Mechanical properties of PVDF/MWCNT fibers prepared by flat/cylindrical near-field electrospinning

Ke, Chien-An 04 September 2012 (has links)
This study presents near-field electrospinning (NFES) on flat and hollow cylindrical process to fabricate permanent piezoelectricity of polyvinylidene fluoride (PVDF)/ multi-walled carbon nanotube (MWCNT) piezoelectric nanofibers. Then the mechanical properties of fibers were measured. PVDF is a potential piezoelectric polymer material combining desirable mechanical, thermal, electrical properties with excellent chemical resistance. The existing researches mostly focused on piezoelectric thin film process. However, the research of characteristic about piezoelectric fiber is little. The methods of measurement of the mechanical properties (Young¡¦s modulus, hardness, and tensile strength¡Belongation) of the electrospun PVDF/MWCN composite nanofiber were carried out by using nano-indention test (MTS Nanoindenter Windows XP System) and tensile test (Microforce Testing System). By setting electric field (1¡Ñ107 V/m), rotating velocity (900 rpm) of the hollow cylindrical glass tube on a motion X-Y stage (2 mm/sec) and PVDF solution concentration (16 wt%), and MWCNT (0.03 wt%), in-situ electric poling, mechanical stretching and morphology of PVDF nanofiber were demonstrated. After the experiments of nano-indention test and tensile strength test, it is suggested that the good mechanical properties in NFES on cylindrical process. The results show that the mechanical properties of composite nanofiber are better than the conventional NFES process. The Young¡¦s modulus of 16% PVDF fiber prepared by cylindrical process is 0.89 GPa and hardness is 26.5 MPa. The mechanical properties were increased 56.2% and 49.4% after adding 0.03% of MWCNT, corresponding to 1.39 GPa and 39.6 MPa. The tensile strength was increased 32.7% and elongation at breaking point was increased 35% after adding 0.03% MWCNT.

The high pressure synthesis, crystal growth and physical properties of transition metal perovskites

Marshall, Luke Gordon 02 March 2015 (has links)
The perovskite structure has an incredible versatility that results in myriad compounds with varied and eccentric behaviors. Perovskite oxides have been extensively studied and used for over 60 years. In order to expand on our already thorough knowledge of these compounds, it is necessary to use modern and creative experimental techniques. High-pressure synthesis and high oxygen-gas pressure annealing techniques are used to synthesize oxygen stoichiometric RNiO₃ (R = lanthanide). The particularly rich phase diagram of this compound allows for the study of the crossover from localized to itinerant electronic behavior and from an enhanced Pauli to a Curie-Weiss law paramagnetism. Single crystals of RFeO₃ are grown in order to analyze the spin canting in these antiferromagnetic samples. The size of the rare earth-cation is used to tune the magnitude of octahedral-tilt distortions. This tuning allows distinguishing between the two possible drivers for spin canting and weak ferromagnetism in these compounds, the octahedral-tilt-dependent single-ion anisotropy and the octahedral-tilt-independent Dzyaloshinskii-Moriya interaction. Although it is a fluoride compound, KCuF₃ has been used as an analogue to transition-metal oxide perovskites such as LaMnO₃ because of the similarity of their orbital ordering. Through the use of high-temperature neutron diffraction, it is shown that the orbital ordering and Jahn-Teller distortion in this compound are not lifted at the predicted temperature. Another mechanism for orbital ordering is identified. La₂[subscript-x] Sr [subscript x] CuO₄ has long been of interest as the progenitor system of the highTc superconductors. Despite having an exceedingly well-studied phase diagram in the over-doped region of its superconducting dome, little is known about this system in the region x > 0.3 because of the difficulty of synthesizing fully oxygen-stoichiometric samples. With high-oxygen-gas-pressure annealing and high-pressure synthesis, the completion of the phase diagram up to x = 1.0 is attempted. Finally, like many iridates, post-Perovskite CaIrO₃ exhibits a very strong spinorbit coupling of its 5d electrons. Because its magnetism is very weak, traditional methods to measure the magnitude of its orbital moment and spin-orbit coupling, such as neutron powder diffraction, are not viable. In order to address this issue, direct measurement of the orbital moments was conducted by using x-ray absorption spectroscopy and x-ray magnetic circular dichroism techniques. / text

