Infecções fúngicas oculares : epidemiologia e etiologia de 23 casos de ceratite fúngica no Rio Grande do Sul / Fungal eye infections: epidemiology and etiology of 23 cases of fungal keratitis in Rio Grande do Sul

A ceratite fúngica (CF) é uma micose ocular oportunística, que tem como sítio de infecção a córnea. Não é uma enfermidade de risco de vida, mas de extremo comprometimento visual e dificuldade terapêutica e, em casos graves, podendo levar à cegueira total ou mesmo a perda do globo ocular. O trabalho objetivou identificar os agentes etiológicos causadores da CF, e descrever os critérios terapêuticos empregados. No período de 1998 a 2011 foram estudados 23 casos de CF diagnosticados no Laboratório de Micologia da Irmandade da Santa Casa de Misericórdia de Porto Alegre / RS. Foram analisados os aspectos demográficos, as doenças de base e os fatores associados ao desenvolvimento da doença, assim como os critérios terapêuticos. No presente estudo, a mediana de idade foi 45 anos, com variação entre 15 e 76 anos, com predomínio do gênero masculino. Os fungos filamentosos figuraram em 78% (18/23) dos casos analisados, sendo o gênero Fusarium spp. o agente etiológico de maior frequência. Concluiu-se que uma compreensão epidemiológica local e a identificação dos fatores de risco, agregados ao diagnóstico micológico precoce e eficaz, são fundamentais na prevenção e correta conduta terapêutica da CF no Rio Grande do Sul, sendo que estas práticas evitarão complicações de perda do globo ocular, melhorando o prognóstico oftalmológico do paciente. / The fungal keratitis is an ocular opportunistic mycosis, which has the cornea as site of infection. Isn’t a life-threatening disease, but with extreme visual impairment and therapeutic difficulty, which can lead to total blindness or even loss of the eyeball, in severe cases. The study aimed to identify the etiologic agents causing fungal keratitis, and describe the therapeutic criteria used. In the period 1998 to 2011 were retrospectively studied 23 cases of fungal keratitis diagnosed at the Mycology Laboratory of Irmandade Santa Casa de Misericórdia Hospital of Porto Alegre / RS. We analyzed the demographics, underlying diseases and the factors associated with disease development, as well as therapeutic criteria. In this study, the median age was 45 years old, ranging between 15 to 76 years old, predominantly male. The filamentous fungus corresponded to 78% (18/23) of the cases analyzed, and Fusarium spp. has been the etiologic agent of highest frequency. It was concluded that an understanding of local epidemiological and identification of risk factors, added to the early and effective mycological diagnosis are essential to prevent and correct therapeutic approach for fungal keratitis in Rio Grande do Sul. These practices will prevent complications of loss of the eyeball, improving the ophthalmological prognosis of the patient.

Desenvolvimento de substrato supressivo à murcha do crisântemo causada por Fusarium oxysporum /

