Vliv potravy na výši fertility u zákeřnic druhu \kur{Platymeris biguttatus} / Effect of food on fertility in assassin bug \kur{Platymeris biguttatus}

This thesis was developed in order to determine whether the selected species most commonly kept feeding insects affect fertility in captive bred assassin bugs \kur{Platymeris biguttatus}. Used feeding insects were \kur{Blaptica dubia} roaches, \kur{Acheta domestica} crickets and larvae of \kur{Tenebrio molitor} bugs. The reason of using assassin bugs \kur{Platymeris biguttatus} was their popularity among breeders, their easy care needs and their superior fertility. Assassin bugs were kept in an air conditioned room with a constant temperature conditions and light regime. It has been shown that the given diet has no effect on fertility in this kind of bugs, also that breeding in groups has negative impact on fertility of this assassin bugs species. The results of this study could demonstrate the importance of proper feeding in keeping these bugs and help breeders to have better results.

Ecology of Chironomids Associated with Myriophyllum Spicatum L. and Heteranthera Dubia Macm

Macroinvertebrate communities inhabiting an exotic, Myriophyllum spicatum, and a native, Heteranthera dubia macrophyte were studied from March 1999 to June 2000 in experimental ponds. Although macrophyte architecture explained some variation in macroinvertebrate abundance between the two macrophytes, most variation was explained by the sampling months. Total number of macroinvertebrates was found to be positively correlated with epiphyton biomass which differed significantly between the two plant types and among sampling months. Taxa richness did not vary between the two plant types. Chironomid larvae were the most abundant organisms and dominated by Apedilum elachistus on both plant communities. Annual production of five chironomid species was estimated by the size-frequency method. Production estimates (P) in g dry wt m-2 yr-1 of plant surface area for the predator Tanypodinae larvae were: Larsia decolarata, P= 0.77 and 0.67, Labrundinia virescens, P= 0.59 and 0.35 on M. spicatum and H. dubia, respectively. Larvae of Cricotopus sylvestris and Psectrocladius vernalis were collected from M. spicatum from March to mid-June. Production of C. sylvestris was found to be 0.46 g dry wt m-2, whereas it was 0.07 g dry wt m-2 for P. vernalis for this period. Apedilum elachistus exhibited the highest productivity: 9.9 g dry wt m-2 yr-1 of plant surface area on M. spicatum, and 8.5 g dry wt m-2 yr-1 on H. dubia. These production estimates are among the highest production values reported for a single species. Additionally, post-ovipositing development times for five chironomid species collected from Myriophyllum and Heteranthera were determined. Three different temperatures (15°, 20° and 25°C) were chosen to rear eggs under 12L: 12D photoperiod. Egg development times ranged between 1-4 days. Larval development times ranged from 44 days at 20°C for Tanypus neopunctipennis to as few as 9 days at 20°C for Larsia decolorata.

Analysis of Zero-Heavy Data Using a Mixture Model Approach

The problem of high proportion of zeroes has long been an interest in data analysis and modeling, however, there are no unique solutions to this problem. The solution to the individual problem really depends on its particular situation and the design of the experiment. For example, different biological, chemical, or physical processes may follow different distributions and behave differently. Different mechanisms may generate the zeroes and require different modeling approaches. So it would be quite impossible and inflexible to come up with a unique or a general solution. In this dissertation, I focus on cases where zeroes are produced by mechanisms that create distinct sub-populations of zeroes. The dissertation is motivated from problems of chronic toxicity testing which has a data set that contains a high proportion of zeroes. The analysis of chronic test data is complicated because there are two different sources of zeroes: mortality and non-reproduction in the data. So researchers have to separate zeroes from mortality and fecundity. The use of mixture model approach which combines the two mechanisms to model the data here is appropriate because it can incorporate the mortality kind of extra zeroes. A zero inflated Poisson (ZIP) model is used for modeling the fecundity in <i> Ceriodaphnia dubia</i> toxicity test. A generalized estimating equation (GEE) based ZIP model is developed to handle longitudinal data with zeroes due to mortality. A joint estimate of inhibition concentration (ICx) is also developed as potency estimation based on the mixture model approach. It is found that the ZIP model would perform better than the regular Poisson model if the mortality is high. This kind of toxicity testing also involves longitudinal data where the same subject is measured for a period of seven days. The GEE model allows the flexibility to incorporate the extra zeroes and a correlation structure among the repeated measures. The problem of zero-heavy data also exists in environmental studies in which the growth or reproduction rates of multi-species are measured. This gives rise to multivariate data. Since the inter-relationships between different species are imbedded in the correlation structure, the study of the information in the correlation of the variables, which is often accessed through principal component analysis, is one of the major interests in multi-variate data. In the case where mortality influences the variables of interests, but mortality is not the subject of interests, the use of the mixture approach can be applied to recover the information of the correlation structure. In order to investigate the effect of zeroes on multi-variate data, simulation studies on principal component analysis are performed. A method that recovers the information of the correlation structure is also presented. / Ph. D.

