Downy mildew (Plamopara viticola) and bunch rot (Botrytis cinerea) are the most important diseases that occur in this winemaking region. The aim of this work was determinate the intensity of downy mildew in leaves and bunch rot in bunches under different training systems, as well the content of total polyphenols in skin of white varieties. Three experiments were done during 2012/2013 and 2013/2014 cycles, two of them in commercials vineyards with Cabernet Sauvignon, Sauvignon Blanc and Chardonnay in São Joaquim/SC subjected to Y-trellis and vertical shoot positioning trellis (VSP) and the third experiment in a experimental vineyard at Santa Catarina State University (CAV/UDESC) in Lages/SC with Merlot and Cabernet Sauvignon varieties, subjected to Simple Curtin, VSP, Geneva Double Curtin (GDC) and T-Trellis(TT) training systems. Based in the data obtained the temporal dynamic of downy mildew were quantified bi-weekly from the first symptoms appearance under natural conditions on leaves distributed in four medium-height branches on each five replications per treatment. The disease was compared by the beginning of symptoms appearance (BSA); time to reach the maximum disease incidence and severity (TRMDI and TRMDS); maximum value of disease intensity and severity (Imax e Smax) and area under the disease progress curve (AUDPC) were calculated. The data were analyzed by linear regression and adjusted for the models Logistic, Monomolecular and Gompertz. The intensity of B. cinerea were evaluate at the harvest time, in five replicants where 30 randomily clusters were evaluated per treatment. The Y-trellis training system provided higher severity of downy mildew and bunch rot at Cabernet Sauvignon variety. The GDC system induced a high intensity of downy mildew at Merlot. Both white varieties evaluated, showed higher intensity of B. cinerea in Y-trellis when it was compared to VSP. The high concentration of total polyphenols was observed in Chardonnay subjected to Y-trellis system. The Gompertz model showed the best data fit. In conclusion, Y-trellis training system favors the development of downy mildew and bunch rot diseases when right management conditions are not adopted. VSP system showed the best disease control in this conditions and it is recommended to growing grapes to produce fine wines in highlands of Santa Catarina State / Regiões de elevada altitude de Santa Catarina (SC) vem se destacando pela produção de vinhos finos de alta qualidade. O míldio (Plamospara viticola) e a podridão cinzenta (Botrytis cinerea) da videira são as principais doenças que afetam o cultivo de uvas viníferas (Vitis vinifera) na região. O objetivo do trabalho foi avaliar a intensidade de míldio em folha e podridão cinzenta no cacho sob diferentes sistemas de sustentação, bem como o teor de polifenóis totais nas cascas de variedades brancas. Foram realizados três experimentos durante os ciclos 2012/2013 e 2013/2014, sendo dois ensaios em vinhedos comerciais em São Joaquim/SC nas variedades Cabernet Sauvignon, Chardonnay e Sauvignon Blanc sob os sistemas de sustentação espaldeira e ypsilon (Y) e o terceiro ensaio em vinhedo experimental da Universidade do Estado de Santa Catarina CAV/UDESC em Lages/SC nas variedades C. Sauvignon e Merlot sob sistemas de sustentação latada descontínua, Geneva Double Curtin (GDC), espaldeira e cortina simples. A dinâmica temporal do míldio foi avaliada quinzenalmente após o início do aparecimento dos sintomas, sob condições de infecção natural, em folhas distribuídas em 4 ramos medianos marcados aleatoriamente com 5 repetições por tratamento. A epidemia foi comparada em relação ao início do aparecimento dos sintomas (IAS), tempo para atingir a máxima severidade e incidência da doença (TAMID e TAMSD), incidência máxima (Ymax), severidade máxima (Smax) e com os dados obtidos calculou-se as áreas abaixo da curva de progresso da incidência e severidade da doença (AACPID e AACPSD). Os dados foram analisados por regressão linear e ajustados para os modelos Logístico, Monomolecular e Gompertz. A intensidade de podridão cinzenta nos diferentes sistemas foi comparada no momento da colheita, em 5 repetições e 30 cachos aleatórios avaliados por tratamento. O sistema em ypsilon (Y) proporcionou uma maior intensidade de míldio e de podridão cinzenta da videira em relação ao sistema espaldeira, nas formas de manejo adotadas no vinhedo da variedade C. Sauvignon. O sistema GDC apresentou maior intensidade de míldio para variedade Merlot. As variedades brancas apresentaram maior intensidade de B. cinerea no sistema em ypsilon (Y). O teor de polifenóis totais foi superior no sistema ypsilon (Y) para variedade Chardonnay. Dos modelos matemáticos testados o modelo Gompertz foi o modelo que permitiu melhor ajuste dos dados. O sistema em ypsilon (Y) favorece o desenvolvimento do míldio e da podridão cinzenta quando técnicas de manejo adequadas não são aplicadas. O sistema espaldeira mostrou maior controle fitossanitário nas condições adotadas, sendo recomendado para produção de uvas viníferas nas regiões de elevada altitude no Planalto Sul de Santa Catarina

Analyse fonctionnelle du rôle CYP76C2 dans les mécanismes de défense des plantes contre les agents pathogènes / Functional analysis of CYP76C2 in plant defense mechanisms against pathogens

Une analyse du transcriptome d’Arabidopsis thaliana soumis à différents stress biotiques a révélé l’activation de certains membres de la famille CYP76, particulièrement celle de CYP76C2 (≈ 50 fois). La caractérisation fonctionnelle de la famille CYP76, et plus particulièrement celle de CYP76C2 a donc fait l’objet de cette thèse. Après confirmation de l’activation sélective de CYP76C2 en réponse aux pathogènes par qRT-PCR, le phénotype de ses mutants d’insertion et de surexpression a été caractérisé sous différentes conditions d’infection par: Pseudomonas syringae pv. tomato DC3000, P. syringae pv. tomato DC3000 avrRpm1 et par Botrytis cinerea. Afin d’identifier la voie métabolique faisant intervenir CYP76C2, un profilage métabolique ciblé et non ciblé a été entrepris, centré sur le(s) métabolite(s) différentiellement accumulés dans les différents mutants en condition d’infection. Alors que des différences subtiles de sensibilité des mutants de CYP76C2 aux pathogènes semblent confirmer son rôle dans la réponse aux pathogènes, les lignées affectées dans son expression ne présentent pas de phénotypes clairement différents de ceux des plantes sauvages. Une analyse non–ciblée en UPLC-MS (Orbitrap) a permis d’identifier un composé absent dans le mutant cyp76c2 qui pourrait correspondre à un dérivé conjugué en C11, sans que sa structure ne puisse pour l’instant être identifiée (formule brute C17H28O9). CYP76C2 ne semble