La recherche actuelle est entreprise dans l'optique de la révision complète du design chimique et des principes d'élaboration des photocatalyseurs composites supportés (PCCS) afin d'améliorer considérablement leurs capacités réactionnelles, la durabilité de leur action en temps et de résoudre le problème de pollution secondaire des milieux à traiter. Les travaux effectués ont permis la mise en œuvre d'un nouveau modèle de PCCS interactifs dont les composants photosensibles sont en fortes interactions chimiques avec leurs supports appartenant au groupe d'acides de Lewis. Cette particularité assure une grande sélectivité d'action des produits développés, leur fonctionnement durable en régime stationnaire, évite la pollution secondaire des effluents à traiter et garanti une performance photocatalytique comparable à celle du produit de référence Degussa P25.Les travaux réalisés ont permis d'acquérir de nouvelles connaissances scientifiques concernant le rôle de l'acidité de surface dans l'action photocatalytique et du taux de dissociation de l'eau adsorbée sur le produit actif. De plus, un ensemble de huit paramètres principaux assurant les meilleures conditions d'exploitation des PCCS a été établi.Les PCCS développés peuvent être appliqués, tout d'abord, dans le domaine de l'incinération photocatalytique des COV. Un prototype de filtre dynamique capable de traiter 50 m3/h de l'air avec une consommation d'énergie modérée, est conçu. Un nouveau type de matériaux composites à vocation germicide à base de polymères synthétiques fait également l'objet de la partie applicative de ces travaux de thèse.

Un nouveau type de matériaux composites à vocation germicide à base de polymères synthétiques fait également l’objet de la partie applicative de ces travaux de thèse. / The current research is undertaken in the context of the complete reconsideration of the supported composite photocatalysts (SCPC) chemical design and elaboration principles in order to improve their reaction capacities, to assure the sustainability of their action in time and to resolve the secondary pollution problem for the media to treat. This work enabled the implementation of a new model of interactive SCPC in which the photosensitive components occur in a strong chemical interaction with their supports belonging to the Lewis acid group. This feature provides the developed products with high selectivity, allows them to function in the stationary conditions, avoids the secondary pollution effect and guarantees the photocatalytic performance comparable with the one of the reference industrial photocatalyst Degussa P25. An important scientific knowledge on the role of the surface acidity in the photocatalytic action and on the dissociation state of the water adsorbed over the active product was acquired. Apart from this, a set of eight main parameters ensuring the best SCPC operation properties was established. The developed SCPC are forecasted to be firstly applied in the photocatalytic VOC oxidation processes. A prototype of such filter capable to treat 50 m3/h of air, having modest energy consumption, was designed. A new composite material with germicidal properties elaborated using synthetic polymer supports, also constitutes a subject of the applications part of this thesis.

Bedömning av gasproduktion i returfiber : En studie för att utvärdera alternativ för att inhibera gasproduktionen inom pappersmassaindustrin / Evaluation of gas production in recycled paper pulp

In the pulp production industry one potential problem is the production of gas from microbial activity in the storage tank. If left unchecked such gas production can in extreme cases lead to an explosion. To prevent any such event the current method of maintaining microbial activity in pulp plants is to use hazardous biocides. Such biocides can be expensive and dangerous to handle – therefore a safer and cheaper alternative would be preferred. This study aims to examine and evaluate two potential alternatives: Tannins extracted from trees with natural antibacterial properties and introduction of Lactobacillus plantarum to see if a more manageable bacteria can out-compete the bacteria already in the pulp. The results were measured by observing gas production in different trials where pulp from a nearby plant was mixed with different concentrations of tannins or Lactobacillus. The results showed that this strand of Lactobacillus plantarum had no effect on gas volume production while the pulp mixed with tannins did have some effect by lowering the amount of gas volume produced. However, the experiments with tannins and pulp showed no clear dose-response relationship which most likely is caused by varying quality and handling of the pulp used in the experiments. In summary this study shows that tannins can have an effect on the volume of gas produced by recycled paper pulp by lowering it – although not completely inhibiting it.

