Vers une analyse fonctionnelle du TALome des souches Africaines de Xanthomonas oryzae pv. oryzae : identification d’une nouvelle stratégie de virulence. Les bactéries du genre Xanthomonas injectent à l’intérieure de la cellule hôte des effecteurs de type TAL (Transcription Activator-Like) qui agissent comme des facteurs de transcription spécifiques à l’aide d’un nouveau domaine de liaison à l’ADN qui est programmable. La bactérie pathogène du riz Xanthomonas oryzae pv. oryzae (Xoo) contient un nombre important de gènes TAL (de 9 à 16) dans leur génome. Tandis que un ou deux de ces gènes codent des facteurs de virulence majeurs, la contribution relative de chacun des autres gènes TALs à la pathogénie de Xoo reste encore obscure. Afin d’élucider cela, le génome complet de trois souches Africaines de Xoo a été analysé à l’aide de la technologie de séquençage PacBio. Une analyse phylogénétique des trois TALomes combinée à des prédictions de cibles des TAL in silico montre que les TALomes Africains sont très conservés et génétiquement distants des correspondants asiatiques. Les gènes TAL individuels des souches Africaines de Xoo MAI1 et BAI3 ont été sous-clonés via une nouvelle méthode d’analyse à moyen débit dans un vecteur d’expression compatible avec des tests de pathogénie. Un test systématique de « gain de virulence » de la fonction de chacun des 11 haplotypes TAL a été réalisé en les introduisant dans la souche de Xoo X11-5A qui est peu virulente et ne contient naturellement pas de TALs, révélant Tal2 comme un nouveau facteur de virulence majeur dans les souches Africaines de Xoo. La prédiction in-silico des cibles de Tal2 dans le promoterome de riz associée à des expériences de QRT-PCR ont mis en évidence deux cibles de virulence candidates de Tal2, OsTFX1 qui code un facteur de transcription de type bZIP déjà rapporté comme gène de sensibilité S commun à différentes souches Asiatiques de Xoo, et Os09g39810 (aussi appelé ici OsERF#123), qui code pour un facteur de transcription de la famille des protéines de type « ethylene response factor ». Des test de pathogénie ont confirmé l’induction spécifique de ce dernier obtenue à l’aide de TAL artificiels confère une sensibilité accrue du riz aux souches africaines de Xoo. Des expériences de type 5’-RACE ont montré que Tal2 induisait l’expression d’un transcrit alternatif caractérisé par une séquence « leader » plus longue. Nous faisons l’hypothèse que l’accumulation de ce transcrit alternatif spécifique de Tal2 interfère avec la fonction endogène de ce facteur de transcription de type ERF, dont nous montrons qu’elle est potentiellement impliquée dans l’induction des réponses de défense de la plante.Un autre axe de recherche de cette thèse a été d’initier la caractérisation des bactérioses du riz au Vietnam, qui est l’un des plus grands exportateurs de riz au monde. Tandis que la bactériose vasculaire due à Xoo a été rapportée plusieurs fois dans ce pays, une description formelle de la bactériose à stries foliaires causée par X. oryzae pv. oryzicola faisait défaut. Ici, nous avons confirmé l’occurrence de la BLS au Nord du Vietnam à l’aide d’une PCR-multiplexée. L’échantillonnage a également montré que les deux pathovars étaient présents dans cette région et parfois retrouvés sur la même feuille. Des analyses de la dynamique des populations de Xo et l’analyse fonctionnelle des TALomes serviront de base pour établir de nouvelles stratégies de lutte contre les xanthomonads infectant le riz au Vietnam. / Towards a functional analysis of African Xanthomonas oryzae pv. oryzae TALomes : identification of a novel virulence strategy Bacterial plant-pathogenic Xanthomonas translocate Transcription Activator-Like (TAL) effectors into plant cells to function as specific plant transcription factors via a novel programmable DNA-binding domain. Rice-pathogenic Xanthomonas oryzae pv. oryzae (Xoo) strains contain multiple TAL genes (from 9 to 16) in their genome. While one or two act as major virulence factors, the relative contribution of each of the other members to Xoo pathogenicity remains unclear. To address that question, three African Xoo TALome have been analyzed using whole genome PacBio sequencing data. A phylogenetic analysis of the three TALomes combined to TAL targets in-silico predictions showed that African TALomes are highly conserved and genetically distant from Asian ones.Individual TAL genes from African Xoo strains MAI1 and BAI3 were directly sub-cloned via a self-developing medium-throughput approach into an expression vector suitable for pathogenicity analysis. A systematic “gain-of-virulence” analysis of the function of each of the 11 TAL effector clusters was assessed in Xoo strain X11-5A which has low virulence and is naturally