Modeling and control of batch pulsed top-spray fluidized bed granulation

In this thesis, a thorough study of the batch top-spray fluidized bed granulation was carried out including experimental study, population balance model (PBM), computational fluid dynamic (CFD) study and control strategy development. For the experimental study, the influence variables of pulsed frequency, binder spray rate and atomization pressure of a batch top-spray fluidized bed granulation process were studied using the Box-Behnken experimental design method. Different mathematical models were developed to predict the mean size of granules, yield, relative width of granule distribution, Hausner ratio and final granule moisture content. Validation experiments have shown the reliability and effectiveness of using the Box-Behnken experimental design method to study a fluidized bed granulation process. The one-dimensional population balance models (ODPBMs) have been developed to model a pulsed top-spray fluidized bed granulation, linking the operating factors of the pulsed frequency, the binder spray rate, and atomization air pressure with the granule properties to predict granule growth behavior at different operating conditions. A multi-stage open optimal control strategy based on the developed ODPBMs was proposed to reduce the model and process mismatch through adjusting the trajectory of the evolution of the granule size distribution at predefined sample intervals. The effectiveness of the proposed modeling and multi-stage open optimal control strategy has been validated by experimental and simulation tests. In addition, an Eulerian-Eulerian two-fluid model (EETFM) was developed to describe the gas-particle two-phase flow in the fluidized bed granulator. By computational fluid dynamic analysis, it has been proven that the fluidized bed granulation system is not homogeneous, based on which a two-compartmental population balance model (TCPBM) was developed to describe the particle growth in the fluidized bed granulation. Validation experiments have shown the effectiveness and superior accuracy of the TCPBM comparing with the ODPBM in predicting the final particle size distribution.

Immobilisation du phosphore par précipitation induite dans un procédé aérobie à biomasse granulaire / Phosphorus removal and induced precipitation in aerobic granular sludge process for wastewater treatment

Depuis une dizaine d'années, les procédés de granulation aérobie sont apparus comme une technologie prometteuse pour le traitement des effluents fortement chargés en azote, phosphore et carbone, tels que ceux issus de l'agro-industrie. La complexité microbienne de ces granules et les mécanismes qui leur donnent des propriétés exceptionnelles de décantation et de cohésion, constituent encore des axes de recherche importants. Dans cette thèse, le travail s'est axé sur un mécanisme encore non étudié : les processus de précipitation des phosphates au cœur des granules microbiennes. Différentes techniques d'analyses spectrales, parfois adaptés pour la première fois à ce type de systèmes, comme la spectroscopie Raman, ont permis de caractériser la nature de ces minéraux formés au cœur des granules. L'analyse menée sur des réacteurs de laboratoires a démontré la présence des phosphates de calcium sous forme d'hydroxyapatite [Ca5(PO4)3(OH)]. Cette précipitation est potentiellement induite par les variations locales de pH et de sursaturation provoqués par les réactions microbiennes à l'intérieur des granules. L'étude des phénomènes de biominéralisation à été étendu aux granules anaérobies issus des réacteurs de type UASB de l'industrie laitière. Un modèle physico-chimique sur les processus de précipitation sous forme matriciel sur AQUASIM®, couplé avec des bases de calcul de sursaturation (PHREEQC®), ont permis d'avancer des hypothèses sur les mécanismes influençant ces processus de biominéralisation, tels que la formation d'un précurseur amorphe de l'hydroxyapatite (ACP), ainsi que d'identifier les constantes de précipitation thermodynamiques (pKsp|20ºC=28.07±0.58) et cinétiques dans différentes conditions opératoires. Grâce au suivi d'un système biologique GSBR (Granular Sludge Sequenced Batch Reactor) pendant plus de 900 jours, la contribution de ce phénomène aux processus de déphosphatation a été estimé (46% dans les conditions testées). L'utilisation de ce processus pour immobiliser efficacement le phosphore et apporter des propriétés physiques stables aux granules a été également discutée. Une évaluation des performances et de la stabilité du réacteur à été mis en œuvre en alternant des cycles anoxies/aérobies ou anaérobies/aérobies vis-à-vis d'une future application industrielle. L'induction locale de la précipitation par les variations de pH et par le relargage des phosphates par les réactions microbiennes, nécessite une modélisation appropriée, qui a été également initiée dans cette thèse / Over the last decade, aerobic granulation processes have araised as a promising technology for treating wastewater effluents containing high nitrogen, phosphorus and carbon concentrations. The microbial complexity of granules and the mechanisms by which they acquire excellent settleability properties, still constitute important research goals to investigate. This thesis is focused on a mechanism that has been little addressed in literature, that is, phosphate precipitation in the core of aerobic granules. Different analytical techniques, sometimes adapted for the first time to this type of systems, like Raman spectroscopy, have let an exhaustive characterization of biominerals in the core of granules. Analyses performed on aerobic granules grown with synthetic fed in a lab-scale SBR (Sequential Batch Reactor), revealed a calcium phosphate core made of hydroxyapatite [Ca5(PO4)3(OH)]. This precipitation phenomenon is induced by local pH and supersaturation gradients issued of biological reactions inside granules. The study of the biomineralization phenomenon has been extended into anaerobic granules coming from UASB reactors at different cheese wastewater treatment plants. A physico-chemical model has been described in a form of matrix with AQUASIM® software, and coupled with a thermodynamic database (PHREEQC®), in an attempt to hypothesize the mechanisms that influence the biomineralization phenomena. It has been proposed the formation of an amorphous precursor (ACP) prior hydroxyapatite precipitation in the core of granules, suggesting the thermodynamic constant (pKsp|20ºC=28.07±0.58) and kinetic constants at different operating conditions. It has been also estimated the contribution of the biomineralization to the overall phosphorus removal process (up to 46% at the operating conditions tested), thanks to the development and study of a GSBR (Granular Sludge Batch Reactor) in labscale, for more than 900 days. The fate of the biomineralization process in granules, regarding the contribution to their stabilization and physical properties, has been also dealt in this thesis. The reactor stability and performances have been assessed by alternating anoxic/aerobic and anaerobic/aerobic cycles, in sights of a future industrial application. The induction of precipitation by local variation of pH and supersaturation issued of biological reactions has been here introduced, although it will need further investigation

