The topic of this thesis is a logical continuation of the bachelor project. In our previous project, using the method of Borrelia cultivation on solid medium, we succeeded obtaining a monoclonal population of B. garinii from the american isolate in which the presence of more borrelia species was confirmed. By this we confirmed the presence so-called "european" Borrelia species in the southeastern United States (specifically in South Carolina). By multilocus analysis of eight housekeeping genes of american isolate it has been shown that the obtained strain of B. garinii is different from the previously described european and asian strains of this species. The main objective of the proposed project, is the analysis of the pathogenic potential of the isolated clone of B. garinii. Validation of the pathogenic potential of American B. garinii isolate in laboratory mice model could be important for the public health of the southeast american population, as this will confirm the possibility of existence of Lyme disease, caused by this spirochete species, with significantly different clinical signs in this region. For this purpose, the method of direct diagnosis (infecting laboratory animals and transferring of infection from infected animals to the tick and vice versa) was used. Laboratory mice were infected subcutaneously, and the spirochete infection was confirmed in ear tissue by PCR. Uninfected Ixodes ricinus larvae were fed on infected mice. After the confirmation of infection in newly hatched nymphs they were fed on naive mice. The ability of transmitted spirochetes to disseminate into different mice tissues was confirmed by PCR. The presence another so-called "European" species, B. afzelii, was confirmed in original rodent associated American isolate. Another aim of the project was to obtain monoclonal populations B. afzelii from co-infected northamerican sample confirming by this way the transoceanic migration of spirochtes.

This thesis focuses on geographical literacy of the general public. It sets two main aims. Firstly to identify geographical community opinion on what knowledge and skills should be these days considered basics of geographical literacy for everyone regardless of their age and achieved education. Secondly to verify the selected basics of geographical literacy with the lay public. The structure of this work follows these aims. The theoretical part concentrates on the explanation of key words and the definition of geographical literacy in accordance with contemporary literature. The other part of the work presents the results of two researches of one's own. The first one was done in the form of an electronic questionnaire and addressed to the members of the geographical community. They firstly used a four-level scale to consider the importance of knowledge and skills that were verified in the research of the geographical literacy of adults in the USA. Secondly, in the form of open-answer questions, they could express their opinion on the core of the monitored assessment reference scope. It proved that assessment statements to most suggested skills are very varying. The target of this thesis is to assess a demographic development of the South Bohemian country since 1989. Basic components of the demographic reproduction will be analyzed in detail in selected classification level within the bounds of context of the social ? economical development. Another part of the thesis is a usage of three-dimensional analysis at GIS. This analysis should serve as a device for forecasts of the future demographic development of the South Bohemian country and also for proposals for improvement of the situation in these parts of the Czech Republic. Considering increasing pressure on a development of provinces, the most of strategic documents are focused on a development of the provinces on a local, regional or supranational level so that a possible potential of this country areas is evident from the point of view of the future development of population.

Epidemiologie roztroušené sklerózy mozkomíšní / Epidemiology of multiple sclerosis

There are approximately 2.5 million cases of multiple sclerosis [MS] in the world. Each year, around 10,000 new cases of MS are diagnosed. There are approximately 0.1%. in the Czech Republic and that means that there are approximately 15,000 people with this disease. Multiple sclerosis is a progressive neurological inflammatory disease of the central nervous system in pathogenesis. The auto-immune mechanisms are appliedinvolves; both the affect the myelin and damaged axons. This damage is responsible for the permanent disability of MS. The main objective of this thesis was to use quantitative research to describe the occurrence of MS in the Regions of South Bohemia and Usti, and within objective to estimate the true prevalence of multiple sclerosis in the South and the Usti Regions. MUDr. Príkaszký and I used the method of descriptive epidemiological studies on the technique of data collection, as well as the analysis and the comparison of two sources using the double-capture method. As a research area, we used a set of patients with multiple sclerosis. The data were gathered in two regions of the Czech Republic: Usti nad Labem Region and South Bohemia. We investigated the number of patients from neurologists and MS centers in both regions.There is also the data on health insurance included with the Ústí Region in this research. According to the data, we estimated the prevalence of neurologists in South Bohemia to be 79.20 per 100,000 people. The MS Center at the hospital in Ceske Budejovice has registered 512 patients: 388 women and 124 men. The values of the MS Centre in Ceske Budejovice received from the neurologists are a total of 882 patients, which is a prevalence of 138.6 patients per 100,000 of the population. Regarding the Usti Region, the prevalence reported by neurologists is 208.2 per 100 000 people. The MS Center at the hospital in Teplice, has registered 1139 sick 730 women and 409 men. According to health insurance data in their database, of 1187 people listed with a diagnosis of G.35, 333 are men and 854 are women. This represents an estimate of the prevalence 305/100000 of the people who are registered in the NGA. We calculated the prevalence of illnesses from those sources in both of these regions is higher than the general estimates. When comparing the data source of health insurance and the model file from the MS center of Ústí Region, we calculated the overall sensitivity of 91.7% within a health insurance group. The calculation of the estimated prevalence has been reached by using the method of double-capture of those sources that provided an estimate of 2,681 patients in the Usti Region. The models of this data are mainly demonstrated as a possible approach to the estimate of the prevalence of this chronic disease in the case of multiple sclerosis. The information has been obtained from the collection of data. The results of this study can be used by other workers who have the same objective as this study.

This diploma thesis deals with the management of tourism development and destination marketing with a focus on the latest trends in the tourism market and their adoption by the destination management organizations (DMOs) in the regions of Czechia. The thesis is divided into two parts. In the theoretical part general preconditions for successful development of incoming tourism are described and the possible use of specific electronic tools for destination marketing at the international level is outlined. The empirical part is dedicated to the critical evaluation of geographic definition of tourist regions in Czechia and geographic nomenclature in English. Tourist web portals in the Czech regions were analyzed in comparison with portals of selected regions abroad. Attention is also paid to the use of social media in destination marketing. Finally, it outlines suggestions for improving the current situation of digital destination marketing in Czech regions.
