O objetivo desse trabalho foi estudar os mecanismos que alteram a produção de melatonina na glândula pineal durante o processo de caquexia associada ao câncer e o papel de citocinas neste contexto. Os resultados mostraram um aumento da produção de melatonina no grupo inoculado com o Tumor de Walker 256 (GT) junto com uma maior atividade e expressão gênica da enzima AA-NAT, principal enzima reguladora da síntese de melatonina. O estudo in vitro mostrou que a glândula do GT produziu menos melatonina que a glândula do grupo controle (GC) após estimulação com noradrenalina (NOR). Além disso, o TNF-<font face=\"symbol\">a foi capaz de modular a síntese de melatonina em cultura, promovendo um efeito estimulatório 2h após o inicio da estimulação com NOR, e um inibitório 4h após essa estimulação. Dessa forma, os resultados indicam que produtos na circulação do rato do GT estão envolvidos na modulação encontrada in vivo e que o TNF<font face=\"symbol\">a- é um forte candidato a participar dessa modulação promovida pelo estabelecimento da síndrome da caquexia sobre a produção de melatonina na glândula pineal. / The purpose of this work was to investigate the mechanisms that modify melatonin synthesis in pineal gland during cancer related cachexia and the involvement of cytokines in this context. The results showed an increment of melatonin production in the group inoculated with Walker 256 Tumor (GT) together with a higher activity and gene expression of AA-NAT enzyme, main enzyme that regulates melatonin synthesis. The in vitro study showed that glands from GT produced less melatonin than glands of the control group (GC) after noradrenalin (NOR) stimulation. Looking for a Simplicity Principle in the Perception of Human Walking Motion

The simplicity principle posits that we interpret sense data as the simplest consistent distal cause, or that our high level perceptual representations of stimuli are optimized for simplicity. The traditional paradigm used to test this principle is coding theory, where alternate representations of stimuli are constructed, simplicity is measured as shortness of representation length, and behavioural experiments attempt to show that the shortest representations correspond best to perception. In this study we apply coding theory to marker-based human walking motion. We compare two representation schemes. The first is based on marker coordinates in a body-centred Cartesian coordinate system. The second is based on a model of 15 rigid body segments with Euler angles and a Cartesian translation for each. Both of our schemes are principal component (PC)-based implementations of a norm-based multidimensional object space – a type of model for high level perceptual schemes that has received attention in the literature over the past two decades. Representation length is quantified as number of retained PC’s, with error increasing with discarded PC’s. We generalize simplicity to efficiency measured as error across all possible lengths, where more efficient schemes admit less error across lengths. We find that the Cartesian coordinates-based scheme is more efficient than the Euler angles and translations-based scheme across a database of 100 walkers. In order to link this finding to perception we turn to the caricature effect that subjects can identify caricatures of familiar stimuli more accurately than veridicals. Our design was to compare walker caricatures generated in our two schemes in the hope of finding that one gives caricatures that benefit identification more than the other, from which we would conclude the former to be a better model of the true perceptual scheme. However, we find that analogous caricatures between the two schemes are only distinguishable at caricature levels so extreme that identification performance breaks down, so our design became infeasible and no conclusion for a simplicity principle in walker perception is reached. A variante A é mais agressiva e pode induzir os efeitos sistêmicos como anorexia, retenção de sódio, líguido de perda de peso e morte. Os mecanismos envolvidos na regressão e progressão tumoral são desconhecidos. No presente trabalho, células esplênicas e tumorais de animais inoculados com variantes A e Ar foram isoladas para avaliar a expressão gênica de TGF-ß, IL-12, TNF-a e IFN-? e componentes de apoptose relacionando com anemia, perda de peso, peso do baço, peso do tumor e fragilidade osmótica comparando controles inoculados com Ringer Lactato. Os resultados mostraram que o grupo inoculado com a variante A comparado com a variante Ar, apresenta altos níveis de expressão de TGF-ß em ambas as células esplênicas e tumorais, ausência de expressão de IFN-? e aumento progressivo dos níveis de IL-12 sem infiltrado inflamatório. Esses resultados sugerem que a agressividade da variante A está relacionada á modulação de citocinas, facilitando o crescimento e escape tumoral. A IL-12 pode ser constitutiva nas linhagens A e Ar. A expressão gênica de Bcl-2 foi progressiva e não houve expressão de Fas nas células tumorais do grupo A. Estes resultados demonstram que na linhagem A a imumogenicidade ocorre por outros mecanismos. A expressão de citocinas e componentes de apoptose pode explicar as diferenças das linhagens A e Ar / Abstract: Two variants of this Walker 256 tumor have been previously reported as Walker 256 A and variant Ar. The variant A is more aggressive and can induce systemic effects as anorexia, sodium and water retention, followed by weight loss and death. The mechanisms involved in enhancing tumor regression and progression in this model is still incompletely understood. In the present study, spleen cells and tumor cells from animals inoculated with variants A and AR, were isolated to investigate the TGF-ß, IL-12, IFN-?, TNF-a gene expression, components of apoptosis and relationship with anemia, weight of animals, weight of spleen, height of tumor and osmotic fragility compared with controls inoculated with Ringer Lactate. Results demonstrate that the group inoculated with variant A compared with variant Ar, shows high levels of TGF-ß gene expression in both tumor tissue and spleen cells, no expression of IFN-? and a progressive and higher levels of IL-12 in tumor tissue without inflammatory infiltrate. These results suggest that the aggressively of variant A is relate to cytokine modulation, facilitating the growth and escape of tumor cells. Furthermore, IL-12 seems to be constitutively expressed in both tumor lineage A and AR. The mRNA expression of Bcl-2 was progressive and there is no expression of Fas on tumoral cells of group A. These results demonstrate that on the lineage A the immunogenicity occurs by others mechanisms. This thesis examines the relationship between Southern literature and socio-cultural realities of the Southern region of the United States of America. Analyzing works of five distinguished Southern writers, this thesis examines the reflection of specific Southern culture features in literature of the region in the period from the end of the American Civil War to the second half of the 20th century. The thesis oppose the opinion that the primary goal of Southern literature was to promote Southern identity and its cultural superiority above the North. The central hypothesis, that is being verified by this thesis, is that despite the indisputable contribution of highly recognized Southern writers to building of Southern identity, these authors expressed in their works also often sharp critiques of the social conditions in the South.

