Nesse trabalho foi avaliado pela primeira vez, a imobilização de células do Botryosphaeria rhodina, um fungo ligninolitico produtor constitutivo de lacases. Três suportes foram avaliados: Fibra Acrílica Fina (FAF); Espuma de Poliuretano Expandido (EPE); Espuma de Poliuretano Fibroso (EPF). O EPF foi o melhor suporte por ter mostrado uma imobilização mais homogenia das células. Um planejamento fatorial foi desenvolvido para otimizar a produção de lacases por células livres, na presença de álcool veratrílico (AV). A análise da superfície de resposta mostrou que 18mM como a melhor concentração de AV para a produção de lacases, usando-se 3 mL de homogeinato de células como inóculo (DOλ400nm 0.4-0.6) para 25 mL de meio de cultura em frascos de 125mL, a 180 rmp, durante 126 horas a 28ºC. O perfil de crescimento do fungo, associado a produção de lacase foram comparados na presença e na ausência de AV, usando-se células livres e células imobilizadas do B. rhodina. A imobilização aumentou aproximadamente 3 vezes a produção de lacases e manteve estável o nível de produção durante 6 reciclos. A imobilização de células do B. rhodina mostrou-se útil uma vez que economizou 72horas para atingir a maior produção de lacase, quando comparada com células livres e também aumentou a tolerância do fungo a concentrações mais altas de AV (500mM) / The use of microbial immobilized cells to increase the production of fungal metabolites in fermentation processes has showed higher yields. This work evaluated by the first time, the immobilization of Botryosphaeria rhodina cells, a ligninolytic fungus that produces laccase. Three carriers were evaluated: acrylic fine fiber (FAF), expanded polyurethane foam (EPE), and fiber polyurethane foam (EPF). The EPF was the best carrier because showed a homogeneous immobilization cells. A factorial design was developed in order to optimize the laccase production by free cells in the presence of the laccase inducer veratryl alcohol (VA). The analysis by response surface answer showed 18 mM as the best VA concentration to produce laccase using 3 mL of a cell homogenate (ODλ400nm 0.4-0.6) as inoculum, to 25 mL of culture medium in shaked flasks (125 mL) at 180 rpm, during 126 hours at 28 °C. A growth profile for laccase and fungal biomass production were compared with and without VA using free and immobilized cells of B. rhodina. The cell immobilization increased approximately 3 folds the laccase production and maintained it stable during 6 consecutive recycles. The cell immobilization of B. rhodina showed to be useful once saved 72 hours to achieve the higher laccase production when compared to the one with free cells, and also increased the fungal cell tolerance at higher VA concentrations (500 mM)

A socialização docente de professores de educação física no início da carreira : um estudo etnográfico em duas escolas da Rede Municipal de Ensino de Porto Alegre - RS

Essa dissertação de mestrado, tem como objetivo central compreender como se constitui o processo de socialização docente de professores de Educação Física em início de carreira em duas escolas da Rede Municipal de Ensino de Porto Alegre - RS. Essa investigação é fruto de inquietações particulares e também se justifica pela sua revisão de literatura, quando apresenta baixa produção sobre o fenômeno estudado no campo da sociologia das profissões, embasada em Dubar (1996), Berger e Luckmann (2012). Ainda, aproximo esse debate com o campo da formação de professores, e passo a compreender a socialização docente como um fenômeno tridimensional, articulada por três elementos centrais que se inter-relacionam: identidades, cultura escolar, e subjetividades dos sujeitos. Também compreendo esse fenômeno como um processo que se faz, desfaz e refaz, de acordo com cada sujeito e cultura. Deste modo, as inquietações a acerca dessa temática se materializou nos seguintes objetivos específicos que auxiliaram na construção das seguintes análises: Identificar e compreender os lugares simbólicos que cada sujeito da escola desempenha no processo de socialização docente dos professores de Educação Física iniciantes; Compreender como o professorado de Educação Física em início de carreira entendem o processo de socialização docente, e identificar em quais /ocus da cultura escolar esse processo está presente; e, Compreender os elementos que constituem o processo de socialização docente dos professores de Educação Física iniciantes na Rede Municipal de Ensino de Porto Alegre. We studied two public schools of Porto Alegre city, and two teachers of Physical Education in the beginning of their carriers. lt was possible to learn during my fieldwork that teaching socialization of teachers happens mediated by some elements: stories and paths of individuais; confrontations