These proteins comprise among others, (i) ribonucleases, (ii) antifungal proteins, (iii) lectins and (iv) trypsin inhibitor with antiviral, antifungal and anticancer activities. The aim of this project was to isolate antipathogenic plant proteins including a ribonuclease from American ginseng branch roots, a trypsin inhibitor from rambutan seeds, defensin-like antifungal peptides from borlotti beans and king pole beans, and lectins from borlotti beans, Japanese large pinto beans and Pseudostellaria heterophylla. / The isolated 26-kDa ginseng branch root ribonuclease was monomeric with a novel N-terminal amino acid sequence. It exhibited maximal robonucleolytic activity toward yeast tRNA at pH 5 and 60℃. It inhibited proliferation of MCF7 human breast cancer cells and HepG2 human hepatoma cells. It also inhibited the activity of HIV-1 reverse transcriptase. / Both borlotti bean lectin and Japanese large pinto bean lectin were dimeric with a subunit molecular mass of 32-kDa. They were stable from 0℃ to 60℃ and from pH 3 to pH 12. Borlotti bean lectin was xylose-specific whereas Japanse large pinto bean lectin was galactose-specific. The 33-kDa Pseudostellaria heterophylla lectin could not be inhibited by the simple sugars tested. It was stable from 0℃ to 60℃ and from pH 2 to 5. All three isolated lectins suppressed proliferation of MCF7 and HepG2 cells and inhibited HIV-1 reverse transcriptase. / The isolated 21-kDa rambutan trypsin inhibitor has relatively high pH stability and thermostability, and exhibited HIV-1 reverse transcriptase inhibitory activity and antiproliferative activity toward a variety of tumor cells. This thesis makes an assessment of the factors influencing the involvement of the Australian Defence Organisation (ADO) in homeland security roles since 11 September 2001 (9/11). This is approached on a largely empirical basis, using document analysis and case studies supported with interviews with key individuals and experts. The thesis commences with an Introduction that provides brief context for the thesis and specifies its central question as ???what factors have shaped the role of the ADO in Australia's response to the homeland security environment that has emerged since 11 September 2001.??? Chapter One provides an historical and theoretical context for the key concepts of homeland security and the challenges confronting Western governments in the homeland security arena. Chapter Two explores the implications of those challenges for Australia, before outlining the research method and providing a literature review. Chapter Three is an historical exposition of homeland security in Australia from British settlement in 1788 until 2001. The Chapter examines events in increasing detail in the 30 years immediately prior to 2001, including a detailed case study of ADO support to the Sydney 2000 Olympic and Paralympic Games, before drawing some broad conclusions on the Australian experience of the involvement of its Defence Organisation in homeland security pre-9/11. Chapter Four establishes the pre-9/11 status quo in relation to the ADO???s involvement in homeland security role before analysing the general pattern of those roles. Chapter Five analyses and draws conclusions about the reasons for the ADO???s pre-9/11 involvement in homeland security roles, introducing an hypothetical construct to explain causal factors. Chapter Six examines the ADO???s involvement in homeland security roles post-9/11, including cases studies of ADO support to the conduct of the 2002 Commonwealth Heads of Government Meeting and the Melbourne 2006 Commonwealth Games. Chapter Seven analyses and identifies the factors led to the ADO???s pattern of involvement in homeland security post-9/11, further developing the hypothetical construct introduced in Chapter 5. Chapter Seven also contains supporting case studies on the ADO???s contribution to Australia???s national chemical, biological, radiological and nuclear response capability and on the state of New South Wales??? homeland security capabilities. Chapter Eight draws overall conclusions, including recommendations for Australian policy development and areas for further research. The essential conclusion reached is that the ADO???s involvement in homeland security roles, both before and since 9/11, has been shaped mostly by pragmatic political and managerial considerations of governments. Developments have normally occurred in an episodic and incremental fashion in response to ???trigger events,??? although 9/11 altered this pattern somewhat by acting as a ???threshold??? event that re-calibrated demands and expectations for ADO involvement. Supporting Appendices provide detail on the Australian Government???s strategic guidance on ADO involvement in homeland security since 1973; on Australia???s policy for Defence Assistance to the Civilian Community and Defence Force Aid to the Civil Authorities; and on the involvement of former Department of Defence employees in non-Defence homeland-security related roles. Plant-Herbivore Interactions and Evolutionary Potential of Natural Arabidopsis lyrata PopulationsPuentes, Adriana January 2012 (has links)
