Dietas e conteúdo alimentar de alguns Carabidae (Coleoptera) observados na região nordeste do Estado de São Paulo /

A determinação do tipo de alimento utilizado por carabídeos encontra-se entre os principais aspectos considerados em estudos sobre hábitos alimentares. O presente estudo teve como objetivo avaliar o efeito de diferentes tipos de dietas sobre o consumo, viabilidade e número de ovos produzidos pelos carabídeos Abaris basistriata Chaudoir, 1873, Selenophorus seriatoporus Putzeys, 1878, Tetracha brasiliensis (Kirby, 1818), Scarites sp.2, Scarites sp.3. e determinar o tipo de material alimentar presente no aparelho digestivo dessas cinco espécies de carabídeos. As dietas avaliadas foram: larvas de Tenebrio molitor Linnaeus, 1758 (Coleoptera, Tenebrionidae), carne moída, ração para gato, pulgão Schizaphis graminum (Rondani, 1852) (Insecta, Hemiptera, Aphididae), sementes de Brachiaria decumbens Stapf (Poaceae) e mistura das dietas. Analisou-se o material presente no inglúvio e proventrículo de 10 carabídeos adultos de cada espécie coletada no campo. As espécies A. basistriata, T. brasiliensis, Scarites sp.2 e Scarites sp.3 não consumiram sementes de B. decumbens. A dieta mais consumida por adultos das cinco espécies foi larvas de T. molitor. As dietas mais favoráveis à capacidade reprodutiva de A. basistriata, S. seriatoporus e T. brasiliensis foram S. graminum e larvas de T. molitor/mistura das dietas, respectivamente. Por outro lado, as dietas de larvas de T. molitor e mistura das dietas foram as mais favoráveis para a criação de A. basistriata, S. seriatoporus e T. brasiliensis em condições de laboratório. No conteúdo alimentar de todas as espécies foram encontrado fluídos e resíduo de tegumento de artrópodes, sem contudo exibirem material vegetal. Foram identificados fragmentos de antenas, pernas, asas e mandíbulas no conteúdo alimentar de A. basistriata, T. brasiliensis, Scarites sp.2 e Scarites sp.3. / Abstract: Ground beetles or carabids are collective terms for the beetle family Carabidae (Insecta, Coleoptera), which contains many species considered important predators associated with agricultural cultures. Among the main factors considered in studies on food habits is determination the type of food used by carabid. The present study aimed to evaluate the effect of different diet types on consumption, viability and number of egg produced by Abaris basistriata Chaudoir, 1873, Selenophorus seriatoporus Putzeys, 1878, Tetracha brasiliensis (Kirby, 1818), Scarites sp.2 e Scarites sp.3. and determination the type of alimentary material in the gut these five carabid. The diets assessed were: larvae of Tenebrio molitor Linnaeus, 1758 (Coleoptera, Tenebrionidae), beef mincemeat, dry cat food, the greenbug Schizaphis graminum (Rondani, 1852) (Insecta, Hemiptera, Aphididae), seeds of the signal grass Brachiaria decumbens Stapf (Poaceae) and a mixture of the diets. The material alimentary in the crop and proventriculus of 10 carabid adults of each species collected in the field we analyzed. Abaris basistriata, T. brasiliensis, Scarites sp.2 and Scarites sp.3 did not consume the B. decumbens seeds. The most consumed diet by five species was T. molitor larvae. Schizaphis graminum and T. molitor larvae / mixture were considered the diets most favourable for the reproductive capacity of A. basistriata and S. Seriatoporus/T. brasiliensis, respectively. On the other hand, T. molitor larvae and mixture of the diets were the most favorable diets for rearing A. basistriata, S. Seriatoporus and T. brasiliensis under laboratory conditions. All species showed fluid and pieces of integument of arthropods in gut, but without displaying the plant material. Some materials gut were identified as fragments of antennae, legs, wings and mandibles in alimentary content of A. basistriata, T. brasiliensis, Scarites sp.2 and Scarites sp.3. / Mestre

Alterações histológicas secundárias à interrupção dos vasa vasorum na aorta descendente : implicações na patogênese da dissecção aórtica