pas impliqué directement dans la synthèse d’une molécule cruciale pour la mise en place du processus de défense, mais exerce plus probablement une fonction spécialisée ou partiellement redondante de défense ou de détoxication. / A transcriptome analysis of Arabidopsis thaliana subjected to biotic stresses has revealed the activation of members of the CYP76 family, especially of CYP76C2 (≈ 50 times). The functional characterization of CYP76C2, has been the objective of this thesis. After confirmation of the selective activation of CYP76C2 by pathogens, the phenotype of its insertion and overexpressor mutants was characterized under infection by Pseudomonas syringae pv. tomato DC3000, P. syringae pv. tomato DC3000 avrRpm1 and Botrytis cinerea. In order to identify the metabolic pathway involving CYP76C2, targeted and non-targeted metabolic profiling was focused on differentially accumulated compounds in the different mutants after infection. Whereas subtle differences of response of the CYP76C2 mutant lines in response to pathogens seemed to confirm its involvement in response to biotic stress, phenotypes strikingly different from those of wild-type plants were not observed. A non-targeted analysis by UPLC-MS (Orbitrap) identified a compound absent in the cyp76c2 line that may correspond to an oxygenated C11 conjugate (raw formula C17H28O9), but its structure was not identified. CYP76C2 thus does not seem directly involved in the synthesis of a molecule crucial for defense responses, but more likely has a role in the synthesis of a potentially redundant specialized defense compound or in a detoxification process.

L'infection des baies de raisin par le champignon Botrytis cinerea, responsable de la pourriture grise, est fréquente et occasionne des dégâts importants. Pourtant, il semble que la baie dispose de moyens de défense parmi lesquels des protéines PR "Pathogenesis-Related". Chez le Pinot Noir, une chitinase (CHI4D) et une thaumatin-like (TL3) s'accumulent naturellement en grande quantité à partir de la véraison et présentent une activité antifongique contre B. cinerea in vitro. L'objectif de ce travail est de comprendre comment B. cinerea peut se développer sur des baies censées disposer de défenses suffisantes. Pour cela, l'expression spatio-temporelle des ARNm et des protéines CHI4D et TL3 a été suivie respectivement par hybridation in situ et immunohistolocalisation dans les baies, au cours de la maturation, à la suite d'un stress abiotique (UV-C) ou d'un stress biotique (B. cinerea). Dans des baies avant véraison (vertes), n'exprimant naturellement que très faiblement CHI4D et TL3, les ARNm et les protéines s'accumulent en grande quantité après application d'un stress abiotique (UV-C) ou biotique (infection artificielle par B. cinerea). Les protéines CHI4D et TL3 sont localisées au niveau des faisceaux conducteurs ainsi qu'au niveau des tissus proches des sites d'exposition aux UV-C (exocarpe) ou au niveau des sites d'inoculation de B. cinerea, suggérant qu'elles sont impliquées dans la défense de la baie avant véraison. Après véraison, les ARNm et les protéines sont naturellement accumulés au niveau de l'exocarpe et des faisceaux conducteurs qui correspondent à des sites potentiels d'entrée ou de propagation des agents pathogènes. Alors que l'application d'un stress UV-C sur ces baies ne provoque qu'un effet mineur sur l'expression de CHI4D et de TL3, au cours de l'infection par B. cinerea, les quantités de transcrits et de protéines diminuent. A un stade précoce d'infection, la diminution de la quantité des deux protéines est observée en avant du front de propagation du champignon, suggérant une dégradation par des protéases sécrétées par B. cinerea. A un stade d'infection plus avancé, cette diminution s'étend à l'ensemble de la baie. La production hétérologue des protéines CHI4D et TL3 nous a permis de confirmer que CHI4D pouvait être dégradée par des protéases aspartiques sécrétées par B. cinerea. La dégradation de TL3 n'a pas pu être reproduite in vitro. Des tests antigerminatifs effectués in vitro avec les protéines hétérologues n'ont pas permis de mettre en évidence d'effet antifongique malgré la présence d'une activité chitinase pour CHI4D et β-1,3,-glucanase pour TL3. Il est donc possible que ces protéines possèdent des fonctions autres que celles impliquées dans la défense. / Grape berries infection by the phytopathogenic fungus Botrytis cinerea, the causal agent of gray mold, is quite common and causes significant damage. However, it seems that berries have a mechanism of defense, among which are pathogenesis related proteins. In Pinot Noir grape berries, a chitinase (CHI4D) and a thaumatin-like (TL3) protein naturally accrue in large amounts from véraison and show in vitro an antifungal effect against B. cinerea. The aim of this work was to understand how B. cinerea can develop on grape berries that seem to have sufficient defense mechanisms. To do so, the spatio-temporal expression of CHI4D and TL3 mRNAs and proteins in berries was studied respectively by in situ hybridization and immunohistolocalization during maturation, after an abiotic stress (UV-C) or a biotic stress (B. cinerea). Before véraison (green berries) while the expression of CHI4D and TL3 is naturally low, mRNAs and proteins have accumulated in large amounts in berries after UV-C exposition or artificial infection with B. cinerea. CHI4D and TL3 proteins have accumulated around vascular bundles as well as near the sites of UV-C exposition (exocarp) or B. cinerea inoculation, suggesting that before véraison these proteins could be involved in the berry defense. After veraison, mRNAs and proteins naturally accumulate in the exocarp and around vascular bundles that correspond to potential sites of penetration or propagation of pathogenic agents. While the application of UV-C stress on these berries causes only a minor effect on the expression of CHI4D and TL3, during infection by B. cinerea, the amounts of mRNA and proteins decreased. At an early stage of infection, the less amounts of both proteins were observed around the fungus propagation area, suggesting that these proteins could be degraded by B. cinerea secreted proteases. At a more advanced stage of infection, the decrease extended to the entire berry.Production of heterologous CHI4D and TL3 proteins allowed us to confirm that CHI4D could be degraded by aspartic proteases secreted by B. cinerea whereas no degradation of TL3 could be observed in vitro. Both heterologous proteins showed no antifungal effect while a chitinase and a β-1,3-glucanase activities were observed respectively for CHI4D and TL3. It is therefore possible that these proteins have other functions than those involved in the defense.

