Syntéza antimikrobiálních látek na bázi derivátů salicylové kyseliny / Synthesis of antimicrobial compounds based on derivatives of salicylic acid

Schiff bases are widely used both in technical practice and in the field of medicine. For their antimicrobial activities are studied as potential antimacterial and abtifungal drugs. With increasing level of resistence to currently drugs is the development of new substances very intense. Salicylanilides are a group of substances with antimicrobial activity and are also intensively studied. Based on that we were synthesized novel Schiff bases resulting from salicylic acid, respectively salicylanilides. It was concretely prepared five compounds: 5- chloro-N-{4-[phenyl(phenylimino)methyl]phenyl}-2-hydroxybenzamide, 5-bromo-N-{4-[ phenyl(phenylimino)methyl]phenyl}-2-hydroxybenzamide, 5-chloro-N-{4-[phenyl(3- chlorfenylimino)methyl]phenyl}-2-hydroxybenzamide, 5-chloro-N-{4-[phenyl(4- chlorfenylimino)methyl]phenyl}-2-hydroxybenzamide, 5-bromo-N-{4-[phenyl(4- chlorfenylimino)methyl]phenyl}-2-hydroxybenzamide. The reaction proceeded at the boiling point of the solvent, with catalytic amount of p-toluensulfonic acid. Most of the compounds were characterized by IR, H1 NMR, C13 NMR and melting point. The prepared compounds showed E/Z isomers on double bond, it is demonstrated on NMR specters.

In this thesis I will deal with people suffering from diabetes mellitus. I will focus on people who are engaged in or have experience with any physical activity, whether recreationally or competitively performed. The primary objective is to determine the habits and practices before, during and after physical activity. Furthermore, the knowledge people affected by the disease itself. So I am going to record how these people with this disease are able to cope by themselves, and how they can regulate their illness. For example, how long they are engaged in physical activity, how often and when they control thyer blood sugar and the like. To obtain information, use the method of surveys. Subsequently, each question will evaluate every particular. People for the poll I will search primarily on Internet forums for diabetics, sports clubs and diabetic clinics. Keywords: Hippeastrum, Amaryllidaceae, alkaloids, antiproliferative activity, Alzheimer's disease, cholinesterase inhibitors, galanthamine The aim of this diploma thesis was to unify current findings about alkaloids isolated from selected plants of the Hippeastrum genus in the Amaryllidaceae family. The fytochemical characteristics of the examined species was introduced, and a group of alkaloids which were isolated from these plant species was composed. Also, the biological activity was evaluated. Up to date, at least 13 plant species of the Hippeastrum genus were examined from the fytochemical perspective. Out of these species examined, 56 different alkaloids with defined structure were isolated. The isolated alkaloids are divided into several groups based on their structure. Namely, these are lycorine, homolycorine, crinine, galanthamine, narciclasine, tazettine, haemanthamine and montanine structural types. Among the many Amaryllidaceae alkaloids, galanthamine has been given a great amount of attention due the fact that it possesses potent acetylcholinesterase inhibition activity, and is distributed worldwide for the treatment of Alzheimer's disease. One of the interesting compounds is haemanthamine, β-crinine-type of Amaryllidaceae alkaloids, which displays significant in vitro cytotoxic activity against several different types of cancer cell lines. The object of this diploma thesis was to prepare several derivatives of alkaloid haemanthamine, and try to find relationship between structure and biological effect. Účinnost specifických aditiv ve výživě sportovních koní

This thesis deals with the use of specific additives in the diet of sport horses. The theme is focused on the peppermint, and its antioxidant activity in horses. There is experiment conducted in 6 different horses to race, sex and age on antioxidant activity of superoxide dismutase SOD and GPx glutathione peroxidase. During the experiment was 4x took blood sample of horses to determine the activity of antioxidant enzymes in period of feeding without additives and feeding with activity use of additives in the diet. The results of experimental monitoring the activity of SOD, as well as in monitoring the activity of GPx enzyme revealed large individual differences between individual animals, which is typical for horses. In our attempt at SOD activity has not been demonstrated statistical relevance. Conversely, it was with the activity of glutathione, here, the results were statistically significant (P <0.05).

