La catégorisation des corps : étude sur l'humain avant la naissance et après la mort / The categorization of bodies : a study on human beings before birth and after deathCarayon, Lisa 12 December 2016 (has links)
Entre les personnes et les choses, les embryons et les cadavres sont souvent représentés comme des objets juridiques non-identifiés. Cependant, une analyse détaillée du travail de la jurisprudence et du législateur dans l’élaboration de leur de leur régime montre que la qualification des corps n’est pas tant imprécise qu’éludée. L’extrême sensibilité politique de la question conduit à la construction d’un droit fractionné, proposant des solutions ponctuelles à des questions précises, sans véritable souci de cohérence catégorielle.Pourtant, présentant la qualification comme une véritable question de connaissance du droit, une partie de la doctrine semble parfois éluder la dimension politique de cette construction. Recourant , pour identifier la nature des corps, à des disciplines extérieures au droit (biologie, psychanalyse, philosophie…) sans toujours mettre en lumière les difficultés épistémologiques liées à cette démarche, les auteurs renouvellent en la matière une approche jusnaturaliste prescriptive qui ne dit pas toujours son nom.Or, l’étude de l’évolution des qualifications et des régimes appliqués aux embryons et aux cadavres montre que le véritable problème est surtout que les catégories établies entre les corps par le droit sont avant tout des classements. Tous les corps ne bénéficient pas de la même protection du droit et le statut des embryons et des cadavres reflète bien souvent les hiérarchies existant entre les personnes : distinctions sociales, religieuses, genrées, racialisées etc.. Se pose alors la question des remèdes possibles à ces multiples catégorisations des corps : assouplissements ponctuels ou transformation radicale du droit ? / Embryos and corpses are often presented as unidentified legal objects, in between people and things. However, a detailed analysis of French case law and statute law shows that the classification of bodies is not unclear, but eluded. The extreme political sensitivity of the matter leads to the construction of a subdivided law, offering one-time solutions to specific questions, without due regard to categorical coherence. Most of the doctrine seems to elude the political dimension of this construction and presents classification as an actual question of legal knowledge. Resorting to extraneous disciplines (biology, psychoanalysis, philosophy…) in order to identify the nature of the bodies, authors seem to repeat a prescriptive approach based on natural law theory even if they often deny it.In contrast, a historical approach of classification and legal systems applied to embryos and corpses reveals that the main issue is that the bodies’ categories induced by law are above all hierarchization. All bodies do not benefit from the same legal protection and the status of embryos and corpses very often reflects existing hierarchies between individuals – distinctions based on social level, religion, gender, ethnicity… Hence a question arises as to possible solutions to this manifold categorization of bodies : should the law be relaxed in isolated cases or should a more radical transformation of the law be considered ?

Background: During the recent years, magnetic resonance imaging (MRI) has evolved to become one of the most commend used diagnostic methods. Although MRI is considered safe and a good diagnostic method of foetuses and pregnant women, the research data is too small to claim that it is completely risk-free. Purpose: The purpose of this literature study was to identify possible risks of foetal defects in connection with MRI examination of pregnant women. Method: This essay is a quantitative systematic literature study where 13 articles were included after being searched in three different databases. Result: No significant differences in biological effects or hearing loss in foetuses exposed to MRI were detected. Heat increase could occur in connection with MRI examination and was influenced by the examination time and the field strength. However, the temperature generally stayed below the approved limit values. The using of gadolinium contrast agents could increase the risk of some foetal disease states as well as miscarriage and stillbirth. Conclusion: MRI could be a good alternative for anatomical imaging of pregnant women, although it should be used with great caution because of the limited research base. Thus, more research is needed in this area to determine the impact of MRI and gadolinium contrast agent on foetal development. / Bakgrund: Under de senaste åren har magnetresonanstomografi (MR) utvecklats till att bli en av de mest använda diagnostiska metoderna. Trots att MR anses vara säkert samt en bra diagnostisk metod av foster och gravida, så är forskningsunderlaget för litet för att påstå att det är helt riskfritt. Syfte: Syftet med denna litteraturstudie var att kartlägga vilka eventuella risker som finns för fosterskador i samband med MR-undersökning av gravida kvinnor. Metod: Detta examensarbete är en kvantitativ systematisk litteraturstudie där 13 artiklar inkluderades efter att ha sökts fram i tre olika databaser. Resultat: Inga signifikanta skillnader gällande biologiska effekter eller hörselnedsättning hos foster som blivit exponerade för MR kunde påvisas. Värmeökning kunde ske i samband med MR-undersökning och påverkades av undersökningstiden samt fältstyrkan. Temperaturen höll sig dock i allmänhet under godkända gränsvärden. Vid användande av gadoliniumkontrastmedel kunde det ge ökad risk för somliga sjukdomstillstånd hos fostret samt missfall och dödföddhet. Konklusion: MR skulle kunna vara ett bra alternativ för anatomisk avbildning av gravida kvinnor, men det bör användas med stor varsamhet på grund av det begränsade forskningsunderlaget. Det behöver således forskas mer inom området för att kunna fastställa MR och gadoliniumkontrastmedels påverkan på fostrets utveckling.