Synthesis and evaluation of an [18F]-labelled antisense oligonucleotide as an imaging probe to measure cellular response to radiation therapy

Koslowsky, Ingrid L 11 1900 (has links)
Antisense oligodeoxynucleotides (asODNs) show strong binding and high selectivity and can be constructed to recognize specific cellular targets such as gene regulated mRNA. Radiolabelled asODNs have the potential to image gene expression through mRNA targeting and could be a valuable tool in the early assessment of outcome to cancer treatment. We have explored the potential of in vivo imaging of p21 gene expression, using fluorine-18 labelled asODNs ([18F]asODNs) and in vitro techniques, recognizing the relationship between the expression of this gene and resistance of cancer cells to radiation therapy. Radiolabelling of fully phosphorothioated, 20-mer ODNs was performed using the [18F]-labelled prosthetic group, 4-N-[18F]fluorobenzyl-2-bromoacetamide ([18F]FBBA). [18F]FBBA was first synthesized in an automated synthesis unit, resulting in a modest radiochemical yield. Methods to improve the yield were investigated using a metal catalyst-assisted borohydride exchange resin. Alkylation of [18F]FBBA to ODN resulted in radiochemical yields of 40%. Cellular uptake and retention studies were performed in human carcinoma cells expressing p21+/+ (HCT116) and the p21 knock-out cell line, 80S4, using both [18F]-labelled antisense and random sequence ODNs. Nonradioactive FBBA-labelled ODNs were used to evaluate the antisense effectiveness and distribution of the FBBA-modified ODNs. In vitro studies demonstrated that FBBA did not interfere with the antisense effect of ODNs against p21 mRNA; however, the probes required a transfection agent to observe an antisense effect. Cell fractionation studies with [18F]ODNs revealed increasing accumulation of liposome-transfected [18F]asODN in the cytoplasm of HCT116 cells over time. A biocompatible spermine-grafted block copolymer (SP) was subsequently evaluated as a potential vector to improve the delivery of [18F]asODN into cells. SP was shown to direct [F]-labelled ODNs to the cytoplasm, whereas naked [F]ODNs remained sequestered in vesicles, and liposome-transfected [F]ODNs localized mostly in the nucleus. Selective uptake and retention of [18F]asODN was observed in p21+/+ cells only when the probe was transfected with SP. Based on these studies, it can be concluded that [18F]asODNs have the potential to image gene expression, however the focus may need to be directed to find an appropriate vector which can rapidly deliver [18F]-labelled asODNs to the target tissue in vivo.

