El papel del parvovirus B19, de los virus herpes y de la metaloproteinasa-2 y 9 en la etiopatogenia de la arteritis de células gigantesRodríguez Pla, Alicia 14 March 2003 (has links)
La etiología de la arteritis de células gigantes (ACG) es desconocida y sus aspectos etiopatogénicos están poco estudiados.Objetivos. 1. Estudiar la presencia de parvovirus B19 y los virus herpes en las arterias temporales positivas y negativas para la ACG. 2. Estudiar la asociación entre la expresión de metaloproteinasa-2 (MMP-2) y 9 (MMP-9) y el resultado de la biopsia de la arteria temporal (BAT), qué células las expresan y su localización. 3. Analizar si la presencia de estas MMP o los virus se relaciona con las alteraciones histológicas y con las variables clínico-epidemiológicas de los pacientes. 4. Conocer cuáles son los factores predictores del resultado de la BAT. Material y métodos. Se incluyeron todas las BAT valorables realizadas de forma consecutiva en el Hospital Universitari Vall d'Hebron por sospecha clínica de ACG entre Enero de 1997 y Marzo del 2002. Como criterios anatomopatológicos se utilizaron los del ACR. La presencia de ADN vírico se determinó mediante reacción en cadena de la polimerasa. Se realizaron nuevas tinciones de hematoxilina-eosina, de fibras elásticas y tricrómicro de Masson. Los macrófagos CD68, la MMP-2 y la MMP-9 se detectaron mediante inmunohistoquímica. De las historias clínicas se obtuvieron datos clínicos y epidemiológicos. Para el ajuste multivariable se utilizó la regresión logística múltiple.Resultados. En el período en estudio se encontraron 147 BAT válidas, 50 (35%) positivas para ACG y 97 (66%) negativas. Se recuperó la historia clínica de 125 pacientes, 46 (36,8%) con BAT (+) y 79 (63,2%) BAT (-). No se encontró presencia de ADN de los virus estudiados en ninguna de las BAT. La MMP-9 se expresaba con más frecuencia en BAT positivas que negativas para ACG (OR=2,93 p=0,005). Para la MMP-2 la diferencia no fue estadísticamente significativa (p=0,08). Ambas MMP se expresaban en macrófagos de la íntima y de la media cercanos a la lámina elástica interna, en células gigantes y en células con apariencia de músculo liso de la media y de la íntima. Le expresión de MMP-2 se relacionó de forma inversa con la afectación exclusiva de la adventicia (p=0,047) y de forma positiva con la fiebre (p=0,025). La MMP-9 se relacionó de forma positiva con: hiperplasia intimal (0,037), degeneración lámina elástica (0,0001), disminución de la luz (0,008), hipercolesterolemia (0,032), hiperestesia craneal (0,009). Ajustando por variables que podrían influir en el resultado de la biopsia, (edad, género, tiempo de evolución de los síntomas hasta la BAT, tratamiento previo con glucocorticoides y año de realización de la biopsia), se mantuvieron los resultados entre la expresión de MMP-2 y MMP-9. El engrosamiento de la arteria temporal, la disminución de su pulso y la pérdida de peso se asociaron de forma independiente con el resultado de la biopsia, ajustando por edad, género, días de evolución, tratamiento previo, año de realización, cefalea, hiperestesia craneal, claudicación mandibular, alteraciones visuales y VSG.Conclusiones. Ninguno de los virus estudiados parece estar implicado en la etiopatogenia de la ACG. La MMP-9, al contrario de la MMP-2, parece tener un papel en dicha etiopatogenia. La MMP-9 se asocia con hipercolesterolemia y la MMP-2 con la fiebre. El engrosamiento, la disminución de la luz de la arteria temporal y la pérdida de peso se han encontrado asociadas a una elevada probabilidad de resultado positivo de la biopsia en los enfermos estudiados. / The etiology of giant cell arteritis (GCA) is unknown and their etiopathogenic aspects have been little studied.Aims. 1. To study the presence of parvovirus B19 and herpes viruses in positive and negative temporal artery biopsies (TAB) for GCA. 2. To study the association between metalloproteinase-2 (MMP-2) and 9 (MMP-9) expression and TAB result, the type of cells expressing them and their location. 3. To analyse if the presence of these MMP or the viruses is related to histologic findings and to clinical-epidemiological variables of the patients. 