Estandarización y validación del método HET-CAM para medir la irritabilidad ocular in vitro de los extractos de cinco frutos nativos del Perú utilizados en la industria cosmética

La presente investigación, provee datos científicos sobre los métodos HET-CAM cualitativo y HET-CAM cuantitativo (CAM-TBS), evaluado en frutos nativos, lo cual puede ser desarrollado profundamente como un método alternativo a la prueba de Draize. El objetivo es estandarizar y validar el método HET-CAM cuantitativo (CAM-TBS) en cinco extractos de frutos nativos del Perú. Los frutos evaluados fueron Physalis peruviana L. (aguaymanto), Myrciaria dubia L. (camu camu), Mauritia flexuosa L. (aguaje), Solanum sessiliflorum D. (cocona) y Passiflora mollisima HBK (tumbo serrano). Estas frutas listadas son comúnmente utilizadas en formulaciones cosméticas. Se prepararon extractos acuosos, hidroalcohólicos y glicólicos. La implementación de la técnica, estandarización y validación, se realizó en el laboratorio de Centro de Investigación de Bioquímica y Nutrición de la Facultad de Medicina Humana de la Universidad de San Martín de Porres (USMP) y en las instalaciones de la empresa privada Ingenioidea SAC, ambas instituciones están localizadas en Lima, Perú. Para la validación del método HET-CAM cuantitativo (CAM-TBS) se evaluaron los parámetros: Linealidad, precisión, exactitud y robustez. Los resultados de estos parámetros se sometieron a pruebas estadísticas demostrando que el método HET-CAM cuantitativo (CAM-TBS) propuesto para la medición de la irritabilidad ocular in vitro es lineal, exacto y preciso pero no robusto.

