A new simple design approach will be described that utilizes test results from the Repeated Load Triaxial Apparatus to establish the risk level of permanent deformations in the unbound granular layers (UGL) in pavement constructions under consideration of the seasonal effects. From this data a serviceability limit line (plastic shakedown limit) stress boundary for the unbound granular materials (UGM) was defined for different moisture contents. Below this line the material will have stable behavior. The serviceability limit line was applied in a finite-element (FE)-program FENLAP to predict whether or not stable behavior occurs in the UGM. To calculate the stress in the UGL, a nonlinear elastic model (Dresden Model), which is described in the paper, was implemented into the FE-program. The effects of changing moisture content during Spring-thaw period and asphalt temperature on pavement structural response were investigated. Additionally, permanent deformation calculations for the UGL were performed taking the stress history into consideration. The results clearly demonstrate that, for pavement constructions with thick asphalt layers, there is no risk of rutting in the granular base, even at high number of load repetitions. The study showed that the proposed design approach is a very satisfactory simple method to assess the risk against rutting in the UGL, even without the calculation of the exact permanent deformation of the pavement construction.

Laboratory characterization of subgrade soils and unbound granular materials is an essential component of the Mechanistic-Empirical Pavement Design Guide (Pavement ME). The design thickness and performance of a pavement structure are highly dependent on the deformation behaviour of subgrade and granular material. Specifications for granular materials vary among transportation agencies based on the availability of materials, climatic conditions, and function. Specifications aim to provide durable materials that meet design requirements and achieve the target design life with cost effective materials.
The objectives of the research are to:
• evaluate resilient modulus of typical fine-grained soils under traffic loading.
• evaluate resilient modulus, permanent deformation, and permeability of typical unbound granular materials.
• evaluate the effect of moisture and fines fraction on the performance of unbound granular materials and subgrade soil.
• develop prediction models for resilient modulus to improve reliability of Level 2 inputs in the Pavement ME.
• provide test data in support of updating Manitoba Infrastructure and Transportation specifications for unbound granular materials to improve the performance of pavement structures.
Resilient modulus tests were conducted on three types of subgrade soil (high plastic clay, sandy clay, and silty sand/sandy silt) at four levels of moisture content. Resilient modulus, permanent deformation and permeability tests were conducted on six gradations representing two types of granular material (100% crushed limestone and gravel) at two levels of moisture content. Prediction models were developed for resilient modulus and compared to the models developed under the Long Term Pavement Performance program. The proposed models provided more reliable predictions with lower root mean square error.
The deformation behaviour of the granular materials was classified according to the shakedown and dissipated energy approaches. Among the tested fines contents, limestone and gravel materials with optimum fines contents of 4.5% and 9%, respectively, had better resistance to plastic deformation and higher resilient modulus. The dissipated energy approach can be used to determine the stress ratio for the boundary between post compaction and stable zones from multistage triaxial testing. Result of permeability tests showed that the hydraulic conductivity of unbound granular material increased as the fines content decreased. / February 2016

Infections in critically ill patients are a problem for the healthcare system and at any one time, 70 % of all intensive care unit (ICU) patients are treated with antibiotics. Antibiotics bind toproteins in the blood, but only unbound drug can diffuse over capillary membranes and bindto the targeted receptor. Standard protein binding percentages for antibiotics have been developed from studies on healthy volunteers and dosing regimens for patients are adapted accordingly. The determination of the total concentration of antibiotics in patients’ bloodsamples is, based on the standard percentages, ordinarily representative for the pharmacological effect of the antibiotic. However, certain conditions that are common incritically ill patients can alter protein binding percentages, resulting in a larger or smaller unbound fraction. This in turn can result in toxicity or therapeutic failure. The aim of this project was to develop an analytical method for the determination of the unbound concentration of the Beta-Lactam antibiotics cefotaxime, flucloxacillin, cloxacillin and piperacillin in plasma. A method was successfully developed using ultrafiltration for the extraction of unbound analytes and ultra high performance liquid chromatography tandem mass spectrometry, UHPLC-MS/MS, for their quantification. The method was partly validated according to the European Medicines Agency’s guidelines on bioanalytical method validation. / Kritiskt sjuka patienter med infektioner är en börda för sjukvården och 70 % av alla patienter på intensivvårdsavdelningar är ordinerade antibiotika. Antibiotika binder till proteiner i blodet, men enbart den icke-proteinbundna (fria) fraktionen kan diffundera över kapillära membran och binda till receptorer. Standardproteinbindningsgrad för olika antibiotika har utvecklats från studier på friska frivilliga och doseringen av läkemedlen är anpassade därefter. Den totala koncentrationen av antibiotika i patienters blod är vanligen representativ för den farmakologiska effekten. Dock kan vissa sjukdomar påverka proteinbindningsgraden vilket resulterar i en större eller mindre mängd fria antibiotika i blodcirkulationen. Det här kan i sintur resultera i toxicitet eller otillräcklig effekt av läkemedlet. Syftet med det här projektet var att utveckla en analytisk metod för att bestämma den fria koncentrationen av Beta-Lactam antibiotikan cefotaxim, flukloxacillin, kloxacillin och piperacillin i plasma. En metod utvecklades med ultrafiltrering för extraktion av den fria fraktionen och högupplösande vätskekromatografi och tandem masspektrometri, UHPLCMS/MS, för kvantifiering av analyterna. Metoden validerades delvis enligt den Europeiska Läkemedelsmyndighetens riktlinjer för bioanalytisk metodvalidering.