Background: The slaughter of pregnant cattle and the fate of the foetuses are relatively new subjects in the field of animal welfare. The Scientific Committee on Veterinary Measures relating to Public Health (SCVPH), however, does not believe this topic to be a critical issue because of the hitherto supposed rare occurrence of this practice. Some previous studies though, contradict this assessment, emphasising its relevance to animal welfare. With regard to the heterogeneous study design of previous investigations, the objective of this study is to evaluate the current situation concerning the slaughter of pregnant cattle in different German abattoirs. Additionally, the prevalence was assessed semi-quantitatively on the basis of a cross-sectional, voluntary and anonymous survey that was conducted amongst senior veterinary students of the University of Leipzig from 2010 until 2013. Results: Of 255 evaluable questionnaires, 157 (63.6 %) mention the slaughter of pregnant cattle, corresponding to 76.9 %
of all visited abattoirs. Slaughter of pregnant cattle is reported often (>10 % of females) in 6 (3.8 %), frequently (1–10 % of females) in 56 (35.7 %), and rarely (<1 % of females) in 95 (60.5 %) of all cases (n = 157) respectively. About 50 % of these animals were reported to be in the second or third stage of gestation. 15 (10.6 %) of 142 questionnaires providing information about the foetus, state that the foetus showed visible vital signs after the death of the mother, but in one case the foetus was euthanized subsequently. Conclusions: The results show that the slaughter of pregnant cattle is a common and widespread practice in German abattoirs. The SCVPH’s assumption that pregnant cattle are only slaughtered in rare exceptional cases can no longer be maintained. The high proportion of foetuses in the second and third gestational stage must also be considered. In this context the implementation of suitable studies and detailed analysis of the current situation is indispensable to ensure the high standards in animal welfare in Germany and Europe.