4. To identify predictive factors of TAB result. Material and methods. All the valid TAB consecutively performed in Hospital Universitari Vall d'Hebron because of clinical suspicion of GCA between January 1997 and March 2002. ACR anatomopathological criteria were used. The presence of viral DNA was determined by means of polymerase chaín reaction. New hematoxilin-eosin, elastic fibers and Masson's trichromic stainings were performed. CD68 macrophages, MMP-2 and MMP-9 were detected by immunohistochemistry. Clinical and epidemiological data were obtained from clinical files. The multiple logistic regression was used for multivariable analysis. Results. During the study period, 147 valid TAB were found, 50 (35%) positive and 97 (66%) negative for GCA. Clinical files of 125 patients were retrieved, 46 (36.8%) with positive TAB and 79 (63.2%) with negative TAB. DNA of the viruses studied was not found in any of the TAB. MMP-9 was more frequently expressed in positive than in negative TAB for GCA (OR=2.93; p=0.005). The difference was not statistically significant for MMP-2 (p=0.08). Both MMP were expressed in macrophages of the intima and the media near internal elastic lamina, in giant cells and in cells of the media and the intima resembling smooth muscle cells. The expression of MMP-2 was inversely related to the isolated involvement of the adventitia (p=0.047) and positively to fever (p=0.025). MMP-9 was positively related to: intimal hyperplasia (0.037), elastic lamina degeneration (0.0001), luminal narrowing (0.008), hypercholesterolemia (0.032), scalp tenderness (0.009). Controlling for variables which could influence the biopsy result (age, gender, evolution of symptoms before TAB, previous glucocorticoids treatment and year of biopsy performance), the results between MMP-2 and MMP-9 expression remained unaltered. Temporal artery thickening, pulse decrease and weight loss were independently associated with biopsy result, controlling for age, gender, days of evolution of symptoms, previous treatment, year of biopsy performance, headache, scalp tenderness, jaw claudication, visual disturbances and erythrocyte sedimentation rate.Conclusions. None of the viruses studied seems to be involved in GCA etiopathogenesis. MMP-9, in contrast to MMP-2, seems to play an etiopathogenic role in GCA. MMP-9 is associated with hypercholesterolemia and MMP-2 with fever. Temporal artery thickening and luminal narrowing and weight loss are associated with a high probability of positive biopsy result in the patients studied.

El objetivo central de esta tesis es intentar contribuir a la comprensión de los procesos que dan lugar a la formación de sistemas planetarios como un todo. Para esto, trabajamos en la construcción de modelos de crecimiento planetario en el contexto de un disco protoplanetario realista, que evoluciona por viscosidad del disco gaseoso como así también por migración de planetesimales. Específicamente calcularemos la interacción que surge entre dos (o más) planetas gaseosos gigantes que crecen simultáneamente en un mismo disco protoplanetario.
Aunque al presente se dispone de una descripción plausible de la formación planetaria, dicha descripción se basa en modelos construidos bajo la hipótesis del crecimiento de un planeta aislado. Esta configuración es poco realista e insuficiente para comprender globalmente tanto la formación del Sistema Solar como la de los sistemas planetarios extrasolares recientemente descubiertos. De los más de 300 exoplanetas descubiertos hasta el momento (The Extrasolar Planets Encyclopaedia, http://exoplanet.eu/catalog.php) muchos de ellos se encuentran en sistemas múltiples (alrededor de 30 sistemas planetarios descubiertos a la fecha); aún más, recientemente Marois et al. 2008 presentaron las primeras imágenes directas (en infrarrojo) de exoplanetas. Estas imágenes muestran tres planetas gigantes orbitando la estrella de tipo espectral A HR 8799, perteneciente a la constelación de Pegasus. Estos datos son determinantes para incluir a la formación simultánea de planetas gigantes en los modelos de formación de sistemas planetarios.

