Kinetika příjmu hydrofobních organických látek v žížale Eisenia andrei v půdách s rozdílným obsahem organického uhlíku / Kinetics of hydrophobic organic compounds uptake in the earthworm Eisenia andrei in soils with different content of organic carbon

This thesis is concerned with the fate of hydrophobic organic pollutants (HOCs) in soil. The theoretical part summarizes the current knowledge about bioavailability, factors influencing the bioavailability and methods for its measurement. Attention is paid to the organic carbon content, which is considered to be one of the key factors and to the influence of microorganisms that can degrade or interact with the substances, thus changing their bioavailable ratio. The practical part of the thesis uses bioaccumulation test with earthworm Eisenia andrei for evaluating the uptake kinetics HOCs in three soils with different organic carbon content. Soils were laboratory contaminated with mixtures of selected HOCs (phenanthrene, pyrene, lindane, p,p'-DDT and PCB 153) and the concentrations of substances in these soils were followed for 217 days. Subsequently, bioaccumulation test was conducted; earthworms were exposed from one to twenty-one days. The collected data were used to model uptake kinetics curves for individual substances. Peak-shaped bioaccumulation were found for pyrene, for lindane a steady-state concentration during exposure was observed, whereas for p,p'-DDT and PCB 153 growing concentrations after twenty days of exposure were detected. The reason for the different behavior of the substances was probably their diverse sorption on the organic material, which together with the different speed of elimination influenced final concentration in earthworms. Phenanthrene was not detected in some samples and therefore the modeling of uptake kinetics was not possible.

Earthworms belonging to oligochaete annelids have been a model for comparative immunology for over 40 years. They possess various defense mechanisms efficiently recognizing and responding to non-self substances. Among these there are molecules with many biological activities including cytolytic, antimicrobial and proteolytic. This work is aimed to compare the immunological features of two closely related earthworm species Eisenia andrei and Eisenia fetida. Due to many morphological and life cycle similarities they have been, until recently, regarded as members of subspecies. Interestingly, their natural habitat varies considerably, and it was of particular interest to investigate how these environmental differences affect the features of innate immunity of both species. A Restauração de Imagens é uma técnica que possui aplicações em várias áreas, por exemplo, medicina, biologia, eletrônica, e outras, onde um dos objetivos da restauração de imagens é melhorar o aspecto final de imagens de amostras que por algum motivo apresentam imperfeições ou borramentos. As imagens obtidas pelo Microscópio de Força Atômica apresentam borramentos causados pela interação de forças entre a ponteira do microscópio e a amostra em estudo. Além disso apresentam ruídos aditivos causados pelo ambiente. Neste trabalho é proposta uma forma de paralelização em GPU de um algoritmo de natureza serial que tem por fim a Restauração de Imagens de Microscopia de Força Atômica baseado na Regularização de Tikhonov. / Image Restoration is a technique which has applications in several areas, e.g., medicine, biology, electronics, and others, where one of the goals is to improve the final appearance of the images of samples, that have for some reason, imperfections or blurring. The images obtained by Atomic Force Microscope have blurring caused by the interaction forces between the tip of the microscope and the sample under study. Moreover exhibit additive noise caused by the environment. This thesis proposes a way to make a parallelization on a GPU of a serial algorithm of which is a Image Restoration of Images from Atomic Force Microscopy using Tikhonov Regularization.

The subject of this thesis is an examination of the acquisition in language of a new country for three Eastern European writers exiled in France. For such writers, art and life become inseparable: just as the experience of geographical displacement liberates the writer so it liberates his language. This new language becomes a field of experimentation, in which the conflicts that precipitated exile are resolved. Departure necessitates the abandonment of the mother tongue: for Cioran, Romanian; for Kundera, Czech; for Makine, Russian. For each of these three writers, studied in this thesis, the adoption of French as the language of literary expression was a decisive act. Geographically and spiritually he and his text are redefined. Separated from familiar landmarks, each finds a new terrain in the language of the creative text, a place, a private space, in which to express the realities of his new self. On the one hand this new paradigm is the expression of a rejection of a past and a tradition; on the other hand it is essential in the process of coming to self-understanding. For Cioran, Kundera and Makine the French language provides a foil to their own ruptured, fragmented, traumatised or guilt-ridden native identities. In each case the adoption of French with its concomitant stereotypical qualities and values constitutes a dialectical process of coming to a clearer sense of self.