Os parâmetros de qualidade de água avaliados na bacia hidrográfica foram: sólidos totais, temperatura, turbidez, demanda bioquímica de oxigênio (DBO), fósforo total,nitrogênio total, oxigênio dissolvido, pH, Escherichia coli e ensaio ecotoxicológico com Ceriodaphnia dubia. Os parâmetros físicos, químicos, microbiológico e ecotoxicológico foram avaliados separadamente e posteriormente o índice de qualidade da água (IQA) foi determinado a fim de se verificar qual a melhor forma de avaliação da qualidade dos recursos hídricos. Os dados referentes aos parâmetros de qualidade da água foram obtidos por meio do Relatório de Qualidade das Águas Superficiais no Estado de São Paulo de 2015, elaborado pela CETESB. O índice de qualidade da água da Bacia Hidrográfica do Rio São José dos Dourados variou de ótima a boa, indicando que o uso e ocupação do solo no entorno da Bacia não tem prejudicado a qualidade de água de forma geral. Entretanto, a análise individualizada dos parâmetros físicos, químicos e microbiológicos permitiu verificar que o fósforo total, oxigênio dissolvido e E. coli excederam os limites estabelecidos pela Resolução CONAMA nº 357/2005, evidenciando que as ações antrópicas estão influenciando a qualidade dos recursos hídricos. Essas alterações podem estar relacionadas com o regime pluviométrico, lançamento de esgotos domésticos e a dissolução de compostos no solo como os fertilizantes, agrotóxicos e excremento de animais. A água da Bacia não apresenta toxicidade aguda para C. dúbia, ou seja, não estão ocorrendo efeitos de transformações das substâncias químicas sobre esses organismos que vivem nos ecossistemas aquáticos. Os ensaios ecotoxicológicos servem como complemento para as análises físicas, químicas e microbiológicas, pois proporcionam abordagens integradas dos efeitos de poluentes, levando em consideração a resposta da comunidade biológica, servindo como um instrumento de controle de poluição. Esse estudo demonstra que, ao utilizar o IQA, perdem-se informações relevantes para a avaliação da qualidade dos corpos d'água, já que, com a aplicação do índice, não se consegue fazer uma interação dos parâmetros analisados. / Qualitative and quantitative monitoring of superficial waters can indicate human activities which present risks to this resource, serving as a main instrument to the management of water bodies. Studies related to hydric resources provide information that can establish criteria for their management and decision-making, considering the framing of the water bodies and the particularities of each watershed. In this context, this study approached the factors that interfere in a negative way in water quality and the damages caused to hydric resources, and the aquatic biota as an effect of those factors. This resource aimed at evaluating the quality and the ecotoxicity of water from the Watershed of the River São José dos Dourados-SP. We used six points of monitoring properly georeferenced from the Integrated Basic Web of CETESB/ANA that exists in UGRHI 18. The parameters of water quality that were evaluated in the watershed are: total solids, temperature, turbidity, biochemical oxygen demand (BOD), total phosphorus, total nitrogen, dissolved oxygen, pH, Escherichia coli and ecotoxicological experiment with Ceriodaphnia dubia. Physical, chemical, microbiological and ecotoxicological parameters were evaluated separately and later the index of water quality (IWQ) was determined to verify the best way to evaluate the quality of hydric resources. The data about water quality parameters were obtained from the Report of Quality of Superficial Waters in the State of São Paulo of the year 2015, elaborated by CETESB. The index of water quality in the Watershed of the River São José dos Dourados had a variation from great to good, what indicates that the land use and zoning land around the Watershed has not damaged the water quality in a general way. However, doing an individualized analysis of physical, chemical and microbiological parameters, allowed to verify that total phosphorus, dissolved oxygen and E. coli exceeded the limits established by the CONAMA Resolution 357/2005. It shows that anthropic actions are influencing the quality of hydric resources. Those alterations may be related to the pluviometric regimen, domestic sewage discharge and the dissolution of composites in the ground, such as fertilizers, pesticides and animal excrements. The water in the Watershed does not present acute toxicity to C. dúbia, i.e., there are no effects of transformation of chemical substances on the organisms living in the aquatic ecosystems. The ecotoxicological experiments serve as a complement to physical, chemical and microbiological analysis, because it provides integrated approaches of the effects of pollutants, considering the response of the biological community, serving as an instrument of pollution control. This research demonstrates that using the IWQ we lose relevant information for water bodies quality evaluation, as applying the index we cannot make an interaction of the analyzed parameters. / CAPES: Código de Financiamento 001

The aim of this thesis was to find out whether selected types of food substrates affect the adult´s productivity, the weight of newborn nymphs and laid oothecas and whether changes in adult´s productivity and the weight of nymphs occur due to one source of food during the experiment. In this experiment, the Blaptica dubia, Nauphoeta cinerea and Schelfordella tartara species were used. The reason for selection of those species was their high rate of reproduction, undemanding breeding and a short period of growth. The experiment was conducted in six types of feed substrates. The cockroaches were kept at a room temperature. The scales accurate to 0,2 g were used for the determination of the weight (AVINET). The Blaptica dubia species proved the dependence of its adults´ productivity and nymphs´ weight on the month of observation and the dependence of nymphs´ growth on a feed. The Nauphoeta cinerea species proved the dependence of nymphs´ growth on a feed. The Schelfordella tartara species proved the dependence of its adults´ productivity and oothecas´ weight on a feed and on the month of observation. The results of this thesis could emphasize the importance of the influence of a feed on the life of cockroaches breeded in captivity and help breeders to their better results with regard to the selection of a suitable food.

Temporal and Spatial Comparisons of Ambient Toxicity of the Trinity River in Relationship to an Effluent