Apesar de ser utilizado na indústria de Fosfato Bicácico (DCP) há vários anos, a microgranulação a partir de misturados horizontais de alto cisalhamento possui fundamentos pouco conhecidos, de maneira que as operações das unidades industriais sejam baseadas em conhecimentos empíricos. Este trabalho busca determinar os mecanismos de granulação associados com o processo de microgranulação de DCP e o perfil de crescimento dos grãos em função dos seguintes parâmetros operacionais: a quantidade de fase líquida, acidez do meio reacional e a velocidade de rotação do eixo central do granulador. Após a análise estatística de dados coletados a partir de um ano de operação em uma unidade industrial, conclui-se que a aglomeração do DCP em ambientes com alta umidade geram preferencialmente partículas grosseiras, que não contribuem para o aumento da eficiência de granulação. O processo de microgranulação de DCP deve ser controlado em uma estreita faixa de condições operacionais, balanceando a quantidade de fase líquida adicionada e a taxa de reciclo de material seco praticada. A redução na umidade de operação, aumenta a qualidade granulométrica do material produzido por este processo. / Despite being used by the Dicalcium Phosphate (DCP) Industry for many years, the fundaments of high shear horizontal mixer microgranulation are poorly known, so the industrial operations are based on empirical knowledge. The goal of this work was to determine the granulation mechanisms associated with the DCP microgranulation process and the particle size enlargement pattern, according with the following operational parameters: the amount of liquid phase, reaction media acidity and the mixer paddles` rotational speed. After statistically analysing one year operational data collected from an industrial facility, it was concluded that the agglomeration of DCP at a high moisture environment promotes preferable formation of coarse particles, which dont increase the overall granulation efficiency. The DCP microgranulated process has to be controlled in very narrow operational condition, balancing the liquid phase and the recycle material returned during the granulation process. The reduction of the process moisture increased the quality of physical quality of the material produced by this process route.