Dessa forma, o presente trabalho teve por objetivo desenvolver um substrato supressivo à murcha do Fusarium em crisântemo com a introdução de matéria orgânica em substratos comerciais. Para tanto, lodo de esgoto e lodo de esgoto compostado; torta de mamona; esterco suíno; cama aviária; compostos comerciais Lanzi®; casca de camarão, biofertilizante e hidrolisado de peixe foram incorporados a substratos à base de casca de Pinus e de turfa em diferentes concentrações e combinações. Os experimentos foram realizados em propriedade produtora de crisântemo Bola-belga com problemas de Fusarium. Em todos os experimentos o número mínimo de repetições foi de 20 vasos por tratamento. Transcorridas 8, 12, 15 e 20 semanas do transplantio foi avaliada a severidade da doença por uma escala de notas de 0 para planta sadia a 5 para planta morta. Com os dados foram calculadas as áreas abaixo da curva de progresso da severidade da murcha de Fusarium. Além disso, foram realizadas análises dos atributos químicos e da atividade microbiana dos substratos bem como do desenvolvimento das plantas. O lodo de esgoto, lodo de esgoto compostado, cama aviária, casca de camarão e o composto Lanzi® induziram a supressividade do substrato à base de casca se Pinus e/ou de turfa, controlando a murcha de Fusarium. Por outro lado, esterco suíno, torta de mamona, hidrolisado de peixe, quitosana e Trichoderma asperellum não interferiram na supressividade à doença. With the advancement of acreage and monoculture, the emergence of several diseases is well known, which has generated losses of quality and productivity of these grains. Among the diseases affecting the corn crop, there is the rot of corn which consequently cause the so-called burning grains, in addition, some of the causative fungi can synthesize mycotoxins that contaminate grain, affecting the quality of these and mainly generating risks animal and human health. In this context the objective of this study is to evaluate the influence of the hybrid, the seeding system, irrigation and foliar application of fungicides in the incidence of fungi that cause the ear rot in corn. The experiments (Experiment I, as irrigation and Experiment II, without irrigation) were installed and conducted in Cruz Alta, RS, in the season 2014/15. The experimental design was randomized blocks in three-factor scheme 2x2x4 corresponding to two hybrids (BG7051H and BG7060HR), two seeding system (direct and conventional) and four treatments (T1: Witness, T2: Pyraclostrobin + Epoxiconazole; T3: Picoxystrobin + cyproconazole; T4: Azoxystrobin + Benzovindiflupir), respectively, in 4 repetitions. The experimental plot had four rows spaced (0.50 m 10.0 m), total area of 20 m2 and floor area of 8 m2, which took place the plant height of reviews and the insertion of the first ear (cm) mass tang (g), number of grains per ear (n), mass of grains per spike (g) of the 1000 seeds (g), grain yield (kg ha-1), incidence of damaged kernels (%) , grain sanity ( "Blotter Test") and in vitro susceptibility of fungi the fungi Penicillium sp., Fusarium spp. and Aspergillus sp. fungicide. / O milho é um dos mais importantes alimentos para humanos e animais, sendo considerado um dos principais cereais cultivados no mundo. Com o avanço das áreas cultivadas e do monocultivo, o surgimento de diversas doenças é notório, o que tem gerado perdas de qualidade e produtividade desses grãos. Dentre as doenças que afetam a cultura do milho, destaca-se a podridão de espiga que por consequência ocasiona os chamados grãos ardidos, além disso, alguns dos fungos causadores podem sintetizar micotoxinas que contaminam os grãos, afetando a qualidade destes e principalmente gerando riscos à saúde animal e humana. Neste contexto o objetivo deste trabalho foi avaliar a influência do híbrido, do sistema de semeadura, da irrigação e da aplicação foliar de fungicidas na incidência de fungos causadores da podridão da espiga na cultura do milho. Os experimentos (Experimento I, como irrigação e Experimento II, sem irrigação), foram instalados e conduzidos no município de Cruz Alta, RS, na safra agrícola 2014/15. O delineamento experimental utilizado foi em blocos casualizados, em esquema trifatorial 2x2x4 correspondente a dois híbridos (BG7051H e BG7060HR), dois sistemas de semeadura (direta e convencional) e quatro tratamentos (T1: Testemunha; T2: Piraclostrobina + Epoxiconazol; T3: Picoxistrobina + Ciproconazole; T4: Azoxistrobina + Benzovindiflupir), respectivamente, em 4 repetições. A parcela experimental apresentou 4 linhas espaçadas (0,50 m e 10,0 m), área total de 20 m2 e área útil de 8 m2, onde realizou-se as avaliações de altura de planta e da inserção da primeira espiga (cm), massa da espiga (g), número de grãos por espiga (nº), massa de grãos por espiga (g), massa de 1000 grãos (g), produtividade de grãos (Kg ha-1), incidência de grãos ardidos (%), sanidade de grãos ( Blotter Test ) e sensibilidade in vitro de fungos dos gêneros Penicillium sp., Fusarium spp. e Aspergillus sp. a fungicidas.

Preliminary investigation of the natural contamination of agricultural crops with selected mycotoxins in northern rural South Africa (Limpopo and Mpumalanga Provinces)