Previous issue date: 2015 / Resumo: A química supramolecular envolve estruturas mantidas por interações intermoleculares. Neste contexto, sistemas que apresentam estruturas auto organizadas são de interesse devido importância como ciência de base bem como aplicabilidade. Neste trabalho foram utilizadas micelas de brometo de hexadeciltrimetilamônio (CTAB) como reticulantes de hidroxietilceluloses hidrofobicamente modificadas. Como as micelas estão em constante quebra e recombinação, reticulações transientes são obtidas. Tais sistemas apresentam comportamento reológico característico, dependente da concentração do surfactante. Obervou-se que a celulose que apresenta maior ramificação hidrofóbica resulta em aumento mais efetivo da viscosidade como consequência do maior grau de reticulação. A Técnica de calorimetria de titulação isotérmica (ITC) foi utilizada de modo a compreender os processos energéticos envolvidos na agregação. Com esta pôde-se obter parâmetros do sistema como cmc, cac e ?H°mic . Elucidou-se também que as interações polímero-surfactante são de origem hidrofóbica. A adição de salicilato de sódio (NaSal) em solução de CTAB leva a formação de micelas gigantes. Determinou-se por ITC a razão, [CTAB]/[NaSal], em que ocorre a formação de micelas gigantes, bem como a variação de entalpia deste processo. Sistemas contendo micelas gigantes e na presença de celulose hidrofóbicamente modificada foram estudados por reologia. Variações em seu comportamento foram claramente observadas / Abstract: Supramolecular chemistry involves structures maintained by the intermolecular interactions. In this context, systems that have self-assembled structures are of interest because of importance as basic science as well as applicability. In this study, we used micelles formed by hexadecyltrimethylammonium bromide (CTAB) as crosslinking agents of hydrophobically modified hydroxyethylcelluloses. As the micelles are constantly breaking and recombination, transient crosslinks are obtained. Such systems have characteristic rheological behavior, dependent on the surfactant concentration. It was observed that the cellulose which is more hydrophobic branching results in more effective increase in viscosity as a result of the higher degree of crosslinking. Isothermal titration Calorimetry (ITC) technique was used to understand the energy involved in the aggregation process. With this it was possible to obtain system parameters such as cmc, cac and ?H°mic. It is also elucidated that the interaction polymer-surfactant are of hydrophobic origin. The addition of sodium salicylate (NaSal) into CTAB solutions leads to formation of giant micelles. It was determined by ITC the ratio [CTAB]/[NaSal] in what occurs the formation of giant micelle, and the enthalpy change of the process. Systems containing giant micelles in the presence of hydrophobically modified cellulose were studied by rheology. A amostra foi constitu?da por 20 esp?cimes teciduais emblocados em parafina de LPCG, 20 de LCCG n?o agressivo e 20 de LCCG agressivo, oriundos do Servi?o de Anatomia Patol?gica da Disciplina de Patologia Oral do Departamento de Odontologia da UFRN. Em rela??o ao GLUT-1, verificou-se uma diferen?a estatisticamente significante (p< 0,05) na quantidade de c?lulas mononucleares imunomarcadas entre a les?o perif?rica (LP) e a les?o central n?o agressiva (LCNA) e entre a LP e a les?o central agressiva (LCA). Estudo microscópico morfométrico e genotípico de pacientes portadores de lesão central de células gigantes / Microscopic morphometric and genotypic assessment of patients with central giant cell lesionsTeixeira, Renata Cordeiro 20 May 2011 (has links)
A lesão central de células gigantes (LCCG) é uma afecção benigna dos maxilares, de comportamento biológico incerto, variando de discreta tumefação assintomática e de crescimento lento à uma forma agressiva, associada a dor, reabsorção radicular e óssea, com destruição cortical. Sua etiologia permanece desconhecida, havendo controvérsias entre processo reacional, neoplásico ou genético. Mutações no gene SH3BP2 foram identificadas em pacientes com querubismo, condição que compartilha várias características clínicas, radiográficas e histopatológicas com a LCCG. Para testar a hipótese de que tais mutações seriam responsáveis por, ou estariam associadas a LCCG e na tentativa de melhor entender a diferenciação microscópica/morfométrica