Influence of Abiotic Environmental Factors on Physiological Responses and Mixotrophy in Freshwater and Marine Chrysophytes

Global climate change represents one of the greatest threats to biodiversity. Phenomena such as rising surface water temperature, increased UV radiation, and ocean acidification have brought negative impacts to ecosystems and their inhabitants. Sensitive to various abiotic factors, microbial eukaryotic communities in aquatic systems are particularly being affected by these environmental changes. Specifically, warming temperature not only can directly affect plankton through limiting growth and inhibiting physiological processes, but can also indirectly impact these organisms by altering light and nutrient availability via loss of sea ice and changes in thermal stratification in various environments. Mixotrophic chrysophytes are an important lineage of protists that often dominate phytoplanktonic blooms in both freshwater and marine systems. Studies have shown mixotrophic organisms’ nutrient-acquiring strategies are influenced by abiotic environmental factors. Temperature in particular, is known to alter growth rate and bacterivory. In response to rising temperature, mixotrophs can either become more phototrophic or more heterotrophic, depending on species, resulting in changes of their role in aquatic food webs and potentially leading to shifts in overall community composition and structure. The objective of this research is to investigate the influence of different environmental factors on primary production and heterotrophic ingestion in marine and freshwater chrysophytes, providing an understanding on how climate change may alter physiological response and survival, with indicative changes in community structures and food webs. The influence of irradiance, nutrient concentrations, and temperature on mixotrophic responses of the Arctic marine chrysophyte Dinobryon faculiferum was investigated, where our results demonstrated an increase in heterotrophic ingestion in response to rising temperature. We also found bacterivory contributes a major proportion of D. faculiferum’s carbon budget in comparison to primary production, which is different from previous studies on Dinobryon species that appeared to be more reliant on phototrophy. Conversely, the freshwater chrysophyte Chrysolepidomonas dendrolepidota, exhibited the opposite temperature effect. The freshwater species was more reliant on primary production and ingested less as temperature increased. Such varying responses showcased diverse nutrient strategies on the mixotrophic spectrum, suggesting generalization of mixotrophic mode in predictive models should be approached with caution. Additional work was done to gain insight on the biogeography of C. dendrolepidota, of which little is known about its distribution. The presence of C. dendrolepidota was not detected through metadata analysis, nor was it detected across several waterbodies sampled in this study. Our results suggested possible rare distribution and endemism of C. dendrolepidota. / Biology

The eastern Mediterranean has been known as the most oligotrophic water body among the worlds oceans and as a result of limited nutrient inputs from terrestrial sources primary productivity and plankton succession are restricted by lack of nutrients especially by phosphorus and nitrogen. Within this domain, Mersin bay forms a so called hot spot (highly sensitive) area where a sharp contrast exists between the coastal area supplied by land-based nutrient sources and the nutrient limited open sea. Excess nutrient enrichment lead for eutrophication in the inner Mersin bay while altering the quality and quantity of flora from shore to offshore. Microorganisms are highly sensitive and profoundly affected by environmental disturbances and are widely used to assess the impact of environmental changes on ecosystem functioning. With this study, it is aimed to investigate responses of the smaller fractions of phytoplankton composed of heterotrophic bacteria, Synechococcus and flagellates to rapidly changing ambient biological, chemical and physical properties of shelf waters over an extended period between 2008 and 2010. Epiflourescence microscopy and image analysis setup were used to enumerate and measure size of cells for biomass estimates.

Zur Bedeutung von Endoparasiten bei Chamäleons (Sauria: Chamaeleonidae) aus Wildfängen und Nachzuchten

