An examination of the relationship between urbanicity and children with emotional disturbances served in restructuring public schools

Efforts to affect changes in student achievement through altering the manners in which schools operate have been countless. However, there are few empirical studies on the relationship between these reform activities and student outcomes, especially outcomes for students with emotional disturbances from geographically diverse locations. The current study was a secondary analysis of data collected as part of the School and Community Study and the Urban School and Community Study conducted by the Research and Training Center for the Childrens Mental Health at the Louis de la Parte Florida Mental Health Institute at the University of South Florida. Both studies examined the relationship between student exposure to school restructuring efforts and change in academic and behavioral functioning. The primary purpose of this study was to investigate the relationship between student outcomes and school reform activities and to compare students attending suburban/rural schools and students attending urban schools on academic achievement, psychopathology, and mental health service utilization. Using baseline data from the School and Community Study to match students from the Urban School and Community Study on the variables gender, income, and age; 66 matches (i.e., 132 students) comprised the study sample. Differences between the suburban/rural students and the matched sample of urban students were statistically significant in reading achievement, math achievement, functional impairment, and mental health service utilization. There were no significant differences between students on the variable of level of behavior problems, all of the students scoring in the clinical range.

Role of mesophyll CO₂ diffusion and large-scale disturbances in the interactions between climate and carbon cycles

Reliable prediction of climate change and its impact on and feedbacks from terrestrial carbon cycles requires realistic representation of physiological and ecological processes in coupled climate-carbon models. This is hampered by various deficiencies in model structures and parameters. The goal of my study is to improve model realism by incorporating latest advances of fundamental eco-physiological processes and further to use such improved models to investigate climate-carbon interactions at regional to global scales. I focus on the CO₂ diffusion within leaves (a key plant physiological process) and large-scale disturbances (a fundamental ecological process) as extremely important but not yet in current models. The CO₂ diffusion within plant leaves is characterized by mesophyll conductance (g[subscript m]), which strongly influences photosynthesis. I developed a g[subscript m] model by synthesizing new advances in plant-physiological studies and incorporated this model into the Community Land Model (CLM), a state-of-art climate-carbon model. I updated associated photosynthetic parameters based on a large dataset of leaf gas exchange measurements. Major findings are: (1) omission of g[subscript m] underestimates the maximum carboxylation rate and distorts its relationships with other parameters, leading to an incomplete understanding of leaf-level photosynthesis machinery; (2) proper representation of g[subscript m] is necessary for climate-carbon models to realistically predict carbon fluxes and their responsiveness to CO₂ fertilization; (3) fine tuning of parameters may compensate for model structural errors in contemporary simulations but introduce large biases in future predictions. Further, I have corrected a numerical deficiency of CLM in its calculation of carbon/water fluxes, which otherwise can bias model simulations. Large-scale disturbances of terrestrial ecosystems strongly affect their carbon sink strength. To provide insights for modeling these processes, I used satellite products to examine the temporal-spatial patterns of greenness after a massive ice storm. I found that the greenness of impacted vegetation recovered rapidly, especially in lightly and severely impacted regions. The slowest rebound occurred over moderately impacted areas. This nonlinear pattern was caused by an integrated effect of natural regrowth and human interventions. My results demonstrate mechanisms by which terrestrial carbon sinks could be significantly affected and help determine how these sinks will behave and so affect future climate. / text

Child, caregiver, and family predictors of rates of growth in clinical and functional outcomes in systems of care

Over the last several decades there has been a growing recognition that children with serious emotional disturbances are considerably underserved. The Comprehensive Community Mental Health Services for Children and Their Family Program is the largest federal grant program to help communities to implement, advance, and evaluate the system of care approach to service delivery. One of the goals of the evaluation has been to determine if children who participate in system of care services demonstrate improved clinical and functional outcomes over time. Prior research has revealed that children do display significant improvements over time. While this research is promising, it is also important to explore the variability in the rates of improvement to determine who benefits the most from system of care services. This dissertation explores the predictive role of a selected group of variables (behavioral and emotional strengths, caregiver strain, and demographic variables) on differential rates of improvements in clinical and functional outcomes over time for children who participate in systems of care. These variables were also examined in relation to differences in levels of clinical and functional impairments at intake into system of care services. The results from the latent variable quadratic growth models indicated that children who are served by systems of care displayed significant improvements in clinical and functional outcomes over time, with the greatest improvement occurring in the first six months. Children's behavioral and emotional strengths, caregiver strain, sex, age, and race significantly predicted differences in instantaneous growth, as well as rates of deceleration, for clinical and functional outcomes. Clinical implications, limitations of the study, and directions for future research are discussed. / text

The work described in this thesis has been carried out over the period 1958-1965, having been begun during the tenure of a residency at the University of Colorado, Denver, U.S.A. At this time, surgery for the closure of septal defects was being actively carried out, and interest was aroused, in two clinical features which were apparent from close contact with this programme. The first was the nature of the chest deformities associated with septal defects; the second, the striking alleviation of symptoms which followed successful closure of these defects. The reasons for these phenomena were by no means clear and the subsequent work has been concerned with an attempt to understand them.