Les amibes appartenant au genre Acanthamoeba sont ubiquitaires et responsables de diverses infections, en particulier de kératite amibienne. Par ailleurs elles sont résistantes à de nombreux traitements de désinfection, aussi bien sous leur forme libre (trophozoite) que sous leur forme enkystée. La plupart des données d’efficacité disponibles ont évalué des biocides utilisés pour la désinfection de l’eau potable et/ou des lentilles de contact, mais peu de données sont disponibles concernant le traitement des surfaces ou des matériels médicaux. Ces amibes sont également capables de servir de réservoir à des bactéries pathogènes, notamment des nouvelles espèces de Chlamydia regroupées sous le terme « Chlamydia-like ». Il s’agit de bactéries découvertes récemment et potentiellement responsables d’infections respiratoires et d’avortements spontanés. Malgré leur importance, peu de données sont disponibles concernant la survie et la résistance aux biocides de ces bactéries. L’objectif de ce travail a été dans un premier temps d’évaluer la résistance des amibes aux traitements de désinfections, aussi bien pour les trophozoites que pour les kystes. Dans un second temps, nous avons étudié la survie et la résistance des Chlamydia-like à la désinfection, ainsi que leurs interactions avec les amibes et avec différentes lignées cellulaires ; Chlamydia trachomatis a servi de contrôle dans cette deuxième série d’expérimentations. Les méthodes de culture et d’enkystement des trophozoites ainsi que le choix des souches testées se sont avérés critiques pour l’évaluation des biocides. Certains traitements de désinfection généralement réputés efficaces contre la plupart des micro-organismes ont montré une efficacité limitée vis-à-vis des kystes amibiens ainsi que des trophozoites (glutaraldehyde). Les Chlamydia-like se sont avérées capables de survivre dans l’environnement pendant de longues périodes mais sont globalement sensibles aux désinfectants. Certaines de ces bactéries sont également capables de survivre dans les kystes d’amibes, ce qui peut leur conférer une résistance accrue vis-à-vis des biocides. / Acanthamoebae are ubiquitous amoebae responsible for several infections, mostly amoebic keratitis. They are also resistant to numerous disinfection treatments, as well under their free shape (trophozoite ) as under their encysted shape. Most of the available data of efficiency estimated biocides used for the disinfection of the drinking water and\or the contact lenses, but few data are available concerning the treatment of surfaces or medical devices. These amoebae are also capable of serving as reservoir for pathogenic bacteria, in particular for new species of Chlamydia named " Chlamydia-like ". These new bacteria recently discovered are potentially responsible for respiratory infections and miscarriages. Despite their importance, only few data are available concerning the survival and the resistance to biocides. The objective of this work was at first time to evaluate the resistance of amoebae to disinfection treatments, as well for the trophozoites as for the cysts. Secondly, we studied the survival and the resistance of Chlamydia-like to disinfection, as well as their interactions with amoebae and with various cellular lineages; Chlamydia trachomatis served as control in this second series of experiments. The methods of culture and encystement of trophozoites as well as the choice of selected strains turned out critical for the evaluation of biocides. Some treatments generally considered as effective treatments of disinfection against most of the microorganisms showed an efficiency limited towards the amoebic cysts as well as the trophozoites ( glutaraldehyde ). Chlamydia-like turned out capable of surviving in the environment during long periods but are globally sensitive to disinfectants. Some of these bacteria are also capable to surve in amoebal cysts, what can confer them a resistance increased towards biocides.

