Estudo da expressão de genes associados ao perfil de ácidos graxos em bovinos Nelore confinados / Study of the gene expression associated to fatty acid profile in Nellore cattle feedlot

Nos últimos anos tem-se destacado a importância dos ácidos graxos presentes na carne e leite, em especial aqueles que poderiam influenciar a saúde humana. A deposição de gordura, e também sua composição, é resultado da interação entre os fatores genotípicos e fenotípicos. Sendo assim, o contínuo melhoramento genético, com a seleção de determinados genes, pode influenciar na qualidade do produto que chega à mesa do consumidor. Objetivou-se com este estudo avaliar os padrões de expressão dos genes diferencialmente expressos relacionados ao perfil de ácidos graxos da carne de bovinos Nelore confinados, por meio da verificação dos padrões de expressão de genes envolvidos no metabolismo lipídico e na síntese dos ácidos graxos palmítico (C16:0), esteárico (C18:0), oléico (C18:1 cis-9), linoléico (C18:2 cis-9 cis- 12), CLA (C18:2 cis-9 trans-11) e linolênico (C18:3). Foram utilizados 48 bovinos machos inteiros, Nelore, com idade aproximada de 24 meses, dos quais foram coletadas amostras do músculo Longissimus para a realização das análises. Verificou-se um total de 1173 genes diferencialmente expressos entre os grupos de baixa e alta concentração de cada um dos ácidos graxos. Os genes diferencialmente expressos identificados no músculo Longissimus foram ACAT1, ACOX2, ACOT11, ACSM3, ACSS1, AGPAT6, BDH1, CIQTNF3, CYP4B1, DGAT2, FABP3, FABP4, FABP7, GK, IGF2, LCAT, LIPE (HSL), LPL, PLIN1, PLIN5, SCD5 e SLC27A6, que possuem ações nas vias metabólica dos ácidos graxos ou nas vias adjacentes, podendo influenciar sobre o perfil lipídico da carne. Estudos com RNAseq envolvendo o perfil de ácidos graxos na carne ainda são escassos, e em animais zebuínos não há relatos. Desta forma, ainda são necessários mais estudos a fim de esclarecer como determinados genes podem influenciar as características desejadas, como por exemplo, o perfil de ácidos graxos da gordura intramuscular. / Recently, the importance of fatty acids on meat and milk has been highlighted, especially those that could affect human health. The fat deposition, and its composition, is a result of the interaction between genotypic and phenotypic factors. In this way, the continuous genetic improvement, with the selection of some genes, can influence the quality of the product acquired by the consumers. The aim of this study was evaluate the patterns of expression of differentially expressed genes related to the fatty acid profile of Nellore cattle feedlot, by verifying the patterns of expression of genes involved in lipid metabolism and in the synthesis of fatty acids palmitic (C16: 0), stearic (C18: 0), oleic (C18: 1 cis-9), linoleic (C18: 2 cis-cis- 9-12), CLA (C18: 2 cis-9 trans-11) and linolenic acid (C18: 3). Forty-eight bovine, male, Nellore, with an average age of 24 months, from which were collected samples of Longissimus Muscle to carry out the analysis. There were a total of 1173 differentially expressed genes between groups of low and high concentration of each of the fatty acids. Differentially expressed genes identified in Longissimus were ACAT1 ACOX2, ACOT11, ACSM3, AGPAT6, BDH1, ACSS1, CIQTNF3, CYP4B1, DGAT2, FABP3, FABP4, FABP7, GK, IGF2, LCAT, LIPE (HSL), LPL, PLIN1, PLIN5, SCD5 and SLC27A6, that are related to metabolic pathways of fatty acids or the adjacent pathways, and may influence on meat lipid profile. Studies with RNAseq involving fatty acid profile in meat are still rare, especially in Zebu cattle, in which there are no reports. In this way, further studies are still needed in order to clarify how certain genes can influence the desired characteristics, for instance, the fatty acid profile of intramuscular fat content.

