Cell wall compositional differences between mealy and non-mealy ‘Forelle’ pear (Pyrus communis L.)

Thesis (PhD(Agric) (Horticulture))--University of Stellenbosch, 2011. / Includes bibliography. / ENGLISH ABSTRACT: Mealiness, a soft, dry textural disorder of ‘Forelle’ pear (Pyrus communis L.), is a problem for the South African fruit export industry. Soft, dry textural disorders seem to be related to changes in cell wall breakdown. The aim of this work was, therefore, to investigate the occurrence of mealiness‐associated changes in the cell wall and elucidate the mechanism by which mealiness occurs in ‘Forelle’ pear, as well as to characterise cell wall changes occurring during normal ripening. Mealy ‘Forelle’ tissues had significantly lower total galacturonic acids associated with the middle lamella (water‐ and CDTA‐soluble fractions). The water‐soluble pectin of mealy tissues was depolymerised at an earlier stage of ripening. The widespread disintegration of cell‐to‐cell adhesion in mealy cell walls only, suggests that the middle lamella and the plasmodesmata are more broken down. In mealy ‘Forelle’ tissues there was no indication of less broken down high molecular weight polyuronides in the CDTA fraction, normally associated with these dry, soft textures. The pectins from mealy tissues were more broken down and both mealy and non‐mealy tissue polyuronides depolymerised. Furthermore, there was a lack of light toluidine staining in the larger air spaces, which would indicate such water‐insoluble pectins. These data suggest that the formation of high molecular weight pectate gels is unlikely in mealy ‘Forelle’ pear. The slight increase in the galactose content in mealy tissues in CDTA‐ and Na2CO3‐soluble fractions and slight decrease in the 1 M KOH glycan fraction during later stages of ripening (6+11, 9+7, 9+11; weeks at ‐0.5°C plus days at 15°C) may indicate that galactose loosely interlinked into the glycan fraction broke down sooner for mealy tissues. This didn’t increase molecular size profiles in the CDTA fraction. Arabinose content was slightly higher in the 4 M KOH fraction and slightly lower in mealy tissues of water‐ and CDTA fractions. This did not influence the molecular weight of the glycans compared to those in the nonmealy tissues. ‘Forelle’ data therefore seem to be more congruent with a decrease in intercellular adhesion as the mechanism by which mealiness occurs, rather than the formation of high molecular weight pectins taking up the cellular fluid. ‘Forelle’ pear water‐soluble pectin content increases with increased ripening. High amounts of watersoluble pectin and low amounts of Na2CO3‐soluble pectin suggests that solubilisation of rhamnogalacturonan‐I pectins must have taken place during early ripening (at a fruit firmness of > 4.7 kg (7.9mm tip). Galactose and glucose in the pectin fraction dramatically decreased after fruit ripened to a firmness of 4.5 kg, whereafter they remained unchanged. This was also the period in which fruit softened the most and the biggest increase in pectin water‐solubility occurred. It is not known whether these events are coincidental, or linked causally. Rhamnose and arabinose extractability increased in the water fraction and xylose, fucose and mannose increased in glycan fractions with ripening. The biggest changes in polyuronide solubilisation and depolymerisation occurred in water‐ and CDTA