Le syndrome CHARGE : étude clinique et moléculaire / Clinical and molecular aspects of CHARGE syndromeLegendre, Marine 14 December 2016 (has links)
Le syndrome CHARGE est une association malformative rare due à une mutation du gène CHD7 dans 60 à 90% des cas. L'objectif de ce travail était d'en décrire les éléments cliniques et moléculaires afin d'optimiser la prise en charge de patients atteints d'un handicap multisensoriel lourd.Le diagnostic anténatal en est difficile et l'étude de 40 fœtus a permis d'affiner la description du phénotype, de décrire de nouveaux éléments cliniques et finalement de proposer un ajustement des critères diagnostiques chez le fœtus.L'étude endocrinienne de 42 patients confirme la présence d'un hypogonadisme hypogonadotrope dans 97% des cas. Souvent méconnu et non traité il peut avoir des conséquences délétères sur la qualité de vie. Nous proposons qu'il soit reconnu comme critère majeur du syndrome.L'étude clinique d'une cohorte française de 119 patients a montré que la surdité et l'atteinte des canaux semi circulaires sont les éléments les plus fréquents du syndrome (suivis de l'atteinte hypophysaire, de l'arhinencéphalie et des anomalies de l'oreille externe), et les seuls significativement associés à la présence d'une mutation dans CHD7. Une étude approfondie des données issues de cette étude est en cours.Sur le plan moléculaire, alors que la plupart des mutations identifiées sont des mutations tronquantes privées apparues de novo, nous avons identifié 4 variants au niveau de l'intron 25, récurrents pour certains, dont l'interprétation était délicate. Leur étude in silico puis par une technique de minigène a permis de mettre en évidence une configuration particulière des séquences impliquées dans l'épissage de l'exon 26 (point de branchement distant) et de démontrer leur pathogénicité. / CHARGE syndrome is a rare disorder of multiple congenital anomalies ascribed to a CHD7 gene mutation in 60% to 90% of cases. The aim of this study was to improve the knowledge regarding molecular and clinical aspects of the syndrome in order to optimize the management of these patients with severe disability. Antenatal diagnosis remains challenging in many instances and a detailed clinicopathological survey in a series of 40 fetuses allowed us to refine the clinical description of CHARGE syndrome in fetuses, describe some novel features and set up diagnostic criteria. An endocrinologic study of 42 patients showed a hypogonadotrophic hypogonadism in 97% of cases. For this reason, it should be considered as a major symptom of the syndrome. An early screening should lead to a hormonal replacement therapy which dramatically impacts the condition.A study of a French cohort of 119 patients found that deafness and semi-circular canals hypoplasia were the most frequent symptoms (followed by hypogonadotrophic hypogonadism, arhinencephaly and external ears anomalies) and the only features statistically associated with a mutation in the CHD7 gene. A detailed study of the data is still going on.The syndrome is mainly due to de novo and private truncating mutations of the CHD7 gene but we report an intriguing hot spot of intronic mutations located in IVS25. Combining computational in silico analysis and ex vivo minigene assays, we explained this mutation hot spot by a particular genomic context, including a distant branch point, and confirmed the pathogenicity of these mutations.

But de l’étudeL’atrésie intestinale est une anomalie congénitale définie par une perte de la continuité digestive. Malgré une restauration chirurgicale précoce de cette continuité, surviennent durant les premiers mois de vie des troubles de la motilité digestive et des surinfections bactériennes chez un tiers des patients. Ces troubles fonctionnels étaient attribués jusque là principalement à des altérations du système nerveux entérique. Le but de cette étude était de confirmer cette hypothèse mais également d’élargir le champ des explorations aux autres composants du tube digestif.Matériel et méthodesLe modèle animal de l’atrésie chez le rat initialement décrit dans notre équipe a été utilisé pour caractériser les anomalies d’expression génique par transcriptomique. L’étude portait également sur la maturation digestive chez des fœtus de rat contrôle entre un stade de développement embryonnaire E15 et E21. Des modifications en amont et en aval de l’obstruction ont été étudiées en prélevant deux segments successifs de 1 cm par cette approche globale transcriptomique puis précisées par RT-PCRq et confirmées par des techniques immunohistochimiques et de microscopie électronique. RésultatsChez les fœtus témoins, l’expression génique montre une décroissance physiologique pour le SNE et une augmentation pour les systèmes neuroendocrine et épithélial de E15 à E21. Concernant les fœtus avec atrésie, les modifications concernent quasi exclusivement le segment d’amont avec une