O Brasil é o maior produtor de laranja doce do mundo, mas a história da citricultura brasileira é marcada por sucessivas perdas devido a pragas e doenças que atacam os pomares. Entre as doenças que afetam os pomares de citros, o huanglongbing (HLB) tem merecido destaque nos últimos anos. O HLB já é conhecido desde 1900 na China, mas no Brasil essa doença está presente desde 2004 e tem causado perdas significativas a citricultura. Essa doença é associada a três espécies de \"Candidatus Liberibacter\", mas no Brasil a espécie Candidatus Liberibacter asiaticus (CLas) é a mais comum. Devido à ausência de plantas de laranja doce resistentes a essa doença, a busca por plantas transgênicas que apresentem resistência a essa doença têm se intensificado nos últimos anos. Uma estratégia para projetar uma construção gênica visando a CLas é a utilização de promotores floema-específico, pois a CLas coloniza o floema das plantas de citros infectadas. Entretanto, para provar que uma sequência promotora funciona é preciso desafiar a construção gênica na presnça da CLas. Portanto, conduziu-se este trabalho com o objetivo de multiplicar plantas de Citrus sinensis Osbeck (L.) cv. \'Hamlin\' transgênicas contendo o gene uidA sob controle dos promotores floema-específicos Arabidopsis thaliana phloem protein 2 (AtPP2), Citrus phloem protein 2 (CsPP2) e Arabidopsis thaliana sucrose transporter 2 (AtSUC2), inocular com CLas por Diaphorina citri, avaliar a interdependência entre a expressão do transgene (gene uidA) e a concentração de CLas, para poder inferir sobre o controle da expressão dos promotores quando as plantas são inculadas com CLas. Para isso, cinco eventos de transformação de cada construção gênica, contendo apenas uma cópia do transgene foram selecionados, multiplicados e inoculados com CLas por Diaphorina citri, dezoito meses após a inoculação, o DNA e o RNA das plantas foram coletados. Foi realizada análise para verificar a concentração de CLas em todas as plantas inoculadas e a expressão do gene uidA foi realizada em uma linhagem transgênica de cada construção gênica. Com o auxílio da análise de coeficiente de correlação de Person, foi possível classificar qualitativamente a interdependência entre a concentração bacteriana e a expressão gênica. A frequência média de inoculação de CLas por Diaphorina citri foi de 30 %, demostrando que é possível incular CLas via Diaphorina citri. A análise de coeficiente de correlação de Person mostrou que nas construções com promotores AtPP2 e CsPP2 há baixa interdependência entre o controle da expressão gênica e a concentração de CLas. A construção sob controle do promotor AtSUC2 apresentou correlação forte e positiva, mostrando que quanto maior a concentração de CLas maior a expressão do transgene. / Brazil is the largest producer of sweet oranges in the world, however the Brazilian citrus history is marked by successive losses due to pests and diseases attacking orchards. Among the diseases affecting citrus groves, the huanglongbing (HLB) has been highlighted in recent years. The HLB is known in China since 1900, although in Brazil this disease has been present since 2004 and has caused significant losses to citrus industry. This disease is associated with three species of \"Candidatus Liberibacter\", however, in Brazil Candidatus Liberibacter asiaticus species (CLas) is the most common. Due to the absence of sweet orange plants resistant to this disease, the search for transgenic plants with resistance to this disease have intensified in recent years. One strategy to design a genetic construct targeting the CLas includes the use of phloem-specific promoters, because the CLas colonizes the phloem of infected citrus plants. On the other hand, to prove that a promoter sequence works it is necessary to challenge the genetic construct in the presence of CLas. Therefore, we conducted this study with the objectives of multiplying transgenic Citrus sinensis Osbeck (L.) cv. \'Hamlin\' bearing the uidA gene under the control of the phloem-specific promoters Arabidopsis thaliana phloem protein 2 (AtPP2), Citrus phloem protein 2 (CsPP2) and Arabidopsis thaliana sucrose transporter 2 (AtSUC2), inoculate CLas with Diaphorina citri, and evaluate the interdependence between the expression of the transgene (uidA gene) and the concentration of CLas, to infer the promoter expression when the plants are inoculated with CLas. Five events processing of each gene construct, containing only one copy of the transgene, and inoculated with CLas by Diaphorina citri. The DNA and RNA of plants were collected after eighteen months of inoculation. The concentration of CLas in the inoculated plants and the expression of the uidA gene were performed in each transgenic line of each gene construct, with the aid of the analysis of Person correlation coefficient. Thus it was possible to classify qualitatively the interdependence between bacterial concentration and gene expression. The average frequency of CLas inoculation by Diaphorina citri was 30%, demonstrating that it was possible to inoculate CLas via Diaphorina citri. Person correlation coefficient values suggested low interdependence between the control of gene expression and the concentration of CLas in constructions with promoters AtPP2 and CsPP2. The construction under AtSUC2 promoter control showed strong positive correlation, indicating that the higher the concentration of CLas the greater is the transgene expression.

