Exploitation de signaux biologiques pour la réalisation de capteurs environnementaux. Application à la construction d'un biocapteur à micro-algues immobilisées et d'une bioélectrode à enzyme immobilisée

Deux biocapteurs, respectivement fondés sur des cellules de micro-algues et sur la polyphénol oxydase (PPO), sont décrits dans cette étude.<br />Le principe du biocapteur algal repose sur le suivi de l'activité photosynthétique des micro-algues par la mesure de la fluorescence chlorophyllienne. Le mode de culture en continu a été choisi afin d'obtenir des suspensions algales dont l'état physiologique des cellules est stable au cours du temps. Les cellules algales sont immobilisées sur- une membrane en fibre de quartz. Le biocapteur comporte cinq membranes algales. L'utilisation simultanée de cinq souches algales différentes permet d'augmenter la représentativité du signal mesuré d'un point de vue statistique ou écotoxicologique. Chlorella vulgaris a été utilisée lors des étapes d'optimisation et de validation du biocapteur. Le comportement de cinq souches algales a été étudié simultanément en l'absence et en la présence de polluants. <br />Le biocapteur enzymatique est fondé sur l'immobilisation de la polyphénol oxydase dans un film électrogénéré de polymère : le poly(3,4-éthylènedioxythiophène) (PEDT). Il permet la détection directe de composés phénoliques et la détection indirecte d'inhibiteurs de cette enzyme. Les conditions d'élaboration et d'utilisation des bioélectrodes ont été optimisées.<br />Par leur spectre de détection et leur sensibilité, ces deux biocapteurs sont des outils complémentaires. Le biocapteur algal est capable de détecter des concentrations en herbicides anti-PSII inférieures au µg.l-1, tandis que la bioélectrode à PPO détecte les composés phénoliques à des concentrations de l'ordre du µg.l-1.

Field assessment of agronomic traits and in vitro acetolactate synthase characterisation of imazapyr herbicide tolerant sugarcane.

Weed control is a major cost for growers in the sugarcane industry, especially for monocotyledonous species such as Cynodon and Rottboellia spp. The introduction of imazapyr-tolerant sugarcane would be advantageous as this herbicide has shown to be effective against the above-mentioned weeds but it also kills sugarcane. In a previous study in our laboratory, several sugarcane putative-mutant lines of variety N12 were generated by in vitro exposure of embryogenic callus to 16 mM ethyl methanesulfonate (EMS), followed by selection on imazapyr-containing medium. Tolerance to a low dose of imazapyr was confirmed in seven of those lines when the herbicide was applied (182 g a.i. ha-1) to 3 month-old plants in pots. The aim of the present study was to identify which of the seven herbicide mutant lines had agronomic characteristics at least equivalent to un-mutated N12. The objectives were to: 1) confirm tolerance to increased rate (312 and 625 g a.i. ha-1) of imazapyr in field plants; 2) measure the agronomic characteristics of these lines; 3) determine the effect of residual soil herbicide activity on germination of sugarcane setts. The seven mutant lines (Mut1-Mut7) and un-mutated N12 were clonally propagated in vitro by shoot multiplication followed by rooting and planted in three plots (untreated, sprayed with 312 or 625 g a.i. ha-1 imazapyr), in the field, in a randomized complete block design. In the untreated control plot there were no significant differences between the control and the mutant plants for agronomic traits (tiller number/plot, stalk height and stalk diameter) or estimated yield (kg/plot) after 10 months, indicating that the mutation process had no effect on general plant phenotype. In the sprayed (312 and 625 g a.i. ha-1) plots, Mut1, Mut4, Mut5, Mut6 and Mut7 plants showed tolerance to imazapyr as the leaves remained green compared with Mut2, Mut3 and N12 control plants, which displayed chlorotic leaves and eventually died in the plot sprayed with 625 g a.i. ha-1. Post-herbicide application, the yields of Mut5, Mut6 and Mut7 (52.33, 43.43 and 41.43 kg/plot, respectively) from the 312 g a.i. ha-1 plot were not significantly different from that of N12 control (53. 