The ecology of Botrytis cinerea on grape in the Western Cape Province

Thesis (MScAgric)--University of Stellenbosch, 2004. / ENGLISH ABSTRACT: Botrytis cinerea Pers.: Fr., a pathogen of grapevine (Vitis vinifera L.), moves mainly through conidia by air currents in vineyards which are deposited intermittently on the surfaces of leaves, inflorescences and bunches. Little is known about the relationship between the inoculum dosage in air and incidence of Botrytis bunch rot, and how the relationship is influenced by environmental and host factors. To better understand this relationship, information is needed on the period over which conidia have accumulated, the time they are able to survive and remain infectious, time of symptom expression in relation to conidium arrival at the infection court and host surface wetness. The aims of this study were (i) to estimate the amount of viable B. cinerea occurring in air in vineyards, and at different positions on leaves, inflorescences and bunches of grape at different phenological stages, (ii) to determine the relationships between the number of B. cinerea colonies recorded on spore traps placed in the bunch zone of vines and the incidence of B. cinerea recorded from the different tissues, and (iii) to compare the efficacy of fenhexamid on leaves and inflorescences carrying natural B. cinerea inoculum with those inoculated with dry, airborne conidia. Different techniques were used to detect viable Botrytis cinerea in air currents and on plant material obtained from table (cultivars Dauphine and Waltham Cross in Paarl- and Worcester-district) and wine grape (cultivars Chardonnay, Sauvignon Blanc and Merlot in Stellenbosch- and Malmesbury district) vineyards in the Western Cape province during 2001-02 and 2002-03. For four consecutive days during prebloorn, bloom, pea-size, bunch closure, veraison and harvest, sets of Petri dishes with freshly prepared Kerssies' B. cinerea selective medium (spore traps) were left overnight in the bunch zone of vines. Plant material was collected from the vines on the fourth day. Leaves, infloresence and bunches were treated with paraquat to terminate host resistance and to promote the development of the pathogen on the tissues. The B. cinerea inoculum dosage in air, and the incidence at which the pathogen was detected at various positions on leaves and in bunches normally differed between vineyards. However, the various tests revealed that the pathogen generally occurred in a consistent pattern in air in the bunch zone of vines, on leaves and in bunches from all vineyards. The inoculum dosage in air in the bunch zone of the vine was generally highest during prebioom or during bloom, it decreased at pea size and mostly remained at a very low level at the later growth stages. The estimations of viable B. cinerea residing naturally on leaves and in bunches, showed that their amounts depicted levels occurring in air in the bunch zone of the vine. Necrotic leaves occurring early season in vineyards were identified as an important source of secondary inoculum for dispersal to the developing bunches. Latent infections at the various positions in bunches were few at véraison and harvest. However, due to the necrotrophic ability of the pathogen, extensive berry rot (due to berry-to-berry contact) and thus severe bunch rot developed from a single berry that become symptomatic at the base of the pedicel/berry attachment zone. The B. cinerea occupation pattern explains why Botrytis bunch rot develops mostly from the inner bunch and why disease management strategies should concentrate on the bloom to pre-bunch closure stage and on inhibiting B. cinerea development in the inner bunch during the early part of the season. Thus, to effectively reduce B. cinerea in grapevine, preventative applications are recommended to reduce two primary infection events: (a) between budding and pre-bloom to counteract primary leaf infection; (b) during late bloom or early pea size stage, to reduce the amount of the pathogen on leaves and infloresences and to prevent colonisation of floral debris. A third spray can be applied at bunch closure to reduce the amount of B. cinerea at various positions of the inner bunch, especially for cultivars with tight bunches. The efficacy of fenhexamid on leaves and inflorescences carrying natural B. cinerea inoculum was compared with those inoculated with dry, airborne conidia. Shoots were obtained during late bloom from a vineyard (wine grape cultivar Merlot) in the Stellenbosch region. The shoots were divided into two main groups. One group of shoots was left uninoculated, the other shoots were inoculated by dusting with dry B. cinerea conidia in a settling tower. Before inoculation, equal numbers of shoots in each main group was sprayed with fenhexamid, or left unsprayed. Following inoculation and incubation, shoots of each treatment were divided in two equal groups. The one lot of shoots were rinsed in water. The other lot of shoots were immersed in paraquat solution to terminate host resistance and to promote the development of the pathogen from the tissues. For both uninoculated and inoculated shoots, irrespective of fungicide treatment, leaves remained asymptomatic at both the blade and petiole position for the water rinse treatment. No symptom of B. cinerea decay developed at any of the positions on leaves from shoots sprayed with fenhexamid. Spraying of shoots with fenhexamid completely suppressed B. cinerea infection and symptom expression on both uninoculated and inoculated inflorescens. For inoculated shoots, B. cinerea developed from approximately 50% of