fractions between storage and ripening durations of 3+7 (4.7 kg) and 6+4 (2.7 kg). / AFRIKAANSE OPSOMMING: Melerigheid, ʼn sagte droë tekstuur afwyking van ‘Forelle’ pere (Pyrus communis L.), is ʼn probleem vir die Suid Afrikaanse vrugte uitvoerbedryf. Sagte, droë tekstuur afwykings blyk betrekking te hê op selwandafbraak veranderinge. Die doel van die studie was dus om die melerigheid‐geassosieerde veranderinge in die selwand te ondersoek, sowel as om vas te stel wat die meganisme betrokke is by melerigheid ontwikkeling in ‘Forelle’ pere. Die selwand veranderinge gedurende normale rypwording is ook gekarakteriseer. Melerige ‘Forelle’ weefsel het betekenisvol laer totale galakturoonsuur wat geassosieer is met die middellamella (water‐ en CDTA‐oplosbare fraksies). Die water‐oplosbare pektien van melerige weefsel was op ʼn vroeër stadium van rypwording gedepolimeriseer. Die wydverspreide disintegrasie van sel‐tot‐sel adhesie, slegs in melerige selwande, dui aan dat die middellamella en die plasmodesmata meer afgebreek is. Daar is geen indikasie van hoë molekulêre massa poliuroniedes in die CDTA fraksie van melerige ‘Forelle’ weefsel, wat gewoonlik geassosieer word met droë, sagte teksture nie. Die pektiene van melerige weefsel was meer afgebreek en melerige en nie‐melerige weefsel se poliurone was gedepolimeriseer. Daar was ook geen ligte toluïdien verkleuring in die groter intersellulêre lugruimtes nie, wat ʼn aanduiding sou wees van wateronoplosbare pektiene. Hierdie data dui dus aan dat die vorming van hoë molekulêre pektien jel in melerige ‘Forelle’ pere onwaarskynlik is. Die klein toename in galaktose inhoud in die CDTA‐ en Na2CO3‐ oplosbare fraksies en ʼn klein afname in 1 M KOH glikaan fraksie tydens latere rypheidstadiums (6+11, 9+7, 9+11; weke by ‐0.5°C plus dae by 15°C), kan beteken dat los verweefde galaktose in die glikaan fraksie vroeër afgebreek het in melerige weefsels. Die molekulêre grootte profiel is nie verander in die CDTA fraksie nie. Arabinose inhoud was bietjie hoër in die 4 M KOH fraksie en bietjie laer in melerige weefsel van die water‐ en CDTA fraksies. Die molekulêre massa van die glikane was klaarblyklik onbeïnvloed hierdeur. ‘Forelle’ data blyk dus meer saam te stem met die meganisme waar ʼn vermindering in intersellulêre adhesie ʼn rol speel in melerigheid, eerder as die meganisme waar hoë molekulêre pektien selvloeistowwe bind. ‘Forelle’ peer water‐oplosbare pektieninhoud neem toe met toenemende rypheid. Hoë vlakke wateroplosbare pektien en lae vlakke Na2CO3‐oplosbare pektien stel voor dat die oplossing van rhamnogalakturonan‐I pektiene gedurende vroeë rypwording moes plaasgevind het (by ʼn fermheid van > 4.7 kg (7.9mm punt). Galaktose en glukose in die pektienfraksie het drasties verminder nadat vrugte tot ʼn fermheid van 4.5 kg ryp geword het, waarna hul onveranderd gebly het. Dit was ook die periode waarin vrugte die meeste sag geword het en die grootste toename in poliuronied wateroplosbaarheid gevind is. Dit is nie bekend of die gebeure toevallig of oorsaaklik verbind is nie. Rhamnose en arabinose ekstraheerbaarheid het vermeerder in die water fraksies, en xylose, fukose en mannose het vermeerder in die glikaan fraksies gedurende rypwording. Die grootste verandering in oplosbaarheid en depolimerisasie het plaasgevind in die water‐ en CDTA fraksies tussen opberging en rypwordingsperiodes van 3+7 (4.7 kg) en 6+4 (2.7 kg).