In der vorliegenden Arbeit wurden 212 Kotproben von Chamäleons auf Parasitenstadien und 75 Tierkörper pathologisch sowie bei einem nachgewiesenen Parasitenbefall histopathologisch untersucht. Ziel war es, anhand dieser Untersuchungen das Vorkommen und die Schadwirkungen von Endoparasiten unter Berücksichtigung der Herkunft, des Alters, des Geschlechts und der Chamäleonart zu beschreiben. Von 212 Kotproben wiesen 55,2% Endoparasitenstadien auf. Bei 54,7% der 64 sezierten und auswertbaren Tiere wurden Endoparasiten nachgewiesen. Der Anteil positiver Proben zeigt zwischen Nachzuchten (55,5%) und Wildfängen (54,1%) keinen wesentlichen Unterschied. In Wildfängen konnten häufiger Endoparasiten mit einem indirekten Lebenszyklus ermittelt werden, Nachzuchten beherbergten dagegen öfter Parasiten mit einem direkten Entwicklungszyklus. In den untersuchten Chamäleons konnten regelmäßig Kokzidien der Gattung Isospora und Oxyuriden nachgewiesen werden. Zestoden konnten in der koproskopischen Untersuchung gar nicht aufgefunden werden, wohingegen sie in der pathologischen Untersuchung sporadisch im Darm diagnostiziert werden konnten. Die Häufigkeit des koproskopischen Nachweises von Parasitenstadien bezogen auf die Gesamtzahl der untersuchten Chamäleons stellte sich wie folgt dar: in 30,4% wurden Protozoon gefunden, 21,7% der Tiere waren mit Kokzidien infiziert (davon 78,3% Isospora spp., 13,0% Choleoeimeria spp., 6,5% Eimeria spp., 2,2% Mischinfektion Isospora spp./Choleoeimeria spp.) und 8,5% mit Flagellaten oder Ziliaten. Bei 83,3% der Tiere mit gastrointestinalen Symptomen konnte ein Befall mit Kokzidien der Gattung Isospora nachgewiesen werden. In 38,7% der koproskopischen Untersuchungen konnten Nematoden (65,9% Oxyuriden, 19,5% Askariden/ Heterakiden, 1,4% Rhabdias sp., 2,8% Strongyloides sp., je 0,5 % Spirurida, Heterakiden/Filarien, Oxyuriden/Strongyloiden) und in 2,8% Trematoden (Digenea) aufgefunden werden. Anamnestisch konnten in 35,8% aller Tiere klinische Symptome beobachtet werden, wovon bei 88,2% der erkrankten Tiere ein Endoparasitenbefall nachzuweisen war. Insgesamt 64,1% der sezierten Chamäleons waren mit Endoparasiten befallen, wovon 68,3% Mono- und 31,7% Mischinfektionen beherbergten. In 31,3% der sezierten Chamäleons wurden Nematoden gefunden und der Befall wurde in 55,0% dieser Fälle als hochgradig eingestuft. Es wurden Befallsraten von 25,0% für Strongyloides sp., 23,4% für Askariden/ Heterakiden,15,0% für Filarien, 5,0% für Rhabdias sp., 9,4% für Zestoden, 10,9% für Digenea registriert. In 11,3% der Fälle lagen Mischinfektionen vor. Damit ist ein Endoparasitenbefall bei Chamäleons häufig und kann zu Erkrankungen führen. Die Exposition unterscheidet sich bei Wildfängen und Nachzuchten aufgrund der unterschiedlichen Umgebungsbedingungen. Auch klinisch unauffällige Tiere waren zu 27,8% mit Parasiten befallen, so dass eine klinische Symptomatik nicht zwingend aus einem Parasitenbefall resultiert. Insgesamt betrachtet verdient der Endoparasitenbefall von Chamäleons das Augenmerk von Tierärzten und Tierhaltern und sollte bei augenscheinlich hohem Infektionsdruck zu Gegenmaßnahmen, insbesondere auch einer verbesserten Hygiene, Anlaß geben. / In the present study 212 chameleon fecal samples were examined for parasite stages and 75 carcasses were examined histopathologically and pathologically in a proven case of a parasite infestation. The basis of this study was to describe the occurrence and harmful effects of internal parasites considering the origin, age and sex of the chameleons. Of the 212 fecal samples 55.2% showed stages of endoparasites. Parasites were detected at 54.7% of 64 evaluated and dissected animals. The proportion of positive samples shows no significant difference between offspring (55.5%) and wild specimens (54.1%). In wild specimens common internal parasites could be determined with an indirect life cycle, however offspring harbored more parasites with a direct life cycle. In the studied chameleons coccidia as the genus Isospora and Oxyurids were regularly detected. In the coprological study Cestodes could not be found, while in the pathological examination they could be diagnosed sporadically in the intestine. Based on the total number of investigated chameleons the frequency of detection of parasite stages are presented as follows: Protozoa were found in 30.4%, 21.7% of the animals were infected with coccidia (of which 78,3% Isospora spp, 13,0% Choleoeimeria spp., 6.5% Eimeria spp., 2.2% polyinfections between Isospora spp./ Choleoeimeria spp.) and 8.5% with flagellates or ciliates. At 83.3% of the animals with gastrointestinal symptoms coccidia of the genus Isospora were detected. In 38.7% of the fecal examination nematodes were determined (65.9% Oxyurids, 19.5% Ascarids/ Heterakis, 1.4% Rhabdias sp., 2.8% Strongyloides sp., 0.5% Spirurida, Heterakids/ Filariae, Oxyurids/ Strongyloides sp.) and Trematodes in 2.8% (Digenea) were found. Prevalences were registered: 25.0% for Strongyloides spp., 23.4% for Ascarids/ Heterakids, 15.0% for Filaria, 5.0% for Rhabdias sp., 9.4% for Cestodes, 10.9% for Digenea. In 11.3% of the cases mixed infections were reported. Thus, endoparasite infestation is common among chameleons and can lead to diseases. Exposure differs from wild-specimens and captive-bred due to the different environmental conditions. Also, 27.8% of clinically healthy animals were also infested with parasites, which means that clinical symptoms are not necessarily the result of a parasitic infestation. Overall, chameleon endoparasites deserve the attention of veterinarians and pet owners and should be treated promptly when there is a high likelihood of infection or hygiene is of concern.