Ziel der vorliegenden Dissertation war die Erweiterung der Kenntnisse über das nichtlinear-elastische Spannungs-Verformungsverhalten von Tragschichten ohne Bindemittel (ToB). Ein analytisches Bemessungsverfahren ist aufgrund der schwierigen Modellierbarkeit des Verformungsverhalten der einzelnen Straßenbaustoffe bisher nicht existent. Die Dissertation soll einen Beitrag zur Entwicklung eines solchen Bemessungsverfahrens leisten. Eine Literaturrecherche zum Verformungsverhalten von ToB bildet die Grundlage für die Festlegung der Vorgehensweise. Basis der weiteren Untersuchungen sind Triaxialversuche, die an der Universität Nottingham an verschiedenen Gesteinskörnungen durchgeführt wurden. Mit Hilfe der Shakedown-Theorie konnte belegt werden, dass sich ToB in unterschiedlichen Beanspruchungsbereichen nach verschiedenen Gesetzmäßigkeiten verhalten und dass Modelle zur Beschreibung des Verformungsverhaltens von ToB bereichsweise verschieden formuliert werden müssen. Somit ist es möglich, Gültigkeitsgrenzen für elastische Stoffmodelle zu ermitteln sowie Beanspruchungsgrenzen für ToB festzulegen. Mit ausgewählten Stoffmodellen erfolgten Beanspruchungsberechnungen nach der Finite-Elemente Methode (FEM). Die beste Annäherung zwischen Messwerten und Rechenergebnissen ergibt sich bei Anwendung des DRESDNER Modells. Berechnungen für Befestigungen nach den RStO 01 haben gezeigt, dass die Beanspruchungen auf den ToB bei Anwendung des DRESDNER Modells stark von denen bei Anwendung linearer Elastizität abweichen können. Durch unterschiedliche Überbauungsdicken der ToB besitzen diese einen verschieden hohen Anteil am Tragverhalten der Befestigung. Abschließend wird eine Methode zur Ermittlung von Sicherheitsniveaus vorgestellt. Bei Kenntnis der entsprechenden Schichtparameter lassen sich die Beanspruchungen jeder beliebigen Bauweise mit denen von Bauweisen nach RStO vergleichen. Als Ergebnis kann festgehalten werden, dass das Sicherheitsniveau einer bestimmten Bauweise nach RStO genauso groß ist, wie eine Befestigung mit einer dünneren Asphalttragschicht, dafür jedoch einer steiferen ToB. / The aim of this thesis was the extension of the knowledge about the resilient stress-deformation behaviour of unbound granular materials (UGM). Due to difficulties in modelling the behaviour of the single materials an analytical design method does not exist at present. Therefore this thesis makes a contribution for developing such a design method. A study of international publications about the current knowledge of the stress-deformation behaviour of UGM and repeated load triaxial tests, which have been performed at Nottingham University, were the base for all further investigations. With the shakedown concept it could be shown that materials behave in a different manner depending on the applied stress level and that material laws for describing the resilient deformation behaviour of UGM have to be formulated separately for different stress ranges. Within this thesis a method is introduced which helps to find boarders for the applicability of different material laws and limiting stress lines where below the lines stable behaviour and admissible deformations for the material are expected. Applying selected material laws finite element calculations have been performed. Comparing calculation results with measurement results it can be shown that the Dresden model offers the best approach. Calculations for pavement constructions applying the Dresden model have resulted that the vertical stresses differ very much in comparison with elastic behaviour for the UGM. It can be concluded that the thickness of the covering bounded layers have an effect to the contribution of the unbound granular layers at the complete deformation behaviour of the pavement construction. Finishing a method is introduced to determine the safety levels of pavement constructions. It seems to be possible to compare the stress-strain levels of any pavement construction with those from the german standard RStO 01. Existing functions could be modified to determine safety coefficients, i. e. ratios between admissible and existing numbers of load cycles. It can be concluded that there are the same safety levels for different pavement constructions ? a thinner asphalt layer can be compensated with a stiffer UGM.