the laterals in the water rinse treatment. However, inflorescences rinsed in water remained asymptomatic. The laboratory studies showed that fungicides, if applied properly to shoots and bunches under controlled conditions, effectively reduced the amount of B. cinerea at the various positions on leaves and inflorescence, and prevented infection and symptom expression at bloom. However, these goals are not achieved in vineyards where the fungicides are applied by conventional spraying methods. Therefore, more work is needed to evaluate fungicide application techniques by conventional spraying methods for proper fungicide coverage, and the reduction of B. cinerea in bunches. / AFRIKAANSE OPSOMMING: Botrytis cinerea Pers.: Fr., 'n patogeen van druiwe (Vilis vinifera L.), beweeg hoofsaaklik deur middel van konidia in lugstrome deur die wingerd, en word dan afwisselend op die oppervlakte van die blare, bloeiwyses en trosse gedeponeer. Die een groep lote is geïnokuleer deur droë B. cinerea konidia in 'n afsettingstoring te strooi, terwyl die ander groep nie geïnokuleer is nie. Vóór inokulasie, is die helfte van die lote in elke groep met fenhexamid behandel, terwyl die ander helfte onbehandeld gelaat is. Ná inokulasie en inkubasie, is lote van elke behandeling verder in twee eweredige groepe verdeel. Die een groep lote is in water gespoel, terwyl die ander groep lote in 'n paraquatoplossing gedompel is om die gasheerweerstand te verwyder, en die ontwikkeling van die patogeen vanuit die weefsels te bevorder. Vir die waterspoelbehandeling van beide ongeïnokuleerde en geïnokuleerde lote, ongeag van die fungisiedbehandeling, het die blare asimptomaties by beide die bladoppervlakte en blaarsteelposisie gebly. Geen simptome van B. cinerea verrotting het by emge van die blaarposisies van die lote, met fenhexamid gespuit, ontwikkel nie. Die spuit van die lote met fenhexamid het die B. cinerea infeksie en die simptoomontwikkeling op beide die ongeïnokuleerde en geïnokuleerde bloeiwyses heeltemalonderdruk. Laboratoriumstudies het getoon dat fungisiedes, indien korrek toegedien op lote en trosse onder gekontroleerde toestande, tot effektiewe vermindering van B. cinerea getalle by die verskillende posisies op blare en bloeiwyses lei, en infeksie en simptoomuitdrukking tydens blom voorkom. Weens die feit dat die doelwitte nie behaal kan word in wingerde waar die fungisiede deur konvensionele spuitmetodes toegedien is nie, moet meer studies gedoen word om fungisied toedieningstegnieke, by konvensionele spuitmetodes, VIr deeglike fungisiedbedekking en die vermindering van B. cinerea in trosse, te evalueer.

Le fractionnement au pressurage, étape clé de la méthode champenoise, et le type de pressurage, conditionnent de façon significative la composition biochimique du moût et du vin. Le pressurage est fractionné en séparant les premiers moûts extraits, qui constituent la "cuvée", et qui proviennent de la pulpe, partie la plus riche en sucre et en acides (tartrique et malique), des volumes suivants, appelés "tailles", qui sont aussi riches en sucre, en sels minéraux (potassium notamment) et en matières colorantes mais moins acides. Les moûts ont des caractéristiques analytiques bien spécifiques. L’évolution de la composition des moûts au cours du pressurage et la mesure des principaux paramètres analytiques permettant de juger de la qualité de l’extraction ont fait l’objet de quelques études (Valade et Blanck, 1989; Blouin, 1998) sur la base d’un fractionnement volumétrique répondant à un cahier des charges champenois. Toutefois, aucune étude portant sur un large nombre de paramètres, dont les propriétés moussantes, n’avait été entreprise avant ce travail de thèse.Une contamination du raisin par le champignon pathogène Botrytis cinerea (pourriture grise) et l’effet de la maturité du raisin, autres paramètres clés dans l’élaboration du Champagne, ont un impact sur la qualité et la composition des moûts et des vins de base et notamment sur les compositions protéique, polysaccharidique et oligosaccharidique. Les essais ont été réalisés avec 2 pressoirs différents : un pressoir pneumatique industriel (capacité 8000 kg) et un pressoir de laboratoire (capacité 6 kg). Ce travail a été réalisé sur deux cépages : Pinot meunier (millésimes 2013 et 2015) et Chardonnay (millésimes 2014 et 2015). Les analyses suivantes ont été réalisées sur les moûts et vins de base : (i) paramètres œnologiques classiques des moûts et des vins, (ii) isolement et analyse des polysaccharides et oligosaccharides solubles des vins de base, (iii) quantification et identification des protéines solubles des vins de base, (iv) mesure de l’activité protéasique des vins de base (impact de Botrytis cinerea), (v) comparaison de la composition des moûts et des vins issus de raisins sains de deux millésimes : 2013 (pressoir industriel) et 2014 (pressoir de laboratoire).La connaissance de l’état sanitaire et l'optimisation du choix de la date des vendanges en fonction de la maturité sont des outils à la disposition de l’œnologue qui lui permettent d’améliorer la qualité des moûts produits au cours du pressurage et par conséquent celle des vins qui en sont issus. A la suite de cette approche, il serait intéressant de voir dans quelles conditions ces paramètres pourraient être reproduits à grande échelle pour une application industrielle. / Press fractioning is a key step in the Champagne method, and the type of pressing will significantly determine the biochemical composition of the juice and the wine. The first pressed juice obtained in the fractioned pressing cycle, called the “cuvée”, is rich in sugar