augmentation du calibre intestinal, de l’épaisseur musculaire et une accélération globale de la maturation. Une redistribution des sous types neuronaux est constatée dans le segment d’amont ainsi qu’une augmentation de l’expression du système neuro endocrine. Pour ces deux systèmes, le segment d’aval est peu modifié. Des modifications importantes du système épithélial sont observées en amont comme en aval avec pour conséquence probable une altération de la barrière intestinale et du système anti infectieux.ConclusionCes résultats montrent que les changements prédominent dans le segment en amont de l’atrésie alors que le segment d’aval était parfois considéré comme le plus pathologique. De plus, il a été retrouvé des changements inattendus du système neuroendocrine et épithélial qui sous tendent une implication non exclusive du SNE. D’autres recherches sont nécessaires pour confirmer ces données et les exploiter dans une démarche thérapeutique. / Aim of the studyIntestinal atresia is a rare congenital affection with postoperative motility disorders, leading sometimes to death. Previous related studies mainly focused on enteric nervous system (ENS) alterations as it was identified to cause abnormal peristalsis. The aim of the study was to expertise the underlying pathological conditions of intestinal atresia using a global approach, before focusing on ENS and neuroendocrine cells in order to precise the presumptive involvement of the different layers of the intestinal wall.MethodsPreliminary transcriptomic approach was elected to screen global gene expression involved in intestinal development and atresia-linked disorders in the rat model previously described by our team. Rat embryos were assigned to atretic group and controls embryos at different stages of development ED15, ED17, ED19 and ED21. Two successive intestine samples of 1 cm were harvested in the proximal segment and in the distal one. The pattern of gene expression was further assessed by immunohistochemistry, electron microscopy and RT-qPCR. Main resultsA physiological decrease in gene expression for enteric nervous system markers and an increase for neuroendocrine and epithelial system was observed on controls from stages ED15 to ED21. Regarding affected embryos, structural modifications concerned the proximal segment with increased muscular layer and a significant disruption including global accelerated maturation was observed in the proximal segment with increased gene expression of neuroendocrine system. Distal segment was comparable to controls for the two systems. Important modifications were noted concerning the epithelial system with consequent abnormalities of the gut barrier and anti infectious functions.ConclusionsFetal intestinal obstruction results in a disrupted gut development predominant in the proximal segment. The distal segment and the ENS were poorly concerned by theses changes. Neuroendocrine and epithelial cells underwent significant unexpected changes, supporting the evidence that ENS do not play an exclusive role in the pathways of intestinal motility disorders.

A oviposição é um estágio vulnerável do ciclo de vida de carrapatos. Rhipicephalus microplus, como todos Ixodidae e Argasidae, possui uma glândula especializada, o órgão de Gené, que produz uma cera que é depositada na superfície do ovo durante a oviposição. Além de restringir a perda excessiva de água, a cera atua como uma barreira contra o ataque de organismos invasores. Em R.microplus, como em outros carrapatos, há poucos estudos demonstrando atividade antimicrobiana em ovos. Ainda mais, não há na literatura relato de atividade antibiofilme em ovos de carrapatos e nem mesmo em qualquer outro artrópode. O objetivo do presente trabalho foi avaliar a hipótese da existência de mecanismos de defesa em ovos de R. microplus contra biofilmes bacterianos. O extrato água/metanol da cera dos ovos apresentou atividade contra o biofilme de Pseudomonas aeruginosa sem afetar a sua viabilidade. Esse extrato também demonstrou efeito antibiótico contra Staphylococcus epidermidis. Nós identificamos a molécula com ambas atividades (antibiofilme e antibiótica) como N-(3-sulfooxy-25-cholest-5-en-26-oyl)-L-isoleucina (boophiline). Na busca por possíveis mecanismos responsáveis pelo efeito antibiofilme de boophiline contra P. aeruginosa, 14 genes foram analisados por qRT-PCR. Boophiline inibe a expressão de fliC (flagelo) e cdrA (componente estrutural da matriz), cujos produtos são necessários para a formação do biofilme de P. aeruginosa. Monosfosfato de guanosina dimérico cíclico (c-di-GMP) é um importante segundo mensageiro característico de bactérias Gram-negativas. Altos níveis intracelulares de c-di-GMP promovem o estilo de vida séssil enquanto baixos níveis induzem o comportamento móvel. De acordo com essa afirmação, nós encontramos que boophiline aumenta a motilidade swarming de P. aeruginosa. Desta forma, nos questionamos