61 kg/plot) in the untreated plot. However, in the 312 g a.i. ha-1 plot, the yield and agronomic trait measurements of the untreated N12 control were significantly higher than those of the herbicide-susceptible plants Mut2 and Mut3. Similarly, in the 625 g a.i. ha-1 plot, the recorded yields for Mut4, Mut6 and Mut7 were 41.60, 43.44 and 36.30 kg/plot, respectively, indicating that their imazapyr tolerance and yield characteristics were comparable to the untreated N12 control. Imazapyr is conventionally applied to a fallow field 3-4 months prior to planting sugarcane as there is residual herbicide activity in the soil that suppresses sugarcane germination and growth. Therefore, in order to establish if the herbicide-tolerant mutants could germinate in iii an imazapyr-treated field, 3-budded setts of the mutant lines (Mut1-Mut7) and N12 control were planted in two plots, one unsprayed and one sprayed with 1254 g a.i. ha-1 imazapyr, 2 weeks previously. Germination was calculated after 3 weeks as the number of germinated setts in each plot/no. germinated setts in unsprayed plot x100. In the sprayed plot, the setts from Mut1, Mut4 and Mut6 displayed the highest germination percentages (60, 71 and 74%, respectively) compared with Mut2 (24%), Mut3 (46%), Mut5 (34%), Mut7 (40%) and the N12 control (12%). The in vitro acetolactate synthase (ALS) enzyme activity of 10 month-old plants from the untreated plot was assessed in the presence of 0-30 μM imazapyr to determine the herbicide concentration that inhibited ALS activity by 50% (IC50). The IC50 values for the mutated lines were between 3 and 30 μM, i.e. 1.5-8.8 times more tolerant to imazapyr than the N12 control plants, with Mut6 displaying the highest IC50 value (30 μM). On the basis of the results, it was concluded that Mut1, Mut6 and Mut7 lines were more tolerant to imazapyr than N12 and the other tested lines. Future work includes phenotypically assessing these lines for traits including sucrose content, fibre content, actual yield (tons cane ha-1) and altered pest and disease resistance. Once isolated and sequenced, the ALS gene conferring imazapyr tolerance can be used in genetic bombardment in the genetic modification approach as the gene of interest or as a selectable marker. A invasibilidade do capim-annoni-2 em agroecossistemas tem sido atribuída as suas características biológicas e relações ecológicas. A ecologia da germinação da espécie e a interação alelopática são pouco compreendidas. Este trabalho objetivou determinar a influência de fatores ambientais na germinação, a fitotoxicidade de extratos de capim-annoni-2, identificar e quantificar aleloquímicos fenólicos presentes em suas frações, determinar a influência da degradação da fitomassa e do solo na fitotoxicidade do extrato aquoso, monitorar a degradação dos compostos presentes nos extratos e identificar e quantificar aleloquímicos no solo de área contaminada. Foram realizados experimentos em laboratório utilizando materiais coletados a campo. As sementes germinaram em temperaturas constantes de 15 até 40 °C, mas as percentagens de germinação foram superiores a 94 % apenas em temperaturas alternadas. A exposição à luz aumentou a frequência de plântulas normais. A germinação aumentou com incremento no potencial de água e o número de plantas normais diminuiu com aumento na concentração de cloreto de sódio. A maior taxa de emergência foi observada com as sementes colocadas na superfície do solo, enquanto que nenhuma emergência ocorreu a 4 cm. Foram identificados e quantificados nove compostos fenólicos com potencial aleloquímico nas frações dos extratos de capim-annoni-2: ácido caféico, ácido cumárico, ácido ferúlico, ácido gálico, ácido vanílico, catequina, epicatequina, resveratrol