A toxicological study was initiated because of concerns about allegations that the Texas Water Commission that effluent from the Dallas Central Wastewater Treatment Plant, which discharges into the Trinity River, was affecting downstream water quality. Monthly, flow-weighted composite effluent samples were collected. Grab samples were also collected upstream and downstream from the effluent from April 1989 to August 1991. Toxicity tests were conducted on these samples using Ceriodaphnia dubia as the test organism. Samples were collected four times during this study in which rainfall occurred prior to sampling. In every instance, this "first flush" of the watershed during a rising hydrograph was toxic to C. dubia upstream. Analyzing toxicity by season resulted in a statistically significantly lower neonate production in the effluent than in the river samples during the months of June, July, and August. This impact on neonate production was suspected of being caused by organic pesticides which are used for insect control on lawns. The effluent was never acutely toxic to C. dubia. Primarily, toxic occurrences in either the effluent or the river samples were primarily of a chronic nature. Overall, survival of C. dubia was affected more frequently at the upstream site than in the effluent or the downstream site. Because EPA's Phase I Acute Toxicity Identification Evaluations (TIEs) methods were designed for identifying acute toxicity, two alternative strategies were attempted to identify chronic toxicity. The first attempt was the modification of the phase I acute TIE methodologies. This was done by processing more sample through the phase I characterization tests. This approach was inadequate due to toxicity that occurred during the last several days of the seven-day C. dubia reproduction test. The second strategy for identifying chronic toxicity within a TIE involved the use of freeze concentration. During this preliminary investigation ofthe efficiency of freeze concentration, four metals and two organic compounds were freeze concentrated.

Ao logo do amadurecimento, as massas e os diâmetros variaram (p ≤ 0,05) somente na progênie 44 e o rendimento em polpa aumentou em todas as progênies, com valores superiores a 50% no estádio maduro. O estádio de maturação não mostrou efeito somente nos teores de umidade, cinzas e pH da progênie 38; cinzas da progênie 44 e lipídios e fibras insolúveis da progênie 17. Os demais resultados variaram com o amadurecimento, porém, com comportamentos diferentes em cada progênie, exceto os teores de açúcares totais, sólidos solúveis e relação SS/ATT, que mostraram tendência de aumento, e a acidez titulável total, que diminuiu em todas as progênies. No geral, foram obtidos maiores conteúdos de vitamina C no estádio verde, com uma primeira etapa de degradação, ao atingir o estádio semimaduro, seguido de uma etapa de síntese até o final da maturação, exceto na progênie 38, que mostrou somente redução. Quanto aos compostos fenólicos, durante o amadurecimento, as três progênies foram caracterizadas por processos de síntese e degradação de compostos fenólicos totais; os flavanóis totais diminuíram; os flavonóis comportaram-se de maneira distinta entre as progênies, com elevação do conteúdo nas progênies 17 e 38, e redução na progênie 44; devido à limitação do método utilizado para pequenas quantidades, foi possível quantificar antocianinas somente nas progênies 17 e 44 no estádio maduro. As capacidades antioxidantes, por meio dos métodos TEAC e DPPH, diminuíram em todas as progênies. Além do efeito significativo do estádio de maturação, os resultados mostraram que a variabilidade genética do camu-camu influenciou de forma significativa nas características físicas dos frutos e nos conteúdos de todos os compostos estudados, além das capacidades antioxidantes obtidas pelos métodos utilizados. / This study aimed to determine physical and physico-chemical characteristics, bioactive compounds and antioxidant capacity from camu-camu pulp of three different progenies, came from Active Germplasm Bank of camucamuzeiro, at Embrapa Amazônia Oriental, in three ripening stages. Over the ripening, the mass and the diameters varied (p ≤ 0.05) just for progeny 44 and the pulp yield increased for all the progenies, with results over 50% in the ripe stage. The ripening stage didn’t show effects in the moisture and ashes content and also pH for progeny 38; ashes for progeny 44 and fat and insoluble fibers for progeny 17. The others results varied with the ripening, but, with different behaviors to each progeny, except for the total sugar, soluble solids contents and the SS/TTA ratio, which showed a tendency to increase, and the total titratable acidity decreased in all the progenies. Overall, a higher level of vitamins C were obtained in the green stage, with a first step of degradation, as it reached the semi-ripe stage, followed by a synthesis step until the end of the ripening stage, except for the progeny 38, which showed a reduction. As for the phenolic compounds, during the ripening stage, the three progenies were characterized for synthesis and degradation processes of the total phenolic compounds; the total flavanols decreased; the flavonols content had a distinct behavior in each progeny, with an increasing in the progenies 17 and 38, and a decreasing in the progeny 44; due to a limitation of the method used to determine small quantities, It was possible to quantify anthocyanins only for progenies 17 and 44 in the ripening stage. The antioxidant capacity, determinated through the methods TEAC and DPPH, decreased in all the progenies. Besides the significant effects of the ripening stage, the results showed that the genetic variability of camu-camu influenced significantly the physical characteristics of the fruits and the continents of all compounds studied, and still the antioxidant capacity obtained through the methods mentioned.

