O camarão rosa (Farfantepenaeus subtilis) explotado pela pesca industrial na plataforma continental amazônica brasileira possui um ciclo de vida curto, mas complexo, habitando áreas oceânicas, mais ao norte da área de ocorrência, na fase adulta e larval, e áreas estuarinas e lagunares na fase de pós-larva e juvenil. O período de maior intensidade de reprodução se estende de maio a setembro e logo após a reprodução as larvas eclodem e iniciam sua migração para áreas costeiras, passando por diversas fases, onde se assentam e residem principalmente entre junho e outubro. A partir de setembro até janeiro do ano seguinte é maior a intensidade de recrutamento de juvenis às áreas oceânicas, onde passam a amadurecer e, a partir de dezembro, começam a ser capturados pela pesca industrial. A maior abundância da população adulta em termos de biomassa vai de março a agosto quando também se verificam as maiores capturas. As fêmeas crescem mais que os machos e estão presentes sempre em maior proporção nas capturas (61%). Os comprimentos assintóticos foram estimados em 231 mm ( k = 1,6 \'ano POT.-1\') e 205 mm (k = 0,94 \'ano POT.-1\'), para fêmeas e machos respectivamente. A população apresenta taxa de mortalidade natural relativamente elevada, 2,53 \'ano POT.-1\' para fêmeas e 1,83 \'ano POT.-1\' para machos, sendo observadas acentuadas flutuações de recrutamento e abundância, com evidências de que são fortemente governadas pelas condições ambientais. O estoque vem sendo explotado em níveis moderados nos anos recentes (E = 0,45), embora tenha sofrido elevadas taxas de explotação na década de 80, o que levou a uma redução do tamanho da população. O rendimento máximo sustentável, considerado uma média de longo prazo, foi estimado em 4.032 toneladas de cauda por ano, para um esforço de pesca de 19.370 dias de mar. Nos últimos anos, se observa uma tendência de recuperação da biomassa populacional, mas com as oscilações anuais características da espécie. A vazão do rio Amazonas é o fator ambiental que governa com mais intensidade as condições do ambiente costeiro na região e verificou-se que suas flutuações estão correlacionadas a alterações na abundância da população da espécie. Postula-se que o aporte e sobrevivência das larvas e pós-larvas no ambiente costeiro seja influenciada pela intensidade da vazão do rio. O período em que se assentam nos berçários na zona costeira coincide com a estação de vazante do rio, sendo a sobrevivência favorecida por vazões abaixo da média e vice-versa. Portanto, medidas de ordenamento voltadas para o uso sustentável do recurso devem estar associadas ao conhecimento das condições ambientais nesta fase, bem como a estudos sobre a abundância de pós-larvas e juvenis na faixa costeira. / The brown shrimp (Farfantepenaeus subtilis) exploited by the industrial fishery on the continental shelf of the Brazilian Amazon has a short but complex life cyele, inhabiting oceanic areas, at the north of the area of occurrence, during the adult and larval stages, and estuarine areas and lagoons in post-larval and juvenile. The period of highest intensity of reproduction extends from May to September and soon after the hatch, the larvae start their migration to coastal areas, passing through several stages, where they settle and remain resident between June and October. From September to January of the following year the intensity of recruitment to ocean areas is higher, and once there they start to mature and are caught by the industrial fishery from December on. The highest abundance of the adult population in terms of biomass is observed from March to August when the largest catches also occur. Females grow larger than males and are always present in greater proportion in catches (61%). The asymptotic lengths were estimated at 231 mm (k = 1.6 \'year POT.-1\') and 205 mm (k = 0.94 \'year POT.-1\') for females and males respectively. The population has a natural mortality rate relatively high, 2.53 \'year POT.-1\' for females and 1.83 \'years POT.-1\' for males, and pronounced fluctuations in recruitment and abundance are observed, with evidence of being strongly governed by environmental conditions. The stock has been exploited at moderate levels in recent years (E = 0.45), although it has suffered high rates of exploitation in the 80\'s, which led to a reduction in population size. The maximum sustainable yield, considered a long-term average, was estimated at 4,032 ton of tail per year for a fishing effort of 19,370 days at sea. In recent years, it is observed a tendency of recovering of the population biomass, but annual fluctuations are characteristics of the species. The flow of the Amazon River is the main environmental facto r that governs the conditions of the coastal environment in the region and it was found that it is correlated with the fluctuatícn of the brown shrimp population abundance. It is postulated that the uptake and survival of larvae and post larvae in the coastal environment is lnfluenced by the intensity of river flow, The period during which they settle at the nurseries in the coastal zone coincides with the dry season and their survival is favored when the flow of the river is below the average, and vice versa. Therefore, management measures aimed at sustainable use of the resource must be associated with the knowledge of environmental conditions during this phase, as well as studies on the abundance of post-larvae and juveniles in the coastal zone.