La presente attività di ricerca ha avuto come oggetto di studio la formazione di biofilm in condizioni dinamiche: per raggiungere tale obiettivo, è stato costruito un idoneo apparato che permettesse di ottenere una valutazione quantitativa sia delle cellule presenti nella soluzione di crescita sia di quelle adese su tre materiali di prova(acciaio inossidabile, PET e rame). Oltre ad esprimere una valutazione dell’adesione batterica alle superfici, si è proceduto ad indagare sui possibili parametri sperimentali che potessero influenzare la formazione di biofilmdi Listeria su superfici abiotiche. Allo stesso tempo l’efficacia battericida di alcuni biocidi è stata valutata sulle cellule planctoniche così come su biofilm batterici sviluppati su superfici abiotiche. Il ruolo del sistema di quorum sensing basato su luxS nella formazione di biofilm batterici: un ceppo di L. innocua con buone capacità adesive è stato confrontato con il suo ceppo deficiente di luxS per la sua suscettibilità ad agenti biocidi così come per la sua capacità di adesione superficiale L’effetto della precendentemente menzionata inattivazione genetica sulla cinetica di crescita batterica e, quindi, sull’efficienza metabolica complessiva è stata esaminate mediante lo strumento BioscreenC. L’effetto inibitorio di due composti polifenolici è stato valutato sul ceppo deficiente di luxS così come sul ceppo originale. / The current research activity was focused on biofilm formation in dynamic conditions. A suitable device was thus constructed, in order to have a quantitative evaluation of the cells freely suspended in the flowing nutrient solution used as well as the sessile cells which adhered on the three tested surfaces (stainless steel, polyethylene therephthalate and copper). During this project, other than of a quantitative assessment of bacterial adhesion, we investigated also on the parameters which could affect significantly the biofilm formation of Listeria on abiotic surfaces. At the same time antimicrobial efficiency of selected biocides was investigated on planktonic cells as well as on bacterial biofilms grown on abiotic surfaces. The role of luxS-based quorum sensing system in the biofilm development process was evaluated: a luxS-null mutant of an adhesive strain of L. innocua was obtained through Campbell-like genetic inactivation and then evaluated for its sensitivity to biocidal agents and adhesion to surfaces, in comparison with its parental strains. Furthermore the effect of genetic inactivation on bacterial growth kinetic and thus on overall bacterial metabolic efficiency was assessed through the apparatus of BioscreenC. Finally two polyphenolic compounds were investigated for their inhibitory effect on Listeria.

Les circuits de refroidissement des centrales nucléaires en bord de rivière sélectionnent par leur mode de fonctionnement des micro-organismes à caractère thermophile, parmi lesquels le micro-organisme pathogène, Legionella pneumophila. Pour lutter contre le développement de ce genre bactérien, un traitement de désinfection de l'eau des circuits de refroidissement à la monochloramine peut être employé. Pour participer à la maitrise des risques sanitaires et environnementaux liés à la modification physico-chimique et microbiologique de l'eau naturelle prélevée, EDF s'est engagé dans une démarche d'amélioration des connaissances sur l'écologie des légionelles dans les circuits de refroidissement et des liens que ce genre bactérien entretient avec son environnement (physico-chimique et microbiologique) favorisant ou non leur prolifération. Ainsi, la diversité et la dynamique des Legionella pneumophila cultivables ont été déterminées dans les quatre centrales nucléaires en bord de Loire pendant un an et leurs liens avec leur environnement physico-chimique et microbiologique ont été étudiés. Cette étude a mis en évidence une forte diversité des sous-populations de Legionella pneumophila et une apparente dynamique qui semble être liée à l'évolution d'un nombre restreint de sous-populations. Les sous-populations de légionelles semblent entretenir des relations souche-spécifiques avec les paramètres biotiques et présenter des sensibilités différentes aux variations physico-chimiques du milieu. La conception des circuits de refroidissement pourrait impacter la communauté de légionelles. L'utilisation de la monochloramine perturbe fortement l'écosystème mais ne sélectionne pas de populations tolérantes au biocide.

