Patogenia experimental da infecção pelo herpesvírus bovino tipo 2 em ovinos e cobaias / Experimental pathogenesis of bovine herpesvirus Type 2 infection in sheep and guinea pigs

The biology and epidemiology of bovine herpesvirus 2 (BoHV-2), the agent of bovine herpetic mammillitis (BHM), remain largely unknown. Thus, the present study aimed at addressing selected aspects of BoHV-2 epidemiology and pathogenesis in sheep and guinea pigs. A serological survey for BoHV-2 antibodies in 2.213 samples from cattle > 24-months-old from 136 counties of seven different mesoregions of Rio Grande do Sul (RS) revealed an overall prevalence of 24.5% (543/2.213). These results demonstrate that BoHV-2 infection is widespread among cattle in RS, and potentially involved in cases of mammillitis frequently described in dairy cows. A second experiment was conducted to characterize the latent BoHV-2 infection in a sheep model. Lactating ewes inoculated with BoHV-2 in the skin of the udder shed virus for up to five days, developed mammillitis and seroconverted. However, attempts to reactivate latent infection by dexamethasone (Dx) administration at day 40 post infection (pi) failed. Nevertheless, viral DNA was detected by PCR in several nerve ganglia and/or regional lymph nodes (LNs) of all animals at day 40 post-reactivation (pr). Lambs previously inoculated with BoHV-2 in the nose also harbored latent viral DNA in trigeminal ganglia, tonsils and regional LNs. These results demonstrated that BoHV-2 establishes latent infection in nerve ganglia and in regional lymphoid tissues, yet virus reactivation is not easily achieved by standard protocols used for other alphaherpesvirus. A third experiment was conducted to evaluate guinea pigs (Cavia porcellus) as an experimental model to study the biology of BoHV-2 infection. Weanling (30-40 daysold) guinea pigs inoculated into the genital area and over the skin of the udder and teats with a viral suspension developed moderate to severe clinical signs, noticeably more pronounced in the genital area. Infectious virus was recovered from swabs collected between days 3 and 7 from genital area (8/12) and less frequently (1/12) from teat skin. All animals seroconverted to BoHV-2 developing neutralizing titers from 16 to 128 at day 30 pi. Histological examination of skin biopsies collected from genital lesions showed intranuclear inclusion bodies and perivascular infiltrates composed by lymphocytes and plasmocytes. PCR examination of tissues collected from animals euthanized at day 35 pi revealed the presence of latent viral DNA in nerve ganglia and LNs. Dx administration at day 35 pi was followed by mild recrudescence of genital disease in some animals, yet virus isolation and/or seroconversion which are usually taken as indicators of virus reactivation were not observed. These results show that guinea pigs may be used as models to study BoHV-2 acute and latent infection and confirm that BoHV-2 reactivation is not easily achieved by using the standard protocols for other alphaherpesviruses. Taken together, these results demonstrate that both sheep and guinea pigs are suitable animal models for BoHV-2 infection. / A biologia e a epidemiologia da infecção pelo herpesvírus bovino tipo 2 (BoHV-2), agente da mamilite herpérica bovina, são pouco conhecidas. Com isso, o presente estudo teve como objetivos elucidar alguns aspectos da epidemiologia e estudar a patogenia da infecção pelo BoHV-2 em ovinos e cobaias. Um estudo sorológico pesquisando anticorpos para o BoHV-2 em 2.213 amostras de bovinos com mais de 2 anos, provenientes de 136 municípios distribuídos em 7 mesoregiões do Rio Grande do Sul (RS), determinou uma soroprevalência de 24,5% (543/2.213). Os resultados demonstram que o BoHV-2 está disseminado no rebanho bovino no RS, com potencial envolvimento em casos de mamilite freqüentemente relatados em rebanhos leiteiros. Um segundo experimento visou caracterizar a infecção latente em