The impacts of street lighting on batsDay, Julie January 2017 (has links)
As human population grows and develops, more urban areas are expanding. Urbanisation has many impacts on the natural environment and one understudied pollutant is artificial light at night. The aims of this thesis were to examine the impacts of street lighting on bats and investigate the exposure of British bat species to artificial light at night and explored the mitigation option of part-night lighting. The current exposure of British bat species to artificial lighting was assessed using roost locations and population sizes from a long-term dataset (1997-2012) from the Bat Conservation Trust’s National Bat Monitoring Programme on seven bat species (Eptesicus serotinus, Myotis nattereri, Pipistrellus pipistrellus, P. pygmaeus, Plecotus auritus, Rhinolophus hipposideros and R. ferrumequinum). These data were combined with satellite imagery in roost sustenance zones and home ranges. Bat roosts were found in areas with brighter light levels than random locations for P. pipistrellus, P. pygmaeus and Plecotus auritus. Species that forage around streetlights (P. pipistrellus and P. pygmaeus) had significantly higher light levels in the landscape around their roosts than species which avoid street lit areas (R. hipposideros, M. nattereri and P. auritus). Colony size was negatively correlated with light levels. This study highlights that different species have different requirements in the landscapes around their roosts. To investigate landscapes effects of artificial light at night on the understudied light avoiding species R. ferrumequinum, eight maternity roosts were surveyed to explore the interaction between habitat features and street lighting. At each maternity roost, bat detectors were deployed at 25 paired street lit and dark locations. Street lighting had a significant negative effect on bat activity. Locations closest to the maternity roost had higher bat activity than those further away and road type had a significant effect on bat activity, with the highest bat activity recorded at minor roads compared with A and B roads. These results highlight the large negative impact street lighting can have on bat activity patterns and the need for mitigation. Several mitigation strategies have been suggested to combat the effects of artificial light at night but few have been tested. One of these suggestions is to restrict the hours of lighting through the night, often called part-night lighting. Part-night lighting has been implemented by many local authorities, often switching the lights off after midnight and switching them back on before dusk. To explore the effects of part night lighting on bats, the hourly patterns of activity for R. ferrumequinum were studied. Bat activity was bimodal, with a peak in the first few hours after sunset followed by a smaller peak before sunrise. To capture more than 50% of bat activity during the dark period of the night, street lights would be required to switch off before 11pm. To explore this further, a before-and-after study of part-night lighting was conducted at towns across Devon. Following the conversion from full-night lighting to part-night lighting, switching street lights off at 2 am, there was a significant reduction in P. pipistrellus and a significant increase for P. pygmaeus and Nyctalus noctule activity. Although part-night lighting is not often operational during peak activity periods for bat species, reducing the duration of lighting at night has impacts on activity patterns for several species. This thesis shows that artificial light at night has impacts on bats across the landscapes around their roosts. Artificial lighting has impacts for species in different ways, depending on whether they forage around street lights or avoid street lit areas. For species that avoid street lit areas such as R. ferrumequinum, street lighting can have very significant negative impacts on the availability of areas around their roosts. This highlights the need for conservation measures to reduce impacts of artificial lighting. Although mitigation schemes such as part-night lighting may help to minimize impacts of nighttime lighting, more tailored schemes for bats should devised to achieve greater conservation impacts.

The major emphasis of the work will be emergent literacy, however, application principles can be applied in all subject areas. Parents who desire to expand their child's literacy skills while exposing the child to an arts enriched home environment need ideas and tools developed with supportive research to strengthen and focus their efforts. These tools will greatly expand parental resources by offering several principles as a guide for adapting existing classroom materials for home use. The following Seven Components form the core foundation for adapting pretend play materials to aid the home learning environment curricula: (1) Child as Active Participant (Vygotsky; Piaget). (2) Parent as Role Model for Dramatic Play and Literacy, serves as Facilitator, and Co-player (Haight and Miller; Vygotsky). (3) Physical environment could be anywhere: car, office, or bedroom. (4) Resource material: should be able to use common found objects in home, with limited preparations or expense. (5) Play Content needed to provide elements of literacy: cognitive learning, symbol representation, oral language, self-expression, listening and comprehension (Goodman). (6) Promote creative and fun learning experience with a relaxed, informal atmosphere. (7) Play should be process and discovery oriented, and not for performance (Brown and Pleydell; Tuge). This thesis will also examine the preliminary results of a study for parents and children who applied a curriculum developed with the core components and explored the participants' interest level in such activities. Recommendations for further research will also be made.

