Quantification and determination of the interindividual variability observed in a group of cockroaches and its implications at a collective level

In this thesis, we used the American cockroach study the relationship between animal personality and collective behaviour. These questions do not only regard domiciliary cockroaches or insects; in fact, the questions we discuss here have a large scope and concern most of the known gregarious species distributed in several taxa. One of our aims is to develop general tools, methods and analyses that could be used for the study of collective behaviour.We show evidence of personality in several behavioural traits and different contexts in the American cockroach. These behavioural differences were observed in a context without social interactions during the daytime (isolated condition) and nighttime (solitary exploration behaviour) and in a social context (rate of joining a shelter and sheltering time). Based on the short-term experiments, this personality can be understood as differences in the probability of joining a shelter. We show that the behavioural variability existing in a population of domiciliary cockroaches is very high. The composition of personalities within a group can lead to group personality – consistent differences in group behaviour. For instance, groups composed of different behavioural profiles show different collective dynamics. Regarding the long-term experiments, our results show that the individuals that were aggregated at a site that was repeatedly disturbed by a lighting stimulus during their resting period showed slow migration to a new shelter, which allowed the initial aggregation site to remain the site of choice for a few days. Moreover, the disturbance regime did not influence the group's global activity rhythm. At the individual level, we observed interindividual differences (personalities) in terms of their position prior to the disturbance but not for the different steps of the fleeing behaviour itself. In addition, we show that thigmotaxis affects the reaction time to the disturbance: individuals near the walls of the shelter react more slowly thanindividuals in the centre. Finally, an approach coupling modelling and experimental data shows that behavioural variability plays a secondary role during migration dynamics, thus highlighting the plasticity of personality traits depending on the context. / Un des comportements collectifs les plus répandus, qu’il s’agisse de vertébrés (mammifères, oiseaux, poissons), d’insectes ou encore de bactéries, est la tendance des individus à se regrouper. Les causes proximales de ces rassemblements ou agrégats, c'est-à-dire les mécanismes à leur origine, ont retenu moins d’attention que l’étude des causes ultimes. De plus, dans la plupart des études portant sur les processus d’agrégation, notamment chez notre organisme modèle, la blatte, la diversité des comportements individuels est souvent sous-estimée ou ignorée. L’objectif de cette thèse est l’étude du rôle des différences interindividuelles, également connues comme personnalité animale ou syndromes comportementales, dans les processus de prise de décision collective et notamment la formation des agrégats.Pour cela, nous avons utilisé des groupes de mâles de la blatte américaine Periplaneta americana. Ces insectes peuvent s’agréger dans un ou plusieurs abris et présenter des différences dans leur réponse au milieu, aux conspécifiques et dans le partage de l’information. Dans le cadre de prises de décisions collectives, nous avons mis en évidence une personnalité individuelle mais également au niveau du groupe. Celles-ci se manifestent dans la recherche d’un abri dont certains individus jouent un rôle clef. Nous avons montré que les groupes présentent une stabilité qui est observable au niveau de la dynamique d'agrégation et qui dépend de la distribution des personnalités au sein de ceux-ci. De plus, nous remarquons que ces différences de personnalités au sein des groupes affectent la prise de décision collective, notamment la vitesse du choix et le nombre total d’individus abrités. De manière surprenante, la composition des groupes n’affecte pas la probabilité que ceux-ci atteignent un consensus: la majorité des blattes étant agrégées sous le même abri.Concernant les influences croisées entre l’effet sociale et la personnalité des blattes, nous montrons que les effets sociaux tendent à supprimer les différences inter-individuelles et créent une corrélation entre le comportement de chaque individu et le comportement du groupe. De plus, nous observons que la présence de différentes personnalités dans un groupe augmente les amplifications sociales, celles-ci étant dues aux interactions entre les membres du groupe. Finalement, nous abordons la question du rôle de la personnalité sur le comportement de fuite lors des perturbations lumineuses et sur la dynamique d’émigration quand le site de repos est régulièrement perturbé. Nous avons montré l’existence de personnalités exprimées pendant la phase active nocturne et de différences interindividuelles au niveau du thigmotactisme pendant la journée (phase passive de repos et d’agrégation). Curieusement, ces différences inter-individuelles ne sont pas observées lors du comportement de fuite et au niveau de la dynamique global d’émigration. Controle de Blattella germanica (L.) com Metarhizium anisopliae e inseticidas reguladores de crescimento. / Control of the german cockroach Blattella germanica with Metarhizium anisopliae and insect growth regulators.