devoid of TAL genes, revealing Tal2 as a novel major virulence factor in African Xoo. In-silico prediction for Tal2 binding sites in the rice promoterome and qRT-PCR analysis highlighted two virulence targets for Tal2, i.e. OsTFX1, which encodes a bZIP transcription factor previously known as a common susceptibility S gene targeted by Asian Xoo, and Os09g39810 (also referred to as OsERF#123), which encodes a putative transcription factor of the ethylene response factor family. Pathogenicity assays further confirmed that designer TALE-mediated specific induction of Os09g39810 confers higher susceptibility of rice to African strains of Xoo. 5’-RACE experiments showed that Tal2 induces the transcription of an alternative Os09g39810 transcript characterized by a longer leader sequence. We hypothesize that accumulation of this Tal2-specific transcript interferes with the endogenous function of this ERF-encoding gene, which is potentially involved with the induction of defense responses. Another project was to initiate the characterization of bacterial diseases of rice in Vietnam, one of the biggest rice exporting country over the world. While the occurrence of bacterial blight which is due to Xoo, was documented in several reports in this country, there was no formal description of bacterial leaf streak which is due to X. oryzae pv. oryzicola. Here, we attempted to confirm the presence of BLS in North Vietnam by using multiplex-PCR assay. The survey also indicated that both pathovars appear in this area and eventually in the same leaf samples. Further analysis of Xo populations dynamic and functional analysis of TALomes will be useful to infer novel strategies to control rice-pathogenic Xanthomonads in this country in the future.

Les légumineuses sont capables de s’associer en symbiose avec des bactéries du sol Rhizobium. Cette interaction culmine par la formation d’un nouvel organe racinaire appelé nodule, à l’intérieur duquel les bactéries différentiées fixent l’azote atmosphérique sous une forme assimilable par la plante hôte. La mise en place de cette association repose sur un dialogue moléculaire entre les deux partenaires, faisant intervenir des signaux bactériens lipo-chitooligosaccharidiques appelés Facteurs Nod (FNods). Chez la légumineuse modèle Medicago truncatula, la perception de ces signaux symbiotiques au niveau de l’épiderme racinaire, initie une voie de signalisation qui conduit à des oscillations calciques nécessaires pour l`activation de gènes de la plante hôte, tel le gène marqueur ENOD11. Il a été montré que les facteurs de transcription ERF ERN1/ERN2, étroitement apparentés, agissent comme des activateurs directs de la transcription d’ENOD11, via leur liaison à la séquence cis régulatrice NFbox. Le mutant ern1 est de manière cohérente requis pour l’activation d’ENOD11 en réponse aux FNods mais également au cours des étapes suivantes d’infection et de développement nodulaire. Cependant, ce mutant présente un phénotype symbiotique partiel soulevant la question d’une redondance fonctionnelle, qui pourrait être attribuée à la présence du facteur ERN2, étroitement apparenté. Ainsi, au cours de ma thèse, j’ai étudié la relation fonctionnelle entre les facteurs ERN1/ERN2 par de approches diverses visant à déterminer leur expression et fonctions relatives au cours de la symbiose rhizobienne. Mon travail de thèse a dans tout d’abord porté sur l’étude des profils d’expression spatio-temporels de gènes ERN au cours de la symbiose rhizobienne, corroborée par la dynamique de localisation cellulaire des protéines de fusions ERN. Ces facteurs sont exprimés de manière séquentielle mais aussi conjointe aux cours de la signalisation FNods et l’infection rhizobienne. Par la suite, des expériences de complémentation croisée, dans le fond mutant ern1, ont montré qu’ERN2 peut remplacer ERN1 pour l’induction d’ENOD11 en réponse aux FNods et pour la formation de nodules, dès lors qu’il est exprimé sous le contrôle du promoteur d’ERN1. Ceci indique que ces facteurs ont des activités biologiques similaires et suggère que l’absence de complémentation d’ern1 par le facteur endogène ERN2 est probablement liée à une régulation transcriptionnelle différentielle de la part de leurs promoteurs. Enfin, nous avons initié la caractérisation phénotypique de nouvelles lignées mutées au niveau du gène ERN2, dans le but d’identifier les fonctions spécifiques de ce facteur au cours de la nodulation. A travers l’analyse d’une lignée Tilling (ern2.1) présentant une mutation ponctuelle dans le domaine de liaison à l’ADN, nous avons mis en évidence un rôle d’ERN2 dans la progression des cordons d’infection au niveau du cortex racinaire. Des études moléculaires ont permis de montrer que l’acide aminé muté est un