ovinos, propostos como modelo experimental. Ovelhas lactantes inoculadas com o BoHV-2 na pele do úbere excretaram o vírus por até 5 dias, desenvolveram mamilite e soroconverteram. Entretanto, a tentativa de reativar a infecção latente pela administração de dexametasona (Dx) no dia 40 pós-infecção (pi) não resultou em reativação. Não obstante, o DNA viral latente foi detectado por PCR em gânglios neurais e/ou em diversos linfonodos (LNs) de todos os animais no dia 40 pós reativação (pr). Cordeiros que haviam sido inoculados com o BoHV-2 pela via nasal igualmente albergavam o DNA viral latente nos gânglios trigêmeos, tonsilas e LNs regionais. Esses resultados demonstram que o BoHV-2 estabelece infecção latente em gânglios neurais e tecidos linfóides regionais, embora a reativação viral não seja facilmente obtida com os protocolos usados em outros alfaherpesvírus. Um terceiro experimento foi realizado para validar cobaias (Cavia porcellus) como modelo experimental para estudo da biologia da infecção com o BoHV-2. Cobaias lactentes (30- 40 dias) inoculadas com o BoHV-2 nas regiões genital e intra-dérmico no tetos e úbere desenvolveram sinais clínicos moderados a severos, notadamente mais pronunciados na região genital. O vírus foi isolado de suabes coletados entre os dias 3 e 7 pi da região genital (8/12) e em menor freqüência da região dos tetos (1/12). Todos os animais soroconverteram para o BoHV-2, apresentando títulos de anticorpos neutralizantes entre 16 e 128 no dia 30 p.i. A análise histológica de biópsias de pele coletada de lesões revelou corpúsculos de inclusão e infiltrado linfocitário e plasmocitário perivascular. PCR realizada com DNA total extraído dos tecidos coletados no dia 35 pi revelou a presença de DNA viral latente em gânglios neurais lombo-sacrais e LNs regionais. A administração de Dx no dia 35 pi cursou com recrudescência clínica na região genital, porém não foram detectados excreção viral nem soroconversão, indicadores de reativação da infecção latente. Estes resultados demonstram que cobaias podem ser usados como modelo para a infecção aguda e latente pelo BoHV- 2, e confirmam que a reativação viral não é facilmente obtida pelo uso de protocolos usuais para outros alfaherpesvírus. Em conjunto, os resultados demonstram a utilidade de ovinos e cobaias como modelo experimental para a infecção pelo BoHV-2.

From leaves of these species was extract the essential oil (XF-OE), that in previous studies, showed spasmolytic activity on phasic contractions induced by CCh or histamine on guinea pig ileum. Considering this premise, the aim of this study was to characterize the mechanism of spasmoliytic action of XF-OE on intestinal smooth muscle. For this, were used functional and cellular methodologies. XF-OE inhibited cumulative concentration-response curves to histamine, and these were shifted to the right, in a non-parallel manner, with Emax reduction, discarding thus a competitive type antagonism and relaxed the guinea pig ileum contracted by KCl (40 mM) or histamine (10-6 M). How the relaxant potency of the essential oil was smaller in organ pre-contracted by KCl, it was hypothesized that XF-OE would be acting as a K+ channels positive modulator. In presence of CsCl (5 mM), a non-selective blocker of these channels, the relaxant potency of XF-OE was not altered, showed a non-participation of K+ channels on the essential oil spasmolytic effect. Thus, we decided to check whether the essential oil would prevent calcium influx through CaV, for this were obtained cumulative concentration-response curves to CaCl2 in depolarizing medium (70 mM KCl) nominally without Ca2+ in absence (control) and presence of different concentrations of XF-OE. The essential oil shifted to the right CaCl2 control curves, with reduction of its Emax, in addition to relaxed the ileum pre-contracted by S-(-)-Bay K8644 (3 x 10-7 M), a CaV1 selective agonist, demonstrating that the essential oil inhibited Ca2+ influx