and clashes with the culture which they make part; and reading, interpretation, understanding, constru ction, and reconstruction of reality. We understand that at first, the teachers go through periods of identification of times and school spaces, in a second time, they incorporate the routines and rituais established in the school, in a third time, it is possible highlight the movement and mobilization of teachers for, who knows, we think about the change of the everyday reality they experience at school. We also learned that the subjects construct interaction networks. These networks are made up of internai and externai elements that directly influence the socialization of teachers. Socialization was interpreted by collaborating teachers, so a li ttle more bureaucratized, and still takes place in two spaces in schools: the formal, which are bui lt by the school organization, and informal, which are built by the subjects of school themselves to meet the personal and professional needs. / This study aims to understand the formation of the process of teaching socialization of Physical Educators in the beginning of their careers in two public schools in Porto Alegre city - RS. This research is the result of private concerns and also it is justified by its literature revision. This research is the result of private concerns and also justified by their literature review, because the literature presents low production on the phenomenon studied in sociology, based on Dubar (1996), Berger and Luckmann (20 12). Still, we approach this debate to the field of teacher training, and we understand the teaching socialization as a three-dimensional phenomenon, articulated by three core elements that are interrelated: identities, school culture, and the subjectivity of individuais. We also understand this phenomenon as a process that is, undo and redo, according to each subject and culture. Thus, the concerns about this issue materialize in the following specific goals that help in the construction of the analysis: identify and understand the symbolic places that each individual in the school plays in the teaching process of socialization of beginner Physical Educators; understanding how the place of Physical Education in early career is understood in the teaching socialization process; identify in which toei of school culture the process is in; and understand the elements of the teaching socialization process of beginning physical educators in the public schools network of Porto Alegre. Methodologically, this research was carried out from a qualitative bias, based on ethnographic study, also linking with the teaching field, it was realized an educational ethnography and criticai method because it allows the researcher get into the school culture, seeking the intension to understand the process of this cultural phenomenon: the teaching socialization. / Esta tesis tiene como objetivo comprender cómo se constituye el proceso de socialización docente de profesores de Educación Física principiantes en dos escuelas de la red de ensefíanza del Ayuntamiento de Porto Alegre. Se trata de una investigación que, por una parte, se justifica por ser producto de indagaciones personales y, por otra parte, por lo aprendido en la revisión de literatura que ha mostrado una baja producción científica sobre el fenómeno estudiado en el campo de la Sociologia de las Profesiones, a partir de Dubar (1996) e Berger e Luckmann (20 12). A lo largo del estudio, acerco este debate en el campo de formación de profesores y entiendo la socialización docente como un fenómeno tridimensional, articulada por tres elementos básicos que están interrelacionados: las identidades, la cultura escolar y las subjetividades de los sujetos. Otra comprensión importante es que la socialización docente se manifiesta en un proceso que se hace, se deshace y se rehace de acuerdo a cada sujeto y su cultura. Así que las preocupaciones acerca del tema de estudio se han materializado en los siguientes objetivos específicos: a) identificar y comprender los lugares simbólicos que cada sujeto de la escuela asume en el proceso de socialización docente de los profesores de Educación Física principiantes; b) comprender como el profesorado de Educación Física prin cipiante entiende el proceso de socialización docente y identificar en cuáles locus de la cultura escolar este proceso está presente, y; d) Comprender os elementos que constituyen el proceso de socialización docente de los profesores de Educación Física principiantes en la red de ensefíanza del Ayuntamiento de Porto Alegre. Desde el punto de vista metodológico, la investigación fue realizada con un enfoque cualitativo y etnográfico. He realizado una etnografía educativa y crítica, lo que permite que el investigador