résidu conservé et important pour la topologie du domaine de liaison à l’ADN, mais également pour l’activité transcriptionnelle d’ERN2 sur ENOD11. Contrairement à ern1, le mutants ern2.1 et ern2.2 (mutant d’insertion) sont capables de former des nodules. Néanmoins, l’infection nodulaire apparait dans les deux cas parfois défectueuse, conduisant à une sénescence précoce. Ces résultats démontrent qu’ERN2 remplit aussi des rôles spécifiques au cours de la nodulation, qui ne sont pas entièrement complémentés par ERN1. Il semblerait donc que les facteurs ERN contrôlent des étapes communes et divergentes de l‘infection rhizobienne, ERN1 ayant un rôle prépondérant dans l`initiation et progression de l’infection tandis qu’ERN2 aurait un rôle secondaire, plus centré dans la progression des cordons. La lignée double mutant ern1ern2.1, ouvre de nouvelles perspectives pour l’étude de la redondance fonctionnelle entre ces deux facteurs au cours des symbioses racinaires / Legumes are able to associate in symbiosis with Rhizobia bacteria in the soil, which culminates in the formation of a new organ referred to as the root nodule, within which differentiated bacteria fix nitrogen to the benefit of the host plant. The establishment of this association relies on a molecular dialogue between the two partners, involving bacterial lipo-chitooligosaccharide signals called Nod factors (NF). In the model legume Medicago truncatula, the perception of these symbiotic signals in the root epidermis, initiates a signaling pathway that leads to calcium oscillation responses required for the activation of downstream genes such as the well-characterized ENOD11. Previously, ERN1 and the closely-related ERN2 transcription factors (TFs) were reported as direct activators of ENOD11 via binding to the NFbox regulatory unit. In addition, phenotypic analysis of the ern1 knockout mutant has confirmed the importance of ERN1 not only during NF signaling but also throughout subsequent infection and nodule development stages. Nevertheless, the ern1 mutant displays a less severe phenotype compared to plants mutated in other NF signaling genes, raising the question of a possible functional redundancy with the endogenous closely-related ERN2 factor. My PhD project was focused on the study of the functional relationship between ERN1 and ERN2 TFs. By using a variety of strategies we aimed at determining both ERN expression profiles and relative functions during nodulation. We first examined the spatio-temporal expression profiles of these genes during rhizobial symbiosis and correlated this with the dynamics of cellular localization of ERN fusion proteins. These analyses revealed that these factors possess both common and distinct expression profiles, correlated with cell-type specific and dynamic in vivo protein accumulation, tightly associated with rhizobial pre-infection and subsequent infection stages in M. truncatula. Further cross-complementation studies in the ern1 mutant background showed that, when ERN2 is expressed under the control of the ERN1 promoter, it can fully restore the ern1 phenotype regarding NF-elicited ENOD11 activation and nodule formation. This indicates that these factors have similar biological activities and suggests that the incapacity of endogenous ERN2 to complement the ern1 mutant is mainly due to differences in their promoter activities. Finally, we also initiated a phenotypic characterization of M. truncatula ern2 mutant lines, in order to get a better insight into ERN2 specific functions during nodulation. The phenotypic analysis of a Tilling line (ern2.1) carrying a point mutation in a conserved amino acid in the ERN2 DNA binding domain, revealed a role for ERN2 during infection thread progression in the root cortex. Further molecular studies demonstrated that this mutated amino acid in the Tilling line is conserved and required for optimal DNA binding domain topology and transcriptional activity of ERN2 on its target ENOD11 gene. In addition, the ern2.1 line and a second ern2.2 insertional mutant line are both capable of forming nodules, in contrast to the ern1 mutant. Nevertheless, these nodules are partly infection defective leading to premature senescence. These findings provide evidence that ERN2 possesses specialized functions during nodulation that cannot be fully complemented by ERN1. This suggests that ERN possess common and divergent functions, ERN1 having a predominant role in rhizobial infection initiation and progression while ERN2 having a secondary and more centered role during infection thread progression. The ern1ern2.1 double mutant line, recently generated during my PhD, opens new perspectives to further study the functional relationship between ERN TFs during root endosymbioses.