A parte inicial deste estudo consistiu da síntese de HMSPP (Polipropileno com Alta Resistência do Fundido) também chamado polipropileno modificado por irradiação gama, a partir de iPP (polipropileno isotático) em presença de acetileno sob pressão de 110 kPa e irradiado com &gamma; (gama) de fonte de 60Co nas doses de 5; 12,5 e 20 kGy. A fração gel das amostras foi determinada pela extração de componentes solúveis em xileno. A parte solúvel das amostras foi decantada com deposição do gel em lâminas de vidro, até total volatilização do xileno à temperatura ambiente de 25 °C. À parte solúvel da amostra com 12,5 kGy adicionaram-se nanopartículas de prata (NPsAg) nas proporções de: 0,25; 0,5; 1,0; 2,0 e 4,0% em massa. Estas amostras foram caracterizadas por: microscopia eletrônica de varredura e espectroscopia de energia dispersiva (MEV/EDS), espectroscopia no infravermelho (FTIR), microscopia eletrônica de varredura com emissão de campo (MEV-EC), microscopia de força atômica (MFA), calorimetria exploratória diferencial (DSC), difração de raios X (DRX), redução da unidade formadora de colônias (UFC) (%) e teste de citotoxicidade. Neste estudo da morfologia, observou-se a formação de microgéis de polipropileno na ordem crescente PP 5 kGy < PP 12,5 kGy < 20 kGy. Constatou-se a existência de estruturas nanométricas de géis de polipropileno (nanogéis e nanofibras) nas amostras de PP 12,5 kGy e PP 20 kGy. Os nanogéis são formações reticuladas, ramificadas, e emaranhadas, nucleadas em regiões de incidência de alta concentração de energia (spurs) em uma amostra irradiada. Nos testes de avaliação da atividade bactericida dos géis com NPsAg observou-se eficiência biocida para E. coli e S. aureus a partir de 1% de NPsAg e no teste de citotoxicidade as amostras foram caracterizadas como não citotóxicas para células de mamíferos. Em uma segunda etapa deste trabalho foram produzidos filmes da blenda de iPP e PP modificado (50/50) em uma extrusora de dupla rosca. Adicionou-se ao processamento NPsAg nas proporções de: 0,1; 0,25; 0,5; 1,0; 1,0 PVP; 2,0 e 4,0% em massa. Os filmes foram caracterizados por: calorimetria exploratória diferencial (DSC), análise de termogravimetria (TGA), difração de raios X (DRX), microscopia eletrônica de varredura (MEV), espectroscopia de energia dispersiva (EDS), espectroscopia de ultravioleta-visível (UV-Vis), microscopia eletrônica de transmissão (MET), redução da unidade formadora de colônias (UFC) (%), teste de sensibilidade a antimicrobianos por disco de difusão e teste de citotoxicidade. Os filmes analisados apresentaram pontos de aglomeração de prata e regiões com distribuição homogênea das partículas. O efeito bactericida com a interação entre a prata, e E. coli, P. aeruginosa e S. aureus foi constatado para o filme de PP 1%NPsAg PVP Poli(N-vinil-2-pirrolidona). No teste de difusão o resultado obtido para PP 1% NPsAg PVP foi 100% positivo com essas bactérias. Os filmes não são considerados citotóxicos para células de mamíferos. / The study consists of the synthesis of HMSPP (polypropylene with high melt strength), also called polypropylene modified by gamma irradiation from iPP (isotactic polypropylene) in presence of acetylene at 110 kPa pressure and irradiated with &gamma; of 60Co source at doses of 5, 12.5 and 20 kGy. The gel fraction of the samples was determined by extracting in xylene. The soluble portion of the samples was decanted to deposition on glass substrate until complete volatilization of xylene at room temperature of 25 °C. On the soluble portion of 12.5 kGy irradiated sample were added silver nanoparticles (AgNPs) in proportions of 0.25, 0.5, 1.0, 2.0 and 4.0 wt%. These gel samples were characterized by: scanning electron microscopy/energy dispersive spectroscopy (SEM / EDS), infrared spectroscopy (FTIR), scanning electron microscopy with field emission (FE-SEM), atomic force microscopy (AFM), differential scanning calorimetry (DSC), X-ray diffraction (XRD), reduction of colony forming unit (CFU) (%) and cytotoxicity assay. In this study of the morphology, it has been observed the formation of microgels in polypropylene increasing with the dose PP 5 kGy < PP 12.5 kGy < 20 kGy. Nanoscale structures of gels polypropylene (nanogels and nanofibers) were found in samples of PP 12.5 kGy and 20 kGy. The nanogels are formation of crosslinking, branching and entanglement that are nucleated in regions of high energy concentration (spurs) of one irradiated sample. Efficiency in tests of bactericide activity of the gels with AgNPs was observed versus E. coli and S. aureus from 1% AgNPs and non cytotoxicity were characterized in those samples for mammalian cells. In a second stage of this work films of the blend of PP and modified PP (50/50) were produced in a twin screw extruder. AgNPs were added to the extrusion processing in proportions of 0.1; 0.25; 0.5; 1.0; 1.0 PVP; 2.0 and 4.0 wt%. The films obtained were characterized by differential scanning calorimetry (DSC), thermogravimetry analysis (TGA), X-ray diffraction (XRD), scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), ultraviolet-visible spectroscopy (UV-Vis), transmission electron microscopy (TEM), reduction of colony forming unit (CFU) (%), antimicrobial susceptibility testing by disk diffusion and cytotoxicity assay. The films analyzed showed agglomeration of silver points and regions with homogeneous distribution of the particles. The bactericide effect with the interaction between silver and E. coli, P. aeruginosa and S. aureus was found for PP film 1% NPsAg Polyvinylpyrrolidone (PVP). In the diffusion test for PP 1% AgNPs (PVP) was obtained 100%positive result for those bacteria. The films were not cytotoxic to mammalian cells.

