Carla Emery and the Recreation of Homesteading

This dissertation examines Carla Emery's The Encyclopedia of Country Living and her contribution to the modern homesteading movement. Emery (1939-2005) advocated the use of traditional methods and antique cookbooks for food self-sufficiency. She first self published and distributed the book in "issues" (1971), which were eventually combined and sold as a single book, originally named The Old Fashioned Recipe Book (1974). A reading community developed around The Encyclopedia and readers participated in its development by submitting recipes and homesteading resources. Several editions have followed and the book remains in print, considered an authoritative reference for modern homesteading. Modern homesteading is rooted in the traditions of homesteading and domestic advice. It represents a consciousness of human relationships with the land for food production and the role domestic advice in cooking and other aspects of home economics, Emery recommended a country living foodways and 19th century methods of food production for homesteading, attempting to de-industrialize the kitchen. Emery represents an important and persistent cultural interest in going back-to-the-land. Modern homesteading practices also provide possible responses to contemporary post-industrial concerns including consumer culture, food insecurity, and the environment.

The role of lamin A and emerin in mediating genome organisation

The nuclear matrix (NM) is proposed to be a permanent network of core filaments underlying thicker fibres, present regardless of transcriptional activity. It is found to be both RNA and protein rich; indeed, numerous important nuclear proteins are components of the structure. In addition to mediating the organisation of entire chromosomes, the NM has also been demonstrated to tether telomeres via their TTAGGG repeats. In order to examine telomeric interactions with the NM, a technique known as the DNA halo preparation has been employed. Regions of DNA that are tightly attached to the structure are found within a so-called residual nucleus, while those sequences forming lesser associations produce a halo of DNA. Coupled with various FISH methodologies, this technique allowed the anchorage of genomic regions by the NM, to be analysed. In normal fibroblasts, the majority of chromosomes and telomeres were extensively anchored to the NM. Such interactions did not vary significantly in proliferating and senescent nuclei. However, a decrease in NM-associated telomeres was detected in quiescence. Since lamin A is an integral component of the NM, it seemed pertinent to examine chromosome and telomere NM-anchorage in Hutchinson-Gilford Progeria Syndrome (HGPS) fibroblasts, which contain mutant forms of lamin A. Indeed, genome tethering by the NM was perturbed in HGPS. In immortalised HGPS fibroblasts, this disrupted anchorage appeared to be rescued; the implications of this finding will be discussed. This study also suggested that telomere-NM interactions are aberrant in X-linked Emery-Dreifuss Muscular Dystrophy (X-EDMD), which is caused by mutant forms of emerin, another NM-associated protein. The positioning of selected genes in control and X-EDMD cell lines was examined in un-extracted nuclei using 2D and 3D FISH. Subtle shifts in the organisation of these genes were detected in diseased cells; however, their expression levels remained unaltered. Furthermore, in order to examine the architectural integrity of the nuclear lamina in lamin A and emerin mutant cell lines, scanning electron microscopy (SEM) was employed. This work revealed that such structures were indeed compromised in disease. The findings presented in this thesis highlight the importance of lamin A and emerin in mediating the organisation of the genome and taken together, promote the hypothesis that dysfunctional NM dynamics may well contribute to disease pathology.

Burgess / Skeletal muscle is a multinucleated cell type in which the many nuclei are precisely positioned to maximize the distance between adjacent nuclei. In order to reach this final positioning, nuclei undergo an elaborate set of movements during muscle development. The disruption of this process is evident throughout muscular dystrophies and myopathies. However, the contribution of aberrant nuclear positioning toward disease progression is unclear and the mechanisms regulating nuclear movement and positioning are poorly defined. The goal of this thesis is to determine the contribution of disease-linked genes to the regulation of nuclear movement and positioning and how these mechanisms are coordinated in skeletal muscle. In this thesis, we utilize Drosophila melanogaster skeletal muscle as an in vivo model system to investigate nuclear positioning throughout muscle development and correlate aberrant nuclear positioning with a decrease in muscle function. We provide the first evidence of distinct mechanisms that are independently regulated by genes that are associated with two different muscle diseases, Emery-Dreifuss muscular dystrophy and Centronuclear myopathy (Chapter 2). We also provide evidence that Emerin-dependent regulation of the LINC complex is a critical determinant of nuclear positioning and for the first time demonstrate a division of Emerin functions among the two Drosophila emerin homologs, bocksbeutel and otefin (Chapter 3). Finally, we conduct a proof-of-concept screen to identify novel regulators of muscle development and function (Chapter 4). Together, the work presented in this thesis provides a framework to further our understanding of the mechanisms regulating nuclear movement and positioning as well as muscle development as a whole. Weighted LP estimates on Riemannian manifolds / Estimations LP à poids sur des variétés riemanniennes