Caractérisation structurale de l'éliciteur du virus X de la pomme de terre (PVX) : recherche, chez les plantes cultivées, de gènes hôtes, impliqués dans la résistance liée à Rx / Structural characterization of the elicitor of the potato virus X (PVX) : research of host genes, in crops, implicated in the Rx mediated resistance

De nos jours, les agents pathogènes des plantes, engendrent encore des pertes agricoles importantes. A terme, la solution la plus adaptée semble être la création de variétés de plantes cultivées présentant une résistance génétique à large spectre et durable. Créer de telles plantes nécessite la compréhension des mécanismes de résistance mis en jeu dans des pathosystèmes modèles, tels que celui impliquant le gène de résistance Rx et le virus X de la pomme de terre. Dans ce système, la protéine Rx intervient dans la perception d’un facteur d'avirulence issu du virus: sa protéine de capside (Cp). Cependant, les mécanismes moléculaires déclenchant la résistance chez la plante exprimant Rx demeurent obscurs. Cette étude s’est focalisée sur l’étape de reconnaissance de l’éliciteur viral par Rx. Une première approche a permis la mise en évidence d’un éliciteur minimal de 90 acides amine. L’étude d’un fragment légèrement plus grand par des méthodes de biochimie structurale, tend à exclure un modèle selon lequel, la différence de structuration tertiaire entre Cp élicitrice et Cp non élicitrice déterminerait la reconnaissance par Rx. Une seconde approche, de biologie moléculaire, a mis en évidence deux protéines hôte interagissant avec ce petit fragment éliciteur. L'étude s’est focalisée sur un gène codant un facteur de transcription, nbERF5, et a révélé que cette protéine interagit aussi bien avec les Cp de souches de Potexvirus avirulentes que virulentes dans la résistance liée à Rx, mais également avec la protéine de résistance Rx et l’intéracteur direct: RanGAP2. La poursuite de la caractérisation de cet ERF, permettra de déterminer son importance dans la résistance liée à Rx. / Nowadays, phytopathogenic agents are still causing significant agricultural losses. The most suitable option appears to be the creation of crop species carrying a genetic durable and broad spectrum resistance. In order to create such varieties, we need to understand the mechanisms underlying resistance, involved in model Pathosystems, Such as the one composed of the resistance gene Rx and the potato virus X. In that system, the host gene encodes a protein assimilated to a receptor implicated in the perception of an avirulence factor produced by the virus: its capside protein (Cp). Nevertheless, the molecular mechanisms triggering the resistance remain largely unknown. This study has been focused on the elicitor recognition mediated by Rx. A first approach led to the identification of a minimal elicitor containing 90 amino acids has. The structural characterization of a slightly larger protein fragment using biochemical methods suggested that the difference in the tertiary structuration of both elicitor and non-elicitor Cp would not be the determinant of Rx mediated recognition. Second, a molecular approach led to the discovery of two host proteins interacting with the small elicitor fragment. The work was focused on a transcription factor, nbERF5 and showed that this protein interacts similarly with elicitor or non-elicitor Cps of Rx mediated resistance. Interestingly, this gene product is able to directly interact with the Rx protein, but also with the direct interactor of Rx: RanGAP2, protein required for the Rx mediated resistance efficiency. Further characterization of this ethylen response factor will help us to understand its role in Rx mediated resistance.