Estudo da monitorização contínua de glicose e das respostas de pressão arterial, frequência cardíaca e de outros parâmetros fisiológicos antes e após treinamento físico em diabéticos tipo II / Study of continuos glucose monitoring and responses in blood pressure, heart rate and others physiological parameters before and after physical training in type II diabetics

Há muitas alterações nos sistemas fisiológicos de indivíduos com diabetes melittus em função dos constantes momentos de hiperglicemia, principalmente alterações relacionadas ao aumento dos riscos cardiovasculares. O objetivo desse estudo foi avaliar as respostas do controle glicêmico pelo monitor contínuo de glicose e da pressão arterial (PA), frequência cardíaca (FC) e sua variabilidade expressa pelos valores de RMSSD em diabéticos tipo II submetidos a testes de avaliação antes e após a realização de treinamento aeróbio e resistido. Participaram desse estudo 9 voluntários diabéticos tipo II do sexo masculino (45 a 65 anos) divididos em 3 grupos: DTA (n=7), diabéticos submetidos a seis semanas de treinamento aeróbio; DTR (n=5), diabéticos submetidos a treinamento resistido e GDC (n=5), diabéticos sem qualquer treinamento regular. Os voluntários realizaram testes laboratoriais, ergoespirometria e teste de fadiga em leg press antes e após o treinamento físico. Os resultados foram analisados estatisticamente pelo teste t de Student e pelo teste de Kruskal Wallis. Os voluntários tiveram a cinética da concentração de glicose mensurada pelo monitor contínuo e analisada qualitativamente antes, durante e após a realização da ergoespirometria e do teste de fadiga por 60 minutos. Como resultados o grupo DTA apresentou menores valores de concentração de glicose pela monitorização contínua e o grupo DTR a melhor resposta na cinética dessa curva, apresentando expressivo decaimento na mesma. Em relação à resposta pressórica, somente a PA diastólica (PAD) foi menor estatisticamente para o grupo DTA pós treinamento aeróbio no repouso. Não houve diferenças entre os valores pré e pós treinamentos em relação à FC e os voluntários do grupo DTA apresentaram maiores valores de RMSSD em repouso e o do grupo DTR incrementos desses valores na recuperação dos testes, mostrando maior ação parassimpática no controle autonômico cardíaco dos diabéticos submetidos a treinamentos. Os indivíduos do grupo GDC apresentaram decremento nesse valor, sugerindo piora no controle autonômico cardíaco. Como conclusão geral, este estudo sugere que indivíduos diabéticos tipo II que realizaram treinamento aeróbio e resistido apresentaram benefícios complementares no controle glicêmico registrado pelo monitor contínuo em repouso e no período de recuperação de exercício, respectivamente, adaptações que parecem estar associadas à melhora da ação parassimpática/vagal no controle autonômico cardíaco e, sugere, também, ser o treinamento físico aeróbio o que permite melhor organização hemodinâmica nas respostas de PAD. / There are many changes in physiological systems of people with diabetes melittus due to the constant moments of hyperglycemia, mainly related to increasing of cardiovascular risk. The aim of this study was evaluate the responses of glycemic control by continuos glucose monitoring and blood pressure (BP), heart rate (HR) and its variability expressed by the values of RMSSD in type II diabetics undergoing evaluation tests before and after performing aerobic and resistance training. Participants were 9 volunteers type II diabetic male (45-64 years) divided in 3 groups: DTA (n=7), diabetics undergoing six weeks of aerobic training; DTR (n=5), diabetics undergoing resistance training and GDC (n=5), diabetics without any regular training. The volunteers underwent laboratory tests, spirometry and fatigue tests on leg press before and after physical training. The results were statistically analyzed by Students t and Kruskal Wallis tests. The volunteers had the kinetics of glucose concentration measured by the continuos monitor and qualitatively analyzed before, during and after the spirometry and the fatigue tests for 60 minutes. As a result the DTA group had lower glucose concentration by continuos monitoring and DTR the best response in the kinetic curve, showing important decrease in it. In relation to the BP response, only diastolic BP (DBP) was statistically lower for the DTA group after aerobic training. There were no differences between pre and post training in HR and the DTA group showed higher RMSSD at rest and the DTR group showed increments of these values in the tests recovery showing higher parasympathetic action on cardiac autonomic control in diabetics patients with training. Individuals in the GDC group showed decrement this value, suggesting deterioration in cardiac autonomic control. As a general conclusion, this study suggests that type II diabetic individuals who performed aerobic and resistance training showed additional benefits in glycemic control by continuos monitor recorded at rest and during exercise recovery, respectively, adaptations that seem to be associated with improvement in parasympathetic action in cardiac autonomic control, and also suggests that aerobic exercise training has better organization hemodynamic in responses of DBP.