se o mecanismo de ação de boophiline estaria envolvido com c-di-GMP já que o sistema quorum sensing não foi afetado pela molécula. Interessantemente, quando os níveis de c-di-GMP foram aumentados pela superexpressão de uma diguanilato ciclase, boophiline não inibiu efetivamente a formação de biofilme. Uma explicação para esse resultado é que boophiline interfere em uma via específica regulada por c-di-GMP, o que explicaria não termos obtido um decréscimo no nível total deste segundo mensageiro. Contrariamente, boophiline foi bactericida contra S. epidermidis. Mudanças morfológicas significativas foram observadas em células tratadas com a molécula, as quais foram severamente danificadas. Boophiline levou a formação anormal de septo, rompimento da membrana bacteriana e extravasamento do material intracelular. Adicionalmente avaliamos o potencial antibiofilme e anti-protozoário de filtrados de cultura obtidos de bactérias isoladas de tecidos de R. microplus. Quatorze filtrados de cultura bacteriano apresentaram notável atividade contra o biofilme de P. aeruginosa e S. epidermidis e foram citotóxicos contra Tritrichomonas foetus. Nosso trabalho é pioneiro em demonstrar a existência de proteção contra biofilmes em ovos de carrapatos bem como de bactérias associadas a carrapatos como produtoras de moléculas bioativas. Além disso, nós demonstramos que boophiline é uma nova molécula antibiofilme, sendo a primeira vez relatado na literatura que um composto age inibindo cdrA. Os dados obtidos em nosso estudo poderiam estimular novas abordagens em áreas como fisiologia e controle de artrópodes, genética e fisiologia de microrganismos e controle de biofilmes. / The oviposition is a vulnerable stage of the tick life cycle. Rhipicephalus microplus, as all Ixodidae and Argasidae, has a specialized gland, the Gene’s organ, which produce a wax that is smeared on egg surface during oviposition. In addition to restricting excessive water loss, wax acts as a barrier to attack by invading organisms. In R. microplus, as in other ticks, there are few studies showing antimicrobial activity in eggs. Moreover, there is no report of antibiofilm activity in tick eggs nor in any other arthropod. The objective of our study was to evaluate the hypothesis of the existence of defense mechanisms against bacterial biofilms in R. microplus eggs. The eggs wax water/methanol extract showed activity against Pseudomonas aeruginosa biofilm without affecting its viability. This extract also presented an antibiotic effect against Staphylococcus epidermidis. We have identified the molecule anti-biofilm and antibiotic as N-(3-sulfooxy-25-cholest-5-en-26-oyl)-L-isoleucine (boophiline). In the search for possible mechanisms responsible by antibiofilm effect of boophiline against P. aeruginosa, 14 genes were evaluated by qRT-PCR. We showed that boophiline inhibits the expression of fliC (flagellum) and cdrA (matrix structural component), whose products are necessary for biofilm formation in P. aeruginosa. Bis-(3’–5’)-cyclic dimeric guanosine monophosphate (c-di-GMP) is an important second messenger, characteristic of Gram-negative bacteria. High intracellular levels of c-di-GMP promote a sessile mode of growth, while low levels promote motile behavior. In line with this, we found that boophiline increases swarming motility, which raised the question whether it acts by altering c-di-GMP levels. Interestingly, when c-di-GMP levels were increased by overexpression of a diguanilate cyclase, boophiline no longer inhibited biofilm formation. One explanation for these results is that boophiline interferes with a specific c-di-GMP-regulated pathway, which would explain we have not obtained a decrease in the total level of this second messenger. Conversely, boophiline had a bactericidal effect against S. epidermidis. Significant morphological changes were observed in the boophiline-treated cells, which appeared to be severely damaged. Boophiline was found to cause abnormal septum formation, bacterial membrane disruption, and extravasation of intracellular material. Additionally, our work also aimed to evaluate the potential antibiofilm and anti-protozoa of culture filtrates obtained from bacteria isolated of R. microplus tissues. Fourteen bacterial culture filtrates showed remarkable activity against of P. aeruginosa and S. epidermidis biofilms, and were cytotoxic against Tritrichomonas foetus. Our work is pioneer in demonstrating the existence of protection mechanisms in tick eggs against biofilms, and ticks-associated bacteria as producers of bioactive molecules. Furthermore, we demonstrated that boophiline is a new antibiofilm molecule, and is the first reported in the literature that a molecule inhibits cdrA. The data obtained in our study could stimulate new approaches in areas such as the physiology and control of arthropods, the genetics and physiology of microorganisms, and biofilm control.