through CaV1. As the Ca2+ mobilization mechanisms into circular and longitudinal muscle layers are differentiated, it was hypothesized that XF-OE would preventing Ca2+ mobilization from intracellular stores in order to promote its spasmolytic effect. To test this hypothesis, in experiments with ileum circular layer, the essential oil antagonized phasic contractions induced by histamine (10-5 M), negatively modulates the Ca2+ release to exert their spasmolytic effect. In cellular experiments, the viability of longitudinal layer myocytes from guinea pig ileum was not altered in XF-OE (81 μg/mL) presence and the fluorescence intensity in these intestinal myocytes stimulated by histamine was reduced by the essential oil, indicating a [Ca2+]c reduction. Thus, the XF-OE spasmolytic action mechanism on guinea pig ileum involves blocking the Ca2+ influx, by CaV1, in addition to negative modulation of Ca2+ release mechanisms from intracellular stores, that lead to reduction of this ion cytosolic levels and consequently muscle relaxation. / A espécie Xylopia frutescens Aubl., conhecida popularmente como “embira”, “semente-de-embira” e “embira-vermelha”, é utilizada na medicina popular como antidiarreica. Das folhas dessa espécie foi extraído o óleo essencial (XF-OE), que, em estudos anteriores, apresentou atividade espasmolítica frente às contrações fásicas induzidas por CCh ou por histamina em íleo de cobaia. Diante dessa premissa, o objetivo desse trabalho foi caracterizar o mecanismo de ação espasmolítica do XF-OE em musculatura lisa intestinal. Para isso, foram utilizadas metodologias funcionais e celulares. O XF-OE inibiu as curvas concentrações-resposta cumulativas à histamina, desviando-as para a direita, de maneira não paralela e com redução do seu Emax, descartando um antagonismo do tipo competitivo, além de relaxar o íleo pré-contraído com KCl (40 mM) ou com histamina (10-6 M). Como a potência relaxante do óleo essencial foi menor quando as contrações foram induzidas por KCl, hipotetizou-se que o XF-OE estaria atuando como um modulador positivo de canais de K+. Na presença do CsCl (5 mM), bloqueador não seletivo desses canais, a potência relaxante do XF-OE não foi alterada, sendo descartada a participação dos canais de K+ no efeito espasmolítico do óleo essencial. Diante disso, decidiu-se verificar se o óleo essencial estaria inibindo o influxo de cálcio através dos CaV, para isso foram obtidas curvas concentrações-resposta cumulativas ao CaCl2 em meio despolarizante (KCl 70 mM) nominalmente sem Ca2+ na ausência (controle) e na presença de diferentes concentrações do XF-OE. O óleo essencial deslocou para a direita as curvas controle de CaCl2 com redução do seu Emax, além de relaxar o íleo pré-contraído com o S-(-)-Bay K8644 (3 x 10-7 M), um agonista dos CaV1, demonstrando que o óleo essencial inibe o influxo de Ca2+ através dos CaV1. Sendo os mecanismos de mobilização de Ca2+ em células musculares das camadas circular e longitudinal diferenciados, hipotetizou-se que o XF-OE estaria impedindo a mobilização de Ca2+ dos estoques intracelulares para promover seu efeito espasmolítico. Para testar essa hipótese, em experimentos utilizando a camada circular do íleo, o óleo essencial antagonizou as contrações fásicas induzidas por histamina (10-5 M), modulando negativamente a liberação de Ca2+ para exercer seu efeito espasmolítico. Nos experimentos celulares, a viabilidade dos miócitos da camada longitudinal do íleo de cobaia não foi alterada na presença do XF-OE (81 μg/mL) e a intensidade de fluorescência nos miócitos intestinais estimulados por histamina foi reduzida pelo óleo essencial, indicando a redução na [Ca2+]c. Assim, o mecanismo de ação espasmolítica do XF-OE em íleo de cobaia envolve bloqueio do influxo de Ca2+, via CaV1, além da modulação negativa dos mecanismos de liberação de Ca2+ dos estoques intracelulares, levando à redução dos níveis citosólicos desse íon e, consequente, relaxamento muscular.

