Nanoparticles as MRI contrast agents and biomarkers – applications in prostate cancer and mild traumatic brain injury

Magnetic Resonance Imaging (MRI) is the most prominent non-invasive technique used in clinical diagnosis and biomedical research. Its development as an imaging technique has been aided by contrast agents (CAs) which enhance contrast to noise ratio in the images. This dissertation deals with paramagnetic lanthanide- and superparamagnetic iron-based nanoparticles (NPs) which are potential CAs at clinical field of 3 T and a high field of 9.4 T. Chapter 1 provides a brief overview of colloidal nanoparticles, with an emphasis on their surface chemistry and magnetic properties for bio-applications. Chapter 2 employs europium as an optical probe to illustrate the contribution of inner, second and outer sphere relaxation towards longitudinal and transverse relaxivities of paramagnetic NP-based CAs. Chapter 3 investigates the positive and the negative contrast enhancement abilities and magnetization of paramagnetic NPs comprising a core of sodium dysprosium fluoride with a sodium gadolinium fluoride shell. Their surface chemistry is tuned to target prostate cancer specifically. The application of these NPs is further extended in Chapter 4 to track an intraneuronal protein called tau following mild traumatic brain injury. Chapter 5 deals with facile synthesis and long-term stability of superparamagnetic iron NPs for their potential application as CAs. Chapter 6 illustrates the concept of MRI correlation using ‘T1-only’ and ‘T2-only’ NPs. Chapter 7 investigates on the dynamics involved in the phospholipids coating the surface of NPs. La bande II est la région spectrale du moyen infrarouge correspondant à la fenêtre de transmission atmosphérique s'étendant de 3 à 5 µm. Ce domaine spectral présente de nombreux champs d'applications dans le domaine militaire (radars laser, contre-mesures) ainsi que pour l'analyse de gaz (contrôle de la pollution atmosphérique). <br /><br />Ce travail de thèse s'inscrit dans la thématique « élaboration de sources lumineuses pour le moyen infrarouge ». Il présente une approche originale puisqu'il allie élaboration des matériaux, caractérisation spectroscopique et modélisation du milieu amplificateur laser. Cette étude, à l'interface entre la chimie des matériaux vitreux et l'optique, s'intéresse à la faisabilité de sources laser fibrées en verres de chalcogénures dopés terres rares pour l'émission entre 3 et 5 µm.<br /><br />L'objectif de ce travail est d'évaluer le potentiel amplificateur des fibres de verre Ge20Ga5Sb10S65 dopées erbium ou dysprosium entre 4 et 5 µm. Pour ceci, une étude spectroscopique a été effectuée sur le matériau massif. Puis, des fibres optiques à saut d'indice et monoindices ont été réalisées et caractérisées optiquement. Enfin, les paramètres obtenus ont été utilisés pour construire un modèle et simuler le fonctionnement d'un amplificateur laser à fibre Ge20Ga5Sb10S65 dopé Er3+ pompé par diode à 807 nm et opérant à 4,6 µm.

Laser processing of Tb0.3Dy0.7Fe1.92 films

In the past decade, there has been an increased interest in magnetostrictive materials for micro actuators and sensors. Of particular importance are the Fe₂R intermetallics, where R = Tb, Dy. In this study, films of Tb[subscript 0.3]Dy[subscript 0.7]Fe[subscript 1.92] were prepared by three laser processing techniques (pulsed laser deposition, flat plate ablation and laser ablation of microparticles) to explore the effect of processing parameters on particle size, crystallinity and magnetic properties. The laser used in the experiments was a KrF laser with a 12 ns pulse width. Pulsed laser deposition of an alloyed target in vacuum produces dense amorphous films with the similar composition to the target, low coercivity (46 Oe) and good magnetostriction ([lambda][subcript two horizontal lines] = 305 ppm at 2300 Oe). Flat plate ablation and laser ablation of microparticles produced amorphous nanoparticles at 1 atm. The particles were subsequently jet deposited onto substrates to form thick films. Nanoparticle films produced by flat plate ablation resulted in oxidized and segregated particles due to extended, non-uniform plume expansion, laser target modification, and open porosity. Laser ablation of microparticles produced thick films with M[subscript s] = 13.8 emu/g. Two types of annealing treatments were performed to close porosity and increase Youngs modulus. Annealing of LAM films at temperatures up to 700°C in-situ and 950°C in a reducing atmosphere did not result in coarsening of the particles or crystallization of the Laves phase due to the core-shell structure of nanoparticles (rare earth oxide shell, Fe rich core) brought about by oxidation-induced segregation.