Para isso investigou-se a experiência emocional vivenciada na entrevista familiar em âmbito nacional (Porto Alegre, Brasil) e internacional (Porto, Portugal). Objetivo geral: propor um modelo de desenvolvimento de perspectiva de autoconhecimento baseada nos referenciais teóricos e nos desvelares promovidos pela produção de dados. Objetivos específicos: 1) promover experimentações estéticas com os coordenadores avançados em transplantes para acessar os conteúdos inconscientes presentes no processo de doação de órgãos; 2) desvelar ações de promoção em saúde mental baseadas no autoconhecimento dos coordenadores avançados em transplantes observadas nos cenários nacionais e internacionais; 3) revelar por meio da produção de dados expressivos evidenciados através da Sociopoética, novas estratégias de enfrentamento das situações difíceis; 4) analisar a repercussão das estratégias de enfrentamento delineadas para as situações de comunicação difícil no cotidiano laboral dos coordenadores avançados em transplantes. Método: Pesquisa de abordagem Sociopoética, com parte de registro em diário de participante. A maior parte dos dados foram produzidos por meio de encontros com o grupo-pesquisador em abril de 2016, e outra parte foi coleta por meio das impressões observadas e registradas de diário de campo de outubro de 2016 à janeiro de 2017. Os participantes foram os coordenadores avançados em transplantes integrantes das equipes que atuam no processo de doação de órgãos em ambos os países investigados. A análise dos dados sociopoéticos foi do tipo, classificatória, transversal e filosófica, enquanto os dados internacionais foram submetidos a análise de conteúdo. Todos os dados produzidos têm como fonte principal de interpretação a perspectiva teórica de Juan Casassus e Jean Watson. Estudo aprovado no CEP respeitando os princípios éticos de pesquisas com seres humanos. Resultados e Discussão: De acordo com os dados foi possível observar a formação de três eixos principais que foram: Autoconhecimento, relacionado ao Coordenador e relacionado às Associações Inconscientes; Estratégias de enfrentamento das situações difíceis, sobre Si Mesmo (Lidar consigo mesmo); e , Ações de Promoção à Saúde Mental. Sendo assim, foi possível estabelecer trocas de saberes, não somente sobre o processo duro, quase aristocrático da doação, mas de saberes outros, socialmente construídos (FREIRE, 2002), muito relacionados ao seio familiar como os que envolvem valores, sentimentos, sensações, medos, inseguranças, que são trivialmente recalcados à turbulência existencial humana. Considerações Finais: Os coordenadores avançados em transplantes iniciaram o movimento de autoconhecimento ao fazerem associações de situações vivenciadas no processo de doação de órgãos com elementos do inconsciente tornando-os acessíveis ao nível consciente, em sequência, ao identificarem o desvelar de suas emoções e as representarem em situações ocorridas frequentemente, conseguiram observar que é preciso se posicionarem mais em relação a si mesmos, e se questionarem como estão enfrentando seus conflitos, aplicando tais conhecimentos na prática, de modo a se reorganizarem e a lidarem melhor com as situações difíceis concernentes a entrevista familiar para doação de órgãos / This study aimed to understand self-knowledge as a strategy for the maintenance of mental health of the transplant teams that perform in family interview for donation of organs. For that, the emotional experience experienced in the family interview at Porto Alegre, Brazil and at Porto, Portugal interviews was investigated. General objective: to propose a model of development of a perspective of self-knowledge based on the theoretical references and on the disclosures promoted by the production of data. Specific objectives: 1) to promote aesthetic experiments with the advanced coordinators in transplants to access the unconscious contents present in the process of organ donation; 2) unveil mental health promotion actions based on the self-knowledge of advanced transplant coordinators observed in national and international settings; 3) to reveal, through the production of expressive data evidenced through Sociopoetics, new strategies for coping with difficult situations; 4) to analyze the repercussion of coping strategies outlined for communication of difficult situations in the daily work of the advanced transplant coordinators.Method: Sociopoetic approach research, with onde part registered in field journal. Most of the data were produced through meetings with the researcher group in April 2016, and another part was collected through observed impressions and wsa registered in a field journal from October 2016 to January 2017. Participants were the Advanced