När människan känner stress aktiveras människans biologiska system för att hantera situationen och stresshormonet kortisol produceras bland annat. The World Health Organisation (WHO) har underrättat om att den materiella stressens roll under graviditeten och uppföljningsstudier har väckt oro för att avkommor från mödrar som utsätts för stress under graviditeten kan ha en ökad risk för specifika sjukdomar såsom missbildningar, astma och psykiska och beteendestörningar. Syfte: Att beskriva om stress under graviditet påverkar fostret och kan kopplas samman med folksjukdomar senare i livet. Metod: En allmän litteraturöversikt har genomförts genom att kunskap inom ett begränsat område kartlagts, beskrivits, kritiskt granskats och analyserats utifrån tio utvalda vetenskapliga artiklar inom ämnesområdet. I analysen valdes fyra teman ut och utifrån dessa fyra teman skapades fyra resultatrubriker. Resultat: Forskning visar att stress under graviditet påverkar barnet och kan kopplas samman med folksjukdomar senare i livet hos barnet. Diskussion: Stress under graviditet är ett komplext problem där många faktorer kan spela roll. Litteraturöversikten visar att de vanligast förekommande folksjukdomarna som diabetes, hjärt-och kärlsjukdom samt fetma, som alla är stora kostnader för samhället, skulle kunna vara resultat av stress under moderns graviditet. / Background: A common part of many people's lives is stress, which can be recognized as acute stress, physical stress or stress that many people experience during their daily lives. When a person feels stressed, the human's biological systems are activated to cope with the situation and the stress hormone cortisol is produced, among other things. The World Health Organization (WHO) has reported that the role of material stress during pregnancy, and follow-up studies has raised concerns that the offspring to mothers exposed for stress during pregnancy may have an increased risk of specific diseases such as malformations, asthma and mental and behavioral disorders. Aim: To describe how stress during pregnancy affects the foetus and can be linked with public illnesses later in the child’s life. Method: A general literature review has been conducted by mapping, describing, critically reviewing and analyzing knowledge in a limited area, based on ten selected scientific articles in the subject area. In the analysis, four themes were selected, and based on these four themes four headings were created. Results: Research shows that stress during pregnancy affects the child and can be linked to public diseases later in life. Discussion: Stress during pregnancy is a complex problem where many factors can play a significant role. The literature review shows that the most common public diseases such as diabetes, cardiovascular disease and obesity, all of which are major costs for society, could be the result of stress during mother's pregnancy.

Ainda mais, não há na literatura relato de atividade antibiofilme em ovos de carrapatos e nem mesmo em qualquer outro artrópode. O objetivo do presente trabalho foi avaliar a hipótese da existência de mecanismos de defesa em ovos de R. microplus contra biofilmes bacterianos. O extrato água/metanol da cera dos ovos apresentou atividade contra o biofilme de Pseudomonas aeruginosa sem afetar a sua viabilidade. Esse extrato também demonstrou efeito antibiótico contra Staphylococcus epidermidis. Nós identificamos a molécula com ambas atividades (antibiofilme e antibiótica) como N-(3-sulfooxy-25-cholest-5-en-26-oyl)-L-isoleucina (boophiline). Na busca por possíveis mecanismos responsáveis pelo efeito antibiofilme de boophiline contra P. aeruginosa, 14 genes foram analisados por qRT-PCR. Boophiline inibe a expressão de fliC (flagelo) e cdrA (componente estrutural da matriz), cujos produtos são necessários para a formação do biofilme de P. aeruginosa. Monosfosfato de guanosina dimérico cíclico (c-di-GMP) é um importante segundo mensageiro característico de bactérias Gram-negativas. Altos níveis intracelulares de c-di-GMP promovem o estilo de vida séssil enquanto baixos níveis induzem o comportamento móvel. De acordo com essa afirmação, nós encontramos que boophiline aumenta a motilidade swarming de P. aeruginosa. Desta forma, nos questionamos se o mecanismo de ação de boophiline estaria envolvido com c-di-GMP já que o sistema quorum sensing