e rutina. A ordem decrescente de fitotoxicidade geral dos extratos brutos no desenvolvimento de plântulas de trigo e alfafa foi acetato de etila, metanólico, aquoso e hexânico. Foi identificada correlação inversamente proporcional entre concentração de ácido vanílico e comprimento de radícula e total de plântulas de alfafa. A simulação da degradação da fitomassa e do solo ocasionou aumento na fitotoxicidade dos extratos sobre a germinação e o desenvolvimento de plântulas, comprovando a liberação dos aleloquímicos e a influência do solo no aumento de sua fitotoxicidade. Extratos etéreos da fitomassa e do solo incubados apresentaram modificações em seus respectivos cromatogramas do primeiro até o sétimo dia de incubação, enquanto a mistura de fitomassa + solo apresentou maiores modificações apenas entre o primeiro e o terceiro dia de degradação. Foram identificados quatro compostos fenólicos com potencial aleloquímico no solo da área infestada com capim-annoni-2: ácido cumárico, ácido ferúlico, catequina e epicatequina. A concentração destes compostos foi maior na superfície do solo e horizontalmente próximo às plantas de capim-annoni-2, evidenciando associação entre as concentrações dos aleloquímicos com a proximidade da planta. / The invasiveness of South African lovegrass in agroecosystems has been attributed to its biological characteristics and the ecological relations. The ecology of species germination and allelopathic interaction are poorly understood. The objective of this work was to determine the influence of environmental factors on germination, phytotoxicity of South African lovegrass extracts, to identify and quantify phenolic allelochemicals present in their fractions, to determine the influence of phytomass and soil degradation on the phytotoxicity of the aqueous extract, to monitor the degradation of the compounds present in the extracts and to identify and quantify allelochemicals in a infested soil. Experiments were carried out in the laboratory using materials collected in the field. Seeds germinated at constant temperatures of 15 to 40 ° C, but the percentages of germination were greater than 94 % only at alternating temperatures. Exposure to light increased the frequency of normal seedlings. Germination increased with increasing water potential and the number of normal plants decreased with increasing sodium chloride concentration. The highest emergence rate was observed with the seeds placed on the soil surface, whereas no emergence occurred at 4 cm. A total of nine phenolic compounds with allelochemical potential were identified in the fractions of extracts of South African lovegrass: caffeic acid, coumaric acid, ferulic acid, gallic acid, vanillic acid, catechin, epicatechin, resveratrol and rutin. The general decreasing order of general phytotoxicity of crude extracts in the development of wheat and alfalfa seedlings was ethyl, methanolic, aqueous and hexane acetate. An inversely proportional correlation was identified between vanillic acid concentration and root and total alfalfa seedlings length. The simulation of phytomass and soil degradation caused an increase in the phytotoxicity of the extracts on germination and seedling development, proving the release of the allelochemicals and the influence of the soil in the increase of its phytotoxicity. The ether extracts of the straw and soil showed changes in their chromatograms from the first to the seventh day of incubation, while the mixture of straw + soil showed greater changes only between the first and third day of degradation. Four phenolic compounds with allelochemical potential were identified in the South African lovegrass infested soil: coumaric acid, ferulic acid, catechin and epicatechin. The concentration of these compounds was higher at the soil surface and horizontally near the South African lovegrass plants, showing an association between concentrations of allelochemicals and plant distance.