For this, a detailed analysis of both works was established as a procedure, highlighting the similarities and, as an intermediate stage, the script of the film to identify the dissimilarities. The theoretical bias used for this work is based on the studies of Haroldo de Campos on transcription, re-creation and poetic translation, as well as the theories of Julio Plaza on Intersemiotic Translation. We will also work on the film elements approached through cinematographic language to perform analyzes and thus better understand the style and poetic optics of Andrei Tarkovski and his other relations with the narrative of the work. At a last moment the analyzes will have as an intention the understanding of the temporal and spatial construction making a parallel between the narratives of the literary work and the cinematographic work. / A presente dissertação tem como objetivo realizar um estudo sobre o processo da tradução escrita para audiovisual entre duas linguagens: literatura e cinema tendo como objeto a novela Piquenique à Beira da Estrada (1972) dos Irmãos Arkadi e Boris Strugatski e sua tradução para o cinema: o filme Stalker (1979) de Andrei Tarkovski. Para isso, estabeleceu-se como procedimento, uma análise detalhada de ambas as obras destacando as semelhanças e como estágio intermediário o roteiro do filme para identificação das dessemelhaças. O viés teórico utilizado para este trabalho, parte dos estudos de Haroldo de Campos sobre a transcriação, recriação e a tradução poética e também as teorias de Júlio Plaza sobre a Tradução Intersemiótica. Serão trabalhados ainda os elementos fílmicos abordados por intermédio da linguagem cinematográfica para realizar análises e assim compreender melhor o estilo e a ótica poética de Andrei Tarkovski e suas demais relações com a narrativa da obra. Em um último momento as análises terão como intuito a compreensão da construção temporal e espacial fazendo um paralelo entre as narrativas da obra literária e da obra cinematográfica.