Iron and steel slag valorization through carbonation and supplementary processes

Alkaline industrial wastes are considered potential resources for the mitigation of CO2 emissions by simultaneously capturing and sequestering CO2 through mineralization. Mineralization safely and permanently stores CO2 through its reaction with alkaline earth metals. Apart from natural formations, these elements can also be found in a variety of abundantly available industrial wastes that have high reactivity with CO2, and that are generated close to the emission point-sources. Apparently, it is the applicability and marketability of the carbonated products that define to a great extent the efficiency and viability of the particular process as a point source CO2 mitigation measure. This project investigates the valorization of iron- and steel-making slags through methods incorporating the carbonation of the material, in order to achieve the sequestration of sufficient amounts of CO2 in parallel with the formation of valuable and marketable products. Iron- and steel-manufacturing slags were selected as the most suitable industrial byproducts for the purposes of this research, due to their high production amounts and notable carbonation capacities. The same criteria (production amount and carbonation capacity) were also used for the selection of the iron- and steel-making slag types that are more suitable to the scope of this work. Specifically for the determination of the slag types with the most promising carbonation capacities, the maximum carbonation conversions resulting from recent publications related to the influence of process parameters on the conversion extent of iron- and steel-manufacturing slags, were directly compared to each other using a new index, the Carbonation Weathering Rate, which normalizes the results based on particle size and reaction duration. Among the several iron- and steel-manufacturing slags, basic oxygen furnace (BOF) and blast furnace (BF) slags were found to combine both high production volumes and significant affinity to carbonation. In the context of this research, two different procedures aiming to the formation of value added materials with satisfactory CO2 uptakes were investigated as potential BF and BOF slags valorization methods. In them, carbonation was combined either with granulation and alkali activation (BOF slag), or with hydrothermal conversion (BF slag). Both treatments seemed to be effective and returned encouraging results by managing to store sufficient amounts of CO2 and generating materials with promising qualities. In particular, the performance of the granulation-carbonation of BOF slag as a method leading to the production of secondary aggregates and the sequestration of notable amounts of CO2 in a solid and stable form, was evaluated in this work. For comparison purposes, the material was also subjected to single granulation tests under ambient conditions. In an effort to improve the mechanical properties of the finally synthesized products, apart from water, a mixture of sodium hydroxide and sodium silicate was also tested as a binding agent in both of the employed processes. According to the results, the granules produced after the alkali activation of the material were characterized by remarkably greater particle sizes (from 1 to 5 mm) compared to that of the as received material (0.2 mm), and by enhanced mechanical properties, which in some cases appeared to be adequate for their use as aggregates in construction applications. The maximum CO2 uptake was 40 g CO2/kg of slag and it was achieved after 60 minutes of the combined treatment of alkali activated BOF slag. Regarding the environmental behavior of the synthesized granules, increased levels of Cr and V leaching were noticed from the granules generated by the combination of granulation-carbonation with alkali activation. Nevertheless, the combination of granulation with alkali activation or that of granulation with carbonation were found not to worsen, if not to improve, the leaching behaviour of the granules with regards to the untreated BOF slag. The formation of a zeolitic material with notable heavy metal adsorption capacity, through the hydrothermal conversion of the solid residues resulting from the calcium- extraction stage of the indirect carbonation of BF slag, was also investigated in this project. To this end, calcium was selectively extracted from the slag by leaching, using acetic acid of specific concentration (2 M) as the extraction agent. The residual solids resulting from the filtration of the generated slurry were subsequently subjected to hydrothermal conversion in caustic solution of two different compositions (NaOH of 0.5 M and 2 M). Due to the presence of calcium acetate in the composition of the solid residues, as a result of their inadequate washing, only the hydrothermal conversion attempted using the sodium hydroxide solution of higher concentration (2 M) managed to turn the amorphous slag into a crystalline material, mainly composed by a zeolitic mineral phase (detected by XRD), namely, analcime (NaAlSi2O6·H2O), and tobermorite (Ca5(OH)2Si6O16·4H2O). Finally, the heavy metal adsorption capacity of the particular material was assessed using Ni2+ as the metal for investigation. Three different adsorption models were used for the characterization of the adsorption process, namely Langmuir, Freundlich and Temkin models. Langmuir and Temkin isotherms were found to better describe the process, compared to Freundlich model. Based on the ability of the particular material to adsorb Ni2+ as reported from batch adsorption experiments and ICP-OES analysis, and the maximum monolayer adsorption capacity (Q0 = 11.51 mg/g) as determined by the Langmuir model, the finally synthesized product can potentially be used in wastewater treatment or environmental remediation applications.