haga una inmersión en la cultura escolar buscando comprender culturalmente el tema de la socialización docente. El estudio fue desarrollado en dos escuelas de la red de ensefíanza del Ayuntamiento de Porto Alegre con dos profesores de Educación Física principiantes. Fue posible aprender que la socialización docente de los profesores ocurre mediada por elementos como: las historias y trayectos de los sujetos, las confrontaciones y choques con la cultura en la que fue insertado, además de la lectura, interpretación, comprensión, construcción y (re)construcción de la realidad. Todavia entiendo que los profesores, en un primer momento, pasan por períodos de identificación de los tiempos y espacios escolares; en un segundo momento, incorporan las rutinas y rituales establecidos en la escuela y, en un tercer momento; reconocen la posibilidad de movimiento hacía el cambio de lo que vive en la realidad cotidiana de la escuela. He aprendido también que los sujetos construyen redes de interacción. Estas redes son constituídas de elementos internos y externos, que influyen directamente en la socialización de los docentes. La socialización fue interpretada por los profesores participantes del estudio de modo burocratizado y ocurre, sobretodo, en dos espacios en las escuelas: en los formales, que son construidos por la institución escolar; y en los informales, construidos por los propios sujetos de la escuela, para satisfacer sus necesidades personales y profesionales.

A terrível moléstia : vacina, epidemia, instituições e sujeitos : a história da varíola em Porto Alegre no século XIX (1846-1874)

Este trabalho tem por objetivo discutir os impactos da varíola em Porto Alegre no século XIX, tendo como ponto de partida as ações protagonizadas pelos diferentes sujeitos que vivenciaram, à época, a experiência da doença e das posturas que se estabelecem contra ela. Em 1846, regulou-se a vacinação contra a varíola como prática de Estado; a recepção da nova lei, entretanto, não alcançou a notoriedade esperada por agentes do governo imperial, já que a ampla maioria da população não estava informada dos propósitos da vacina e não conferia legitimidade à medida. Observando o perfil social dos variolosos percebe-se que aqueles sujeitos, protagonistas dos conflitos de seu tempo foram capazes de elaborar respostas próprias às demandas da doença, tendo buscado os préstimos do hospital Santa Casa de Misericórdia de Porto Alegre com vistas a aliviar seus sintomas. A recusa à vacinação foi constatada por meio de muitos relatórios governamentais no período, mas também ficou evidente mediante a observância da varíola como uma doença endêmica na cidade, que alcançou surtos epidêmicos graves, como no ano de 1874, quando 1% da população faleceu em função do contágio. Analisando as repostas elaboradas pelo incipiente “poder público” para o combate à epidemia, observou-se a inoperância das medidas profiláticas vigentes, já que o isolamento de pessoas, tido como necessário para a não propagação da doença, era regra para uma parte da população, majoritariamente sujeitos livres pobres e escravos, enquanto a doença atingiu indiscriminadamente todos os setores ou classes sociais. / This dissertation aims to discuss the impact of smallpox in Porto Alegre in the nineteenth century taking as its starting point the actions played by different subjects who experienced at the time the experience of illness and the attitudes that were established against it. In 1846, smallpox vaccination was regulated as a State practice, but the reception of the new law, however, did not reach the expected notoriety bythe agents of the imperial government, since the vast majority of the population was not informed of the purpose of the vaccine and did not confer legitimacy tothis measure. Observing the social profile of the carriers of smallpox, one realizes that those subjects, protagonists of conflicts of his time, were able to develop their own answers to the demands of the disease, searching the good services of the Hospital Santa Casa de Porto Alegre in order to alleviate their symptoms. The refusal of vaccination was found by many governmental reports of the period, but was also evident by the observance of smallpox as an endemic disease in the town. Indeed, smallpox reached serious epidemic outbreaks, as in 1874, when 1 % of the population died due of contagion. Analyzing the responses prepared by the incipient public power to fight the epidemic at the time, we can observe the ineffectiveness of current prophylactic measures, since the isolation of persons was deemed necessary to not spread the disease. It was the rule for a part of the population, mostly poor free subjects and slaves, while the disease indiscriminately affected all sectors or social classes.