and acids (tartaric and malic). The second pressed juice, called the “tailles”, is as rich in sugar, mineral salt (potassium in particular) and colorant materials as the first one but less acidic. Must has specific analytical characteristics. The evolution of composition in the must during the pressing cycle and the measure of current analytical parameters, allowing the understanding of grapes extraction, have led to little studies in the Champagne region (Valade et Blanck, 1989; Blouin, 1998). These studies have followed the changes between the Cuvée and the Tailles, according to the rules applied for Champagne production. Nevertheless, no study had considered a large number of parameters, including the foaming properties, before this thesis.The contamination of grapes by the pathogenic Botrytis cinerea (gray mold) and the effect of grape maturity, which are other key parameters in the elaboration of Champagne, have an impact on the quality of must and base wine, especially on proteins, polysaccharides and oligosaccharides. The essays were carried out in two different presses: an industrial automatic press (capacity 8000 kg) and a laboratory press (capacity 6 kg). This study was made on two grape varieties: Pinot meunier (vintages 2013 and 2015) and Chardonnay (vintages 2014 and 2015). The following analyzes were effectuated on must and base wine: (i) determination of current oenological parameters of must and base wine, (ii) isolation and analysis of soluble polysaccharides and oligosaccharides in base wine, (iii) analysis, quantification and identification of soluble proteins in base wine, (iv) quantification of protease activity in base wine (the impact of Botrytis cinerea), (v) comparison of composition of must and base wine from healthy grapes of two vintages: 2013 (industrial press) and 2014 (laboratory press).The knowledge of the sanitary state and the optimization of the harvest date are tools used by oenologist to improve the quality of must, obtained during the pressing cycle, and therefore of wine elaborated from them. Following this study, it could be interesting to examine in which conditions these parameters may be reproduced at a bigger scale for forward industrial applications.

Paraburkholderia phytofirmans souche PsJN est une bactérie endophyte capable d’induire une promotion de croissance de la vigne et de lui conférer une tolérance vis-à-vis des basses températures. Puisque la bactérie PsJN induit également une tolérance de la vigne contre la pourriture grise causée par le champignon Botrytis cinerea, l’objectif de ce travail était de caractériser les mécanismes de la résistance induite par P. phytofirmans suite à une infection par B. cinerea chez la vigne.Nos résultats indiquent que l’inoculation des racines de vitroplants par la bactérie PsJN entraîne des modifications du métabolisme phénolique au niveau des plantules bactérisées. En effet, des tanins d’origine gallique et ellagique sont accumulés au niveau des racines en réponse à PsJN alors qu’une accumulation de lignine a été observée dans la tige et d’anthocyanes dans les feuilles. De même, PsJN induit une résistance contre B. cinerea en potentialisant l'expression de gènes de défense dépendants de l’acide salicylique et de l’acide jasmonique dans les feuilles de vigne. Parallèlement, une accumulation de callose et d’H2O2 a été observée ainsi qu’une meilleure mobilisation des glucides dans les plantules bactérisées, corrélée à une réduction des dommages de l’activité photosynthétique.Suite aux résultats obtenus avec le modèle vitroplant, nous avons optimisé la bactérisation des boutures fructifère et évaluer l’impact de cette bactérie sur la résistance des inflorescences contre B. cinerea. Les résultats présentés dans ce travail permettent d’envisager l’utilisation de la bactérie PsJN comme agent de lutte biologique. / Paraburkholderia phytofirmans strain PsJN is an endophytic beneficial bacterium able to promote the growth of grapevine and to induce the plant resistance to biotic and abiotic stresses. Indeed, the interaction between PsJN and grapevine confers to the grapevine a protection towards cold stress (4 °C), by reducing damages on the photosynthetic system and modulating the carbon metabolism. The PsJN strain also induces a protection against gray mold caused by Botrytis cinerea. In this context, the aim of this work was to better characterize this P. phytofirmans strain PsJN-induced resistance in grapevine against B. cinerea.Our results indicate that the inoculation of in vitro-plantlets roots with PsJN altered the phenolic metabolism in bacterized-plantlets. Gallic and ellagic derived tannins are clearly accumulated in roots of bacterized-plantlets whereas an accumulation of lignin in stems and anthocyanins in leaves was observed. We also showed that PsJN induces a protection against B. cinerea by priming the expression of genes involved in salicylic acid and jasmonic acid pathways. In parallel, an accumulation of callose and H2O2, as well as a better mobilization of carbohydrates in bacterized plantlets were observed. In addition to the defense mechanisms setting up by plants, we demonstrated that PsJN reduces in vitro and in vivo development of B. cinerea, in a dose-dependent manner.Following results obtained with the in vitro-plantlets model, we optimized the protocol of bacterization of grapevine fruiting cuttings by the PsJN strain, and evaluate the impact of the bacteria on to induce the resistance of inflorescences to B. cinerea. Our results provide new insights for the use of PsJN and suggest its commercial use as a biological control agent.