Avaliação do efeito do cloridrato de papaverina na reparação de feridas cirúrgicas abertas em dorso de ratos / Effect of papaverine hydrochloride in open surgical repair of wounds in rats

Os objetivos deste estudo foram avaliar o efeito da aplicação subcutânea do cloridrato de papaverina no processo de reparação de feridas cirúrgicas abertas no dorso de ratos e avaliar pelos métodos histomorfológico, histomorfométrico e imunoistoquímico eventos biológicos do processo de reparação. Foram realizadas feridas dérmicas padronizadas com punch, 5mm de diâmetro e 2mm de profundidade, no dorso de ratos. Os animais foram divididos em dois grupos conforme o tratamento realizado: no grupo controle, 25 ratos foram tratados através da injeção de cloreto de sódio 0,9% e no grupo teste, 25 ratos foram tratados através da injeção da solução de cloridrato de papaverina a 50mg/mL de cloreto de sódio a 0,9%. Em ambos os grupos foi aplicado subcutâneo (por quadrante da ferida), 0,25mL da solução correspondente aos frascos do grupo teste ou grupo controle totalizando 1 mL da solução. Os 50 espécimes foram processados para as análises macro e microscópica. Para análise do cálculo do edema, foram utilizados 10 ratos (5 animais do grupo teste e 5 animais do grupo controle). Fragmentos de pele padronizados (3cm2) foram removidos da área da ferida e pesados no período de 6 horas. Para análise morfométrica do fechamento da ferida/formação de cicatriz, 10 ratos foram utilizados (5 animais do grupo teste e 5 do grupo controle). As feridas cirúrgicas padronizadas foram fotografadas nos períodos de 0h, 3,7,14 e 21 dias pós-cirúrgico e as imagens foram analisadas por software de morfometria (ImageLab2000®) quanto a área, perímetro e fator de forma. Pela técnica de coloração da hematoxilina e eosina, a análise histomorfológica (análise qualitativa descritiva) e histomorfométrica (análise quantitativa em relação à reepitelização, formação do tecido de granulação, edema, celularidade e matriz colagênica) foram realizadas nos períodos de 6h, 3,7,14 e 21 dias e analisadas sob microscopia de luz. Reações de imunoistoquímica com o anticorpo anti-actina de músculo liso foi realizada para identificação e contagem do número de miofibroblastos nos períodos de 3, 7, 14 e 21 dias. A partir do modelo experimental avaliado, comprovou-se a ação vasodilatadora da papaverina. Os fragmentos padronizados do grupo teste apresentaram maior peso em relação ao grupo controle (p= 0,047). As feridas do grupo teste mostraram um fechamento maior e menor formação de cicatriz quando comparado ao grupo controle no período de 21 dias. No grupo teste, uma maior quantidade de edema (p = 0,028) e uma menor quantidade de matriz colagênica (colágeno) (p = 0,028) foram encontradas no período de 6 horas. Houve maior reepitelização no grupo teste no período de 7 dias e menor formação de tecido de granulação nos períodos de 14 e 21 dias para o mesmo grupo. No grupo controle, um maior número de miofibroblastos foi encontrado quando comparado ao grupo teste nos períodos de 14 e 21 dias (p=0, 016). Dentro dos limites deste estudo, a aplicação subcutânea do cloridrato de papaverina parece acelerar o processo de reparação de feridas cirúrgicas abertas no dorso de ratos. Sua aplicação promove vasodilatação e um maior exsudato inflamatório no início do processo de reparação. Tal efeito parece estar relacionado com a maior velocidade de reparação observada macro e microscopicamente. / The objectives of this study were to evaluate the effect of subcutaneous administration of papaverine hydrochloride in the process of open surgical repair of wounds in the back of rats and evaluate the methods histomorphological, immunohistochemical and histomorphometric biological events of the repair process. Standardized wounds were inflicted by dermal punch, 5mm in diameter and 2mm deep, in the backs of rats. Animals were divided into two groups according to treatment: control group, 25 rats were treated by injection of sodium chloride 0.9%) and test group (25 rats treated by the injection of papaverine hydrochloride 50 mg / mL sodium chloride 0.9%). In both groups was administered subcutaneously (by quadrant of the wound), 0.25 mL of the solution corresponding to bottles of the test group or control group, totaling 1 mL of the solution. The 50 specimens were processed for macro and microscopic analysis. For analysis of the calculation of edema, we used 10 rats (5 animals in the test group and 5 control animals). Standardized skin fragments (3cm2) were removed from the wound area and heavy during the 6 hours. For morphometric analysis of the closure of the wound / scar formation, 10 rats were used (five animals in the test group and 5 in the control group). Surgical wounds were photographed in standardized periods of 0h, 3,7,14 and 21 days after surgery and the images were analyzed by morphometry software (ImageLab2000 ®) as area, perimeter and form factor. By staining with hematoxylin and eosin, the histomorphologic analysis (descriptive qualitative analysis) and histomorphometric (quantitative analysis in relation to reepithelialization, formation of granulation tissue, edema, cellularity and collagen matrix) were recorded at 6h, 3.7 , 14 and 21 days and analyzed under light microscopy. Immunohistochemical reactions with anti-smooth muscle actin was performed to identify and count the number of myofibroblasts at 3, 7, 14 and 21 days. From the experimental model evaluated, proved the vasodilator papaverine. The fragments of the standardized test group had a higher weight in the control group (p = 0.047). The wounds of the test group showed a greater closing and less scarring compared to control group within 21 days. In the test group, a greater amount of edema (p = 0.028) and a smaller amount of collagen matrix (collagen) (p = 0.028) were found within 6 h. A greater reepithelialization in the test group after 7 days and less formation of granulation tissue during periods of 14 and 21 days for the same group. In the control group, a greater number of myofibroblasts was found when compared to the test group during the periods of 14 and 21 days (p = 0, 016). Within the limits of this study, subcutaneous administration of papaverine hydrochloride appears to accelerate the repair of surgical wounds opened in the back of rats. Its application promotes vasodilation and a greater inflammatory exudate in the early repair process. This effect appears to be related to a faster repair observed macroscopically and microscopically.