Croton nepetaefolius Baill à um arbusto aromÃtico do Nordeste brasileiro, conhecido como âmarmeleiro sabiÃâ, utilizado na medicina popular como antiespasmÃdico e carminativo. Os principais constituintes do seu Ãleo essencial sÃo 1,8-cineol, metil-eugenol, xantoxilina e terpineol. Recentemente, demonstraram-se propriedades antiespasmÃdica intestinal, hipotensiva, antiinflamatÃria e analgÃsica para o Ãleo essencial do Croton nepetaefolius (OECN). Neste trabalho, caracterizamos os efeitos farmacolÃgicos do OECN sobre o mÃsculo liso respiratÃrio e vascular de rato e cobaio e sobre o funcionamento elÃtrico de neurÃnios de gÃnglio celÃaco de cobaio. Usamos preparaÃÃes in vitro de vasos sanguÃneos de ratos e cobaios machos (aorta e vasos mesentÃricos) e anÃis de traquÃia, mantidos em soluÃÃo nutridora, aerada, pH 7,4, a 37 oC, para registro isomÃtrico das contraÃÃes musculares. Usamos tambÃm gÃnglios celÃacos intactos de cobaios, mantidos em temperatura ambiente in vitro, para os registros eletrofisiolÃgicos pela tÃcnica do microeletrodo intracelular. In vivo, avaliamos as aÃÃes do OECN sobre a pressÃo arterial mÃdia e alguns parÃmetros cardÃacos, respiratÃrios e hematolÃgicos em ratos. O OECN (0,1 - 1000 microgrma/ml) relaxou o tÃnus basal (5 mM [K+]) e o tÃnus aumentado por K+ (60 mM) em traquÃia de cobaio, de maneira dependente de concentraÃÃo (CE50 de 4 e 63 micrograma/ml, respectivamente). Em tecidos de cobaios previamente sensibilizados, o OECN (300 e 600 micrograma/ml), reduziu a contraÃÃo induzida pela apresentaÃÃo do antÃgeno sensibilizante (ovalbumina). Na faixa de concentraÃÃo de 100 a 400 micrograma/ml, bloqueou as contraÃÃes induzidas por histamina e PGF2 alfa. O OECN inibiu as contraÃÃes induzidas por histamina, carbacol e KCl com CI50 na faixa de 100 - 130 micrograma/ml. Em aorta de rato e cobaio, relaxou a contraÃÃo induzida por 60 mM de K+ (CI50 de 32 e 200 micrograma/ml, respectivamente). Em rato, mas nÃo em cobaio, este relaxamento foi parcialmente inibido pela retirada do endotÃlio vascular ou pela adiÃÃo de 100 micrograma M de L-NAME. Em aorta de cobaio, o OECN inibiu as contraÃÃes independentes de Ca2+, induzidas por dibutirato de forbol e por K+ hiperosmolar na soluÃÃo nutridora. O OECN diminuiu preferencialmente a pressÃo arterial em ratos DOCA-sal do que em ratos nefrectomizados e, bloqueou mais potentemente as contraÃÃes induzidas pela fenilefrina em aorta de rato DOCA do que dos animais nefrectomizados. O OECN, metil-eugenol e terpineol, aumentam o fluxo de lÃquido pela circulaÃÃo mesentÃrica de rato, sendo este efeito parcialmente inibido pela presenÃa de L-NAME. Em nenhuma dessas preparaÃÃes o OECN produziu hiperpolarizaÃÃo do potencial transmembrana. Em neurÃnios fÃsicos de gÃnglio celÃaco de cobaio, o OECN diminuiu significativamente o aumento da excitabilidade produzido pela histamina sem alterar as propriedades passivas e ativas dos neurÃnios. Em conclusÃo, o OECN relaxa o mÃsculo liso das vias aÃreas, à um agente hipotensor e vasorelaxante e diminui a excitabilidade induzida por histamina em neurÃnios autonÃmicos. Seus efeitos sÃo, provavelmente, intracelulares ou mediados por proteÃna quinase C. / Croton nepetaefolius is an aromatic bush found in brazilian Northeast region, called âmarmeleiro sabiÃâ, and it is used in folk medicine as an antispasmodic and carminative agent. Its essential oil is comprised of 1,8-cineole, methyl-eugenol, xanthoxylin, terpineol and others constituents. Recent studies showed some pharmacological activities of the essential oil of Croton nepetaefolius (EOCN) as an intestinal antispasmodic, hypotensive, anti-inflammatory and analgesic agent. Our aim in this work was to evaluate the effects of EOCN on airway and vascular smooth muscle and also on autonomic neurons. We used in vitro models of rat and guinea-pig isolated vessels and guinea-pig tracheal rings for isometric recording of the smooth muscle contractions. Guinea-pig celiac ganglion, was used for intracellular microelectrode recording of electricophysiological signals. Moreover, mean arterial pressure, cardiovascular, respiratory and hematologic parameters were measured in vivo in rats. EOCN (0,1 â 1000 microgram/ml) relaxed basal and K+-increased guinea-pig tracheal tonus (EC50 = 4 and 63 microgram/ml, respectively), in a concentratation-dependent manner. In ovalbumin-sensitized guinea-pig tissues, EOCN inhibited the antigen-induced contraction. EOCN (100 - 400 microgram/ml) blocked the histamine- and PGF2alpha -induced contractions. The contractions induced by histamine, carbacol and KCl were inhibited by EOCN with IC50s between 100-130 microgram/ml. In rat and guinea-pig aortic rings, EOCN relaxed the 60 mM K+-induced contractions (IC50 = 32 and 200 microgram/ml, respectively). Only in rat tissues, this EOCN-induced relaxation was partially inhibited by both L-NAME (100 microgram M) or endothelium lack. In guinea-pig aortic rings, EOCN inhibited the Ca2+-independent phorbol esther- and hyperosmotic K+- induced contractions. EOCN, preferably, diminished the mean arterial pressure and inhibited the aortic rings phenylephrine-induced contractions in DOCA-salt treated rats rather than uninephrectomized rats. Both EOCN, methyl-eugenol and terpineol increased the flow through rat mesenteric bed. This effect was partially blocked by L-NAME (50 microgram M). EOCN did not produce hyperpolarization of the transmembrane potential. In celiac ganglion phasic neurons, EOCN signicantly inhibited the histamine-induced increase of the neuronal excitability. In conclusion, EOCN is an airway smooth muscle relaxant, hypotensor and vasorelaxant agent, and it is a blocker of the stimulant histamine activity on autonomic neurons. Its effects are, probably, mediated by an intracellular action or protein C kinase modulation.