However poor storage practices and contamination with mycotoxins, particularly fumonisins and aflatoxins impacts adversely on production, food safety and food security. Mycotoxins are toxic natural food-borne compounds which frequently contaminate agricultural produce worldwide. They are hazardous to humans and animals and result in significant production losses for farmers. This study focused on former Bantustans in Northern South Africa, namely Vhembe District Municipality (Limpopo) and Gert Sibande District Municipality (Mpumalanga). The aim was to assess mycological and mycotoxin contamination of crops grown by subsistence farmers. A semi-structured questionnaire was administered to randomly thirty-nine households. Data on demographics, storage practices and production during period of 2011 and 2012 cropping seasons were collected. One hundred and fifteen (115) crop samples (maize, beans and peanuts) were collected for analysis. Standard mycological methods and validated mycotoxin analysis methods (HPLC and LC- MS/MS) were used. It was found that maize was the staple food in both provinces, with a significant difference (p = 0.0184) in its production between the two districts; Vhembe produced 0.6 tonnes compared to 2.4 tonnes in Gert Sibande. The majority of the farmers for storage used traditional open wooden cribs (15/20) and steel tanks (5/20) while VDM farmers used sealed store houses 5/19 and 15/19 used polystyrene sacks. Aflatoxin occurrence was low with <1% of GSDM samples contaminated compared to 11% of VDM samples. No significant difference (p > 0.05) was observed in the aflatoxin contamination in VDM samples between the year 2011 and 2012. Samples from VDM households had higher Aspergillus fungal infection (maximum incidence 69%) compared to GSDM (27%) over both seasons. The most frequently isolated Fusarium species in VDM samples was F. verticillioides (92%; 93%), and F. subglutinans (97%; 80%) in GSDM samples over seasons 2011 and 2012, respectively. Highest levels of fumonisins (FB1+ FB2) ranged between 1010 μg/kg and 12168 μg/kg with less than 30% extremely contaminated above the regulated limit in 91% of samples from Limpopo over both seasons (2011 and 2012). Fumonisin levels between the two seasons in VDM showed no significant difference (p>0.05). Only three (less than 5%) from 68% GSDM contaminated maize samples were above the FB1 and FB2 limit. In 2011, there were two highly contaminated maize samples (1762 μg/kg and 4598 μg/kg) with the other samples less than 600 μg/kg, whereas in season two (2012) all samples were below 200 μg/kg, except one highly contaminated sample (26115 μg/kg). None of the beans and peanuts from Mpumalanga was contaminated with mycotoxins above the recommended limit, but from Limpopo 1/5 peanuts was found contaminated with aflatoxin G1 (41 μg/kg). Natural occurrence and contamination of both fumonisin and aflatoxin in stored home-grown maize from VDM was significantly (p < 0.0001) higher than GSDM over both seasons. In general, Limpopo farmers’ experience lower harvests and greater mycotoxin contamination of agricultural produce. This may be attributed in part to poor storage practices and environmental and climatic conditions in that agro-ecological zone.

FUNCTIONAL DIVERSITY OF FUNGI ASSOCIATED WITH DURUM WHEAT ROOTS IN DIFFERENT CROPPING SYSTEMS

Differences in pea (Pisum sativum L.) and chickpea (Cicer arietinum L.) microbial compatibility and/ or their associated farming practices may influence root fungi of the following crop and affect the yield. The main objective of this research was to explain the difference in durum wheat (Triticum turgidum L.) yield the year after pea and chickpea crops through changes in the functional diversity of wheat root fungi. The effect of fungicides used on chickpea on the root fungi of a following durum wheat crop was studied using plate culture and pyrosequencing. Pyrosequencing detected more Fusarium spp. in the roots of durum wheat after fungicide-treated chickpea than in non-fungicide treated chickpea. Plate culture revealed that the functional groups of fungi responded differently to fungicide use in the field but the effect on total community was non-significant. Highly virulent pathogens were not affected, but antagonists were suppressed. More fungal antagonists were detected after the chickpea CDC Luna than CDC Vanguard. Fungal species responded differently to the use of fungicides in vitro, but the aggregate inhibition effect on antagonists and highly virulent pathogens was similar. The effect of chickpea vs. pea previous crop and different chickpea termination times on root fungi of a following durum wheat crop was studied. The abundance of Fusarium spp. increased after cultivation of both cultivars of chickpea as compared to pea according to pyrosequencing and was negatively correlated with durum yield. Plate culture analysis revealed that fungal antagonists were more prevalent after pea than both cultivars of chickpea and chickpea CDC Vanguard increased the abundance of highly virulent pathogens. The abundance of highly virulent pathogens in durum wheat roots was negatively correlated to durum yield. Early termination of chickpea did not change the community of culturable fungi in the roots of a following durum crop. It is noteworthy that Fusarium redolens was identified for the first time in Saskatchewan and its pathogenicity was confirmed on durum wheat, pea and chickpea. The classical method of root disease diagnostics in cereals is based on the examination of the subcrown internode. I evaluated the method by comparing the fungal communities associated with different subterranean organs of durum wheat. The fungal community of the subcrown internode was different from that of roots and crown, suggesting cautious use of this method.

Untersuchungen zu qualitätsbeeinflussenden, nacherntephysiologischen und phytopathologischen Prozessen bei Convenience-Produkten während der Kurzzeitlagerung am Beispiel von Spargel (Asparagus officinalis L.)