Os decisionistas portugueses entre o direito comum e o direito pátrio / Portuguese decisionistas in the early modern age

O objetivo desta tese é determinar se é possível considerar as obras de decisiones como um gênero específico da literatura jurídica portuguesa no Antigo regime, para, em seguida, tentar traçar um perfil das características e dos elementos que as diferenciem dos demais gêneros doutrinários, especialmente dos consilia. O trabalho foi dividido em três partes. A primeira será dedicada aos pressupostos da discussão, que envolvem a opinio communis e a pluralidade de contribuições doutrinárias, característica fundamental do ius commune, e o desenvolvimento da alta jurisdição na Europa do final do Medievo e no começo da Modernidade, focando na tensão entre a indeterminação do direito certo e a existência de tribunais com poderes de decidir em última instância. Em seguida, serão apresentadas as principais características das decisiones no contexto europeu, partindo da comparação entre as obras dos autores mais importantes, como Matthaeus De Afflictis, Guido Papa, Octavianus Cacherano dOsasco, etc, e diferenciando essas obras de gêneros próximos. A segunda parte consistirá na análise aprofundada das obras dos seis mais importantes autores de decisiones em Portugal entre os séculos XVI e XVII (António da Gama, Álvaro Valasco, Jorge de Cabedo, Belchior Febo, Gabriel Pereira de Castro e António de Sousa de Macedo). De cada uma das obras se falará sobre a biografia do autor, os aspectos gerais da obra (quantidade de edições e locais de impressão, por exemplo), a estrutura formal dos textos, o uso dos fundamentos para a construção da argumentação (precedentes, legislação e doutrina) e as matérias abordadas nas decisiones. Por fim, a terceira parte tentará estabelecer as relações entre as decisiones portuguesas do período e o Brasil colonial. / The main purpose of this thesis is to determine whether or not are the decisiones a specific genre in Portuguese legal literature during the Ancient Régime, and then try to describe the most important characteristics and elements that differentiate them from other doctrinal genres. This work is divided into three parts. The first one will deal with the premises of this text, the opinion communis and its plurality of doctrinal works, which was a key feature of ius commune, and the development of high jurisdiction in Europe at the end of Middle Ages and Early Modernity; the focus will be the tension between indeterminacy of ius certum and the existence of certain courts with powers to decide ultimately. Then the main features of European decisiones will be presented from the comparison among the works of the most importante authors such as Matthaeus De Afflictis, Guido Papa, Octavianus Cacherano dOsasco, etc, and later distinguishing these works from some similar genres. The second part will consist in an analysis of the six most important Portuguese works on decisiones during 16th and 17th centuries (António da Gama, Álvaro Valasco, Jorge de Cabedo, Belchior Febo, Gabriel Pereira de Castro and António de Sousa de Macedo); each analyse will approach the following points: authors biography, general aspects of the book (amount of editions and place of the printings, for exemple), the formal strucute of the texts, the arguments in which the decisiones are grounded (Case Law, Statutes/Statutory Law and legal literature) and the subject of the decisio (Public Law, Civil Procedure, Private Law and Criminal Law). Finally, the third part will try to establish the relationship between the Portuguese decisiones and Colonial Brazil.

Avaliação da resistência mecânica à compressão axial da poliuretana de mamona e quitosana associada a cimento de fosfato de cálcio no preenchimento de falhas ósseas do terceiro metacarpiano de equinos / Mechanical evaluation of strength of ricinus communis polyurethane and chitosan with calcium phosphate cement on filling of bone gap in equine third metacarpal bone

Os equinos são animais que, quando adultos, apresentam elevado peso corpóreo e ossos bastante resistentes para sustentar esse peso. Dessa forma, para que ocorra fratura nos ossos do cavalo é necessário um trauma de elevada energia resultando em fraturas cominutivas, que podem formar falhas ósseas impedindo o contato entre os fragmentos ósseos, o que compromete a estabilidade e resistência da osteossíntese. Devido à complexidade desse tipo de fraturas e aos implantes disponíveis apresentarem baixa resistência para o uso nessa espécie e devido a seu comportamento, o tratamento de fraturas em equinos apresenta baixos índices de sucesso. Dessa maneira, o uso de um material osteocondutor com resistência mecânica para o preenchimento dessas falhas poderia