coordinators in transplantation of the teams that work in the process of donating organs in both countries investigated. The analysis of sociopoetical data was of the type, classificatory, transversal and philosophical, while international data were submitted to content analysis. All the data produced have as main source of interpretation the theoretical perspective of Juan Casassus and Jean Watson. Study approved in the CEP respecting the ethical principles of researches with human beings. Results and Discussion: According to the data it was possible to observe the formation of three main axes that were: Self-knowledge, related to the Coordinator and related to the Unconscious Associations; Strategies to cope with difficult situations, about Self (Dealing with oneself); And, Actions to Promote Mental Health. Thus, it was possible to establish exchanges of knowledge, not only about the hard, almost aristocratic process of donation, but also about other socially constructed knowledges (FREIRE, 2002), which are closely related to the family as those involving values, feelings, Fears, insecurities, which are trivially repressed by human existential turbulence. Final Considerations: Advanced transplant coordinators initiated the self-knowledge movement by making associations of situations experienced in the process of organ donation with elements of the unconscious making them accessible to the conscious level in sequence by identifying the unveiling of their emotions and representing them in situations that have occurred frequently, have been able to observe that they need to position themselves more in relation to themselves, and question how they are facing their conflicts, applying such knowledge in practice, in order to reorganize themselves and to deal better with the difficult situations concerning the interview for organ donation

Recentemente desenvolvida, a biotecnologia denominada por Microarrays permite o monitoramento simultâneo dos valores de expressão gênica de centenas de milhares de genes, fator este que traz uma nova interpretação aos resultados obtidos em pesquisas desenvolvidas nas mais diversas áreas do conhecimento incluindo, por exemplo, a Farmacologia e Medicina, uma vez que os resultados obtidos são interpretados ao nível molecular. Contudo, apesar de muita tecnologia ser empregada à técnica de Microarrays, sua aplicação ainda ocasiona algumas complicações decorrentes, por exemplo, das inúmeras fontes de variação existentes, da escala das respostas ou da natural dificuldade de se analisar uma grande quantidade de fragmentos genéticos avaliados sob poucas unidades experimentais. Frente a estas complicações, atualmente, muitas são as propostas metodológicas de análises estatísticas para atenuar ou eliminar os problemas inerentes à técnica de Microarrays e propiciar a extração de resultados mais confiáveis a partir dos valores de expressão gênica, porém muitos desafios ainda persistem. Sob esta colocação, o presente trabalho procurou explorar duas metodologias de análise estatística alternativas no que diz respeito a seus conceitos, embora ambas tenham sido contextualizadas ao problema de Microarrays e aplicadas para se atingir o mesmo objetivo: possibilitar a identificação dos genes diferencialmente expressos sob distintas condições experimentais. A primeira metodologia consistiu da aplicação de Modelos de Análise de Variância de efeitos fixos com a adoção de modificações nas estatísticas de teste, metodologias de correções para múltiplos testes e a construção de gráficos vulcão. Já, a segunda metodologia consistiu da contextualização e aplicação da Teoria da Resposta ao Item TRI aos experimentos de Microarrays, abordagem esta pouco explorada na análise deste tipo de dado, mas a qual possibilita a seleção de genes diferencialmente expressos a partir de uma medida latente estimada para cada gene e a construção de uma escala para as categorias de resposta de expressão gênica. A motivação para este trabalho originou de um experimento de Microarrays com ratos congênicos disponibilizado pelo Laboratório de Cardiologia e Genética Molecular do Instituto do Coração (InCor-USP) cujo objetivo é identificar genes associados à hipertensão. / Recently developed, the biotechnology denominated Microarrays permits a simultaneous monitoring of the gene expression values of hundred thousands of genes; fact that introduces a new interpretation of the results obtained in researches developed in many distinct areas including, for example, Pharmacology and Medicine, once the obtained results are read according to the molecular level. However, despite the fact that much technology is used in the Microarrays technique, its application still causes some implications, for