No. of bitstreams: 1 Arquivototal.pdf: 1506813 bytes, checksum: 01a6e65a669f2f0616d92f5db1de2aaf (MD5) Previous issue date: 2011-12-15 / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / The objective of this study was to evaluate the effect of CPP-ACP in dental erosion control. For this purpose two studies. (1) The objective the first study was to evaluate in vitro the effect of pastes containing CPP-ACP and fluoride dentifrice in controlling dental erosion with or without abrasion. Eighty enamel blocks were divided into 10 groups (n = 8) according to the type of product used: MI Paste ® (G1/G6), Mi Paste Plus ® (G2/G7), Sensodyne Pro-Enamel ® (G3 / G8), Crest ® (G4/G9) and artificial saliva (G5/G10) and the experimental procedure: erosion with dripping with solution of the product (G1-G5) and erosion abrasion with dripping of the solution of the product (G6- G10). The specimens were subjected to simulated erosion using a soft drink as an agent of erosion, for three cycles of erosion of five minutes under constant agitation, for ten days. The surface roughness (Ra) was used to quantify the changes of the enamel surface eroded. For qualitative assessment of enamel was used SEM. There was a statistically significant difference (p <0.05) between groups G1-G4 in the control group G5 after the erosive challenge. The same occurred between groups G6-G9 and G10 when the erosion control was followed by abrasion. However, the products did not differ when they were compared in two conditions of erosion. In the evaluations of the SEM observed change in the surface of all experimental groups compared to healthy surface in each specimen surface. It can be concluded that the CPP-ACP and CPP-ACPF, showed ability to reduce the deleterious effects of erosion similar to fluoride toothpastes. (2) The objective of the second study was to evaluate in situ the effect of CPP-ACP incorporated into a chewing gum in reducing the increase of surface roughness of enamel after being subjected to erosive challenge with or without abrasion. This study presented a blind crossover design in which eight volunteers used intraoral palatal appliances containing six specimens. Each volunteer was subjected to three experimental periods of seven days each, with seven-day wash out between them. In each period were observed three conditions of erosion: erosion only, immediate erosion associated with brushing and erosion associated with brushing after an hour. The surface roughness (Ra, Rz and Rt) was used to analyze the surface changes. In all specimens there was an increase in roughness of enamel after the erosive challenges. Statistically significant difference was found between the experimental periods "no gum" and "gum with CPP-ACP" when erosion was followed by immediate brushing: Rz (p = 0.0148) and Rt (p = 0.036) and one hour after brushing Ra (p = 0.0312), Rz (p = 0.0264) and RT (p = 0.498). Among the three conditions of erosion in each period, the roughness values always followed the same pattern of presentation: erosion <erosion with abrasion after an hour <erosion with abrasion immediately, but there was no statistically significant difference. The data suggest that the use of chewing gum with CPP-ACP after ingestion of erosive beverages, contributes to reduction of dental enamel erosion, especially when the challenge is followed by the erosive abrasion. Thus, it is concluded that the CPP-ACP incorporated into folders or chewing gum was able to reduce the deleterious effects of dental erosion.

January 2004 (has links)
Abstract: The aim of this study was to evaluate the effect of pH of the demineralizing solution in a cycling model on surface hardness, fluoride release and surface aspects of restorative materials (Ketac-Fil Plus, Vitremer, Fuji II LC, Freedom and Fluorofil). Thirty-four specimens of each material were made and surface hardness was measured. Then, the specimens were randomized in five groups (n=30), according to pH (4.3; 4.6; 5.0; 5.5; and 6.2) of the demineralizing solution and submitted to a cycling model during 15 days. The specimens remained in the demineralizing solution for 6 hours and in the remineralizing solution for 18 hours. After that, surface hardness was measured again and surface aspects were observed (n=24). Fluoride concentration was determined daily in all solutions (De+Re). Data from SMH (before and after pH-cycling) and %SMHC were analyzed by analysis of variance (p<0.05); Kruskal-Wallis test was carried out for the results of fluoride release. For glass ionomer cements, the variation of pH led to a direct relationship when hardness was compared and to an inverse relationship with regards to fluoride release. For polyacid-modified resin composites, an inverse relationship was found regarding fluoride release; no correlation was observed with regards to hardness. Surface aspect was influenced by pH variation: an acidic pH led to a greater alteration, except for polyacid-modified resin composites. It was possible to conclude that pH of the demineralizing solution did influence fluoride release from the tested materials. The pH variation altered hardness and surface aspects of glass ionomer cements, but it did not influence polyacid-modified resin composites in those analysis. / Orientador: Denise Pedrini / Coorientador: Alberto Carlos Botazzo Delbem / Mestre

