The role of host switching in the evolution of the fungus-gardening ant symbiosis

The fungus-growing ants have long provided a spectacular example of co-evolutionary integration between distantly related taxa. Their ecological success has been thought to depend largely on the evolutionary alignment of reproductive interests between ants and fungi, following vertical transmission and the ancient suppression of fungal sexuality. In my dissertation I explored the role of lateral cultivar switching on the evolution of the fungus-gardening ant mutualism. First, I provided the first evidence for sexual reproduction in the attine cultivars, together with evidence of extensive independent long-distance horizontal transmission of fungal genes. In fact, fungi have greater gene flow relative to their host ants, crossing the Gulf of Mexico between Latin America and Cuba, over which the ants cannot readily disperse. Second, for the special case of leaf-cutting ants, I show that the cultivar population was largely unstructured with respect to host ant species, and leaf-cutting ants interact largely with a single species of fungus. Finally, I examined the effect of post-glacial expansion on the population structure of the northern fungus-gardening ant Trachymyrmex septentrionalis and compared it with that of its two microbial mutualists: a community of lepiotaceous fungal cultivars and associated antibiotic-producing Pseudonocardia bacteria. This comparison allowed me to examine the effect of historical biogeographic forces, such as climate-driven range shifts, on the population structure of the ants and their microbial symbionts. While neither the cultivar nor the Pseudonocardia genetic structure was correlated with that of the ants, they were significantly, though weakly, correlated with each other. These results suggest that biogeographic forces may act differently on macro- and microscopic organisms, even in the extreme case where some microbial mutualists may be vertically transmitted from generation to generation and share the same joint ecological niche. Thus, binding forces that appear to enforce host fidelity are relatively weak and pairwise associations between cultivar lineages and ant species have little opportunity for evolutionary persistence. Taken together, my studies suggest that mechanisms other than long-term pairwise interactions between ants and fungi (so-called partner fidelity feedback) govern the evolution of the mutualism over evolutionary time. / text

Effect and underlying mechanisms of cultivar mixtures on weed and disease suppression in field pea (<i>Pisum sativum</i>)

Field pea is an important annual crop due to its contribution to soil fertility and other rotational benefits. However, weeds and ascochyta blight limit pea yield, particularly in organic systems. Leafed and semi-leafless pea types differ in lodging resistance, and may affect weeds and disease through differences in canopy light penetration and air flow. Mixtures of the two leaf types may improve weed and disease suppression and yield compared with monocultures of the same cultivars. To test this hypothesis, replicated field experiments were conducted under organic and conventional management in Saskatoon and Vonda, SK, in 2011 and 2012. Mixtures of a leafed and semi-leafless cultivar, CDC Sonata and CDC Dakota, were sown in ratios of 0:100, 25:75, 50:50, 75:25, and 100:0 leafed to semi-leafless pea, at target seeding rates of 88 and 132 plants m-2. Conventionally managed plots were inoculated with ascochyta blight-infested pea straw and received overhead irrigation to encourage disease. Mixtures of 50% or more semi-leafless pea adopted the greater lodging resistance and weed suppression of the semi-leafless cultivar. Mixtures comprised of 25% leafed and 75% semi-leafless pea increased both seed and biomass yield compared with either cultivar grown alone. Yield enhancement was attributed to the leafed cultivar, whose seed yield was 76% higher in mixture than expected based on monoculture yield. Ascochyta blight epidemics were of moderate severity, and leafed and semi-leafless monocultures reached 36 and 43% necrosis in 2011, and 33 and 38% necrosis in 2012, respectively. The disease reaction of mixtures fell between the two component cultivars. At disease onset in 2012, lower light interception and shorter moisture durations coincided with the lower ascochyta blight severity of leafed monocultures. In 2011 and the later phase of the 2012 epidemic, disease severity was negatively associated with vine length, and positively associated with number of nodes and tissue senescence. Despite the advantages of leafed and semi-leafless pea mixtures, the limited selection of leafed cultivars impedes adoption of this technique by growers. For pea breeders, developing mixtures of pea lines isogenic for leaf type may increase yield compared with single cultivars.