The central hypothesis is that the Tarkovskian cinematic works were made through the articulation of procedures inherent to the world view model organized through an reverse perspective, an articulation which opened possibilities for aesthetic experimentations associated to the predominant model, organized by the linear perspective. The underlying hypothesis is that the construction strategies in Tarkóvsky s cinema present themselves as a form o resistance to the Soviet cannons of arts and communication. The primary objective is to understand how these world view models connect with the visible world to produce visual messages and in which way such models problematize the concept of realism in the Arts and one of its derivatives, the Socialist Realism. The secondary objective is to discuss the procedures that intersect in time and space in distinct constructions: the Russian Avant-Gardes, medieval painting and Andriêi Tarkóvski s cinema. The final objective is to demonstrate how associative models dynamized the construction of this cinema through the use of the reverse perspective as a procedure, counterposing itself to the cannons established by the Socialist Realism. The theoretical basis of this research lies on the studies of the dialogical processes realized by the Russian theorist Mikhail Bakhtin, especially regarding his concepts of Small and Great Time, Chronotopy and Active Comprehension, in dialog with the manifests and writings of the Russian Avant- Gardes, taking mainly into perspective the conceptual field worked by the Russian thinker Pável Floriênski concerning the Reverse Perspective, and with the thoughts of Gilles Deleuze about the Ritornello. Methodologically, sequences of the seven fulllength features are selected in light of the concepts in which this work is based on. From these sequences the paradigms that trigger the association between the world view models have been highlighted: the potential lines that connect spaces; the formation of multiple centers and the densening of the centres or chronotopical extracts. Finally, this study stands out as it contributes to the problematization of guiding models of the totalitarian aesthetics that direct artistic and communicational creation and others, that direct strategies that aim at resisting artistic and communicational totalitarianism nowadays / Essa tese propõe estudar estratégias de construção no cinema do cineasta russo (soviético) Andriêi Arsiénievitch Tarkóvski, estabelecendo o campo de análise nos sete longas-metragens realizados por ele entre 1962 e 1986. A hipótese central é que a obra cinematográfica tarkóvskiana se fez, a partir da articulação de procedimentos inerentes ao modelo de visão de mundo organizado pela perspectiva inversa, articulação essa que abriu possibilidades para experimentações estéticas associativas ao modelo predominante, organizado pela perspectiva linear. A hipótese subjacente é que as estratégias de construção do cinema de Tarkóvski configuram-se como resistência aos cânones da arte e da comunicação soviéticas. Objetiva-se compreender, primeiramente, como esses modelos de visão de mundo se conectam com o mundo visível para produzirem mensagens visuais e de que forma tais modelos problematizam o conceito de realismo nas artes e seus derivados, o Realismo Socialista. Em seguida, pretende-se identificar e discutir os procedimentos que se intersectam no tempo e no espaço nas seguintes construções distintas: Vanguardas Russas, pintura de ícones medieval e cinema de Andriêi Tarkóvski. Finalmente, demonstrar como modelos associativos dinamizaram a construção desse cinema através do uso da perspectiva inversa como procedimento, contrapondo-se aos cânones estabelecidos pelo Realismo Socialista. Elege-se como campo teórico, o conjunto de estudos dos processos dialógicos na arte realizados pelo teórico russo Mikhail Bakhtin, principalmente os conceitos de Pequeno e Grande Tempo, Cronotopia e Compreensão Ativa, em diálogo com os manifestos e escritos das Vanguardas Russas, privilegiando-se o campo conceitual trabalhado pelo pensador russo Pável Floriênski acerca da Perspectiva Inversa e ainda com o pensamento de Gilles Deleuze sobre o Ritornelo. Metodologicamente são selecionadas seqüências dos sete longas-metragens à luz dos conceitos que embasam esse trabalho. Dessas seqüências destacam-se os paradigmas que desencadeiam a associação entre os modelos de visão de mundo: as linhas potenciais conectivas de espaços; formação de centros múltiplos e o adensamento dos centros ou extratos cronotópicos. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Problemas durante o processo de aquisição de imagens de AFM têm feito com que pesquisas na área de nanotecnologia busquem a utilização de ferramentas para minimizar esses efeitos degenerativos. Neste sentido, foram desenvolvidas ferramentas computacionais de restauração destas imagens degradadas. Neste trabalho é utilizado o método baseado na Regularização de Tikhonov, cuja aplicação está concentrada principalmente em restaurações de imagens biológicas. A proposta deste trabalho é a utilização deste regularizador também em imagens de interesse em engenharia. Em alguns casos, um pré-processamento anteriormente à aplicação do algoritmo, apresenta boa resposta na restauração das imagens. Na fase de préprocessamento foram utilizados alguns filtros como, filtro de média, filtro de mediana, filtro laplaciano e filtro de média pontual. Com a aplicação deste regularizador em imagens foi possível obter perfis de distribuição dos pixels onde é mostrado que na medida em que se aumenta a carga de dissolução de ferro puro em ácido sulfúrico, percebe-se que a razão de aspecto aumenta e características de superfície ficam mais visíveis. / Problems during the process of acquisition of images of AFM have been doing with that research in the nanotechnology searchs the use of tools to minimize those degenerative effects. Computational tools for restoration of these degraded images have developed, in this work the method is used based on Regularization of Tikhonov. In some cases, a previously processing to the application of the algorithm, it presents good answer in the restoration of the images. The previously processing phase some were used filters as, average filter, median filter, laplacian filter and filter of punctual average, besides combination of filters.