Flagellates in the marine microbial food web : the ecology of a mixotrophic nanoflagellate, Ochromonas sp.

Nanoflagellates were found to be abundant in a coastal area of the northern Bothnian Sea. The maximum concentration of nanoflagellates, approximately 8000 cells ml-1, was observed in July, coinciding with a decrease in the abundance of cyanobacteria. Pigmented and non-pigmented nanoflagellates were approximately equally distributed throughout the year. Most of the identified genera are known as being phagotrophic, independent if autotrophic or not. A non-cyst-forming pigmented flagellate, Ochromonas sp., was isolated and nutritionally characterized. This chrysophycean flagellate was shown to be a mainly heterotrophic organism: Photosynthesis was too poor to support multiplication of the cells, whereas when feeding on bacteria, high growth rates were obtained. The biological function of the photosynthetic apparatus is suggested to be a survival mechanism during poor bacterial conditions. The flagellate grazed bacteria selectively, preferring cyanobacteria and large cells of heterotrophic bacteria, presumably depending on size-selective grazing. Despite higher growth rates of the bacteria in the sea during summer (July) than spring (May), heterotrophic bacteria in the sea was observed to be smaller in the summer. Nanoflagellates showed a maximum in July, and by selective grazing of large bacteria they might have caused the decrease in the average size of the bacteria and the decrease in the abundance of cyanobacteria. During the consumption of bacteria the flagellate was shown to remineralize nutrients at high rates and excrete dissolved free amino acids. Assuming the existence of a protozoan predator-prey chain of several trophic levels, it seems likely that a significant part of the nutrients fixed by primary producers is remineralized in the euphotic zone. Furthermore, data from this work indicate that flagellate activity may be a significant source of dissolved free amino acids, utilizable for the heterotrophic bacteria. / digitalisering@umu

Avaliação do consumo de bactérias pelo zooplâncton em um reservatório eutrófico raso (Reservatório do Monjolinho São Carlos SP)

This study aimed to evaluate the relative importance of protozooplanktonic (ciliates and heterotrophic nanoflagellates) and metazooplanktonic organisms (rotifers, cladocerans and copepods) as bacterial consumers throughout a year, which included a rainy and a dry season, in a eutrophic environment (Monjolinho reservoir-SP) by quantifying their ingestion rates. For this purpose, grazing experiments were carried out bimonthly during one year at the sub-surface of a central station of the reservoir using fluorescently labeled bacteria (FLB) dyed with DTAF. In each survey, besides the experiments, limnological (pH, dissolved oxygen, temperature, conductivity, water transparency and nutrients) and biological (chlorophyll-a and phaeophytin concentrations, bacterioplankton, protozooplankton and metazooplankton density) water parameters were also sampled. The heterotrophic nanoflagellates (HNF) showed higher population ingestion rates than the ciliates in dry and rainy seasons, and the HNF smaller than 5μm were the main consumers of bacteria. Among the metazooplanktonic organisms, the rotifers dominated the bacterial ingestion, and the organisms belonging to the genus Filinia and Keratella and to the families Testudinellidae and Brachionidae were the main bacterial consumers. Besides ingesting bacteria, however, protozoa and rotifers population also seemed to be controlled by cladocerans and copepods predation, which, in turn, seemed to be regulated by predation of larger organisms, such as small vertebrates. Thus, the predation control (top-down) occured both on bacterial communities and protozooplanktonic and metazooplanktnic communities and bacteria were regulated both by direct and indirect predation of zooplanktonic population. The resources control (bottom-up) semmed to be higher during the dry season, while in the rainy season bacterial communities were more controlled by predation. The greatest impact on bacterial communities was caused by HNF and ciliates during the rainy season and by HNF and rotifers during the dry one. / As bacterias, encontradas em ambientes terrestres e aquaticos, alem de possuirem importante papel na decomposicao da materia organica e na ciclagem de nutrientes, sao fonte de alimento para organismos de niveis troficos superiores, tais como protozoarios e organismos do metazooplancton. O presente trabalho avaliou a importancia relativa de organismos protozooplanctonicos e metazooplanctonicos (rotiferos, cladoceros e copepodes) como consumidores de bacterias em um ambiente eutrofico (Reservatorio do Monjolinho-SP) pela quantificacao das taxas de consumo durante o periodo de um ano, que englobou uma estacao seca e uma chuvosa. Para isso, na superficie de um ponto central no reservatorio, foram realizados experimentos de bacterivoria (grazing) bimestralmente durante um ano, utilizando bacterias marcadas fluorescentemente (FLB) com o corante DTAF. Nos dias dos experimentos foram tambem analisados parametros limnologicos (pH, oxigenio dissolvido, temperatura, condutividade, transparencia da agua, e nutrientes) e biologicos (concentracoes de clorofila-a e feofitina, densidade do bacterioplancton, protozooplancton e metazooplancton). Os nanoflagleados heterotroficos (NFH), quando comparados aos ciliados, apresentaram maiores taxas populacionais de ingestao de bacterias tanto no periodo seco quanto no chuvoso, sendo os NFH menores que 5 μm os maiores consumidores de bacterias. Dentre os organismos metazooplanctonicos, os rotiferos dominaram o consumo de bacterias, sendo os representantes dos generos Filinia e Keratella e das familias Testudinellidae e Brachionidae os maiores consumidores de bacteria. Os dados obtidos indicaram que, alem de predadores de bacterias, as populacoes de protozoarios e rotiferos pareceram ser reguladas pela predacao de cladoceros e copepodes, os quais, por sua vez, parecem sofrer pressao de predacao por organismos maiores, como provavelmente pequenos vertebrados. Assim, o controle por predacao (top-down) ocorreu tanto sobre as comunidades bacterianas quanto sobre as protozooplanctonicas e metazooplanctonicas, sendo as comunidades bacterianas reguladas tanto pela predacao direta quanto indireta das populacoes zooplanctonicas. O controle por disponibilidade de recursos (bottom-up) pareceu ter maior influencia no reservatorio no periodo seco, enquanto no periodo chuvoso, as comunidades bacterianas pareceram mais controladas por predacao, sendo que maiores impactos sobre as comunidades bacterianas foram causados pelos NFH e ciliados no periodo chuvoso, e pelos NFH e rotiferos no periodo seco.