Baigiamajame darbe pateikiami lauko eksperimento, vykdyto Aleksandro Stulginskio universiteto bandymų stotyje 2010–2011 m., duomenys. Dirvožemis karbonatingas sekliai glėjiškas išplautžemis (Calc(ar)i-Epihypogleyic Luvisol). Tyrimo tikslas – įvertinti sėjos laiko įtaką linijinių ir hibridinių žieminių rapsų derliaus formavimuisi skirtingos trukmės šiltėjančiu rudens – žiemos periodu. Sėjos laikas ir veislė turėjo esminės įtakos žieminių rapsų pasėlių tankumui rudenį. Rugpjūčio 20 d., rugsėjo 5 ir 10 d. sėtų skirtingų veislių žieminių rapsų pasėlių tankumas skyrėsi esmingai – tankesni buvo linijinės veislės rapsų pasėliai. Žieminių rapsų sudygimui įtakos turėjo meteorologinės sąlygos dygimo metu, ypač – kritulių kiekis ir intensyvumas. Sėjos laikas turėjo esminės įtakos rapsų biometriniams rodikliams rudenį prieš žiemojimą. Veislės esminė įtaka nustatyta tik lapų skaičiui pasėjus žieminius rapsus rugpjūčio mėnesį. Pasibaigus atsinaujinusiai vegetacijai rudens–žiemos laikotarpiu, sėjos laiko įtaka rapsų biometriniams rodikliams išliko esminė. Didesnę antžeminės dalies masę, lapų skaičių bei šaknų masę užaugino ‘Kronos’ veislės žieminiai rapsai. Atsinaujinus vegetacijai pavasarį, sėjos laikas turėjo esminės įtakos antžeminės dalies masei bei šaknų masei, tuo tarpu lapų skaičiui esminės įtakos nebuvo. ‘Kronos’ veislės žieminiai rapsai suformavo esmingai didesnę šaknų masę, juos sėjant rugpjūčio 10 d. Soil of the experiment field - (Calc(ar)i-Epihypogleyic Luvisol). Aim of the research – to estimate the effect of the linear and hybrid winter rape yield formation during the warming autumn – winter period. Sowing time and variety had significant effect on winter rape crop thickness in autumn. Thickness of the crop of the considered winter rape varieties sown August 20, September 5 and 10 differed, considerably. Thicker crop was recorded for the linear rape variety. Meteorological conditions especially precipitation amount and intensity were found to have significant effect on winter rape germination. Sowing time contributed significantly to the biometric rape indices before wintering. The obtained data revealed the effect of variety on the number of winter rape leaves sown in August. The effect of the sowing time after the end of renewed vegetation in autumn-winter period was found to be significant. ‘Kronos’ variety winter rape produced higher above-ground mass, leaves number and root mass. The sowing time was found to have a significant effect not only on the above-ground mass but also on root mass. ‘Kronos’ variety winter rape sown August 10 were recorded to produce considerably higher root mass. It is a crop of low productivity, and may be superior when management in fertilization is appropriate. However, more information about the nutrition of the crop is needed. Nitrogen is one of the nutrients that most limits the production of sesame because it is responsible for important functions of metabolism and nutrition of the crop. Therefore, the objective of this work was to evaluate the cultivars of irrigated sesame in the function of nitrogen fertilization in two agricultural crops. Experiments were conducted at the Experimental Farm Rafael Fernandes, belonging to the Federal Rural Semi-Arid University, Mossoró-RN, from February to May (1st crop) and July to October (2nd crop) in 2016. The experimental design was in randomized complete blocks with four replications in a subdivided plots scheme, being allocated in the plots five nitrogen doses (0, 30, 60, 90 and 120 kg ha-1), and the four sesame cultivars in the subplots (CNPA G2 , CNPA G3, CNPA G4 and BRS Silk). The variables evaluated were: nitrogen content in the leaf diagnosis, plant height, stem diameter, number of leaves and capsules, dry mass (leaf, stem, capsules and plants), seed productivity, agronomic efficiency, oil and protein content. The dose of 120 kg ha-1 of nitrogen provided the best agronomic performance for sesame. The cultivar CNPA G4 had the highest productive performance. The cultivar BRS Silk had a higher oil content. The second crop provided better agronomic performance to the sesame crop / O gergelim é uma oleaginosa que apresenta