Materiais compósitos biocidas baseados em resinas reticuladas contendo nanopartículas de prata / Biocide composite materials based on crosslinked resins containing silver nanoparticles

Nesta dissertação, duas resinas reticuladas comerciais (denominadas, AmberliteGT73 e AmberliteIRC748) foram empregadas para suportar partículas de prata. Os grupos tiol da GT73 e ácido iminodiacético da IRC748 presentes nessas resinas foram empregados para a fixação de íons Ag+ a partir de solução aquosa. Posteriormente, os íons Ag+ foram reduzidos pelo emprego de três redutores diferentes em pH alcalino, denominados hidrazina, hidroxilamina e formaldeído (pH &#61504; 12). A morfologia e a impregnação de prata dos materiais binários assim obtidos foram avaliadas por meio de microscópio eletrônico de varredura equipado com detector de elétrons retro-espalhados (SEM-BSE). O detector de espectrometria de energia dispersiva de raios-X (EDAX) acoplado ao SEM permitiu a observação de partículas de prata. Os espectros de raios-X revelaram a presença do metal nas superfícies interna e externa das microesferas dos compósitos. A quantidade de prata incorporada foi determinada pelo método titulométrico, empregando solução padrão de tiocianato de potássio. As características antibactericidas dos compósitos foram avaliadas em colunas contendo pérolas de resina por onde foram percoladas suspensões da bactéria Escherichia coli auxotrópica AB1157 (tipo selvagem) nas concentrações de 103 a 107 células/mL. A avaliação biocida mostrou que estes materiais foram completamente bactericidas, sendo efetivos na eliminação da bactéria em poucos minutos. Esta ação biocida foi atribuída à combinação da atuação da prata e dos grupos funcionais das resinas / In this work, two different commercial cross-linked resins (namely, Amberlite&#61666; GT73 and Amberlite&#61666;IRC748) were employed for anchoring silver. The SH and the N(CH2COOH)2 groups, respectively present on these resins, were used for the Ag+ chelation from aqueous solution. The Ag+ ions were reduced by employing three different reductants, namely hydrazine, hydroxylamine and formaldehyde (under alkaline pH). The produced composites were characterized by thermogravimetry (TG-DTG) and scanning electron microscopy equipped with backscattered scanning electron detector (SEM-BSE). Energy dispersive X-ray spectroscopy (EDS) coupled to SEM allowed the observation of submicron particles of silver and chemical microanalysis of emitted X-rays revealed the presence of metal on the internal and external surfaces of the composite microspheres. The amount of incorporated silver was determined by titration. The antibacterial activity of the silver/resin composites were determined towards 103 to 107 cells/mL dilutions of the auxotrophic AB1157 Escherichia coli strain, showing that the networks containing anchored submicron silver particles were completely bactericidal within a few minutes due to the combined action of silver and functional groups of the resins

Estas amostras foram caracterizadas por: microscopia eletrônica de varredura e espectroscopia de energia dispersiva (MEV/EDS), espectroscopia no infravermelho (FTIR), microscopia eletrônica de varredura com emissão de campo (MEV-EC), microscopia de força atômica (MFA), calorimetria exploratória diferencial (DSC), difração de raios X (DRX), redução da unidade formadora de colônias (UFC) (%) e teste de citotoxicidade. Neste estudo da morfologia, observou-se a formação de microgéis de polipropileno na ordem crescente PP 5 kGy < PP 12,5 kGy < 20 kGy. Constatou-se a existência de estruturas nanométricas de géis de polipropileno (nanogéis e nanofibras) nas amostras de PP 12,5 kGy e PP 20 kGy. Os nanogéis são formações reticuladas, ramificadas, e emaranhadas, nucleadas em regiões de incidência de alta concentração de energia (spurs) em uma amostra irradiada. Nos testes de avaliação da atividade bactericida dos géis com NPsAg observou-se eficiência biocida para E. coli e S. aureus a partir