Spectroscopy of selected metal-containing diatomic molecules

Fourier transform infrared emission spectra of MnH and MnD were observed in the ground <em>X</em><sup>7</sup>&sigma;<sup>+</sup> electronic state. The vibration-rotation bands from <em>v</em> = 1 to 0 to <em>v</em> = 3 to 2 for MnH, and from <em>v</em> = 1 to 0 to <em>v</em> = 4 to 3 for MnD were recorded at an instrumental resolution of 0. 0085 cm<sup>-1</sup>. Spectroscopic constants were determined for each vibrational level and equilibrium constants were found from a Dunham-type fit. The equilibrium vibrational constant <em>&omega;<sub>e</sub></em> for MnH was found to be 1546. 84518(65) cm<sup>-1</sup>, the equilibrium rotational constant <em>B<sub>e</sub></em> was found to be 5. 6856789(103) cm<sup>-1</sup> and the equilibrium bond distance <em>r<sub>e</sub></em> was determined to be 1. 7308601(47) Å. <br /><br /> New high resolution emission spectra of CoH and CoD molecules have been recorded in the 640 nm to 3. 5 <em>µ</em>m region using a Fourier transform spectrometer. Many bands were observed for the <em>A</em><sup>'3</sup>&phi;-<em>X</em><sup>3</sup>&phi; electronic transition of CoH and CoD. In addition, a new [13. 3]4 electronic state was found by observing the [13. 3]4- <em>X</em><sup>3</sup>&phi;<sub>3</sub> and [13. 3]4-<em>X</em><sup>3</sup>&phi;<sub>4</sub> transitions in the spectrum of CoD. Analysis of the transitions with &delta;&omega; = 0, ±1 provided more accurate values of spin-orbit splittings between &omega; = 4 and &omega; = 3 components. The ground state for both molecules was fitted both to band and Dunham-type constants. The estimated band constants of the perturbed upper states were also obtained. <br /> <br /> The emission spectrum of gas-phase YbO has been investigated using a Fourier transform spectrometer. A total of 8 red-degraded bands in the range 9 800 ? 11 300 cm<sup>-1</sup> were recorded at a resolution of 0. 04 cm<sup>-1</sup>. Because of the multiple isotopomers present in the spectra, only 3 bands were rotationally analyzed. Perturbations were identified in two of these bands and all 3 transitions were found to terminate at the <em>X</em><sup>1</sup>&sigma;<sup>+</sup> ground electronic state. The electronic configurations that give rise to the observed states are discussed and molecular parameters for all of the analyzed bands are reported. <br /><br /> Electronic spectra of the previously unobserved EuH and EuD molecules were studied by means of Fourier transform spectroscopy and laser-induced fluorescence. The extreme complexity of these transitions made rotational assignments of EuH bands impossible. Catharanthus roseus (Madagascar periwinkle) is the exclusive source of an array of terpenoid indole alkaloids (TIAs) that are used in the treatments of hypertension and certain types of cancer. TIA biosynthesis is under stringent spatiotemporal control and is induced by jasmonate (JA) and fungal elicitors. Tryptamine, derived from the indole branch, and secologanin from the iridoid branch are condensed to form the first TIA, strictosidine. Biosynthesis of TIA is regulated at the transcriptional level and several transcription factors (TFs) regulating the expression of genes encoding key enzymes in the pathway have been isolated and characterized. The JA-responsive APETALA2/ETHYLENE RESPONSE FACTOR (AP2/ERF), ORCA3, and the basic helix-loop-helix (bHLH) factor, CrMYC2, are the key activators of the TIA biosynthesis. Recently, two other TFs, the bHLH IRIDOID SYNTHESIS 1 (BIS1) and BIS2 were also identified as regulators of TIA pathway. Analysis of C. roseus genome sequence has revealed that ORCA3 forms a physical cluster with two uncharacterized AP2/ERFs, ORCA4 and ORCA5. In plants, physically linked clusters of TFs are less characterized. Moreover, the regulation of TF clusters is relatively unexplored. My research uncovered that the ORCA gene cluster is differentially regulated. ORCA4 and ORCA5, while functionally overlapping with ORCA3, regulate an additional set of TIA pathway genes. ORCA4 or ORCA5 overexpression has resulted in significant increase of TIA accumulation in C. roseus hairy roots. In addition, ORCA5 directly regulates the expression of ORCA4 and indirectly regulates ORCA3, likely via unknown factor(s). Interestingly, ORCA5 also activates the expression of ZCT3, a negative regulator