grande potencial econômico, devido às possibilidades de exploração. É uma cultura de baixa produtividade, podendo ser superior quando há manejo adequado na adubação. Contudo, maiores informações sobre a nutrição da cultura precisam ser obtidas. O nitrogênio é um dos nutrientes que mais limitam a produção do gergelim, por ser responsável por importantes funções do metabolismo e da nutrição da cultura. Diante disso, o objetivo deste trabalho foi avaliar cultivares de gergelim irrigado em função de adubação nitrogenada em duas safras agrícolas. Os experimentos foram conduzidos na Fazenda Experimental Rafael Fernandes, pertencente à Universidade Federal Rural do Semi-Árido, Mossoró-RN, no período de fevereiro a maio (1ª safra agrícola) e julho a outubro (2ª safra agrícola) de 2016. O delineamento experimental foi em blocos completos casualizados com quatro repetições em esquema de parcelas subdivididas, sendo alocadas nas parcelas cincos doses de nitrogênio (0, 30, 60, 90 e 120 kg ha-1), e nas subparcelas as quatros cultivares de gergelim (CNPA G2, CNPA G3, CNPA G4 e BRS Seda). As variáveis avaliadas foram: teor de nitrogênio na folha diagnóstico, altura de plantas, diâmetro do caule, número de folhas e cápsulas, massa seca (folha, caule, cápsulas e plantas), produtividade de sementes, eficiência agronômica, teor de óleo e proteína. A dose de 120 kg ha-1 de nitrogênio propiciou o melhor desempenho agronômico para o gergelim. A cultivar CNPA G4 teve o maior desempenho produtivo. A cultivar BRS Seda teve maior teor de óleo. The aim of this diploma thesis was to evaluate the effects of conventional and organic crop management on cooking quality of potatoes. In one-year potato field trials (year 2010) with different crop management (conventional versus organic), 13 table potato genotypes (Adéla, Bionta, Karin, KE 100/10, Laura, Madona, Magda, Princess, Red Anna, Rosara, Solara, Satina,Terka) were used. The cooking quality (i.e. colour of the flesh, consistency, mealiness, moistness, flavor and darkening) was subjectively assessed using sensory analysis according to VORAL (1996) method. Only structure of boiled potato tubers was significantly affected by different crop management (4.8 % from total variability). Genotypically declared cooking types in used potato genotypes were confirmed in both production systems.

Conclui-se que a adubação nitrogenada e fosfatada aplicada na sua maior dose (120 kg ha-1) e (309 kg ha-1), respectivamente, obtiveram maiores produtividades, o híbrido ou cultivar não influenciaram a produtividade e o lucro operacional no tratamento mais oneroso foi negativo R$ -430,11/ha, apresentando a cultura uma dificuldade de subsistir nesse sistema de cultivo / The objective was to determine nitrogen and phosphorus combined with hybrid and the castor bean cultivar planted in conservation tillage, and carry through economic analysis of production costs and profitability. It was developed at the Experimental Farm UNESP - Single Island, located in Selvíria (MS), presenting as geographic coordinates 51 o 22 'W and 20 o 22' S, altitude approximately 335 m in Haplorthox. The experimental design was randomized blocks. The treatments were a combination of a hybrid and a cultivar (Lyra and IAC-2028) respectively, 2 P rates (0 and 309 kg ha-1 of single super phosphate) and five nitrogen rates (0, 15, 30, 60 and 120 kg ha-1). Sowing occurred on 07/06/2009 and crop was irrigated by center pivot system using the espaçamneto 0.9 m between rows and 0.9 m between plants. This thesis deals with an inventory survey of tree species in the castle park in Žinkovy. The park was founded in 1896 - 1898 and is one of the best preserved castle parks on the southern Pilsen (Czech Republic). Since its inception the owners, management and appearance of the park have been changed several times. The inventory survey covers 81 tree species including their cultivar varieties. Furthermore the thesis compares the results with previous inventory surveys, and preserved documents and provides concept of new management measures. A list of the tree species and these comparisons are summarized in the appendix of the thesis.