Mangled Bodies, Mangled Selves: Hurston, A. Walker and Morrison

Indiana University-Purdue University Indianapolis (IUPUI) / Broken bodies litter the landscape of African American women’s literature. Missing limbs and teeth, paralyzed appendages, lost hair, and deformities appear frequently in the works of authors like Nella Larsen, Zora Neale Hurston, Ann Petry, Dorothy West, Alice Walker, Toni Morrison, Pearl Cleage, and Octavia Butler. While many white authors also include broken bodies in their works, Hemingway’s preoccupation with synecdoche in terms of body parts perhaps being the most notable example, the motif permeates the tradition of African American women’s fiction like no other genre, appearing in the work of almost every major African American woman author. In the case of some authors, Morrison and Walker for example, broken bodies appear in every novel of their corpuses. In fact, every story in Walker’s first collection of short stories, In Love and Trouble: Stories of Black Women, features a broken body. Several questions arise from the ubiquity of this motif in the texts of African American women authors: Where did the motif originate? Why does the motif persist? Do the authors use the motif in the same way? What does the trail of broken bodies reveal about how African American women authors interpret the relationship between body and self? Surprisingly, given the prevalence of the motif and the number of critical comments on one or another text, no critic has essayed a comprehensive examination of the motif in African American literature. While this paper does not have the scope to cover the African American canon as a whole, it will discuss the motif across the works of Zora Neale Hurston, Alice Walker and Toni Morrison.

Construction of a synthetic ribosome using DNA as the building material

This thesis forms part of an ongoing project in the DNA Group to build and operate a synthetic ribosome. We present two synthetic ribosome designs that can be combined with DNA-templated chemistry to generate libraries of functional synthetic small molecules. In Chapter 2 we use the DNA strand displacement technique to construct a mechanism that is capable of moving along a DNA track. We explore ways to control the speed and the driving force of the mechanism, and present a mathematical model of the system. We discuss the ability of the design to incorporate chemically-functionalised DNA strands. In Chapter 3 we use a 2D DNA origami tile as the basis of the synthetic ribosome mechanism. Functionalised DNA strands are arranged on the surface of the tile, and we demonstrate the ability to template reactions between the strands, and discuss the possibility of creating a library of distinct chemical products from a single origami tile.