La palourde japonaise, Ruditapes philippinarum, a été introduite en France en 1972 suite à une volonté de diversification de la production aquacole des bivalves. A la fin des années 1980, des épisodes de mortalité massive ont été observés dans les parcs vénéricoles du pays des Abers (Finistère, France). La mortalité massive des palourdes a été associée à la maladie de l’anneau brun (MAB), une vibriose dont l’agent étiologique est Vibrio tapetis. Le développement de la MAB en milieu naturel a été associé à la modulation de certains facteurs environnementaux tels que la température ou la présence de ressource trophique. Dans le cadre de ce travail et dans un premier temps, des infections expérimentales de palourdes avec différentes souches de V. tapetis ont été effectuées conjointement à une acclimatation des animaux à deux températures contrastées. L’augmentation de la température de 15 à 22 °C a été associée à de plus fortes activités enzymatiques de la superoxyde dismutase (SOD), impliquée dans le système antioxydant, et de la phénoloxydase (PO), impliquée dans le système immunitaire inné. L’augmentation de la température a également conduit à une diminution de la virulence de Vibrio tapetis. Ensemble, ces résultats permettent d’expliquer la baisse de la prévalence et de l’intensité de la MAB à 22 °C. Dans un second temps, des palourdes sexuellement matures nourries avec deux algues contrastées en termes de composition lipidique ont été infectées par V. tapetis. Si la qualité des microalgues n’a pas conduit à des différences de prévalence et d’intensité de la MAB, le statut de reproduction des animaux a eu une influence sur l’intensité de la maladie. Les réponses métaboliques de R. philippinarum exposée à V. tapetis n’ont pas été influencées par la qualité de la nourriture mais par le développement de la MAB. Ces réponses consistaient en une modulation de l’activité d’enzymes antioxydantes (SOD, catalase, glutathion peroxydase, glutathion réductase,glutathion-S-transférase) et d’enzymes reliées au système immunitaire inné (PO et oxyde nitrique synthase inductible) en fonction de la présence de signes cliniques de la MAB. Finalement, l’utilisation de ces indicateurs biochimiques pourrait permettre d’établir des critères de sélection d’individus résistants à la MAB. / The Manila clam, Ruditapes philippinarum, was introduced in France in 1972 following the willingness of bivalve aquaculture diversification. In the late 1980s, episodic mass mortality events were observed in ponds of the “Pays des Abers” region (Finistère, France). The massive mortality of clams was associated brown ring disease (BRD), a vibriosis which causative agent is Vibrio tapetis. BRD development in field has been associated with the modulation of environmental factors such as temperature or the presence of trophic resource. Firstly in the frame of the present work, experimental infections of clams with different strains of V. tapetis were performed together with animal acclimation at two contrasted temperatures. The increase of temperature from 15 to 22 ° C was associated with higher enzymatic activities of superoxide dismutase (SOD), involved in the antioxidant system, and the phenoloxidase (PO), involved in the innate immune system. Temperature increase also led to a decrease in virulence of V. tapetis. Together, these results might explain the decline in BRD prevalence and intensity observed at 22 ° C. Secondly, sexually mature clams fed with two microalgal diets contrasted in terms of lipid composition were infected with V. tapetis. Although microalgae quality did not lead to any difference in BRD prevalence and intensity, the reproductive status of clams influenced BRD intensity. Metabolic responses of R. philippinarum exposed to V. tapetis were not influenced by the food quality but mainly by BRD development. These responses consisted in a modulation of the activity of antioxidant enzymes (SOD, catalase, glutathione peroxidase, glutathione reductase, glutathione S-transferase) and enzymes related to innate immune system (PO and inducible nitric oxide synthase) according to the presence of BRD clinical signs. Finally, the use of these biochemical indicators could allow for new criteria for selection of BRD resistant clams.