Avaliou-se o efeito de Metarhizium anisopliae e dos inseticidas triflumuron e pyriproxifen para adultos e ninfas de Blattella germanica, e a eficiência da associação desses agentes no controle de colônias da praga. Doses elevadas do patógeno ocasionaram altos índices de mortalidade de adultos e ninfas. Não foi observada mortalidade significativa para insetos que se alimentaram de iscas contaminadas com o patógeno. Adultos e ninfas expostos à superfície contaminada com o fungo na formulação pó (5%) apresentaram mortalidade de 93,7% e 82,3% após 9 dias da aplicação, respectivamente. As CL25 para ninfas tratadas com triflumuron e pyriproxifen foram de 0,19% e 0,45% de ingrediente ativo, respectivamente, para um período de alimentação de 72 horas na isca contaminada. Os inseticidas afetaram a produção de ootecas viáveis e o número de ninfas produzidas por ooteca. Após 60 dias, colônias de laboratório não tratadas apresentaram 6,55 vezes mais insetos em relação à população inicial, com uma média de 233,9 insetos. As colônias apresentaram crescimento menor quando submetidas aos inseticidas triflumuron e pyriproxifen, com 60,8 e 81 insetos em média, respectivamente. Os melhores resultados foram obtidos nos tratamentos em que os inseticidas foram associados ao patógeno, resultando populações de 41,8 e 57,7 insetos para triflumuron + M. anisolpiae e pyriproxifen + M. anisopliae, respectivamente. Duas aplicações associando-se o patógeno aos inseticidas na mesma isca atrativa, em uma câmara de infecção, controlou totalmente uma população em campo. Insetos da criação infectados por Gregarina sp. mostraram-se mais sensíveis quando expostos a M. anisopliae e a triflumuron em relação a insetos sadios. Estudos mais detalhados sobre como os inseticidas podem interferir no processo de doença casusada pelo fungo no inseto, além de aspectos relacionados à formulação do micoinseticida e da sua contribuição na mortalidade de populações de B. germanica em campo são ainda necessários. / It was evaluated the effect of Metarhizium anisopliae and the insect growth regulators triflumuron and pyriproxifen for adults and nymphs of the German cockroach Blattella germanica, and the efficacy of its association in the control of this urban pest. High doses of the pathogen caused high mortality of adults and nymphs. There was no mortality of B. germanica nymphs exposed for 72 hours to the food contaminated with M. anisopliae. The mortality of adults and nymphs was of 93.7% and 82.3%, respectively, when exposed to a surface that had been contaminated with the fungus formulation (5%), as early as 9 days after the application. Charakterisierung von Transportmechanismen in der Speicheldrüse der Schabe Periplaneta americana / Characterisation of transport mechanisms in salivary glands of the cockroach Periplaneta americana

Die Aktivierung der Speichelsekretion erfolgt in der innervierten Speicheldrüse der Schabe <i>Periplaneta americana</i> durch die biogenen Amine Dopamin (DA) und Serotonin (5-HT). Die Acini der Speicheldrüse sezernieren einen Primärspeichel, der in den Ausführgängen modifiziert wird. Die durch DA und 5-HT aktivierten Signalwege sowie die an der Elektrolyt- und Flüssigkeitssekretion bzw. Speichel-modifikation beteiligten Transportmechanismen sind weitgehend unbekannt.<br> Mikrofluorometrische Ca<sup>2+</sup>-, Na<sup>+</sup>- und pH-Messungen in Kombination mit pharmakologischen Experimenten, biochemische Messungen der Aktivitäten von Ionentransport-ATPasen sowie videomikroskopische Analysen zu transepithelialen Wasserbewegungen wurden in dieser Arbeit durchgeführt. Sie sollten Informationen über die an der Speichelbildung und -modifikation beteiligten Transportmechanismen und die Signalwege liefern, welche durch DA und/oder 5-HT aktiviert werden. <br><br> Wesentliche Ergebnisse dieser Arbeit waren:<br><br> <ul> <li>Messungen des intrazellulären pH (pH<sub>i</sub>) in Gangzellen zeigten, dass isolierte Ausführgänge mit Acini bei Stimulierung mit DA und 5-HT stark ansäuerten. In isolierten Ausführgängen ohne Acini verursachte nur DA eine schwache Ansäuerung. Da nur die Ausführgänge dopaminerg innerviert sind, die Acini jedoch dopaminerg und serotonerg, zeigt dieses Ergebnis, dass die DA- und/oder 5-HT-induzierte Primärspeichelbildung die Ursache für die pHi-Änderungen in den Gangzellen ist. pH<sub>i</sub>-Messungen in den Gangzellen geben also auch Hinweise auf Transportvorgänge in den Acini.