Introdução: Existem vários fatores a serem considerados na fisiopatologia da dissecção aórtica tais como: distúrbios hemodinâmicos e os fatores relacionados às propriedades mecânicas e geométricas da parede aórtica. Os Vasa Vasorum (VV) parecem ter um importante papel em várias doenças, pois o fluxo sangüíneo insuficiente através dos VV aórticos está envolvido na fisiopatologia da aterosclerose e pode contribuir para a necrose da túnica média da aorta. Objetivo: O objetivo era demonstrar pela primeira vez as alterações histológicas que ocorrem na parede aórtica a longo prazo, ou seja, além das 2 semanas de interrupção do fluxo sangüíneo através do VV da aorta descendente em modelo suíno. Método: O experimento foi conduzido com 8 suínos jovens, idade média de 8 semanas resultantes do cruzamento das raças Landrace e Large White, pesando em média 25 Kg (21 – 27Kg) e todos os animais eram fêmeas. Por meio de uma incisão tóraco-lateral à esquerda no 6º espaço intercostal, a camada adventícia era incisada transversalmente em cerca de 1/3 da circunferência da aorta. Era então realizada a ligadura das artérias intercostais e a retirada da camada adventícia, por cerca de 5cm na porção média da aorta descendente. Após a retirada da camada adventícia a aorta era envolvida com o pericárdio bovino ao redor da área sem adventícia e então era realizado o fechamento da cavidade torácica. Após a eutanásia em duas, quatro, seis e oito semanas era realizada a retirada do segmento aórtico envolvido por pericárdio e cerca de 5 cm de aorta proximal e distal. A seguir, uma lâmina de cada segmento (proximal, médio e distal) das aortas foi corada pela Hematoxilina e Eosina e uma pelo método de Weigert – van Gieson, para coloração das fibras elásticas. Resultados: Após duas semanas observamos fibrose subendotelial com preservação da lâmina elástica interna, assim como da camada média nos seus 2/3 internos. Havia um moderado desarranjo das fibras elásticas no 1/3 externo da camada média com um leve infiltrado de linfócitos e neutrófilos. Identificaram-se também focos de angiogênese no 1/3 externo da camada média. Nos animais sacrificados após quatro semanas persistia a fibrose subendotelial com a lâmina elástica interna preservada, assim como a camada média no seu 1/3 interno. Foi identificado um desarranjo estrutural das fibras elásticas e colágenas nos 2/3 externos da camada média. Em relação à inflamação foi observado um infiltrado de linfócitos, histiócitos e neutrófilos nos 2/3 externos da camada média. Quanto à angiogênese foram observados focos nos 2/3 externos da camada média. Após seis semanas os seguintes achados foram identificados: necrose da parede da aorta com fibrose subendotelial. Em relação à inflamação foi identificada necrose com infiltrado linfocítico, histiocitário e neutrofílico em toda a camada média. Quanto à angiogênese observaram-se focos em toda a camada média e na porção subendotelial. Finalmente, após oito semanas havia fibrose de toda a parede da aorta com destruição da lâmina elástica interna. Observamos uma área de inflamação moderada próxima ao endotélio. Havia focos de angiogênese em toda a camada média e na porção subendotelial. Conclusão: O nosso estudo confirma que a isquemia induzida pela interrupção total do fluxo sanguíneo através dos vasa vasorum adventiciais leva à degenerações da túnica média, determinando perda da estrutura normal da parede aórtica que é variável em sua localização, intensidade e forma, dependendo do tempo de evolução da lesão. Em resumo, estudamos as alterações histológicas decorrentes da interrupção dos VV da aorta descendente em suínos desde a segunda semana da lesão, até que ocorresse a fibrose completa da parede. Este estudo pode contribuir para um melhor entendimento do papel dos VV na degeneração da parede aórtica e dos mecanismos envolvidos na dissecção. / Introduction: There are several factors to consider in the pathophysiology of aortic dissection, including: hemodynamic disorders and factors related to the mechanical properties and geometry of the aortic wall. The Vasa Vasorum (VV) seems to have an important role in various diseases, since insufficient blood flow through VV is involved in the pathophysiology of atherosclerosis and may contribute to necrosis of the media. Objective: To demonstrate the histological alterations that occur in aortic wall over the long term, i.e. more than two weeks after the interruption of blood flow through the VV of the descending aorta in a porcine model. Method: The experiment was conducted with eight young female pigs, resulting from the crossing of two breeds, Landrace and Large White, with a mean age of 8 weeks, weighing on average 25 kg (21 - 27 kg). Through a left thoracoabdominal incision through the 6th intercostal space, the adventitia was incised transversely in about one third of the circumference of the aorta. The intercostal arteries were then ligated and an area of the adventitia about 5 cm in length was removed from the middle portion of the descending aorta. Then, the area from which the tunica adventitia had been removed, was wrapped in bovine pericardium, after which the chest cavity was closed. Following euthanasia, at two, four, six and eight weeks, the aortic segment wrapped in pericardium was removed together with portions about 5 cm long of proximal and distal aorta. Next, slices from each segment (proximal, middle and distal) of the aorta were stained with hematoxylin and eosin (HE) and Weigert - Van Gieson method for elastic fiber staining. Results: After two weeks, subendothelial fibrosis was observed with preservation of the internal elastic lamina and the inner 2/3 of the tunica media. There was a slight cellular breakdown in the outer 1/3 of the media with some lymphocyte and neutrophil infiltration. In addition, foci of angiogenesis were identified in the outer 1/3 of the tunica media. In animals sacrificed at four weeks, persisted a subendothelial fibrosis with preservation of the internal elastic lamina and of inner 1/3 of the tunica media. There was structural breakdown of elastic fibers and collagen fibers in the outer 2/3 of the tunica media. Regarding inflammation, lymphocyte and neutrophil infiltration was observed in the outer 2/3 of the tunica media. Foci of angiogenesis were observed in the outer 2/3 of the tunica media. After six weeks, there was necrosis of the aortic wall with subendothelial fibrosis. Regarding inflammation, there was lymphocytic, histiocytic and neutrophil infiltration with necrosis throughout the media. Foci of angiogenesis were observed throughout the tunica media and in the subendothelial portion. Finally, after eight weeks, there was fibrosis of the entire wall of the aorta with destruction of the internal elastic lamina. We observed an area of moderate inflammation near the endothelium. In relation to angiogenesis, foci were observed throughout the tunica media and in the subendothelial portion. Conclusion: Our study confirms that the ischemia induced by complete interruption of blood flow through the vasa vasorum (VV) leads to degeneration of the media, determining loss of normal structure of the aortic wall that is variable in its location, intensity and shape, depending on the length and duration of the ischemic insult. In summary, we studied the histological changes caused by the interruption of the VV of the descending aorta in pigs from the second week of the injury until the occurrence of complete fibrosis of the wall at eight weeks. This study may contribute to a better understanding of the role of VV in the degeneration of the aortic wall and the mechanisms involved in aortic dissection.