In this thesis, I combined field, greenhouse and common-garden experiments to examine the ecological and evolutionary consequences of plant-herbivore interactions and the genetic architecture of fitness-related traits in the insect-pollinated, self-incompatible, perennial herb Arabidopsis lyrata. More specifically, I examined (1) whether damage to leaves and inflorescences affects plant fitness non-additively, (2) whether trichome production is associated with a cost in terms of reduced tolerance to leaf and inflorescence damage, (3) whether young plant resistance to a specialist insect herbivore varies among populations, and (4) whether the evolution of flowering time, floral display and rosette size is constrained by lack of genetic variation or by genetic correlations among traits. A two-year field experiment in a Swedish population showed that damage to rosette leaves and to inflorescences can affect both current and future plant performance of A. lyrata, and that effects on some fitness components are non-additive. A two-year field experiment in another Swedish population indicated that trichome-producing plants are not less tolerant than glabrous plants to leaf and inflorescence damage. In a greenhouse experiment, acceptability of young plants (5-6 weeks old) to ovipositing females and damage received by Plutella xylostella larvae varied considerably among twelve A. lyrata populations. Both oviposition and leaf damage were positively correlated with rosette size, but trichome density in the trichome-producing morph was apparently too low at this developmental stage to influence resistance to P. xylostella. In a common-garden experiment, flowering time, floral display and rosette size varied among four Scandinavian A. lyrata populations, and displayed significant additive genetic variation in some populations. Yet, strong genetic correlations between flowering start and number of flowers, and between petal length and petal width suggest that these traits may not evolve independently. Taken together, the results indicate the need to consider possible long-term and non-additive effects of herbivore damage to different plant parts, that there is no trade-off between trichome production and tolerance to herbivory, that the importance of morphological defenses against herbivory may change through plant ontogeny, and that considerable genetic variation for traits such as flowering time and floral display can be maintained in natural plant populations.

Chemical cues affecting susceptibility of gorgonian corals to fungal infectionHicks, Melissa Kathryn 28 November 2005 (has links)
Coral diseases have become more prevalent and destructive over the past 20 years, possibly due to an increase in stressful environmental factors that may weaken corals defenses against disease. Aspergillosis is a disease caused by the fungus Aspergillus sydowii, which apparently infects only two species of gorgonian corals in the Caribbean Ocean (Gorgonia ventalina and G. flabellum). We hypothesized that the differential resistance to infection is caused by differences in chemical defenses among gorgonians. Freeze-dried gorgonian powders and extracts deterred fungal growth, but potencies varied among gorgonian species and among fungi. Extracts and powders generated from G. ventalina all strongly inhibited fungal growth. Since G. ventalina was predicted to have weak antifungal chemical defenses compared to gorgonians not known to suffer from aspergillosis, we concluded that gorgonian susceptibility to fungal infection is determined by factors other than, or in addition to, chemical defenses.
In order to investigate specific gorgonian antifungal strategies, we attempted to use bioassay-guided fractionation to isolate antifungal compounds from four gorgonians: Gorgonia ventalina, Briareum asbestinum, Eunicea succinea, and Pseudopterogorgia americana. We succeeded in isolating two antifungal compounds, diastereomers of 9,11-seco-24-hydroxydinosterol, from the gorgonian Pseudopterogorgia americana. This compound was previously identified by other groups, but this study is the first to establish its antifungal activity. At natural concentration, one diastereomer of 9,11-seco-24-hydroxydinosterol inhibited the growth of three different fungi, suggesting that at least this diastereomer may possess broad-spectrum antifungal activity.
The results from our survey of gorgonian chemical defenses indicate that susceptibility to aspergillosis cannot be explained by chemical growth inhibition alone. Further areas of investigation include induction of gorgonian chemical defenses, examination of growth-inhibiting mechanisms of antifungal metabolites, and identification of non-chemical factors affecting gorgonians vulnerability to fungal infection.