Buscou-se produzir híbridos somáticos entre laranjas doces (Citrus sinensis) com tangerinas, mexericas (C. reticulata, C. reshni, C. sunki, C. deliciosa), tangores (C reticulata x C. sinensis) ou tangelo ‘Orlando’ (C. reticulata x C. paradisi), além da avaliação da resistência do híbrido somático laranja ‘Hamlin’ (C. sinensis) + mexerica ‘Montenegina’ (C. deliciosa) a Xanthomonas axonopodis cv. citri e Xylella fastidiosa. Foram utilizados como fonte de protoplastos calos embriogênicos e folhas coletadas de plântulas germinadas in vitro e de plantas cultivadas em casa-de-vegetação. Após o isolamento, protoplastos provenientes de calos e/ou suspensões embriogênicas foram fundidos quimicamente por polietilenoglicol (PEG) com protoplastos não embriogênicos oriundos de mesofilo foliar. Os calos provenientes da fusão foram induzidos a formação de embriões somáticos para posterior germinação e regeneração de plantas. As plantas regeneradas foram individualizadas, enraizadas ou microenxertadas e aclimatizadas em casa-de-vegetação. A confirmação da hibridação somática foi feita por análise morfológica, análise do DNA com marcadores moleculares do tipo RAPD, determinação da ploidia com a contagem do número de cromossomos e/ou citometria de fluxo. Foram obtidos dois híbridos somáticos do genitor embriogênico laranja ‘Hamlin’ com os genitores não embriogênicos mexerica ‘Montenegrina’ e tangerina ‘Dancy’. Ambos híbridos somáticos obtidos podem apresentar características complementares dos genitores, podendo ser utilizados diretamente como copa e em programas de melhoramento de copa de citros. O híbrido somático laranja ‘Hamlin’ com mexerica ‘Montenegrina’ foi avaliado mostrando resistência a Xanthomonas axonopodis cv. citri e a Xylella fastidiosa. / This research aimed to produce new somatic hybrid combinations between sweet orange (Citrus sinensis) with mandarins (C. reticulata, C. reshni, C. Sunki, C. deliciosa), tangors (C reticulata x C. sinensis) or ‘Orlando’ tangelo (C. reticulata x C. paradisi), and also evaluate the somatic hybrid ‘Hamlin’ sweet orange (Citrus sinensis) + ‘Montenegrina’ mandarin (Citrus deliciosa) for resistance to Xanthomonas axonopodis pv. citri and Xylella fastidiosa. Protoplast sources included embryogenic calli or suspension-culture derived calli, and leaves collected from plants cultivated in vitro or in screenhouses. After protoplast isolation, embryogenic protoplasts were chemically fused by polyethylene glycol (PEG) with mesophyll-derived non-embriogenic protoplasts. Fusion-derived calli were further cultured to embryo induction, germination, and plant regeneration. Regenerated plants were individually rooted or micrografted, and further acclimated in screenhouse. Somatic hybridization was confirmed by analysis of leaf morphology, molecular analysis by RAPD markers, ploidy determination by chromosome counting or flow cytometry. The producion of the somatic hybrids ‘Hamlin’ sweet orange + ‘Montenegrina’ mandarin and ‘Hamlin’ sweet orange + ‘Dancy’ mandarin was confirmed. These hybrids may have complementary traits from both progenitor and be used directly as scion cultivars or as parental lines in scion improvement programs. ‘Hamlin’ sweet orange + ‘Montenegrina’ mandarin somatic hybrid was resistant to Xanthomonas axonopodis pv. citri and Xylella fastidiosa.