This thesis deals with the physical activity group of seniors who attend senior social organization in the Czech Budejovice. The theoretical part is focused on physical activity and its effects on health and lifestyle in connection with the old man, specifically senior. The work also deals with the process of aging in general and to him belonging symptoms and problems or complications of aging. In conclusion, the theoretical part deals with the care of the elderly, their needs and also the services that are provided for them. In the research was conducted research on physical activity of seniors. Here was performed data collection and analysis of results IPAQ - long (International Physical Activity Questionare) questionnaire and monitoring using a pedometer. Determining the level of physical activity was carried out using a questionnaire IPAQ - long by 120 probands and monitoring the number of steps per day for 1 week. The monitoring was conducted in 30 subjects, using pedometers which are individually adjustable. Data obtained by processing the survey and monitoring were recorded, evaluated and presented in graphs with commentary, discussion then took place and were determined conclusions.

Flavonoidy a antioxidační aktivita červených vín

The literary part of the thesis brings the insight into the issue of free radicals and their impact on the health of the organism. It deals with the specific abilities and the role of antioxidants in the protection against the negative impact of radicals. It also pays attention to flavonoids as major polyphenols in red wines which significantly influence the antioxidative activity, and thus the health benefits of wine. The practical part shows, using the spectrophotometric DPPH and FRAP methods, the comparison of antiradical activity and reducing power in selected samples of red wines made by different technologies of maceration. The results show the direct relationship between the maceration method and the measured values of the antioxidative potential of the red wines. The discussion and the final part of the thesis suggest new questions and possible directions for further research.

Studium antioxidačních látek u hroznových semen

It is known, that grape seeds are rich in significant antioxidant. The issue of dealing with the analysis and comparison of antioxidant components in the seeds of Vitis vinifera in the individual varieties has not been sufficiently studied, yet. In the first part of the thesis is an extensive literature focused on a summary of the work, which study substances in the vine seeds and their effect on human health. Experimental work is focused on the study of antioxidant components in the seeds of Vitis vinifera. The experiment was used extracts of the three "normal" (Frankova, Ryzlink vlašský and Cabernet Moravia) and three interspecific varieties (Nativa, Marlen and Kofranka). Spectrometric, using three fundamentally different methods - DPPH, ABTS and FRAP was determined antioxidant activity . Spectrophotometry was further investigated the content of total polyphenolic compounds and total content of flavanols . Technique of high- performance liquid chromatography with mass spectroscopy were analyzed nine major flavonoids (apigenin, astragalin, hyperoside, isorhamnetin, kaempferol, myricetin, quercetin, quercitrin and rutin) and two procyanidins (procyanidin A2 and procyanidin B1). The highest values of antioxidant reached interspecific varieties Marlen and Nativa, at the least antioxidant components were recorded in the variety Cabernet Moravia. However, in complex evaluation, between studied of varieties were not significant differences.

Vplyv fortifikácie v rôznych štádiách macerácie/fermentácie na antioxidačné vlastnosti vín

Diploma thesis dealt with studying and evaluating the impact of fortification in different stages of maceration/fermentation on antioxidant properties of wine. The literary part of the work was clearly described chapters on various production methods and types of fortified wines, different classes of phenolic substances present in wine and study of antioxidant activity of wine and methods of assessment. In the practical part, were selected three blue grape variety from which it was successively produced 9 samples fortified wines. Time fortification took place in three different stages of maceration. The first was the fortification to the must before the start of fermentation, a second fortification during the fermentation of sugar - fermentation to about 7 % by volume alcohol, and the third fortification after fermentation in young dry wine. Fortification was carried out by adding the calculated amount of spirit in the volume of wine or must, at the same level of spirituosity all the resulting wine samples. After some time joining the distillate was carried out basic analysis of wine samples and determination of the profile of phenolic compounds. Spectrophotometric method was further assessed total content of phenolic compounds, flavanols, anthocyanins and antioxidant activity, which was determined by two different methods. Basic analysis and spectrophotometric measurements were performed in triplicate and the results were averaged. Samples of fortified wines were finally sensory evaluated participation of 12 tasters, and all the results summarized in tables and graphs, statistically evaluated and described in drawing conclusions for practical use.