example, the countless sources of existing variance, the scale of answers or the natural difficulty in analyzing a large number of genetic fragments measured by few experimental units. Facing such complications, a lot of methodologies were suggested in order to reduce or eliminate the problems caused by the Microarrays technique and also foster the obtainment of more reliable results from the gene expression values, yet many challenges still persist. Under this perspective, the present work aimed at exploring two alternative methodologies regarding concepts, despite both were contextualized according to the Microarrays problem and applied with the same objective: enabling the identification of the genes differently expressed under different experimental conditions. The first methodology was composed by the application of Analysis of Variance Models of fixed effects with changes in the test statistics, correction methodologies for multiple tests and volcano plot. The second methodology consisted of the contextualization and application of the Item Response Theory IRT towards the Microarrays experiments, being this one not much explored in analysis that use this kind of data, but enabling the selection of genes differently expressed from an estimated latent trait for each gene and the construction of a scale for the categories of gene expression answers. The motivation for the present work came from an experiment of Microarrays with congenic mice made available by the Cardiology and Molecular Genetics Laboratory of the Heart Institute (InCor-USP) that aimed at identifying genes associated with hypertension.

Obesity leads to a reduction in life expectancy through its association with chronic diseases of lifestyle. The prevalence of obesity is rapidly increasing throughout the world. It is now accepted that most cases of obesity result from an interaction between genetic and environmental factors. This rapid increase in obesity generally leads to an increase in morbidity and mortality from chronic diseases such as cardiovascular disease, type 2 diabetes, osteoarthritis and cancer of which obesity is a risk factor. There is a lack of information in molecular research to explain how obesity predisposes individuals to these diseases. Proteomics is a molecular tool and a set of techniques used to identify changes at protein level from a diseased state. This study aims to identify differentially expressed proteins in serum of obese rats fed different isocaloric diets using proteomics.

Long-term Outcome, Suicidal behaviour, Quality of Life and Expressed Emotion in Adolescent Onset Psychotic Disorders

<p>This study investigated a consecutive cohort of 88 youngsters with onset of a psychotic disorder at age 15.7 (sd 1.5) years and followed-up 10.6 (sd 3.6) years after first admission at the age of 26.5 (sd 3.7) years. A subsample of 15 subjects were assessed with the Five Minute Speech Sample for measuring Expressed Emotion and subsequent recording of relapses during a two year period.</p><p>A diagnostic split between schizophrenia spectrum psychosis and affective psychotic disorder was usually stable over time. The main diagnostic shift was an influx to schizophrenia spectrum disorder of subjects with a better premorbid function and less insidious onset as compared to those with a stable schizophrenia diagnosis.</p><p>Early onset schizophrenia spectrum disorder usually had a poor functional outcome. Most subjects needed support in the form of a disability pension. Early onset affective psychotic disorder usually had a good functional outcome. Most subjects worked and enjoyed regular friendships. The functional level before onset of illness was the best predictor of future functional level in psychotic disorders. A family history of non-affective psychosis predicted a worse function in schizophrenia. Frequent episodes and low intelligence predicted a worse function in affective disorders.</p><p>Four men (4.5% of the sample) committed suicide. The risk of suicide was increased about 30 times. Almost a third of subjects attempted suicide. Females made more attempts. Suicide attempts were related to more depressive symptoms but less negative symptoms at first episode, to readmissions and to dependence on nicotine. </p><p>Subjects with schizophrenia spectrum psychoses were less satisfied with life than those with affective psychotic disorder. Subjective satisfaction in schizophrenia was strongly associated to depressive mood while in affective disorders it was associated to degree of employment.</p><p>Adolescents with psychosis in families rated high or borderline high in Expressed Emotion either during first episode or after discharge had an increased risk of relapse.</p>