This thesis deals with factors structuring phytoplankton flagellates. The aim of the thesis was to investigate the role of microhabitats in spatio-temporal differentiation of lentic communities of these flagellates, which has not been rigorously examined to date. The study was conducted during 2010-2013 in the shallow littoral sites of Horní rybník, Rybníčky u Podbořánek Nature Reserve. Phytoplankton flagellates showed cyclic seasonal dynamics, which was reflected by significantly different community structure and species richness during the season. In contrast, silica-scaled chrysophytes showed a non- cyclic seasonal dynamics. The impact of the season was strongly correlated with the impact of fluctuating environmental factors - pH, conductivity and temperature. Gradient of interrelated environmental factors between the northern and southern shore together with microhabitat - substrate type played the major role and accounted for 64.4 - 75.8 % variability in the data. Microhabitat type (plankton, metaphyton, epipelon) significantly affected species richness and community structure also in the course of the season. Mechanisms structuring the pelagic microbial food web : Importance of resource and predation

<p>Temporal and spatial variations of pelagic microorganisms in the northern Baltic Sea were studied, as well as factors influencing their abundance and growth rates. Three main questions were asked 1) How does increased productivity influence the structure of the microbial food web? 2) Does predation limitation vary between trophic levels? 3) What is the relative importance of resource and predation limitation at different trophic levels?</p><p>A field study in the northern Baltic Sea showed that dominating protozoa, flagellates and ciliates, increased with increasing primary productivity from north to south. Furthermore, relatively small protozoan cells dominated in the low productive north, while larger cells became more dominant in the south. The relationship between plankton size structure and productivity was further studied in an experimental system. In agreement with present theories regarding nutrient status of pelagic food webs, increased productivity caused a lengthening of the food chain as well as a change in plankton size structure. While microplankton dominated in nutrient rich treatments pico- and nanoplankton dominated during nutrient poor treament. The flagellate community was dominated by a potentially mixotroph, <i>Chrysochromulina</i> sp., at low nutrient concentrations. To our knowledge this is the first experimental study showing that <i>Chrysochromulina</i> sp. in resemblance with other mixotrophs is favoured by nutrient poor conditions compared to strict autotrophs and heterotrophs.</p><p>During a stratified summer period autotrophic microorganisms in the northern Baltic Sea did not respond to removal of potential predators, indicating that they were primarily limited by inorganic nutrients. An exception was small eucaryotic picoplankton that showed a large response to predator removal. Among the heterotrophic microorganisms direct effect of predation seemed to increase from ciliates, heterotrophic bacteria, small heterotrophic flagellates, medium flagellates to large flagellates. No quick indirect effect was observed, but after four days trophic cascades were detected.</p><p>The relative importance of resource and predation limitation was studied among heterotrophic bacteria, flagellates and ciliates in the northern Baltic Sea. For all these groups, resource limitation seemed to prevail during the summer period. The results also indicated that the relative importance of predation increased with the productivity of the system. To our knowledge there are no earlier measurements on the relative importance of resource and predation limitation for micoorganisms in the pelagic environment.</p>