Survey of brucellosis among people at risk in Lagos, Nigeria

Brucellosis is one of the neglected diseases in Nigeria. In Lagos, the commercial capital of Nigeria with about twenty one million people, a descriptive cross-sectional study was carried out in order to determine the sero-prevalence of brucellosis among people at risk in some selected abattoirs and secondary health care facilities (hospitals) in the state. Mixed sampling method was employed at the abattoir while convenient sampling method was used in sampling the respondents at the hospitals. Sera samples from three hundred and one (n=301) abattoir-based workers and traders; and one hundred and twenty one (n=121) hospital-based individuals which include people with febrile illnesses and blood donors were tested for brucellosis using Rose Bengal Plate test (RBPT), with indirect ELISA being used as a confirmatory test. Of the 301 abattoir-based workers and traders, 27 (8.97%) were sero-positive to the infection when Rose Bengal Plate test antigen was used. The twenty seven individuals consists of fifteen (15) butchers; four (4) veterinarians; two (2) meat transporters and bone/cow horn dealers each as well as one each of blood meal producer, abattoir engineer, water seller and meat supplier. When blood samples from the sero-positive individuals were subjected to ELISA, 3 (11.1%) were sero-positive to the brucellosis, while one is equivocal. These results confirm that agglutination observed on RBPT might be related to unknown cross-reactions and confirmation with a different test was necessary. None of the hospital-based respondents is sero-positive to the infection. The clinical signs significant for the infection in this study were fever, joint pain, lower backache, regular headache and miscarriage. Brucellosis awareness level among the respondents was very low. Data was analysed using (SPSS) version 20.0 at α0.05 significant level. The significant risk factors for human brucellosis according to this research are consumption of fura (unpasteurized milk) and wara (fresh cheese). The study revealed that brucellosis is not only an occupational disease but can also affect people who trade or live in proximity with infected animals. / Agriculture, Animal Health and Human Ecology / M. 台灣自1987年開放天空政策以來,各大小航空公司紛紛成立。低成本航空(LCC)也開始積極進入台灣市場。台灣與日本於2011年11月10簽屬日航空協定,此後,捷星(Jetstar)、樂桃(Peach)等低成本航空如雨後春筍般進入台灣,進而激起了許多低成本航空對於傳統航空(FSC)衝擊之討論。然而,低成本航在日本航載客率上是否真的對傳統航空造成影響?具體影響的程度為何?鮮有文獻深入探討。 本文針對大阪、成田、名古屋、琉球四條航線,透過敘述性統計、相關分析、多元迴歸分析之方法,將產業變數之、總體環境變數分別對華航、長榮載客率2011年至2016年之月資料進行分析。並進一步透過BCG矩陣歸納出兩家航空公司四條航線的成長率與市占率分布情形。 研究結果顯示,在日本四條航線上,低成本航空皆沒有與傳統航空載客率造成負向影響,反觀整體旅日市場上升,兩者市場區隔,形成二種經營型態共榮共存的局面。攸關日本線最顯著之變數為淡旺季、次者為消費者信心指數。在BCG矩陣研究結果顯示,中華航空四條航線市場占有率高、成長率低的情形;長榮航空相對市場佔有率低於中華航空,但是大阪線與琉球線的成長率卻高。 根據研究結果,分別對中華航空、長榮航空四條航線上提出各項建議,供航空業者、相關研究人員未來研究之參考。 / Since Taiwan agreed to initiate its Open-skies Policy in 1987, various airline companies began to form, including low-cost carriers (LCC) which eagerly entered Taiwanese markets. Taiwan and Japan reached aviation agreements on November 10, 2011; many LCCs such as Jetstar and Peach emerged as rising competitors in Taiwan, sparking a discussion about their impact on other full-service carriers (FSC). Do LCCs really pose a threat to FSCs in air travel between the two countries? To what extent do they affect those companies? Few sources have explored in detail. This research focuses on four destinations - Osaka, Tokyo Narita, Nagoya and Okinawa - based on descriptive statistics, correlation analysis and multiple regression analysis (MRA), while taking a close look at marketing and overall environmental factors in travel data between China Airlines and EVA Air (2011-2016, monthly). The growth and market share distributions of these four routes, operated by both companies, can be furthermore described using BCG matrices. The research concludes that, with respect to these four Japanese travel routes, LCCs did not create a negative impact on FSCs. Increasing traffic between Taiwan and Japan, in fact, facilitated prosper coexistence between the two types of carriers. Among the most significant factors are peak and low seasons, in addition to consumer confidence. BCG matrix analyses indicate that China Airlines had greater market shares and smaller growth rates; although EVA Air possessed fewer market shares than China Airlines, their Osaka and Okinawa routes grew at a rapid rate. The results of these observations may serve as suggestions to China Airlines and EVA Air, as well as references for aviation professionals and future researchers.