Currently, the population density of many pollinators are being reduced to levels that can impair pollination services. The decline of honeybee Africanized (Apis mellifera L.) populations impacts global agricultural production and has a clear impact on both food production and the economy. One of the probable causes for this decline is the indiscriminate use of pesticides. Thus, there was comparative levels of exposure to pesticides among bees (Apis mellifera L.) that are used to pollinate melon crops, bees (Apis mellifera L.) kept in the bush, and Jandaira bees (Melipona subnitida ) kept in the bush. To do this, honey samples were collected from 23 colonies of Apis mellifera used to pollinate melon palntações, 20 colonies of Apis mellifera foraging forest and 10 colonies of Melipona subnitida foraging in the forest. The level of exposure to pesticides was determined by measuring the residual levels of 152 compounds in honey using a multiresidue analysis technique in UFLC-MS system. Honey samples from the present study contained 19 different pesticides, 13 of which were present in honey from bees pollinating melon crops. The levels of some compounds were sufficiently high to promote adverse effects in the bees. Thus, crop pollination offers a great toxicological risk to bees that may contribute to colony collapse / Os polinizadores estão entre os componentes essenciais para o funcionamento dos ecossistemas em geral, pois é essencial para a reprodução e manutenção da diversidade de espécies de plantas e provê alimentos para humanos e animais, influenciando, também, o aspecto quantitativo e qualitativo da produção. Atualmente, a densidade populacional de muitos polinizadores está sendo reduzida a níveis que podem comprometer os serviços de polinização. O declínio das populações de abelhas (Apis mellifera L.) tem impactado na produção agrícola mundial e tem um impacto claro sobre a produção de alimentos e da economia. Uma das causas prováveis para este declínio é o uso indiscriminado de agrotóxicos. Desta forma, fez-se estudo comparativo dos níveis de exposição aos pesticidas entre as abelhas Apis mellifera L. (africanizadas) que são usadas para polinizar plantações de melão, as abelhas Apis mellifera L.(africanizadas) mantidas na caatinga, e as abelhas jandaíra (Melipona subnitida) mantidas na caatinga. Para isso, amostras de mel foram coletadas de 23 colônias de abelhas africanizadas Apis mellifera utilizadas para polinizar plantações de melão, 20 colônias de abelhas africanizadas Apis mellifera que forrageiam de na caatinga e 10 colônias de Melípona subnitida que forrageiam na caatinga. O nível de exposição aos pesticidas foi determinado medindo os níveis residuais de 152 compostos no mel, utilizando uma técnica de análise de multiresíduos no sistema UFLC-MS. As amostras de mel estudadas apresentaram 19 pesticidas diferentes, 13 dos quais estavam presentes no mel de abelhas polinizadoras das plantações de melão. Os níveis de alguns compostos foram suficientemente elevados para promover efeitos adversos em abelhas. This diploma thesis deals with the cultivation of fast-growing wood and concentrates on the influence of particular factors like e.g. location, agrotechnology and fertilizers, which have a crucial impact on the phytomass production. Moreover, the thesis evaluates the economic efficiency of this type of sustainable energy ressource, which can be a source of local business. Introductory theoretical chapters describe essentials rules of the cultivation of fast-growing wood. Furthermore the thesis presents general information in this field and proposes suitable wood clones. In addition, the thesis outlines the process of choice and preparation of the agricultural parcel, the usage of various fertilizes, herbicides and agrotechnology. The applied core characterises the business case itself. The primary research compares financial investment and its return of two examined agricultural parcels under the prerequisit of suitable choice of location and agrotechnology. A trail phase depictes the cultivation of fast-gworing wood and discuss presumptions for succesfull business. This thesis is mainly based on primary research. The acquired data are further analyzed and the final chapter attempts to outline the results and suggest measures.

