Previous issue date: 2014-04-18 / Não informada / Tuberculosis is an ancient infectious disease caused by Mycobacterium
tuberculosis. With 9.2 million cases worldwide each year by 2008, According to
WHO and the gradual loss of effectiveness of existing drugs through the
resistance of microorganisms, deserves special attention from our country's
authorities and encourages research in discovering new drugs that may be
successful in treating this disease. For this, a widely used approach is to search
for the pathogens metabolism inhibitors. The shikimic acid pathway is a
biosynthetic pathway critical to maintaining the bacteria, and thus the enzymes
belonging to the shikimate pathway are a prime target in the design of new
antibacterial agents. The enzyme chorismate synthase catalyzes the seventh and
final step of this route, converting the 5-enolpyruvylshikimate-3-phosphate
(EPSP) in chorismate, this is an important precursor in other metabolic pathways,
such as aromatic amino acids, folate, ubiquinone, vitamin K and other products.
Understanding the chorismate synthesis reaction is controversial because there is
no unanimity regarding the mechanism or the role of amino acids involved. The
elucidation of it will bring a significant scientific advance, helping to understand
the complexity involved in this biochemical step, and also contribute to the
development of new enzyme inhibitors. The study of the synthesis reaction in
question was developed employing the use of quantum chemistry computational
in DFT level using the functional B3LYP and 6-31 + G (d, p) basis set. From the
orbital Homo-Lumo orbitals and optimization of transition state between reactant
and reaction product (QST2), it was possible to clarify some important points
about the reaction . It was possible to propose a reaction mechanism in the light
of this theoretical chemical study . / A tuberculose é uma doença infecciosa muito antiga causada pelo
Mycobacterium tubrculosis. Com 9,2 milhões de casos no mundo a cada ano até
2008, segundo a OMS e a gradual perda de eficácia dos fármacos existentes frente
à resistência dos microorganismos, merece especial atenção das autoridades de
nosso país e motiva pesquisas na descoberta de novos fármacos que venham a ser
bem-sucedidos no tratamento dessa enfermidade. Para isso, uma abordagem
muito usada é a busca de inibidores do metabolismo dos agentes patogênicos. A
via do ácido chiquímico é uma rota biossintética fundamental para a manutenção
da bactéria e, assim, as enzimas pertencentes à via do chiquimato constituem um
excelente alvo no desenho de novos agentes antibacterianos. A enzima corismato
sintase catalisa a sétima e última etapa dessa rota, convertendo o 5-
enolpiruvilchiquimato-3-fostato(EPSP) em corismato, este é um importante
precursor em outras rotas metabólicas, como as de aminoácidos aromáticos,
folato, ubiquinona, vitamina K e outros produtos. A compreensão da reação de
síntese do corismato é controversa, pois não há unanimidade com relação ao
mecanismo nem quanto ao papel dos aminoácidos envolvidos. A elucidação da
mesma trará um significativo avanço científico, ajudando a entender toda a
complexidade envolvida nessa etapa bioquímica, e também contribuirá para o
desenvolvimento de novos inibidores da enzima. O estudo da reação de síntese
em questão foi desenvolvido empregando o uso da química quântica
computacional em nível DFT utilizando o funcional B3LYP e a base 6-31+G(d,p).
A partir dos orbitais Homo-Lumo e da otimização do estado de transição entre
reagente e produto da reação (QST2), foi possível esclarecer alguns pontos
importantes sobre a reação. Foi possível propor um mecanismo de reação à luz
deste estudo químico teórico.