of the TIA pathway. In addition CrMYC2 is capable of activating ORCA3 and co-regulating pathway genes concomitantly with ORCA3. Several lines of evidence suggest that, in addition to the transcriptional control, biosynthesis of TIAs is also controlled at the posttranslational level, such as protein phosphorylation. Available literature indicates that a mitogen-activated protein kinase (MAPK) cascade is involved in this process. Analysis of C. roseus MAP kinome, identified two independent MAPK cascades regulating the indole and iridoid branches of the TIA pathway. We showed that the ORCA cluster and CrMYC2 act downstream of a MAP kinase cascade consisting of CrMAPKK1, CrMAPK3 and CrMAPK6. Overexpression of CrMAPKK1 in C. roseus hairy roots upregulates TIA pathway genes expressions and boosts TIA accumulation. The other cascade, consisting of CrMAPKK6 and CrMAPK13, mostly regulates the iridoid branch of the TIA pathway. Overexpression of CrMAPK13 in C. roseus hairy roots significantly upregulates iridoid pathway genes and boosts tabersonine accumulation. Moreover, we recently identified the third MAPK cascade, consisting of CrMAPKK1 and CrMAPK20, that negatively regulates the indole branch of the TIA pathway. Overexpression of CrMAPK20 in C. roseus hairy roots represses the genes regulated by CrMYC2-ORCAs and reduces catharanthine accumulation. These findings significantly advance our understanding of transcriptional and post-translational regulatory mechanisms that govern TIA biosynthesis in C. roseus.

Thesis (MA)--Stellenbosch University, 2013. / ENGLISH ABSTRACT: Traditionally the body has been neglected within Western philosophical thought. In this tradition man is seen as a dualist organism whose mind is more highly valued than his or her body. Since the start of the twentieth century, especially, there has also been Western thinkers who deconstruct the traditional, opposisional view of the mind and body. Most of these thinkers are men, and they argue from a male perspective. The deconstruction of the opposition between mind and body is, however, also useful for feminism, as the woman was traditionally seen as more bodily, and less rational, than the man. This view of woman can be used to support arguments that her place is within the home, where she should look after the bodily and emotional needs of her husband and children. First and second wave feminists criticised above-mentioned traditional view of women by arguing that the woman is just as rational as the man. Other feminists make use of the 'male' criticism of the body-mind opposition by arguing that traditionally female attributes and activities are not less valuable than male ones. In this thesis it is argued that the novellas Klaaglied vir Koos, Erf and Louoond should be read against this background as feminist texts in which the traditionally female space of the kitchen is celebrated as a space in which the female subject can create art and develop a perspective on the outside world. Focusing on the body and on traditionally female values (such as the domestic space) have implications for, amongst others, epistemology, ethical thought and literary theory. The way these implications are developed in the three novellas are explored in this thesis. Domestic spaces are not only celebrated in these texts, they are also represented as problematical, limited spaces which isolate the protagonists of the novellas and withhold them from the potential of political solidarity. The novellas are therefore treated in this thesis as complex texts in which the literary focus on domestic space, female embodiment and man‘s relationship with food is both celebrated and problematised. / AFRIKAANSE OPSOMMING: Tradisioneel is die liggaam afgeskeep in die Westerse filosofie. Binne hierdie tradisie is die mens beskou as ‘n dualistiese organisme wie se verstand meer waardevol is as sy of haar liggaam. Veral sedert die begin van die twintigste eeu is daar ook Westerse denkers wat die tradisionele, opposisionele siening van die liggaam en die verstand dekonstrueer. Die meeste van hierdie denkers is mans en neem slegs die manlike perspektief in ag. Die dekonstruksie van die opposisie tussen liggaam en verstand is egter ook nuttig vir die feminisme, omdat die vrou tradisioneel gesien is as meer liggaamlik en minder rasioneel as die man. Hierdie tradisionele siening van die