Key words: Magnolia solulangeana, secondary metabolites, alkaloids, biological activity. Secondary metabolites of plants are responsible for various biological activities. Alkaloids were described as a potentially suitable for Alzheimer's disease therapy (AD) through their inhibition activities against cholinesterases. Nowadays, these substances are important medicine for AD, therefore a screening of herbal drugs is still a current topic. An alkaloid extract of Magnolia × soulangeana flowers was tested in a preliminary testing on anticholinesterase activity. Because of the promising results, it was chosen for an isolation and identification possible effective alkaloids. The extract was separated by a column chromatography using aluminium oxide and a step gradient elution. Alkaloids were isolated by a repeated preparative thin-layer chromatography. Title: Developing a dancing-motion programme for senior citizens Objectives: The objective of this diploma work was to develop and apply a dancing-motion programme for seniors and to characterize its benefits for senior individuals aged between 60 and 90. Methods: Using quantitative and qualitative methods we evaluated the quality of developing a dancing-motion programme for seniors. We used the method of inquiry with the techniques of survey, observation and group interview. The praxix took place in a chosen retirement home during the time period of three months. Results: The achieved results of my work are described in detail in the result part of my work and summarized in its discussion part. The results of our research showed that although the developing and the application of the dancing-motion programme was positively evaluated by the probational group, its level of difficulty did not quite suit the seniors' psysical abilities and that is why it was found difficult by them. The principle aim of this thesis is to create an unified methodical framework for a smaller Software Testing Department. Furthermore, its aim is to define and describe an important element of testing - the software mistake, define the way of its reporting and finally with the view of it to specify the troubleshooting areas of web applications. The last aim is to find a solution of how to publish this methodology. The aims of this thesis were reached by studying available theoretical findings and applications of principles that are known from notorious and approved methodologies which complexly focus on the software development. These principles were confronted with author's practical experience. Based on these processes, the methodology described above has been originated The contribution of the thesis is in the inner structure of the methodology, summarization of the most important information, application of practical personal experiences and adapting some elements of the methodology according to its usage in a small team. Other strong points are the proposals and recommendations of how to improve the situation on the testing department of a particular company, how to publish the methodology and how to maintain it in the future. The thesis is composed of three main parts. In the first chapter, the essential characteristics of testing, models of the life cycle of software development, sorts and levels of tests are given. The second chapter is the crucial part of the thesis. It describes the whole methodology - the main workflow and its details (processes), activities made during these processes, roles occurring in the methodology and the description of its responsibility (for activities and artefacts), artefacts made on the testing department, full description of mistake and its reporting and finally the summary of mistakes appearing in the web applications environment. The last chapter attends to the way of putting the methodology into operation - technical coverage of particular parts of the methodology in the present time, suggestion of improvement the testing in the future and possibilities of the publication of the methodology.

The financial analysis is elaborated for the needs of the company owner and it covers the period of time 2005-2009. To evaluate the company financial position I use standard methods, e.g. - horizontal and vertical analysis, calculations of profitability, activity, liquidity, productivity of labour and indebtedness. At the end of the thesis I give a complex evaluation of the company and suggest some measures for the financial innovation of the company.

The aim of this thesis is to analyze the structure of the population of the Czech Republic in terms of education and economic activity and then draw out its future development. To this variations of the projection is included the development of economic activity in the population compared to the standard demographic projections. This approach allows better assessment of future development economically active parts of the population by its actual participation in the labor market, and not only by age group, as it is in most demographic projections. This work also includes analysis of sex-education-age structure of the population and analysis the reasons for economic inactivity.

Practical part of the thesis contains analysis of financial statements of the company from 2010 to 2014. This part is focused on horizontal and vertical analysis and also analysis of ratios from categories profitability, activity, liquidity and solvency. Closing chapter covers the summary of findings of the financial analysis.

The aim of the thesis is a complex mapping of the cultural life of national minorities living in the Czech Republic. It focuses on the possible ways of their support by the state as well as local governments and on introduction of these national minorities and presentation of their specific culture. The first part defines the relevant terms and legal framework. Further it describes the possibilities of financial and other support of their cultural activities. Integral part is formed by the characteristics of individual national minorities and description of their cultural activities. The second part shows the analysed possibilities on the example of the Greek minority and existing Greek cultural organizations in the Czech Republic, particularly it analyses activities of the organization Lyceum Řekyň v České republice (Lyceum Club of Greek Women in the Czech Republic). This part of the thesis results above all in the assessment of functioning of the organisation as well as in recommendations for its further development.

Author: Bc. Kateřina Humlová Title: Exercise and physical activities during pregnancy from the perspective of physiotherapist in the Czech Republic. Objectives: The aim of this diploma thesis is evaluate the opinions, experiences and competencies of physiotherapist from the Czech Republic in relation to work with pregnant woman, which were collected during pre- and postgraduate education. Methods: A non-standardized questionnaire was created for the Leeds of the research. The research was taken by 171 probands (physiotherapists of the Czech Republic). The data were evaluated using Microsoft Excel and mathematical software R. Results: In the examined number of physiotherapists in the Czech Republic (n = 171) it was found that over 85 % (n = 146; 85,4 %) of participating physiotherapists rand the evaluation of informatik during undergraduate study and the posibility of working with pregnant woman as insufficient. There were no statistically signifiant differences between the evaluations of respondents from different schools or between the time from graduation. More than 81 % of respondents (n = 139) were interested in pregnancy education. Interest of women dominated over interest of men. Over 71 % of respondents (n = 119) work regurarly with pregnant women and more than 73 % of respondents (n = 126)...

Title: Use of Jr. NBA League programme for the Czech basketball Objectives: The main goal of the thesis is to completely analyze all four seasons of Jr.NBA in Czech Republic and to suggest specific recommendations for organizers. Methods: To achieve the goal of the thesis there were used methods such as primary external data analysis, in-depth interview, online survey, participant observation and SWOT analysis Results: Aspects of communication of the organizational matters and organization of Draft and The Finals are among the strengths of the programme. Weaknesses such as the number of schools and high competitive game requirements for schools participating in the programme are found to be severe. The biggest opportunity of the project is in getting bigger and more intense cooperation of clubs both within the project and in a way of approaching more kids for their youth programmes or after-school activities. Sport events which will be taking place next year or years after can be significantly used for promotion of the project and Czech basketball as well. Possibly the next waves of global pandemic or some other consequences related to Mother Nature can be considered for real threats.

Pohybová aktivita žáků staršího školního věku v době nouzového stavu / Physical activity of pupils of older school age at the time of emergency

The diploma thesis deals with the psysical aktivity of pupils of older school age, ie pupils of the second grade of primary school, during an emergency caused by a coronavirus pandemic. The aim of the work is based on theoretical analysis and the results of our own research to map and compare the programof teaching psysical education in distance form in selected schools. The theoretical part deals with the issue of physical activity, the period of older school age, the state of emergency and the associated excessive sitting and social isolation. The practical part specifically focuses on the content, volume, intensity of physical education, communication between teachers and students in the distance form, but also on leisure physical activity. To find out the results for the practical part, I chose a questionnaire survey of pupils in the second grade od selected primary schools and interviews with physical education teachers. Based on these results, a specific program for teaching physical education in distance for was created.