Índice de seleção em cultivares de algodoeiro herbáceo / Selection index in upland cotton cultivars

Os índices de seleção são combinações lineares de valores fenotípicos, resultando numa medida que concentra em único valor, os atributos positivos e negativos de cada genótipo para vários caracteres. Por necessitarem das estimativas das variâncias e covariâncias genotípicas e fenotípicas dos caracteres, esses índices são mais indicados para programas de seleção recorrente. Contudo, existem também outros tipos de índices não lineares, conhecidos como não paramétricos que, por não precisarem das estimativas dos parâmetros genéticos, podem ser utilizados na seleção de cultivares em fase final dos programas de melhoramento. Para essa situação, uma alternativa bastante promissora é o emprego do índice de seleção de cultivares proposto por Garcia (1998) que é uma medida de distância genética entre cada genótipo a um ideótipo. Esta técnica permite a aplicação de teste de comparação de médias e a identificação de genótipos com desempenhos abaixo dos critérios mínimos definidos pelo melhorista. Este trabalho teve como objetivos avaliar a viabilidade do índice proposto por Garcia (1998) na identificação de genótipos superiores de algodão, estimar os parâmetros de estabilidade do índice e verificar quanto da variação fenotípica do índice é de natureza genotípica. Foram utilizados quinze genótipos de algodoeiro do Ensaio Estadual de Algodoeiro Herbáceo (2000/01) avaliados em 27 locais sob o delineamento de blocos ao caso com quatro repetições. Os caracteres avaliados foram produtividade de algodão em caroço (kg/ha), altura de planta (cm), peso de um capulho (g), porcentagem de fibra (%), resistência da fibra (gf/tex), comprimento da fibra (mm), finura da fibra (IM), ramulose (notas) e bacteriose (notas) Os resultados obtidos evidenciaram que o índice proposto por Garcia (1998) identificou com eficiência as cultivares superiores com destaque para BRS AROEIRA, CNPA 97- 4565 e DELTA OPAL. As cultivares mais estáveis foram: CNPA CO 99-01, BRS IPÊ e BRS JATOBÁ, para o índice de seleção e BRS AROEIRA, BRS IPÊ e CNPA 99-01, para a produtividade. Os índices de seleção apresentaram médio e alto coeficiente de determinação genotípica, indicando que é possível antever o sucesso com a seleção utilizando as estimativas do índice proposto por Garcia (1998). Neste aspecto, recomenda-se complementar o emprego do índice de seleção com informações sobre a estabilidade e adaptabilidade das cultivares selecionadas sob condições favoráveis e desfavoráveis. / Selection indices are linear combinations of phenotypic values, which combine in one unique value the positive and negative attributes of each genotype for the characters evaluated. Because these indices need estimates of genetic and phenotypic variances and covariances, their use is best suited in recurrent selection programs. However, other types of non-linear indices also exist, and are known as non-parametric indices; these can be used in selection of cultivars at the final stage of breeding programs, since genetic parameter estimates are not a requirement. In this situation, the selection index developed by Garcia (1998) is a promising alternative. This index is an estimate of the genetic distance from each genotype to an “ideotype”. This technique allows the comparison of means and the identification of genotypes with performances that meet the minimum requirements chosen by the breeder. The present work had the following objectives: to evaluate the viability of the Garcia index for the identification of superior genotypes, to estimate the stability parameters of the index and verify how much of the phenotypic variation is genotypic. Fifteen genotypes of the Upland cotton Variety Trials (Ensaio Estadual de Algodoeiro Herbáceo) – 2000/01 were evaluated in 27 locations in randomized complete block design with four replications. Characters assessed were: seed cotton yield (kg/ha), plant height (cm), boll mass (g), lint percentage (%), fiber strength (gf/tex), fiber length (mm), lint micronaire (IM), ramulosis (scores) and bacterial blight (scores). The data showed that the Garcia index identified superior cultivars successfully, as BRS AROEIRA, CNPA 97-4565 and DELTA OPAL. The most stable cultivars were: CNPA CO 99-01, BRS IPÊ and BRS JATOBÁ for selection index and BRS AROEIRA, BRS IPÊ e CNPA 99-01 for yield. The selection indices showed medium and high coefficient of genotypic determination indicating that is possible to expect selection success when the Garcia index is used. We suggest to complementing the use of the selection index with informations about stability and adaptability of the selected cultivars at favorable and unfavorable environments.