Aspectos descritivos e quantitativos da anatomia macroscópica e microscópica do nervo vestíbulo-coclear de cobaias / Descriptive and Quantitative Aspects of macroscopic and microscopic anatomy of the vestibulocochlear nerve of guinea pigs

O nervo vestíbulo-coclear da cobaia possui peculiaridades não encontradas em outros nervos periféricos. Não foram encontradas informações detalhadas sobre os aspectos morfométricos do VIII nervo craniano em cobaias adultas na literatura. A avaliação descritiva e quantitativa no presente estudo, evidencia informações que precedem o estudo das alterações que ocorrem em modelos experimentais de neuropatias do VIII nervo craniano e as doenças ou lesões que possam afetar o homem. Foram utilizadas 8 cobaias adultas, perfundidas com glutaraldeído a 2,5% em tampão cacodilato de sódio (0,025 M). Seus nervos direito e esquerdo (n=6, 4 nervos direito e 2 esquerdos) foram dissecados na região mediana do nervo e pós fixado com tetróxido de ósmio (OsO4) a 1% e incluídos em resina epóxi Poly/Bed 812®. Os fragmentos foram cortados em secções transversais semifinas seriadas (6 nervos) com uma espessura de 0,5 μm e corados com azul de toluidina para a microscopia de luz. Foram analisados os aspectos histológicos descritivos e topográficos do nervo vestíbulo-coclear em cobaias adultas, bem como os aspectos histométricos na parte mediana do nervo, no referente a densidade de fibras mielínicas, a distribuição dos diâmetros de tal tipo de fibras mielínicas, ao cálculo da razão G e o número de vasos sangüíneos encontrados dentro da área de cada nervo. O nervo coclear é envolvido pelo vestibular na porção inicial da junção de ambos. O conjunto, nervos coclear e vestibular e nervo vestíbulo-coclear têm a aparência bifurcada da letra Y, na horizontal com os ramos lateralizados, possuindo em média 5 mm de comprimento. É ricamente vascularizado e documenta uma completa interposição das fibras dos nervos coclear e vestibular, quando o fascículo do VIII nervo está completamente constituído. As suas fibras mielínicas dispõem-se paralelamente em seu sentido longitudinal e são circundadas por escasso tecido endoneural. Não foram evidencias fibras amielínicas no VIII nervo. A distribuição das fibras mielínicas no segmento mediano do VIII nervo craniano foi unimodal, havendo predomínio de fibras com 3,5 m de diâmetro. O número de fibras foi de 5.390 ± 1.504 fibras. O diâmetro das fibras variou de 1,5 a 6 m de diâmetro. Os axônios com 2,5 m de diâmetro foram predominantes quantitativamente: 6.757 ± 1.922 axônios. O diâmetro axonal variou entre 1 a 4,5 m. A densidade média das fibras do VIII nervo em seu segmento mediano foi de 41.474 ± 4384 fibras/mm2. O diâmetro fascicular foi de 0,77 ± 0,04 mm. A razão G varia de 0,2 a 0,9. O maior número de fibras tem razão G de 0,7 e 0,6, respectivamente, predominando as fibras com 0,7. Os resultados descritos neste trabalho científico são originais, contendo alguns resultados inéditos e pouco estudados na literatura científica sobre a anatomia e morfometria do VIII nervo craniano em cobaia. O nervo vestíbulo-coclear da cobaia é um nervo sensitivo aferente especial, e bem diferente em sua anátomo-fisiologia de todos os outros nervos sensitivos e motores encontrados nos mamíferos. A cobaia adulta é um excelente modelo experimental para o estudo do nervo vestíbulo-coclear, semelhantes aos de humanos. / The vestibulocochlear (VIIIth cranial nerve) nerve of guinea pigs have special features not common for peripheral nerves in general. There are no detailed reports on the morphometric characteristics of the VIII nerve in adult guinea pigs. This is a descriptive and qualitative study that shows normal parameters that are useful in experimental models of the VIIIth nerve neuropathy. Eight adult guinea pigs were perfused with 2.5% glutaraldehyde in isotonic cacodilate buffer (0.025M). The right (N=4) and left (N=2) nerves were dissected in the median region, post-fixed in 1% osmuin tetroxide and embedded in epoxy resin. Semithin (0.5 µm) serial transverse sections were stained with 1% toluidine blue for light microscopy study. The descriptive histology and the topographical and histometric aspects of the median region of the nerve were analyzed, including myelinated fiber density, myelinated