relational and interactive, feeling safe, accepted, and welcome to be yourself. Detractors interfere with positive meaning making and are identified in examples of contemporary effects of historical trauma. Also included are participant recommendations for community building, descriptions of holistic wellbeing, and examples of many ways urban Aboriginal Two-Spirit and LGBTQ people are creating communities of Two-Spirit vitality and resurgence (Simpson, 2011).

Guernsey children and the Second World War

From June 1940 until May 1945, Guernsey children either lived under German occupation or were evacuated to England for the duration of the war. This thesis presents a small case study that uses oral testimony and resilience theory to describe Guernsey children’s experiences during World War Two. Its intent is to contribute towards the larger picture of British children’s experiences during this period. This thesis also aims to understand how the majority of those who were children on Guernsey during this time judged that, despite the obvious challenges related to wartime, their experiences had a net positive effect on their lives. Findings suggest that, consistent with resilience theory, children found the support they needed both internally using optimism, empathy, comparison, and the attitude of ‘getting on with it’ and externally from family, teachers, and the local people with whom they lived during evacuation or occupation. / Graduate

Nonlinear interfacial waves in two-phase flow

Large amplitude interfacial waves are an important feature of annular gas-liquid two-phase flow. They act as a source of entrainment for liquid droplets. They occur for liquid flow rates above a critical value which depends on the gas flow rate. This thesis examines the formulation of a mathematical model to describe the behaviour of these nonlinear waves. Attention is focussed on the case of vertical upwards flow with reference to the experimental conditions for the rig at AERE Harwell. A comprehensive account is given of the limitations and similarities of mathematical models proposed by earlier research workers and their applicability to vertical two-phase flow. The most suitable approaches are found to be kinematic wave theory and an integral method. Experiments have been carried out at AERE Harwell to determine the relationship between liquid flux and film thickness required by kinematic wave theory and also to test some of the theory's predictions. There is a discussion of the difficulties involved in modelling the stresses exerted by the turbulent gas core on disturbance waves. The applicability of Benjamin's 'quasi-laminar' theory is considered. A linear stability analysis indicates that the interface is always unstable. The linear theory cannot provide a criterion for disturbance wave inception. Alternative explanations for wave inception are suggested. The SMAC (Simplified Marker And Cell) numerical method has been developed to model the time dependent behaviour of large amplitude waves in vertical annular two-phase flow. Finally, it is proposed that any realistic mathematical model for disturbance waves should be based upon kinematic wave theory and should take account of wave-breaking.

Experimental studies of viscous effects on cavitation

The work reported in this thesis falls into three distinct, though intimately related parts. Part I is concerned with the production of a variable temperature two-phase flow test facility for studying single and two-phase flows around arbitrary two-dimensional bodies. It includes the initial design study required to define an operational envelope for the facility, and the design and constructional features of its main components and ancilliary systems. The design of a two-phase flow vapour ventilation experiment is described, together with the operating procedure for the facility. Part II reports single and two-phase experiments with a two-dimensional circular cylinder, the two-phase flows being generated by inducing natural cavitation. The cavitation inception flow regime was found to be strongly influenced by viscous effects. Three forms of incipient cavitation were observed, two attached or very close to the cylinder surface, and one of a detached nature, occurring well downstream in the wake of the cylindrical test body. These inception modes have been related to the fully wetted viscous flow around the cylinder in the Reynolds number range 10<sup>5</sup> <R<sub>d</sub><10<sup>6</sup>. The development of cavitation from these three aforementioned incipient states was investigated. Viscous effects were found to influence both the limited and developed cavitation flow regimes. For development of cavitation at Reynolds numbers corresponding broadly to the supercritical range for fully wetted flow, a critical cavitation number was found at which the apparent free stream lines of the flow changed from a concave to a convex disposition. For the experimental configuration used, this critical cavitation number was independent of Reynolds number, for Reynolds numbers above the critical value. With development of cavitation at Reynolds numbers corresponding broadly to the subcritical range for fully wetted flow, no such gross changes in flow pattern were observed, the displacement of the apparent free streamlines of the flow between the limited and fully developed cavitating states being minimal. Part III contains recommendations for modifications to the experimental facility and suggestions for further studies arising from the results reported herein.