elevar a resistência da osteossíntese, levando a melhores índices de sucesso na correção dessas fraturas. O presente estudo teve como objetivo a avaliação biomecânica em laboratório de dois biomateriais substitutos ósseos no preenchimento de falha óssea. Para isso foram utilizados 30 ossos terceiro metacarpiano de equinos que foram submetidos a osteossíntese com placa LCP em sua face dorsal e criada uma falha óssea transversal completa de um centímetro na diáfise média. Dez dessas peças foram submetidas a ensaios biomecânicos não destrutivos até a carga de 1000N, onde foram avaliadas a rigidez, as deformações da peça inteira, da placa e do osso individualmente em diferentes regiões. As outras 20 dessas peças foram submetidas a ensaios destrutivos, onde foram avaliadas a carga e deformação suportada no limite elástico e no ponto de ruptura. Dessa forma, pôde-se observar que houve aumento na rigidez de 699,39N/mm para 2905,38N/mm e 4274,93N/mm devido ao preenchimento da falha com poliuretana de mamona e quitosana, respectivamente. A peça inteira teve sua deformação diminuída de 1,73mm para 0,5mm e 0,35 com carga de 1000N, devido ao preenchimento da falha com poliuretana de mamona e quitosana, respectivamente. A placa teve sua deformação diminuída de 2260,64µd para 320,25µd pelo preenchimento da falha com poliuretana de mamona e para 89,88µd com o preenchimento com quitosana durante a aplicação de 1000N. O osso próximo à falha sofreu maiores deformações tanto com poliuretana de mamona quanto com quitosana, contudo não apresentou maiores deformações em região distante da falha. A peça inteira teve aumento da carga suportada em seu limite elástico de 1008N para 8804N apenas com o preenchimento da falha com quitosana. A peça inteira teve sua deformação diminuída de 1,64mm para 1,26 no limite elástico apenas devido ao preenchimento da falha poliuretana de mamona. A peça inteira teve aumento da força suportada no momento de ruptura de 1660N para 15187N e 11012N com o preenchimento da falha com poliuretana de mamona e quitosana, respectivamente. A peça inteira teve sua deformação máxima no ponto de ruptura diminuída de 5,4mm para 2,16mm apenas com o preenchimento da falha com quitosana. / Adult horses are heavy animals equipped with bones strong enough to support their body weight. High energy trauma is required to produce comminuted fractures, where bone loss may prevent proper fracture reduction and compromise osteosynthesis resistance and stability. In horses, aside from behavioural issues, low success rates associated with comminuted fracture resolution may be due to the complexity of such fractures or the low resistance of bone implants. Therefore, osteoconductive biomaterials with good mechanical resistance would potentially benefit treatment outcomes. This ex vivo study evaluated the biomechanical behaviour of two bone surrogate biomaterials in experimental third metacarpal fractures in horses. Thirty equine third metacarpal bone specimens were submitted to dorsal LCP (locking compression plate) osteosynthesis following creation of a transverse 1 cm wide middiaphiseal defect. Defects were filled either with Ricinus communis polyurethane or calcium phosphate cement-chitosan composite. Ten specimens were submitted to non-destructive biomechanical testing under 1000N maximum load; construct stiffness, construct deformation and isolated deformation of LCP and third metacarpal bone in different regions were evaluated. The remaining 20 specimens were submitted to destructive biomechanical testing; maximum load and deformation within the elastic limit, and to failure were documented. Bone defect repair with RCP or CPC-chitosan composite increased construct stiffness from 699,39N/mm to 2905.38N/mm and 4274.93N/mm and decreased construct deformation under 1000N from 1.73mm to 0,5mm and 0.35 respectively. LCP deformation under 1000N decreased from 2260.64d to 320.25µd and from 2260.64µd to 89.88d following filling of the bone defect with RCP or CPC-chitosan composite respectively. Bone deformation around the defect increased following treatment with RCP or CPC-chitosan composite. However bone deformation away from the defect remained unchanged. Maximum load within the elastic limit increased from 1008N to 8804N when the experimental defect was filled with chitosan composite. Conversely, construct deformation within the elastic limit decreased from 1.64mm to 1.26mm following treatment with RCP. Maximum load to construct failure increased from 1660N to 15187N and 11012N following bone defect repair with RCP or calcium phosphate cement-chitosan composite respectively. However, construct maximum deformation decreased from 5.4mm to 2.16mm when calcium phosphate cement-chitosan composite was used.

