This thesis deals with the possibilities of using on-line diagnostic tools in selecting employees. The theoretical part of the thesis summarizes the theoretical basis for selection of employees, psychodiagnostics and on-line diagnostics. Attention is paid to psycho diagnostic characteristics and determination of preferences and risks of on-line diagnostics. The practical section examines and assesses the individual tools on-line diagnostics available for selection of employees on the Czech market - specifically, by the company Access Assessment s.r.o., Assessment Systems s.r.o., cut-e czech s.r.o and www.SCIO.cz, s.r.o. The tools are evaluated on the basis of psychometric features, factor is the standardization, objectivity, reliability and validity, as these characteristics determine the quality of the questionnaire or test. The result is a recommendation of an on-line questionnaire and tests that are for standard practice in the selection procedure most applicable.

Thesis deals with the design, implementation and verification of functionality of station designed for measuring of acoustic emissions from pneumatic cylinders for the purpose of fault diagnosis. The first part of this thesis includes an overview of previous work dealing with the diagnosis of pneumatic systems, also works dealing with the diagnosis of various machine parts using acoustic emission. Further work contains a description of the proposed conceptual design and final draft is described in detail, including occurred complications and their solutions. The last part describes the results of measurements of acoustic emissions from damaged and undamaged cylinders. Functionality of the station is verified by comparing the frequency spectra and envelopes of measured acoustic emission signal.

Title of diploma thesis: Determination of biochemical parameters and their diagnostic and prognostic value in sepsis Sepsis is a relatively common complication that may be signed by the increased mortality of patients. It affects all ages, regardless of gender. Occurrence is more frequent in polymorbid and immunosuppressed patients. The most effective method to prevent the development of septic complications is quick and accurate diagnosis. In the period from 1.6.2012 to 01.4.2014, we observed 697 patients hospitalized due to the development of septic complications and we analyzed statistically tracked markers: presepsin, procalcitonin, C - reactive protein, interleukin - 6, lactate, D - dimer, fibrinogen and quantity of leukocytes to determine their predictive and diagnostic value. Largest correlation was observed between comparison of presepsin with procalcitonin and presepsin with C - reactive protein, which reflected rapidly changing status of patients. Cercariae of the genus Trichobilharzia can penetrate not only the skin of definitive hosts (ducks), but they are also able to penetrate the skin of accidental hosts (mammals). As a result of the penetration, the inflammatory response known as cercarial dermatitis appears. The goal of our thesis is to detect parasite DNA in the serum and cerebrospinal fluid of infected ducks, and also in the serum of infected mice. By using PCR with primers designed for a tandem repeated sequence, we were able to detect 1 femtograms of parasite DNA isolated from sera of infected ducks. We were able to amplify parasite DNA only from 16 samples of sera and cerebrospinal fluid of the infected ducks, but we were not able to do so with the serum of mice. Sera of infected mice were also tested by ELISA and Western blot. The homogenates of T. regenti (TRhc), T. szidati (TShc) and S. mansoni (SMhc) cercariae were selected as an antigen. The results showed progressive increase in the level of IgM antibody from 10 days after 1st infection and also increase of the level of IgG from the 2nd infection. 10 days after the 4th infection, the level of IgM and IgG gradually declined, but the level of antibodies 100 days after the 4th infection was still higher if compared to uninfected mice. Submitted diploma thesis deals with laboratory diagnostics of filamentous micromycetes (moulds) genus Fusarium in the laboratories of medical mycology. In the first part of the diploma thesis is dedicated space for general familiarization with filamentous microscopic fungi. Next separate chapter describes the genus Fusarium, especially in relation to human medicine. The final part of the diploma thesis summarizes current possibilities of laboratory diagnostics of filamentous micromycetes, focusing on the genus Fusarium. Invasive infections caused by filamentous fungi are increasingly common complication, especially in immunocompromised patients. In most cases they affect the patients with hematologic malignity and patients after hematopoietic stem cell transplantation or solid organ transplantation. The most common etiological agent of such infections is unambiguously the Aspergillus genus. However, in recent years, increasingly cases of infections due to less common filamentous fungi, including Fusarium spp. are described. Invasion fungal infections are associated with very high mortality. The basic requirement thus becomes the speed and accuracy of diagnosis. Bird schistosomes of the genus Trichobilharzia are known as causative agents of hyper-immune skin reaction called cercarial dermatitis (swimmer's itch). They use pulmonary water snails from family Lymnaeidae as the intermediate host and mostly anatid birds as the definitive host. The first larva, miracidium, actively moves in water environment, penetrates the snail and develops to the mother sporocyst. Then the daughter sporocysts are formed and migrate to the hepatopancreas of the snail where the high number of cercariae is assexually produced. Cercariae leave the intermediate host, actively move in a water and penetrate the skin of definitive host. Within a host body they mature and lay eggs. Cercariae can penetrate also the mammalian skin, including human, where they are immediately eliminated by the immune system of the host, which is followed by inflammatory reaction. Until now, for humans, there is no effective method enabling to differ cercarial dermatitis from other hyper-immune skin reactions and for birds the reliable diagnostic method of trichobilharziasis is missing. The aim of the study was to identify the relationship between selected fitness skills and the performance demonstrated in nomination races (NR) to the Czech Republic national team (NT) and for the Olympic Games. At the same time, we wanted to determine the optimal level of conditioning assumptions and define a model single-canoe sportsman. We were also searching to determine the differences between Senior NT and Junior NT. The research sample (n=17) of the Czech highest competitions series (Czech Cup) participated in testing to determine level of strength, endurance and partly speed assumptions. There were also measured anthropometric parameters (body dimensions and composition) in detail. Testing took place only 5 weeks before NR. The condition indicators were subsequently correlated with the NR performance. For indicators for which significant and at least moderate correlation were found, the possibility of performance prediction was also found. The highest correlations with performance were found in case of on-water sprints: 20m (rs=0,65), 40m (rs=0,86, resp. rs=0,62), 80m (rs=0,58) and 200m (rs=0,795).