Produ??o e avalia??o de nanoemuls?o cati?nica como poss?vel vetor n?o-viral para pIRES2-EGFP

To achieve this goal, efficient vehicles, viral or non-viral, should be developed. The aim of this work was to produce and evaluate a nanoemulsion system as a possible carrier for no-viral gene therapy able to load a plasmid model (pIRES2-EGFP). The nanoemulsion was produced by the sonication method, after been choose in a pseudo-ternary phase diagram build with 5 % of Captex 355?, 1.2 % of Tween 80?, 0.8 % of Span 80?, 0.16% of stearylamine and water (to 100 %). Measurements of droplet size, polydispersity index (PI), zeta potential, pH and conductivity, were performed to characterize the system. Results showed droplets smaller than 200 nm (PI < 0.2) and zeta potential > 30 mV. The formulation pH was near to 7.0 and conductivity was that expected to oil in water systems (70 to 90 &#956;S/s) A scale up study, the stability of the system and the best sterilization method were also evaluated. We found that the system may be scaled up considering the time of sonication according to the volume produced, filtration was the best sterilization process and nanoemulsions were stable by 180 days at 4 ?C. Once developed, the complexation efficiency of the plasmid (pDNA) by the system was tested by agarose gel electrophoresis retardation assay.. The complexation efficiency increases when stearylamine was incorporated into aqueous phase (from 46 to 115 ng/&#956;L); regarding a contact period (nanoemulsion / pDNA) of at least 2 hours in an ice bath, for complete lipoplex formation. The nanoemulsion showed low toxicity in MRC-5 cells at the usual transfection concentration, 81.49 % of survival was found. So, it can be concluded that a nanoemulsion in which a plasmid model was loaded was achieved. A possibilidade de escalonamento, a estabilidade do sistema e a melhor forma de esteriliza??o tamb?m foram avaliadas onde observou-se que o sistema pode ser escalonado adequando o volume produzido ao tempo de sonica??o e a forma mais eficiente de esteriliza??o foi a filtra??o em membrana. O sistema apresentou estabilidade maior que 180 dias quando armazenado a 4 ?C. Uma vez desenvolvido, o sistema foi testado quanto ? capacidade de compactar o modelo plasmidial (pDNA). Eletroforese em gel de agarose foi a metodologia empregada para este fim, onde verificou-se que o poder de compacta??o aumenta (de 46 para 115 ng/&#956;L) quando a estearilamina ? incorporada na fase aquosa, respeitando um per?odo de contato (nanoemuls?o/pDNA) de pelo menos 2 horas, em banho de gelo. A nanoemuls?o mostrou-se pouco t?xica nas concentra??es usuais para ensaios de transfec??o em c?lulas do tipo MRC-5, apresentando 81,49 % de viabilidade celular. Portanto, o processo de produ??o de um sistema que complexa eficientemente o modelo plasmidial foi obtido.