Nesse sentido, objetivou-se com o presente trabalho avaliar a influência do espaçamento entre fileiras e da população de plantas no crescimento e produtividade de cultivares de mamona de porte baixo na safra de verão (novembro) e em safrinha (março). Para tanto, foram realizados quatro experimentos, com dois cultivares (FCA-PB e IAC 2028), em duas épocas de cultivo (safra de verão e safrinha), conduzidos durante os anos agrícolas 2007/08 e 2008/09. O delineamento experimental utilizado em todos os experimentos foi o de blocos casualizados, em esquema de parcelas subdivididas, com quatro repetições. As parcelas foram constituídas por quatro espaçamentos entre fileiras (0,45, 0,60, 0,75 e 0,90 m) e as subparcelas por quatro populações iniciais de plantas (25.000, 40.000, 55.000 e 70.000 plantas ha-1). O trabalho de pesquisa foi conduzido em condições de campo, na Fazenda Experimental Lageado da Faculdade de Ciências Agronômicas – UNESP, campus de Botucatu, SP. Na safra de verão foi possível observar que: para a cultivar FCA-PB, o aumento da população de plantas, independente do espaçamento entre fileiras, diminuiu a população final de plantas, a matéria seca da parte aérea, o número de racemos por planta e de frutos por racemo; o aumento da população de plantas nos espaçamentos de 0,60 e 0,75 m entre fileiras aumentou a altura de inserção do primeiro racemo; a maior produtividade de grãos e de óleo da mamoneira FCA-PB foi obtida com população inicial entre 55.000 e 70.000 plantas... / The release of new dwarf castor bean cultivars and indehiscent fruit is suitable for growth in large scale with and mechanized harvest, therefore there is a need to establish which row spacing and plant population is the best for each of these materials can achieve the maximum yield. In this sense, the objective of this study was to evaluate the influence of the row spacing and plant population in the growth and yield of dwarf castor bean cultivars in the summer (November) and out-of-season (March) cropping. Therefore, four experiments were performed, i.e., two cultivars (FCA-PB e IAC 2028) in two cropping seasons (summer and out-of-season), conducted during 2007/08 and 2008/09 agricultural years. The experiment was arranged in a randomized block design with split-plot and four replications. Plots comprised four row spacings (0.45, 0.60, 0.75, and 0.90 m), and subplots comprised four initial plant populations (25,000, 40,000, 55,000, and 70,000 plants per hectare). In summer cropping season was observed: in the FCA-PB cultivar, plant population increasing, regardless of row spacing, decreased final plant population, aboveground dry matter, stem diameter, racemes per plant and fruits per raceme; the plant population increasing, at 0.60 and 0.75 m row spacings, increased the height of the first raceme insertion; the highest grain and oil yields were obtained with a population among 55,000 and 70,000 plants per hectare and 0.45 or 0.60 m row spacings. Metodologia para quantificar ricina em sementes de mamona com o uso de Caenorhabditis elegans /

Resumo: A mamona pode ser utilizada para mais de 700 finalidades diferentes, porém a sua toxidez é o limitante principalmente para o uso restrito da torta originada após a extração do óleo. Para as análises utilizou-se um organismo com o modo de reação a medicamentos e toxinas semelhante ao ser humano, o nematóide Caenorhabditis elegans, o qual tem sido amplamente utilizado pela indústria farmacêutica. O trabalho teve inicio com ensaios preliminares os quais se utilizaram culturas de C. elegans. Estas foram expostas às toxinas que foram colocadas em pequenas cavidades no meio de cultura, porém, devido à falta de praticidade do método, à difícil leitura e sua degradação pela E. coli, passou-se a realizar os testes em placas de 24 poços transparentes contendo apenas água, o nematóide e a toxina. Os resultados mostraram que o teste utilizando placas de 24 poços para exposição do nematóide, e a extração feita através da homogeneização, seguida por banho-maria seguida por centrifugação se mostrou eficiente para medir a ricina sendo semelhantes aos obtidos pelos testes de radioimunodifusão e Elisa, testes bastante específicos, porém de custo mais elevado. O presente trabalho teve por objetivo desenvolver um bioensaio para quantificar a ricina tóxica da mamona, pois a relação entre as análises realizadas por diferentes métodos de quantificação existentes, nem sempre refletem a toxidez real, podendo ocorrer que sementes analisadas por estes métodos, apresentem menor teor de ricina, embora sejam mais tóxicas do que outras sementes que tenham maior teor de ricina quando analisada pelo método. Isto se deve aos métodos empregados que medem além da ricina o RCA Ricinus Communis Aglutimina, um composto menos tóxico do que a ricina pura. / Abstract: The castor oil can be used to 700 different products but its toxicity limits mainly the use of the castor bean cake, the product you have after extract the oil. The present reseach developed a bioassay to quantify the ricin content on castor