Cervicocephalic artery dissection:radiological study with clinical outcome

Abstract The aim of this study was to analyze angiographic findings and the presence and topography of cerebral ischemic and/or hemorrhagic lesions in cerebral CT or MRI, and to assess the long-term clinical outcome of a series of 136 consecutive cervicocephalic artery dissection (CCAD) patients. Pulsatile tinnitus was evaluated as a symptom of CCAD. Medical records and films were reviewed retrospectively. Irregular stenosis was found in angiography in 50% and occlusion in 33% of the dissected cervicocephalic arteries. Irregular stenosis normalized in 81% and occlusion recanalized in 34%. Other findings, such as pseudoaneurysms, intimal flaps, double lumens, and irregular dilatations were rare and often remained unchanged in follow-up. Pulsatile tinnitus was a presenting symptom in 12% of the CCAD patients, but the majority of patients had concomitant head or neck pain, ischemic brain symptoms, Horner's syndrome, or cranial neuropathies. Of the 131 patients who underwent brain imaging, 73 (56%) had signs of infarction in cerebral CT or MRI. Occlusion of the dissected vessel was accompanied by infarction in 76%, irregular stenosis in 40%, and other findings only rarely. Of the anterior circulation infarctions, 95% (39/41) were territorial, subcortical, or territorial infarctions with fragmentation and could thus be considered embolic. Subarachnoid hemorrhage was found in CT in 5 of the 22 patients (23%) with intracranial dissection. The patient's long-term clinical outcome was assessed using two methods: a classification into categories based on neurological symptoms and defects and the modified Rankin Scale (mRS). Of the 136 CCAD patients, 60% recovered with no or mild disability and 79% scored 0–2 on mRS. In the case of dissection of one or more cervicocephalic arteries without occlusion, the figures were 75% and 89%. In the case of occlusive dissection of one or more arteries, only about 35% of the patients recovered well, having no or mild disability, and 61% scored 0–2 on mRS. No significant differences were seen in recovery after intra- and extracranial dissections. In conclusion: irregular stenosis, which is the most common angiographic finding in CCAD, is associated with brain infarction less frequently than occlusion, and the long-term clinical outcome is good in most cases. Occlusion of the dissected vessel causes more brain infarctions, and only about 35% of the patients recover well, having no or mild disability. More than 10% of CCAD patients have pulsatile tinnitus as a presenting, and sometimes the only symptom.