Certains champignons sont très dommageables pour les plantes mais aussi pour la conservation après récolte des produits végétaux. Les restrictions réglementaires concernant l’utilisation des fongicides nécessitent de trouver des solutions alternatives. L'amélioration de la résistance des plantes en utilisant des méthodes physiques, comme les rayonnements UV, peut présenter une alternative prometteuse en association avec d’autres méthodes dans le cadre d’une protection intégrée des cultures. L'objectif de cette thèse a été d’étudier l’effet des rayonnements UV-C sur la sensibilité des plantes aux agents pathogènes. Les rayonnements UV-C ont été appliqués durant la culture, seuls ou en combinaison à des agents de biocontrôle, ou en post-récolte. Les expérimentations ont été menées sur deux espèces végétales : le fraisier et la tomate.Sur fraisier, des traitements UV-C à une dose de 1,70 kJ/m² entraînent une diminution systématique d'environ 20 % de la sensibilité des feuilles à Botrytis cinerea et diminuent jusqu’à 50 % le nombre de feuilles attaquées par Podosphaera aphanis, l’agent causal de l’oïdium. Chez la tomate, la protection des feuilles contre B. cinerea atteint 43 % avec une dose d’UV-C cumulée de 4,0 kJ/m². Ces mêmes traitements UV-C appliqués sur fruits réduisent la sensibilité des fraises vis-à-vis de Rhizopus de 75 % mais aucun effet n’est observé vis-à-vis de B. cinerea. Au contraire, sur fruits de tomate, la sensibilité est diminuée d'environ 63 % contre cet agent pathogène. Parallèlement, des tests biochimiques et gustatifs suggèrent que la qualité des fruits n’est pas significativement améliorée ni altérée par les expositions UV-C. Malgré un effet germicide marqué des rayonnements UV-C sur feuilles de fraisiers, aucun effet bénéfique de l’application de ces rayonnements sur le niveau d’efficacité d’agents de lutte biologique n’a été mis en évidence.Nos observations sont encourageantes pour envisager une utilisation de cette méthode dans un schéma de protection intégrée des cultures. Cependant, une compréhension claire de la façon dont les rayonnements UV-C sont perçus par les plantes et traduits en mécanismes de défense est nécessaire avant que leur utilisation ne soit recommandée sur le terrain. / Some fungi are very damaging during plant cultivation and also in post-harvest storage of products. Regulatory restrictions on fungicide use require alternative solutions. Improving plant resistance using physical methods, such as aplication of UV radiation, can be a promising alternative in association with other methods as part of an integrated pest management strategy. The objective of the present thesis was to study the effect of UV-C aplication on the sensitivity of plants to fungal pathogens. UV-C radiation was applied during plant cultivation, alone or in combination with biocontrol agents, or in post-harvest. Experiments were conducted on two plant species: strawberry and tomato.On strawberry plants, UV-C treatments at 1.70 kJ/m² result in a systematic 20 % decrease in leaves sensitivity to Botrytis cinerea and in a reduction by up to 50 % of the number of leaves infected by Podosphaera aphanis, the causal agent of powdery mildew. In tomato, leaves protection against B. cinerea reaches 43 % with a cumulative dose of 4.0 kJ/m². UV-C treatments applied on fruits reduce the strawberry sensitivity to Rhizopus by 75 % but no effect is observed against B. cinerea. On the contrary, sensitivity against B. cinerea is reduced by 63 % on tomato fruit. Biochemical and taste tests suggest that fruit quality is not significantly improved or altered by UV-C treatment. Despite a marked germicidal effect of UV-C radiation on strawberry leaves, it has no significant effect on the protective efficacy of biocontrol agents.Our observations are encouraging to consider the use of this tool in an integrated crop protection scheme. However, a clear understanding of how UV-C radiations are perceived by plants and their translation into defense mechanisms is necessary before their use is recommended in the field.

O nitrogênio pode ser perdido facilmente no solo, seja por volatilização ou por lixiviação, sendo indicado que seu fornecimento ocorra juntamente com a aplicação de materiais ricos em matéria orgânica, parte no plantio e parte em cobertura. Diante do exposto, objetivou-se com este estudo avaliar o efeito das doses e épocas de aplicação de torta de mamona em cobertura na produção, características físico-químicas e acúmulo de macronutrientes na cultura da couve-flor em manejos orgânicos. Foram realizados dois experimentos um no município de São Manuel, SP, e o outro em Botucatu, SP. Em São Manuel, SP foram estudados 14 tratamentos, em esquema fatorial 3 x 4 + 2, sendo três doses de torta de mamona (2150, 3225 e 4300 kg ha-1), combinadas com quatro parcelamentos em cobertura (¼ + 2/4 + ¼; 1/3 + 1/3 + 1/3 e 1/3 + 2/3 e 2/3 + 1/3), além de duas testemunhas (sem adubação em cobertura e com adubação inorgânica). Em Botucatu, SP, foram 10 tratamentos, em esquema fatorial 3 x 3 + 1, sendo três doses de torta de mamona (1290, 2580 e 3870 kg ha-1) combinadas com três épocas de aplicação em cobertura (única aos 30 ou aos 45 dias após o transplante (DAT) e parcelada aos 30 e 45 DAT), além de uma testemunha (sem adubação em cobertura). Os experimentos foram no delineamento experimental de