Decay Fungi from New Zealand Leaky Buildings: Isolation, Identification and Preservative ResistanceStahlhut, Dirk January 2008 (has links)
Leaky buildings are those that show elevated moisture contents of the framing timber, which can subsequently lead to the establishment of fungal and bacterial decay. Prior to this study, the causative agents of the decay in these leaky buildings were unknown, though it was suspected to be one or more species of decay fungi. Therefore, the overall goal of this multi-disciplinary PhD thesis research was to determine the causative agents of decay in leaky buildings of New Zealand in an effort to develop solutions for both their remediation and future prevention. Use of molecular biology methodology and classical mycological techniques based on morphology enabled identification of decay fungi from framing timber and air samples of leaky New Zealand buildings and provided insight into relative importance based on isolation frequency. In most cases, fungi colonising Pinus radiata D. Don were isolated to produce pure cultures. Mycelia from these cultures on agar media were collected to extract DNA. To identify the fungi to the species level, polymerase chain reaction (PCR) with fungal specific DNA primer pairs were performed followed by DNA sequencing of the internal transcribed spacer (ITS) region. Identification was by BLAST (Basic Local Alignment Search Tool) search on sequences in known GenBanks. In total, 421 samples from leaky buildings were processed, predominately untreated P. radiata decayed framing timber and also fibre cement boards and building paper. From these, sixty-eight fungal identifications were made. The only taxa that were isolated with significant frequency were identified as 4 basidiomycete species, as follows, along with the number of times they were isolated from the 421 samples: • Gloeophyllum sepiarium (Wulf.: Fr.) Karst. 13x • Oligoporus placenta (Fries 1865) Gilb. In Ryv.1985 11x • Antrodia sinuosa (Fr.) Karst. 8x • Gloeophyllum trabeum (Fr.) Murr. 4x Although these species were identified repeatedly, in total they represent less than 10% of the total samples and, therefore, it is concluded that the leaky building decay samples represent high fungal biodiversity. An aerial spore study of internal air, wall cavity air and exterior air of leaky buildings was carried out using a Merck MAS-100 instrument which collects spores directly onto selective media plates. Viable fungal aerial spores were detected at every sampling location tested at the leaky buildings, by the criteria of culturing, with a highest mean of 3714 colony-forming units (CFU) per cubic metre found in the cavities of water-damaged walls. This aerial spore study in conjunction with isolation from decayed wood samples from the same leaky buildings enabled identification of G. sepiarium and A. sinuosa at the same test site. The use of carboxymethylcellulose medium further demonstrated the presence of potential cellulose-degrading fungi within and around the location. Overall, the combination of direct sampling of timber and air sampling proved useful for detection of fungal species variability at a multi-unit building. Four decay fungi isolated from New Zealand leaky buildings and two standard control decay fungi (Coniophora puteana and Serpula lacrymans) were submitted to laboratory wood block testing to determine the effectiveness of currently used wood framing preservatives under laboratory conditions before and after a standard leaching regime. P. radiata blocks were treated with water based boron copper azole and solvent based IPBC propiconazole plus tebuconazole (1:1) preservatives and exposed to the basidiomycetes for 12 weeks. Mass loss for the fungal decay-infected samples was recorded of up to 55% for preservative-treated samples, up to 62% mass loss for leached samples and up to 58% mass loss for un-preservative treated samples. Additionally, well defined dosage responses and approximate toxic thresholds were obtained for all preservatives tested. Results suggested that the minimum IPBC retention specified by Hazard Class 1.2 of NZS3640:2003 (0.025% m/m) is on the low side, and demonstrated after the 2 week leaching regime complete loss of efficacy of boron at 0.4% m/m boric acid equivalent (BAE). This PhD research gave a first overview of fungi occurring in New Zealand leaky buildings, and it demonstrated the following key aspects of wood preservation: 1. The isolated test fungus Antrodia sinuosa was more difficult to control with propiconazole plus tebuconazole at retention 0.007% m/m than the known tolerant fungus Oligoporus placenta; 2. Boron at Hazard Class 1.2 retention of 0.4% m/m BAE was not toxic to Oligoporus placenta; 3. Serpula lacrymans exhibited tolerance to the highest retention of 0.06 %m/m tebuconazole plus propiconazole; and 4. Gloeophyllum species appeared susceptible to all wood preservatives. In order to correlate fungal colonisation and wood decay, colonised wood blocks were studied using light microscopy (LM) and field- emission scanning electron microscopy (FE-SEM). Microscopic observations of P. radiata wood blocks following a standard wood decay test of twelve weeks of fungal colonisation by Serpula lacrymans, Antrodia sinuosa, Oligoporus placenta and Gloeophyllum sepiarium revealed that the two microscopic techniques employed were complementary by allowing features such as pit membranes, chlamydospores or S3/S2 compound middle lamella interface to be photographed in greater detail, allowing for more precise analyses and interpretation of key findings, as follows: 1. Brown rot fungi directly target their apical growth towards degraded pit apetures; 2. Reliance on light microscopy and observed birefringence as a tool to record changes in cell wall crystallinity associated with brown rot decay alone could be misleading; 