From the experimental model evaluated, proved the vasodilator papaverine. The fragments of the standardized test group had a higher weight in the control group (p = 0.047). The wounds of the test group showed a greater closing and less scarring compared to control group within 21 days. In the test group, a greater amount of edema (p = 0.028) and a smaller amount of collagen matrix (collagen) (p = 0.028) were found within 6 h. A greater reepithelialization in the test group after 7 days and less formation of granulation tissue during periods of 14 and 21 days for the same group. In the control group, a greater number of myofibroblasts was found when compared to the test group during the periods of 14 and 21 days (p = 0, 016). Within the limits of this study, subcutaneous administration of papaverine hydrochloride appears to accelerate the repair of surgical wounds opened in the back of rats. Its application promotes vasodilation and a greater inflammatory exudate in the early repair process. This effect appears to be related to a faster repair observed macroscopically and microscopically.

Sistemas monolíticos particulados contendo os constituintes ativos veiculados na forma de grânulos revestidos - pellets - têm recebido crescente atenção nos últimos anos, em função da otimização na biodisponibilidade e segurança na liberação do fármaco. A utilização destas unidades, como componentes de comprimidos traz, como principal vantagem, a divisibilidade da forma sem a perda do perfil biofarmacêutico desejado para o fármaco. Para sua produção, é indispensável a manutenção da integridade do revestimento daquelas unidades. Uma estratégia para o alcance deste objetivo envolve a utilização de grânulos inertes deformantes, comprimidos em conjunto com os grânulos revestidos, que atuam como um sistema de amortecimento das forças de compressão. Neste trabalho investigou-se a produção de grânulos deformantes através de dois métodos de granulação por via úmida, avaliando a influência de adjuvantes sobre as características dos produtos obtidos. Empregando a técnica de extrusão/esferonização obtiveram-se grânulos com propriedades de fluxo, empacotamento e resistência mecânica aceitáveis. O efeito dos adjuvantes sobre as etapas tecnológicas foi estudado por meio de um planejamento fatorial. Testaram-se duas variedades de celulose microcristalina, os desintegrantes croscarmelose sódica e crospovidona e soluções aglutinantes aquosas e hidroetanólicas de povidona. Para o desenvolvimento dos comprimidos utilizaram-se, como modelo, grânulos revestidos gastro-resistentes contendo omeprazol. A influência da composição dos grânulos deformantes sobre a liodisponibilidade do fármaco dos comprimidos foi avaliada através de análise fatorial 23. Os grânulos deformantes protegeram o revestimento polimérico dos pellets com diferentes intensidades. / Monolythic particulate systems containing the active constituents as coated pellets became great interest due to the improvement of safety and bioavailability. The use of such units as components of tablets shows as main advantages the divisibility of the pharmaceutical dosage form without loosing the desired biopharmaceutical profile of the drug. Consequently for the tablet production, the integrity of the polymeric film must be attained. A strategic option involves the utilization of inert soft pellets, which could be compressed together with the film coated pellets, absorbing the compaction forces. In this work the production of soft pellets was investigated using two wet granulation methods and evaluating the influence of formulation adjuvants on the pellets properties. The extrusion/spheronization technique yielded pellets with acceptable flow, packing and mechanical characteristics. The influence of the adjuvants on the technological steps was carried out through a statistical designed experiment. Microcrystalline cellulose from two producers, the disintegrants sodium croscarmellose and crospovidone, and aqueous and hydroethanolic dispersions of povidone, as binder, were tested. For the tablets development omeprazol gastroresistant film coated pellets were used as model. Aiming at the study of the influence of the soft pellets composition on drug lyoavailability was performed a 23 factorial experiment. The soft pellets protected at different intensities the polymeric coating of the gastroresistant pellets.

Avaliação da aspirina e do ácido cafeico fenetil estersobre o reparo tecidual cutâneo / Evaluation of aspirin and caffeic acid phenethyl ester on cutaneous wond healing