</li> <li> Der Na<sup>+</sup>-K<sup>+</sup>-2Cl<sup>-</sup>-Symporter und der Cl<sup>-</sup>-HCO<sub>3</sub><sup>-</sup>-Antiporter, gekoppelt mit dem Na<sup>+</sup> H<sup>+</sup>-Antiporter (NHE) waren an der NaCl-Aufnahme in die peripheren Zellen der Acini zur Bildung des NaCl-reichen Primärspeichels beteiligt. Die Aktivität dieser Transporter hing von der CO<sub>2</sub>/HCO<sub>3</sub><sup>-</sup>-Verfügbarkeit ab und war Ca<sup>2+</sup>-abhängig.</li> <li>Die starke Ansäuerung in den Gangzellen hing nicht von der Aktivität der apikalen vakuolären Protonen-ATPase (V-H<sup>+</sup>-ATPase), aber von der Aktivität der basolateralen Na<sup>+</sup>-K<sup>+</sup>-ATPase ab, die anscheinend in den Ausführgängen die Speichelmodifikation energetisiert.</li> <li>In isolierten Ausführgängen mit Acini waren die V-H<sup>+</sup>-ATPase und Na<sup>+</sup>-abhängige Transporter (u. a. NHE) an der Erholung von einer DA-induzierten oder einer NH<sub>4</sub>Cl-Vorpuls-induzierten Ansäuerung in den Gangzellen beteiligt. Bei der Regulation des pH<sub>i</sub> in unstimulierten Gangzellen spielten diese Transporter keine Rolle.</li> <li>In isolierten Ausführgängen mit Acini induzierte DA in den Gangzellen einen Anstieg der [Na<sup>+</sup>]<sub>i</sub> und, zeitlich verzögert, auch der [Ca<sup>2+</sup>]<sub>i</sub>. Der [Na<sup>+</sup>]<sub>i</sub>-Anstieg war von der Aktivität der Acini abhängig und erfolgte möglicherweise über apikale Na+-Kanäle. Der [Ca<sup>2+</sup>]<sub>i</sub>-Anstieg war graduiert und tonisch. Der DA-induzierte [Na<sup>+</sup>]<sub>i</sub>-Anstieg in den Gangzellen und deren Depolarisation führten dazu, dass der basolaterale Na<sup>+</sup>-Ca<sup>2+</sup>-Antiporter in den Ca<sup>2+</sup>-Influx-Modus umkehrte. Die daraus resultierende tonische [Ca<sup>2+</sup>]<sub>i</sub>-Erhöhung könnte an der Regulation der Na<sup>+</sup>-Rückresorption beteiligt sein.</li> <li>Zum Nachweis transepithelialer Flüssigkeitsbewegungen in isolierten Ausführgängen wurde eine videomikroskopische Methode entwickelt. Isolierte Ausführgänge ohne Acini resorbierten im unstimulierten Zustand Flüssigkeit aus dem Ausführganglumen. Möglicherweise sezernieren die Acini auch im unstimulierten Zustand mit geringerer Rate einen Primärspeichel, der in den Ausführgängen resorbiert wird. Die Resorption war ATP-abhängig. Der ATP-verbrauchende Transportmechanismus konnte nicht identifiziert werden. Weder die Na<sup>+</sup>-K<sup>+</sup>-ATPase noch die V-H<sup>+</sup>-ATPase waren an der Resorption beteiligt.</li> </ul> <br> Diese Arbeit trug zur Kenntnis der komplexen Funktionsweise von Speicheldrüsen in Insekten bei und erweiterte das lückenhafte Wissen über die zellulären Wirkungen biogener Amine in Insekten. Zudem wurden in dieser Arbeit viele Parallelen zu Funktionsweisen der Speicheldrüsen in Vertebraten deutlich. / The acinar salivary glands in the cockroach <i>Periplaneta americana</i> are innervated by dopaminergic and serotonergic fibers and secrete a NaCl-rich primary saliva upon stimulation with the biogenic amines dopamine (DA) or serotonin (5-HT). The ducts downstream of the acini are thought to modify the primary saliva by Na<sup>+ </sup>reabsorption and K<sup>+</sup> secretion. The electrolyte and fluid transport processes activated by DA and 5-HT as well as the second messenger pathways mediating between the biogenic amine receptors and the effector transport mechanisms are poorly understood.In this sudy, microfluorometrical Ca<sup>2+</sup>, Na<sup>+</sup> and pH measurements were performed in combination with pharmacological experiments. Furthermore, ATPase activity assays and microscopical analyses of transepithelial fluid transport were done. The aim of this work has been the characterisation of the DA-induced transport mechanisms in the cockroach salivary glands in order to improve our understanding of the cellular actions of biogenic amines in insects. <br><br> Intracellular pH measurements in duct cells of isolated small lobes of salivary glands consiting of several acini and ducts showed a strong intracellular acidification upon DA or 5-HT stimulation. On the other hand, only a small intracellular acidification could be recognised in isolated ducts without acini. The acini are innervated by dopaminergic and serotonergic fibers, whereas the ducts are innervated only by dopaminergic fibers. Thus, this result demonstrates, that the DA- or 5-HT-induced production of primary saliva in the acini causes the intracellular pH changes in the ducts. Consequently, intracellular pH measurements in ducts are also useful to characterise transport processes in the acini.