não foi afetado pela molécula. Interessantemente, quando os níveis de c-di-GMP foram aumentados pela superexpressão de uma diguanilato ciclase, boophiline não inibiu efetivamente a formação de biofilme. Uma explicação para esse resultado é que boophiline interfere em uma via específica regulada por c-di-GMP, o que explicaria não termos obtido um decréscimo no nível total deste segundo mensageiro. Contrariamente, boophiline foi bactericida contra S. epidermidis. Mudanças morfológicas significativas foram observadas em células tratadas com a molécula, as quais foram severamente danificadas. Boophiline levou a formação anormal de septo, rompimento da membrana bacteriana e extravasamento do material intracelular. Adicionalmente avaliamos o potencial antibiofilme e anti-protozoário de filtrados de cultura obtidos de bactérias isoladas de tecidos de R. microplus. Quatorze filtrados de cultura bacteriano apresentaram notável atividade contra o biofilme de P. aeruginosa e S. epidermidis e foram citotóxicos contra Tritrichomonas foetus. Nosso trabalho é pioneiro em demonstrar a existência de proteção contra biofilmes em ovos de carrapatos bem como de bactérias associadas a carrapatos como produtoras de moléculas bioativas. Além disso, nós demonstramos que boophiline é uma nova molécula antibiofilme, sendo a primeira vez relatado na literatura que um composto age inibindo cdrA. Os dados obtidos em nosso estudo poderiam estimular novas abordagens em áreas como fisiologia e controle de artrópodes, genética e fisiologia de microrganismos e controle de biofilmes. / The oviposition is a vulnerable stage of the tick life cycle. Rhipicephalus microplus, as all Ixodidae and Argasidae, has a specialized gland, the Gene’s organ, which produce a wax that is smeared on egg surface during oviposition. In addition to restricting excessive water loss, wax acts as a barrier to attack by invading organisms. In R. microplus, as in other ticks, there are few studies showing antimicrobial activity in eggs. Moreover, there is no report of antibiofilm activity in tick eggs nor in any other arthropod. The objective of our study was to evaluate the hypothesis of the existence of defense mechanisms against bacterial biofilms in R. microplus eggs. The eggs wax water/methanol extract showed activity against Pseudomonas aeruginosa biofilm without affecting its viability. This extract also presented an antibiotic effect against Staphylococcus epidermidis. Bis-(3’–5’)-cyclic dimeric guanosine monophosphate (c-di-GMP) is an important second messenger, characteristic of Gram-negative bacteria. High intracellular levels of c-di-GMP promote a sessile mode of growth, while low levels promote motile behavior. In line with this, we found that boophiline increases swarming motility, which raised the question whether it acts by altering c-di-GMP levels. Interestingly, when c-di-GMP levels were increased by overexpression of a diguanilate cyclase, boophiline no longer inhibited biofilm formation. One explanation for these results is that boophiline interferes with a specific c-di-GMP-regulated pathway, which would explain we have not obtained a decrease in the total level of this second messenger. Conversely, boophiline had a bactericidal effect against S. epidermidis. Significant morphological changes were observed in the boophiline-treated cells, which appeared to be severely damaged. Boophiline was found to cause abnormal septum formation, bacterial membrane disruption, and extravasation of intracellular material. Additionally, our work also aimed to evaluate the potential antibiofilm and anti-protozoa of culture filtrates obtained from bacteria isolated of R. microplus tissues. Fourteen bacterial culture filtrates showed remarkable activity against of P. aeruginosa and S. epidermidis biofilms, and were cytotoxic against Tritrichomonas foetus. Our work is pioneer in demonstrating the existence of protection mechanisms in tick eggs against biofilms, and ticks-associated bacteria as producers of bioactive molecules. Furthermore, we demonstrated that boophiline is a new antibiofilm molecule, and is the first reported in the literature that a molecule inhibits cdrA. The data obtained in our study could stimulate new approaches in areas such as the physiology and control of arthropods, the genetics and physiology of microorganisms, and biofilm control.