vrou kan gebruik word om te argumenteer dat haar plek in die huishoudelike ruimte is, waar sy kan omsien na die liggaamlike en emosionele behoeftes van haar man en kinders. Eerste- en tweedegolf-feministe het bogenoemde tradisionele opvattings veral gekritiseer deur te argumenteer dat die vrou net so rasioneel soos die man is. Latere feministe bou egter voort op 'manlike‘ kritiek teen die opposisie tussen liggaam en verstand deur te argumenteer dat tradisioneel vroulike eienskappe en aktiwiteite nie minder waardevol is as sogenaamde manlike eienskappe nie. In hierdie tesis word geargumenteer dat die novelles Klaaglied vir Koos en Erf deur Lettie Viljoen en Louoond deur Jeanne Goosen teen hierdie agtergrond gelees kan word as feministiese tekste waarin die tradisioneel vroulike ruimte van die kombuis gevier word as ‘n ruimte waarin die vroulike subjek ‘n perspektief op die buitewêreld kan ontwikkel en kuns kan skep. ʼn Fokus op die liggaam en op tradisioneel vroulike elemente (soos die huishoudelike ruimte) behels implikasies vir onder andere die epistemologie, etiese denke en literêre teorie. Hierdie implikasies word verken soos wat hulle na vore kom in die drie novelles. In die novelles word die huishoudelike ruimte egter ook uitgebeeld as ʼn problematiese, beperkende ruimte wat die hoofkarakters van die novelles afsny van die buitewêreld en van die moontlikheid van politieke solidariteit. Daar word geargumenteer dat die novelles dus komplekse tekste is waarin literatuur wat fokus op die huishoudelike ruimte, vroulike liggaamlikheid en die mens se verhouding met kos sowel gevier en geproblematiseer kan word.

Frequently, the main determinant of gene expression is its transcriptional rate. Thus, molecular mechanisms underlying transcription regulation have become an important topic in genetic studies of ethylene signaling. The present work aimed to investigate the ERF (Ethylene Response Factor) family employing bioinformatic tools, integrating publicly available datasets from the model species Arabidopsis thaliana and phylogenetic analyses to help elucidating the biological roles of the family in apple. The preliminary survey of the ERF sequences in Malus has provided basic information to be incorporated in further studies of the functional role of ERFs in this perennial species. Expression analyses of MdERF1 and MdERF in apple fruits suggest that other factors, besides ethylene, are involved in their transcriptional regulation in Malus. The second chapter reports the investigation of the transcriptional profiling of those ERF genes in response to pathogen attack, using a biological assay of in vitro propagated plants inoculated with the fungus Venturia inaequalis (apple scab disease). The study has provided evidences of the involvement of MdERF1 in eliciting the plant response; whereas, MdERF2 does not appear to be participate in the pathogenesis. / Moléculas que participam dos processos regulatórios, como os fatores de transcrição, têm recebido atenção especial, pois uma das principais ações dos estímulos hormonais é a modulação da expressão gênica. Como a taxa de transcrição de um gene é o maior determinante da sua expressão, os mecanismos moleculares pelos quais a transcrição gênica é regulada têm se tornado um dos tópicos principais de estudos em genética molecular envolvendo o hormônio etileno. O objetivo deste trabalho foi realizar análises de bioinformática para a família ERF (Ethylene Response Factors), integrar bases de dados existentes na internet no modelo Arabidopsis, bem como análise filogenética que permitam avaliar os papéis dos diferentes membros da família. Este levantamento preliminar das seqüências ERF em Malus forneceu informação básica para estudos posteriores mais aprofundados, com relação aos mecanismos moleculares da família nesta importante cultura perene. A análise da expressão de MdERF1 e MdERF2 em frutos de maçã indica que outros fatores além do etileno estão envolvidos na regulação da transcrição dos ERF em Malus. O segundo capítulo refere-se à resposta dos ERF frente ao ataque de patógenos. Para isso, foram infectadas plantas de macieira provenientes de cutivo in vitro com o fungo Venturia inaequalis (sarna da maçã). As evidências desses estudos sugerem o envolvimento do gene MdERF1 no processo de patogênese, enquanto que o gene MdERF2 parece não estar envolvido no processo.