Hodnocení subchronického působení atrazinu na raka (Cherax destructor)

The evaluation of the sub-chronic exposure to atrazine on crayfish The aim of this study is to evaluate the sub-chronic effect of atrazin on a behaviour, oxidative stress, antioxidant enzyme aktivities and biochemical profile of haemolymph in. These complex data should help to appraise the impact of this substance in the environment. The total test duration was 28 days and was divided into two periods. The first 14 days the crayfish were exposed to two concentrations of atrazine: 6.86 micrograms per liter (ATRenv = environmental concentration in the water in the Czech Republic) and 1.21 milligrams per liter (ATR10% = is coincident to 10% LC50). After the atrazine treatments the depuration 2 weeks phases in water without any chemicals followed. The results indicate that sub-chronic effect of atrazine influenced neither the behaviour of the crayfish nor the level of oxidative stress (measured by TBARS), whereas the changes of superoxiddismutase (SOD) were observed in all tissues (muscles, gills and hepatopancreas). The changes of enzyme activity were observed in catalase (CAT; hepatopancreas and the muscle tissue), glutathione S-transferase (hepatopancreas and the gills tissue), glutathione reductase (GR; the hepatopancreas tissue) and reduced glutathione (the muscle tissue). The influence of ATRenv on the biochemical profile of haemolymph at the following parameters was estimated only for lactate and alkaline, however phosphatase changes made by ATR10% were significant for glucose, ammonia, lactate and alkaline phosphatase measurements. The sub-chronical effect changed the activity of all antioxidant enzymes in hepatopancreas, muscles and the gills tissue of the observed crayfish. The presented results in this study are giving compact information of impact of atrazine on the crayfish and the whole water environment. The suggestion of using the crayfish for tests of toxicity looks like an ideal supplement for triazine herbicide estimations.

In Brazil there are few studies linking pesticide toxicity with fish. For this reason, two experiments were conducted to evaluate possible effects of herbicides on some toxicological parameters in carp (Cyprinus carpio). The fish were exposed to two commercial herbicides, one composed of a mixture formulated imazetapir and imazapic and the second composed of clomazone. The periods of exposure were seven days in the laboratory and seven, 30 or 90 days on rice field condition. After the experimental period, metabolic and toxicological parameters in the liver, brain and muscle tissues were evaluated. The enzymatic parameters analyzed in different tissues of this species were acetylcholinesterase activity (AChE), catalase (CAT) and glutathione S-transferase (GST). Also, oxidative stress parameters, such as carbonyl protein and TBARS levels in hepatic tissue, were analyzed. The metabolic parameters analyzed in liver and muscle of carp were glucose, glycogen, lactate, protein, amino acids and ammonia. Moreover, metabolic assessments were made (glucose, lactate and protein) in carp plasma. The results showed that AChE activity was increased in carp brain after seven days of exposure to a commercial formulation containing imazethapyr and imazapic herbicides both under field and laboratory conditions. In the same period of exposure, muscle tissue showed reduced activity of AChE after exposure to all herbicides (imazetapir and imazapic and clomazone) under laboratory conditions. At 30 days, in the field, AChE activity was reduced in brain and enhanced in muscle of carps exposed to imazethapyr and imazapic. Fish exposed to clomazone herbicide did not show any change in the activity of this enzyme in this period. On the other hand, at 90 days of exposure, only the fish exposed to the herbicide clomazone, showed a reduction in muscle AChE activity. The antioxidant enzyme catalase showed increased activity in liver tissue after seven days of exposure under both experimental conditions. At 30 days of exposure, catalase activity showed no further changes and after 90 days of exposure activity was reduced in liver of carp exposed to clomazone herbicide. The enzyme glutathione S-transferase was altered only after 30 and 90 days of exposure in the field. GST activity was reduced in liver tissue after exposure to both herbicides tested. TBARS levels were increased in almost all periods, terms of exposure and tissues considered, with the exception of brain tissue, which presented a decrease in