The presented diploma thesis is focused on the preparation and properties of transparent polycrystalline ceramic materials based on Al2O3. Theoretically, the most important technological aspects of the processing of these materials are presented. Detailed attention is given to transparent Al2O3 polycrystalline ceramics doped with rare earth elements. The influence of microstructural parameters on the optical properties (represented by RIT) is investigated on Al2O3 samples doped and codoped with dysprosium, terbium and chromium. A significant effect of the average grain size on the light transmittance of all samples is observed. The highest RIT = 55 % (measured by a laser beam with a wavelength of 632,8 nm) was achieved by an optimized preparation process for a sample doped with 0,05 at. % of dysprosium. For all samples photoluminescent properties are also analysed. The photoluminescent emission spectra correspond to the activation of doping elements. In case of the terbium and chromium codoped samples, the differences in the activation of individual dopants depending on different excitation wavelengths were demonstrated, resulting in different colour emissions for different excitation wavelengths.

No / Two new complexes, [Fe3(μ3-O)(inicH)6(H2O)3][Gd(NO3)6]·(NO3)4·nH2O (1) and [Fe3(μ3-O)(inicH)6(H2O)3][Dy(NO3)5 (H2O)]·(NO3)5·n(H2O) (2) with two isolated complex moieties, were generated when isonicotinic acid was treated with iron(III) nitrate and the corresponding lanthanide(III) nitrate in water. The structures were determined by single-crystal X-ray diffraction studies. In these compounds, the anionic lanthanide complexes are encapsulated by trinuclear [Fe3(μ3-O)(inicH)6(H2O)3]7+ cationic cluster units, which is facilitated by hydrogen-bonding interactions. Securing rare earth elements (REE) for a stable supply require sustainable management strategies in Europe due to a missing local primary production and a dependence on China as the main producer of REE. These elements, like neodymium (Nd) and dysprosium (Dy), are contained in permanent magnets (PM) (mostly NdFeB-magnets) in wind turbines. Addressing the question whether PM-material, Nd- and Dy-contents from wind turbines could help to meet future demands of REE in Europe while reducing simultaneously the import dependence, the purpose of the present work was to analyze the urban mining opportunities, recovery - and recycling potentials for REE from end-of-life (EoL) wind turbines. This thesis aimed to identify current and upcoming stocks as well as material flows of the PM and their containing REE in the wind energy sector. Two European countries, Germany and Denmark, were chosen as case studies to be compared based on created future scenarios and the modeling of the theoretical recycling potential of Nd and Dy in both countries. It could have been identified that the German anthropogenic stock contains greater amounts of NdFeB-magnets and REE compared to the Danish stock. Overall it could be concluded that the countries’ demand could partly be met by using secondary Nd and Dy from the EoL-wind turbines. Although future scenarios were used, the results realistically illustrate the German and Danish anthropogenic stock until 2035 by relying on data of already installed turbines up to 2018, which makes an evaluation of capacities and EoL-turbines, which need to be decommissioned by 2035, achievable. The provided information is valuable for further investigations regarding recovery strategies, feasibility analysis, and future decision-making processes. / För att säkra tillgången på jordartsmetaller (REE) i Europa krävs hållbara beslutsstrategier. Detta på grund av avsaknaden av en inhemsk primärproduktion samt ett beroende av Kina som en huvudprodu-cent av REE. Jordartsmetaller som neodymium (Nd) och dysprosium (Dy), finns kvar i permanenta magneter (PM) (mestadels NdFeB-magneter) i vindturbiner. För att ta itu med frågan om huruvida Nd- och Dy-innehållet i PM-material, från vindturbiner skulle kunna bidra till att uppfylla framtida efter-frågan på REE i Europa samtidigt som importberoendet skulle minskas, var syftet med detta arbete att analysera möjligheterna till urban utvininng, återvinning och materialutnyttjande av REE från vindtur-biner i uttjänt tillstånd (EoL).Syftet med denna uppsats var att identifiera nuvarande och kommande tillgångar samt materialflöden av PM och därav följande REE inom vindkraftsektorn. Två europeiska länder, Tyskland och Danmark, valdes ut som fallstudier och jämfördes i framtida scenarier och mo-dellering av Nd -och Dy teoretiska återvinningspotential i båda länderna. Det kunde konstaterats att det tyska antropogena beståndet innehåller större mängder NdFeB-magneter och REE än det danska beståndet. Sammanfattningsvis kan man dra slutsatsen att ländernas efterfrågan delvis kunde tillgodo-ses genom att man använde sekundär Nd och Dy från EoL-vindturbiner. Även om framtida scenarier användes illustreras resultatet på ett realistiskt sätt det det antropogena lagret i Tyskland och Danmark fram till 2035 genom att man förlitar sig på uppgifter om redan installerade turbiner fram till 2018, vilket gör det möjligt att göra en utvärdering av kapaciteten och antal EoL-turbiner, som måste av-vecklas senast 2035. Informationen är värdefull för ytterligare utredningar om återvinningsstrategier, genomförbarhetsanalys och framtida beslutsprocesser.