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Numerosos modelos in vitro e in vivo foram desenvolvidos para estudar o reparo de lesões e identificar os mecanismos chave deste processo. Visando avaliar o processo de cicatrização utilizamos um modelo de lesão excisional total e um modelo de queimadura promovida por escaldamento. No estudo utilizando o modelo de lesão excisional total, abordamos o uso da aspirina (um inibidor não seletivo da COX) e seu efeito diferenciado sobre os sexos na cicatrização cutânea de camundongos. Observamos que os grupos fêmea controle e tratado apresentaram contração atrasada comparado aos grupos macho controle e tratado, respectivamente. Entre os grupos fêmea e macho controles, as fêmeas apresentaram menor atividade da mieloperoxidase e menor quantidade de células MIF-positivas do que os machos controle. Já entre os grupos fêmea e macho tratados, foi observado que nas fêmeas tratadas, a atividade da mieloperoxidase e a quantidade de macrófagos F4/80-positivos estavam maiores do que no grupo macho tratado. Ainda entre os grupos tratados, as fêmeas apresentaram menores níveis de hidroxiprolina e maior expressão proteica de vWF e VEGF comparado aos machos. No estudo das lesões causadas por queimadura, avaliamos as propriedades anti- inflamatórias e antioxidantes do ácido cafeico fenetil ester (CAPE) no reparo destas lesões e observamos que em 7, 14, 21 e 70 dias após a queimadura, o grupo queimado+CAPE apresentou menor área lesada, além de menor atividade da mieloperoxidase e dos níveis de nitrito do que o grupo queimado. Também foi observado que no grupo queimado+CAPE a expressão proteica de CD68 e de PECAM-1 estava reduzida comparada ao grupo queimado. Analisando os parâmetros de dano oxidativo foi observado que os níveis de MDA e de proteínas carboniladas estavam menores no grupo queimado+CAPE do que no grupo queimado, tanto no plasma quanto na lesão. Em suma, nosso estudo avaliou o processo de cicatrização de dois modelos de lesão, em roedores de diferentes espécies e em abordagens distintas. No modelo de lesão excisional total em camundongos observamos que a administração da aspirina prejudicou o processo de cicatrização em camundongos Balb/c fêmeas, mas não afetou esse processo nos machos da mesma linhagem, o que pode ser explicado pela redução dos níveis de estrógenos nas fêmeas. A utilização do CAPE na cicatrização de queimaduras em ratas comprovou sua eficácia anti-inflamatória e antioxidante devido ao grupo queimado+CAPE ter apresentado maior taxa de cicatrização, redução da resposta inflamatória, além da redução do dano oxidativo em lipídeos e proteínas comparado ao grupo queimado. / Several in vitro and in vivo models have been developed to study wound healing and to identify key mechanisms of this process. The most of these studies use animals models to reproduce the human physiology and possible therapeutic treatments. In order to evaluate the wound healing process in different wound models, we use a full-thickness excisional wound model and a burn model promoted by scalding. In the full-thickness excisional wound study, we approach the aspirin use (COX non-selective inhibitor) and its distinct effect on the gender in cutaneous wound healing on mice. It was observed that both female control and treated groups presented smaller wound area than male control and treated groups, respectively. Between female and male control groups, the females presented smaller myeloperoxidase activity and MIF-positive cells than control males. Comparing female and male treated groups it was observed that in female treated group, the myeloperoxidase activity and the F4/80-positive macrophages amount were greater than male treated group. Also between the treated groups, the females presented smaller hydroxyproline levels and greater vWF and VEGF protein expression compared to the males. In study of the burns performed by scalding, it was evaluated the caffeic acid phenethyl ester (CAPE) anti-inflammatory and antioxidant properties on repair of these lesions. It was observed that at 7, 14, 21 and 70 days after burning, the burn+CAPE group presented smaller wound area, beyond smaller myeloperoxidase activity and nitrite levels than burn group. It was also observed that the burn+CAPE group presented smaller CD68 and PECAM-1 protein expression compared to burn group. Analyzing the oxidative damage parameters, it was observed that the MDA and carbonilated proteins levels were greater in the burn group compared to burn+CAPE group. In conclusion, our study evaluated the wound healing process in 2 wound models on distinct rodent species with distinct approaches. In mice full-thickness wound model, under gender and aspirin action influence, it was seen that the aspirin administration impaired the wound healing process on Balb/c female mice, but did not alter the wound repair on males of the same lineage, what can be explained by reduction of the estrogen levels in these females. The CAPE use on rat burning healing demonstrated its anti-inflammatory and antioxidant efficiency because the burn+CAPE group presented greater burning healing, smaller inflammatory response, beyond smaller oxidative damage at lipid and protein level compared to burn group