TBARS levels at 30 days of exposure. Protein carbonyl was reduced in liver of carp exposed to clomazone herbicide after seven days under laboratory conditions. Moreover, at 30 and 90 days of exposure, the protein carbonyl levels were increased in the liver of fish exposed to both herbicides. After exposure to both herbicides, the carp showed metabolic disorders. In general, protein and amino acid levels were reduced and the amount of glycogen was enhanced in liver and muscle of carp exposed to both herbicides at all times tested. However, ammonia, lactate and glucose levels were increased and reduced according to the tissue considered, time of exposure and herbicide tested. These results indicate that the parameters measured may be good indicators of contamination of these commercial herbicides in Cyprinus carpio. / Os pesticidas podem afetar parâmetros toxicológicos e ainda causar estresse oxidativo em peixes. No Brasil existem poucos estudos relacionando toxicidade de pesticidas com peixes. Por esta razão foram conduzidos dois experimentos com objetivo de avaliar possíveis efeitos dos herbicidas sobre alguns parâmetros toxicológicos em carpas (Cyprinus carpio). Os peixes foram expostos a dois herbicidas comerciais, um composto pela mistura formulada de imazetapir e imazapic e o segundo composto por clomazone. Os períodos de exposição foram de sete dias em laboratório e por sete, 30 ou 90 dias em condição de lavoura de arroz. Após os períodos experimentais, nos tecidos hepático, cerebral e muscular foram avaliados parâmetros metabólicos e toxicológicos. Os parâmetros enzimáticos analisados foram a atividade da acetilcolinesterase (AChE), catalase (CAT) e glutationa S-transferase (GST) em diferentes tecidos desta espécie. Além disso, analisaram-se parâmetros de estresse oxidativo, como a carbonilação de proteínas e níveis de TBARS no tecido hepático. Os parâmetros metabólicos analisados foram glicose, glicogênio, lactato, proteína, amônia e aminoácidos em fígado e músculo de carpas. Ademais, foram feitas avaliações metabólicas (glicose, lactato e proteína) em plasma de carpas. Os resultados mostraram que a atividade da AChE foi aumentada em cérebro de carpas, após sete dias de exposição a formulação comercial contendo os herbicidas imazetapir e imazapic, em condições de campo e laboratório. Neste mesmo período de exposição, o tecido muscular mostrou redução da atividade da AChE após exposição as duas formulações comerciais em laboratório. Aos 30 dias em campo, a atividade da enzima AChE foi reduzida em cérebro e aumentada em músculo de carpas expostas a formulação contendo a mistura dos herbicidas imazetapir e imazapic. Os peixes expostos ao herbicida clomazone não mostraram alteração na atividade desta enzima, neste período. Já aos 90 dias de exposição, somente os peixes expostos ao herbicida clomazone, mostraram uma redução na atividade da AChE muscular. A enzima antioxidante catalase mostrou sua atividade aumentada no tecido hepático após sete dias de exposição em ambas as condições experimentais e às duas formulações comerciais testadas. Aos 30 dias de exposição, a enzima não foi alterada e após 90 dias de exposição a sua atividade foi reduzida em fígado de carpas expostas ao herbicida clomazone. A enzima glutationa S-transferase só foi alterada após 30 e 90 dias de exposição em campo. Sua atividade foi reduzida no tecido hepático após exposição às formulações comerciais dos herbicidas testados. Os níveis de TBARS foram aumentados em praticamente todos os períodos, condições de exposição e tecidos considerados, com exceção do tecido cerebral que aos 30 dias de exposição, mostrou os níveis de TBARS diminuídos. Os níveis de proteína carbonil foram diminuídos em fígado de carpas expostas por sete dias ao herbicida clomazone em laboratório. Por outro lado, aos 30 e 90 dias de exposição os níveis de proteína carbonil foram aumentados no fígado de peixes expostos as duas formulações comerciais. Após exposição aos dois herbicidas comerciais, as carpas demonstraram desordens metabólicas. De uma maneira geral, os níveis de proteína e aminoácidos foram diminuídos e o conteúdo de glicogênio aumentado em fígado e músculo de carpas expostas aos dois herbicidas comerciais e tempos testados. Porém, os níveis de amônia, lactato e glicose mostraram alterações variáveis de acordo com o tecido considerado, com o tempo de exposição e com o herbicida testado. Estes resultados indicam que os parâmetros medidos podem ser bons indicadores da contaminação destes herbicidas comerciais em Cyprinus carpio.