bean seed because the relationship between the analyses performed by different quantifying methods not always reflect the actual toxicity, witch can occur that seeds analysed by these methods have lower ricin levels although are more toxic than other seeds, because when it test not only the ricin but also the complex ricin + RCA Ricinus Communis Algutimin is measured and the RCA is less toxic than the pure ricin. That is why it was used an organism that has similarities in the way the human body reacts to drugs and toxin, choosing to use the Caenorhabditis elegans that has been used to the pharmaceutical industry. This assay started with preliminary assays using C. elegans colonies that were exposed to the toxins placed in small holes did in the media , but it was hard to read and the E. coli degradation, the method changed to the 24 well plates with water, nematode and the ricin. Caracterização filogenética das proteínas inativadoras de ribossomos (RIPs) de mamona (Ricinus communis L.) e análise da expressão dos genes Rcom RIPs durante o desenvolvimento da semente

As Proteínas Inativadoras de Ribossomos (RIPs) compreendem uma família de enzimas que inibem a síntese protéica através da depurinação de uma adenina específica do RNA ribossomal. Os membros desta família são classificados como RIPs do tipo I, quando possuem somente o RNA-N-Glicosidase e RIPs do tipo II quando além do domínio glicosidase, também apresentam um domínio de lectina. As RIPs foram mais estudadas em plantas, onde a ricina e a aglutinina, ambas RIP do tipo II de mamona (Ricinus communis), estão entre as primeiras descritas. O presente trabalho teve o objetivo de identificar parálogos da ricina e aglutinina, bem como RIPs do tipo I de mamona e analisar as suas relações filogenéticas. Além disso, validar o uso de 14 potenciais genes de referência para qRT-PCR em cinco estádios do desenvolvimento da semente de mamona. O padrão de expressão gênica por RT-qPCR de todas RIPs de mamona identificadas, também foram analisados nestes mesmos estádios. Um total de 18 genes de RIPs foi identificado em mamona (Rcom RIPs), dos quais 10 foram classificados como do tipo II e 8 do tipo I. As topologias das árvores filogenéticas sugerem que as Rcom RIPs foram originadas a partir de múltiplos eventos de duplicação gênica. Dois modelos evolutivos foram propostos para a radiação das Rcom RIPs, baseados em processos de fusão gênica associado ou não a eventos de duplicação parcial. Os genes Act 2/7, EF β, Ubi, TIP e UBC foram os que apresentaram perfil de expressão mais estável e foram selecionados para subsequente normalização dos dados de expressão das Rcom RIPs. Os genes que codificam as Rcom RIPI 3, 4, 5, 7 e 8 e as Rcom RIPII 1, 2, 4, 5, 6 e 8 são transcritos em sementes, sendo que a Rcom RIPII 1 (ricina) e a Rcom RIPII 2 (aglutinina) foram as mais expressas. O presente trabalho apresenta um modelo evolutivo das Rcom RIPs, o qual pode ser extrapolado para outras espécies de plantas. Este trabalho também demonstra o primeiro esforço para a padronização de genes de referência para RT-qPCR em mamona e o primeiro que apresenta a expressão outras Rcom RIPs, além da ricina e aglutinina. / Ribosome inactivating proteins (RIPs) comprise a family of enzymes that inhibit protein synthesis, after depurination of an adenine-specific ribosomal RNA. The members of this family are classified as type I RIPs, which have a RNA-Nglycosidase domain and type II RIPs encompassing a RNA-N-glycosidase and a lectin domain.The RIPs were more studied in plants, where ricin and agglutinin, both type II RIP of castor bean (Ricinus communis), were the first to be described. This work aimed to: 1) identifine paralogous of ricin and agglutinin, as well as the type I RIPs of castor bean; 2) analyze their phylogenetic relationships; 3) validate the use of 14 potential housekeeping genes for qRT-PCR for five developmental stages of R. communis seeds; 4) analyze the pattern of gene expression by RTqPCR of all RIPs castor identified in these same stages. A total of 18 genes that encode RIPs were identified in castor bean (Rcom RIPs), 10 of which were classified as type II and 8 as type I. The phylogenetic trees topologies suggest that Rcom RIPs were originated from multiple events of gene duplications. Two evolutionary models have been proposed for the radiation of Rcom RIPs based on gene fusion processes associated or not to events of partial duplication. The genes Act 2/7, EF β, Ubi, TIP and UBC presented the more stable expression profile and were selected for further RT- qPCR normalization experiments. The Rcom RIPI 3, 4, 5, 7 and 8 and Rcom RIPI 1, 2, 4, 5, 6 and 8 genes are actively transcribed in seeds, whereas the Rcom RIPI 1 (ricin) and Rcom RIPI 2 (agglutinin) were the most expressed. This paper presents an evolutionary model of Rcom RIPs, which can be extrapolated to other plant species. Also, corresponds to the first effort to standardize housekeeping genes for RT-qPCR in castor bean and the first that shows the expression Rcom RIPs, other than ricin and agglutinin.