fiber diameter distribution, g ratio and number of endoneural capillary vessels. The cochlear nerve is enveloped by the vestibular nerve when they join together. Both, the cochlear and the vestibular nerves join to form the vestibulocochlear nerve with a Y shape, with approximately 5 mm in length. The nerve is widely vascularized and presents a complete mixture of the vestibular and cochlear myelinated fibers when the VIII nerve fascicle is constituted. The myelinated fibers are longitudinally oriented and present few endoneural connective tissue in between. No unmyelinated fibers were evidenced in the VIII nerve. The myelinated fiber diameter distribution was unimodal, with a peak at 3.5, and intervals between 1,5 and 6 m. The average number of fibers was 5.390 ± 1.504. The myelinated axon diameter distribution was also unimodal, with a peak at 2.5 µm of diameter were predominant quantitatively: 6.757 ± 1.922, and intervals between 1 and 4.5 µm. The average myelinated fiber density was 41.474 ± 4384 mm2, spread in a fascicular diameter of 0.77 ± 0.04 mm. The g ratio values varied from 0.2 to 0.9 and most of the myelinated fibers showed g ratio values of 0.7. Our results are original and only few parameters studied here were described in the literature. The vestibulocochlear nerve of the guinea pigs is a special afferent nerve, differing from other peripheral nerves (sensitive and motor) in its main anatomic and physiological characteristics, compared to other nerves from mammals. The adult guinea pig is an excellent model for the vestibulocochlear nerve neuropathy studies once it is similar to the human nerve.

Modulation of serous salivary gland function by the sympathetic nervous system : a biochemical and ultrastructural study with special reference to β-adrenoceptor subtypes

The aim of the present investigation was to study the influence of the sympathetic nervous system and of various adrenoceptor agents on enzyme secretion and morphology in rat parotid and guinea-pig submandibular glands. Biochemical methods were combined with electron microscopical techniques. Two different in vitro systems were employed, batch-incubation and microperifusion, to characterize the sympathetically evoked amylase release and its correlation to cyclic AMP. By using various selective β-adrenoceptor agonists and antagonists a dominance of the β1-adrenoceptor over the β2 - in regulating amylase release - was establ ished. Continuous noradrenaline perifusion caused a rapid initial amylase discharge, closely correlated to tissue levels of cyclic AMP; no correlation between the two was observed during the later phase. Prenalterol (a β1-agonist) failed to elevate glandular cyclic AMP. This was in contrast to its potent secretagogic effect. On the other hand, terbutaline (a β2-agonist) was a weak secretagogue but markedly raised the levels of cyclic AMP. Thus, β-adrenoceptor activation may lead to release of large amounts of amylase despite minimal or no increase in cyclic AMP. Moreover, these effects seemed to be dissociated in salivary glands with regard to the β-adrenoceptor subtypes. This was further substantiated by the findings that repeated injections of prenalterol induced qualitative changes in the granule populations, similar to those caused by the non-selective β-agonist isoprenaline. Terbutaline was without effect. However, acinar cells size was increased following both prenalterol and terbutaline treatment. The data suggest that the 3-adrenergic effects on acinar cell size and granule population may be independently regulated. A decreased sympathetic activity of long duration was induced by neonatal or adult extirpation of the superior cervical ganlion on one side. Acinar cell size, as well as granule and amylase content was reduced 9 weeks after neonatal denervation. Ganglionectomy performed in adult animals was without significant effects. The secretory behaviour of neonatally denervated glands was characterized by an increased postjunctional sensitivity to 3-adrenoceptor agonists. Of special interest was the finding that neonatal denervation seemed to transform terbutaline from a partial to a full secretory agonist, thus changing its effects in the direction of those of prenalterol and noradrenaline. Moreover, increased levels of cyclic AMP as well as an enhanced response to DBcAMP were noted in the denervated glands as were intracellular changes. The denervation supersensitivity after neonatal denervation seems to differ from that observed in adult denervated glands. Cryoconservation des ressources génétiques chez le cochon d’Inde (Cavia porcellus) : production et congélation des embryons / Cryopreservation of guinea pig (Cavia porcellus) genetic resources : embryos production and freezing