das lesões agressivas e não agressivas, vinte e cinco pacientes portadores de LCCG foram selecionados para o estudo. O DNA foi obtido através do sangue e de espécimes em blocos de parafina, oriundos de biópsias e tratamento cirúrgico. Um estudo microscópico morfométrico foi paralelamente realizado, para avaliar o número de células gigantes e densidade de volume das mesmas nas lesões agressivas e não agressivas. O sequenciamento genético dos treze exons do gene SH3BP2 nos vinte e cinco pacientes estudados evidenciou uma alteração no códon do exon 4 em 10 pacientes. A densidade de volume de células gigantes foi maior nas lesões agressivas quando comparadas às não agressivas (p=0,013). Não houve diferença significante quanto ao número de células gigantes/mm2 em lesões agressivas e não agressivas (p =0,245). / Central giant cell lesion (CGCL) is a benign disease of the jaws, with uncertain behavior, ranging from mild asymptomatic slow-growing swelling to an aggressive form, with pain, radicular and bone resorption and cortical destruction. Its aetiology is still unknown and there is discussion whether it is a reactive, neoplastic or genetic disease. Mutations on gene SH3BP3 were identified in patients with cherubism, which shares several clinical, radiographic and histopathological features with CGCL. In order to test the hypothesis that such mutations would be responsible for or would be related to CGCL and also in order to better understand microscopic morphometric differentiation of the aggressive and non-aggressive lesions, 25 patients with CGCL were selected to this study. DNA was extracted from blood samples and from tissue samples, obtained by biopsy or surgical treatment. Microscopic morphometric assessment was also performed, in order to evaluate the number and the volume density of the giant cells in aggressive and in non-aggressive lesions. Gene sequencing of all 13 exons in gene SH3BP3, performed on each of the 25 patients, showed an alteration in one codon from exon 4, in ten patients. Volume density of giant cells was greater in aggressive lesions than in non-aggressive ones (p=0,013). There was no significant difference on the number of giant cells per mm2 when comparing aggressive and non-aggressive lesions.

Expressão das conexinas 32 e 43 em células trofoblásticas da placenta bovina em cultura celular / Expression of the connexins 32 and 43 in trophoblast cells of the bovine placenta in cellular cultureBirck, Arlei José 05 December 2007 (has links)
A expressão da conexina 32 e 43 nas células gigantes trofoblásticas foi analisada em condições de cultura celular com e sem influência de hormônios sexuais. Placentônios bovinos foram coletados de vacas prenhes em abatedouro nos diferentes períodos gestacionais e divididos em três grupos, primeiro terço (I), segundo terço (II), terceiro terço (III) e transportados ao laboratório em condições assépticas à temperatura de 4ºC em solução de PBS com antibiótico. No laboratório as células foram isoladas e cultivadas em meio D-MEM com 10% SFB por cinco dias. As detecções das conexinas 32 e 43 foram realizadas através de munofluorescencia, pelo método de amplificação da tiramida-fluoresceina, utilizando anticorpo primário policlonal a imunoglobulina de coelho, anti-conexina 32 e 43 de camundongo. Os resultados mostraram que as células gigantes trofoblásticas expressam conexinas 32 e 43 nos três períodos gestacionais com exceção da Cx32 no primeiro terço gestacional sem adição de hormônios, a qual passou a expressar fluorescência após a adição de hormônios. A distribuição da Cx43 evoluiu com a progressão da gestação, permanecendo limitada ao interior das células gigantes trofoblásticas, sem formar junções comunicantes. O terceiro terço gestacional mostra a Cx43 na CGTT localizada no interior do núcleo. A adição de hormônios ao meio de cultura vem confirmar que a progesterona e o estrógeno podem ter um papel no controle da Cx32. Para Cx43 onde se adicionou progesterona os níveis de expressão foram baixos no início aumentando no decorrer da gestação, o mesmo foi encontrado para o conjugado de progesterona/ estrógeno. Para o estrógeno no grupo I os níveis de expressão foram menores se comparados aos três grupos diminuindo no grupo II e voltando a aumentar no grupo III. / The trophoblast cells expression of connexins 32 and 43 was studied in cell culture conditions with or without influence