blocos ao acaso, com quatro repetições. Foram analisadas as seguintes características: ciclo (DAT), número de folhas, diâmetro e altura da inflorescência, massa da matéria fresca da inflorescência e massa da matéria fresca da parte vegetativa (caule e folhas), pH, acidez titulável (AT), ácido ascórbico (AA), sólidos solúveis (SS), índice de maturação (IM)/“Ratio”, teores de açúcares redutores (AR), proteína, massa da matéria seca, teor e acúmulo de macronutrientes na inflorescência da couve-flor. Foram realizadas análises de variância, de regressão, teste de Tukey e de Dunnet. Em São Manuel, obteve-se aumento linear da massa fresca da inflorescência quanto maior a dose de torta de mamona, enquanto para a altura e o diâmetro da inflorescência o efeito foi quadrático, com máximos valores estimados para as doses de 4300 e 3856 kg ha-1, respectivamente. Os fatores não afetaram o teor de sólidos solúveis, entretanto, o aumento das doses elevou os valores de pH, índice de maturação, proteína bruta e teor de açúcares redutores, enquanto reduziu a acidez titulável na inflorescência. A ordem decrescente dos nutrientes acumulados na inflorescência foi K>N>P≈Ca>Mg>S com máximas de 1,99; 1,94; 0,28; 0,27; 0,12 e 0,09 g planta-1. Em Botucatu, a máxima massa fresca da inflorescência foi estimada com a dose de 3015 kg ha-1, enquanto a altura e o diâmetro máximos foram atingidos nas doses de 3785 e 3335 kg ha-1, respectivamente. Os valores de sólidos solúveis e proteína bruta não foram afetados pelos fatores avaliados, enquanto que o aumento das doses de torta de mamona em cobertura elevou os valores de acidez titulável e reduziu o de açúcares redutores, ácido ascórbico e o índice de maturação da inflorescência de couve-flor. A ordem decrescente dos nutrientes acumulados em Botucatu foi K>N>P≈Ca>Mg>S, com máximas de 1,48; 1,03; 0,15; 0,14; 0,08 e 0,06 g inflorescência-1. / Currently, conventional cauliflower cultivation is still predominant; however, cultivation in alternative systems, such as organic, has been growing. Compared to other brassicas, it is the most demanding in fertile soils with a high content of organic matter. Nitrogen and potassium are the most accumulated nutrients in cauliflower. Nitrogen can be easily lost in the soil, either by volatilization or by leaching, and it is indicated that its supply occurs along with the application of materials rich in organic matter, part in planting and part in cover. In view of the above, this study aimed to evaluate the effect of doses and times of application of castor bean cake on yield, physical-chemical characteristics and accumulation of macronutrients in cauliflower crop in organics managements. Two experiments were carried out, one in the municipality of São Manuel, SP, and the another in Botucatu, SP. In the experiment 1 (São Manuel, SP), 14 treatments were studied in a 3 x 4 + 2 factorial scheme, with three doses of castor bean (2150, 3225 and 4300 kg ha-1), combined with four installments (¼ + 2/4 + ¼; 1/3 + 1/3 + 1/3; 1/3 + 2/3 and 2/3 + 1/3), besides two controls (without cover fertilization and with inorganic fertilization). In experiment 2 (Botucatu, SP), 10 treatments were used, in a 3 x 3 + 1 factorial scheme, three castor bean doses (1290, 2580 and 3870 kg ha-1) combined with three installments (one application at 30 or at 45 days after transplanting (DAT) and parceled at 30 and 45 DAT), in addition to one control (no fertilization in coverage). The experiments were in a randomized complete block design, with four replications. The following characteristics were analyzed: cycle (DAT), number of leaves, inflorescence diameter and height, inflorescence fresh matter weight and fresh matter weight of the vegetative part (stem and leaves), pH, titratable acidity (TA), acid ascorbic acid (AA), soluble solids (SS), maturation index (MI) / "Ratio", reducing sugars (RS), protein, dry weight of inflorescence, macronutrient content and accumulation in cauliflower inflorescence . Analyzes of variance, regression, Tukey and Dunnet tests were performed. It was observed that, for most of the characteristics, the best dose ranged between 3.2 and 3.9 t ha-1 of castor bean cake in coverage for both sites, whereas the parceling only had an effect on the cultivation of São Manuel, where 2/3 + 1/3 of the total dose applied at 20 and 40 DAT increased the fresh weight of the inflorescence. In general, the effects were more pronounced in S. Manuel, where soil is poorer in organic matter. Regarding the physico-chemical characteristics, it was verified that only pH was influenced by the splitting in both places. For the experiment carried out in São Manuel, the increase in doses reduced only the titratable acidity, while the other characteristics (SS, pH, RS and MI) increased. On the other hand, in Botucatu, TA increased with increasing doses and AA, RS and MI reduced. The difference in values between the sites was minimal, indicating that for these physico-chemical characteristics, the difference in organic matter content in the soil had little influence. The decreasing order of nutrients accumulated in the inflorescence in São Manuel and Botucatu was K>N>P≈Ca>Mg>S and K≈N>P≈Ca>Mg>S, respectively, with maximums of 1.99; 1.94; 0.28; 0.27; 0.12 and 0.09 g plant-1 in São Manuel and 1.48; 1.03; 0.14; 0.15; 0.08 and 0.06 g inflorescence-1 in Botucatu.