Cette thèse s'inscrit dans le domaine de l'analyse harmonique et plus exactement, des estimations à poids. Un intérêt particulier est porté aux estimations Lp à poids des transformées de Riesz sur des variétés Riemanniennes complètes ainsi qu'à l'optimalité des résultats en terme de la puissance de la caractéristique des poids. On obtient un premier résultat (en terme de la linéarité et de la non dépendance de la dimension) sur des espaces pas nécessairement de type homogène, lorsque p = 2 et la courbure de Bakry-Emery est positive. On utilise pour cela une approche analytique en exhibant une fonction de Bellman concrète. Puis, en utilisant des techniques stochastiques et une domination éparse, on démontre que les transformées de Riesz sont bornées sur Lp, pour p ∈ (1, +∞) et on déduit également le résultat précèdent. Enfin, on utilise un changement élégant dans la preuve précèdente pour affaiblir l'hypothèse sur la courbure et la supposer minorée. / The topics addressed in this thesis lie in the field of harmonic analysis and more pre- cisely, weighted inequalities. Our main interests are the weighted Lp-bounds of the Riesz transforms on complete Riemannian manifolds and the sharpness of the bounds in terms of the power of the characteristic of the weights. We first obtain a linear and dimensionless result on non necessarily homogeneous spaces, when p = 2 and the Bakry-Emery curvature is non-negative. We use here an analytical approach by exhibiting a concrete Bellman function. Next, using stochastic techniques and sparse domination, we prove that the Riesz transforms are Lp-bounded for p ∈ (1, +∞) and obtain the previous result for free. Finally, we use an elegant change in the precedent proof to weaken the condition on the curvature and assume it is bounded from below.

Cette thèse traite de plusieurs sujets d'analyse dans les espaces métriques mesurés, en lien avec le transport optimal et des conditions de courbure-dimension. Nous considérons en particulier les équations de continuité dans ces espaces, du point de vue de fonctionnelles continues sur les espaces de Sobolev, et du point de vue de la dualité avec les courbes absolument continues dans l'espace de Wasserstein. Sous une condition de courbure-dimension, mais sans condition de doublement de mesure ou d'inégalité de Poincaré, nous montrons également l'identification des p-gradients faibles. Nous étudions ensuite les espaces de Sobolev sur le produit tordu de l'ensemble des réels et d'un espace métrique mesuré. En particulier, nous montrons la propriété Sobolev-à-Lipschitz sous une certaine condition de courbure-dimension. Enfin, sous une telle condition et dans le cadre d'une théorie non-lisse de Bakry-Emery, nous obtenons une inégalité améliorée de Bochner et proposons une définition du N-tenseur de Ricci. / This thesis concerns in some topics on calculus in metric measure spaces, in connection with optimal transport theory and curvature-dimension conditions. We study the continuity equations on metric measure spaces, in the viewpoint of continuous functionals on Sobolev spaces, and in the viewpoint of the duality with respect to absolutely continuous curves in the Wasserstein space. We study the Sobolev spaces of warped products of a real line and a metric measure space. We prove the 'Pythagoras theorem' for both cartesian products and warped products, and prove Sobolev-to-Lipschitz property for warped products under a certain curvature-dimension condition. We also prove the identification of p-weak gradients under curvature-dimension condition, without the doubling condition or local Poincaré inequality. At last, using the non-smooth Bakry-Emery theory on metric measure spaces, we obtain a Bochner inequality and propose a definition of N-Ricci tensor.

In this essay I have read the novels Wide Sargasso Sea (Jean Rhys 1966), Desirada (Maryse Condé 1997) and Invisible Cities (Italo Calvino 1972) through Édouard Glissant’s notion of the primary scene in Philosophie de la relation. Poésie en étendue. I have examined how the primary scene can be seen as a political/an aesthetical strategy in the three novels. This was done in order to question the blunted tool of the notion of the identity, which is often considered in studies regarding ”postcolonial novels”. Invisible Cities is not belonging in this tradition, the method I used was ”The travelling theory” of Edward W. Saïd that proposes to let theories travel through different fields instead of trying to understand their ”original” meaning. I compared these novels in accordance to Glissants theory of questioning the notion of origins. The analysis compares how the novels depending on in wich tradition they have been read is being seen with different eyes. Novels interpreted as ”postcolonial novels” are usually read as a symptom of lack of origin. Whereas the protagonist tryes to understand the loss of identity. Glissants theory eases the analysis to reach other possible meanings of the novels, where they both meet and differs. My results indicate that The Wide Sargasso Sea and Desirada create a complex relation to time and space, where the primary scenes in these novels are steeped through different time levels. The two novels present a new way to understand time that rejects historicity, create new tools for thinking and evaluates new strategies to exist in the world, without origins in the classical meaning; Rhys through her imagery and Condé through her weave of times. The same goes for Invicible Cities, only though Venice the primal scene in the novel stays the center of interest, and even though borders of time and space are being questioned, the novel tends to make Venice the guzzling synthesis. / Édouard Glissant, Jean Rhys, Maryse Condé, Italo Calvino, Édward W.