Foram coletados frutos de três genótipos, escolhidos ao acaso, em três estádios de maturação. Os resultados foram expressos em base seca e analisados por meio de Análise de Variância, teste de comparação de médias de Tukey e Coeficiente de Correlação de Pearson, com intervalo de confiança de 95%. Para as análises físico-químicas, houve um comportamento diferente para cada progênie, em função da maturação, com exceção do teor de carboidratos que aumentou em todas as progênies. Para vitamina C, as progênies 38 e 44 apresentaram uma elevação do teor de ácido ascórbico, comparando os estádios verde e maduro, e a progênie 17 demonstrou diminuição durante a maturação; o maior teor de vitamina C foi encontrado na progênie 44, estádio maduro (24,02 g/100g). Todas as progênies apresentaram uma elevação no teor de fenólicos totais e antocianinas ao longo da maturação, sendo que os maiores valores foram demonstrados pela progênie 17 (3298,98 mg AGE/100g) e 44 (165,91 mg/100g) respectivamente. Os flavonóis e carotenoides apresentaram um comportamento distinto e estatisticamente diferente em cada progênie, sendo que o maior teor de flavonóis foi encontrado na progênie 17, no estádio verde (343,63 mg QE/100g) e de carotenoides na progênie 44, estádio maduro (105,88 mg/100g). Em relação à atividade antioxidante baseada no método DPPH, as progênies 38 e 44 apresentaram o mesmo comportamento ao longo da maturação, com maior atividade antioxidante no estádio semimaduro e menor no verde, sendo que a maior atividade antioxidante foi encontrada na progênie 17, no estádio maduro (38,95 g fruta/g). Já pelo método ABTS, a atividade antioxidante foi estatisticamente diferente para todas as progênies e em todos os estádios de maturação, com maior valor apresentado pela progênie 44, no estádio maduro (1701,63 μM trolox/g). Avaliando-se a relação entre os compostos bioativos e a atividade antioxidante, a progênie 17 obteve mais correlações estatisticamente significativas. Pode-se concluir que o fator variedade genética causa diferenciações na síntese de diversos compostos durante a maturação de frutos de camucamuzeiro. / The presence of bioactive compounds suffers greatly influenced by environmental and genetic factors, and behave differently in each part of the plant. However, there are few studies which deal these aspects of camu-camu. Thus, this study aimed to analyze the physical, physico-chemical composition, the main bioactive compounds and antioxidant capacity of camu-camu bark, depending on the stage of ripeness in fruits of different genotypes from plant matrices Bank Active Germplasm Camucamuzeiro of Embrapa Eastern Amazon. Fruits of three genotypes, selected at random in three maturity stages were collected. Results were expressed on a dry basis and analyzed by Analysis of Variance, Comparison Test Tukey's and Pearson's Correlation Coefficient, with a confidence interval of 95%. For physico-chemical analysis, there was a different one for each progeny, depending on maturity, except that the carbohydrate content increased in all progenies behavior. For vitamin C, the progenies 38 and 44 showed a high content of ascorbic acid, comparing the green and ripe stages, and 17 progeny showed a decrease during maturation; the highest content of vitamin C has been found in 44 progeny, ripe stage (24,02 g/100g). All progenies showed an increase in the content of total phenolics and anthocyanins throughout maturation, with the highest values were demonstrated by progeny 17 (3298,98 AGE/100g mg) and 44 (165,91 mg/100g) respectively. Flavonols and carotenoids showed a distinct and statistically different behavior in each progeny , with the highest content of flavonols was found in 17 progeny in the green stage (343,63 mg QE/100g) and carotenoids in 44 progeny, ripe stage (105,88 mg/100g). Regarding the antioxidant activity based on the DPPH method, the progenies 38 and 44 showed the same behavior throughout maturation, with higher antioxidant activity half ripe and smallest stage in green, with the highest antioxidant activity was found in 17 progeny in the ripe stage (38,95 g fruit/g). Already by ABTS method, the antioxidant activity was statistically different for all progenies and in all stages of maturation, higher value submitted by 44 progeny at maturity (170,63 μM trolox/g). Evaluating the relationship between bioactive compounds and antioxidant activity, the 17 progeny won more statistically significant correlations. It can be concluded that various factors cause differences in the synthesis of various compounds during fruit ripening of camucamuzeiro.
