Aplikace spektrálních a nespektrálních analytických metod pro sledování kvality kečupů / Application of spectral and non-spectral analytical methods for monitoring the quality of ketchups

The aim of the master´s thesis was to verify the applicability of elemental analysis and molecular spectrometry for examining the authenticity of ketchup. For verify of share refractometric dry matter introduced into ketchup from tomatoes was used analysis of potassium and lycopene content. Ketchups with different tomato contents were prepared and subsequently models were created, which were used to verify the proportion of refractometric dry matter introduced into tomato ketchup in ketchups from the store. The results of the work showed that the amount of lycopene is a problematic marker for verifying the authenticity of ketchup, because during the production of ketchup, lycopene is degraded.In contrast, the amount of potassium in ketchup has been shown to be a good marker for monitoring the authenticity of ketchup. The model based on the potassium content of ketchup worked reliably, the deviations obtained from the quantity of tomatoes used for the production of ketchup indicated on the packaging were less than 10%. Higher deviations were achieved in ketchups with a minimum permitted tomato content of up to 20%. The concentration of potassium in ketchup with a lower content of used tomatoes can be significantly affected by the addition of other ingredients to the ketchup (onion, garlic, carrot, apple, etc.). In addition to the ketchup production technology used, the ripeness of tomatoes also has a significant effect on the model for verifying the authenticity of ketchup.

Stanovení autenticity potravinářských výrobků s ovocnou složkou / Analysis of autenthicity of food products with fruit component

The aim of this thesis was to determine the authenticity of fruit food for infants using molecular and instrumental methods. In the experimental part, plant DNA isolations from fruit leaves (peaches, apricots, plums and apples) and bananas were performed. Further, DNA was isolated also from five commercial products, and from model mixtures that were prepared in terms of content identical to the commercial mixtures. The isolated DNA was characterized and verified by qPCR with plant DNA-specific ITS2 primers. Three triple primer pairs were selected, and their specificity was evaluated when performing multiplex PCR. This method makes it possible to detect more types of fruit in one reaction, reducing the economic and time requirements for detection. As none of the selected primer pairs were sufficiently specific for the apricot, the evidence from the plum and peach was further realized using duplex PCR. High resolution melting curve analysis was used for better DNA type recognition. Subsequently, agarose gel electrophoresis was performed to analyse the fragment lengths. Furthermore, experiments have been made to identify some specific phenolic substances in commercial and model fruit mixtures by HPLC. Since phenolic substances are degradable under unsuitable storage conditions, the presence of individual compounds was not detected by this method.

For decades, brands and companies have been engaging in Corporate Social Responsibility, which has recently become highly visible through social media. While not brands, SMIs have also started engaging in and creating social responsibility content on social media platforms, such as Instagram. An experimental design study of 421 participants was conducted to examine and compare consumer perceptions. Using self-presentation theory as a framework and experimental design, this study analyzes consumer perceptions of brand and SMI credibility and authenticity when engaging in social responsibility content on social media.

This diploma thesis focuses on validation of RP–HPLC method for determination of organic acids in fruit juices. The aim of this thesis was to determine suitability of the method for verification of the fruit juices authenticity based on analysis of organic acids. The theoretical part describes chemical composition of fruit and fruit juices and focuses on organic acids. It provides description of various methods of organic acids analysis with more detailed description of High Performance Liquid Chromatography and its validation. The experimental part focuses on optimalization and validation of the method. Samples of malic, citric, succinic and ascorbic acid were analysed. The optimalized method was validated and following performance parameters were determined: repeatability, linearity, limit of detection (LOD) limit of quantitation (LOQ). RP–HPLC method was applied to real samples of 100% fruit juices (apple, orange and grapefruit) and the authenticity parameters were verified.

The thesis deals with the issue of authentic or inauthentic work of art in the context of philosophy of authentic / inauthentic mode of existence (Dasein) as it was developed by Martin Heidegger in his Time and Being. The reason for addressing this issue is an effort to open the possibility of understanding authenticity or inauthenticity of individual Dasein in the manner which grasps it in the act of its performance, i.e. as a singular event. Thinking of authentic / inauthentic work of art demonstrates the limits of approach which understands authenticity only as certain originality or "selfness" which should belong to something authentic or inauthentic, almost like its accident. This thesis is therefore concerned with what is revealed if the concept of authenticity is transposed to a work of art. Since it is the subject who can make the act of performance of authenticity happen, the author and the recipient of the authentic / inauthentic work of art are problematized as well. When dealing with the work of art with respect to its temporality (historicality) or its being, the thesis also provides room for the issue of perception of the work of art. The resemblance of the author and the recipient is thus revealed and so is the necessity to learn to see not only artworks as such but to see them...

The work is focused on the evolution of Patočka's conception of movements of the human existence throughout his thoughts. My aim is to show sources of this conception, its explicite expressions and their structure and finally the meaning of this conception for the human life and its status in Patočka's whole-life philosophical project, all this by the analysis of three metodologically defined periods of Patočka's work. The inevitable past of this kind of rethinking of Patočka's conception is dealing with the question of succesion of his two great teachers, Edmund Husserl and Martin Heidegger,and showing, that his conception of movements of the human existence can't be explained simply from the polarity of uncritical bulilding on them, neither the complete refusing them. Also I would like to show the conceptional kinship with other thinkers of western philosophical tradition, whose selection is selective, but in my opinion meaningful. I also hope, that this rethinking of the conception of movements of the human existance will show the place, which belongs to Patočka's thougt in the western philosophical tradition and, first of all, it's meaning for human life. Prague as space, inside of which music of various genres is being created and performed, linked to different values, being created on foundations of distinct concepts and serving as a medium of various strategies, is characterized by fluid and difficult to grasp soundscape (Shellemay 2006). This environment allows the creation and negotiation of traditions (Glassie 1995), moving into different directions. One of them, which have a recursive character and serves its actors as the construction of meaning and carrying out sociocultural change for its actors, is connected with the concept of folklore. The presented fieldwork examines this part of contemporary Prague soundscape in the field, framed by one of the music and dance events. The regular event, named by the organizers "Folklorní mejdlo", defines itself as a connection of the world of modernity, which is currently offered in the metropolis, and the world of traditions, authenticity and folklore. The fieldwork aims its focus on the way the concepts of authenticity and legitimacy are negotiated, its assigning to individual actors and objects and generally how the actors of the event undertake the construction of folklore and how the final product looks like.