Assim como outros frutos, o pêssego apresenta compostos bioativos, mas que ainda pouco têm se estudado, principalmente a estabilidade destes compostos frente ao processamento. Assim, o objetivo deste estudo foi avaliar o impacto do processamento de pêssego sobre o conteúdo de compostos bioativos em pêssego em calda e pessegada e avaliar a estabilidade destes compostos durante o período de estocagem destes produtos. Para isto foram utilizadas três cultivares de pêssegos (Prunus pérsica): Santa Áurea, Esmeralda e Maciel, frutos provenientes de cinco produtores da região de Pelotas/RS, e cultivados sob sistema de rastreabilidade da fruta, sendo avaliados durante duas safras consecutivas. Os pêssegos foram processados na forma de doce em calda e de pessegada, estocados e posteriormente avaliados. Foram realizadas as análises de pH, acidez titulável (AT), sólidos solúveis (SS), relação SS/AT, total de carotenoides, carotenoides individuais, total de compostos fenólicos, compostos fenólicos individuais, vitamina C e total de antocianina s. A cultivar Esmeralda destacou-se entre as demais estudadas demonstrando relação SS/AT:20; representando excelente qualidade para processamento, seguido da cultivar Santa Áurea e Maciel. Ocorreram perdas significativas de compostos bioativos durante a elaboração e estocagem de pêssego em calda, sendo a vitamina C (40% perdas) e os carotenoides (77%perdas) os componentes mais instáveis. Após elaboração de pessegada não foram verificadas perdas significativas de compostos fenólicos sendo intensificadas durante o período de estocagem, atingindo valores acima de 57% de perdas. A escolha de cultivares para elaboração de pêssego em calda e doce em massa apresentou valorização das características sensoriais, físico-químicas e bioativas. / The Brazilian market for products of temperate climate fruits is very promising, but there is no uniformity on fruit quality, processing failures that favor waste in the field and in the industry, generating correction of defective lots. As other fruits, peach presents bioactive compounds, but so far still not studied, mainly the stability of these compounds against processing. The objective of this study was to evaluate the impact of peach processing on the content of bioactive compounds in peach compotes and in sweet peach mass, and evaluate the stability of these compounds during the storage of these products. For this, three cultivars of peach (Prunus persica L.) were used: Santa Áurea, Esmeralda and Maciel, fruits were obtained from five producers of Pelotas / RS, and grown under fruit traceability system, being evaluated during two consecutive harvests. The peaches were processed as sweet peach mass and peach compote, stored and later evaluated. Analyzes of pH, titratable acidity (TA), soluble solids (SS), SS / TA, total carotenoids, individual carotenoids, total phenolic compounds, individual phenolic compounds, vitamin C and total anthocyanin were analysed. The Esmeralda cultivar stood out among the others showing SS / TA ratio: 20; representing excellent quality for processing, followed by cv. Santa Aurea and Maciel. Significant losses of bioactive compounds during the preparation and storage of peaches compotes were observed, with vitamin C (40% loss) and carotenoids (77% loss) the most unstable components. After sweet peach mass processing, no significant losses of phenolic compounds was observed, but the losses were intensified during storage period, where values above 57% losses were observed. The choice of cultivars for preparation of peaches compotes and sweet peach mass, enhanced the sensory, physicochemical and bioactive characteristics.