In Deutschland nimmt der Anbau von Bleichspargel (Asparagus officinalis L.) sechzehn Prozent der Gesamtgemüseanbaufläche ein. Die Qualitätssicherung von Spargel, insbesondere aber von geschältem (Convenience-) Spargel stellt wegen der hohen Stoffwechselaktivität nach der Ernte eine Herausforderung dar. Insbesondere gilt es, die Textur und die Inhaltsstoffe vor qualitätsmindernden Veränderungen und verderbsförderndem Pilzbefall zu bewahren. Als Verpackung dienen in der Regel Folienverpackungen, die aber oft für das empfindliche Gemüseprodukt nicht geeignet sind. Daher wurde der Einfluss von unterschiedlichen Folienverpackungen (vier Polypropylenfolien, zwei biologisch abbaubare Folien und ein Oberflächencoating) mit unterschiedlicher Permeabilität für Sauerstoff und Kohlendioxid auf die Veränderungen der Qualitätsparameter ( Farbe, Textur, Frischmasse, Trockensubstanz, Gerüstkohlenhydrate ( Pectine, Lignin, Hemicellulose, Cellulose, Saccharose, Fructose, Glucose) von nicht geschälten und geschälten Spargel unmittelbar nach der Ernte und nochmals nach drei (bzw. vier) Lagertagen (2°C, 10°C, 20°C Lagertemperatur) ermittelt. Direkt nach der Ernte und nach drei Lagertagen wurde bei 10°C und 20°C Lagertemperatur die Kontamination mit Pilzen und der eventuell damit verbundene Gehalt an Fumonisin B1 geprüft. Die Lagertemperatur von 10°C (2 d Lagerdauer) erwies sich als geeignet zur Qualitätserhaltung von Convenience – Spargel. Bei 2°C und 20°C geschältem Spargel waren die Pectinfraktionen, bei nicht geschältem Spargel war der Hemicellulosegehalt Veränderungen unterworfen. Die Veränderungen dieser Qualitätsparameter waren mit den Veränderungen der Textur korreliert. Die Spargelspitze ist bei der Lagerung (2°C und 20°C) stoffwechselaktiver, als die restliche Spargelstange. Es zeigte sich, dass das Verhältnis von O2 zu CO2 (RQ) in der Verpackungsfolie signifikanten Einfluss auf die Qualitätsparameter von geschältem Spargel ausübte. Die geringsten stoffwechselphysiologischen Veränderungen wurden bei einem RQ von 0,65 festgestellt. Folienverpackungen mit RQ von 0,03-0,65 hatten sich für das endophytische Pilzwachstum als hemmend erwiesen. Das Mykotoxin Fumonisin B1 wurden in gesundheitlich unbedenklichen Mengen (< 1,67 mg * kg TS-1) in nicht gelagerten und in gelagerten Spargelstangen nachgewiesen. / The production area of white asparagus comprises 16 % of all vegetable crops in Germany. The economic important asparagus (Asparagus officinalis L.) is known for its high quality loss in postharvest. In order to protect convenience asparagus, i.e. fresh-cut peeled white asparagus from rapid deterioration in respect to phytopathological fungi, nutritive and sensory compounds the commercial use of film packaging might be an important tool. However, the film packaging materials used commercially do often not fulfil product physiological concerns. Therefore, the influence of different film packaging materials (four polypropylene-films, two biological degradable films and one coating) with different permeabilities for CO2 and O2 was investigated for unpeeled and peeled white asparagus during storage (2, 3, and 4 days) at temperatures of 2°C, 10°C, 20°C. Changes in the following quality attributes were studied: colour, texture, fresh weight, dry weight, structural carbohydrates (pectic substances, lignin, hemicellulose, cellulose), mono- and disaccharides (fructose, glucose, sucrose). Moreover, at harvest and after three days of storage the contamination with fungi and the content of the mycotoxin Fumonisin B1 was observed. The tests for contamination of fungi were conducted with slight nutrient agar (SNA) for seven days at 20°C under 14 h UV light and 10 h darkness (Nirenberg, 1976) Quality changes were most inhibited at a storage temperature of 10°C for two days. The ratio of O2 to CO2 (RQ) within the film packaging had a pronounced effect on the quality attributes of peeled asparagus. Changes in dry weight, fresh weight, lignin content were low at a RQ of 0,65 (P-Plus 2 film), whereas the water-soluble/insoluble pectin ratio (1 : 0,8) remained constant during the entire storage period. A low correlation was found between texture and hemicellulose, glucose, sucrose and the ratio of water-soluble to insoluble pectin. The meristematic zone of the asparagus tip revealed a higher metabolic activity than other morphological parts of the spear. Endophytic fungi, e.g. Fusarium spp., a precursor of mycotoxin, was found in control asparagus spears. During storage, the development of Fusarium spp. could be inhibited by all film packaging revealing a RQ of 0,003 - 0,65. The mycotoxin Fumonisin B1 occurred in all asparagus spears (control and stored spears), however the content (< 1,67 mg * kg TS -1) did not reach health risk threshold values.
