Lucie Cahlíková, Ph.D. Title of Diploma thesis: Preparation of lycorine derivatives and their biological activity The plants of the Amaryllidaceae family are one of the most important sources of biologically active alkaloids. Lycorine, a phenanthridine alkaloid, isolated from various species of the Amaryllidaceae plant family, has attracted considerable attention due to its promising biological activities. Specifically, its anticancer properties have been evaluated in vitro and in vivo in various preclinical models of human cancers. Further biological effects manifested by lycorine are: antiviral, antibacterial, antifungal, antiplasmodial, anti-oxidant, anti-inflammatory and insect antifeedant effects, as well as ascorbic acid biosynthesis and RNA inhibitory activity. So far, lycorine was used for preparation of many derivatives by modification of different functional groups in its molecule, and screened for a various biological activities such as anticancer activity, inhibition of cholinesterases, antiplasmodial, antitrypanosomal, antiviral and anti-Trichomonas vaginalis activity. Title of diploma thesis: Alcaloids of Vinca minor (Apocynaceae) and their biological activities The aim of this diploma thesis was to process the assigned fraction isolated from the Vinca minor L. plant and to isolate at least one alkaloid to test its biological activity. Using preparative TLC, the alkaloid was sequentially isolated from the fraction, which was then subjected to a structural analysis. NMR and EI-MS methods were used herein. Thanks to these analyzes, the alkaloid was identified as venoterpine. Furthermore, the alkaloid was prepared to test for biological activity. Thus, the isolated alkaloid was identified as venoterpine and tested for biological activity. The observed inhibitory concentration (IC50) to HuAChE and HuBuChe was more than 500 μM, indicating that venoterpine does not rank as a significant inhibitor of cholinesterase. Likewise, the value of cytotoxic activity, when this activity was found to be greater than 10 μM to colorectal cancer cells as well as to healthy cells, is not significant. Author: Adéla Hodulová Title: Alkaloids of Amaryllidaceae family, genus Nerine Diploma thesis Charles University, Faculty of pharmacy in Hradec Králové, Department of Pharmaceutical botany 2018, 51 p. The aim of this diploma thesis was to summarize up the findings about alkaloids which where isolated from genus Nerine plants of Amaryllidaceae family. There was introduced a botanical characteristic of the phytochemical studied species of the genus Nerine. The most studied species was Nerine bowdenii. All the knowledges about mentioned alkaloids were sum up. Ten species have been phytochemical studied out of the genus Nerine until now. In total, 40 alkaloids were isolated from these plants. Alkaloids which where isolated from the Nerine are divided into several structural types. Anticancer, acetylcholinesterase-inhibitory of the alkaloids were described. The biological activity is connected with their stucture. The most significant anticancer activity was observed in alkaloids from crinine- and lycorine type . Acetylcholinesterase-inhibitory activity was pronounced the most in crinine-type alkaloids. Keywords: Nerine, Nerine bowdenii, Amaryllidaceae, alkaloids

My thesis is focused on the weekly physical activity range of students of the University of South Bohemia and the impact of these activities on their health. The reference set are students of full time bachelor and engineering degree programs of the University of South Bohemia in České Budějovice. Physical activity is defined within the theoretical part of the thesis, and is briefly elaborated beyond its basic classification. The remaining chapters are mostly devoted to the influence of physical activities on the diseases called ?civilization diseases?. Other diseases are also described in the context of physical activity in relation to health. In the practical part of the thesis, the results of physical activities are processed and the health status of the students put into context. The aim is to map the range of physical activities of students of the South Bohemian University, determine their impact on health and to make a connection between the range of these activities and BMI values. The research has shown a correlation between the extent of physical activity and health status. This piece of work could be used as a comprehensive source of information relating to the impact of physical activity on human health, and as motivational material for parts of the population living a sedentary life. The thesis is also a documentation of South Bohemian University students´ physical activity in 2012, and sets out information indicative of their health.

Physical activity has been lately discussed around the whole world mainly because of its impact on human health. Due to huge technical development and current life style it turns out that the society is almost inactive. Physical activity is completely insufficient which brings a lot of negative effects on an individual and their health. The aim of thesis was to study the specialized literature in the field of physical activity and its impact on human health, processing of the given literature in the ´research part´ of my thesis and summary of that literature. Further parts of thesis are devoted to gathering of the necessary data, analyzing the results from IPAQ - long (International Physical Activity Questionnaire) questionnaire and monitoring the results from pedometer. To ascertain the amount of physical activity I opted for the IPAQ-long fulfilled by 120 probed people and also for the monitoring of the number of steps a day for a period of one week. The monitoring was performed on 30 individuals with the help of pedometers. The pedometers were individually set for the given person. According to the information gathered from the questionnaires and monitoring with the pedometer is obvious that some individuals miss physical activity in the field of intensive load and the number of steps in a day. Overall, it is possible to say, that according to the recommendation made by USDHHS, all respondents comply with the standard recommended for and adult individual in case of staying healthy.

The aim of the diploma paper was to test a complete fitness of pupils in the 6th class in the primary school in České Budějovice and to affect forming of the correct carriage of body habit by pointed motion activities. The objects of my research were two groups of probands from the 6th class of the primary school. Altogether, 40 probands were submitted to the research. They were separated to the two equal groups, as the experimental one and as the control one. At the beginning, I tested, found out and documented the complete fitness and mainly I found out the correct carriage of body of the both groups. Inclusion of compensatory and toning exercises for the correct carriage of body into the PE for the six-week period by the exercise plan done by myself followed. I did exercises for strengthening the abdominal and back muscles and exercises for stretching the muscles along the vertebral column with the experimental group of 20 probands. The control group had during the exercise plan ordinary PE lessons by school curriculum led by the PE teacher. The Matthias method was used for the evaluation of the carriage of body and the Ruffier test was used for finding the complete fitness. Everything was written down, compiled and evaluated in charts and graphs for its objectification. The results showed that the pupils in the 6th class in the primary school have nowadays very bad complete fitness and they cannot assume the correct upright posture. Also weakened abdominal muscles and shortened quadrates lumbar muscle were showed up on the most of the pupils.

The theoretical part contains the theoretical notion of physical activity, its importance, cutting, recommendations for school children. It also includes health risks associated with hypokinesis effect of physical activity on company health is lifestyle. Deals with the characteristic physical, psychological social characters child of that age. The notion of sporting leisure, this section works also defined. The practical part is mainly focused on practical implementation of our empirical research, Which is based on theoretical knowledge gained in previous chapters. The first part describes the research sample, methods and procedures of work. Furthermore, an analysis, analysis of the evaluation questionnaire. The last chapters are devoted to the discussion by drawing conclusions questionnaires.