Salt-dependent regulation of a CNG channel subfamily in Arabidopsis

Background: In Arabidopsis thaliana, the family of cyclic nucleotide-gated channels (CNGCs) is composed of 20 members. Previous studies indicate that plant CNGCs are involved in the control of growth processes and responses to abiotic and biotic stresses. According to their proposed function as cation entry pathways these channels contribute to cellular cation homeostasis, including calcium and sodium, as well as to stress-related signal transduction. Here, we studied the expression patterns and regulation of CNGC19 and CNGC20, which constitute one of the five CNGC subfamilies. Results: GUS, GFP and luciferase reporter assays were used to study the expression of CNGC19 and CNGC20 genes from Arabidopsis thaliana in response to developmental cues and salt stress. CNGC19 and CNGC20 were differentially expressed in roots and shoots. The CNGC19 gene was predominantly active in roots already at early growth stages. Major expression was observed in the phloem. CNGC20 showed highest promoter activity in mesophyll cells surrounding the veins. Its expression increased during development and was maximal in mature and senescent leaves. Both genes were upregulated in the shoot in response to elevated NaCl but not mannitol concentrations. While in the root, CNGC19 did not respond to changes in the salt concentration, in the shoot it was strongly upregulated in the observed time frame (6-72 hours). Salt-induction of CNGC20 was also observed in the shoot, starting already one hour after stress treatment. It occurred with similar kinetics, irrespective of whether NaCl was applied to roots of intact plants or to the petiole of detached leaves. No differences in K and Na contents of the shoots were measured in homozygous T-DNA insertion lines for CNGC19 and CNGC20, respectively, which developed a growth phenotype in the presence of up to 75 mM NaCl similar to that of the wild type. Conclusion: Together, the results strongly suggest that both channels are involved in the salinity response of different cell types in the shoot. Upon salinity both genes are upregulated within hours. CNGC19 and CNGC20 could assist the plant to cope with toxic effects caused by salt stress, probably by contributing to a re-allocation of sodium within the plant.

Memoria para optar al Título Profesional de Ingeniero Agrónomo / Botrytis cinerea Pers. causa niveles importantes de pudrición, los que pueden superar el 10% en peras de exportación en Chile, principalmente en la variedad Packham’s Triumph. La enfermedad postcosecha más importante producida por B. cinerea en peras almacenadas en frío corresponde a la pudrición calicinal, cuyo control se realiza en floración, período en que el hongo produce infecciones latentes que se manifiestan en pudrición tras la madurez de la fruta. En años con primaveras húmedas se ha observado un aumento de la incidencia de la enfermedad, lo cual ha generado cuestionamientos acerca de los tratamientos de control e incertidumbre sobre la capacidad del patógeno de infectar los frutos durante su crecimiento. Los objetivos del presente estudio fueron determinar la ocurrencia de infecciones en campo durante el período de crecimiento de frutos bajo condiciones artificiales de alta humedad con y sin inoculación del patógeno, y determinar el nivel de infección en precosecha para evaluar la efectividad de las medidas de control realizadas por los productores en dos huertos comerciales de la zona central de Chile. En uno de los huertos estudiados se determinó que las inoculaciones lograron generar infecciones en el período de crecimiento de frutos, lo que permite concluir que el patógeno efectivamente puede infectar peras cv. Packham’s Triumph durante el crecimiento de los frutos. Por otro lado, el patógeno fue recuperado desde la cavidad calicinal de fruta no inoculada en crecimiento en ambos huertos, observándose también pudrición calicinal en postcosecha, lo que sería consecuencia de la inefectividad de las aplicaciones de fungicidas realizadas en floración por los productores. Este trabajo establece una base para la realización de estudios en campo en pudrición calicinal de perales en Chile. / Botrytis cinerea Pers. causes significant levels of postharvest decay that may be over 10% in pear fruit in Chile, especially in the cultivar Packham's Triumph. Calyx-end rot is the most important postharvest disease caused by B. cinerea in stored pears, and its control is achieved in the flowering period, when the fungus produces latent infections that manifest in fruit decay after ripening. It has been observed that there is an increased incidence of the disease in years with humid and rainy conditions during spring and summer, which has raised questions about control treatments and pathogen's ability to infect fruit during its growth under these conditions. The objectives of this study were to determine the occurrence of infections in the orchard during fruit growth under artificial high humidity conditions with and without pathogen inoculation, and to determine the level of preharvest infection in order to evaluate the efficacy of control measures taken by the growers in two commercial orchards of central Chile. In one of the studied orchards, inoculations resulted in infections during fruit growth, leading to the conclusion that the pathogen can effectively infect Packham's Triumph pears during this period. Moreover, the pathogen was isolated from the calyx cavity of non-inoculated fruit during the growth period, and postharvest calyx-end decay was also observed, which would indicate the inefficacy of fungicide applications performed by the growers. This work provides basic information for further field studies on calyx-end rot of pears in Chile.

