Bakterien Legionella pneumophila orsakar sjukdomen Legionella vilket har ökat i världen de senaste 20 åren och har troligtvis ett ännu större mörkertal. Den frodas i vattenrör mellan temperaturen 25–50 grader och vid installation av varmvattenrör i byggnader måste det tas hänsyn i riskzoner. Ett speciellt riskområde är varmvattenavstick som skapas för att kunna dra vidare vattenrör i framtiden. Där utsätts vattnet för större risk för stillastående och sjunkande temperaturprofil beroende på längd. Med hjälp av COMSOL Multiphysics® Modelling Software har ett teoretiskt ramverk för dimensionering av varmvattenavstick i byggnader skapats. Resultatet visar på hur stor strömningshastighet i vattenflödet, isoleringstjocklek och luftrörslängd har påverkan på hur långt avsticket kan vara utan att understiga kritisk temperatur. Resultatet kan användas som beslutsstöd givet att liknande avstickmodell och flödesprofil används samt om strömningshastighet i röret inte är längre än det i rapporten. / The bacterium Legionella pneumophila causes the disease Legionella which has seen an increase in the world over the last 20 years and likely has an even greater number of unrecorded cases. It thrives in water pipes between the temperature of 25-50 degrees and when installing hot water pipes in buildings certain factors must be taken into account. A special risk area is hot water branches that are created to be able to extend water pipes in the future. There, the water is at greater risk of stagnant and decreasing temperature profile depending on its length. With the help of COMSOL Multiphysics® Modelling Software, a theoretical framework for the dimensioning of hot water distances in buildings has been created. The result shows how the velocity in the water flow, the insulation thickness and the length of the air pipe have a great influence on how long the branch can be without falling below the critical temperature. The results can be used as decision support if one has a similar branch model and flow profile and also given a that a lower flow rate is not used.

Text in Afrikaans / The purpose of the study is to examine the extent and demands volunteers have in a managerial function. Volunteers are responsible for the management of the branches of the ACVV, functioning in a specific environment. The variables of these environments have a definite influence on the functioning of a branch and place specific demands on the members. The group of respondents consisted of chairpersons of branches of the ACVV in the West Coast region. The findings of the empirical studies are that all levels of the environment bring pressure to bear upon volunteers. The demands placed upon them have a bearing on the managerial tasks and have a negative effect on the willingness of volunteers to serve as board members. It is imperative that training and development of volunteers, in welfare organizations, must be ongoing in order to empower and give them confidence for these managerial tasks. / Social Work / M. Arbuscular mycorrhiza in Medicago truncatula