3. Presence of fine (≤ 1 m) to wide (≥ 3.5 m) bore-hole and hyphal size ranges, and nearly unchanged cell wall thickness of all wood/test fungal combinations, confirmed active decay at moderate to late stages; 4. Some ray parenchyma cells for Antrodia sinuosa, Oligoporus placenta and Gloeophyllum sepiarium colonised blocks were intact throughout late stages of decay, outlining that they were not preferentially degraded early in the brown rot decay process, and 5. Presence of bore-holes, clamp and medallion clamp formation and resting spores (chlamydospores and arthrospores) are fungal specific, can aid in their differentiation and identification, and should be recorded during wood decay studies, as especially resting spores are an important factor when planning remediation strategies. In summary, this PhD thesis research provided the first comprehensive investigation into the biodiversity of fungi from leaky New Zealand buildings, identified the dominant species and presented details about their micromorphology and their decay patterns. It also demonstrated substantial differences in efficacy of preservative formulations currently (December 2008) approved for framing treatments in New Zealand and possible deficiencies where framing may be subjected to severe leaching. This study also provided the first comparative analyses of viable fungal aerial spores between leaky wall cavities and the surrounding air environment. Subsequently, this research added to the knowledge of the decay fungal species diversity in and around New Zealand leaky buildings, outlined their capabilities to degrade treated and un-treated P. radiata framing timber and illustrated the efficacy of New Zealand approved wood preservatives for their potential as remedial treatment and future prevention.

Odour plays an important role in many predator/prey interactions. In the northern hemisphere, many mammalian prey species have been shown to respond to predator odours. It is also widely assumed that mammalian predators utilise odours to locate their prey. This thesis explores the importance of odour in Australian mammalian predator/prey interactions. Responses of native Australian species to the faecal odour of two predators; the native tiger quoll Dasyurus maculatus and the introduced red fox Vulpes vulpes, were evaluated through live-trapping and focussed behavioural studies of captive animals. Tiger quoll responses to prey olfactory cues were investigated in a captive experiment. Native rodents (bush rats Rattus fuscipes, swamp rats R. lutreolus and eastern chestnut mice Pseudomys gracilicaudatus) equally avoided traps scented with either quoll or fox faeces, and in captive experiments, bush rats and swamp rats reduced their average speed in response to both predator odours. Of the marsupial species, northern brown bandicoots Isoodon macrourus and common brushtail possums Trichosurus vulpecula were captured more frequently in quoll-scented traps than unscented traps or foxscented traps, while captures of brown antechinus Antechinus stuarttii, long-nosed bandicoots Perameles nasuta and southern brown bandicoot I. obesulus were unaffected by the either predator odour. In captive experiments, brown antechinus, long-nosed and northern brown bandicoots decreased their foraging in response to both predator odours, and spent less time in areas scented with quoll faeces. Tiger quolls didn't appear to detect odour sources from a distance of >65 cm, but they did follow scent trails and spent more time in areas scented with the urine and faeces of potential prey. Chemical analysis revealed no common components in fox and quoll odour which prey species could be responding to. Therefore, these native species have evolved to respond to fox odour since foxes were introduced to Australia 130 years ago. The stronger response of native rodents to fox odour may be a legacy of their co-evolution with canid predators prior to entering Australia. A better understanding of how odour is utilised in Australian predator/prey interactions may lead to a greater ability to protect Australia's unique mammalian fauna from introduced predators.

The solvent-induced swelling behaviour of Victorian brown coals was examined in detail to probe the bonding mechanisms in very low rank coals (in this case Victorian brown coal). Correlation of solvent properties with differences in observed swelling behaviour were interpreted in terms of the coal structure, and means of predicting the observed behaviour were considered. Modification of the coal structure via physical compression (briquetting), chemical digestion, thermal modification, and functional group alkylation was used to further elucidate those structural features which govern the swelling behaviour of Victorian brown coals. Briquette weathering (i.e. swelling and disintegration of briquettes when exposed to variations in humidity and temperature) was examined by making alterations to briquette feed material and observing the effects on swelling in water.
The application of solubility parameter alone to prediction of coal swelling was rejected due to the many exceptions to any proposed trend. Brown coal swelling showed a minimum when the solvent electron-donor number (DN) minus its electron-acceptor number (AN) was closest to zero, i.e. when DN and AN were of similar magnitude. The degree of swelling increased either side of this point, as predicted by theory. In contrast to the solubility parameter approach (which suffers from the uncertainty caused by specific interaction between coal and solvent), the electron donor/acceptor approach is about specific interactions. It was concluded that a combination of total and three-dimensional solubility parameters and solvent electron donor/acceptor numbers may be used to predict solvent swelling of unextracted brown coals with some success.