Apesar de ser utilizado na indústria de Fosfato Bicácico (DCP) há vários anos, a microgranulação a partir de misturados horizontais de alto cisalhamento possui fundamentos pouco conhecidos, de maneira que as operações das unidades industriais sejam baseadas em conhecimentos empíricos. Este trabalho busca determinar os mecanismos de granulação associados com o processo de microgranulação de DCP e o perfil de crescimento dos grãos em função dos seguintes parâmetros operacionais: a quantidade de fase líquida, acidez do meio reacional e a velocidade de rotação do eixo central do granulador. Após a análise estatística de dados coletados a partir de um ano de operação em uma unidade industrial, conclui-se que a aglomeração do DCP em ambientes com alta umidade geram preferencialmente partículas grosseiras, que não contribuem para o aumento da eficiência de granulação. O processo de microgranulação de DCP deve ser controlado em uma estreita faixa de condições operacionais, balanceando a quantidade de fase líquida adicionada e a taxa de reciclo de material seco praticada. A redução na umidade de operação, aumenta a qualidade granulométrica do material produzido por este processo. / Despite being used by the Dicalcium Phosphate (DCP) Industry for many years, the fundaments of high shear horizontal mixer microgranulation are poorly known, so the industrial operations are based on empirical knowledge. The goal of this work was to determine the granulation mechanisms associated with the DCP microgranulation process and the particle size enlargement pattern, according with the following operational parameters: the amount of liquid phase, reaction media acidity and the mixer paddles` rotational speed. After statistically analysing one year operational data collected from an industrial facility, it was concluded that the agglomeration of DCP at a high moisture environment promotes preferable formation of coarse particles, which dont increase the overall granulation efficiency. The DCP microgranulated process has to be controlled in very narrow operational condition, balancing the liquid phase and the recycle material returned during the granulation process. The reduction of the process moisture increased the quality of physical quality of the material produced by this process route.