Quinze voluntários participaram do estudo. / The protective effect of fluoride against erosion has been related mainly to the formation of calcium fluoride deposits and the adsorption of fluoride ions on the dental surface, so it was hypothesized that the incorporation to the Carbopol 980 polymer could potentiate the protective effect of fluoride solutions in the control of the enamel erosive wear with the formation of a protective film. The proposal was divided in two stages. The first stage consisted of an in vitro study in which the association of Carbopol polymer to sodium fluoride (NaF) and sodium fluoride with tin chloride (NaF + Sn) was tested in the presence of acquired pellicle. The ph stat test was used to test the protective potential of these components isolated or associated, in relation to the dissolution of hydroxyapatite, thus predicting the protective potential of the solutions tested. The remineralizing and protective effect against initial erosion measured by microhardness was investigated. Cylindrical polished bovine enamel specimens (3 mm diameter) was prepared and demineralized with 0.3% citric acid (pH 2.6) for 2 minutes (n=15). The specimens were immersed in human saliva for 2 hours to allow acquired pellicle formation and then, treatment with experimental solutions for 2 minutes were performed [NaF (500 ppm F), NaF + Carbopol (0,1%), NaF + Sn (500 ppm F + 800 ppm Sn), NaF + Sn + Carbopol], deionized water (negative control) and comercial mouthrinse (Elmex – positive control). A new immersion in human saliva (2 h) for remineralization and then new acid challenge were performed. The Knoop microhardness was measured at different times: baseline, after the first acid challenge, after treatments (remineralization potential), and after second acid challenge (protective potential). Additionally, a new immersion in test solutions was conducted for the measurement of alkali-soluble fluoride on enamel surface. ANOVA and Tukey tests were applied (5%). It was observed that the dissolution of hydroxyapatite was lower in the groups treated with NaF + Sn + Carbopol compared to the other groups. This result was also observed with the remineralizing and protective potentials, measured by microhardness. The measurement of alkali-soluble fluoride on enamel surface was also higher to the NaF + Sn + Carbopol association group compared to the other groups. The second stage was an in situ study carried out with the experimental solutions that present the greatest protective potential in the previous in vitro study. For this, polished enamel specimens were divided into three groups (n = 60): NaF + Sn (positive control), NaF + Sn + Carbopol, and deionized water (negative control). An in situ model with three stages was tested. Fifteen volunteers participated in the study. In each phase, volunteers used a palatal appliance containing 4 specimens each, in which two were submitted to an erosion and remineralization cycle for 5 days. This cycle consisted of using the appliance for 2 hours for pellicle formation, followed by extra oral immersion in 1% citric acid (pH 2.3 - 5 minutes - 4x/day) with intervals of 1 hour of in situ saliva exposure between challenges and treatment with the solutions 2x/day for 1 minute. The other two specimens were subjected to erosion/abrasion/remineralization cycle, in which abrasion was performed 2x/day - 15 seconds, with active electric toothbrush, before the daily treatment with the solutions, and erosion/remineralization as previously described. At the end of the experiment, enamel surface loss (µm) was evaluated by profilometry and the data were analyzed by two-way ANOVA and Tukey tests (5%). There were differences between the challenges (erosion and erosion + abrasion), as well as treatments with fluoride solutions were able to protect the enamel against erosive wear, significantly different from the negative control. It can be concluded that for both the in vitro and the in situ study, the association of NaF to Sn, and these to Carbopol, presented promising results reducing the development of erosive wear. / FAPESP 2015/19585-4

Apesar do aumento no fluxo... / In previous research we found that spontaneously hypertensive rats (SHR) have lower stimulated salivary flow than normotensive Wistar, which could lead to a higher rate of caries in these animals. The aim of this study was to evaluate the effect of chronic treatment with sodium fluoride (NaF) in the biochemical parameters of saliva and plasma, bone and mineralization in incisor teeth of SHR. Were used SHR and Wistar rats at 3 months of life. The treatment was made with NaF solution (20 ppm) for 30 days in drinking water. The measurement of systolic blood pressure (SBP) was performed by the indirect method of tail plethysmography. To collect saliva, salivary flow was stimulated with pilocarpine nitrate and the animals were placed on an inclined board and saliva collected for 15 minutes. Salivary and plasma concentrations of fluoride were determined with specific electrodes and calcium using a specific commercial kit. The determination of total protein concentration was performed by the method of Lowry and salivary amylase activity using a commercial kit. The analysis of fluoride on the surface of the femur was made by the direct method and analysis in the femur by the method of Taves. The microhardness of incisors was determined using Knoop microhardness with indenter type. The results were evaluated and differences were considered when p <0.05. The SBP and the weight of the animals were not affected by treatment. The reduced salivary flow in SHR were enlarged in treated SHR. The fluoride concentration in saliva and plasma increased with treatment in Wistar rats, however, in SHR the increase was only observed in plasma. Despite the increase in salivary flow that occurs after treatment with NaF, the concentration of fluoride in saliva is not altered in SHR. The calcium concentration decreased in saliva and plasma in SHR group after treatment, what ... (Complete abstract click electronic access below)