Solvent access to chemically densified coal was found to be insensitive to a reduction in pore volume, and chemical effects were dominant. Thermal modification of the digested coal resulted in reduced swelling for all solvents, indicating that the structure had adopted a minimum energy configuration due to decarboxylation and replacement of hydrogen bonds with additional covalent bonds. Swelling of oxygen-alkylated coals demonstrated that the more polar solvents are able to break relatively weak hydrogen bonded crosslinks.
The large difference between the rate and extent of swelling in water (and hence weathering) of Yallourn and Morwell briquettes was shown to be almost entirely attributable to exchanged magnesium. Magnesium exchange significantly increases the rate and extent of swelling of Yallourn coal. It was also shown that the swelling of briquettes due to uptake of water by magnesium-exchanged coals is reduced significantly with controlled ageing of the briquettes.
The solvent swelling behaviour of Victorian brown coals is consistent with the notion that coal is a both covalently and non-covalently crosslinked and entangled macromolecular network comprising extractable species, which are held within the network by a wide range of non-covalent, polar, electron donor/acceptor interactions. Solvents capable of significant extraction of whole brown coals are also capable of significant swelling, but not dissolution, of the macromolecular coal network, which supports the view that the network is comprised of both covalent and ionic bonding. Victorian brown coals have also been shown to exhibit polyelectrolytic behaviour due to a high concentration of ionisable surface functionalities.

Cette thèse a pour objet l'étude du transport électronique dans les nanotubes de carbone monoparois par l'intermédiaire des fluctuations du courant. L'étude se place dans le cadre de la physique mésoscopique dans des conducteurs balistiques. Dans ce type de conducteur, plusieurs régimes diff´erents peuvent apparaître : blocage de Coulomb, transport modulé par les interférences quantiques, effet Kondo. Nous avons étudié les fluctuations du courant dans un régime d'interféromètre de type Fabry-Pérot électronique qui se présente comme une situation id´eale afin de sonder le régime où l'effet des interactions est faible. Les fluctuations du courant ont été analysées dans le formalisme de Landauer-Büttiker et nous obtenons une bonne correspondance entre la théorie et l'expérience. Nous avons ainsi observé la suppression du bruit dans les régimes de transmission unitaire et, par le biais des données combinées de la conductance et du bruit, nous avons pu déterminer les transmissions pour des canaux de conduction non dégénérés. Par ailleurs, le régime de l'effet Kondo a fait l'objet d'une étude dans laquelle nous avons observé des comportements universels dans la conductance et le bruit. Nous avons ajusté ces différentes grandeurs avec une théorie de bosons esclaves de champ moyen. Finalement, nous avons étudié une configuration de type Hanbury Brown et Twiss : un nanotube monoparoi sur lequel nous avons déposé un multiparoi qui nous sert de sonde afin d'injecter des électrons sur le conducteur.

Carbon Nanotubes as Cooper Pair Beam SplittersHerrmann, Lorentz 07 July 2010 (has links) (PDF)
We report on conductance measurements in carbon nanotube based double quantum dots connected to two normal electrodes and a central superconducting finger. By operating our devices as Cooper pair beam splitters, we provide evidence for Crossed Andreev Reflection (CAR). We inject Cooper pairs in the superconducting electrode and measure the differential conductance at both left and right arm. The contacts split the device into two coupled quantum dots. Each of the quantum dots can be tuned by a lateral sidegate. If the two sidegates are tuned such that both quantum dots are at a transmission resonance, a considerable part of the injected Cooper pairs splits into different normal contacts. On the contrary, if only one of the two dots is at resonance, nearly all pairs tunnel to the same normal contact. By comparing different triple points in the double dot stability diagram, we demonstrate the contribution of split Cooper pairs to the total current. In this manner, we are able to extract a splitting efficiency of up to 50% in the resonant case. Carbon Nanotubes ensure ballistic transport and long spin-flip scattering lengths. Due to these properties they are promising candidates to investigate EPR-type correlations in solid state systems.