of sexual hormones. Bovine placentomes were collected from pregnant cows in slaughterhouses in different gestational periods and so divided into three groups, first term (I), second term (II), and third term (III), and transported to the laboratory in aseptic conditions at a temperature of 40C in PBS solution with antibiotics. At the laboratory the cells were isolated and cultivated in D-MEM environment with 10% SFB for five days. The detections of connexins 32 and 43 were achieved through immunofluorescence, by the tyramide-fluorescein amplification method, using a rabbit polyclonal primary antibody anti-connexin 32 and 43 of mice. The results showed that the trophoblast cells expressed connexins 32 and 43 in the three gestational periods with an exception of the Cx32 at the first gestational period. However, after the addition of sexual hormones, they began to express the connexins in the cytoplasm in the first stage. The distribution of the Cx43 evolved with the progression of the gestation, remaining limited to the trophoblast`s cells interior, without formation of gap junctions. The third gestational term shows the Cx43 located inside the nuclei of the CGTT. Addition of hormones in the culture environment confirmed that estrogen and progesterone may have an important action controlling Cx32. For Cx43, the expression was low after prosterone was added to the culture medium, but increased as the gestation evolved, the same was found for a combined compost of estrogen\\ progesterone. For estrogen, in group I the expression levels were inferior when compared to the three groups decreasing in group II and increasing again in group III. We may conclude that the sexual hormes, specially estrogen, affect the expression of connexins in trophoblastic cells.

O presente trabalho estudou a expressão da conexina 43 nas células trofoblásticas gigantes bovinas, em especial nas células trofoblásticas gigantes binucleadas. Foram utilizadas 12 amostras de placenta bovina, divididas em três grupos segundo a fase gestacional, primeiro terço, segundo terço e terceiro terço da gestação. O tecido placentário foi fixado em solução metacarn e, posteriormente, submetido ao método tradicional de inclusão em parafina. A detecção da conexina 43 foi realizada através de imuno-histoquímica, pelo método de amplificação da tyramida, utilizando como anticorpo primário policlonal a imunoglobulina de coelho anti-conexina 43 de camundongo. Os resultados deste trabalho indicaram que as células trofoblásticas gigantes bovinas são capazes de expressar a conexina 43 nas três fases da gestação. A distribuição da conexina 43 evoluiu com a progressão da gestação, mas ficou limitada ao interior das células trofoblásticas gigantes bovinas, sem formar marcações pontuais na membrana citoplasmática que pudessem indicar a formação de junções comunicantes. A avaliação de diferentes protocolos histológicos e imuno-histoquímicos permitiu estabelecer a melhor metodologia para a preservação do arranjo tecidual da placenta bovina e a identificação da conexina 43 nas células trofoblásticas gigantes bovinas / The present study has researched the expression of Connexin-43 in bovine trophoblast giant cells, especially in binucleate trophoblast giant cells. Twelve samples of bovine placenta were used, divided into three groups according to the gestational phase, first part, second part and third part of gestation. The placental tissue was fixed in Methacarn solution and later submitted to the traditional method, that is, embedded in paraffin. The detection of Connexin-43 was done through immunohistochemistry by the tyramide amplification method, using as polyclonal primary antibody, rabbit immunoglobuline anti-mouse Connexin-43. The results of this study have revealed that the bovine trophoblast giant cells are capable of the expression of Connexin-43 in the three phases of gestation. The distribution of Connexin-43 evolved as gestation progressed, but was limited to the interior of the bovine trophoblast giant cells without punctate markings in the cytoplasmic membrane which could indicate the formation of communicating junctions. The assessment of different histological and immunohistochemical protocols allowed us to establish the best methodology for the preservation of the tissue arrangement of bovine placenta and the identification of Connexin-43 in the bovine trophoblast giant cells