This work was carried out in Fazenda Experimental Vale do Curu, Pentecoste, Ceará, Brazil and the objective of the present study was to evaluate the productive characteristics of three castor cultivars of high agricultural importance (IAC Guarani, Mirante 10 e BRS Paraguaçu) under different levels of irrigation. The experimental delineation used was of the blocks at random with subdivided parcels, composed of five treatments in the parcels, with three cultivars in the sub parcels and three repetitions. The treatments was constituted in five levels of irrigation based on the evaporation of the tank Class “A” ECA (T1 = 0,25 ECA; T2 = 0,50 ECA; T3 = 0,75 ECA; T4 = 1,00 ECA; T5 = 1,25 ECA). The result, found a significant effect between cultivars. The variables length and number of fruits per racemes were higher for the IAC Guarani, while the tenor of oil and number of racemes per plant were higher for the Mirante 10. However the Paraguaçu BRS stood out of the rest of cultivars as for the mass of the raceme, mass of the fruits, mass of hundred seeds and productive potential. There was no significant effect of variable tenor of oil with the blades of irrigation. The Mirante 10 presented the largest tenor of oil (40,64 %), with the maximum blade used of 913,4 mm. The best efficiency of the use of the water was obtained by the treatment T5 (913,4 mm) in the BRS Paraguaçu, what turned the productivity of 2872,42 Kg ha -1. The secondary racemes were that more contributed with the total productivity. / O uso da irrigação em cultivares melhoradas tende a favorecer o aumento da produtividade da mamoneira no semi-árido. Assim, esta pesquisa teve como objetivo avaliar o comportamento das variáveis produtivas de três cultivares de mamoneira de importância agrícola (IAC Guarani, Mirante 10 e BRS Paraguaçu) a cinco níveis de irrigação por gotejo. O experimento foi realizado na Fazenda Experimental Vale do Curu, Pentecoste, Ceará, Brasil. O delineamento experimental foi o de blocos ao acaso com parcelas subdivididas, composto de cinco tratamentos nas parcelas, com três cultivares nas subparcelas e três repetições. Os tratamentos constituíram-se em cinco níveis de irrigação baseados na evaporação do tanque Classe “A” ECA (T1 = 0,25 ECA; T2 = 0,50 ECA; T3 = 0,75 ECA, T4 = 1,00 ECA; T5 = 1,25 ECA). Houve diferença estatística entre as cultivares, as variáveis comprimento e número de frutos por racemos foram maiores para a cultivar IAC Guarani, enquanto o teor de óleo e número de racemos por planta foram maiores para cultivar Mirante 10. Já a BRS Paraguaçu destacou-se das demais cultivares quanto a massa do racemo, massa dos frutos, massa de cem sementes e potencial produtivo. Não houve efeito significativo da variável teor de óleo com as lâminas de irrigação. Verifica-se maior teor de óleo (40,64 %), nas sementes da cv. Mirante 10, com a lâmina máxima aplicada de 913,4 mm. A melhor eficiência do uso da água foi obtida com o tratamento T5 (913,4 mm) na cultivar BRS Paraguaçu, que resultou na produtividade de 2872,42 kg ha-1. Os racemos secundários foram os que mais contribuíram com a produtividade total.