Physiological adaptations in two ecotypes of Fucus vesiculosus and in Fucus radicans with focus on salinityGylle, A Maria January 2011 (has links)
The in origin intertidal marine brown alga Fucus vesiculosus L. grow permanently sublittoral in the brackish Bothnian Sea, side by side with the recently discovered F. radicans L. Bergström et L. Kautsky. Environmental conditions like salinity, light and temperature are clearly different between F. vesiculosus growth sites in the Bothnian Sea (4-5 practical salinity units, psu; part of the Baltic Sea) and the tidal Norwegian Sea (34-35 psu; part of the Atlantic Ocean). The general aims of this thesis were to compare physiological aspects between the marine ecotype and the brackish ecotype of F. vesiculosus as well as between the two Bothnian Sea species F. vesiculosus and F. radicans. The result in the study indicates a higher number of water soluble organic compounds in the marine ecotype of F. vesiculosus compared to the brackish ecotype. These compounds are suggested to be compatible solutes and be due to an intertidal and sublittoral adaptation, respectively; where the intertidal ecotype needs the compounds as a protection from oxygen radicals produced during high irradiation at low tide. The sublittoral ecotype might have lost the ability to synthesize these compound/compounds due to its habitat adaptation. The mannitol content is also higher in the marine ecotype compared to the brackish ecotype of F. vesiculosus and this is suggested to be due to both higher level of irradiance and higher salinity at the growth site. 77 K fluorescence emission spectra and immunoblotting of D1 and PsaA proteins indicate that both ecotypes of F. vesiculosus as well as F. radicans have an uneven ratio of photosystem II/photosystem I (PSII/PSI) with an overweight of PSI. The fluorescence emission spectrum of the Bothnian Sea ecotype of F. vesiculosus however, indicates a larger light-harvesting antenna of PSII compared to the marine ecotype of F. vesiculosus and F. radicans. Distinct differences in 77 K fluorescence emission spectra between the Bothnian Sea ecotype of F. vesiculosus and F. radicans confirm that this is a reliable method to use to separate these species. The marine ecotype of F. vesiculosus has a higher photosynthetic maximum (Pmax) compared to the brackish ecotype of F. vesiculosus and F. radicans whereas both the brackish species have similar Pmax. A reason for higher Pmax in the marine ecotype of F. vesiculosus compared to F. radicans is the greater relative amount of ribulose-1.5-bisphosphate carboxylase/oxygenase (Rubisco). The reason for higher Pmax in marine ecotype of F. vesiculosus compare to the brackish ecotype however is not due to the relative amount of Rubisco and further studies of the rate of CO2 fixation by Rubisco is recommended. Treatments of the brackish ecotype of F. vesiculosus in higher salinity than the Bothnian Sea natural water indicate that the most favourable salinity for high Pmax is 10 psu, followed by 20 psu. One part of the explanation to a high Pmax in 10 psu is a greater relative amount of PsaA protein in algae treated in 10 psu. The reason for greater amount of PsaA might be that the algae need to produce more ATP, and are able to have a higher flow of cyclic electron transport around PSI to serve a higher rate of CO2 fixation by Rubisco. However, studies of the rate of CO2 fixation by Rubisco in algae treated in similar salinities as in present study are recommended to confirm this theory. / Fucus vesiculosus L. (Blåstång) är en brunalg som i huvudsak växer i tidvattenzonen i marint vatten men arten klarar också att växa konstant under ytan i det bräckta Bottenhavet. Norska havet och den del av Bottenhavet, där algerna är insamlade i denna studie, har salthalterna 34-35 psu (praktisk salthaltsenhet) respektive 4-5 psu. F. radicans L. Bergström et L. Kautsky (Smaltång) är en nyligen upptäckt art (2005) som har utvecklats i Bottenhavet. F. radicans och Bottenhavets ekotyp av F. vesiculosus växer sida vid sida och har tidigare ansetts vara samma art. Sett till hela Östersjön, så ändras ytans salthalt från 25 till 1-2 psu mellan Östersjöns gräns mot Kattegatt och norra Bottenviken. Den låga salthalten i Östersjön beror på det höga flödet av sötvatten från älvarna och på ett litet inflödet av saltvatten i inloppet vid Kattegatt. Salthaltsgradienten är korrelerad med antalet arter som minskar med minskad salthalt. Östersjön är ett artfattigt hav och de arter som finns är till stor del en blandning av söt- och saltvattenarter. Det finns bara ett fåtal arter som är helt anpassade till bräckt vatten och F. radicans är en av dem. Exempel på miljöskillnader för F. vesiculosus i Norska havet