No entanto, o uso contínuo de fungicidas pode promover a seleção de fungos resistentes além de causar a contaminação de ecossistemas. Com o objetivo de encontrar uma solução para esse problema, diversos estudos tem se concentrado na busca de novas alternativas de controle, como por exemplo, as proteínas de plantas com atividades antifúngicas (AFPs). Nesse sentido, uma seleção de peptídeos antimicrobianos de flores, folhas e sementes de espécies do gênero Lippia bem como o isolamento de peptídeos antifúngicos de flores de Lippia sidoides foi o objetivo desse trabalho. Neste trabalho, a purificação e identificação de dois novos peptídeos de aproximadamente 10 kDa e 15 kDa de flores de Alecrim-pimenta (Lippia sidoides) foi descrito. As flores de L. sidoides, depois de secadas em estufa a 25o por cinco dias, foram submetidas à extração de proteínas com solução de HCl 0,1% e NaCl 0,6M, seguido por precipitação com sulfato de amônia (100%). Após a precipitação, o extrato foi dialisado contra água destilada (cut off 1,0 kDa) e liofilizado. A fração rica foi aplicada em cromatografia hidrofóbica Octyl-sepharose, seguida por cromatografia de fase-reversa de HPLC (Vydac C18-TP). Bioensaios com EB, e fração PR in vitro indicaram a inibição do crescimento do fungo Botrytis cinerea. As sequências N-terminal, obtidas por degradação de Edman, seguidas por alinhamento indicam que esses dois peptídeos podem ser classificados com proteínas R NBS-LRR. Esta descoberta pode contribuir, futuramente, para o desenvolvimento de produtos biotecnológicos como drogas antifúngicas e plantas transgênicas com resistência elevada a fungos patogênicos. / One of the major global issues in agriculture could be directly related to the severe production crop losses caused by phytopathogenic fungi, especially when infection affects post-harvest cultures. In this view, the fungus Botrytis cinerea is able to cause gray mold in more than 200 species of plants, being considered a major problem for the agribusiness. The use of fungicides is a primary fungi control method in plants, due to velocity and facility of application. However, the continuous use of fungicides may promote the selection of resistant fungi and also the ecosystems contamination. Aiming to find different solutions to this problem, several studies have focused on the search for new alternatives to fungi control, such as plant proteins with antifungal activities (AFPs). In this view, a selection of antimicrobial peptides from flowers, leaves and seeds from Lippia genus and further isolation of antifungal peptides from Lippia sidoides flowers was focused in this work. In this work, the purification and identification of two novel peptides of approximately 10 kDa and 15 kDa from flowers of rosemary-pepper (L. sidoides) was described. L. sidoides flowers were oven dried at 25 oC for 5 days, following protein extraction with a solution containing 0.1% HCl 0.6 M NaCl, and further ammonium sulfate precipitation (100%). After precipitation the extract was dialyzed against distilled water (cut off 1.0 kDa) and lyophilized. The rich fraction was applied onto an Octyl-Sepharose hydrophobic chromatography, followed by HPLC reversed-phase chromatography (Vydac C18-TP). Bioassays using crude extract and in vitro PR fraction indicated the inhibition of Botrytis cinerea growth. N-termini sequences, obtained by the Edman degradation, followed by alignment indicate that these two peptides can be classified as NBS-LRR R proteins. This discovery may help in a near future to the development of biotechnology products such as antifungal drugs and transgenic plants with enhanced resistance to fungal pathogens.

Étude de l'implication des transporteurs de sucres dans l'interaction entre Arabidopsis thaliana et le champignon nécrotrophe Botrytis cinerea / Role of sugar transporters in the interaction between Arabidopsis thaliana and the necrotrophic fungus Botrytis cinerea

Au cours des interactions plante/agent pathogène, la disponibilité en sucres est un des enjeux majeurs pour les deux partenaires. Il s'établit donc une compétition vis-à-vis des ressources carbonées entre l'agent pathogène hétérotrophe pour le carbone et la plante consommant de l'énergie pour se défendre. Les transporteurs de sucres sont les acteurs moléculaires qui interviennent dans cette compétition et probablement dans le devenir de l'interaction. L'objectif de cette étude est de déterminer l'implication des transporteurs de sucres au cours de l'interaction entre la plante modèle Arabidopsis thaliana et le champignon nécrotrophe Botrytis cinerea.Parmi la famille des transporteurs d'hexoses (STPs) d'A. thaliana, l'expression du gène STP13 est régulée positivement durant l'infection par B. cinerea. Le rôle potentiel de STP13 au cours de cette interaction a donc été étudié à l'aide de plantes transgéniques (Knock-Out et surexpresseur). Le suivi du développement des symptômes et la mesure d'absorption du glucose ont permis de montrer des modifications phénotypiques entre les différents génotypes étudiés. Les résultats indiquent une corrélation entre le niveau d'expression de STP13, le transport de glucose et le développement du champignon, confortant ainsi le rôle de STP13 dans la tolérance face à B. cinerea.Les résultats préliminaires de l'étude du transport de glucose à l'échelle cellulaire montrent une inhibition dans des conditions mimant l'infection. Ces analyses ont été effectuées grâce à un modèle constitué d'une suspension cellulaire d'A. thaliana subissant un traitement éliciteur à partir d'un extrait protéique de B. cinerea.Nous nous sommes également intéressés au transport de saccharose à l'échelle de la plante infectée. Nos résultats suggèrent que l'inoculation par le champignon modifie les flux de saccharose classiquement observés créant ainsi une nouvelle force d'appel. Ainsi, la feuille infectée se comporte comme un nouveau puits. Ces travaux de recherche s'inscrivent dans la nécessité d'une meilleure compréhension des mécanismes de transport des sucres qui permettra à terme d'agir sur les capacités de résistance des plantes vis-à-vis d'agents pathogènes. / During plant/pathogen interactions, sugar availability is one of the major issues for both partners. There is a competition for the same carbohydrates necessary for carbon supply on the pathogen's side and to support the additional energy demand for plant's defense. Sugar transporters are the molecular actors in this competition which is determinant for the final outcome of the interaction. In this study, we characterized the implication of sugar transporters in the interaction between the model plant Arabidopsis thaliana and the necrotrophic fungus Botrytis cinerea.Among the A. thaliana hexose transporter family (STPs), STP13 is induced during B. cinerea infection. A potential role of STP13 in this interaction was investigated using transgenic plants (Knock-Out and over-expressor lines). Disease symptoms characterization and glucose uptake assays showed phenotypical variations between the different genotypes. It seems that STP13 expression, glucose uptake and fungus spreading are correlated pointing to a role of STP13 in tolerance to B. cinerea. Other preliminary results showed an inhibition of the cellular glucose uptake upon condition mimicking B. cinerea infection. These analyses were performed on a model composed of an A. thaliana cell suspension elicited with a proteinaceous extract from B. cinerea.We also studied sucrose fluxes in the whole infected plant. Our results suggest that fungus inoculation modifies the usual fluxes creating a new sink.This study may lead to a better understanding of sugar transport mechanisms to improve plant resistance capacity against pathogens in the future.