Investigação e documentação de flagelados no tubo digestório de flebotomíneos / Investigation and documentation of flagellates in the gut of sand fly phlebotomines

Introdução. Leishmanioses são zoonoses que têm como agentes etiológicos tripanossomatídeos de várias espécies do gênero Leishmania. Elas são transmitidas aos hospedeiros vertebrados por Phlebotominae (Diptera, Psychodidae). As infecções por Leishmania podem se manifestar nas formas visceral (LV) e tegumentar (LT); ambas são sérios problemas de saúde pública no Brasil, que se encontra entre os países do mundo com as mais altas incidências. Dentre as atividades de controle das leishmanioses, as da vigilância entomológica estão voltadas para a redução da taxa de contato dos vetores com os hospedeiros vertebrados e identificar áreas de risco de transmissão. Neste sentido, a detecção de fêmeas de flebotomíneos naturalmente infectadas por Leishmania constitui-se numa das estratégias. Essa detecção pode ser realizada apenas por meio de análises moleculares ou estas serem precedidas de dissecção das fêmeas para observação da presença de flagelados em seus tratos digestórios. A observação dessas formas no trato digestório pode indicar a capacidade do parasita em resistir aos processos de sua eliminação, dependendo do local onde se encontra, e a que subgênero (Leishmania ou Viannia) pertence. No Brasil, a LV tem sido associada a espécie Leishmania (Leishmania) infantum. A LT tem como agentes espécies de ambos os subgêneros. O presente projeto teve como objetivo desenvolver um manual no qual são descritos os processos para se realizar a investigação da infecção natural por flagelados em fêmeas de flebotomíneos. Materiais e Métodos. Para a elaboração do manual foram examinadas fêmeas selvagens e outras provenientes de colônias alimentadas em hamsters infectados experimentalmente por Leishmania braziliensis e, posteriormente, dissecadas e examinadas em microscópio. Os parasitas utilizados nos experimentos de infecção em laboratório são oriundos de cultivos mantidos em laboratório. As espécies examinadas foram Lutzomyia longipalpis, Nyssomyia neivai e Pintomyia fischeri. Foram descritos os cuidados necessários à preservação dos espécimes em campo e transporte ao laboratório, de modo a tornar viável a dissecção e exame dos espécimes. As capturas realizadas em campo para o estudo permitiram tanto a obtenção dos espécimes como a elaboração do item que trata dos cuidados com as amostras. A documentação das múltiplas etapas de dissecção e exame dos espécimes necessários para o estudo foram realizados por meio de câmera fotográfica acoplada a microscópio óptico. Resultados. São apresentados na forma de manual. Neste são descritos e ilustrados aspectos das técnicas de captura, cuidados com os insetos no campo e laboratório, transporte de amostras vivas, o processo de dissecção da fêmea e a observação de seu tubo digestório para a investigação da presença de flagelados, e de estruturas de sua genitália, para a identificação da espécie de flebotomíneo. Apresenta-se ainda um protocolo a ser utilizado na preservação dos espécimes que forem encontrados infectados para posterior análise molecular e identificação específica do parasita. Conclusões. Com os tópicos apresentados no manual, espera-se contribuir com o programa vigilância epidemiológica das leishmanioses, em atividades da vigilância entomológica voltadas para a identificação de áreas de risco de transmissão dessas doenças e obter evidências que apontem para o potencial de espécies que possam estar atuando como vetores permissíveis de Leishmania spp. / Introduction. Leishmaniases are zoonoses having trypanosomatids of several species of the genus Leishmania as etiological agents, transmitted to vertebrate hosts by the bite of females of Phlebotominae (Diptera, Psychodidae). These parasitoses can manifest in visceral (VL) and tegumentary (TL) forms, both being serious public health problems in Brazil, which is among the countries of the world with the highest incidence rates. Among the activities of control of leishmaniases, those of entomological surveillance are aimed at reducing the contact rate of the vectors with the vertebrate hosts and also to identify areas in risk of the disease transmission. In this sense, the detection of sand fly females naturally infected with Leishmania is one of the strategies. This detection can be carried out exclusively by means of molecular analyzes or these being preceded by dissection of the females to observe the presence of flagellates in their guts. The observation of flagellate forms in the gut indicates the ability of the parasites to resist the processes of their elimination, and also to identify the subgenus (Leishmania or Viannia) to which the parasites belong. In Brazil, the VL infection is attributed to Leishmania (Leishmania) infantum, while in TL, there are various agents belonging to both subgenera. Objective. To develop a manual in which the processes for the investigation of the natural infection by flagellates in sand fly females are described and illustrated. Material and Method. For the construction of the manual, observation of infection by flagellates was made through the dissection of wild or laboratory females of Lutzomyia longipalpis, Nyssomyia neivai and Pintomyia fischeri, experimentally infected with Leishmania braziliensis. The description of care in the preservation of the specimens in the field and in their transport to the laboratory, in order to make feasible the dissection, was made based on the collection undertaken in this project. The documentation was made by means of a photographic camera coupled to an optical microscope. Result. A manual is presented describing and illustrating aspects of collection techniques, insect care in the field and laboratory, transport of the live samples, the dissection process and observation, respectively, of the female gut for the investigation of the flagellate presence and genitalia structure for the identification of the phlebotomine species. Protocol is also presented to be used in the preservation of the specimens found infected for subsequent molecular analysis and the specific identification of the parasite. Conclusions. With the topics presented in this manual, we intend to contribute to the program of epidemiological surveillance of leishmaniases, in activities of entomological surveillance, in the identification of areas of risk of transmission of these diseases, as well as to obtain evidence about the potential of species that may act as permissible vectors of Leishmania spp.