och i Bottenhavet är salthalten, tidvattnet, ljuset och temperaturen. Tidvattnet i Norska havet gör att algerna växlar mellan att vara i vattnet och på land, vilket utsätter algerna för stora ljusskillnader, snabba och stora temperaturväxlingar samt även torka. De alger som växer i Bottenhavet har däremot en jämnare och lägre temperatur, istäcke på vintern och mindre tillgång på ljus eftersom de alltid lever under vattenytan. Skillnaderna i miljön mellan växtplatserna leder till skillnader i fysiologiska anpassningar. Anledningen till att F. vesiculosus och F. radicans valdes som studieobjekt i denna avhandling är att de är viktiga nyckelarter i Bottenhavet. F. vesiculosus och F. radicans är de enda större bältesbildande alger som finns i det artfattiga ekosystemet och de används därför flitigt som mat, gömställe, parningsplats och barnkammare för t.ex. fisk. Att de är nyckelarter gör det angeläget att försöka förstå hur algerna är anpassade och hur de reagerar på miljöförändringar för att få veta hur de kan skyddas och bevaras. F. radicans inkluderades även för att se hur en naturlig art i Bottenhavet är anpassad i jämförelse med den invandrade F. vesiculosus. Marin F. vesiculosus inkluderades för att vara en artreferens från artens naturliga växtplats. Studien visar att det finns fler vattenlösliga organiska substanser (finns vissa organiska substanser som har en proteinskyddande funktion) i den marina ekotypen av of F. vesiculosus än i Bottenhavets ekotyp. Anledningen till detta föreslås vara en anpassning till att växa i tidvattenzonen. Vid lågvatten utsätts F. vesiculosus från Norska havet för starkt ljus, uttorkning, och snabba temperatur- växlingar vilket gör att den kan behöva dessa organiska substanser som skydd mot fria syreradikaler som bildas under lågvattenexponeringarna. F. vesiculosus från Bottenhavet har troligen mist förmågan att syntetisera dessa substanser på grund av anpassning till att hela tiden växa under ytan. Mängden mannitol (socker) är högre i den marina ekotypen av of F. vesiculosus än i Bottenhavets ekotyp. Detta föreslås bero på högre fotosyntetiskt maximum i F. vesiculosus från Norska havet jämfört med ekotypen från Bottenhavet. Skillnaden i fotssyntetiskt maximum är bland annat kopplat till ljus- och salthaltskillnaden på algernas växtplatser. Denna teori styrks av att både fotosyntesen och halten av mannitol ökar i Bottenhavets ekotyp när den behandlas i högre salthalt. Studien visar även att båda ekotyperna av F. vesiculosus samt F. radicans har ett ojämnt förhållande mellan fotosystem II och I (PSII och PSI) med en dominans av PSI. Denna slutsats är baserad på fluorescens emissions mätningar vid 77 K (-196 °C) och mätning av den relativa mängden D1 protein (motsvarar PSII) och PsaA protein (motsvarar PSI). F. vesiculosus från Bottenhavet visar ett emission spektrum som pekar mot en jämnare fördelning av PSII och PSI jämfört med den marina ekotypen och F. radicans. Detta stämmer dock inte med förhållandet mellan D1/PsaA som indikerar att alla tre har mer PSI än PSII. Förklaringen till avvikelsen mellan metoderna antas vara att F. vesiculosus från Bottenhavet har större ljus-infångande antennpigment än marin F. vesiculosus och F. radicans. De tydliga skillnaderna i 77 K fluorescens emission spektra mellan Bottenhavets F. vesiculosus och F. radicans visar att denna metod kan användas som säker artidentifiering. Den marina ekotypen av F. vesiculosus har högre fotosyntetiskt maximum än de båda arterna från Bottenhavet. Mätningar av den relativa mängden av enzymet Rubisco, viktigt för upptaget av koldioxid hos växter och alger, visar att mängden enzym är en sannolik förklaring till skillnaden i fotosyntetiskt maximum mellan den marina ekotypen av F. vesiculosus och F. radicans och detta är troligen en normal artskillnad. Mängden Rubisco kan dock inte förklara skillnaden i fotosyntetiskt maximum mellan de båda ekotyperna av F. vesiculosus. För att undersöka vad skillnaden mellan dessa två beror på så föreslås istället mätningar av Rubisco’s koldioxidfixeringshastighet. Det är en ökning av fotosyntetiskt maximum i Bottenhavets ekotyp av F. vesiculosus när den behandlas i högre salthalt (10, 20 och 35 psu) och det högsta fotosyntetiska maximumet uppmättes i alger som behandlats i 10 psu. Denna ökning beror inte på ökning i den relativa mängden av Rubisco. Ökningen i fotosyntesen speglas dock av en ökning av den relativa mängden PsaA. Detta antas bero på att det behövs mer energi i form av ATP och att en ökning av detta kan ske på grund av att mer PsaA kan driva den cykliska elektrontransporten i fotosyntesreaktionen. Ökat behov av ATP antas bero på en ökning av Rubisco aktiviteten men mätning av aktiviteten krävs för att bekräfta detta.