Die arbuskuläre Mykorrhiza (AM) ist eine mutualistische Symbiose, die die Phosphataufnahme und Pathogenresistenz von Pflanzen verbessern kann. In der vorliegenden Doktorarbeit wurde die Rolle der Protonen-pumpenden ATPase MtHA1 für die AM Symbiose in Medicago truncatula untersucht. In MtHA1 Mutanten konnten AM Pilze nur noch verkürzte Arbuskel ohne typische Verzweigungen ausbilden. Dies zeigte sich auch in Expressionsmustern von Genen, die für Proteine in verschiedenen Bereichen der periarbuskulären Membran kodieren. Außerdem waren AM Pilzbesiedelung, die verbesserte Nährstoffaufnahme und die Wachstumsförderung in MtHA1 mutierten Pflanzen reduziert. Die Mykorrhiza-induzierte Resistenz (MIR) wurde näher in M. truncatula Pflanzen untersucht, die von Aphanomyces euteiches infiziert waren, dem Erreger einer Wurzelfäule in Leguminosen. In einem geteilten Wurzelsystem, das eine hohe Expression von Verteidigungsgenen aufwies, unterdrückte ein AM Pilz diese Expression und erhöhte in Folge die Empfindlichkeit für das Pathogen. In Wurzeln von Topfkulturen dagegen konnte eine typische MIR beobachtet werden, die wahrscheinlich auf erhöhter Aktivität der Jasmonat/Ethylen-regulierten Verteidigungsantwort beruht, verursacht durch eine Unterdrückung der Salizylsäuresynthese. Im Ergebnis zeigt diese Arbeit die bedeutende Rolle des Gens MtHA1 für die Bildung und Funktion der arbuskelhaltigen Zellen. Die Mutation des Gens führt zur verminderten Arbuskelverzweigung, reduzierter Phosphataufnahme und Wachstumsförderung in der Mykorrhiza und schließlich zu einer geringeren Gesamtbesiedelung durch den AM Pilz. Genexpressionsanalysen weisen darauf hin, dass unterschiedliche Mechanismen den lokalen und systemischen Wechselwirkungen zwischen AM Pilzen und Pathogenen in der Wurzel zu Grunde liegen. Verschieden physiologische Zustände von geteilten Wurzelsystemen und Wurzeln in Topfkulturen erschweren allerdings einen direkten Vergleich der beiden experimentellen Ansätze. / Arbuscular mycorrhiza (AM) is a wide spread mutualistic symbiosis, which can improve phosphate acquisition and pathogen resistance of plants. In the current Ph.D. thesis the role of a proton pumping ATPase (MtHA1) for the AM symbiosis in Medicago truncatula was investigated. In MtHA1 mutant plants, different AM fungi only developed truncated arbuscules without forming typical hyphal branches, and this phenotype was mirrored by expression patterns of genes for proteins located in different areas of the periarbuscular membrane. AM fungal colonization, improved phosphate uptake and plant growth promotion were reduced in MtHA1 mutant plants. Mycorrhiza-induced resistance (MIR) and the nodule symbiosis were, however, not affected. MIR was further analyzed in the M. truncatula infected with Aphanomyces euteiches which causes a root-rot disease in legumes. In a split root system showing high levels of defense-gene expression, colonization of an AM fungus reduced this expression and in consequence increased susceptibility of the roots for the pathogen. In roots of pot cultures, however, a typical MIR was observed and could be based on the higher activity of jasmonate/ethylene-regulated defense responses due to suppression of salicylic acid biosynthesis. In conclusion, this work shows that the gene MtHA1 encoding a proton pumping-ATPase plays a critical role in the formation and function of arbuscule-containing cells. Expression of the mutated gene results in reduced formation of arbuscule branches. This in turn negatively influences mycorrhizal phosphate uptake, plant growth promotion and overall mycorrhizal colonization of the roots. Gene expression analyses indicate that different mechanisms underlay local and systemic interactions between the mycorrhizal fungus and the root pathogen. The different physiological stages of pot culture and split root system make a comparison of the two experimental approaches, however, difficult.

The theses presents results of experiments with spring barley started in years 2017 and 2018. The experiments were put into practice at Mr Jiří Nepovím's (BSc) place in Opatovice near Světlá nad Sázavou. Laudis 550 was used as the experimental variety of spring barley and the objective of the experiment was to observe the increasing amount of nitrogen and its effect on the resulting quantity and quality. The experimental amounts of nitrogen were set to 60 kg.ha-1, 75 kg.ha-1, 90 kg.ha-1, 105 kg.ha-1 and 120 kg.ha-1. With the increasing amount of nitrogen the crop increased of 0,2 - 0,4 t.ha-1. The share of nitrogen substances in grain increased of 0,2-0,5 %.