La conservation de la biodiversité génétique du cochon d’Inde (Cavia porcellus) est importante en tant qu’animal de consommation endémique de la région andine ainsi qu’en tant qu’animal modèle précieux pour la recherche biomédicale.L’objectif de ce travail était de mettre en place les protocoles qui permettront une conservation ex situ des ressources génétiques de cette espèce par la voie femelle. Les étapes nécessaires à la concrétisation d’un projet de cryobanque sont :- la maîtrise de la production d’embryons grâce à des traitements hormonaux de synchronisation du cycle œstral et de superovulation des femelles,- la cryoconservation dans l’azote liquide des embryons obtenus et leur transfert dans des femelles receveuses.Une méthode standardisée de synchronisation des chaleurs basée sur l’administration d’altrenogest per os pendant 15 jours a été définie. Les chaleurs des femelles donneuses et receveuses d’embryons apparaissent dans les 4 à 5 jours qui suivent l’arrêt du traitement de ce progestagène.Des traitements de superovulation basés sur 3 injections à 24 heures d’intervalle d’hMG, ou 6 injections à 12 heures d’intervalle de FSH-recombinante humaine, permettent une augmentation du taux d’ovulation de l’ordre de fois 3 à fois 4 par rapport à des femelles non-traitées. Toutefois les réponses au traitement restent variables et il faudra réitérer l’expérience sur de plus grands lots d’animaux afin d’ajuster les doses et les moments d’application.La méthode de congélation lente, utilisant l’éthylène glycol comme cryoprotecteur, a permis d’obtenir des taux de survie embryonnaire satisfaisants (70,3%). La méthode de vitrification a également donné de bons résultats, avec un taux de survie embryonnaire de 41,7%.Le premier transfert au monde d’embryons frais réalisé avec succès dans une femelle receveuse préalablement synchronisée a été obtenu lors de ce travail. Les transferts d’embryons décongelés n’ont pas encore donné lieu à des gestations. / The preservation of the guinea pig (Cavia porcellus) genetic biodiversity is important as a native source of protein for many highlanders in the Andean region and as a precious laboratory animal for biomedical research.The aim of this work was to establish the protocols that will enable an ex situ preservation of the genetic resources of this species by the female way.The necessary steps for the achievement of a cryobank project are:- the control of the embryo production thanks to hormonal treatments to synchronize the estrous cycles and to superovulate the females,- the cryopreservation in liquid nitrogen of the obtained embryos and their transfer into recipient females.A standard method for synchronization of heat periods based on the per os administration of altrenogest during 15 days has been defined. The heat periods of the females, donors and recipients of embryos, appears 4 to 5 days after the end of the progestagen treatment. Superovulation treatments, based on 3 injections at 24-hour intervals of hMG or 6 injections at 12-hour intervals of human recombinant-FSH, lead to an increase of the ovulation rate of about 3 to 4 times when compared to untreated females. However, the responses to the treatments remain variable and further studies involving a larger number of animals should be carried out in order to adjust the dose and the moment of application of the treatment.The slow-freezing method, using ethylene glycol as cryoprotectant, enables the attainment of a satisfactory in vitro embryo survival rate (70,3%). The vitrification method also gives good results, with a 41,7% in vitro embryo survival rate.The first successful transfer of fresh embryos into a synchronized recipient female has been achieved in this study. The transfers of frozen-thawed embryos did not yet lead to gestation.