Identifying Target Genes related to Respiratory Network Dysfunction in a Mouse Model for the Rett Syndrome

Das Rett Syndrom (RTT) gehört zu den tiefgreifenden Entwicklungsstörungen des Gehirns von dem fast ausschließlich Mädchen betroffen sind (ICD-10, F84.10). Ursächlich für die Pathogenese sind Mutationen im X-chromsomalen MECP2-Gen, welches für den Transkriptionsfaktor Methyl-CpG binding protein 2 (MeCP2) kodiert. Unterschiedliche neurologische Symptome treten zwischen 6 und 18 Monaten nach der Geburt auf, wobei schwere Rhythmussstörungen der Atmung für ein Viertel plötzlicher Todesfälle bei Rett-Patientinnen verantwortlich gemacht werden. Der neuronale Atmungsrhythmus bei Säugern wird in verschieden Regionen des ponto-medullären Hirnstammes generiert, wobei der Prä-Bötzinger Komplex als essentiell für die Rhythmogenese der Atmung angesehen wird. Mittels Genexpressionsstudien in der Ventralen Respiratorischen Gruppe (VRG), die den Prä-Bötzinger-Komplex einschließt, zeigte sich eine massiv erhöhte, pathologische Expression des Serotoninrezeptor 5B sowohl auf mRNA-, als auch auf Proteinebene bei MeCP2-defizienten Mäusen zum postnatalen Entwicklungstag P40. Der Serotoninrezeptor 5B (5-HTR5B) gehört zur Klasse der G-Protein-gekoppelten Rezeptoren. Durch detaillierte Analysen des 5-HTR5B-Proteins konnte eine natürliche Trunkierung des Rezeptors nachgewiesen werden. Des Weiteren wurde eine ungewöhnliche intrazelluläre Lokalisierung in Membranen von vesikulären und tubulären Kompartimenten beobachtet. Trotz dieser ungewöhnlichen Eigenschaften besitzt der Rezeptor weiterhin die Fähigkeit, das inhibitorische G-Protein Gαi3 konstitutiv zu aktivieren und somit den Anstieg von cAMP zu verhindern. Durch genetisches Ausschalten des 5-HTR5B Proteins (knockout) konnte gezeigt werden, dass die durch 5-HTR5B-verminderte cAMP-Konzentration in der VRG ursächlich für den gestörten Atmungsrhythmus MeCP2-defizienter Mäuse ist. Die sich aus diesen Ergebnissen ableitende pharmakologische Strategie, die cAMP Konzentration zu erhöhen, führte zu einem deutlich verbesserten Atmungsrhythmus. Die Ergebnisse dieser Arbeit implizieren neue Therapieansätze zur Behandlung der Atmungs-störungen von Rett-Patienten.

Nuclear magnetic resonance studies of modified eukaryotic cytochrome C

The central theme of this thesis is a study of the structural changes accompanying chemical modification and denaturation of eukaryotic cytochrome c as characterised by <sup>1</sup>II nuclear magnetic resonance (n.m.r.) spectroscopy. First, however, it was necessary to obtain and confirm assignments for individual resonances; this was achieved by a novel method of cross assignment between ferricytochrome c and ferrocytochrome c and by double resonance techniques. A variety of perturbations were caused to native cytochromes c, which ranged in degree from the elevation of temperature for ferrocytochrome c to the complete denaturation of the protein with urea or methanol. Modification at single sites both on the surface (e.g. Met 65, Tyr 74) and in the core ( e.g. Tyr 67) of the molecule were found to cause only small local effects to the structure, although the dynamic features of the molecules were altered. One single site modification, the breaking of the iron - sulphur cross linking bond, caused considerable disruption to one side of the molecule, although hydrophobic domains in the other side were preserved; this state of the molecule is analogous to the penultimate state in the refolding pathway. Modification of all the charged lysine residues caused small changes to the surface structure of the molecule, though the complete reversal of the charges in maleyl cytochrome c produced a species which unfolded reversibly from a native configuration with the increase of temperature. The unfolding of the protein is virtually identical with both methanol and urea, but the pathways are shown to differ for the oxidised and reduced proteins.

INITIAL RESPONSE OF INVASIVE EXOTIC PLANT SPECIES TO TIMBER HARVESTING IN SOUTHEASTERN KENTUCKY FORESTS

The responses of invasive exotic plant species (IES) to silvicultural treatments one growing season after timber harvesting were examined in the Cumberland Plateau region of Southeastern Kentucky. Treatments included a commercial deferment harvest and unharvested control applied to five watersheds within University of Kentucky’s Robinson Forest. The effects of harvesting were compared between treatments and between preharvest and postharvest samplings. The spatial distribution and abundance of targeted IES throughout forest sites and trail systems were calculated from several sampling schemes. Additional analyses were performed to quantify forest disturbances derived from harvest activities to determine the relationships between soil, light levels, and other environmental characteristics and IES cover. Logistic and multivariate analysis techniques were used to analyze differences in IES distribution between pre-harvest and post-harvest units to relate post-harvest IES to microsite conditions. Microsite conditions within the forest and along the trail system proved important for explaining the presence and distribution of IES. Timber harvesting caused a significant increase in both Ailanthus altissima and Microstegium vimineum within harvested areas. However, many other identified IES did not initially respond to disturbances. Throughout the treatment units, species were influenced by disturbance type and intensity, as well as proximity to reclaimed surface mined land.