Previous issue date: 2017-03-29 / Conselho Nacional de Pesquisa e Desenvolvimento Científico e Tecnológico - CNPq / Paracoccidioides fungi are the causative agents of paracoccidioidomycosis, an endemic disease in Latin America with great socioeconomic importance. Inhalation of spores, the infectious particles of the fungus, is a common way to get the infection. Its treatment is slow and generates long-term side effects making it necessary to study new metabolic pathways that can be potential targets for antifungal development. In this context, stands out the chiquimate pathway in Paracoccidioides spp., related to the production of chorismate and production of aromatic amino acids, and the chorismate synthase involved in the last stage of this pathway. The chorismate synthase from Pb18 was cloned into pGEX-4T3 vector, expressed in pLysS cells and purified on a glutathione-sepharose column. Antibodies were produced from the immunization of mice with the recombinant protein obtained and used in Western blot assay to confirm protein expression. For characterization of the enzyme, its modeling was carried out. Pull-down-GST assay was performed to identify interactions between the chorismate synthase and soluble proteins present in total protein extract of Pb18. The interactions found included proteins of different functional categories related to protein synthesis, related with metabolism of common intermediates such as folate and quinolinate or related with the availability of ATP, phosphoenolpyruvate and erythrose-4-phosphate required in the chiquimate pathway. Unexpected interactions with proteins of the cell cycle and also with regulating proteins of the cell division process were observed. / Fungos do gênero Paracoccidioides são os agentes causadores da paracoccidioidomicose, uma doença endêmica na América Latina de grande importância socioeconômica. A inalação de esporos, partículas infecciosas do fungo, é uma forma comum
de adquirir a infecção. O seu tratamento é lento e gera efeitos colaterais tornando necessário o estudo de novas vias metabólicas que sejam potenciais alvos para o desenvolvimento de antifúngicos. Nesse contexto, destaca-se a via do chiquimato em Paracoccidioides spp., relacionada à obtenção de corismato e produção de aminoácidos aromáticos, sendo corismato sintase a enzima envolvida na última etapa dessa via. A corismato sintase proveniente de Pb18 foi clonada em vetor pGEX-4T3, expressa em células pLysS e purificada em coluna de glutationa-sefarose. Anticorpos foram produzidos a partir da imunização de camundongos com a proteína recombinante obtida e utilizados em ensaio de Western blot a fim de confirmar a expressão proteica. A modelagem da corismato sintase foi realizada e os aminoácidos envolvidos no sítio ativo foram identificados. Ensaios de pull-down-GST foram realizados a fim de identificar os complexos de interações entre a corismato sintase e proteínas solúveis presentes em extrato proteico total de Pb18. As interações encontradas incluíram proteínas de diferentes categorias funcionais relacionadas à síntese proteica, ao metabolismo de intermediários comuns como folato e quinolinato ou ainda com a disponibilidade de ATP, fosfoenolpiruvato e eritrose-4-fosfato necessários na via do chiquimato. Interações inesperadas com proteínas do ciclo celular e também com proteínas reguladoras do processo de divisão celular foram observadas.

Yeast Chorismate Mutase: Molecular Evolution of an Allosteric Enzyme / Die Chorismatemutase der Bäckerhefe: Molekulare Evolution eines Allosterischen EnzymsHelmstaedt, Kerstin 31 October 2002 (has links)
Die Chorismatmutase (CM, EC 5.4.99.5), kodiert durch ARO7, katalysiert die Claisen-Umlagerung von Chorismat zu Prephenat in der Biosynthese von Tyrosin und Phenylalanin. Das relativ kleine, dimere Enzym der Hefe Saccharomyces cerevisiae wird allosterisch durch Tryptophan aktiviert und allosterisch durch Tyrosin inhibiert. In der vorliegenden Arbeit wurde die Theorie widerlegt, dass die Chorismatemutase an der Osmoregulation und Vakuolenentstehung beteiligt ist. Die Analyse einiger Stämme mit punktmutiertem oder deletiertem ARO7-Gen zeigte ausschließlich eine Funktion in der Aminosäure-Biosynthese. Die Fusion an das grün-fluoreszierende Protein ermöglichte die Lokalisierung der CM in Cytoplasma und Kern der Hefezelle.Auf Proteinebene wurde der intramolekulare Signalübertragungsweg von den allosterischen zu den aktiven Zentren näher untersucht. Es wurden Chimären-Enzyme hergestellt, in denen das molekulare Scharnier L220s zwischen der katalytischen und allosterischen Domäne ausgetauscht wurde gegen den entsprechenden Bestandteil homologer Pilzenzyme. Die kinetische Analyse zeigte, dass dieser Proteinteil essentiell ist für die Unterscheidung zwischen dem Signal Aktivierung bzw. Inhibierung. Diese Region ist auch für die Dimerisierung der CM von Bedeutung. Durch Austausch hydrophober Aminosäuren gegen geladene Reste in und in der Nähe dieses Scharniers wurde eine stabile, monomere Enzymvariante hergestellt. Diese CM zeigte reduzierte Aktivität und keine Regulation, aber das kodierende Gen komplementierte die Tyrosin- und Phenylalanin-Auxotrophie der Zellen. Diese Ergebnisse unterstützen die Theorie, dass das Hefeenzym durch gleichzeitige Evolution von Regulations- und Stabilisierungsmechanismen aus einem monomeren, unregulierten Vorläuferprotein entstanden ist, welches dem der Escherichia coli CM ähnlich war. Um weitere Erkenntnisse über die Prinzipien der Proteinstabilisierung zu erhalten, wurde auch die Chorismatmutase on Thermus thermophilus charakterisiert, nachdem das kodierende Gen kloniert war. Dieses Enzym ist ähnlich zu der strukturell einzigartigen Chorismatmutae aus Bacillus subtilis, wird aber, im Gegensatz zu letzterem durch Tyrosin in seiner Aktivität gehemmt. Modellierungsstudien zeigten, dass wie auch bei anderen Proteinen verstärkte Hydrophilität von Oberflächen, erhöhte Hydrophobizität innerhalb der Struktur wie auch die Versteifung von Loops in der Nähe des aktiven Zentrums zur Stabilisierung dieser Proteinfaltung beitragen.