de 1% de NPsAg e no teste de citotoxicidade as amostras foram caracterizadas como não citotóxicas para células de mamíferos. Em uma segunda etapa deste trabalho foram produzidos filmes da blenda de iPP e PP modificado (50/50) em uma extrusora de dupla rosca. Adicionou-se ao processamento NPsAg nas proporções de: 0,1; 0,25; 0,5; 1,0; 1,0 PVP; 2,0 e 4,0% em massa. Os filmes foram caracterizados por: calorimetria exploratória diferencial (DSC), análise de termogravimetria (TGA), difração de raios X (DRX), microscopia eletrônica de varredura (MEV), espectroscopia de energia dispersiva (EDS), espectroscopia de ultravioleta-visível (UV-Vis), microscopia eletrônica de transmissão (MET), redução da unidade formadora de colônias (UFC) (%), teste de sensibilidade a antimicrobianos por disco de difusão e teste de citotoxicidade. Os filmes analisados apresentaram pontos de aglomeração de prata e regiões com distribuição homogênea das partículas. O efeito bactericida com a interação entre a prata, e E. coli, P. aeruginosa e S. aureus foi constatado para o filme de PP 1%NPsAg PVP Poli(N-vinil-2-pirrolidona). No teste de difusão o resultado obtido para PP 1% NPsAg PVP foi 100% positivo com essas bactérias. Os filmes não são considerados citotóxicos para células de mamíferos. / The study consists of the synthesis of HMSPP (polypropylene with high melt strength), also called polypropylene modified by gamma irradiation from iPP (isotactic polypropylene) in presence of acetylene at 110 kPa pressure and irradiated with &gamma; of 60Co source at doses of 5, 12.5 and 20 kGy. The gel fraction of the samples was determined by extracting in xylene. The soluble portion of the samples was decanted to deposition on glass substrate until complete volatilization of xylene at room temperature of 25 °C. On the soluble portion of 12.5 kGy irradiated sample were added silver nanoparticles (AgNPs) in proportions of 0.25, 0.5, 1.0, 2.0 and 4.0 wt%. These gel samples were characterized by: scanning electron microscopy/energy dispersive spectroscopy (SEM / EDS), infrared spectroscopy (FTIR), scanning electron microscopy with field emission (FE-SEM), atomic force microscopy (AFM), differential scanning calorimetry (DSC), X-ray diffraction (XRD), reduction of colony forming unit (CFU) (%) and cytotoxicity assay. In this study of the morphology, it has been observed the formation of microgels in polypropylene increasing with the dose PP 5 kGy < PP 12.5 kGy < 20 kGy. Nanoscale structures of gels polypropylene (nanogels and nanofibers) were found in samples of PP 12.5 kGy and 20 kGy. The nanogels are formation of crosslinking, branching and entanglement that are nucleated in regions of high energy concentration (spurs) of one irradiated sample. Efficiency in tests of bactericide activity of the gels with AgNPs was observed versus E. coli and S. aureus from 1% AgNPs and non cytotoxicity were characterized in those samples for mammalian cells. In a second stage of this work films of the blend of PP and modified PP (50/50) were produced in a twin screw extruder. AgNPs were added to the extrusion processing in proportions of 0.1; 0.25; 0.5; 1.0; 1.0 PVP; 2.0 and 4.0 wt%. The films obtained were characterized by differential scanning calorimetry (DSC), thermogravimetry analysis (TGA), X-ray diffraction (XRD), scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), ultraviolet-visible spectroscopy (UV-Vis), transmission electron microscopy (TEM), reduction of colony forming unit (CFU) (%), antimicrobial susceptibility testing by disk diffusion and cytotoxicity assay. The films analyzed showed agglomeration of silver points and regions with homogeneous distribution of the particles. The bactericide effect with the interaction between silver and E. coli, P. aeruginosa and S. aureus was found for PP film 1% NPsAg Polyvinylpyrrolidone (PVP). In the diffusion test for PP 1% AgNPs (PVP) was obtained 100%positive result for those bacteria. The films were not cytotoxic to mammalian cells.