<br><br> The Na<sup>+</sup>-K<sup>+</sup>-2Cl<sup>-</sup> cotransport and/or the Cl<sup>-</sup>-HCO<sub>3</sub><sup>-</sup> exchange combined with the Na<sup>+</sup> H<sup>+</sup> exchange (NHE) were responsible for the NaCl uptake at the basolateral membrane in the peripheral cells of the acini during production of primary saliva. The activity of these transporters was regulated by the CO<sub>2</sub>/HCO<sub>3</sub><sup>-</sup>-availability and was Ca<sup>2+</sup>-dependent. The activity of the basolateral Na<sup>+</sup>-K<sup>+</sup>-ATPase, but not of the apical vacuolar-type proton pump (V-H<sup>+</sup>-ATPase) in the duct cells was necessary for the strong intracellular acidification in the ducts with acini. Thus, the Na<sup>+</sup>-K<sup>+</sup>-ATPase seems to energise the saliva modification in the ducts. In ducts with acini, the V-H<sup>+</sup>-ATPase and Na<sup>+</sup>-dependent transporters (e.g. NHE) were responsible for the pH-recovery after a DA- or NH<sub>4</sub>Cl-induced intracellular acidification in the duct cells. In the regulation of the intracellular resting pH these transporters played a minor role. In addition, DA induced an increase in the intracellular Na<sup>+</sup> concentration, followed by an increase in the intracellular Ca<sup>2+</sup> concentration in duct cells with acini, but never in duct cells without acini. The Na<sup>+</sup> elevation was probably the result of the activity of apical Na<sup>+</sup> channels. The DA-induced Na<sup>+</sup> elevation and a depolarisation of the basolateral membrane of the duct cells reversed a Na<sup>+</sup>-Ca<sup>2+</sup> exchange activity into the reverse mode causing a graded Ca<sup>2+</sup> elevation in duct cells. The Ca<sup>2+</sup> elevation is probably involved in the regulation of the Na<sup>+</sup> reabsorption during saliva modification. Transepithelial fluid transport in isolated ducts was detected with a fluorescent microscopical method. Already unstimulated isolated ducts reabsorbed fluid from the duct lumen to the bath side. Perhaps unstimulated acini possess a basic secretion rate and this primary saliva is than reabsorbed in the ducts. The fluid reabsorption was ATP-dependent, but the ATP-consuming transport mechanism could not be identified. Neither the basolateral Na<sup>+</sup>-K<sup>+</sup>-ATPase, nor the apical V-H<sup>+</sup>-ATPase were involved in fluid reabsorption. This work extends our knowledge about the complex function of insect salivary glands and about the cellular action of biogenic amines in insects. Additionally, it indicates lots of similarities between the functions of salivary glands in vertebrates and invertebrates.

The thesis is focused on anatomy and implementation of dissections of 4 selected model invertebrates in school practice. The selected model species include swan mussel (Anodonta cygnea), stick insect (Medauroidea extradentata), cockchafer (Melolontha melolontha) and sea urchin (Echinus esculentus). In case of the cockchafer, the text is based on this model species, but the supporting photographs of dissection are taken from the tropical cetoniid beetle Pachnoda, which has very similar internal anatomy with the cockchafer. For all selected animals, I describe external and internal morphology as well as recommended methods of obtaining the material, proper killing of specimens, and dissection techniques. The text is supplemented with original digital macrophotographs. The thesis includes also an overview of primary and secondary school textbooks with focus on anatomy of the selected animals. The text is supplemented by a glossary of morphological terminology used in this thesis. Attached is a photographic atlas of dissections, which is a combination of this master thesis and my bachelor thesis (defended in 2010), in which I dealt with five model invertebrates in the same way. The analysis is anchored in Michael Hardt and Antonio Negri’s reconceptualization of Michel Foucault’s notion of biopolitics. Hardt and Negri combine the concept of biopolitics with contemporary theories of cognitive capitalism and immaterial labour, to illuminate how subjects are subsumed into a system of biopower in which capitalistic production has become biopolitical production. I argue that the Brexit novels examined in this thesis demonstrate how the intrinsic bond between production and life shapes the characters’ relationship to the referendum. As the characters are caught between individual goals and communal values, in a system that demands that they take sole responsibility for their own success while also being responsible democratic citizens, the referendum produces conflicted subjects that experience deep internal and external conflicts in relation to Brexit.