Building user interactive capabilities for image-based modeling of patient-specific biological flows in single platform

In this work, we have developed user interactive capabilities that allow us to perform segmentation and manipulation of patient-specific geometries required for Computational Fluid Dynamics (CFD) studies, entirely in image domain and within a single platform of ‘IAFEMesh'. Within this toolkit we have added commonly required manipulation capabilities for performing CFD on segmented objects by utilizing libraries like ITK, VTK and KWWidgets. With the advent of these capabilities we can now manipulate a single patient specific image into a set of possible cases we seek to study; which is difficult to do in commercially available software like VMTK, Slicer, MITK etc. due to their limited manipulation capabilities. Levelset representation of the manipulated geometries can be simulated in our flow solver (SCIMITAR-3D) without creating any surface or volumetric mesh. This image-levelset-flow framework offers few advantages. 1) We don't need to deal with the problems associated with mesh quality, edge connectivity related to mesh models, 2) and manipulations like boolean operation result in smooth, physically realizable entities which is challanging in mesh domain. We have validated our image-levelset-flow setup with the known results from previous studies. We have modified the algorithm by Krissian et al. and implemented it for the segmentation of Type-A aortic dissection. Finally, we implemented these capabilities to study the hemodynamics in Type-A aortic dissection. Our image based framework is a first of its kind and the hemodynamic study of Type-A dissection too is first study onto the best of our knowledge.

Echerichia coli Biofilm Formation in Musca domestica Crops

The house fly, Musca domestica can transmit human pathogens including Escherichia coli O157:H7 through regurgitation of ingested bacteria from the crop which is a foregut organ of house fly and stores the excess ingested nutrients. Interactions between the ingested bacteria and the crop have a direct influence on bacteria persistence, survival and ultimately fly vector competence. In this research, in situ crop vessel assay was developed to investigate bacterial growth within fly crops up to 48 hours post-ingestion. Flies were fasted for 12 h prior to feeding E. coli O157:H7 pEGFP and then fed bacteria with red food color which was added to confirm that flies had consumed the bacteria. After feeding, flies with red abdomens were aseptically dissected and crops were removed and maintained in sterile phosphate buffered saline in microtiter plates held at 32˚C. For each time point (0, 24 and 48 hours post-ingestion), five crops were homogenized individually using a tissue grinder and bacterial levels (CFU/crop) were monitored using plate counts. Confocal microscopy of intact crops was used to monitor biofilm development. There was no statistical difference in cell numbers (CFU/crop) over the 48 h incubation period. Microscopy showed that upon prolonged incubation, GFP-expressing E. coli within the crop produced biofilms. This method showed greater reproducibility in studying crop bacteria level than using a live fly feeding study. But this system was not recommended to study the interaction between bacteria and the crop of housefly.

Recreational Firewood Movement as a Vector of Non-Native Woodborers in Mississippi

Recreational firewood collected from campers in Mississippi State Parks was investigated for factors associated with insect presence and their diversity. Insects were found in 20% of firewood and evidence of past feeding was found in 64.8%. Representatives of 35 families of insects were reared from collected firewood. These included representatives of Buprestidae, Cerambycidae, Curculionidae, Formicidae, and Rhinotermitidae. The effects of firewood age, moisture content, and source were also examined as effects on insect presence in firewood. At the same time, a survey of campers’ beliefs and attitudes about non-native woodborers was conducted. Exposure to public awareness campaigns had the strongest association on reported sources of firewood and support for regulations on firewood movement. However, no association was found between attitudes and beliefs and camper firewood habits. No association was found between camper responses to survey questions and biological factors from firewood collected from them.