Moisture Influence on Structural Behaviour of Pavements : Field and Laboratory InvestigationsSalour, Farhad January 2015 (has links)
The structural behaviour of pavements in cold regions can considerably be affected by seasonal variation in environmental factors such as temperature and moisture content. Along with the destructive effect of heavy traffic loads, climatic and environmental factors can considerably contribute to pavement deterioration. These factors can influence the structural and functional capacity of the pavement structures which, as a result, can trigger and accelerate pavement deterioration mechanisms. Studies on the influence of variation of the environmental factors on the response and behaviour of pavement materials have shown that proper consideration to these factors must be given in realistic pavement design and analysis. In flexible pavement structures, particularly with a thin hot mix asphalt (HMA) layer, unbound materials and subgrade soil largely contribute to the overall structural behaviour of the pavement system. In unbound materials, moisture content and its variation can significantly affect pavement layer stiffness and permanent deformation characteristics. Therefore, the moisture condition of pavements and its influence on the mechanical behaviour of pavement materials has been of interest among the pavement research community. A proper understanding of moisture transformation in pavement systems and its effects on pavement performance are important for mechanistic pavement design. The present summary of this doctoral thesis is based on four main parts. The first part of the thesis covers field measurements and findings from a test section along county road 126 in southern Sweden and consists of two journal papers (paper I and II) tackling different aspects of the research topic. This test section is located in a relatively wet ground condition and consists of a thin flexible pavement structure with a deep drainage system. It is instrumented with subsurface temperature, volumetric moisture content and groundwater probes. The mechanical response of the pavement structure was investigated using Falling Weight Deflectometer (FWD) measurements. The second part of the thesis (paper III and IV) are based on laboratory experiments and investigates different recent approaches that have been proposed to apply principles of unsaturated soil mechanics for incorporating seasonal variation of moisture content into the resilient modulus models using matric suction. The third part of the thesis (paper V) builds a bridge that spans between the laboratory and field investigations with an attempt to evaluate one of the predictive models presented in Paper III. The fourth part of the thesis (paper VI) mainly focuses on the laboratory-based investigation of the permanent deformation characteristic of subgrade soils. In this part, the permanent deformation characteristics of two different silty sand subgrade soils were investigated and modelled using the data obtained from repeated load traxial tests. Paper I mainly focuses on the spring-thaw weakening of the pavement structure. The environmental data collected using different sensors and the FWD tests were used to investigate variations in moisture content with thaw penetration and its influence on the stiffness of unbound layers and the pavement’s overall bearing capacity. Using the backcalculated layer stiffness and corresponding in situ moisture measurements in the unbound layers, a degree of saturation-based moisture-stiffness model was developed for the granular material and the subgrade. In Paper II, the drainage system of the structure was manually clogged during a three month period in summer to raise the groundwater level and increase the moisture content of the layers. Along with the subsurface groundwater level and moisture content monitoring, the structural response of the pavement was studied. In this research work, the FWD tests were conducted at three different load levels. The stress dependent behaviour of the unbound granular layer and the subgrade soil were further studied using the multilevel loads FWD test data. Additionally, parameters of a nonlinear stress-dependent stiffness model were backcalculated and their sensitivity to in situ moisture content was studied. In Paper III and IV, series of suction-controlled repeated load triaxial (RLT) tests were conducted on two silty sand (SM) subgrade materials. Several resilient modulus prediction models that account for seasonal moisture content variation through matric suction were summarized and after optimizing the model parameters, the capability of the prediction models in capturing the material response were evaluated. In Paper V, an attempt was made to evaluate the proficiency of one of the suction-resilient modulus models using the field moisture content and FWD measurements from the Torpsbruk test site. The backcalculated subgrade stiffness dataset at different moisture contents were compared with resilient modulus models obtained from the suction-resilient modulus predictive model. Paper VI presents an evaluation of several permanent deformation models for unbound pavement materials that incorporate the time-hardening concept using a series of multistage repeated load triaxial (RLT) tests conducted on silty sand subgrade materials. The permanent deformation tests were conducted at four different moisture contents with pore suctions measurement throughout the test. The effect of moisture content (matric suction) on the permanent deformation characteristics of the materials and the predictive model parameters were further investigated. / <p>QC 20150324</p>