To begin with an analysis was made of the rules present in the Federal Constitution, in order to identify the minimum content of the income concept and verification of the compatibility between the Constitution and the pertinent rules contained in the Brazilian Tax Code ( Código Tributário Nacional ). With reference to the structural rule of corporate income tax, we analyzed the calculation basis of the tax, examining at greater depth the negative elements in its composition. Given the need of fixing the content of the vocabulum used in the Federal Constitution for the identification of the elements of the income tax levy event, we resumed the question of the limits imposed on the statutory legislator in the exercise of his taxing power upon producing the institutor laws of this tax, especially in relation to the income , profit , assets and liabilities and legal entity concepts. We verified the existing relationship between the taxpayer and the fact described in the structural rule of income tax on the profits of legal entities, in an effort to identify the limits in the election of the debtor and the corresponding effect on the calculation basis, specifically, in the cases wherein it, the legal entity, applies the material criterion: earn income . We address the case of a legal entity owner of several establishments and the respective legal implication for the purposes of income tax. Finally, always through the application of a systematic method of interpretation, we study the rules that discipline the income earned by a legal entity domiciled in Brazil through a branch based overseas / O objetivo deste trabalho é sistematizar as normas referentes ao imposto sobre a renda incidente sobre os resultados de pessoa jurídica domiciliada no Brasil auferidos através de filial localizada no exterior. Partiu-se da análise das normas presentes na Constituição Federal na busca da identificação do conteúdo mínimo do conceito de renda . Verificou-se a compatibilidade entre a Constituição e as normas pertinentes veiculadas no Código Tributário Nacional. Com referência à regra-matriz do imposto sobre a renda, analisou-se a sua base de cálculo, com aprofundamento sobre os elementos negativos na sua composição. Dada a necessidade de fixação do conteúdo dos vocábulos utilizados na Constituição Federal para a identificação dos elementos da hipótese de incidência do imposto sobre a renda, retomamos o questionamento dos limites impostos ao legislador ordinário no exercício de sua competência tributária ao produzir normas instituidoras desse imposto, especialmente em relação aos conceitos e conteúdos de renda , lucro , patrimônio , pessoa jurídica .Verificamos a relação existente entre o contribuinte e o fato descrito no antecedente da norma jurídica tributária do imposto de renda sobre o lucro da pessoa jurídica, em busca da identificação dos limites na fixação do sujeito passivo, bem como, refletimos sobre a manipulação nessa escolha e o correspondente efeito na base de cálculo, especificadamente, nas hipóteses em que ela, pessoa jurídica, realiza o critério material: auferir renda . Abordamos a hipótese de pessoa jurídica titular de diversos estabelecimentos e a respectiva implicação legal para efeitos do imposto sobre a renda. The aim of the diploma thesis was to determine, which nutritional supplements are selected by athletes of strength and endurance sports branches and what is the reason for their option. The theoretical part characterizes the 8 selected sports sector from point of view of the sport performance structure. There is also mentioned the issue of nutritional supplements, their division, legislation or general recommendations for the use in sport. The last chapter deals with the requirements for the use of nutritional supplements in different periods with respect to the branche of sport. Research part has specifically determined the nutritional strategy for athletes in the field of mentioned food supplements at strength and endurance-oriented sports disciplines by using the questionnaire. The research is supported by an interview with an expert nutritionist and sports through collaboration with the staff of the Centre for Sports of the Ministry of Interior

Financial services institutions invest in self–service technologies for various reasons. These include the demands to rationalise costs and to meet the channel preferences of a „technology- savvy‟ client base. Some advantages of self–service technologies (“SSTs”) include the optimisation of staff activities and faster and improved customer services. Retail banks experience various migration-related costs when migrating customers to an SST environment; in terms of both branch infrastructure and the development of employee skills. Some customers continue to favour face-to-face service interactions, which necessitates an identification and evaluation of the necessary skills required by employees to facilitate this migration process. This study aims to both identify and classify the requisite skills needed by financial services professionals to enable them to migrate customers from physical to electronic service channels; including ATMs. With the appropriate training and competencies, employees can guide customers more effectively through the migration process in a non-judgemental way. This would, in turn, address the lack of self-service technology understanding among customers in the longer term. The lack of support from skilled service employees has, in many instances, led to customers paying higher transactional fees and experiencing inconvenience at physical channels, thereby resulting in overall lower self-service usage. / Business Management / DBL