Localization of Cholinergic Innervation in Guinea Pig Heart by Immunohistochemistry for High-Affinity Choline Transporters

Objective: Previous studies have used acetylcholinesterase (AChE) histochemistry to identify cholinergic nerves in the heart, but this enzyme is not a selective marker for cholinergic neurons. This study maps cholinergic innervation of guinea pig heart using a new antibody to the human high-affinity choline transporter (CHT), which is present only in cholinergic nerves. Methods: Immunohistochemistry was used to localize CHTs in frozen and paraffin sections of heart and whole mount preparations of atrial ganglionated nerve plexus. AChE-positive nerve fibers were identified in sections from separate hearts for comparison. Results: Control experiments established that the antibody to human CHT selectively labeled cholinergic neurons in the guinea pig. CHT-immunoreactive nerve fibers and AChE-positive nerves were very abundant in the sinus and AV nodes, bundle of His, and bundle branches. Both markers also delineated a distinct nerve tract in the posterior wall of the right atrium. AChE-positive nerve fibers were more abundant than CHT-immunoreactive nerves in working atrial and ventricular myocardium. CHT-immunoreactive nerves were rarely observed in left ventricular free wall. Both markers were associated with numerous parasympathetic ganglia that were distributed along the posterior atrial walls and within the interatrial septum, including the region of the AV node. Conclusions: Comparison of labeling patterns for CHT and AChE suggests that AChE histochemistry overestimates the density of cholinergic innervation in the heart. The distribution of CHT-immunoreactive nerve fibers and parasympathetic ganglia in the guinea pig heart suggests that heart rate, conduction velocity, and automaticity are precisely regulated by cholinergic innervation. In contrast, the paucity of CHT-immunoreactive nerve fibers in left ventricular myocardium implies that vagal efferent input has little or no direct influence on ventricular contractile function in the guinea pig.

Por ello, nace Cuy’s Burger, hamburguesas basadas en carne de cuy con sabor casero. Teniendo en cuenta que existen muy pocos lugares donde ofrecen este tipo de hamburguesas, decidimos proyectar esta idea de negocio. Además, el análisis de los resultados de las entrevistas, que no solo nos permitió validar el problema que enfrenta el cliente al buscar carne de cuy en presentación de hamburguesas, sino también plantear una serie de soluciones como las alianzas estratégicas con supermercados más conocidos en el ámbito local y plantear supuestos riesgos. En la actualidad, existe una demanda insatisfecha en nuestro mercado local, personas que buscan alimentarse sanamente. Finalmente, nosotros somos un equipo altamente capacitado con una amplia experiencia en diferentes áreas como Finanzas, Negocios Internacionales, Logística, Comercial. Comprehensive Assessment of the Virulence Factors sub 3, sub 6 and mcpA in the Zoonotic Dermatophyte Trichophyton benhamiae Using FISH and qPCR

Skin infections by keratinophilic fungi are commonly referred to as dermatophytosis and represent a major health burden worldwide. Although patient numbers are on the rise, data on virulence factors, their function and kinetics are scarce. We employed an ex vivo infection model based on guinea pig skin explants (GPSE) for the zoonotic dermatophyte Trichophyton (T.) benhamiae to investigate kinetics of the virulence factors subtilisin (sub) 3, sub 6, metallocarboxypeptidase A (mcpA) and isocitrate lyase (isol) at gene level for ten days. Fluorescence in situ hybridization (FISH) and quantitative polymerase chain reaction (qPCR) were used to detect and quantify the transcripts, respectively. Kingdom-spanning, species-specific and virulence factor-specific probes were successfully applied to isolated fungal elements showing inhomogeneous fluorescence signals along hyphae. Staining results for inoculated GPSE remained inconsistent despite thorough optimization. qPCR revealed a significant increase of sub 3- and mcpA-transcripts toward the end of culture, sub 6 and isol remained at a low level throughout the entire culture period. Sub 3 is tightly connected to the de novo formation of conidia during culture. Since sub 6 is considered an in vivo disease marker. However, the presented findings urgently call for further research on the role of certain virulence factors during infection and disease.
